Tetracycline hydrochloride loaded citric acid functionalized chitosan hydrogel for wound healing

Hao Chen,Biyun Li,Bei Feng,Hao Wang,Huihua Yuan,Zhiwei Xu

Citric modified chitosan (CC) hydrogel containing antibacterial drugs is developed by the freezing and thawing treatment method. The SEM image of the CC hydrogel revealed a porous structure. The rheological properties, porosity, swelling rate, water uptake, tensile properties and in vitro degradation were found to be tunable via CC concentration. To enhance antibacterial properties, tetracycline hydrochloride (TH) representing the drug model, was integrated into the CC hydrogel. The cumulative release of drug was also tunable via CC concentration. The drug loaded CC hydrogel showed enhanced antimicrobial activity against E. coli and S. aureus. In animal tests, it was found the TH loaded CC hydrogel accelerated the healing of the wounds created on rats. These results suggest that the drug loaded CC hydrogel has a promising future in wound healing as a wound dressing.

A functional polysaccharide film forming by pectin, chitosan, and tea polyphenols

Hao-Xiang Gao,Zheng He,Qun Sun,Qiang He,Wei-Cai Zeng

The formation, properties and functions of a polysaccharide film (CPFT) were investigated. CPFT was created by loading TP into pectin and chitosan matrix by blending method. Comparing with control, the thickness and the water vapor permeability of CPFT were increased with the addition of TP, and the tensile strength of CPFT was reduced by 17% while its elongation at break was increased by 37%. Then, fourier transform infrared (FTIR) spectrum and scanning electron microscope (SEM) results showed that TP had interacted with the film substrates and might made CPFT more complex. Furthermore, CPFT showed the remarkable slow-release, antioxidant and antiseptic activities, which were all concentration-dependent. Moreover, the degeneration of color in fresh beef could be effectively inhibited by CPFT during the 8 days storage. All present results suggested that CPFT showed the potential value to be used as a functional edible polysaccharide film applied in food industries.

Preparation and evaluation of porous starch/chitosan composite cross-linking hemostatic

Yan Chen,JunQing Qian,ChangYan Zhao,LieChao Yang,Jing Ding,Hui Guo

Composite porous starch (PS) with chitosan (CS) was easily cross-linked by sodium trimetaphosphate (STMP) to obtain a hemostat called STMP/PS/CS (SPC) with ideal hemostatic effect. The water absorption ratio and swelling ratio of SPC reach up to 150.8%, 355.0% respectively and it was observed by scanning electron microscopy (SEM) that a large amount of CS was combined with PS, which indicated the composite effect was ideal. Synergistic hemostatic effect of SPC was indicated in blood cells evaluation experiment and the activation of platelets triggers a multistep cascade that produces a clot by adsorbing more blood cells on the surface of SPC. The average hemostasis time of SPC in tail amputation 1 cm, 2 cm and liver laceration was shortened by 29%, 42%, 37% respectively compared with blank control, and had better hemostatic effect than CS, PS in the hemostatic performance on the rat tail amputation and liver laceration. Besides, we found SPC displayed a superior adhesiveness in rat injuries.

Chitosan Use in Dentistry: A Systematic Review of Recent Clinical Studies

Marco Cicciù,Luca Fiorillo,Gabriele Cervino

This study aims to highlight the latest marine-derived technologies in the biomedical field. The dental field, in particular, uses many marine-derived biomaterials, including chitosan. Chitosan that is used in different fields of medicine, is analyzed in this review with the aim of highlighting its uses and advantages in the dental field. A literature search was conducted in scientific search engines, using keywords in order to achieve the highest possible number of results. A review of randomized controlled trials (RCT) was conducted to evaluate and process all the relevant results for chitosan and oral health. After a screening and a careful analysis of the literature, there were only 12 results highlighted. Chitosan performs different functions and it is used in different fields of dentistry in a safe and effective way. Among the uses of chitosan, we report on the remineralizing property of chitosan which hardens tissues of the tooth, and therefore its role as a desensibilizer used in toothpastes. According to our systematic review, the use of chitosan has shown better surgical healing of post-extraction oral wounds. Furthermore, some studies show a reduction in bacterial biofilm when used in dental cements. In addition, it has antibacterial, antifungal, hemostatic and other systemic properties which aid its use for drug delivering. Keywords:chitosan;dentistry;oral surgery;dental material;marine drugs

Preparation and characterization of arginine-modified chitosan/hydroxypropyl methylcellose antibacterial film

Jinxing Song,Huafeng Feng,Mengqi Wu,Linxiao Chen,Wei Xia,Wenqing Zhang

Chitosan and its derivatives are widely used in medical, cosmetic and food fields. In this study, chitosan-N-arginine (CSA) was synthesized and characterized by Fourier-transform infrared (FT-IR), 1 H NMR, gel permeation chromatography (GPC), thermogravimetric analysis (TGA) and scanning electron microscopy (SEM). A novel antibacterial composite film consisting of CSA, hydroxypropyl methylcellose (HPMC) and glycerol was then prepared. The transparent and homogeneous film presented good compatibility between CSA and HPMC, confirmed by SEM. The thickness of the film was about 44.8 μm and its moisture content was 23.0%. Antimicrobial evaluation of CSA/HPMC film showed 9.0 mm bacteriostatic diameter zone against E. coli and 10.5 mm one against S. aureus . The film exhibited cell biocompatibility and promoted proliferation with L929 cell cytotoxicity test . Both antibacterial and cytotoxic results showed that the CSA/HPMC film was a promising material for medication, cosmetics and food preservation applications.

Rongli Zhang,Hui Hao,Cuige Zhang,Renchun Yang,Mingyue Sun,Ching-Ping Wong,Ying Xu

A kind of bioadhesive hydrocaffeic acid modified chitosan colloidal particles (HCA-CS/TPP CPs) containing synthetic catecholamine groups was prepared via amidation reaction and ionic gelation. The formation process and property of the HCA-CS/TPP CPs were characterized by ultraviolet visible spectrophotometry and Zeta PALS instruments. The prepared HCA-CS/TPP CPs were then used as particulate emulsifier to stabilize Pickering emulsions. The stability of the Pickering emulsion droplet was characterized by digital camera and optical microscope. The results revealed that HCA-CS/TPP CPs exhibited good emulsifying property at a wide pH value range, which will endow the potential application of the Pickering emulsions stabilized by HCA-CS/TPP CPs in various fields.

Preparation and physicochemical properties of antioxidant chitosan ascorbate/methylcellulose composite films

Wenqiang Tan,Jingjing Zhang,Xiang Zhao,Qing Li,Fang Dong,Zhanyong Guo

Polysaccharide-based biodegradable films have been considered as the promising candidates for food packaging industry instead of petroleum-based packaging materials. Here, we reported a class of edible composite films based on chitosan ascorbate and methylcellulose prepared by mixing different ratios (1,0, 4:1, 2:1, 1:1, 1:2, 1:4, and 0:1) of the biopolymers using the casting technique. Their physicochemical properties as well as the DPPH radical scavenging ability and reducing power were investigated. All physicochemical properties and antioxidant activities were significantly affected by the chitosan ascorbate/methylcellulose ratio in the matrix. The increases in tensile strength and elongation at break values, maximum decomposition temperatures, whitish index, compactness, moisture content, and a reduction in water vapor permeability were observed as the proportion of methylcellulose increased in the matrix. But the composite films containing a greater proportion of chitosan ascorbate exhibited the better barrier properties against UV–vis light and the stronger DPPH radical scavenging effect and reducing power. The chitosan ascorbate/methylcellulose composite films with interesting physicochemical properties and strong antioxidant action showed the potential value as biodegradable and edible biomaterials for food packaging.

Silver nanoparticles incorporated chitosan/calcium pyrophosphate hybrid microflowers for antibacterial applications

Shangpeng Liu,Jianwei Ma,Shuang Wang,Shaojuan Chen,Bin Wang,Jiwei Li

In this work, silver nanoparticles (Ag NPs) were incorporated into the flower-like hierarchical chitosan/calcium pyrophosphate hybrid microflowers (CS-CaP) via an in situ reduce process by polydopamine (PDA). Scanning electron microscope (SEM) observation, Energy dispersive spectroscopy (EDS), X-ray diffraction (XRD), and X-ray photoelectron spectroscopy (XPS) analysis confirmed the formation of Ag NPs on the petals of CS/CaP (Ag@CS/CaP). Since the hierarchical structure of Ag@CS/CaP facilitates the contact of Ag NPs with bacteria, results of the antibacterial test and cell culture showed that the Ag@CS-CaP has efficient antibacterial properties to both E. coli and S. aureus bacteria without causing cytotoxicity. These results demonstrate the Ag@CS-CaP has the potential capacity for biomedical applications.

Chitosan based polymer-lipid hybrid nanoparticles for oral delivery of enoxaparin

Wei Dong,Xiuhua Wang,Chang Liu,Xiao Zhang,Xin Zhang,Xianzhi Chen,Yongqiang Kou,Shirui Mao

In the present study, chitosan based polymer-lipid hybrid nanoparticles (PLNs) were prepared by a self-assembly method and their use as the carrier for oral absorption enhancement of enoxaparin was evaluated. The enoxaparin-loaded nanoparticles were composed of chitosan as the polymer and glyceryl monooleate as the lipid with a lipid/polymer mass ratio ranging from 0 to 0.3, and F127 was added as a stabilizer. It was found that the PLNs showed a higher surface hydrophobicity but mucoadhesive properties similar to those of chitosan based nanocomplexes. Results from DSC experiments and NMR solvent relaxation study indicate that glyceryl monooleate was completely incorporated into the nanoparticles and the lipid/polymer ratio affected the extent of lipid-polymer interaction inside the nanoparticles and the resultant internal structures. The stability of the PLNs in simulated gastrointestinal fluids was also affected by the lipid/polymer ratio; the best stability was shown by nanoparticles with a lipid/polymer ratio of 0.2. Nanoparticles with the optimal composition significantly enhanced the oral bioavailability of enoxaparin with a 4.5-fold increase in AUC in comparison with a solution of enoxaparin. In conclusion, GMO/CS based PLNs can provide a new insight to develop orally applicable delivery system for hydrophilic macromolecules. Their absorption can be enhanced with proposed PLNs and preparation of this PLNs was also found to be easy comparing to other similar methods.

Preparation and properties of chitosan/guar gum/nanocrystalline cellulose nanocomposite films

Yanjun Tang,Xinqi Zhang,Ruonan Zhao,Daliang Guo,Junhua Zhang

Nanocrystalline cellulose (NCC) has shown great potential for reinforcing nanocomposite systems, particularly in biopolymer-based nanocomposites. In the present work, transparent and biodegradable chitosan (CH)/guar gum (GG)/NCC nanocomposite films were designed and prepared. The effect of NCC loadings on the rheological behavior of the as-prepared nanocomposite suspensions was investigated, the overall properties of the resultant nanocomposite films as a function of various amounts of NCC were determined, and the surface morphology of various nanocomposite films was characterized by scanning electron microscopy (SEM). Results indicated that the increased NCC loadings increased the shear viscosity of the nanocomposite suspensions. Moreover, as expected, the obtained CH/GG/NCC nanocomposite films were found to exhibit improved tensile strength and reduced air permeability in comparison to pure CH/GG films. This work supported the conclusion that NCC was effective in reinforcing the CH/GG composite films, which may be a promising material for food packaging applications.

Preparation and BSA Adsorption Behavior of Chitosan-arginine Based Nanofiber Membranes

Li Xiaojie,Chao Guiqun,Wang Liangan,Xu Xianlin,Cai Zhanjun,Shi Lei,Zhuang Xupin,Cheng Bowen

In this work, an affinity nanofiber membrane was successfully prepared by solution blowing of arginine-modified chitosan (CS-Arg) for bovine serum albumin (BSA) adsorption. CS-Arg was firstly synthesized by coupling L-arginine onto chitosan backbone. Then, CS-Arg nanofiber membranes (CANFs) were fabricated using solution blowing process with Polylactide (PLA) as assistant polymer. The results showed that CANFs effectively adsorbed BSA, and the adsorption capacities were influenced by the degrees of substitution (DS) of arginine in CS, pH value, contact time, and initial protein concentration. The highest adsorption capacity of 445.19 mg/g was achieved uvnder the following conditions: DS of 43.7 %, pH of 7.14, and initial concentration of 3.0 mg/ml. BSA adsorbed on the CANFs membrane conformed to Langmuir model, and the adsorption rate was consistent with the second-order kinetics model. This work implies that an arginine-modified chitosan nanofiber-based novel biomaterial has a potential application in adsorption of BSA.

Electrospinning of Carboxymethyl Chitosan/Polyoxyethylene Oxide Nanofibers for Fruit Fresh-Keeping

Yue Tian-Tian,Li Xiao,Wang Xiao-Xiong,Yan Xu,Yu Miao,Ma Jian-Wei,Zhou Yu,Ramakrishna Seeram,Long Yun-Ze

Electrospinning provides an effective method for generating nanofibers from solution of carboxymethyl chitosan/polyoxyethylene oxide (CMCS/PEO). The goal of this work is to explore the potential application of electrospun CMCS/PEO nanofiber membrane in fruit fresh-keeping. The microstructure, antibacterial activity, hydrophilia, and air permeability of the nanofiber membrane have been tested. For comparison, the fresh-keeping effects of commercial cling wrap and CMCS/PEO nanofiber membranes on strawberries’ rotting rate and weight loss rate have been studied. The results indicate that the electrospun CMCS/PEO membrane could effectively avoid water loss in strawberries and has a remarkable effect to prolong strawberries’ shelf life due to its breathability and antibacterial activity. In addition, the composite CMCS/PEO, nanofiber membrane is non-poisonous and edible, which can be used in food industry.
