A smart Zr/Co porphyrin MOFs-based nanotheranostics: Triggering ROS storm for potentiated tumor therapy

Lu Zhao,Aiai Wang,Yaru Li,Yunfeng Bai,Feng Feng

Reactive oxygen species (ROS)-mediated therapy utilizes the slightly acidic environment of tumor microenvironment and the high amount of H 2 O 2 to disrupt the redox balance of tumor cell while minimizing damage to normal cell. However, this strategy was greatly compromised by the powerful antioxidant capacity of tumors and the limited ROS generation rate of bimetallic metal-organic frameworks (BMOFs). In pursuit of innovative strategies for precise diagnosis and effective treatment, a smart nanotheranostics Zr-TCPP(Co)/ICG/PEG/Apt-M (ZTCIPA) based on Zr/Co porphyrin MOFs was designed. Under 808 nm laser irradiation, ZTCIPA demonstrated exceptional photothermal conversion efficiency alongside robust singlet oxygen ( 1 O 2 ) generation, ensuring potent tumor ablation. Meanwhile, the temperature elevation significantly accelerated the Co 2+ mediated catalytic decomposition of H 2 O 2 to generate •OH. Large amounts of •OH and 1 O 2 formed ROS storm effectively damaged mitochondria and further resulted in tumor cell death. Furthermore, ZTCIPA achieved tumor targeted specificity while enabling real-time fluorescence imaging (FLI)/photothermal imaging (PTI) for accurate tracking. The results in vitro and in vivo demonstrated that ZTCIPA could effectively inhibit tumor growth by photothermal amplified ROS storm under 808 nm laser irradiation. Thus, this novel strategy has enormous potential by solving the bottleneck of ROS-mediated therapies.

Electrochemical Biosensor Based on Ultrathin 2D CN-rGO@AgNPs for ROS Detection in Leukemia Cells

Mingxuan Huang,Huafeng Wang,Shicong Gui,Chujia Xu,Yi Chen,Shaohua Hu,Yubo Li

Accurate monitoring of ROS levels is essential for oxidative stress (OS) management, but their low concentration and instability pose significant detection challenges. Here, we developed a highly sensitive and selective electrochemical biosensor for real-time, micro-volume ROS detection in leukemia cells. The sensor was fabricated using ultrathin g-C₃N₄-rGO (CN-rGO) nanosheet synthesized through a calcination and hydrothermal approach. These nanosheets were deposited onto a screen-printed carbon electrode (SPE) and further modified with silver nanoparticles (AgNPs) to construct a CN-rGO@AgNPs/SPE biosensor. The porous nanostructure significantly enhanced the catalytic surface area and electron transfer efficiency, achieving high sensitivities towards H₂O₂ and O₂·⁻ detection. Using a window titration platform, the sensor detected ROS in 0.1 mL lysates of leukemia cell lines (U-937, THP-1, and MOLM-13) with concentrations of 7.38 ± 4.81 μM, 11.74 ± 0.985 μM, 19.84 ± 3.73 μM, respectively. Furthermore, the platform allowed precise monitoring of doxorubicin (DOX)-induced OS, providing valuable insights for guiding leukemia chemotherapy. Additionally, Lipopolysaccharide (LPS) stimulation experiments demonstrated the sensor's exceptional real-time performance, enabling dynamic detection of H₂O₂ release kinetics and ROS generation rates at the cellular level. Combining high sensitivity, low detection limits, and micro-volume analysis capabilities, this sensor holds great potential for clinical auxiliary diagnosis and drug efficacy assessment in leukemia research.

Platinum Nanozyme-Loaded Dissolving Microneedles Scavenge ROS and Promote Lineage Progression for Androgenetic Alopecia Treatment

Weitong Hu,Shuhan Shi,Yihua Xu,Yunting Zhang,Jingyi Hu,Qiong Bian,Xiaolu Ma,Yuxian Ye,Shengfei Yang,Xiaoxia Sheng,Guang Liang,Tianyuan Zhang,Haibin Wu,Jianqing Gao

Androgenetic alopecia (AGA) is a prevalent issue affecting the physical and mental health of individuals but with fewer current treatments. Platinum nanozymes (PtNZs) are known for their excellent ability to reduce and modulate the high oxidative stress environment in AGA pathology. And microneedles are used to overcome the skin barrier due to the poor permeability of PtNZs. Herein, dissolving microneedles loaded with PtNZs (Pt-MNs) are designed and successfully induced hair regeneration in the AGA model. Pt-MNs possessed adequate mechanical strength to breach the skin barrier for effective PtNZs delivery. In vivo, PtNZs first reduced reactive oxygen species (ROS) to oxygen, which recovered the AGA pathological environment. And the oxygen then increased oxidative phosphorylation, promoting the differentiation of hair follicle stem cells to achieve hair regeneration. The group treated with Pt-MNs with a dosing frequency of once every three days achieved faster hair growth than the daily application of the positive drug minoxidil. Further safety experiments showed that the application of Pt-MNs locally opened temporary and recoverable skin channels, with no retention of Pt in major organs, indicating high safety. In conclusion, this study indicated the potential of Pt-MNs as an effective method for treating AGA.

Novel drug-free cascaded nanoparticles induce tumor-specific ROS storms via multimodal synergistic anticancer therapy

Wen Mingsen,Chen Hongwei,Xu Song,Yang Shanyi,Guan Xuan,Wang Xuancheng,She Zhiyong,Wei Zhijuan,Tong Ying,Luo Jichu,Qin Qixuan,Lin Xueting,Tan Yuru,Nong Yanying,Zhang Qisong

Reactive oxygen species (ROS), generated by sonosensitizers, play a pivotal role in tumor cell apoptosis during sonodynamic therapy (SDT), particularly for tumors located deep within tissues. Nevertheless, conventional sonosensitizers present limitations including inadequate ROS generation, insufficient tumor-specific accumulation, and associated adverse effects, significantly restricting their clinical applicability. To address these limitations, novel drug-free multifunctional nanoparticles (designated HGMP NPs) were synthesized. These NPs consist of mesoporous polydopamine (MPDA)-loaded protoporphyrin IX (PpIX), further surface-modified with glucose oxidase (GOx) and hyaluronic acid (HA), to achieve integrated photothermal, sonodynamic, and starvation-based tumor therapy. Upon exposure to near-infrared (NIR) irradiation (808 nm) combined with ultrasound (US), HGMP NPs exhibited pronounced synergistic anticancer effects. Specifically, the photothermal effect triggered by NIR irradiation effectively enhanced local oxygen supply within tumor sites, thus significantly augmenting ROS production and improving the therapeutic outcomes of SDT. Concurrently, GOx-mediated glucose depletion induced tumor starvation and produced hydrogen peroxide (H 2 O 2 ), further exacerbating oxidative stress within the tumor microenvironment. Transcriptomic analysis revealed that ROS and TNF signaling pathways represented key mechanisms underlying tumor elimination by this multimodal synergistic strategy. Real-time PCR analysis and ELISA assays further validated activation of the TNF signaling pathway. Importantly, this study first confirmed the high biocompatibility and biosafety of HGMP NPs via serum metabolomics, demonstrating no detectable systemic metabolic perturbations. Collectively, the prepared HGMP NPs provide a rational paradigm for synergistic anticancer therapy. These findings highlight the potential of HGMP NPs as an exceptionally safe and effective nanoplatform for cancer treatment, offering valuable insights into future developments in cancer nanomedicine. Ultrasound Activated Copper-Based Sonosensizer for Dual-Amplified ROS-Mediated Tumor Therapy and Cuproptosis Induction

Penghui Wang,Runming Zhong,Yingzhe Wu,Lingli Jin,Yanan Zhang,Kailong Zhang,Fan Zhao

Sonodynamic therapy (SDT) is a promising noninvasive cancer treatment with deep tissue penetration, but its therapeutic efficacy is limited by insufficient reactive oxygen species (ROS) production. Herein, we developed copper-based sonosensitizer (CCG MPNs) by coordinating copper ions (Cu2+) with chlorin e6 (Ce6) and gallic acid (GA) for dual-amplified ROS-mediated tumor therapy and cuproptosis induction. CCG MPNs dissociate in the high-glutathione (GSH) tumor microenvironment, releasing Cu ions, Ce6, and GA. GA rapidly depletes GSH, enhancing tumor cell sensitivity to ROS and cuproptosis. Additionally, the released Cu+ ions generate cytotoxic hydroxyl radicals via a highly efficient Fenton reaction and ultrasound (US) activation of Ce6 produces singlet oxygen, amplifying ROS-mediated apoptosis by evoke chemodynamic therapy (CDT) and SDT. Moreover, the yielded ROS would cut off the ATP supply, resulting in a decrease in the expression of the copper transporter protein ATP7A. Large accumulations of intracellular Cu+ ions can be more readily induce cuproptosis by promoting lipoylated protein aggregation. In vitro and in vivo anti-tumor studies showed that CCG MPNs group under US treatment exhibited better anti-tumor performance. This multimodal approach overcomes the limitations of the tumor microenvironment, offering a synergistic strategy to enhance tumor therapy efficiency through combined ROS generation and cuproptosis activation.

Sonotheranostic nanosideromycin eradicates bacterial biofilm infections via ultrasound-detonated ROS generation and ferroptosis-like death

Xin Pang,Chang Zhang,Qicai Xiao,Yi Cheng,Qixuan Dai,Hu Chen,Sijia Tan,Gang Liu,Yun Zeng

Biofilm formation poses a severe challenge to antibacterial stewardship. While siderophore-antibiotic conjugates (termed as sideromycins) offer a promising solution, their efficacy is inherently limited by antibiotic resistance. To transcend this barrier, we pioneer a transformative siderophore-sonosensitizer conjugate through covalent linkage of a catechol siderophore to purpurin 18 (a sonosensitizer). This novel conjugate further self-assembles with iron(III) ions, forming the first-reported carrier-free nanosideromycin—an all-in-on iron-siderophore-sonosensitizer nanoplatform. This design enables ultrasound-denotated reactive oxygen species (ROS) generation and ferroptosis-like amplication. Capitalizing on bacteria-specific siderophore uptake and pH-responsive assembly/disassembly, the nanosideromycin enables precision delivery and active internalization of sonosensitizers into bacteria. This strategy permits real-time localization of infections via concurrent fluorescence/photoacoustic and magnetic resonance imaging. Upon ultrasound irradiation, dual antimicrobial mechanisms of sonosensitizer-mediated sonodynamic therapy and siderophore/iron-augmented sono-Fenton catalysis are stimuonously unleashed, synergistically tirggering an explosive ROS burst and potent ferroptosis-like bacterial death. As a result, mice with multidrug-resistant biofilm-induced pyomyositis were completely cured. Collectively, this first-in-class theranostic nanosideromycin integrates highly-targeted imaging diagnostics, cost-effective yet ultra-efficient ROS generation, and ferroptosis-like bacterial killing, establishing a paradigm-shifting strategy for biofilm therapy with spatiotemporal controllability.

Fe-modified biochar-driven ROS generation in the rhizosphere and their role in microplastic transformation

Zulqarnain Haider Khan,Han Yu,Hui Li,Syed Shabi Ul Hassan Kazmi,Ruixia Han,Muhammad Azeem,Xin Sun,Zia Ur Rehman,Gang Li

Reactive oxygen species (ROS) are critical mediators of soil biogeochemical processes. While the production of ROS with biochar (BC) in the rhizosphere has not been explored. We demonstrate that BC and Fe-modified biochar (FeBC), prepared at 400°C and 600°C, influence ROS generation in paddy soil containing biodegradable (polybutylene succinate: PBS) and conventional (polystyrene) microplastics (MPs). FeBC, particularly FeBC prepared at 600°C, enhance ROS (O 2 •− , H 2 O 2 , • OH) production in paddy soil. The results of rhizosphere-focused pot experiment simulating field-like flooded conditions highlight that FeBC significantly increased O 2 •− , H 2 O 2 , • OH concentrations, especially with PBS (25.63, 27.69, 35.82 µM/kg, respectively) surpassing control by 3.1–3.6-folds. Iron plaque formation, water-soluble phenols, and microbial activity further influenced ROS dynamics and accelerated MP degradation via oxidative fragmentation (SEM surface fissures), dissolved organic carbon release (484 mg/kg), and SUVA 254 decline (4.6 folds). Microbial shifts toward Actinobacteria (37–43 %) and Firmicutes (25–28 %) correlated with ROS dynamics, linking biotic-abiotic degradation pathways. This work reveals the novel role of FeBC in rhizosphere ROS generation and the environmental behavior of MPs in BC-treated agricultural systems.

Light-switchable lignin-based nanospheres for photodynamic eradication of drug-resistant pathogens and ROS scavenging

Qian Wang,Mingjie Chen,Junwei Yao,Gang Zhou,Dandan Zhang,Qingshan Shi,Qianwen Mai,Xiaobao Xie

The increasing prevalence of drug-resistant bacteria and the ecological burden of synthetic antimicrobials demand sustainable biomaterials with superior antibacterial properties. Natural lignin, an amorphous polymer, presents considerable potential as a structural building block for sustainable biomaterial. Herein, inspired by antimicrobial photodynamic inactivation, kraft lignin was first reacted with formaldehyde and ethylenediamine to synthesize aminated lignin, the aminated lignin was further reacted with acetic anhydride, resulting in modified lignin. Thereafter, the modified lignin functions as a carrier, encapsulating the hydrophobic photosensitizer chlorin e6 to improve its dispersibility and aqueous solubility. Chitosan was further utilized to coat the nanoparticles, improving bacterial uptake, ultimately obtaining the desired final nanospheres (CS-ML-Ce6). Upon laser irradiation, CS-ML-Ce6 achieved >99.99 % bactericidal efficiency against methicillin-resistant Staphylococcus aureus . Additionally, it demonstrated a minimum inhibitory concentration of modified lignin of 67.71 μg/mL when exposed to light. It could result in membrane disruption and intracellular component leaking, while bacterial adsorption increased to 47.36 %. The dead/live rate of CS-ML-Ce6 against drug-resistant pathogens was almost 8-fold to modified lignin control. Concurrently, hemolysis assays with rabbit red blood cells confirmed exceptional biocompatibility, with hemolysis rates maintained below 5 % even at 6 × MIC concentrations. Considerable reactive oxygen species scavenging effectiveness at both the solution and cellular levels was also observed of CS-ML- Ce6, this dual-function approach uniquely combines light-activated bactericidal properties and dark-phase antioxidant repair. Utilizing lignin, an industrial byproduct, provides an efficient and promising antimicrobial alternative to prevent overuse of antibiotics and has significant potential for human healthcare.

Targeting the ROS-ferroptosis-inflammation cycle with a nanozyme-functionalized hydrogel for intervertebral disc repair

Wang Yanqiu,Tan Lu,Yang Yi,Duan Hongli,Liu Chao,Zhao Jing,Zhou Yue,Li Changqing,Liu Minghan

Intervertebral disc degeneration is a major cause of low back pain and is driven by a vicious cycle of reactive oxygen species (ROS), ferroptosis, and inflammation, afflicting millions of people worldwide. Breaking this cycle represents a significant therapeutic challenge. Here, we develop a ROS-responsive hydrogel loading a resident nanozyme system composed of nucleus pulposus cell membrane-coated black phosphorus@cerium oxide to disrupt this degenerative cascade. Specifically, the nanozyme exhibits a self-sustaining cerium redox cycle due to the incorporation of black phosphorus nanosheets into cerium oxide, conferring durable antioxidant capacity for scavenging ROS. Furthermore, it suppresses the inflammatory cytokine IL6 by inhibiting HuR-mediated mRNA stabilization, thereby blocking the pro-inflammatory and pro-ferroptotic IL6/STAT3 axis. Together, this dual mechanism enables our nanozyme-functionalized hydrogel to break the ROS-ferroptosis-inflammation feedback loop, thus effectively promoting structural and functional disc repair.

Biofilm-disrupting heterojunction microneedles: dual ROS amplification and glucose deprivation for accelerated diabetic wound healing

Wenjie You,Feng Xiao,Zichao Cai,Jiaxin Zhao,Zhengyao Zhang,Weikang Hu,Yun Chen,Kwang Leong Choy,Zijian Wang

Rationale: Diabetic wound healing process is critically hindered by bacterial infection, bacterial biofilm formation, and persistent hyperglycemia. Biomolecular microneedles represent a promising alternative to conventional therapies such as antibiotics and antibiotic-loaded wound dressings, owing to the advantages like reduced risk of drug resistance and enhanced long-term efficacy. However, the microneedles that fulfill the clinical needs of diabetic wounds have rarely been reported. Methods: A glucose oxidase (GOx)-laden Ti 3 C 2 /In 2 O 3 (INTG) heterojunction was engineered as a nano-micro platform for reactive oxygen species (ROS) amplification and glucose deprivation, and subsequently immobilized onto the gelatin methacryloyl (GelMA) microneedle tips to obtain double-layer microneedles (GITG microneedles). Their physiochemical properties and biomedical applications were comprehensively investigated. Results: For INTG heterojunction, the formation of Schottky structure significantly improved the oxygen absorption capacity, facilitated the generation and migration of photogenerated electron-hole pairs, thereby promoting the ROS generation. Besides, under near-infrared (NIR) irradiation, GITG microneedles effectively inhibited bacterial proliferation and survival by generating ROS, thereby preventing the formation of bacterial biofilm. Additionally, GITG microneedles accelerated wound closure and facilitated skin tissue regeneration in a rat model through multiple mechanisms. Conclusion: This study developed an advanced microneedle platform enabling on-demand multimodal treatment, demonstrating significant potential for clinical diabetic wound management.

ROS-responsive nanogels for brain targeted delivery of icariin in the treatment of Parkinson’s disease

Xue Li,Jian Huang,Yan-Ming Zhou,Xue Chen,Qin-Wei Dong,Yuan-Lu Cui,Yi-Bing Chen

Excessive reactive oxygen species (ROS)-induced nigrostriatal dopaminergic neuron degeneration is a cardinal pathological feature of Parkinson’s disease (PD). Although icariin, a natural antioxidant capable of scavenging ROS, shows therapeutic potential, it remains underutilized in clinical settings. This translational gap primarily stems from two pharmacological limitations: (1) inadequate blood–brain barrier (BBB) penetration that prevents effective delivery of icariin to the brain, and (2) the lack of targeted drug release at pathological sites, thereby diminishing its local neuroprotective efficacy against ROS-mediated neurodegeneration. To overcome these challenges, we developed a ROS-responsive selenocysteamine-alginate nanogel (ASeNG-ICA) that bypasses the BBB via nose-to-brain delivery and enables pathology-triggered drug release through diselenide bond cleavage in the high-ROS microenvironments characteristic of PD. In vitro studies demonstrated that the nanogels undergo ROS-responsive disintegration, resulting in sustained icariin release under oxidative conditions. Following intranasal administration in mice, ASeNG-ICA achieved rapid brain biodistribution. In a PD mouse model, this delivery system significantly reduced striatal malondialdehyde (MDA) levels, regulated antioxidant enzymes (HO-1, SOD) expression, alleviated oxidative stress and improved behavioral disorders, surpassing conventional free icariin therapy. Overall, ASeNG-ICA resolves critical delivery barriers in PD therapy, offering a new paradigm for neurodegenerative disease treatment.

A cyclodextrin/ROS-responsive Pydiflumetofen nanoparticle for enhanced fungicidal activity against Sclerotinia sclerotiorum in soybean

Qian Zhao,Liyan Zhang,Ziyu Wang,Weibo Han,Jianzhong Wu

Sclerotinia stem rot (SSR), caused by Sclerotinia sclerotiorum , significantly reduces soybean yields. Conventional SSR-based pesticide application presents environmental and health hazards, whereas ROS-responsive nanocarrier systems demonstrate superior efficacy as a sustainable alternative. In this study, PYD@OxbCD NPs, encapsulating Pydiflumetofen (PYD), were synthesized and showed ROS-triggered release and strong in vitro activity against SSR. This targeted release mechanism reduces ecological risk by minimizing pesticide exposure to non-target organisms. PYD@OxbCD delivers PYD more precisely to infected soybean tissues, outperforming traditional Pydiflumetofen suspension concentrate (SC) in antifungal efficacy. It also upregulates defense-related genes and reduces disease incidence and severity in soybean plants. PYD@OxbCD also mitigates the phytotoxic effects of PYD SC on seedling growth. Overall, PYD@OxbCD shows strong potential as an effective nanoplatform for managing SSR in soybeans, enhancing disease control and pesticide utilization for sustainable agriculture.
