Ofloxacin is one kind of quinolone antibiotic drugs, the abuse of ofloxacin in livestock and aquaculture may bring bacterial resistance and healthy problem of people. The illegally feeding cattle with ofloxacin will help it keep health, but the sedimentation of ofloxacin could bring problem in food safety. The accurate, simple and instant monitoring ofloxacin from beef by portable sensor was of vital issue in food quality. A simple and reliable method was proposed for instant and quantitative detecting ofloxacin in beef, in which the thin-layer chromatography (TLC) -surface-enhanced Raman scattering (SERS) spectroscopy was in tandem with machine learning analysis base one principal component analysis-back propagation neural network (PCA-BPNN). The TLC plate was composed with diatomite, that was function as the stationary phase to separate ofloxacin from beef. The real beef juice was directly casted onto the diatomite plate for separating and detecting. The directly monitor ofloxacin from beef was achieved and the sensitivity down to 0.01 ppm. The PCA-BPNN was used as reliable model for quantitative predict the concentration of ofloxacin, that shown superior accuracy compared with the traditional model. The results verify that the diatomite plate TLC-SERS combined with machine-learning analysis is an effective, simple and accurate technique for detecting and quantifying antibiotic drug in meat stuff to improve the food safety.

In this paper, we demonstrate a surface-enhanced Raman spectroscopy (SERS) biosensor based on the self-assembly of gold nanorods (AuNRs) for the specific detection of airway inflammatory factors in diluted sputum. The AuNR surface was modified with an antibody that was able to specifically recognize an airway inflammatory factor, interleukin-5 (IL-5), so that a end-to-end self-assembly system could be obtained, resulting in an order of magnitude amplification of the Raman signal and greatly improved sensitivity. Meanwhile, the outer layer of the biosensor was coated with silicon dioxide, which improved the stability of the system and facilitated its future applications. When the detected concentration was in the range of 0.1–50 pg/mL, the SERS signal generated by the sensor showed a good linear relationship with the IL-5 concentration. Moreover, it had satisfactory performance in diluted sputum and clinical subjects with asthma, which could achieve sensitive detection of the airway inflammatory factor IL-5. Overall, the developed biosensor based on the SERS effect exhibited the advantages of rapid and sensitive detecting performance, which is suitable for monitoring airway inflammatory factors in sputum.

Colorectal cancer (CRC) remains a significant contributor to the global mortality rate, and a single biomarker cannot meet the specificity required for CRC screening. To this end, we developed a multiplexed, pump-free surface-enhanced Raman scattering (SERS) microfluidic chip (LoC-SERS) using a one-step recognition release mechanism; the aptamer-functionalized novel Au nanocrown array (AuNCA) was used as the detection element embedded in the detection zone of the platform for rapid and specific detection of protein markers in multiple samples simultaneously. Here, the corresponding aptamer specifically captured the protein marker, causing the complementary strand of the aptamer carrying the Raman signal molecule to be shed, reducing the SERS signal. Based on this platform, sensitive and specific detection of the target can be accomplished within 15 min with detection limits of 0.031 pg/mL (hnRNP A1) and 0.057 pg/mL (S100P). Meanwhile, the platform was consistent with ELISA results when used to test clinical. By substituting different aptamers, this platform can provide a new solution for the rapid and sensitive detection of protein markers, which has promising applications in future disease detection.

Ferroptosis is a non-apoptotic cell death regulated by iron-dependent lipid peroxidation. Glutathione (GSH), a key antioxidant against oxidative damage, is involved in one of the most important metabolic pathways of ferroptosis. Herein, an excellent plasmonic nanoprobe was developed for highly sensitive, in situ, dynamic real-time monitoring of intracellular GSH levels during ferroptosis. A nanoprobe was prepared by functionalizing gold nanoparticles (AuNPs) with the probe molecule crystal violet (CV). The fluctuation in the SERS signal intensity of CV via the competitive displacement reaction can be used to detect GSH. The advantages of the plasmonic nanoprobe including low-cost production techniques, outstanding stability and biocompatibility, high specificity and sensitivity towards GSH with a detection limit of 0.05 μM. It enables real-time dynamic monitoring of GSH levels in living cells during erastin-induced ferroptosis. This approach is expected to provide important theoretical support for elucidating the GSH-related ferroptosis metabolic mechanism and advancing our understanding of ferroptosis-based cancer therapy. Graphical abstract Overview of the workflow of sensing principle for highly sensitive, in situ and dynamic tracking of intracellular GSH levels during drug-triggered ferroptosis process.

Circulating tumor cells (CTCs) are cancer biomarkers for the diagnosis at early stage. However, it’s still a great challenge to develop the cytosensor for CTCs detection due to its ultralow counts in human peripheral blood. Herein, a highly sensitive cytosensor based on surface-enhanced Raman scattering (SERS) was developed. The tumor cells were first identified by a strand replacement reaction (SDR) that released the initiator of catalytic hairpin assembly (CHA) reaction. Subsequently, the initiator opened the residue hairpin DNA outside the silver coated gold nanoparticles ( [email protected] NPs). The double strand DNA (dsDNA) CHA products drove the [email protected] NPs towards the same side of dsDNA to form a zipper-like structure. This structure brought about hotspots area for SERS quantitative analysis of A549 tumor cells. The designed cytosensor exhibited good linearity ranging from 10 to 500 cells/mL with a calculated limit of detection (LOD) of 8 cells/mL. A SERS cytosensor with high sensitivity and good reproducibility was obtained in this work. In addition, this research presents a new method for rapid tumor cell detection in biological samples for cancer diagnosis.

Multiplexed detection of Parkinson’s disease (PD) biomarkers is of great significance for early diagnosis and personalized treatment. In this study, we fabricated a robust surface-enhanced Raman scattering-enabled lab-on-a-chip (LoC-SERS) platform for simultaneous quantification of α-synuclein, phosphorylated tau protein 181, osteopontin, and osteocalcin. Herein, the antibody–DNA conjugate was designed to introduce the catalytic hairpin self-assembly (CHA) amplification into the protein detection. Au nano-stars (AuNSs) modified with Raman reporter molecules and hairpin-structure DNA 1 were applied as the SERS nanotags. Au-coated silicon nanocone array (Au/SiNCA) fabricated based on the maskless plasma etching-prepared high-density Si nanocone array (SiNCA) and surface ion sputtering was used as the capture substrate after the modification of hairpin-structure DNA 2. Benefitting from the antibody–DNA conjugate-induced CHA amplification, numerous AuNSs can be connected to the Au/SiNCA surface, which significantly amplify the plasmonic coupling effect for ultrasensitive SERS detection, and the limit of detection was less than the pg/mL level. The application of highly uniform Au/SiNCA and antibody–DNA conjugate endows the LoC-SERS platform excellent analytical performance, including superior reproducibility, satisfactory universality, and high sensitivity. In addition, a 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced PD mice model was established, and satisfactory results were obtained in real sample analysis with the LoC-SERS platform, which may be enlightening for exploiting protein biomarkers in PD monitoring.

Herein, a SiO 2 @Ag NPs core/shell nanoparticles were synthesized to fabricate a surface-enhanced Raman spectroscopy (SERS) sensor for the simultaneous determination of histamine (HIS) and tyramine (TYR) based on specific aptamer recognition and ratiometric strategy. SiO 2 @Ag NPs with 4-thiosaminophenol (4-ATP) and Nile blue A (NBA) molecules were used as an internal standard (IS) and labeled with aptamers corresponding to HIS and TYR, respectively. Raman reporter molecules ROX and Cy5 labeled complementary DNA (cDNA) were then hybridized with aptamers to form rigid double-stranded DNA. After the HIS and TYR were captured by their aptamers, resulting in the dissociation of cDNA and separated from the SERS substrate. Therefore, the SERS signal intensity at 1503 cm −1 of ROX and 1358 cm −1 of Cy5 tagged on the terminal of cDNA decreased with the concentration of HIS and TYR increasing, while the SERS signal intensity at 1079 cm −1 of 4-APT and 592 cm −1 of NBA on the substrate remain stable. Thus, the concentrations of HIS and TYR can be determined by the I 1503 /I 1079 and I 1358 /I 592 values, respectively. This sensing strategy achieves a lower detection limit of 0.2 ng/mL for HIS and 0.05 ng/mL for TYR, respectively, demonstrating promising applications in sensitive detection of BAs in animal-derived foodstuff.

The surface-enhanced Raman scattering (SERS) technique is a promising method for the detection of explosives such as 2,4,6-trinitrotoluene (TNT) and 3-nitro-1,2,4-triazol-5-one (NTO) because of its high sensitivity to trace substances. However, most SERS detection processes are often nonautomated as well as exhibit low efficiency and toxic exposure, which often poses potential danger to operators. Herein, we propose the integration of SERS with digital microfluidics (SERS-DMF) for automated, high-throughput, and high-sensitivity detection of explosives. First, we carefully designed a DMF chip comprising 40 drive electrodes and 8 storage electrodes to achieve a high-throughput process. And different concentrations of target molecules, silver nanoparticles (Ag NPs), and salts were loaded into the DMF chip. Then, the droplet aggregation, incubation, and detection processes were automatically controlled using the SERS-DMF platform. In addition, Ag NPs were efficiently aggregated by screening different types and concentrations of salts, resulting in “hotspots” and the SERS effect. With the help of the SERS-DMF platform, two explosive samples were automatically detected with high throughput and high sensitivity. The detection limits of TNT and NTO were 10–7 and 10–8 M, respectively. In addition, compared with nonautomatic operations, the SERS-DMF platform exhibited better reproducibility and higher efficiency for the detection of explosives. The proposed SERS-DMF thus has considerable potential as an analytical technique for detecting hazardous substances.

Tyrosinase (TYR) which can catalyze the oxidation of catechol is recognized as a significant biomarker of melanocytic lesions, thus developing powerful methods for the determination of TYR activity is highly desirable for the early diagnosis of melanin-related diseases, including melanoma. Herein, we develop a novel portable and recyclable surface-enhanced Raman scattering (SERS) sensor, prepared by assembling gold nanoparticles and p-thiol catechol (p-TC) on an ITO electrode, for detecting TYR activity via the SERS spectral variation caused by the conversion of p-TC into its corresponding quinone under TYR catalysis. The developed SERS sensor has a rapid response to TYR within 1 min under the optimized conditions and shows high selectivity for TYR with the detection limit at 0.07 U/mL. Importantly, this SERS sensor can be easily regulated by applying negative voltage to achieve circular utilization, favoring the automation of SERS detection. Furthermore, the presented recyclable SERS sensor can perform well on both the determination of TYR activity in serum and the assessment of TYR inhibitor, demonstrating huge potential in the sensitive, selective, and facile detection of TYR activity for disease diagnosis and drug screening related with TYR.

Molecularly imprinted polymers (MIPs) have the ability of predesigned specific recognition that can be selected to construct reliable functional materials. Combining the MIPs with magnetic nanoparticles results in magnetic MIPs (MMIPs) which possess both facile manipulation and high selectivity. In this report, MMIP microspheres are fabricated by coating a layer of MIPs on Fe3O4@Ag for SERS-based sensing of molecular species. The obtained Fe3O4@Ag@MIPs are further applied for simple, rapid, ultrasensitive, and label-free SERS detection of sibutramine (SIB, illegal additive), with a low LOD of 1.0 × 10–9 M. Moreover, the MMIP microspheres exhibit ultrahigh adsorption efficiency, selectivity, reusability, and good structural stability, which demonstrates the potential application for SERS. Particularly, the proposed approach of MMIPs-SERS can be successfully applied to quick screening of SIB in slimming supplements such as capsules, tea powder, and tablets, and the content for those is 6.18, 13.61, and 3.09 mg g–1, respectively. These properties make MMIP microspheres a promising SERS substrate to detect illegal additives in health-related products and chemical pollutants in the environment.

A three-dimensional surface enhanced Raman scattering (SERS) substrate based on graphene nanofoam (GNF)/AgNPs hybrid nanostructure has been prepared using a typical liquid phase reduction synthetic method. The synthesized materials which utilized the three-dimensional framework structure of NF with micropore diameter 50–500 μm were illustrated by scanning electron microscope (SEM) images. Meanwhile, the NF was coated with graphene oxide (GO) evidenced by energy dispersive spectra (EDS) images. In addition, the composite was modified with Ag nanoparticles (AgNPs) whose average size was about 500 nm. The preparation process is very economic and environmental-friendly. The SERS experiment of hybrid nanostructure substrate using Rhodamine 6 G (R6G) as probe molecules showed that the concentration of GO aqueous solution and the Ag NPs growth time were two key factors to determine the intensity of SERS signal and the limit of detection of the hybrid nanostructure is as low as 1 × 10−8 M. 30 different test areas were selected to evaluated the homogeneous SERS platforms, and the RSD is only 8.2%, indicating highly uniformed and reproducibility. In summary, the three-dimensional (3D) substrates by a simple method showed a good sensitivity, stability and reproducibility, and it is promising for application in the field of rapid trace molecule detection in the realistic environment.

In this work, we prepared graphene oxide (GO)-decorated silver-coated gold nanobones (AuNBs@Ag@GO) for obtaining optimal surface-enhanced Raman scattering (SERS) activity. 4-Mercaptobenzoic acid (4-MBA) was adsorbed on the interface between gold and silver to provide more space for GO decoration on the outer surface of silver. Silver was deposited on the middle and end surfaces of the gold nanobones (AuNBs) with negative curvature, which resulted in stubby bimetallic nanocuboids inducing transverse surface plasmon resonance (SPR); the intermediate SPR merged into a new plasmon resonance. The SERS activity of silver-coated gold nanobones (AuNBs@Ag) changed non-monotonically with an increase in the silver coating, which reached up to 8 times larger than that of bare AuNBs. This is better performance than that of silver-coated gold nanorods (AuNRs@Ag). GO hybridized with AuNBs@Ag by electrostatic adherence could further improve the SERS activity of AuNBs@Ag, and the concentration of GO was adjusted for the best SERS performance, which was 4 times higher than that of naked AuNBs@Ag. AuNBs@Ag@GO can be used as a SERS substrate to detect glucose in urine. The mechanism is that the higher concentration of liquid glucose induced a large density of coverage, forming a glucose partition layer on the AuNBs@Ag@GO substrate: with an increase in the glucose concentration, the thickness of the glucose partition layer increased. This reduced the intensity of incident light and resulted in a decrease in the Raman signal of 4-MBA. Moreover, the strong SERS activity of the AuNBs@Ag@GO substrate can provide a wider space to achieve a lower limit of detection (LOD) value for glucose. The intensity of the characteristic Raman peak at 1585 cm−1 linearly decreased with an increase in the glucose concentration in the range of 10–107 nmol L−1 with a low LOD value (S/N = 3) of 2.61 nmol L−1. At the same time, it is confirmed that the SERS substrate can be selective and reliable and shows promising potential and broad applications in glucose detection.
