Vortex-Assisted Liquid–Liquid Microextraction for the Determination of Chloramine-T in Cosmetics Coupled with High-Performance Liquid Chromatography

Juan Wang,Zhihua Li,Yaling Yang

Vortex-assisted liquid–liquid microextraction is reported for the preconcentration of chloramine-T before determination by high-performance liquid chromatography with a diode array detector. Magnetite nanoparticles were used to remove hydrophobic additives in cosmetics by physical adsorption. The method used sonication for 30 min at 45°C in 40% hydrochloric acid to convert the analyte to p-toluenesulfonamide. 1-Hexanol was the extraction solvent with vortex mixing. The extraction conditions were optimized to provide a linear dynamic range from 0.5 to 500.0µg/mL. The recovery of chloramine-T was from 90.8 to 100.9% with a relative standard deviation from 3.7 to 6.5%. The limit of detection was 8.2 mg/kg and the enrichment factor (EF) was 100. Good repeatability, quantitative recovery, and a high EF demonstrated the suitability of the procedure for the determination of chloramine-T in cosmetics.

Solid–Liquid Phase Equilibrium of N,N′-Piperazinebis(neopentylglycol)phosphoramidate in Selected Solvents from T = (293.15 to 333.15) K

Dong-Fang Zhao,Li-Sheng Wang,Jian Sun,Chuan-Ming Chen

A phosphorus flame retardant named N,N′-piperazinebis(neopentylglycol)phosphoramidate (PBNGP) was synthesized and characterized by elemental analysis, Fourier transform infrared spectroscopy, mass spectra, and nuclear magnetic resonance. Its thermal properties were evaluated via thermogravimetric analysis and differential scanning calorimetry. The solubilities of PBNGP in selected solvents at T = (293.15 to 333.15) K and atmospheric pressure were measured via a static-analytic method. The experimental data were correlated with Scatchard–Hildebrand, Buchowski-Ksiazczak, Wilson, nonrandom two liquid, and universal quasichemical equations. The calculated results showed that all of these models can reproduce the experimental data well, and the Wilson model gives the best correlation, with an overall relative standard deviation of 0.70%. The solubility parameter of PBNGP was calculated using the Scatchard–Hildebrand methodology.

Activity Coefficients of [Cnmim]Br (n = 3 to 8) Ionic Liquids in Aqueous Fructose Solution at T = 298.15 K

Kelei Zhuo,Hao Ren,Yujing Wei,Yujuan Chen,Jingjing Ma

Activity coefficients of the 1-alkyl-3-methylimidazolium bromide [Cnmim]Br (n = 3 to 8) ionic liquids (ILs) in fructose + water mixed solvents at 298.15 K were determined by cell potential measurements. The molalities of [Cnmim]Br ranged from (0.005 to 0.1) mol·kg–1 and those of fructose from (0.2 to 0.8) mol·kg–1. Gibbs free energy interaction parameters were also obtained together with salt constants. The interactions between [Cnmim]Br and fructose are mainly controlled by electrostatic interactions. Gibbs free energy interaction parameters (gES) and salting constants (kS) are negative for the ILs (n = 3 to 6), indicating fructose are salted-in by the ILs in water, whereas fructose are salted-out by the ILs (n = 7 and 8).

Densities, Viscosities and Excess Properties of Binary Mixtures of 1,1,3,3-Tetramethylguanidinium Lactate + Water at T = (303.15 to 328.15) K

Shidong Tian,Shuhang Ren,Yucui Hou,Weize Wu,Wei Peng

Densities and viscosities of 1,1,3,3-tetramethylguanidium lactate ([TMG]L) (1) + H2O (2) with different compositions were determined at temperatures from (303.15 to 328.15) K. Densities of [TMG]L (1) + H2O (2) decrease with the increase of temperature. However, when the mole fraction of water is lower than 0.5, the water content does not have an obvious influence on the density of [TMG]L. When the mole fraction of water is greater than 0.5, the densities of [TMG]L (1) + H2O (2) decrease obviously with further increasing the mole fraction of water. Viscosity of [TMG]L decrease dramatically with the increase of temperature and the water content. Excess molar volumes (VE) and viscosity deviations (Δη) of the binary mixtures were calculated by the experimental data and fitted to the Redlich–Kister equation with four parameters. The result shows that the values of VE and Δη for the binary mixtures are negative over the whole composition range and temperatures from (303.15 to 328.15) K. The negative of the excess properties indicates a strong interaction between the ionic liquid and water. The partial molar volumes (V̅i), excess partial molar volumes (V̅iE), Gibbs energy of activation for viscous flow (ΔG*), and excess Gibbs energy of activation for viscous flow (ΔG*E) of the binary mixtures were also calculated based on the densities and viscosities data. Besides, the Jouyban–Acree model was used to correlate the densities and viscosities of the binary mixtures with respect to the mixture composition and temperature simultaneously.

A High Sensitivity Electrochemical Sensor Based on Fe3+-Ion Molecularly Imprinted Film for the Detection of T-2 Toxin

Xing Gao,Wenying Cao,Miaomiao Chen,Huayu Xiong,Xiuhua Zhang,Shengfu Wang

The excellent detection sensitivity in various matrices of T-2 toxin (T-2), which has cytotoxic and immunosuppressive effects in DNA and RNA synthesis, is a highly desirable characteristic. A sensitive molecularly imprinted electrochemical sensor was constructed for the selective detection of T-2. In this study, iron ions (Fe 3+ ) were introduced to increase the chelation of the metal ions and templates for preparing molecularly imprinted polymers (MIPs). With the increased chelation of the metal ions and templates, the selectivity and sensitivity of the MIPs were effectively improved. The imprinted sensor was successfully employed to detect T-2 in cereals and human serum samples.

Phase Diagrams and Physicochemical Properties for the Ternary System (CsCl + NaCl + H2O) at T = (298.15, 308.15, and 318.15) K

Yunxue Gao,Shuni Li,Quanguo Zhai,Yucheng Jiang,Mancheng Hu

Complete phase diagrams and physico-chemical properties for the ternary system (CsCl + NaCl + H2O) were determined at T = (298.15, 308.15, and 318.15) K, respectively. The X-ray diffraction method and Schreinemaker’s wet residue method were used to verify the solid phase. The phase diagrams show this ternary system is a complex type. Three crystallization regions were found corresponding to the single salt NaCl, a compound CsCl·2NaCl·2H2O, and a solid solution series [Cs1–x(Na·H2O)x]Cl (x in [Cs1–x(Na·H2O)x]Cl changed from 0 to 0.43 (298.15 K), 0.39 (308.15 K), and 0.36 (318.15 K) at each temperature. With rising temperature, the crystallization area of NaCl increased, but CsCl·2NaCl·2H2O and [Cs1–x(Na·H2O)x]Cl decreased simultaneously. However, density and refractive index increased with increasing CsCl concentration.

Generating Immunological Memory Against Cancer by Camouflaging Gold-Based Photothermal Nanoparticles in NIR-II Biowindow for Mimicking T-Cells

Xuefan Xiong,Ying Zhang,Xinqi Huang,Shusheng Zhang,Qiong Li

Photothermal therapy (PTT) against cancer not only directly ablates tumors but also induces tumor immunogenic cell death (ICD). However, the antitumor immune response elicited by ICD is insufficient to prevent relapse and metastasis because of the immunosuppressive tumor microenvironment (TME). A biomimetic nanoplatform (bmNP) mimicking cytotoxic lymphocytes (CTLs) for combinational photothermal-immunotherapy to effectively regulate the immunosuppressive TME is reported here. The bmNP is constructed by wrapping the T -cell membrane onto a new type of photothermal agents, spherical Au-based PNCs (sAuPNCs). Similar to T -cells, the bmNP enhanced accumulation at the tumor site by targeting the tumor via adhesion proteins on T -cell membrane. The obtained sAuPNCs have a wide absorption band in the second near-infrared (NIR-II) region with a high photothermal conversion efficiency (PCE) up to about 75% and excellent photostability. The bmNP with a smaller size is more superior compete with T -cells to bond with tumor cells via PD-1/PD-L1 interaction to effectively block the PD-1 checkpoint of T -cells for preventing T -cell exhaustion. Furthermore, in vivo studies reveal the immunological memory effect is significantly elicited in mice received bmNPs therapy. Collectively, bmNPs show great potential in photothermal-enhanced immunotherapy.

Identification of genetic modifiers enhancing B7-H3-targeting CAR T cell therapy against glioblastoma through large-scale CRISPRi screening

Li Xing,Sun Shiyu,Zhang Wansong,Liang Ziwei,Fang Yitong,Sun Tianhu,Wan Yong,Ma Xingcong,Zhang Shuqun,Xu Yang,Tian Ruilin

Background Glioblastoma multiforme (GBM) is a highly aggressive brain tumor with a poor prognosis. Current treatment options are limited and often ineffective. CAR T cell therapy has shown success in treating hematologic malignancies, and there is growing interest in its potential application in solid tumors, including GBM. However, current CAR T therapy lacks clinical efficacy against GBM due to tumor-related resistance mechanisms and CAR T cell deficiencies. Therefore, there is a need to improve CAR T cell therapy efficacy in GBM. Methods We conducted large-scale CRISPR interference (CRISPRi) screens in GBM cell line U87 MG cells co-cultured with B7-H3 targeting CAR T cells to identify genetic modifiers that can enhance CAR T cell-mediated tumor killing. Flow cytometry-based tumor killing assay and CAR T cell activation assay were performed to validate screening hits. Bioinformatic analyses on bulk and single-cell RNA sequencing data and the TCGA database were employed to elucidate the mechanism underlying enhanced CAR T efficacy upon knocking down the selected screening hits in U87 MG cells. Results We established B7-H3 as a targetable antigen for CAR T therapy in GBM. Through large-scale CRISPRi screening, we discovered genetic modifiers in GBM cells, including ARPC4 , PI4KA , ATP6V1A , UBA1 , and NDUFV1 , that regulated the efficacy of CAR T cell-mediated tumor killing. Furthermore, we discovered that TNFSF15 was upregulated in both ARPC4 and NDUFV1 knockdown GBM cells and revealed an immunostimulatory role of TNFSF15 in modulating tumor-CAR T interaction to enhance CAR T cell efficacy. Conclusions Our study highlights the power of CRISPR-based genetic screening in investigating tumor-CAR T interaction and identifies potential druggable targets in tumor cells that confer resistance to CAR T cell killing. Furthermore, we devised targeted strategies that synergize with CAR T therapy against GBM. These findings shed light on the development of novel combinatorial strategies for effective immunotherapy of GBM and other solid tumors.

Amplification-free sensitive detection of Staphylococcus aureus by spherical nucleic acid triggered CRISPR/Cas12a and Poly T-Cu reporter

Zhang Xiaoyu,Sun Ruimeng,Zheng Haoran,Qi Yanfei

A spherical nucleic acid (SNA, AuNPs-aptamer) into CRISPR/Cas12a system combined with poly T-template copper nanoparticles as fluorescence reporter was fabricated to establish an amplification-free sensitive method for Staphylococcus aureus ( S. aureus ) detection. This method, named PTCas12a, utilizes the concept that the bifunction of SNA recognizes the S. aureus and triggers the Cas12a cleavage activity. Then, the Cas12a enzyme cleaves the Poly T40 to generate a signal change in Poly T-Cu fluorescence, indicating the presence or absence of the target bacteria. The PTCas12a platform demonstrated a detection limit as low as 3.0 CFU/mL (3 N/S) in a wide response range of 1.0 × 10 1 –1.0 × 10 6  CFU/mL for S. aureus detection, which holds significant potential in ensuring food safety and preventing the spread of diseases. Facile and Rapid Synthesis of Cerium-Modified UiO-66-NH2 for Efficient Removal of Safranine T from Aqueous Solutions

Li Zhengjie,Fang Chunxia,Ma Xilong,Liu Yixian,Xie Juan,Li Pengfei,Zhang Zhikun,Zhao Xudong,Sun Hua,Han Jilong

Herein, a facile and rapid preparation method was developed to synthesize Ce-doped UiO-66-NH 2 through a one-pot solvothermal method. This approach simultaneously introduces functional groups onto organic ligands and incorporating a novel metal into metal clusters. The incorporation of Ce ions effectively enlarged the pore size and increased the pore volume, thereby facilitating the diffusion of specific adsorbate molecules into the inner adsorption sites provided by the –NH 2 groups. Due to the favorable pore structures and more exposed adsorption sites, the safranine T (ST) adsorption capacity of Ce-doped UiO-66-NH 2 was ten times higher than the pristine UiO-66-NH 2 and reached 1340 mg/g at optimal adsorption conditions, surpassing most of the reported porous adsorbents. The kinetic and isotherm fitting results showed that the pseudo-second-order and Langmuir models described the adsorption behaviors of ST best, indicating a homogeneous, endothermic, and spontaneous monolayer adsorption process. The adsorption mechanism showed that the hydrogen bonding interaction and pore filling effect are identified as the main mechanism whilst electrostatic attraction interaction further enhances the adsorption capacity at optimal pH. Therefore, Ce-doped UiO-66-NH 2 can serve as a promising adsorbent for the removal of cationic dyes from textile wastewater, and the developed facile and rapid method offers a viable strategy to construct novel MOF-based adsorbents.

Piceatannol improves autoimmune hepatitis by inhibiting the immune activities of T cells and macrophages through binding with c-Jun

Xu Cong,Lu Chenqi,Wang Wu,Wang Zhimin,Qiu Yang,Han Min,Yang Jun,Li Shanglin

Autoimmune hepatitis (AIH) is an immune-mediated liver disease that currently lacks viable drug treatment methods. This study is to explore the role of piceatannol (PIC) in ConA-induced AIH and the related mechanisms. A mouse model of AIH was established by injecting ConA (i.v.), and PIC was administered as an intervention. The protective effect of PIC was evaluated by the liver function, liver pathology, and serum levels of inflammatory factors. Subsequently, network pharmacology was used to predict the pathways and targets of PIC in the treatment of AIH, and the predicted results were validated using flow cytometry, molecular docking, surface plasmon resonance (SPR) and so on. Finally, the immunosuppressive effect of PIC was further validated in a mouse heart transplantation model. PIC can improve liver function decline and reduce pathological liver damage, as well as inhibit the increase of serum inflammatory factor levels in mice with AIH induced by ConA. The protective effect is achieved by suppressing the immune activity of T cells and macrophages through binding to c-Jun. PIC can also extend the survival of cardiac allografts and inhibit acute rejection reactions. These results indicated that PIC can significantly improve ConA-induced AIH in mice by inhibiting the immune activity of T cells and macrophages through binding to c-Jun. PIC can also extend the survival of cardiac allografts in mice and inhibit acute rejection responses. The above results indicated that PIC may serve as a promising immunosuppressant and be effective for AIH.

Baicalein prevents pregnancy loss by maintaining regulatory T cell activation through inhibition of the TLR4/NF-κB signaling pathway

Caiqun Huang,E Fen Cheng,Jinping Ni,Ying Lyu

Introduction Unexplained recurrent spontaneous abortion (URSA) involves multifactorial etiologies, with regulatory T cells (Tregs) playing a pivotal role in maintaining immune tolerance during pregnancy. Baicalein, a flavonoid from Scutellaria baicalensis, exhibits anti-inflammatory and immunomodulatory properties. This study evaluates baicalein's therapeutic potential in mitigating URSA via the TLR4/NF-κB signaling pathway. Methods A URSA mouse model was used, and baicalein was administered intraperitoneally. Pregnancy outcomes, abortion rates, and placental morphology were assessed on gestational day 14 (G14). Treg cells were quantified via flow cytometry, and gene/protein expression levels were analyzed by immunohistochemistry, Western blotting, and real-time PCR. In vitro experiments on ihESCs further investigated the role of the TLR4/NF-κB pathway. Results Baicalein reduced abortion rates from 33.3 % in URSA mice to 21.6 % (low dose) and 14.8 % (high dose), improved embryonic development by altering placental structure and decidual morphology, and reduced inflammation at the maternal-fetal interface. It expanded Treg cells and enhanced the expression of IGFBP-1 and PRL, markers of endometrial decidualization, and decreased TLR4, p-P65, and P65 expression. In vitro, baicalein's effects were abrogated by TLR4 inhibition, confirming pathway specificity. Discussion Baicalein improved pregnancy outcomes by enhancing Treg function and promoting decidualization via TLR4/NF-κB pathway inhibition. These findings highlight baicalein's potential as a therapeutic agent for URSA.
