Molecularly imprinted polymers and PEG double engineered perovskite: an efficient platform for constructing aqueous solution feasible photoelectrochemical sensor

Xiaoyu Yang,Lu Chen,Xiuxiu Xiong,Yun Shu,Dangqin Jin,Yang Zang,Wei Wang,Qin Xu,Xiao-Ya Hu

Herein, a kind of dual polymers, molecularly imprinted polymers (MIPs) and polyethylene glycol (PEG), coated perovskite (MIP-PEG/CH 3 NH 3 PbI 3 ) was synthesized and used to construct a photoelectrochemical (PEC) sensor for salicylic acid (SA) detection. A selective MIP layer for SA recognition was anchored on the PEG modified CH 3 NH 3 PbI 3 through a facile UV-polymerization process. PEG benefits the water-resistant ability of CH 3 NH 3 PbI 3 and MIPs endows the photoactive CH 3 NH 3 PbI 3 good selectivity. Thanks to the protection from PEG and MIPs, the MIP-PEG/CH 3 NH 3 PbI 3 product showed stable photocurrents in aqueous solution. Furthermore, methylammonium iodide (MAI) contained electrolyte was used in which MAI could restore the degraded part of CH 3 NH 3 PbI 3 to improve its stability. SA was detected by monitoring the photocurrent signal changes of the PEC sensor resulting from the reaction between SA and MIPs. The as-fabricated PEC sensor showed good performance in terms of sensitivity and selectivity. The linear range between the photocurrent changes and the concentrations of SA was from 1.0 × 10 -15 to 7.0 × 10 -11 M. This work unveiled the promise of organic-inorganic hybrid lead halide perovskite based structure for PEC sensors and inspired more interests in the development of diverse perovskite-based materials for advanced PEC analysis.

RhB-encapsulating silica nanoparticles modified with PEG impact the vascular endothelial function in endothelial cells and zebrafish model

Shuang Liang,Yueyue Chen,Shiming Zhang,Yuanyuan Cao,Junchao Duan,Yapei Wang,Zhiwei Sun

Silica nanoparticles (SiNPs) have been widely used in human health related products, such as food additives, cosmetics and even drug delivery, gene therapy or bioimaging. Recently, a first-in-human clinical trial based on polyethylene glycol (PEG)-modified SiNPs had been approved by US FDA to trace melanoma. However, as a nano-based drug delivery system, its biocompatibility and vascular toxicity are still largely unknown. Thus, we synthesized the fluorescent SiNPs to explore the biocompatibility and vascular endothelial function, and compare different biological effects caused by PEG-modified and unmodified SiNPs in cells and zebrafish model. The characterizations of SiNPs and PEG-modified SiNPs were analyzed by TEM, SEM, AFM and DLS, which exhibited relatively good stable and dispersive. Compared with SiNPs, PEG-modified SiNPs had markedly reduced the inflammatory response and vascular damage in Tg (fli-1: EGFP) and Tg (mpo: GFP) transgenic zebrafish lines, respectively. Consistent with the in vivo results, the PEG-modified SiNPs had been found to significantly decline the levels of ROS, inflammatory cytokines and mitochondrial-mediated apoptosis in vascular endothelial cells compared to SiNPs, and the ROS scavenger NAC could effectively alleviate the above adverse effects induced by nanoparticles. Our results suggested that the PEG-modified SiNPs could become more safety via increasing the biocompatibility and decreasing cellular toxicities in living organisms.

Fabrication of macroporous protein-containing films through the reverse emulsions approach featuring β-cyclodextrin-conjugated PEG-PLGA copolymers

Wen Ji-jie,Lu Hong-guang,Liu De-e,Gao Hui

A series of β -cyclodextrin-conjugated 4-arm poly(ethylene glycol)-poly(lactide- co -glycolide) (4-arm PEG-PLGA) copolymers were synthesized by a ring-opening polymerization of D,L-lactide and glycolide using 4-arm PEG as initiator, and then conjugated with mono(6-ethylenediamine-6-deoxy)- β -cyclodextrin (CDen) or ethylenediamino-bridged bis- β -CD (BCDen). The chemical structures of copolymers were confirmed by 1 H-NMR and FTIR spectroscopy. The β -CD-conjugated PEG-PLGA formed stable reverse micelles due to the formation of β -CD and bovine serum albumin (BSA) inclusion complexation, which could accommodate BSA in the organic solvent with improved encapsulation efficiency. Moreover, we demonstrated a one-step approach to construct macroporous protein-containing films using these reverse micelles. The films with ordered pore arrays were directly prepared from reverse micelles. Interestingly, the protein was totally located in the whole matrix except for the pores.

Rongbin Huang,Lei Xiang,Ying Liu,Xiangping Deng,Xuan Cao,Lanfang Li,Cuiyun Yu,Yanming Chen,Guotao Tang

ObjectivesTo develop an acid trigger release of antitumour drug delivery carriers, pH-sensitive amphiphilic poly (ethyleneglycol)-imine-benzoic-dipalmitate (PEG-I-dC16) polymers were designed and synthesized and the drug-loaded micelles were evaluated in vitro.MethodsPEG-I-dC16 synthesized by Schiff base synthetic method and characterized by 1H-NMR. To determine the drug-loading capacity, doxorubicin (DOX) was encapsulated in the micelles using membrane dialysis method. Zeta potential, particle size, drug-loading capacity, in vitro drug release in different pH conditions and cytotoxicity evaluation of micelles were carried out comparing with non-acid liable PEG–amide–benzoic–dipalmitate (PEG-A-dC16) polymers micelles. The cellular uptake and intracellular distribution of DOX were detected by flow cytometry and confocal laser scanning microscope.Key findingsDrug-loading capacity and encapsulation efficiency of micelle (PEG molecular weight 2k) were 12.7 ± 1.1% and 49.8 ± 2.2%, respectively. The average particle size was 72.3 ± 2.5 nm. The DOX release rate of PEG-I-dC16 micelles is much higher at pH 6.5 than at pH 7.4. DOX cellular uptake and nuclear accumulation of PEG-I-dC16 micelles were more efficiency than that of PEG-A-dC16 micelles.ConclusionThe pH-sensitive PEG-I-dC16 micelles could be a promising drug delivery system for anticancer drugs.

Efficient nanobiocatalytic systems of nuclease P1 immobilized on PEG-NH2 modified graphene oxide: effects of interface property heterogeneity

Wei Zhuang,Linjiao He,Jiahua Zhu,Jianwei Zheng,Xiaojing Liu,Yihui Dong,Jinglan Wu,Jingwei Zhou,Yong Chen,Hanjie Ying

The use of graphene oxide (GO) nanosheets for functional enzyme support has attracted intensive interest owing to their unique planar structure and intriguing physical and chemical properties. However, the detailed effects of the interface properties of GO and its functionalized derivatives on active biomolecules are not well understood. We immobilize nuclease P 1 , a common industrial nucleic acid production enzyme, on pristine and amino poly(ethylene glycol) (PEG-NH 2 ) modified GO nanosheets with interface property heterogeneity using two approaches, physical adsorption and chemical crosslinking. It is demonstrated that nuclease P 1 could be stable immobilized on the surface of pristine GO by physical adsorption and on the edge of modified GO nanosheets by chemical crosslinking. The resultant loading capacity of nuclease P 1 on pristine GO is as high as 6.45   mg/mg as a consequence of strong electrostatic and hydrophobic interactions between the enzyme and carrier. However, it is determined that the acid resistance, thermal stability, reusability and degradation efficiency of the immobilized enzyme on PEG-NH 2 -modified GO are obviously improved compared to those of the enzyme immobilized on pristine GO. The enhanced catalytic behavior demonstrates that GO and its derivatives have great potential in efficient biocatalytic systems.

Targeted Soft Biodegradable Glycine/PEG/RGD-Modified Poly(methacrylic acid) Nanobubbles as Intelligent Theranostic Vehicles for Drug Delivery

Yongjing Li,Jiaxun Wan,Zihao Zhang,Jia Guo,Changchun Wang

The development of multifunctional ultrasound contrast agents has inspired considerable interest in the application of biomedical imaging and anticancer therapeutics. However, combining multiple components that can preferentially accumulate in tumors in a nanometer scale poses one of the major challenges in targeting drug delivery for theranostic application. Herein, reflux-precipitation polymerization, and N-(3-(dimethylamino)propyl)-N′-ethylcarbodiimide-meditated amidation reaction were introduced to effectively generate a new type of soft glycine/poly(ethylene glycol) (PEG)/RGD-modified poly(methacrylic acid) nanobubbles with a uniform morphology and desired particle size (less than 100 nm). Because of the enhanced biocompatibility resulting from the glycine modification, over 80% of the cells survived, even though the dosage of glycine-modified polymeric nanobubbles was up to 5 mg/mL. By loading doxorubicin as an anticancer drug and perfluorohexane as an ultrasound probe, the resulting glycine/PEG/RGD-modified nanobubbles showed remarkable cancer therapeutic efficacy and a high quality of ultrasonic imaging; thus, the ultrasonic signal exhibited a 1.47-fold enhancement at the tumor site after intravenous injection. By integrating diagnostic and therapeutic functions into a single nanobubble, the new type of theranostic nanobubbles offers a promising strategy to monitor the therapeutic effects, giving important insights into the ultrasound-traced and enhanced targeting drug delivery in biomedical applications.

Poly(vinyl methyl ether/maleic anhydride)-Doped PEG–PLA Nanoparticles for Oral Paclitaxel Delivery To Improve Bioadhesive Efficiency

Qian Wang,Chan Li,Tianyang Ren,Shizhu Chen,Xiaoxia Ye,Hongbo Guo,Haibing He,Yu Zhang,Tian Yin,Xing-Jie Liang,Xing Tang

Bioadhesive nanoparticles based on poly(vinyl methyl ether/maleic anhydride) (PVMMA) and poly(ethylene glycol) methyl ether-b-poly(d,l-lactic acid) (mPEG-b-PLA) were produced by the emulsification solvent evaporation method. Paclitaxel was utilized as the model drug, with an encapsulation efficiency of up to 90.2 ± 4.0%. The nanoparticles were uniform and spherical in shape and exhibited a sustained drug release compared with Taxol. m-NPs also exhibited favorable bioadhesive efficiency at the same time. Coumarin 6 or DiR-loaded nanoparticles with/without PVMMA (C6-m-NPs/DiR-m-NPs or C6-p-NPs/DiR-p-NPs) were used for cellular uptake and intestinal adhesion experiments, respectively. C6-m-NPs were shown to enhance cellular uptake, and caveolae/lipid raft mediated endocytosis was the primary route for the uptake of the nanoparticles. Favorable bioadhesive efficiency led to prolonged retention in the intestine reflected by the fluorescence in isolated intestines ex vivo. In a ligated intestinal loops model, C6-m-NPs showed a clear advantage for transporting NPs across the mucus layer over C6-p-NPs and free C6. The apparent permeability coefficient (Papp) of PTX-m-NPs through Caco-2/HT29 monolayers was 1.3- and 1.6-fold higher than PTX-p-NPs and Taxol, respectively, which was consistent with the AUC0–t of different PTX formulations after oral administration in rats. PTX-m-NPs also exhibited a more effective anticancer efficacy, with an IC50 of 0.2 ± 1.4 μg/mL for A549 cell lines, further demonstrating the advantage of bioadhesive nanoparticles. The bioadhesive nanoparticles m-NPs demonstrated both mucus permeation and epithelial absorption, and thus, this bioadhesive drug delivery system has the potential to improve the bioavailability of drugs that are insoluble in the gastrointestinal environment.

Targeted delivery of ginsenoside compound K using TPGS/PEG-PCL mixed micelles for effective treatment of lung cancer

Lei Yang,Zhenghai Zhang,Jian Hou,Xin Jin,Zhongcheng Ke,Dan Liu,Mei Du,Xiaobing Jia,Huixia Lv

Ginsenoside compound K (CK) is one of the effective ingredients in antitumor composition of ginsenoside. However, the poor water solubility and significant efflux have limited the widespread clinical use of CK. In this study, preparation of novel CK-loaded d -alpha-tocopheryl polyethylene glycol 1,000 succinate/poly(ethylene glycol)-poly(ε-caprolactone) mixed micelles (CK-M) is discussed to solve the above problems. Particle size, zeta potential, and morphology were characterized using dynamic light scattering and transmission electron microscopy. CK-M are spherical shaped with an average particle size of 53.07±1.31 nm with high drug loading of 11.19%±0.87% and entrapment efficiency of 94.60%±1.45%. Water solubility of CK was improved to 3.78±0.09 mg/mL, which was ~107.35 times higher than free CK. A549 and PC-9 cells were used to evaluate in vitro cytotoxicity and cellular uptake. IC 50 values of CK-M in A549 and PC-9 cells (24 h) were 25.43±2.18 and 18.35±1.90 μg/mL, respectively. Enhanced cellular uptake of CK-M was observed in both cells. Moreover, CK-M promoted tumor cell apoptosis, inhibited tumor cell invasion, metastasis, and efflux through regulation of Bax, Bcl-2, matrix metalloproteinase-2, Caspase-3, and P-glycoprotein. In vivo imaging indicated that CK-M has excellent tumor targeting effect within 24 h, and the relative tumor inhibition rate of CK-M was 52.04%±4.62% compared with control group ( P <0.01). Thus, CK-M could be an appropriate delivery agent for enhanced solubility and antitumor effect of CK.

Redox-sensitive mesoporous silica nanoparticles functionalized with PEG through a disulfide bond linker for potential anticancer drug delivery

Huameng Gong,Zhifei Xie,Mingxing Liu,Hongda Zhu,Honghao Sun

In this paper, redox-sensitive mesoporous silica nanoparticles functionalized with polyethylene glycol (PEG) through a disulfide bond linker (MSNs–SS–PEG) were successfully synthesized using silica nanoparticles modified with a thiol group (MSNs–SH) and thiol-functionalized methoxy polyethylene glycol (MeOPEG–SH). Meanwhile, the particle size, pore size and structural properties of these materials were characterized by transmission electron microscopy (TEM), field emission scanning electron microscopy (FE-SEM), dynamic light scattering (DLS), Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD) and nitrogen adsorption–desorption measurements. Furthermore, the in vitro drug release behaviour of DOX-loaded MSNs–SS–PEG (DOX@MSNs–SS–PEG) was investigated. It was shown that DOX release was markedly accelerated with the increasing concentration of glutathione (GSH), while DOX was not released from the carrier materials in the absence of GSH. Cytotoxicity evaluation revealed the good biocompatibility of the blank nanoparticles and the DOX@MSNs–SS–PEG exhibited comparative anticancer activity to free DOX towards BEL-7402 cells. Therefore, the MSNs–SS–PEG might be a great potential carrier for anticancer drug delivery.

Preparation and Performance of Amphiphilic Polyurethane Copolymers with Capsaicin-Mimic and PEG Moieties for Protein Resistance and Antibacteria

Xi Chen,Guangfa Zhang,Qinghua Zhang,Xiaoli Zhan,Fengqiu Chen

Antifouling (AF) coatings have been a research focus on effectively resisting biocontamination of equipment applied in the aquatic environments. The amphiphilic ternary copolymers P(H–P–A) composed of N-(4-hydroxy-3-methoxybenzyl)acrylamide (HMBA), poly(ethylene glycol) methyl ether methacrylate, and 2-hydroxyethyl acrylate were synthesized by free-radical polymerization. Then the polyurethane (PU) copolymer coatings with capsaicin and poly(ethylene glycol) (PEG) moieties were prepared by cross-linking with polymeric phenyl methanediisocyanate. Fourier transform infrared and 1H NMR were employed for chemical structural analysis of the products. The surface composition and concentration of nitrogen atoms on the copolymer films were quantitatively analyzed by X-ray photoelectron spectroscopy. The AF properties of the copolymers were assessed by static protein adsorption and antibacterial tests. The targeted copolymer film samples can achieve an average antibacterial efficiency of 93.3% and a protein-resistant rate of 97.3%, which demonstrated that P(H–P–A) is a potential environmentally friendly material for AF application.

Salt and pH sensitive semi-interpenetrating polyelectrolyte hydrogels poly(HEMA-co-METAC)/PEG and its BSA adsorption behavior

Yan-Yan Hu,Jing Zhang,Qi-Chen Fang,Dong-Mei Jiang,Chu-Cheng Lin,Yi Zeng,Ji-Sen Jiang

A novel semi-interpenetrating poly(2-hydroxyethyl methacrylate) (pHEMA) based polyelectrolyte hydrogel [p(HEMA- co -METAC)/PEG] was prepared by copolymerizing HEMA with the cationic monomer 2-methacryloyloxyethyltrimethyl ammonium chloride (METAC) in the presence of polyethylene glycol (PEG) with different content and molecular weight (MW 4000 and 400). The chemical structure of the gels was confirmed by FT-IR spectroscopy, morphology study was performed by scanning electron microscope (SEM), thermal stability was revealed by thermogravimetric analysis (TGA), and the mechanical properties were determined by electronic universal testing machine. Swelling studies showed introduction of cationic monomer METAC led to high water content, and the obvious salt and pH sensitive properties were observed which proved the smart behavior of the semi-interpenetrating polymer networks (IPNs) gels. In addition, the effect of temperature and some important biological solution on swelling behavior were reported. Cytotoxicity test demonstrated that synthesized gels owned satisfactory cytocompatibility and were convenient for the application as biomaterials. Finally, the weak bovine serum albumin (BSA) adsorption on semi-IPNs by introducing METAC and controlling the content of PEG in gels demonstrated that they were of good protein resistance effect in biomedical applications. © 2014 Wiley Periodicals, Inc. Fluorescent probe based on GO/g-C3N4-PEG@Cu NPs/MIP for the detection of dopamine in banana

Yaru Wang,Chaohai Pang,Xionghui Ma,Mingyue Wang,Xiaopeng Wu,Sixin Liu,Shuhuai Li

Graphene oxide (GO) and copper nanoparticles (Cu NPs) were incorporated to modulate and enhance the fluorescence properties of pegylated graphite phase carbon nitride (g-C 3 N 4 -PEG). Combined with the specific recognition capability of a molecular imprinted polymer (MIP), a highly sensitive and selective fluorescent molecular imprinted probe for dopamine detection was developed. The fluorescent g-C 3 N 4 -PEG was synthesized from melamine and modified with GO and Cu NPs to obtain GO/g-C 3 N 4 -PEG@Cu NPs. Subsequently, MIP was prepared on the surface of GO/g-C 3 N 4 -PEG@Cu NPs using dopamine as the template molecule. Upon elution of the template molecule, a dopamine-specific GO/g-C 3 N 4 -PEG@Cu NPs/MIP fluorescence probe was obtained. The fluorescence intensity of the probe was quenched through the adsorption of different concentrations of dopamine by the MIP, thus establishing a novel method for the detection of dopamine. The linear range of dopamine detection was from 5 × 10 −11 to 6 × 10 −8 mol L −1 , with a detection limit of 2.32 × 10 −11 mol L −1 . The sensor was utilised for the detection of dopamine in bananas, achieving a spiked recovery rate between 90.3% and 101.3%. These results demonstrate that the fluorescence molecular imprinted sensor developed in this study offers a highly sensitive approach for dopamine detection in bananas.
