Staphylococcus aureus enterotoxin B (SEB) is a leading cause of foodborne illness. In this study, we developed a time-resolved fluorescent immunochromatography test strip (TRFITS) combined with a hydrogel microneedle patch (HMNP) and machine learning (ML) for the rapid detection of SEB in salmon. The developed HMNP, fabricated from methacrylated gelatin, can automatically extract 37 mg of interstitial fluid within 6 min. We then conducted TRFITS analysis on salmon samples to detect SEB and ML algorithms so as to predict its concentration. This method enabled efficient sampling and accurate quantification of SEB within 32 min, with a limit of detection (LOD) of 4.63 ng/mL. In addition, ML-assisted TRFITS applied to salmon demonstrated that the YOLOv11 model achieved a prediction accuracy of 91.3% and a lower LOD of 2 ng/mL. These findings highlight a novel approach toward on-site SEB detection with significant implications for food safety monitoring and point-of-care diagnostic applications.

Multidrug-resistant bacterial infections are a grand challenge to global medical and health systems. Therefore, it is urgent to develop versatile antibacterial strategies that can combat bacterial resistance without displaying toxicity. Here, we synthesize antibacterial polypeptide-conjugated gold nanoparticles that exhibit potent antibacterial activities against clinically isolated multiple drug resistance Gram-positive bacteria, such as methicillin-resistant Staphylococcus aureus, and excellent in vitro and in vivo biocompatibility. The antibacterial mechanism study indicates that over-production of reactive oxygen species results in the killing of bacteria. The overall antibacterial performance of these polypeptide-conjugated gold nanoparticles and the convenient synthesis of these polypeptides via lithium hexamethyldisilazide-initiated fast ring-opening polymerization on α-amino acid N-carboxyanhydride imply the potential application of this strategy in treating bacterial infections.

Accurate and rapid identification of Staphylococcus aureus ( S. aureus ) is of great significance for controlling the food poisoning and infectious diseases caused by S. aureus . In this study, a novel strategy that combines lysin cell-binding domain (CBD)-based magnetic separation with fluorescence detection was developed for the specific and sensitive quantification of S. aureus in authentic samples. The S. aureus cells were separated from the sample matrix by lysin CBD-functionalized magnetic beads. Following lysis by lysostaphin, intracellular catalase was released from S. aureus cells and detected by a fluorometric system composed of horseradish peroxidase (HRP), hydrogen peroxide (H 2 O 2 ), and Amplex Red. S. aureus was quantified via the inhibitory effect of the released intracellular catalase on the fluorometric system since the catalase could decompose the H 2 O 2 . Optimized conditions afforded a calibration curve for S. aureus ranging from 1.0 × 10 2 to 1.0 × 10 7 CFU mL −1 . The detection limit was as low as 78 CFU mL −1 in phosphate-buffered saline (PBS), and the total detection process could be completed in less than 50 min. Other bacteria associated with common food-borne and nosocomial infections negligibly interfered with S. aureus detection, except for Staphylococcus epidermidis , which may have slightly interfered. Moreover, the potential of this proposed method for practical applications has been demonstrated by detection assays of sterilized milk and human serum.

Fructus crataegi (hawthorn) is the common name of all plant species in the genusCrataegusof the Rosaceae family. In the present study, three monomers of (+)-catechin (C), (−)-epicatechin gallate (ECg) and (−)-epigallocatechin (EGC) were isolated from the hawthorn under the guide of antibacterial sensitization activity. The bioactivity of the composite fraction in enhancing the antibacterial effect of oxacillin against methicillin-resistantStaphylococcus aureus(MRSA) was greater than that of the individual monomer of the hawthorn extractin vitro. Two-fold dilution and checkerboard methods were used to analyze antibacterial activity and screen for the combination and proportion of monomers with the best bioactivity. The result showed that C (128 mg/L) combined with ECg (16 mg/L) had the greatest effect and the combination also reduced the bacterial load in blood of septic mice challenged with a sublethal dose of MRSA, increased daunomycin accumulation within MRSA and down-regulated the mRNA expression ofnorA,norCandabcA, three important efflux pumps of MRSA. In summary, C and ECg enhanced the antibacterial effect of β-lactam antibiotics against MRSAin vitroandin vivo, which might be related to the increased accumulation of antibiotics within MRSA via suppression of important efflux pumps’ gene expression. Keywords:fructus crataegi;(+)-catechin;(−)-epicatechin gallate;synergistic effect;MRSA;drug accumulation

The healing of wounds infected with drug-resistant bacteria has become a serious issue for the medical system, while designing an all stage biofilm-resistant wound dressing which can ideally address this problem still remains a challenge. Herein, we designed a photothermal/photodynamic/inherent synergistic antibacterial hydrogel based on caffeic acid grafted ε-poly- l -lysine (CE)-modified Cu-doped carbon quantum dots (CQDs-CE) and chitosan-phenylboronic acid (CSP)/oxidized dextran (OD)-based dual-dynamic-bond cross-linked hydrogels (CSP/OD/CQDs-CE) for the healing of methicillin-resistant Staphylococcus aureus ( MRSA ) infected wounds. The dual dynamic network crosslinked by pH-sensitive boronate esters and Schiff base bonds offers the hydrogel excellent self-healing properties and on-demand painless removal capacity. Mechanistically, antibacterial activity relied on membrane disruption by CE, reactive oxygen species generation from Cu-CQDs, photothermal/photodynamic damage mediated by CQDs, and electrostatic interactions from positively charged chitosan. Notably, the hydrogel achieved biofilm clearance ratios of 91.1 % in vitro and 90.6 % in vivo , markedly higher than those of the 6S Ag + group (54.8 % and 64.0 %, respectively). Besides, the hydrogel exhibited good biocompatibility, injectability, antioxidant properties, and hemostatic properties. In MRSA -infected full-thickness skin defect models, wounds treated with CSP/OD/CQDs-CE1 + NIR showed a final healing ratio of 99.7 %, significantly surpassing that of Tegaderm™ hydrocolloid dressing (83.0 %). While effectively inhibiting infection, reduce inflammation, accelerate angiogenesis, and promote collagen deposition, suggesting great potential for the treatment of wounds infected with drug-resistant bacteria.

Honokiol targets HptA to block G‑6‑P uptake and induce lethal carbon starvation. • Carbon starvation collapses MRSA central metabolism and weakens biofilm formation. • Metabolic inhibition lowers NADPH, amplifies ROS, and drives oxidative damage. • Honokiol reduces MRSA burden, dampens TLR2/NF‑ κ B, and accelerates wound healing. • HptA is absent in mammals, giving HNK high target selectivity and low toxicity. Background Methicillin-resistant Staphylococcus aureus (MRSA) compromises chronic wound healing, and growing antibiotic resistance necessitates novel agents that target unconventional bacterial vulnerabilities. Purpose To investigate the anti-MRSA efficacy of honokiol and to characterize hexose phosphate transport regulatory protein A (HptA), a regulator of alternative carbon source uptake under nutritional stress, as a therapeutic target. Methods Anti-MRSA activity was evaluated in vitro , with MIC determination. HptA protein was purified, and HptA–honokiol interactions were characterized using molecular docking, bio-layer interferometry, and related techniques. Multi-omics analysis were performed to elucidate metabolic responses. Therapeutic efficacy was assessed in mouse model of MRSA-induced wound infection. Results Honokiol exhibited moderate anti-MRSA activity with an MIC of 36 µg/ml. Mechanistically, honokiol allosterically locks HptA into a nonfunctional conformation, blocking glucose-6-phosphate uptake under nutrient-limited wound conditions. This induces carbon starvation, redox imbalance, and a ROS burst, leading to membrane damage, DNA fragmentation, and biofilm eradication. Local bacterial clearance subsequently reduced peptidoglycan-driven TLR2/NF- κ B activation, attenuated inflammation, and significantly accelerated wound healing in vivo . Conclusion HptA is identified as a potential antibacterial target against MRSA. By blocking alternative carbon uptake, honokiol induces carbon starvation‑driven lethal stress; lowered bacterial burden then inhibits TLR2/NF‑ κ B signaling, accelerating wound healing through a localized antibacterial action. Susceptibility point-of-care testing (POCT) and bacterial inactivation are crucial for food safety and clinical diagnosis since bacteria pose a severe hazard to public health and healthcare. To enable photothermal determination and hyperthermal inactivation of bacteria, vancomycin-modified near-infrared (NIR)-responsive copper selenide nanoparticles (Cu 2−X [email protected] NPs) with excellent photothermal conversion are synthesized here. Cu 2−X [email protected] NPs can be trapped on the surface of Gram-positive bacteria such as Staphylococcus aureus ( S. aureus ), forming a Cu 2−X [email protected] NPs/ S. aureus complex. Suspension (Cu 2−X [email protected] NPs) and sediment (Cu 2−X [email protected] NPs/ S. aureus ) can convert the NIR laser energy into a thermal signal that can be detected by the thermal camera at concentrations as low as 6.0 CUF mL −1 . Meanwhile, S. aureus can be effectively inactivated by localized temperature rises during temperature-based signal detection. The antibacterial efficiency can reach as high as 99.53%. The proposed strategy not only provides a sensitive POCT strategy for bacterial determination but also implements efficient inactivation of bacteria simultaneously.

Immunomagnetic separation based on Fe3O4 magnetic nanoparticles (MNPs) has been widely performed in sample pretreatment. The oriented conjugation strategy can achieve a better capture effect than the N-(3-dimethylamlnopropyl)-N'-ethylcarbodiimide hydrochloride (EDC) /N-hydroxysuccinimide (NHS) method. However, immunoglobulin yolk (IgY) cannot be oriented through an SPA strategy like immunoglobulin G (IgG). In this article, an oriented conjugation nanoprobe was prepared for the enrichment of bacteria based on pH adjusting. The main factors affecting the enrichment efficiency were studied, such as the pH of the buffer system, the concentration of IgY, the concentration of nanoprobe, and the enrichment time. Under the optimal conditions, the enrichment efficiency toward target bacteria could reach 92.8%. Combined with PCR, the limit of detection (LOD) was found to be 103 CFU/ml, which was lower than the PCR only. In conclusion, we provided a new protocol for the oriented conjugation of IgY and high sensitivity detection with simple pretreatment.

Although bacteriophage-based biosensors are promising tools for the rapid, convenient, and sensitive detection of S. aureus in food products, the effect of biosensors using temperate phages as the biorecognition element to detect viable S. aureus isolates remains unclear. In this study, four temperate Staphylococcus aureus phages were isolated, and their biological features were compared. The selected phage, SapYZUs7, was immobilised on an MnCo 2 O 4 nanozyme to prepare SapYZUs7@MnCo 2 O 4 , and the application of SapYZUs7@MnCo 2 O 4 in the S. aureus detection was evaluated. Compared to the other three S. aureus phages, SapYZUs7 exhibited a higher titre, broader host range, greater pH tolerance (3–12, > 91 %), higher absorption rate (71.6 %, 6 min), and more unique gene content. The characterisation of SapYZUs7@MnCo 2 O 4 revealed its successful immobilisation, excellent biological activity, and peroxidase-mimicking ability. Moreover, the SapYZUs7@MnCo 2 O 4 exhibited high sensitivity and specificity for the rapid colorimetric determination of viable S. aureus counts, with a limit of detection of 0.77 × 10 2 CFU/mL. In addition, SapYZUs7@MnCo 2 O 4 was successfully employed for the colorimetric detection of S. aureus in food samples, regardless of different food additives , NaCl concentrations, and pH values. In conclusion, the SapYZUs7@MnCo 2 O 4 is a powerful tool for the detection of viable S. aureus in food matrices.

A one-step, one-tube colorimetric assay is described for the detection of bacterial double-stranded DNA (dsDNA). It utilizes a G-quadruplex DNAzyme produced by an asymmetric polymerase chain reaction (As-PCR) that catalyzes the oxidation of cysteine to form cystine. This results in the formation of oligonucleotide-modified gold nanoparticles via triplex formation, and eventually in a color change from red to blue that occurs within about 10 mins. This can be measured by ratiometric colorimetric (at 525 and 600 nm). The limit of detection (LOD) for the model analyte (dsDNA of Staphylococcus aureus ( S. aureus )) is as low as 0.28 pg per 0.05 mL with a good linear response ranging from 16.0 fg·μL −1 to 1.6 ng·μL −1 . This is much lower than previously reported LODs. The assay is highly selective for S. aureus dsDNA over a range of other bacterial DNAs. Conceivably, it provides an attractive alternative tool for rapid detection of bacterial dsDNA as required in pathogen screening in the food industry.

The present study investigated the antibacterial efficacy of telomycin nanoparticles (TNPs). Telomycin, a bioactive metabolite of Streptomyces abietis PP752347, was loaded onto chitosan nanoparticles. The minimum inhibitory concentration (MIC 50 ) of telomycin against pathogenic strains of Staphylococcus aureus was determined to be 1.0 ± 1.00 μg/mL and 2.0 ± 1.00 μg/mL, respectively, against different strains. Meanwhile, the (MIC 50 ) of TNPS against the same strains was .125 ± 1.00 μg/mL and 1.0 ± 1.00 μg/mL, respectively. Morphological changes in S. aureus treated with TNPS were examined using scanning electron microscopy (SEM). Additionally, the bioactive metabolite extracts were characterized using X-ray diffraction (XRD) and Fourier-transform infrared spectroscopy (FTIR), while SEM was employed to characterize the nanoparticles. For the in vivo experiment, 120 African catfish ( Clarias gariepinus ) were divided into four groups: the first group served as the negative control (neither challenged nor treated); the second group (TNPS) received TNPS treatment (100 μg/L); the third group (SA) represented the positive control, infected with S. aureus ; and the fourth group (SA + TNPS) was infected with S. aureus and treated with TNPS. Infected fish (SA and SA + TNPS groups) received an intraperitoneal injection of S. aureus (1.6 × 10 6 CFU/mL). The findings demonstrated that all infected groups exhibited noticeable clinical signs resulting from S. aureus infection. TNPS administration via immersion significantly reduced the negative impacts of infection on liver and kidney biomarkers, innate immune responses, antioxidant levels, serum gonadotropic hormones, and hepatic expression levels of both Caspase-3 and TNF-α. TNPS treatment significantly ( p < 0.05) alleviated S. aureus –induced clinical symptoms and hepatorenal dysfunction. It enhanced innate immunity (lysozyme, nitric oxide, MPO, and IL-1β expression), antioxidant capacity, and sex hormone levels, while reversing histopathological alterations in the liver, kidney, spleen, and ovaries. Furthermore, TNPs reduced hepatic expression of both Caspase-3 and TNF-α. These findings indicate that TNPs (100 μg/L) is safe for African catfish and exhibits potent antibacterial activity, supporting its potential as a promising and eco-friendly therapeutic agent for aquaculture.

Rhinitis is a widespread immune disorder that affects over 40% of the global population. It is typically managed with noninvasive inhaled antihistamine medications. However, frequent use of these drugs can cause significant irritation, making the development of targeted drug delivery systems a major challenge. Recent advancements in Chitosan (CS) biopolymer-based nanogels present a safer and more effective method for controlling rhinitis caused by MRSA. In this study, the antibiotic cefminox was encapsulated within a chitosan nanogel to achieve pH-responsive drug release aimed at eradicating Methicillin-resistant Staphylococcus aureus (MRSA). This CEF@CS nanogel effectively delivers cefminox under acidic conditions. The cleavage of the chitosan polymer outer layer within the CEF@CS nanogel facilitates the release of cefminox at the sites of bacterial infection. The chitosan coating enhanced MRSA growth reduction and improved the nanogel’s biocompatibility in L929 cells. Results from antibacterial assays indicated that cefminox’s pH-responsive and sustainable release effectively eradicated MRSA bacterial growth, even at low doses. The nanoscale antibiotic molecules can easily penetrate the bacterial cell wall, leading to ROS-mediated cell death. Additionally, the antibiofilm assay suggested that CEF@CS nanogel treatment could potentially prevent the formation of MRSA biofilms. The findings from the L929 cytotoxicity assay demonstrated that CEF@CS nanogel is a highly biocompatible therapeutic material for managing rhinitis. Overall, this research highlights the CEF@CS nanogel as an effective pH-responsive drug delivery system for controlling MRSA-related rhinitis and for nursing care applications.
