Multifunctional MIL-101 nanoparticles with Fenton-like reactions to Co-deliver LL-37 peptide and Vancomycin for targeted NIR imaging and Drug-resistant bacteria treatment

Luogen Lai,Wanqing Zou,Yan Zhang,Yuanbiao Tu,Simeng Li,Tongxuan Xin,Tianyue Zhou,Shan Xu,Pengwu Zheng,Qingshan Pan,Wufu Zhu

Bacterial infections and antibiotic resistance have become a global healthcare crisis. There is an urgent need to develop visualized therapeutic platforms that allow accurate imaging of the bacterial infections, successful eradication of bacteria and real-time monitoring of treatment progress. In this work, we report a novel NIR imaging and synergistic antibacterial nano-system ( [email protected] ) for resistance bacterial infection therapy. The multifunctional nano-system is fabricated by a MIL-101 core, which is covalently connected with Vancomycin and modified with targeted antimicrobial peptides LL-37 on the surface. The ability of monitoring the progress of infection treatment and effective promoting wound healing of bacterial infection are proved by in vivo MRSA -infected mice models. Under the environment of endogenous H 2 O 2 overexpression and slightly acid bacterial infection, the excellent Fenton-like reaction activity of [email protected] could effectively catalyze the decomposition of H 2 O 2 to produce hydroxyl radicals (•OH), which trigger highly efficient chemo-dynamic therapy (CDT) and tissue protection effect. By combining CDT with antibacterial peptides and antibiotics therapy, special targeting and synergistic killing of MRSA are successfully achieved in in vitro and in vivo antibacterial assays with excellent biocompatibility. Due to this, this study proposes a novel, high-efficient, multifunctional imaging and therapy system, which will open a new avenue for the design of synergistic antibacterial and diagnostic platforms in the future.

Multifunctional Surface Enhanced Raman Scattering Substrate Fe 3O4 @AgNPs@MIL-101 for Pretreatment and Rapid Detection of Pesticide Residues on the Surface of Fruit Peels

Mingyu Zhang,Yizhuo Tian,Sijia Liu,Yu Wang,Haiyan Li,Yafei Chen,Qing Gao,Xinli Wang,Mingli Chen

A multifunctional surface-enhanced Raman scattering substrate Fe 3 O 4 @AgNPs@MIL-101 was prepared. Rapid SERS detection of pesticide residues was realized by direct pre-enrichment and separation on the peel surface. MIL-101 has an ortho-octahedral framework and large pore size, which endowed Fe 3 O 4 @AgNPs@MIL-101 with the ability to rapidly adsorb and separate positively charged targets. The introduction of tannic acid realized the in situ growth of AgNPs on the backbone, to modulate the electromagnetic enhancement. Pesticide molecules were adsorbed onto the surface of AgNPs mediated by central S atoms, accompanied by the interaction between pesticide molecules and AgNPs, the corresponding SERS signals of different pesticides were observed. Together with the introduction of magnetic coating Fe 3 O 4 , the molecules were enriched in the hotspot and separated to further enhance the SERS performance. Magnet instead of centrifugation was used to simultaneously perform surface extraction and sample separation for a noninvasive, rapid, immediate, and portable assay. The method was accomplished in measurement of thiram and thiabendazole on apple and tangerine epidermis, and the limits of detection (LODs) were 20 ng/cm 2 and 4 μg/cm 2 , respectively. The recovery was reasonable, and it showed that the procedure is valuable for the rapid and nondestructive surface analysis of residual chemicals.

Optimization and mechanism exploration of NH2-functionalized MIL-101(Fe) for enhanced adsorption of antibiotics in single and binary systems: Experimental analysis and DFT calculations

Si Duan,Yuanhua Lin,Guihong Lan,Yudi Jing,Mohd Talha

Antibiotic pollution and resistance spread pose urgent environmental risks, compounded by coexisting multiple antibiotics. To address this, this study aims to enhance the adsorption of typical antibiotics (tetracycline, TC; ciprofloxacin, CIP) through structural and functional modulation of metal-organic frameworks (MOFs). Focusing on MIL-101(Fe), a series of NH 2 -MIL-101(Fe) materials with tunable amination degrees were prepared via a mixed-ligand strategy, which involves adjusting the molar ratio of terephthalic acid (H 2 BDC) to 2-amino terephthalic acid (NH 2 -H 2 BDC). This strategy breaks through the limitations of traditional single-ligand systems. Results revealed that the NH 2 -MIL-101(Fe)-3 (H 2 BDC: NH 2 -H 2 BDC = 1:3) exhibited remarkable adsorption performance in both single and binary systems. The maximum adsorption capacities for TC and CIP reached 827.75 mg/g and 806.26 mg/g, respectively, representing significant increases of 46.34 % and 52.44 % compared to pristine MIL-101(Fe). Although the adsorption capacities decreased in the competitive binary system, they still achieved 641.17 mg/g and 588.43 mg/g for TC and CIP, respectively, outperforming most reported adsorbents. Additionally, NH 2 -MIL-101(Fe)-3 demonstrated robust tolerance to interference from inorganic ions and natural organic matter, along with excellent cyclic regeneration performance. FTIR, XPS, and density functional theory (DFT) calculations clarified mechanisms: -NH 2 groups enhanced the material's polarity and hydrophilicity, facilitating hydrogen bonding; Fe 3 + -antibiotic chelation dominated, supplemented by π-π stacking and pore-filling effects. This multi-mechanism synergy, quantified by DFT-derived binding energies (coordination > hydrogen bonding > π-π interactions). Overall, this study underscores the potential of mixed-ligand functionalized MOFs in addressing complex pollution systems, providing a theoretical foundation for high-performance adsorbent design and antibiotic pollution control.

Incorporation of hierarchical porous MIL-101@MXene into double-network hydrogel for an enhanced strategy for selective and effective removal of hazardous Hg2+ from water

Jialin Pang,Yao Zhu,Meiqian Li,Jicheng Xu,Ziyu Yuan,Yilmaz Yurekli,Yuting Dai,Tao Zhang,Dongya Yang,Fengxian Qiu

Hg 2+ contamination in aquatic environments represents a significant threat to human health and ecological systems due to its pronounced toxicity and enduring nature. It is imperative to develop a high-performance, recyclable adsorbent for Hg 2+ removal to enable effective treatment of wastewater. Herein, MIL-101@MXene was synthesized through in-situ hydrothermal growth of MIL-101 on MXene substrates, and was subsequently incorporated into a double-network hydrogel matrix comprising polyvinyl alcohol (PVA) and chitosan (CS), resulting in the formation of the MIL-101@MXene/hydrogel adsorbent. A comprehensive characterization of MIL-101@MXene/hydrogel was conducted utilizing a series of analytical techniques, including X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), Brunauer-Emmett-Teller (BET) analysis, and X-ray photoelectron spectroscopy (XPS). The porous hydrogel network of MIL-101@MXene/hydrogel, along with the micro-mesoporous structure of MIL-101@MXene, creates a hierarchical pore architecture. In this configuration, the macropores of the hydrogel facilitate efficient diffusion of Hg 2+ into the material's interior while promoting physical adsorption. Meanwhile, the micro-mesopores not only provide a substantial specific surface area but also reveal numerous active adsorption sites, thereby enhancing the overall adsorption capacity. Batch adsorption experiments demonstrated that MIL-101@MXene/hydrogel exhibited optimal performance for the removal of Hg 2+ at a pH of 5, achieving a maximum adsorption capacity. In addition, MIL-101@MXene/hydrogel demonstrated competitive selectivity and reusability, with only about a 25 % reduction in Hg 2+ adsorption capacity in the presence of various competing ions, and retaining over 55 % of its initial capacity after six successive adsorption-desorption cycles. The adsorption mechanism was further elucidated through FT-IR and XPS analyses conducted both before and after the adsorption process. The results highlighted the coordination of Hg 2+ with unsaturated Fe 3+ sites and carboxyl oxygen atoms, as well as surface complexation between Hg 2+ and the functional groups on MXene. Physisorption also contributed to the overall adsorption process. Additionally, there were interactions characterized by hydrogen bonding and coordination between the hydrogel matrix and Hg 2+ . This work indicates that MIL-101@MXene/hydrogel is an effective adsorbent with practical potential for Hg 2+ -contaminated wastewater remediation.

A ratiometric fluorescence sensor based on HFeO-modified NH2-MIL-101(Fe) and Ru-embedded for supersensitive and visual detection of As(V) in water

Yin Fengqin,He Xudong,Liu Xueting,Guo Dong,Wang Jiahui,Cui Xinxin,Fang Zongping,Ye Tai,Cao Hui,Hao Liling,Xu Fei

An enhanced ratiometric fluorescence sensor Ru@HFeO@NH 2 -MIL-101 (Fe) for the determination of arsenate was constructed by using HFeO-modified NH 2 -MIL-101 (Fe) as the response signal and Ru(bpy) 3 2+ as the reference signal, which can be used for sensitive and visual detection of As(V). Because the HFeO nanoparticles were cleverly in-situ generated within the NH 2 -MIL-101(Fe) framework, the adsorption capacity of As(V) was enhanced 32% and further increased fluorescence ratio. The sensor exhibited a good linear relationship within As(V) concentration range 0.05-1 and 3–30 µM, with a detection limit (LOD) of 0.039 µM. The sensor also exhibited good selectivity and practical application performance, and can be utilized by a smartphone for visual detection of As(V). This provides an effective method for the sensitive detection of As(V) in complex environmental conditions. Graphical

Tuning the primary selective nanochannels of MOF thin-film nanocomposite nanofiltration membranes for efficient removal of hydrophobic endocrine disrupting compounds

Dai Ruobin,Han Hongyi,Zhu Yuting,Wang Xi,Wang Zhiwei

Metal organic framework (MOF) incorporated thin-film nanocomposite (TFN) membranes have the potential to enhance the removal of endocrine disrupting compounds (EDCs). In MOF-TFN membranes, water transport nanochannels include (i) pores of polyamide layer, (ii) pores in MOFs and (iii) channels around MOFs (polyamide-MOF interface). However, information on how to tune the nanochannels to enhance EDCs rejection is scarce, impeding the refinement of TFN membranes toward efficient removal of EDCs. In this study, by changing the polyamide properties, the water transport nanochannels could be confined primarily in pores of MOFs when the polyamide layer became dense. Interestingly, the improved rejection of EDCs was dependent on the water transport channels of the TFN membrane. At low monomer concentration (i.e., loose polyamide structure), the hydrophilic nanochannels of MIL-101(Cr) in the polyamide layer could not dominate the membrane separation performance, and hence the extent of improvement in EDCs rejection was relatively low. In contrast, at high monomer concentration (i.e., dense polyamide structure), the hydrophilic nanochannels of MIL-101(Cr) were responsible for the selective removal of hydrophobic EDCs, demonstrating that the manipulation of water transport nanochannels in the TFN membrane could successfully overcome the permeability and EDCs rejection trade-off. Our results highlight the potential of tuning primary selective nanochannels of MOF-TFN membranes for the efficient removal of EDCs.

Efficient monitoring of trace phthalate esters in environmental waters using multi-walled carbon nanotubes@Fe3O4@MIL-101(Fe) nanocomposites based on magnetic dispersive solid-phase extraction

Guobin Huang,Zhonghua Pan,Qilong Wang,Qingqing Liang,Jiayi Wu,Yanmei Zhou,Tianju Chen,Wangchuan Xiao

Background Phthalate acid esters (PAEs) are endocrine-disrupting contaminants that require reliable trace-level monitoring in environmental waters. Magnetic dispersive solid-phase extraction (MDSPE) offers a green sample-preparation approach, but its performance is often constrained by the limited adsorption capacity, poor structural integrity, limited reusability, and high material cost of existing sorbents. Developing a stable, magnetically responsive, and high-affinity nanocomposite to overcome these limitations remains a critical challenge. Results A novel ternary magnetic MWCNTs@Fe 3 O 4 @MIL-101(Fe) nanocomposite was successfully synthesized via a facile hydrothermal strategy, where multi-walled carbon nanotubes (MWCNTs) serve as a conductive scaffold intertwining Fe 3 O 4 nanoparticles and MIL-101(Fe) crystals. This unique architecture endows the sorbent with high saturation magnetization (58.9 emu/g) for rapid separation, a large specific surface area (1127.28 m 2 /g), and abundant π-electron-rich sites for enhanced adsorption. Under optimized MDSPE conditions coupled with gas chromatography-flame ionization detection (GC-FID), the method achieved wide linear ranges (0.05-2500 μg/L), ultra-low limits of detection (0.005-0.05 μg/L), and satisfactory absolute recoveries (89.5%-110.6%) in real environmental water samples. The sorbent exhibited good reusability (≥90% after five cycles) and excellent precision (RSDs <9.0%). Greenness assessment using the AGREE metric yielded a score of 0.65, reflecting a moderate-to-high level of environmental sustainability. Significance and novelty This work presents a novel “MWCNT-bridged” strategy that successfully integrates magnetic responsiveness, high porosity, and π-π interaction sites into a single, stable sorbent, resolving the common trade-off between functionality and recoverability in magnetic MOF composites. The rationally designed nanocomposite provides a robust, highly sensitive, and environmentally sustainable platform for monitoring trace PAEs in complex aqueous matrices, advancing the development of functional materials for green analytical chemistry.

Enhanced treatment of high-algae water and membrane fouling mitigation by SPDA@MIL-101-modified polyethersulfone ultrafiltration membranes with magnetic field regulation and magnetic flocculant assistance

Meng Yao,Ang Li,Zhilin Ran,Xiaoqing Dong,Ruiyan Xiang,Ting Chen,Gang Liu

Harmful algal blooms threaten drinking water safety. Although ultrafiltration can effectively retain algae cells and algal-derived pollutants, such as extracellular polymeric substances, it still faces challenges such as severe membrane fouling and insufficient contaminant removal. To address these issues, this study developed an integrated “membrane material–flocculant–magnetic regulation” process to enhance algae-laden water treatment and mitigate fouling. Nanoparticles of sulfonated polydopamine (SPDA)-modified Materials of Institute Lavoisier-101 (MIL-101), denoted SPDA@MIL-101, were utilized to functionalize a polyethersulfone (PES) ultrafiltration membrane, enhancing its hydrophilicity and negative surface charge. Meanwhile, a magnetic flocculant (PCNF), composed of polyferric sulfate (PFS), sodium carboxymethyl cellulose (CMC-Na), and iron(II,III) oxide (Fe₃O₄) nanoparticles, was synthesized. Under an external magnetic field, a combined coagulation–ultrafiltration process was employed to actively regulate the cake layer, forming a magnetically responsive cake layer with a graded structure of lower density at the bottom and higher density at the top. The SPDA@MIL-101/PES membrane exhibited significantly improved hydrophilicity, indicated by the reduced contact angle of 54.89°, as well as excellent removal efficiency toward algae-derived fluorescent organic matter. Guided by the magnetic field, the PCNF flocculant formed a loose and porous cake layer, leading to a membrane flux recovery rate of 89% and a reduction in irreversible fouling to 11%. Mechanistic analysis revealed that the magnetic field-mediated dynamic reconstruction of the cake layer optimized the pore network, simultaneously improving contaminant retention and flux recovery performance. This study provides a feasible strategy for the treatment of high-algae water by combining material innovation and process regulation.

Deep Learning Sensing Platform for Simultaneous Detection of Kanamycin and Kanamycin B Based on Dual-Recognition and Catalytic Modulation Mechanism of Aptamer-Functionalized MIL-101 Nanozyme

Yadi Qin,Jie Qian,Jian Sun,Qinqin Li,Jun Yao,Minwei Zhang

Extensive antibiotic use has raised concerns over residues in food, necessitating rapid and accurate detection. This study integrates smartphone-assisted deep learning with aptamer-functionalized MIL-101(Fe) nanozymes to build a dual-recognition biosensor capable of simultaneously identifying and quantifying kanamycin (KAN) and kanamycin B (KMB). MIL-101(Fe) exhibited strong adsorption energies and high Langmuir-fitted capacities (28.39 mg/g for KAN; 31.36 mg/g for KMB), indicating monolayer binding at Fe─OH sites that partially suppressed catalytic activity. Aptamer modification enabled selective molecular recognition and accelerated electron transfer, synergistically enhancing nanozyme performance. Coupled with a ResNet-based multitask model, smartphone imaging achieved accurate classification of single and mixed samples and precise quantification over 5–1000 nM. Tests in honey and beef showed recoveries of 84.71%–111.50%, confirming stability and practicality. Overall, the aptamer-enhanced MIL-101(Fe) nanozyme combined with deep learning provides a sensitive, portable approach for on-site monitoring of antibiotic residues in food.

A dispersive solid-phase extraction method for the determination of Aristolochic acids in Houttuynia cordata based on MIL-101(Fe): An analytes-oriented adsorbent selection design

Rui Cheng,Xuejin Mao,Jiaying Yu,Fan Liu,Lan Guo,Dongmei Luo,Yiqun Wan

In view of the molecular structure of Aristolochic acid I (AA-I) and Aristolochic acid II (AA-II), MIL-101(Fe) was selected as the sorbent to develop a dispersive solid-phase extraction (d-SPE) method for capturing the two analytes from Houttuynia cordata. The interactions between the sorbent and analytes were investigated by FT-IR, XPS and UV–Vis DRS spectra. The optimized method demonstrated good linearity with R 2  > 0.9999. The limit of detections (LODs) were 0.007 mg/L and 0.014 mg/L for AA-I and AA-II, respectively, lower than the limit stipulated by Chinese Pharmacopoeia (0.001 %, w/w). The recoveries for AA-I and AA-II were within the range of 73.3–106.4 %. The precisions of intra-day and inter-day were 0.9–5.8 % and 2.1–5.8 %, respectively. Thus, the established method demonstrated to be efficient and reliable to determine AA-I and AA-II in Houttuynia cordata.

MIL-101(Fe)@TiO2 nanotube composite material is used for the solid phase extraction of non-steroidal anti-inflammatory drugs under the synergy of multiple interactions

Hongmei Liu,Shihao Dang,Mingdeng Li,Baogui Ye

Increasing the adsorption sites and effective interactions between sorbents and the targets can improve the solid-phase extraction (SPE) efficiency. Herein, based on the advantages of MOFs and TiO2 nanotubes (TiO2 NTs), an MIL-101(Fe)@TiO2 NT composite was prepared and applied to extract non-steroidal anti-inflammatory drugs (NSAIDs) from water samples coupled with high performance liquid chromatography (HPLC). Through characterization, it was established that MIL-101(Fe) was effectively composited on the surface and inside the TiO2 nanotubes, increasing effective adsorption sites. The obtained composite material well retains the structure and functional groups of the two original materials, and while retaining the original force with the target, it achieves a synergistic effect and produces more interactions with the target. Therefore, the extraction efficiency was greatly improved. The recovery efficiency reached 97.7–105.1% with an RSD of less than 6.71%, the detection limit was 0.1–0.2 μg L−1, and the linear range was 1–200 μg L−1 with a determination coefficient of 0.9972–0.9994. Owing to the stability of the two original materials, the composite material could be recycled and reused to extract NSAIDs up to 15 times without a loss of the recovery rate. Satisfactory results were obtained when it was used to extract NSAIDs from the Yellow River. These results indicate that the synthesized MIL-101(Fe)@TiO2 NT material is a promising sorbent to extract NSAIDs at trace concentrations with high efficiency and long lifetimes.

A dual-signal self-checking photoelectrochemical immunosensor based on the sole composite of MIL-101(Cr) and CdSe quantum dots for the detection of α-fetoprotein

Xiaolin Zhong,Min Zhang,Lu'an Guo,Yongze Xie,Renfeng Luo,Wenxue Chen,Faliang Cheng,Lishi Wang

Designing a photoelectrochemical (PEC) immunosensor that can produce dual photocurrent signals which can refer to each other is a great importance but a big challenge. In this manuscript, a novel dual photocurrent signals immunosensor was constructed for the detection of α-fetoprotein (AFP). Unlike the usual method of using two composite materials to provide cathode and anode photocurrent respectively, this work applies only one compound of MIL-101 (Cr) and CdSe quantum dots (QDs). Thereinto, we found that the photocurrent polarity of MIL-101(Cr) would switch by adjusting applied voltage . And then CdSe QDs was introduced by simple ultrasound mixing to boost the dual photocurrent signals. Furthermore, in the composite of M&C, the electron transfer path between MIL-101(Cr) and CdSe QDs may switch between “Z-type” and “Ⅱ-type” by adjusting voltage. Benefiting by the dual signals, the proposed sensor can not only perform sensitively quantitative detection of α-fetoprotein (AFP), but also can intuitively estimate the accuracy and reliability of the test result by determining whether the corresponding relationship of “cathode photocurrent-analyte concentration-anode photocurrent” is established. The linear ranges of the sensing electrodes as cathode and anode are the same, both from 0.1 to 300 ng mL −1 . The limit of detection (LOD) is 0.082 ng mL −1 (S/N = 3) when it used as an anode, and the LOD is 0.054 ng mL −1 (S/N = 3) when it served as cathode. Furthermore, this sensor showed acceptable stability, reproducibility, specificity, and feasibility of detecting AFP in human serum, which has broad development prospects in the early clinical diagnosis.
