Er, Y:CaF 2 particles were synthesized using the chemical precipitation method, and as-synthesized particles were calcined at different temperatures. Fourier-transform infrared spectra confirmed the elimination of residual nitrates from Er, Y:CaF 2 phosphors calcined at 600 °C. Measurements revealed that the calcined Er, Y:CaF 2 phosphors displayed a single cubic-fluorite crystal structure with regular polygon shapes. We investigated the sensitivity effects of introducing non-active Y 3+ ions as buffers on the luminescence characteristics of Er, Y:CaF 2 phosphors under 488 nm and 980 nm excitation. Two emission bands were detected in luminescence spectra. For phosphors under 980 nm excitation, a three-photon UC mechanism is identified to be involved in the generation of green and red emissions. The findings provide insights into enhancing the 2 H 11/2 / 4 S 3/2 → 4 I 15/2 transitions in Er, Y:CaF 2 phosphors by introducing Y 3+ ions as buffers.

A series of lanthanide-containing water-resistant nitrates, namely RE(OH)2NO3 (RE = Tb(1), Dy(2), Ho(3), and Er(4)), was obtained through the hydrothermal process. As possible nonlinear optical materials, they feature a layered isomorph composed of an [REO3(OH)6] polyhedron and an [NO3] triangle, and the synergistic arrangement of the [REO3(OH)6] and [NO3] groups in their structures leads to their obvious second-order nonlinear optical effect. Nevertheless, the unique optical absorption caused by the electronic transitions on 4f–4f orbitals of lanthanides results in their second harmonic generation responses of different strengths, with 1 exhibiting 5.07 times that of KDP, but 2–4 showing less than half of KDP. In addition, 1 possesses an outstanding water-resistant capacity and a transparent cut-off edge around 300 nm, foreshadowing its potential value as a nonlinear optical crystal. Moreover, 1 is found to emit characteristic green fluorescence due to the typical 5D4 → 7F5 transitions of the excited Tb3+ ions

Graphical NaGdF 4 ,Yb,Er@NaGdF 4 ,Yb,Tm@NaYF 4 core@shell@shell three-layer structure of upconversion nanoparticles coated with Fe−TCPP metal-organic frameworks nanocomposite was designed and constructed for multimodal imaging and synergetic chemodynamic therapy/photodynamic therapy of tumors. Here, NaGdF 4 ,Yb,Er@NaGdF 4 ,Yb,Tm@NaYF 4 core@shell@shell three-layer structure of upconversion nanoparticles (UCNPs) coated with Fe−Tetrakis (4-carboxyphenyl) porphine (TCPP) metal-organic frameworks (Fe-MOFs) nanocomposite (UCNPs@MOFs) was designed and constructed for multimodal imaging and synergetic chemodynamic therapy (CDT)/photodynamic therapy (PDT) of tumors. The UCNPs@MOFs were successfully applied for tumor cells imaging in vitro and in vivo in near-infrared (NIR) region. The doped Gd was used as contrast agent for the magnetic resonance imaging (MRI) of mouse tumors. The luminescence in the UV-vis region was absorbed by the Fe-MOFs to produce singlet oxygen ( 1 O 2 ) for PDT. The Fe 3+ doped in the MOFs can catalyze H 2 O 2 to produce oxygen and hydroxyl radical (⋅OH). Hydroxyl radical is used in CDT and cooperates with the 1 O 2 of PDT. Based on the CDT/PDT synergistic effects, the UCNPs@MOFs nanocomposite had obviously enhanced tumor inhibitory efficiency in vivo. These results described that the asprared UCNPs@MOFs nanocomposite have great potential in the effective multimodal imaging and treatment of tumors.

Bisphenol A (BPA) is a widely used endocrine disruptor , which has attracted much attention due to its harmful effects on male reproduction. To investigate the interference of BPA on steroid synthesis in males, male rare minnows ( Gobiocypris rarus ) were exposed to 15 μg L −1 BPA for 7, 14 and 21 d. Meanwhile, a positive control group was performed with 25 ng L −1 17α-ethynyl estradiol (EE2). Results showed that BPA exposure induced lower testosterone (T) levels, while affecting the transcripts of steroidogenic gene StAR . Moreover, BPA induced abnormal germ cells proliferation in the testis in rare minnow. Transcriptome analysis showed that 354 transcripts significantly differentially expressed after BPA exposure for 14 d, several of them were enriched in the signaling pathways of cell cycle process, PPAR signaling pathway, the steroid synthesis pathway and estrogen signaling pathway. BPA significantly increased estrogen receptor (ER) levels and induced abnormal protein levels of PPARγ. BPA disrupted the StAR expression by interfering ER enrichments within StAR 5′ flanking region . Additionally, our study also revealed that BPA and EE2 might have different mechanisms for interfering with steroid hormone levels and germ cells proliferation in the testis.

Upconversion (UC) luminescence materials with dynamically adjustable activity under external stimuli are of great interest owing to their potential applications in optical storage and high-sensitivity optical switches. Recently, photochromic (PC) effect was considered as an alternate way to regulate the UC luminescence of inorganic materials; however, external-field-induced bleaching was routinely completed by single stimulation, which limited its practical application. In this work, one kind of smart phosphor, Er-activated Bi7Ti4NbO21 intergrowth bismuth layer-structured ceramics that exhibited both intense UC green emission and prominent PC reaction, was synthesized. The UC luminescence, PC effect, and their coupling properties were researched. Interestingly, light–heat and light–light dual-controlled UC luminescence and PC processes were realized. Accompanying the PC effect, the UC luminescence can be reversibly modified by alternating 405 and 532 nm light stimulation (or thermal stimulation). Under 405 nm irradiation, the UC luminescence quenching ratio can reach up to 80.8%. Meanwhile, the UC luminescence raise ratio can respectively reach up to 426.9 and 118.1% under 250 °C thermal treatment and 532 nm light bleaching. The UC luminescence modulation was exposed to be tightly related to the irradiation coloration and thermal/light decoloration processes. Good fatigue resistance of the UC luminescence switching was also achieved. The mechanisms related to the UC luminescence modulation and PC process were discussed. These results indicated that Er-activated Bi7Ti4NbO21 samples can be adopted as a kind of optical data memory materials.

Yb2(MoO4)3:Ln3+ (Ln = Ho, Tm, Er) phosphors were synthesized by a hydrothermal method with further calcination. The crystal structure and morphology of the samples were characterized by X-ray diffraction and scanning electron microscopy. The effects of rare earth ion interaction in the Yb2(MoO4)3:Ln3+ phosphors on the upconversion (UC) luminescence properties and energy transfer process at different doping concentrations were discussed. It is concluded that by doping Ho3+, Tm3+, and Er3+ in the Yb2(MoO4)3 host, the phosphors emit orange-yellow, blue and green light, respectively, and the energy transfer from Yb3+ → Ho3+/Er3+ and Yb3+ → Tm3+ is a two and three photon process, respectively. Under the excitation of a 980 nm laser, by changing the Ho3+/Tm3+ ratio in doped Yb2(MoO4)3, we have realized the transition of phosphors from cold white light to warm white light. The temperature sensitivity of the 2H11/2 and 4S3/2 energy levels of Yb2(MoO4)3:Er3+ in the range of 323–573 K was studied by the FIR technology. The experimental results show that the maximum absolute sensitivity was 0.0081 K−1 at 573 K and the maximum relative sensitivity was 0.01 K−1 at 323 K. Compared with other materials, the Er3+ doped Yb2(MoO4)3 phosphor is an excellent optical temperature sensing material.

Temperature is one of the physical parameters that affects the emission luminescence properties of upconversion (UC) materials. Temperature-dependent emission color tuning and good temperature sensitivity in multifunctional β-NaYF4:Yb/Er@β-NaYF4:Yb/Tm microcrystals were achieved under 980 nm excitation. Based on the different emission bands (blue, green and red bands) and spectral sensitivities of the core–shell structured microcrystals to temperature, intense white-light emission was obtained at room temperature and a real-time naked-eye recognizable emission color change from white to the purplish-pink region was realized with the temperature increasing from 300 to 500 K. Furthermore, optical temperature sensing properties of the sample with good sensitivity (from 0.0034 to 0.0044 K−1) were investigated by using a fluorescence intensity ratio between 521 nm (2H11/2 → 4I15/2 transition) and 540 nm (4S3/2 → 4I15/2 transition) in the temperature range from 300 to 500 K. The results indicate that the sample has high sensitivity in a relatively wide range of temperature intervals, especially in the low temperature region. We believe that the multifunctional core–shell structured microcrystals exhibit promising applications in white-light emitting diodes, the anti-counterfeiting field, and temperature sensing.

Endoplasmic reticulum (ER) stress is a cellular state that results from the overload of unfolded/misfolded protein in the ER that, if not resolved properly, can lead to cell death. Both acute lung infections and chronic lung diseases have been found related to ER stress. Yet no study has been presented integrating metabolomic and transcriptomic data from total lung in interpreting the pathogenic state of ER stress. Total mouse lungs were used to perform LC–MS and RNA sequencing in relevance to ER stress. Untargeted metabolomics revealed 16 metabolites of aberrant levels with statistical significance while transcriptomics revealed 1593 genes abnormally expressed. Enrichment results demonstrated the injury ER stress inflicted upon lung through the alteration of multiple critical pathways involving energy expenditure, signal transduction, and redox homeostasis. Ultimately, we have presented p-cresol sulfate (PCS) and trimethylamine N-oxide (TMAO) as two potential ER stress biomarkers. Glutathione metabolism stood out in both omics as a notably altered pathway that believed to take important roles in maintaining the redox homeostasis in the cells critical for the development and relief of ER stress, in consistence with the existing reports.

Septic cardiomyopathy is associated with mitochondrial damage and endoplasmic reticulum (ER) dysfunction. However, the upstream mediator of mitochondrial injury and ER stress has not been identified and thus little drug is available to treat septic cardiomyopathy. Here, we explored the role of B-cell receptor-associated protein 31 (BAP31) in septic cardiomyopathy and figure out whether melatonin could attenuate sepsis-mediated myocardial depression via modulating BAP31. Lipopolysaccharide (LPS) was used to establish the septic cardiomyopathy model. Pathway analysis was performed via western blot, quantitative polymerase chain reaction and immunofluorescence. Mitochondrial function and ER stress were detected via enzyme-linked immunosorbent assay, western blot, and immunofluorescence. After exposure to LPS, cardiac function was reduced due to excessive inflammation response and extensive cardiomyocyte death. Mechanistically, melatonin treatment could dose-dependently improve cardiomyocyte viability via preserving mitochondrial function and reducing ER stress. Further, we found that BAP31 transcription was repressed by LPS whereas melatonin could restore BAP31 expression; this effect was dependent on the MAPK-ERK pathway. Inhibition of the ERK pathway and/or knockdown of BAP31 could attenuate the beneficial effects of melatonin on mitochondrial function and ER homeostasis under LPS stress. Altogether, our results indicate that ERK-BAP31 pathway could be used as a critical mediator for mitochondrial function and ER homeostasis in sepsis-related myocardial injury. Melatonin could stabilize BAP31 via the ERK pathway and thus contribute to the preservation of cardiac function in septic cardiomyopathy.

Purpose It is demonstrated that unsaturated fatty acids can counteract saturated fatty acids-induced lipotoxicity, but the molecular mechanisms are unclear. In this study, we investigated the protective effects of monounsaturated oleic acid (OA) against saturated palmitic acid (PA)-induced cytotoxicity in rat β cells as well as islets, and mechanistically focused on its regulation on endoplasmic reticulum (ER) stress. Methods Rat insulinoma cell line INS-1E cells and primary islets were treated with PA with or without OA for 24 h to determine the cell viability, apoptosis, and ER stress. SD rats were fed with high-fat diet (HFD) for 16 w, then, HFD was half replaced by olive oil to observe the protective effects of monounsaturated fatty acids rich diet. Results We demonstrated that PA impaired cell viability and insulin secretion of INS-1E cells and rat islets, but OA robustly rescued cells from cell death. OA substantially alleviated either PA or chemical ER stressors (thapsigargin or tunicamycin)-induced ER stress. Importantly, OA attenuated the activity of PERK-eIF2α-ATF4-CHOP pathway and regulated the ER Ca 2+ homeostasis. In vivo, only olive oil supplementation did not cause significant changes, while high-fat diet (HFD) for 32 w obviously induced islets ER stress and impaired insulin sensitivity in SD rats. Half replacement of HFD with olive oil (a mixed diet) has ameliorated this effect. Conclusion OA alleviated PA-induced lipotoxicity in INS-1E cells and improved insulin sensitivity in HFD rats. The amelioration of PA triggered ER stress may be responsible for its beneficial effects in β cells.

Ethnopharmacological relevance Oxidative damage to osteoblasts was a key factor in the development of osteoporosis. Er-Xian Decotion (EXD) is widely used in China for the treatment of osteoporosis, which has a variety of antioxidant active ingredients. EXD may be an important source of protection against oxidative damage in osteoblasts, but the anti-osteoporotic active components of EXD is currently unclear. Aim of the study This work established an effective and reliable drug screening method to find main active ingredients in EXD (M-EXD) that can protect osteoblasts against oxidative stress and achieve anti-osteoporosis effects. Materials and methods H 2 O 2 -induced osteoblast cell fishing with UHPLC-QTOF/MS was firstly used to discover the potential active components from EXD. Afterword, the EXD compound-osteoporosis target network was constructed using network pharmacology , thus potentially anti-osteoporosis ingredients were founded, and their combination were defined as the M-EXD. Finally, pharmacology effects of M-EXD was evaluated by ovariectomized rats, prednisolone induced-zebrafish and H 2 O 2 -induced osteoblasts. Results 40 candidate active ingredients in EXD were initially screened out via pathological cell fishing. According to network pharmacology result, M-EXD consisted of 13 ingredients since they had a close relationship with 65 osteoporosis-related targets. Pharmacological evaluation showed that M-EXD significantly ameliorated oxidative stress in H 2 O 2 -induced osteoblast model, evidently reversed the activity of ALP , ROS , GSH-px, NO and MDA compared with the model group. M-EXD showed better anti-oxidative activities than individual ingredients, presenting obvious synergetic effects. In osteoporosis rat and zebrafish models, M-EXD also demonstrated good anti-osteoporotic properties by mitigating the osteoporosis bone loss and increasing serum bone morphogenetic protein 2 , and reversing osteocalcin expression in bone tissue. It significantly ameliorated oxidative stress in the in-vivo models. Moreover, M-EXD and EXD showed similar anti-osteoporotic and anti-oxidative properties, while the rest components of EXD had no satisfactory anti-osteoporotic efficacy. Conclusions Our work successfully identified the main active components in EXD, which could represent the efficacy of EXD on treating osteoporosis, and meanwhile, it also provided an effective strategy to investigate active ingredients from natural medicines, which might be helpful for drug development and application.

Autophagic degradation of the endoplasmic reticulum (ER-phagy) has been found to play a critical role in human sensory neuropathy. So far, however, specific and efficient intervention means for ER-phagy remain unexplored. Herein, brefeldin A (BFA), a blocking agent on protein transport between the ER and Golgi, was screened from ER stress inducers. BFA was then delivered to the perinuclear area co-localized with the ER by a mesoporous silica nanoparticle-based drug-carrier functionalized with autophagy-inducing peptides of TAT-beclin 1 (MSNs-BFA), to evoke a perturbation of ER-phagy. The molecular mechanism of ER-phagy regulated by BFA was explored by biochemical evaluation including time-lapse live-cell fluorescence imaging. We found that MSNs-BFA treatment caused a lower mRNA/protein expression level of FAM134b even under a compensation of autophagic flux in U2OS cells, and resulted in ER-expansion. The fragmentation of the ER was blocked as a response to ER stress mediated by inactivation of the AKT/TSC/mTOR pathway. Our work developed an efficient external manipulation strategy to regulate ER-phagy and may contribute to the therapeutic application of autophagy-related major human diseases.
