Novel Ferrocenyl-Terminated Linear–Dendritic Amphiphilic Block Copolymers: Synthesis, Redox-Controlled Reversible Self-Assembly, and Oxidation-Controlled Release

Xueyi Chang,Renfeng Dong,Biye Ren,Zhiyu Cheng,Jun Peng,Zhen Tong

Novel linear–dendritic amphiphilic block copolymers with hydrophilic poly(ethylene glycol) (PEG) block and hydrophobic Percec-type dendrons containing ferrocenyl terminals were synthesized by the esterification reaction of poly(ethylene glycol) methyl ether with ferrocenyl-terminated alkyl-substituted benzoic acid dendrons. On the basis of the results that the critical aggregation concentration (CACox) of the oxidation state polymer is much higher than CACred of the corresponding reduction state, these polymers can reversibly self-assemble into various aggregates, such as spherical, wormlike micelles, and vesicles, and also disassemble into irregular fragments in aqueous solution by redox reaction when changing the polymer concentrations. Copolymer PEG45-b-Fc3 (3) with 3,4,5-tris(11-ferrocenylundecyloxy) benzoic acid (2) can self-assemble into nanoscale wormlike micelles when the polymer concentration in aqueous solution is above its CACox. These wormlike micelles can be transformed into nanosized vesicles by Fe2(SO4)3 and regained by vitamin C. Interestingly, copolymer PEG45-b-Fc2 (5) with 3,5-bis(11-ferrocenylundecyloxy) benzoic acid (4) can reversibly self-assemble into spherical micelles with two different sizes by redox reaction above the CACox, indicating that the terminal hydrophobic tail number of dendrons plays a key role in determining the self-assembled structures. Furthermore, rhodamine 6G (R6G)-loaded polymer aggregates have been successfully used for the oxidation-controlled release of loaded molecules, and the release rate can be mediated by the concentrations of oxidant and copolymers. The results provide an effective approach to the reversible self-assembly of linear–dendritic amphiphilic block copolymers and also promise the potential of these novel redox-responsive amphiphilic block copolymers in drug delivery systems, catalyst supports, and other research fields.

Fe3O4@mSiO2 core–shell nanocomposite capped with disulfide gatekeepers for enzyme-sensitive controlled release of anti-cancer drugs

Chunyu Yang,Wei Guo,Liru Cui,Na An,Ting Zhang,Gang Guo,Huiming Lin,Fengyu Qu

Multifunctional nanocarriers based on the magnetic Fe3O4 nanoparticle core and bis-(3-carboxy-4-hydroxy phenyl) disulfide (R–S–S–R1) modified mesoporous silica shell (Fe3O4@mSiO2@R–S–S–R1) were synthesized for cancer treatment through passive targeting and enzyme-sensitive drug release. Anti-cancer drug doxorubicin (DOX) was used as the model cargo to reveal the release behavior of the system. The drug loading system (DOX–Fe3O4@mSiO2@R–S–S–R1) retains the drug until it reaches the tumor tissue where glutathione reductase (GSH) can degrade the disulfide bonds and release the drug. Furthermore, the grafting amount of R–S–S–R1 can be used to adjust the release performance. All the release behaviors fit the Higuchi model very well and the release kinetics are predominated by disulfide bond degradation and mesoporous structure. With good bioactivity and targeted release performance, the system could play an important role in the development of intracellular delivery nanodevices for cancer therapy.

Ecofriendly Controlled-Release Insecticide Carrier: pH-/Temperature-Responsive Rosin-Derived Hydrogels for Avermectin Delivery against Mythimna separata (Walker)

Shihao Gu,Mei Liu,Renle Xu,Xu Han,Yuhang Lou,Yue Kong,Yanqing Gao,Shibin Shang,Zhanqian Song,Jie Song,Jian Li

The exploration of environmentally friendly, less toxic, sustained-release insecticide is increasing with the growing demand for food to meet the requirements of the expanding population. As a sustained-release carrier, the unique, environmentally friendly intelligent responsive hydrogel system is an important factor in improving the efficiency of insecticide utilization and accurate release. In this study, we developed a facile approach for incorporating the natural compound rosin (dehydroabietic acid, DA) and zinc ions (Zn2+) into a poly(N-isopropylacrylamide) (PNIPAM) hydrogel network to construct a controlled-release hydrogel carrier (DA-PNIPAM-Zn2+). Then, the model insecticide avermectin (AVM) was encapsulated in the carrier at a drug loading rate of 36.32% to form AVM@DA-PNIPAM-Zn2+. Surprisingly, the smart controlled carrier exhibited environmental responsiveness, strongly enhanced mechanical properties, self-healing ability, hydrophobicity, and photostability to ensure a balance between environmental friendliness and the precision of the drug release. The release experiments showed that the carboxyl and amide groups in the polymer chains alter the intermolecular forces within the hydrogel meshes and ingredient diffusion by changing temperatures (25 and 40 °C) and pH values (5.8, 7.4, and 8.5), leading to different release behaviors. The insecticidal activity of the AVM@DA-PNIPAM-Zn2+ against oriental armyworms was good, with an effective minimum toxicity toward aquatic animals. Therefore, AVM@DA-PNIPAM-Zn2+ is an effective drug delivery system against oriental armyworms. We anticipate that this ecofriendly, sustainable, smart-response carrier may broaden the utilization rosin and its possible applications in the agricultural sector.

Mesoporous Silica with Dual Stimuli-Microenvironment Responsiveness via the Pectin-Gated Strategy for Controlled Release of Rosmarinic Acid

Lina Song,Wenxian Chen,Xinbo Lu,Ziqiang Liu,Qingqing Rao,Weiqiang Xiao,Xiaoli Zhan,Shengxiang Yang,Feng Gao,Qinghua Zhang

Traditional drug-delivery methods are limited by low bioavailability and nonspecific drug distribution, resulting in poor therapeutic efficacy and potential risks of toxicity. Mesoporous silica nanoparticles (MSNs) have attracted wide attention as drug-delivery carriers due to their large specific surface area, adjustable pore size, good mechanical strength, good biocompatibility, and rich hydroxyl groups on their surface. In this paper, MSNs were synthesized by a template method, and the morphology and pore structure were regulated. The obtained particles possessed a narrow particle size distribution and good sphericity with a diameter of 878 nm, and their growth process was consistent with the La-Mer growth mechanism. Furthermore, MSNs were modified with amino acids and loaded with rosmarinic acid (RosA), accompanied by the “outer packaging” of pectin to obtain RosA@MSNs-Pec. RosA was added at 200, 150, and 100 mg within 72 h, with a drug loading of 193, 175.5, and 156 mg/g and an encapsulation rate of 19.3, 17.6, and 15.6, respectively. Interestingly, RosA@MSNs-Pec demonstrated the dual pH and pectinase response property, and its release curve conformed to the Higuchi model, indicating that its drug-controlled release was based on Fick’s law.

Plant-based delivery systems for controlled release of hydrophilic and hydrophobic active ingredients: Pea protein-alginate bigel beads

Qianzhu Lin,Xiaojing Li,David Julian McClements,Zhengyu Jin,Chao Qiu,Guanghua Li

Oral delivery systems for nutrients and nutraceuticals are required for personalized nutrition applications. In some applications, these delivery systems need to encapsulate both hydrophilic and hydrophobic bioactive agents. In this study, bigel beads (2 mm) were prepared using pea protein (PP) and sodium alginate (SA) to form the hydrogel phase, and glyceryl monostearate and soybean oil to form the oleogel phase. The hydrogel phase was crosslinked by using Ca 2+ ions to form electrostatic bridges between the alginate molecules. The bigel beads had an oil-in-water type structure consisting of an oleogel dispersed phase within a hydrogel continuous phase. These beads were used to encapsulate and control the release of model hydrophobic (curcumin) and hydrophilic (epigallocatechin gallate) nutraceuticals. The textural and microstructural characteristics of the bigel beads were evaluated by dynamic shear rheology, textural profile analysis, cryo-electron microscopy, and fluorescence confocal microscopy. As the concentration of SA increasing from 0.2 to 1.0%, the hardness of bigel beads increased from 306.75 g to 792.40 g. In vitro digestion experiments showed that most of the nutraceuticals were released in the small intestine, rather than the stomach. Nutraceutical release could be controlled by manipulating bead composition, with the extent of nutraceutical release decreasing with increasing alginate concentration. The bigel beads developed in this study could be used as easy-to-swallow carriers for the controlled release of hydrophilic and hydrophobic bioactive ingredients. These kinds of products may be especially useful for people with dysphagia.

Multifunctional nanocoatings with synergistic controlled release of zinc ions and cytokines for precise modulation of vascular intimal reconstruction

Jianying Tan,Huanran Wang,Sainan Liu,Li Li,Hengquan Liu,Tao Liu,Junying Chen

Vascular stent implantation remains the major therapeutic method for cardiovascular diseases currently. We here introduced crucial biological functional biological function factors (SDF-1α, VEGF) and vital metal ions (Zn 2+ ) into the stent surface to explore their synergistic effect in the microenvironment. The combination of the different factors is known to effectively regulate cellular inflammatory response and selectively regulate cell biological behavior. Meanwhile, in the implemented method, VEGF and Zn 2+ were loaded into heparin and poly- l -lysine (Hep-PLL) nanoparticles, ensuring a controlled release of functional molecules with a multi-factor synergistic effect and excellent biological functions in vitro and in vivo . Notably, after 150 days of implantation of the modified stent in rabbits, a thin and smooth new intima was obtained. This study offers a new idea for constructing a modified surface microenvironment and promoting tissue repair.

The mechanism of activity-controlled release of 2,2′,4,4′-tetrabromodiphenyl degradation by polystyrene microcapsulated nanoscale zero-valent iron

Junyao Yang,Kuang Wang,Chengjie Xue,Zhanqiang Fang

Nano zero-valent iron (nZVI) is widely used for polychlorinated biphenyl (PBDE) remediation due to its cost-effectiveness and strong reduction capacity. However, its practical application is limited by poor stability, mobility, and antioxidant performance, as well as high reactivity that leads to side reactions and activity loss. To overcome these challenges, a poly(styrene)-encapsulated nZVI (PS-nZVI) core-shell structure was developed using dispersion polymerization. The resulting 10–15 nm polystyrene coating improved antioxidation, stability, mobility, and pH resistance (pH 5–9) compared to conventional nZVI. Incorporating a controlled release agent (20% acetone) at the start and midpoint of the reaction enhanced BDE-47 removal efficiency by 2.64 and 2.02 times, respectively, achieving nearly 100% activity release. Mechanistic studies revealed that the release agent promotes microcapsule corrosion and improves nZVI-BDE-47 interaction, enhancing both removal and debromination efficiency. PS-nZVI thus mitigates activity loss during storage and transport while enabling controlled reactivity for effective remediation.

Zein/sodium abietate complex stabilized pickering emulsion as a delivery system for controlled release of hydrophobic photosensitive pesticide

Jiayuan Tian,Jiyun Xie,Li Hao,Huayao Chen,Xinhua Zhou,Hongjun Zhou

Inefficient use of pesticides and the consequent environmental problems have led to the development of low-cost, environment-friendly and efficient pesticide delivery systems. Pickering emulsions (PK) stabilized by zein are considered to have many application prospects in the delivery of pesticides. Herein, the function of zein was improved by non-covalent binding with sodium abietate (RA-Na). Tea tree oil (TTO) was used as the solvent for hydrophobic photosensitive pesticide, avermectin (AVM), and also as a component of the oil phase of emulsion. The emulsion AVM@PK was oil in water (O/W) type that had certain foliar affinity. Under the same conditions of UV irradiation, the AVM residual rate of AVM@PK system was 2.4 times that of commercial emulsion (EC), showing superior UV resistance. AVM@PK released AVM at a faster rate under acidic/ basic conditions in vitro than under neutral conditions, with a certain pH response. In vitro bioactivity tests showed that AVM@PK had good antibacterial activity and insecticidal activity equivalent to EC. This system is expected to be used to prevent some plant pests while also preventing or treating some bacterial diseases. It is a simple, environment-friendly, low-cost, effective, and practical strategy for pesticide delivery.

Preparation of gelatin/sodium carboxymethyl cellulose-based probiotic controlled-release tablets via complex coacervation and the double emulsification method

Yu Wang,Yuhe Dong,Bingmin Wu,Yao Liu,Zhenzi Yun,Xi Yu,Quancai Sun,Ying Xiao,Tian Zhong

To increase the survival and controlled-release properties of probiotics during storage and gastrointestinal digestion, this study develops a novel probiotic intestinal-targeted controlled-release tablet using a gelatin/sodium carboxymethyl cellulose (CMC)-based complex coacervation and double emulsification method integrated with spray drying and direct compression technology. The effects of three different CMCs on the complex coacervation process, as well as the microstructure, physicochemical characteristics, gastrointestinal controlled-release behavior, and storage stability of the resulting microcapsule tablets, were systematically evaluated. The results demonstrated that the complex coacervates formed from medium-viscosity CMC and gelatin exhibited the highest probiotic encapsulation efficiency (EE) (87.27%) in the double emulsion system. Following spray drying, the microcapsule powders exhibited favorable physicochemical properties. Second, the microcapsule tablets formulated with medium-viscosity CMCs provided the most effective protection for probiotics, with a survival rate of 48.74%, which was significantly higher than that of the unencapsulated tablet group (21.09%, p < 0.05). These advantages can be attributed to the moderate molecular chain length and steric hindrance exhibited by medium-viscosity CMC, which enable the coacervate to achieve sufficient emulsification performance and protective capability. In simulated gastrointestinal digestion experiments, the survival rate of probiotics in the microcapsule tablets at the end of intestinal digestion ranged from 75.41% to 85.68%, which was also markedly higher than that of the unencapsulated group (11.46%, p < 0.05). Furthermore, after storage at 37°C for six months, the viable bacterial count in the microcapsule tablets decreased by only 0.9×10 9 to 1.57×10 9 CFU/tablet. This study systematically investigated the impact of CMC viscosity on the encapsulation and controlled-release performance of a gelatin/CMC (G/CMC) complex coacervation system, leading to the successful development of a probiotic controlled-release tablet with high survival rates, enhanced stability, and targeted delivery.

High bio-content polyurethane coated controlled-release fertilizer with metal coordination layer for prolonged nutrient release and enhanced degradability

Shuqi Dong,Yan Gao,Jungang Yang,Guoyuan Zou,Wentian He,Liang Yuan,Bing Cao

Controlled-release fertilizers (CRFs) offer significant potential for reducing environmental pollution and enhancing fertilizer utilization efficiency. However, achieving improved controlled-release and degradability in high bio-content polyurethane (HB-PU) coated fertilizers remains challenging, limiting their further agricultural application. In this work, HB-PU coatings were synthesized via a one-pot process primarily using castor oil (CO), lysine diisocyanate (LDI), and polymethylene polyphenyl polyisocyanate (PAPI). Results demonstrate that the enhanced controlled-release performance of the HB-PU coating is associated with the type and concentration of metal ions within the complexation layer formed on the surface of the urea. After constructing the Cu coordination layer, the nutrient-controlled release period of castor oil-based polyurethane with bio-content over 70 % (PCU1-Cu) coated urea with 2.5 % coating percentage was extended by 20 days compared to the control. The chemical structure, thermodynamic stability, and microscopic morphology of the HB-PU with Cu coordination layer were analyzed. XPS confirms that the metal ion coordination was mainly enriched in the inner layer of the coating material. The Fenton-like reaction confirmed the presence of Cu ions in the PCU1-Cu coating and with catalytic reactivity. Degradation studies in NaOH solution demonstrated that introducing Cu ions accelerates the HB-PU degradation. The application of PCU1-Cu coated urea on Pakchoi ( Brassica rapa ssp. chinensis ) exhibited increased yield. This study prepared HB-PU controlled-release fertilizers with excellent controlled-release properties by constructing a metal coordination layer, while also providing a novel design method for supplementing essential trace metal elements for plant growth.

Dual–responsive microgel beads prepared from pectin–stabilized emulsions for the controlled release of synbiotics to ameliorate ulcerative colitis

Xian He,Wenxin Dai,Shaoxian Liang,Yunyun Qin,Haoyue Liu,Kang Cheng,Mingyu He,Wanshui Yang,Xinsheng Qin

The benefits of synbiotics in ulcerative colitis are now supported by a growing amount of evidence. However, achieving its targeted delivery in vivo and reaching effective doses for therapeutic effects remains to be explored. To further explore the potential role of synbiotics in alleviating ulcerative colitis, this study designed a dual-response “egg–box” shaped microgel microsphere for targeted delivery and precise release of synbiotics. The internal “egg” structure was constructed using an apple pectin–zein hybrid particles (AZHPs) stabilized W 1 /O/W 2 double emulsions system, while a dense network formed by poly–L–lysine (PLL) and calcium alginate constituted the protective “box” shell. We investigated the physicochemical characteristics of W 1 /O/W 2 double emulsions prepared with different concentrations of AZHPs. The synbiotics–loaded W 1 /O/W 2 double emulsions were coated with PLL and calcium alginate to prepare microgel beads. In vitro digestion results demonstrated that the survival rate of probiotics within the microgel beads was significantly higher than that in a free state. Oral microgel beads alleviate colitis by modulating tight junction proteins (ZO–1, Claudin 1 and Occludin), reducing inflammatory factors (IL–1β, IL–6 and TNF–α), promoting the production of short–chain fatty acids. After treatment with EPMB, the number of Firmicutes in the intestinal flora increased while the number of Bacteroidetes decreased, and synbiotics intervention successfully restored the abundance of gut microbiota. This indicated that the loaded synbiotics “egg–box” shaped microgel beads offer a new option for the current treatment of colitis.

Insights into ultrasound-modified soy protein isolate-inulin hydrogels: Focus on binding mechanisms, stability, and riboflavin controlled-release behavior

Sai Yang,Ziteng Lian,Yifei Chi,Yaqi Tang,Mengmeng Wang,Jiexu Zhang,Xiaohong Tong,Huan Wang

The loose structure of natural soy protein isolate (SPI) gels limits their application in food nutrient delivery systems. This study investigated the effects of varying inulin (INU) concentrations combined with ultrasound treatment on the structural characteristics, stability, and riboflavin digestion and release of SPI-based hydrogels. Increasing INU concentration enhanced the textural properties and water-holding capacity (WHC) of the composite gels, converting free water into immobilized water. The addition of 2 % INU with ultrasound treatment (SU-2) markedly improved rheological properties, yielding a smoother, denser microstructure and increasing protein disorder. Molecular docking and molecular dynamics simulations indicated that the SPI-INU gel structure was stabilized primarily through hydrogen bonding and hydrophobic interactions. The modified gels improved riboflavin loading and stability, with SU-2 exhibiting the highest bioaccessibility (85.82 %) and chemical stability (76.01 %). This study contributes to the development of natural protein/polysaccharide gel carriers for targeted slow release of hydrophilic bioactives.
