Manganese induces tumor cell ferroptosis through type-I IFN dependent inhibition of mitochondrial dihydroorotate dehydrogenase

Shanlong Zhang,Li Kang,Xiaoxue Dai,Junlan Chen,Zhengtao Chen,Meixiang Wang,Huantong Jiang,Xin Wang,Suqin Bu,Xinyuan Liu,Guohui Zhang,Hua Tang

Ferroptosis is a novel form of regulated cell death characterized by the iron-dependent accumulation of lipid peroxides to lethal levels, which is morphologically, biochemically, and genetically distinct from apoptosis, necroptosis, autophagy, and pyroptosis. Manganese play an important role in innate immunity and antitumor immunity. Many manganese-based nanomaterials induce tumor cell death by catalyzing the production of reactive oxygen species (ROS) within the tumor. However, the exact underlying mechanisms remain unclear. As research on ferroptosis advances and its regulatory mechanisms in tumors continue to be refined, more evidence has suggested that triggering ferroptosis in tumor cells is an effective strategy for tumor treatment. In this study, we found that administration of MnCl 2 to tumor cells resulted in lipid peroxidation and increased the levels of mitochondrial ROS, consequently leading to ferroptosis. Dihydroorotate dehydrogenase (DHODH)-mediated ferroptosis defence is a targetable vulnerability in cancer. We show that MnCl 2 downregulated DHODH expression in tumor cells, resulting in increased mitochondrial ROS and lipid peroxidation to induce ferroptosis. In addition, MnCl 2 enhanced the phosphorylation levels of STING, TBK1, and IRF3 and upregulated the expression of type-I interferon (IFN), produced by the cGAS–STING signaling pathway. When inhibiting the cGAS–STING signaling pathway or type-I IFN, DHODH expression was restored, reversing lipid peroxidation and ROS production and rescuing MnCl 2 -induced ferroptosis.. Knockout of IFNAR1 or overexpression of DHODH weakens the antitumor effect of MnCl 2 . Mechanistically, these results revealed that Manganese treatment-activated cGAS–STING signaling promote mitochondrial lipid peroxidation and ROS production by releasing type-I IFNs that reduce DHODH function and thereby inducing ferroptosis in tumor cells. This may provide a new strategy to complement existing antitumor treatment regimens.

Development of Stable, Efficient, and Recyclable Amine-Containing Microspheres for Gold(I) Thiosulfate Complex Recovery

Li Zhao,Xianzhi Hu,Futing Zi,Shuliang Chen,Huiling Cheng,Peng Yang,Yan Zhang,Yunlong Chen,Yunshu Jiang,Xinrong Li,Yue Lin,Zhujuan Li,Juan Li,Haiwen Wang,Yufang Li

It is desirable to develop a highly stable, cost-effective, and recyclable adsorbent to adsorb Au(I) from thiosulfate solutions. In this study, reliable aminated microspheres (PS–TETA) were synthesized via a one-pot method that profited from active chloromethyl group-induced nucleophilic substitution between chloromethylated polystyrene (PS–Cl) and triethylenetetramine (TETA). These microspheres were used to recover a gold(I) thiosulfate complex. Scanning electron microscopy, Fourier transform infrared spectroscopy, Brunauer–Emmett–Teller technique, and X-ray photoelectron spectroscopy were adopted to analyze the surface elements and structure of PS–TETA. Additionally, influencing parameters, such as pH, thiosulfate concentration, and temperature, were investigated. The results indicated that the synthetic process and gold-adsorption behavior of PS–TETA was directly related to the surface chlorine and amine groups. The adsorption process was exothermic and consistent with the Freundlich isotherm model and pseudo-second-order kinetics. Complete adsorption of Au(I) was achieved with a maximum capacity of 23.79 kg t–1. Na2SO3 proved to be an effective desorbent and exhibited a satisfactory adsorption ability of the adsorbent even after five cycles. Anion exchange as a reasonable Au(S2O3)23– adsorption mechanism of PS–TETA is proposed. This study offers a practical method for synthesizing aminated microspheres with excellent properties to recover Au(I) from thiosulfate solutions.

MXene-mediated electron transfer in Cu(II)/PMS process: From Cu(III) to Cu(I)

Haoran Song,Shiting Pan,Yuwei Wang,Yang Cai,Wei Zhang,Yongming Shen,Changping Li

There exists an obvious contradiction between Cu(III) generation and Cu(I) contribution in reductants enhanced Cu(II)/peroxymonosulfate (PMS) process because the involved electron transfer pathways were always ignored in previous studies. This study investigated the MXene-mediated electron transfer mechanism in Cu(II)/PMS for the first time. The introduction of MXene significantly promoted atrazine degradation under acidic condition because the in-situ generation of Cu(I) was an excellent activator for PMS. Under alkaline condition, although addition of MXene generated a large amount of Cu(I), the degradation of atrazine was inhibited due to that the conversion of Cu(I) to Cu(III) was interrupted. The strong metal-support interaction (SMSI) altered the reactive intermediate species from Cu(III) (two-electron transfer) to hydroxyl radical (HO • ) and sulfate radical (SO 4 •- ) (one-electron transfer) under alkaline condition. This work proposed a novel MXene-mediated electron transfer mechanism and improved the understanding of Cu(III)/Cu(I) transformation pathway.

Stabilization and Fluorescence Enhancement of CsPbX3 (X = Cl, Br, I) Perovskite Nanocrystals with Tungstosilicic Acid

Shuoshuo Zang,Guoqing Xu,Renci Yang,Qinyi Zhu,Hewen Liu

It is still challenging to achieve both good stability and high PLQYs for perovskite NCs because of their ionic nature and structural defects. In this work, tungstosilicic acid (TSA) is used for the surface passivation of cesium lead halide perovskite NCs. Near-unity PLQY is achieved for CsPbBr3 NCs after surface passivation with a suitable amount of TSA. XRD in conjunction with XPS analysis shows that TSA binds hydrophobic chains of OAm onto the surface of CsPbBr3 NCs. XPS results also demonstrate that TSA passivation can change the charge distribution of the cations on the surface layers, which can enhance the interaction with surface traps. PL decay analysis reveals that trap states arising from the surface defects are significantly decreased after TSA passivation. TSA-passivated CsPbBr3 NCs demonstrate long-term ambient stability and good resistances to polar solvents and maintain about 80% of their initial PL intensity after storage under ambient conditions for 50 days. It is further found that TSA passivation is able to improve the PLQYs of a broad spectrum of CsPbX3 (X = Cl, Br, I) NCs without hindering the tunability of the band gaps and PL colors of CsPbX3 NCs via anion exchange.

Photocatalytic Destruction of Gaseous Benzene Using Mn/I-Doped TiO2 Nanoparticle Catalytic Under Visible Light

Yue Sun,Lijiang Tian,Bingkun Liu,Fulong Chen,Sijie Ge

In this article, Mn/I-TiO2 nanocatalyst was prepared by sol-gel method and used as the photocatalyst to discompose the gaseous benzene in the visible light. The crystal phase composition and grain size of the photocatalyst were investigated by X-ray diffraction and Raman, respectively. X-ray photoelectron spectroscopy and ultraviolet-visible diffuse reflectance spectroscope were used to study the surface elements and bandgap width of the photocatalyst. The research findings suggested that the crystal type of all catalysts was anatase. TiO2 nanoparticle doped by Mn/I could reduce its crystallite size and augment lattice distortion because of the formation of oxygen vacancies. The generation rate, migration rate, and disintegration rate of electron-hole pairs were increased by modification of TiO2. Meanwhile, the Mn-TiO2 was recognized with the smallest bandgap among four kinds of catalyst. Mn-TiO2 catalyst exhibited the highest efficiency on gaseous benzene removal (37.86%) among pure-TiO2 (3.33%), I-TiO2 (30.21%), and Mn-I-TiO2 (31.56%) under visible light. Correspondingly, the photocatalytic ability of pure-TiO2, I-TiO2, Mn-I-TiO2, and Mn-TiO2 was 51.25 μg C6H6/(g·h), 424.36 μg C6H6/(g·h), 493.61 μg C6H6/(g·h), and 245.43 μg C6H6/(g·h), respectively.

Harmonious K–I–O co-modification of g-C3N4 for improved charge separation and photocatalysis

Shifei Kang,Zhihao Zhang,Maofen He,Zirou Fang,Di Sun,Lulu Zheng,Xijiang Chang,Lifeng Cui

Co-modification of graphitic carbon nitride (g-C3N4) photocatalysts can maximally optimize their intrinsic photoelectric structures, but usually involves complex multistep reactions, and thus is challenging because structural collapse and active site decay usually happen due to the complex process, over-treatment or conflict of different modification strategies. In this work, potassium iodate (KIO3), an edible salt additive, was used as a critical waste-free modifier in the thermal polymerization of g-C3N4, leading to a fast and coordinated co-modification of g-C3N4 photocatalysts. The final g-CN-KIO3 photocatalyst demonstrated a superior and balanced optical absorption ability and semiconductor band structure. In visible-light-driven photocatalytic model tests, g-CN-KIO3 showed a 3.25 times higher Cr(VI) photoreduction rate and five orders of magnitude lower residual bacteria in a photocatalytic sterilization within 120 min compared with raw g-C3N4. The photocatalytic performance of g-CN-KIO3 also outperformed all of the controls without maximized K–I–O co-modification (CN-K, CN-I, CN-O, and CN-KI). Photocurrent and electrochemical impedance experiments suggest that the co-modified photocatalysts have a faster charge separation ability, which is attributed to the multiple K–I–O co-modifications inducing an optimized surface electronic structure and slower carrier recombination. This work provides a new way to simplify and coordinate complex multi-factor regulation of materials.

Synthesis and characterization of a luminescent diphosphine and aromatic N-heterocyclic copper(I) coordination polymer

Ming-Ming Wang,Xiaofei Kuang,Ting-Ting Li,Hui Yang,Ying-Hao Mi,Ying-Chao Xu,Can-Zhong Lu

Luminescent Cu(I) complexes have drawn considerable attention as alternative materials to precious transition metal complexes due to their low cost and electron configuration. Discrete heteroleptic diamine-diphosphine copper(I) complexes have been investigated owing to their outstanding photophysical features. However, heteroleptic copper(I) coordination polymers remain mostly unexplored. Herein, we report the synthesis and photophysical characterization of a heteroleptic copper(I) coordination polymer by employing bidentate oxybis(2,1-phenylene)bis(diphenylphosphine) (POP) diphosphine and nitrogen heterocyclic 5-[4-(1-imidazolyl)phenyl]-2H-tetrazole (ipt) as ligands. By utilizing the mixed-solvent strategy, a one-dimensional (1D) linear chain was constructed under solvothermal reaction conditions. Temperature-dependent emission spectra combined with decay time measurements demonstrate that emission originated from triplet phosphorescence at ambient temperature.

From Type‑I to S‑Scheme: Coordination-bridged CuInS₂-CdS heterojunction for enhanced photocathodic protection

Shaoming Yang,Yaqi Wang,Wenling Zha,Leyan Xiong,Minyang Dai,Yongxin Liu,Ji Zhao,Xi Chen,Liangpeng Zeng,Haijiao Xie

Coordination CIS-EDTA-CdS S-scheme heterojunction for PEC cathodic protection. • EDTA: bifunctional layer, passivating defects and modulating band positions. • S-scheme charge transfer enhances carrier separation and suppresses recombination. The development of photoanodes combining broad-spectrum light response with efficient charge separation is crucial for photoelectrochemical cathodic protection. In this work, an atomic-scale S-scheme heterojunction was constructed between narrow-bandgap CuInS₂ (CIS) and CdS via coordination-bond interface engineering. Specifically, EDTA was first employed as a small-molecule bridging agent to passivate dangling bonds and surface defects on CIS through coordination interactions. Subsequently, CdS was grown in situ onto the CIS-EDTA surface, yielding the CIS-EDTA-CdS composite photoanode. Results demonstrate that EDTA modification not only suppresses non-radiative recombination but also directionally tailors the band structure of CIS, transforming the heterojunction from a Type-I to an S-scheme configuration. The optimized composite photoanode achieves a photocurrent density of 1.3 mA·cm⁻² under light and shifts the corrosion potential of the coupled 304 stainless steel to −1.15 V (vs. NHE). Mechanistic investigations reveal that the S-scheme charge-transfer pathway promotes the recombination of conduction-band electrons from CdS with valence-band holes from CIS-EDTA, while preserving the high-energy electrons on the CIS-EDTA side for effective cathodic protection. This study provides a new paradigm for the application of atomic-scale interface engineering in photoelectrochemical anticorrosion, highlighting the potential of coordination-bridge strategies in designing high-performance photoanodes. Multifunctional Ca2+ nanomodulators regulate type I interferon pathway to remodel tumor microenvironment for cancer immunotherapy

Shulin Qin,Lichun Ma,Runfeng Li,Pengfei Yuan,Yunfeng Shi,Xin Ji,Wei Xue,Yanli Li,Wen Liu

Although immune checkpoint blockade (ICB) therapy has been widely used for the treatment of a variety of cancers, its therapeutic efficacy is severely impaired by low tumor immunogenicity and the immunosuppressive microenvironment. Herein, we designed multifunctional immunomodulatory calcium nanoparticles to reshape the immunosuppressive tumor microenvironment. Our multifunctional calcium nanoparticles, CD@CaCO 3 P, are designed to release calcium ions and doxorubicin (DOX) into the acidic tumor microenvironment, which synergistically increase the tumor cell calcium ion concentration, while the release of combretastatin A4 phosphate (CA4P) further enhances T cell infiltration. Specifically, CD@CaCO 3 P effectively mediates the immunogenic cell death (ICD) of tumor cells and triggers the activation of nuclear factor kappa-B (NF-κB)/interferon regulatory factor 3 (IRF3), thereby regulating type I interferon (IFN-I) signaling pathways, and ultimately reprogramming the immunosuppressive tumor microenvironment by inducing M1-like macrophage polarization, promoting dendritic cell maturation, and increasing the infiltration of CD8 + T cells. Furthermore, CD@CaCO 3 P significantly increased tumor susceptibility to α-PD-1 checkpoint blocking immunotherapy and enhanced systemic immunotherapy performance against both primary and metastatic tumors. Our work provides new insights for the use functional calcium nanoparticles that modulate the tumor microenvironment to enhance the efficacy of cancer immunotherapy.

A facile method to synthesize nZVI-doped polypyrrole-based carbon nanotube for Ag(i) removal

Wen-Juan Zhang,Ye-Zhi Ding,Ya-Xian Wang,You-Liang Wang,Yu-Long Fei,Meng-Yu Ran

The pollution of silver ion (Ag( i )) has become a serious environmental threat and hazard to ecosystem and human health. Thus, the nanoscale zero valent iron (nZVI)-doped polypyrrole-based carbon nanotube (nZVI/CNT) composites were synthesized by a facile method to remove Ag( i ) from wastewater due to the toxicity and scarcity nature of Ag( i ). In this process, Fe 3+ initiated the self-assembly of polypyrrole tubes in the presence of methyl orange, while it also served as an iron source generated nZVI/CNTs by carbothermal reduction method. The nZVI/CNT composites exhibited a homogeneous tubular structure, and the nZVI formed were uniformly dispersed in the nZVI/CNT composites. The nZVI/CNT composites were used as an adsorbent for the removal of Ag( i ) and showed a higher adsorption capacity compared to nZVI and CNTs, with a maximum adsorption capacity of 522.41 mg g −1 . Ag( i ) was adsorbed on nZVI/CNT composites by ion exchange and chelation, where Ag( i ) was reduced to non-toxic Ag due to the redox reaction among pyrrolic-N, nZVI, and Ag( i ). The adsorption process of Ag( i ) on nZVI/CNT composites was dominated by monolayer adsorption. According to our results, nZVI/CNT composites can be used as economical treatment for wastewater containing Ag( i ).

Thermosensitive and magnetic molecularly imprinted polymers for selective recognition and extraction of aristolochic acid I

Huihuang Xiong,Yong Fan,Xuejin Mao,Lan Guo,Aiping Yan,Xian Guo,Yiqun Wan,Hao Wan

Recently, the natural compound of aristolochic acid I (AAI) has attracted wide attentions due to its strong nephrotoxicity and carcinogenicity. However, the extraction of AAI based on conventional molecularly imprinted polymers (MIPs) are tedious with extensive eluent, causing secondary pollution and poor regeneration. Herein, thermosensitive and magnetic MIPs (TMMIPs) were synthesized by a surface imprinting method, which achieved thermosensitive capture/release of AAI, along with rapid magnetic separation, significantly shortening the elution time and reducing organic-solvent consumption. TMMIPs with dual-stimuli responses exhibited superior affinity, selectivity, kinetics, and regeneration ability towards AAI. TMMIPs were applied to analyze AAI in Houttuynia cordata via dispersive solid-phase extraction (d-SPE) coupled with high performance liquid chromatography (HPLC), yielding satisfactory recoveries (79.03–99.67%) and relative standard deviations (≤5.78%). The limit of detection of AAI was as low as 26.67 µg/L. TMMIPs demonstrate great applicability for fast, selective and eco-friendly extraction of AAI in complicated matrices.

Highly efficient and blue-excitable Mn-doped PEA2Pb(Br/I)4 perovskite for solid lighting

Zhenxing Fang,Kuang Wu,Le Wang,Da Xu,Weiwei Wang,Yuhang Lin,Xiaohui Liu,Chun Sun

Mn doped two-dimensional (2D) layered perovskites , are becoming excellent candidates as red phosphor because of their high quantum yields (QY) and enhanced stability. However, these Mn doped 2D layered perovskites can only be excited by the near UV light , which largely limits the development of perovskites. Herein, blue-excitable Mn-doped PEA 2 (Pb/Mn)(Br/I) 4 PQDs are synthesized from anion exchange surface engineering strategy. After anion exchange, orange Mn 2+ emission from exciton-to-Mn energy transfer is acquired under the excitation of 450 nm. The PEA 2 (Pb/Mn)(Br/I) 4 compounds exhibit strong luminescence with photoluminescence (PL) QY of 52%, which is comparable to most of the reported red emitting phosphors. A prototype white light-emitting diode (WLED) with good optical performance was fabricated using commercial (Sr,Ba) 2 SiO 4 :Eu phosphor and as-prepared PEA 2 (Pb/Mn)(Br/I) 4 as green and orange color converters, showing great potential in the applications of solid state lighting.
