Multiwalled carbon nanotubes noncovalently functionalized by electro-active amphiphilic copolymer micelles for selective dopamine detection

Xiaoma Fei,Jing Luo,Ren Liu,Jingcheng Liu,Xiaoya Liu,Mingqing Chen

We have synthesized an electro-active amphiphilic copolymer with carbazole side chains via free radical polymerization using 7-(4-vinylbenzyloxy)-4-methyl coumarin and 9-(4-vinylbenzyl)-9H-carbazole as the monomers. The copolymer can self-assemble to form micelles (termed EACMs) in aqueous solution and can adsorb onto the surfaces of MWCNTs via π–π interactions and thereby cause the efficient dispersion of the MWCNTs in aqueous solution. The coumarin groups in the copolymer undergo UV-induced photo-crosslinking, which further improves the stability of the suspension. Moreover, the electro-active carbazole moieties in the EACMs can undergo electropolymerization to form a conducting network on the MWCNTs that significantly accelerates electron transfer. The EACM/MWCNTs hybrid was applied to the amperometric sensing of dopamine (DA) as a model analyte. After electropolymerization, the electrode exhibited good sensitivity and selectivity toward the determination of dopamine with a 0.2 μM detection limit and a wide linear range. The method described here provides a viable route to water-dispersible and stable carbon nanotubes while preserving their outstanding electrical properties. We presume that the composite described here represents a valuable tool for the construction of electrochemical sensors and electronics.

Changeable wettability of electrospun membrane by adjusting self-assembly micelles structure of amphiphilic block copolymer

Lingxiao Wang,Xinyue Liang,Guilong Xu,Jian Hu

Fiber membranes with high specific surface area can be produced when using electrospinning technology combined with amphiphilic block polymers. Due to the solubility difference between the two segments of amphiphilic block polymers, it is easy to be self-assembly in solvents, which affects the morphology of the spinning fiber and the surface properties of the membrane. In this work, the relationship between the self-assembly of the amphiphilic block polymer and the morphology of the spinning fiber and the wetting performance of the spinning membrane was explored. Membrane is produced by electrospinning amphiphilic block copolymer PS-b-PHEMA (polystyrene-block-poly(2-hydroxyethyl methacrylate) by using co-solvent system. PS-b-PHEMA with controlled molecular weight (Mw) and polymer dispersity index (PDI) was synthesized via atom transfer radical polymerization (ATRP) and characterized by nuclear magnetic resonance (NMR) and gel permeation chromatography (GPC). The electrospun membrane have both super-hydrophobic and super-hydrophilic by adjusting the ratio of mixed solvents, demonstrating that the wetting performance of the fiber membrane mainly depends on the shell structure of the self-assembly micelles.

Construction of curcumin-loaded micelles and evaluation of the anti-tumor effect based on angiogenesis

Liu Rui,Liu Zhongyan,Guo Xueli,Kebebe Dereje,Pi Jiaxin,Guo Pan

Objective: Inhibition of tumor angiogenesis has become a new targeted tumor therapy. In this study, we established a micellar carrier with a tumor neovascularization-targeting effect modified by the neovascularization-targeting peptide NGR. Methods: The targeted polymer poly(ethylene glycol)-b-poly(lactide-co-glycolide) (PEG-PLGA) modified with Asn–Gly–Arg (NGR) peptide was prepared and characterized by 1 H nuclear magnetic resonance and Fourier-transform infrared spectrometry. NGR-PEG-PLGA was used to construct curcumin (Cur)-loaded micelles by the solvent evaporation method. The physicochemical properties of the micelles were also investigated. Additionally, we evaluated the antitumor efficacy of the polymer micelles (PM) using in vitro cytology experiments and in vivo animal studies. Results: The particle size of Cur-NGR-PM was 139.70 ± 2.51 nm, and the drug-loading capacity was 14.37 ± 0.06%. In vitro cytological evaluation showed that NGR-modified micelles showed higher cellular uptake through receptor-mediated endocytosis pathways than did unmodified micelles, leading to the apoptosis of tumor cells. Then, in vivo antitumor experiments showed that the modified micelles significantly inhibited tumor growth and were safe. Conclusions: NGR-modified micelles significantly optimized the therapeutic efficacy of Cur. This strategy offers a viable avenue for cancer treatment. Export

Self-assembled polymeric micelles based on THP and THF linkage for pH-responsive drug delivery

Fangxia Zhu,Qinglai Yang,Yuan Zhuang,Yuanqing Zhang,Zhifeng Shao,Bing Gong,Yu-Mei Shen

Developing smart nanocarriers for drug delivery system is advantageous for many kinds of successful biomedicinal therapy. In this study, we designed an amphiphilic block copolymers containing pH-responsive tetrahydropyran (THP) and tetrahydrofuran (THF) linkage. Their structures were confirmed by 1 H NMR and gel permeation chromatography (GPC). The release rate of encapsulated drugs depends upon the pH value and pH sensitive linkage in the backbone of copolymers . For PLA–THP–PEG micelles the cumulative release amount of doxorubicin (DOX) was 62% at pH 5.0, which is about four times higher than that at pH 7.4. Under the same conditions the release rate for PLA–THF–PEG micelles is a little faster than that of the PLA–THP–PEG micelles. Cellular uptake study demonstrates that DOX-loaded micelles can easily enter the cells and produce the desired pharmacological action and minimizing the side effect of free DOX. These findings indicate that THP and THF linked diblock copolymer micelles is a promising candidate for drug carrier.

Fluorinated reversed micelles by polymerization-induced self-assembly with main-chain-type semifluorinated alternating copolymer

Jinying Wang,Jiannan Cheng,Kai Tu,Yuxue Wang,Qing Yu,Lifen Zhang,Zhenping Cheng

In this work, based on the semifluorinated macro-RAFT (reversible addition-fragmentation chain transfer) agent and polymerization-induced self-assembly (PISA) strategy, we established a mild and facile method to prepare reversed micelles up to 40 wt% of solid content with main-chain-type semifluorinated alternating copolymer as shell and hydrophilic polymer as core for the first time. The fluorinated macro-RAFT agent was synthesized by combination of step transfer-addition & radical-termination (START) polymerization and post-modification method. The reversed micelles were generated by the PISA of N,N -dimethacrylamide (DMA) in the presence of the fluorinated macro-RAFT agent in the non-polar solvent toluene at 70 °C, and fluorinated reversed micelles can be fixed in the presence of crosslinker ethyleneglycol dimethacrylate (EGDMA). This strategy will facilitate to open a new way in designing novel functional synthetic fluorinated nanomaterials.

The characterisation, pharmacokinetic and tissue distribution studies of TPGS modified myricetrin mixed micelles in rats

Chunmei Wei,Qilong Wang,Wen Weng,Qiuyu Wei,Yujiao Xie,Michael Adu-Frimpong,Elmurat Toreniyazov,Hao Ji,Ximing Xu,Jiangnan Yu

This study was designed to investigate the bioavailability and targeting of myricetrin-loaded ternary micelles modified with and without TPGS. The particle diameters of myricetrin-loaded micelles and myricetrin-loaded-TPGS micelle were 30.93 ± 1.34 nm and 26.42 ± 0.89 nm, respectively, while their respective encapsulation efficiencies (m/m, %) were 83.3 ± 1.08 and 93.8 ± 1.18. The release rate of myricetrin in the micellar system clearly exceeded the free myricetrin in the three media (pH 6.8 phosphate buffer, pH 1.2 HCl solution and double distilled water). In vivo studies displayed that the bioavailability of myricetrin mixed micelles was remarkably improved than the free drug after oral administration. Moreover, the results of tissue distribution showed that myricetrin-loaded-TPGS micelles accumulated well in the liver tissue. Based on these results, it was speculated that myricetrin-loaded-TPGS micelles might act as a promising carrier for liver targeting with improved hepatic concentration of myricetrin compared with the myricetrin-loaded micelles.

MMP-Responsive TAT-Containing Polymeric Micelles for Cascade Delivery of Oxaliplatin in Gastric Cancer Therapy

Zou Jiaxin,Li Yanan,Kou Zhaojun,Wang Dayang,Lu Linlin,Cong Hailin,Yu Bing,Ma Xuezhen

Although nanodrug delivery systems have advanced cancer therapy, limited tumor penetration and inefficient intracellular transport still restrict their therapeutic efficacy. Herein, we constructed a matrix metalloproteinase (MMP)-responsive, TAT-containing polymeric micelle, O@MTPP, for cascade delivery of oxaliplatin in gastric cancer. O@MTPP integrates an MMP-2/9-cleavable peptide, MTP (GPLGIAG), with the cell-penetrating and nuclear-localization-associated TAT peptide (CYGRKKRRQRRR). In the tumor microenvironment, MMP-2/9-mediated cleavage triggers charge conversion, enhancing tumor-cell internalization and penetration-associated distribution. After cellular uptake, TAT further promotes nuclear-associated accumulation of the nanocarrier-associated payload, supporting improved intracellular delivery of oxaliplatin. This cascade delivery strategy enhanced tumor accumulation, strengthened antitumor activity, and induced immunogenic cell death (ICD)-associated immune responses, including calreticulin exposure, high mobility group box 1 (HMGB-1) expression, increased tumor necrosis factor-α (TNF-α), interferon-γ (IFN-γ), and interleukin-12 (IL-12) levels, and decreased interleukin-6 (IL-6). Overall, O@MTPP provides an enzyme-activated cascade delivery strategy for overcoming tissue-level and intracellular barriers in oxaliplatin-based gastric cancer therapy.

pH/ROS Dual-Responsive Zwitterionic Polymer Micelles with Enhanced Mucus Penetration for Bacterial Pneumonia Therapy

Yang Xue,Huang Menglin,Wu Hongjing,Li Wen,Chen Hongqi,Dai Xiaomei,Gao Feng

Bacterial pneumonia, a prevalent respiratory disease with high global morbidity and mortality rates, remains a formidable therapeutic challenge. Herein, pH/reactive oxygen species (ROS) dual-responsive zwitterionic polymer (PODEA- b -P(ROS-CA-EG)n) micelles are developed for bacterial pneumonia therapy. The zwitterionic shell can facilitate the micelles to effectively penetrate through the mucus. These micelles exhibit pH-responsive charge reversal performance, which is conducive to the cellular internalization and bacterial killing. Meanwhile, the micelles respond to the acid and overexpressed ROS microenvironment at the lesion site to release cinnamaldehyde (CA), enhance ROS generation, and activate an immune reaction to eliminate extracellular and intracellular bacteria collaboratively. In bacterial pneumonia, PODEA- b -P(ROS-CA-EG)n micelles can effectively eliminate bacteria and alleviate lung inflammation. These pH/ROS dual-responsive zwitterionic polymer micelles could provide a synergistic therapeutic strategy against bacterial pneumonia.

Chitosan composite packaging film loaded with luteolin-superoxide dismutase nano-micelles: Enhancing food preservation performance

Pei Wu,Yilin Che,Rui Zhou,Yixin Tang,Lei Guo,Baiqiao Chen,Xinran Yan,Shanli Wu,Chang Liu,Mujie Kan,Jizhou Zhang,Xi Yang,Xin Jiang,Zhanxin Wang,Erlei Wang,Tianhao Yan,Caina Xu

Novel chitosan-based composite film with Luteolin-SOD nano-micelles via nano-encapsulation for food preservation. • This composite film outperforms conventional packaging: superior inhibition of oxidation, delays spoilage, biodegradable. • Natural-ingredient film: promising substitute for traditional packaging, meeting eco-friendly and sustainable demands. Chitosan-based films are promising alternatives to petroleum-based food packaging materials but suffer from insufficient mechanical strength, poor water resistance, and limited antioxidant capacity. To address these issues, we synthesized chitosan-based nano-polymers loaded with luteolin (Lut) and superoxide dismutase (SOD) (CLS nano-micelles). We then incorporated these nano-micelles into a chitosan-based film to produce a CLS film. The results showed that, compared with the pure chitosan film, the mechanical properties of CLS film significantly improved, with tensile strength increasing from 6.14 MPa to 37.50 MPa and elongation at break increasing from 17.4% to 34.6%. The barrier properties of the CLS film were also enhanced, as both water vapor permeability and solubility decreased. Notably, the CLS film also exhibited excellent antioxidant properties (with DPPH inhibition rates increasing from 4.4% to 48.3% and ABTS inhibition rates rising from 7.2% to 74.6%) and demonstrated potent antibacterial activity against E. coli , S. aureus , and B. subtilis . It also exhibited excellent biocompatibility and demonstrated favorable biodegradability in soil. Grape preservation assays further confirmed its efficacy in maintaining fruit freshness. These results demonstrate that integrating CLS nano-micelles into a chitosan matrix significantly improves its mechanical, barrier, antioxidant, and antibacterial properties. This makes the CLS film a promising eco-friendly alternative for sustainable food packaging applications, offering an efficient and environmentally friendly solution. Self-Assembly of Alginate-Grafted Penicillanic Acid Derivatives into Antibacterial Micelles for Controlled Drug Delivery

Xiuqiong Chen,Jiayi Liu,Jiji Fan,Yanan Bu,Qian Wang,Huiqiong Yan,Changjiang Yu,Qiang Lin

The self-assembly and drug release of amphiphilic polymers are critically controlled by synergistic molecular forces and polymer architecture. Therefore, elucidating the self-assembly behavior of amphiphilic polymers, based on the entropy effect mediated by water molecules, is of significant importance for designing functional nanomicellar carriers. To advance the design of functional nanomicellar carriers, this work innovatively exploited the dual functionality of 6-aminopenicillanic acid (6-APA), a key semisynthetic penicillin intermediate, by grafting it onto alginate via acylation, creating an amphiphilic alginate-grafted penicillanic acid derivative (AM-Alg-APA). This molecular design uniquely integrates inherent antibacterial activity with tunable self-assembly driven by entropy-mediated hydrophobic effects in aqueous systems. The synthesized AM-Alg-APA was able to autonomously form spherical micelles, displaying a hydrodynamic diameter of 568.69 nm with a polydispersity index (PDI) of 0.28 and a zeta potential of −34.8 mV in aqueous solution. This system further demonstrated responsive colloidal behavior to variations in the pH and ionic strength under simulated physiological conditions. Critically, micellar morphologies arose from synergistic intermolecular forces and segmental entropy optimization, enabling efficient encapsulation of hydrophobic triclosan (TCA) via high-shear processing and its controlled release via non-Fickian diffusion. Beyond demonstrating cytocompatibility with MC3T3-E1 cells, the micelles leveraged the grafted 6-APA to confer significant antibacterial activity against S. aureus and E. coli. The multifunctionality, including entropy-regulated self-assembly, stimuli-responsive drug delivery, intrinsic antimicrobial action, and biocompatibility, establishes AM-Alg-APA micelles as innovative platforms for wound-targeted drug delivery in advanced medical dressings.

Coacervates to wormlike micelles transition enables lower-temperature-triggered pesticide release for nocturnal pest management

Keming Zhou,Yuchun Han,Chengyun Li,Ming Yang,Jie Wang,Yilin Wang

Temperature-triggered coacervates to micelles transition is realized. • Coacervates possess high encapsulation capacity for hydrophobic pesticides. • Disintegrated coacervates boost pesticide release against nocturnal pests. • Assemblies exhibit excellent spreading and deposition on leave. • Calorimetry provides thermodynamic insights into aggregate transition. To enhance pesticide release efficiency at lower temperature while keeping pesticide encapsulated at higher temperature would largely improve pesticide utilization against Noctuidae pests active at night. However, such a kind of carrier has been rarely reported. Herein, a temperature-induced reversible aggregate transformation is adopted to fabricate temperature-responsive pesticide carrier by cationic surfactant N -dodecyl- N, N -2-dihydroxyethyl- N -methyl ammonium bromide (DDHAB) and 8-hydroxypyrene-1, 3, 6-trisulfonic acid trisodium salt (HPTS). At higher temperatures (> 25 °C), the HPTS/DDHAB complexes associate into coacervates with high encapsulation efficiency for hydrophobic pesticide, Emamectin Benzoate. At lower temperatures (≤ 25 °C), the coacervates transform into wormlike micelles and facilitate rapid pesticide release. Thus, the system shows an advantage of killing Noctuidae pests with high activity at nights. The median lethal concentration (LC 50 ) at 25 °C toward two kinds of pests ( Helicoverpa armigera and Spodoptera exigua ) are reduced by 42.4% and 45.9% relative to those at 35 °C. The HPTS/DDHAB solution droplets exhibit superior spreading and deposition behavior on both hydrophilic and hydrophobic leaves. Furthermore, DDHAB achieves better biocompatibility after binding with HPTS. This study establishes a method to realize intelligent temperature-response pesticide release with application potential for Noctuidae pest control. pH-Responsive Biomimetic Polymeric Micelles as Lymph Node-Targeting Vaccines for Enhanced Antitumor Immune Responses

Xiqin Yang,Tong Yu,Yingping Zeng,Keke Lian,Xueqing Zhou,Jia Ke,Yinghong Li,Hong Yuan,Fuqiang Hu

Lymph nodes are proposed as the intriguing target in cancer immunotherapy, and cellular immunity is vital for vaccines to fight against cancer. However, inefficient delivery of vaccines to lymph nodes and deficient lysosomal escape of antigens result in weak cellular immunity, which restrains the strength of vaccines in inducing antitumor immune responses. Hence, dendritic cell membrane (DCM)/histidine-modified stearic acid-grafted chitosan (HCtSA)/ovalbumin (OVA) micelles, as pH-responsive biomimetic vaccines, were fabricated to target lymph nodes and induce cellular immunity for enhanced antitumor immune responses. DCM/HCtSA/OVA micelles exhibited pH-dependent antigen release behavior, which resulted in efficient escape of antigens from dendritic cell (DC) lysosomes. Besides, DCM/HCtSA/OVA micelles accumulated and reserved in the lymph nodes, which ensured effective uptake by DCs. Importantly, DCM/HCtSA/OVA micelles induced potent T cell immune responses, promoted secretion of antitumor-related cytokines, and notably inhibited tumor growth. Overall, DCM/HCtSA/OVA micelles exhibit great potential in targeted immunotherapy and can provide guidance for the design of vaccines.
