pH- and NIR light responsive nanocarriers for combination treatment of chemotherapy and photodynamic therapy

Sheng Wang,Weitao Yang,Jing Cui,Xue Li,Yan Dou,Lin Su,Jin Chang,Hanjie Wang,Xiaodong Li,Bingbo Zhang

The side effects of antitumor drugs and low treatment efficacy are two major challenges of current chemotherapy. To address these issues, we developed a new kind of smart nanocarriers that combine pH-responsive chemotherapy and near-infrared (NIR) light triggered photodynamic therapy. These nanocarriers were based on upconversion nanoparticle (UCN)-loaded folate-conjugated polymeric lipid vesicles (UFPLVs). Merocyanine 540 (MC540), as a photosensitizer, was loaded in the UFPLVs; doxorubicin hydrochloride (DOX), as an antitumor drug, was conjugated to the surfaces of UFPLVs by pH-sensitive hydrazone bonds. The newly developed MC540&DOX–UFPLVs had a nanosized structure with targeting ligand modification, so they had the potential to accumulate into tumor sites via a combination of passive and active targeting effects. An in vitro singlet oxygen test showed that the nanocarriers can generate cytotoxic singlet oxygen successfully under the irradiation of NIR light. In vitro DOX release profiles demonstrated that the nanocarriers can achieve a pH-triggered drug release. It has been demonstrated by a cellular uptake study that the nanocarriers can efficiently deliver drugs and photosensitizers into tumor cells. In vitro and in vivo combination treatments evidenced the high antitumor effects of MC540&DOX–UFPLVs under NIR light irradiation. These results suggest that the MC540&DOX–UFPLVs may be promising nanocarriers for tumor combination therapy applications.

Enhanced-permeability delivery system for hydroxyl radical-responsive NIR-II fluorescence-monitored thrombolytic therapy

Nan Lv,Shaodong Zhai,Jun Xiong,Nan Hu,Xiang Guo,Zhida Liu,Ruiping Zhang

Pathological thrombus can cause serious acute diseases that present a significant threat to human health, such as myocardial infarction and stroke. Challenges remain in achieving effective thrombolysis and real-time monitoring of therapeutic effects while minimizing side effects. Herein,a multifunctional nanoplatform (TG-OPDEA@UK/MnO 2 -H1080) with enhanced thrombus-permeability was developed to monitor the therapeutic effect of antioxidant-thrombolysis by hydroxyl radical-responsive NIR-II fluorescence imaging. The polyzwitterion poly (oxidized N,N-Diethylaminoethyl methacrylate- co -n-butyl methacrylate) (OPDEA) was prepared as the matrix of nanoparticles to simultaneously loading urokinase (UK) and MnO 2 QDs, as well as NIR-II fluorescent molecule, H-1080. Subsequently, the fibrin targeted peptide CREKA was modified on the surface of the nanoparticles. OPDEA exhibits efficient loading capacity while endowing nanoparticles with the ability to effectively increased penetration depth of UK by 94.1 % into the thrombus, for extensive thrombolysis and fluorescence monitoring. The loaded UK exhibited good thrombolytic effect and greatly reduced the risk of bleeding by 82.6 %. TG-OPDEA@UK/MnO 2 -H1080 showed good thrombolytic efficacy and specific thrombus monitoring in the mouse carotid artery thrombosis model induced by ferric chloride (FeCl 3 ). This work prepares a nanoplatform for thrombolytic therapy and real-time efficacy assessment based on an independent externally forced thrombus penetration delivery strategy.

De Novo siRNA nanoplatform with NIR-II imaging for treatment of carotid atherosclerotic plaques

Jialu Gao,Hua Zhu,Huanhuan Cai,Qihang Ding,Xiaoxing Xiong,Tong Zhang,Bo Chen,Wumei Wang,Xinpeng Ju,Jianying Huang,Xiaodong Zeng,Jong Seung Kim,Xuechuan Hong

Reducing foam cell formation and accumulation within plaques is a critical strategy for treating atherosclerosis. Although current lipid-lowering therapies have significantly improved the management of atherosclerosis, the risk of cardiovascular events remains. Thus, it is necessary to explore next-generation therapies to reduce foam cell formation, inflammation, and oxidative stress. To address these challenges, we present an innovative near-infrared II (NIR-II) fluorescent dye coupled with a lesion-targeting peptide, forming a molecular self-assembly nanoplatform for effective in vitro small interfering RNA (siRNA) delivery. This platform downregulates Epsin1/2, a family of ubiquitin-binding endocytic adaptors, upregulates the expressions of ABCG1 and LRP-1 proteins, and markedly reduces foam cell formation. Additionally, this system enables real-time tracking of plaque accumulation, therapeutic response, and efficacy through in vivo noninvasive NIR-II imaging. Systemic siRNA delivery significantly suppresses lipid accumulation, plaque size, and necrotic core area in the carotid plaque model of ApoE −/− mice. These results suggest that this multi-functional nanoplatform could become an effective tool for NIR-II image-guided siRNA delivery for personalized treatment of atherosclerosis.

Extending visual range of bacteria with upconversion nanoparticles and constructing NIR-responsive bio-microrobots

Wei Xu,Zhen Liu,Jing Wang,Kai Jin,Lulu Yue,Lin Yu,Luqi Niu,Qingqing Dou,Jinliang Liu,Yuzhe Zhang,Xiaohui Zhu,Yihan Wu

The motility of bacteria is crucial for navigating competitive environments and is closely linked to physiological activities essential for their survival, such as biofilm development. Precise regulation of bacterial motility enhances our understanding of these complex processes. While optogenetic tools have been used to control and investigate bacterial motility, the excitation light in most existing systems are limited to the visible light spectrum. Here, we introduce a new type of bio-microrobot comprising genetically engineered E. coli cells and orthogonally emissive upconversion nanoparticles that can respond to both 980 nm and 808 nm NIR light. This system allows toggling of bacterial states between tumbling and swimming via simply alternating the NIR light between different wavelengths. It is believed that the use of NIR light with deeper tissue penetration suggests potential applications for these bio-microrobots in areas like targeted drug delivery.

Lipase-Entrapped Polydopamine (PDA) Nanobottles for Near-Infrared (NIR)-Triggered Pickering Interfacial Biocatalysis (PIB)

Xingxing Ding,Zhonglin He,Jin Mao,Qi Zhou,Qianchun Deng,Lei Zheng,Jie Shi

In this work, we presented an external stimulating Pickering interfacial biocatalyst (PIB), which is a gelatin and Candida rugosa lipase (CRL) complex containing polydopamine (PDA) nanobottles (PDA@G-CRL). This created PDA nanobottles are employed as emulsifiers and enzyme carriers for the PIB system, showing an excellent energy saving effect during the reaction. They are made utilizing the polystyrene (PS) template technique, and after lipase is entrapped with gelatin gel assistant, by exploiting the photothermal properties of PDA under near-infrared (NIR) irradiation, these nanobottles may efficiently stabilize the oil-in-water (O/W) emulsion and cause the release of gelatin–lipase complex, producing the PIB effect. The encapsulated lipase rate and minimum loading rate of lipase in PDA nanobottles are 50.76% and 263 mg g–1, respectively, which is 3.59 times higher than that of free lipase. In the biosynthesis phytosterol, the esterification rate of PIB reached 78% after 34 h of reaction, which was 3.4 times higher than that of free lipase in a single-phase system, and after 5 cycles, the relative activity remained at 51.33%. In particular, in terms of energy consumption, our proposed method is 227 times lower than that of traditional heating methods. It is believed that this smart PIB is able to contribute insights into green biocatalysis areas.

Controlled drug release and antibacterial enhancement via NIR-activated CuS-modified TiO2 nanotubes arrays

Qin Huang,Chunling Xie,Xiufeng Xiao

Two primary obstacles impede the longevity of implants: bacterial infection and fixation loosening. This research introduces an innovative near-infrared (NIR) light-activated drug delivery system tailored to enhance the antimicrobial efficacy and bone integration of titanium-based implants. Titanium dioxide nanotube arrays (TNTs) were modified with epoxy for covalent bonding of therapeutic agents, and copper sulfide nanoparticles (CuS NPs) were added for regulated drug release. Alendronate sodium (ALN), an osteoporosis medication, was successfully bonded to the nanotubes. In vitro experiments showed that the release rate of ALN from TNTs-ALN reached 94 % within 36 h. In contrast, minimal release was observed in TNTs-ECH-ALN-CuS, which effectively addresses the prevalent issue of abrupt drug discharge typical in conventional delivery systems. In the context of NIR light exposure, the TNTs-ECH-ALN-CuS exhibited a notable ALN release efficacy of 48 %, demonstrating effective control over drug delivery. Additionally, the generation of ROS, coupled with temperature elevation under NIR exposure, endowed the modified TNTs with exceptional antibacterial efficacy, exceeding 99 % inhibition against E.coli and S.aureus . We observed a pronounced enhancement in the drug delivery mechanism, characterized by a reduction in the initial burst release and a sustained, controlled release, coupled with robust antibacterial action and notable biological performance.

Design of donor–acceptor conjugated polymers based on diketopyrrolopyrrole for NIR-II multifunctional imaging

Mengyu Cao,Xuezhen Ma,Chang Wang,Wentao Zou,Fang Wang,Bing Yu,Hailin Cong,Youqing Shen

Diketopyrrolopyrrole (DPP) is an excellent photosensitizer and photothermal agent with the advantages of good planarity, strong electron affinity, high electron mobility, easy purification, easy structural modification and high molar absorption coefficient. It is regarded as one of the ideal choices for the design and synthesis of efficient organic photovoltaic materials. Therefore, two kinds of donor–acceptor (D–A) conjugated polymers were designed and synthesized with DPP as the acceptor, and their optical properties and applications in the near-infrared region were studied. The quantum yield (QY) of PBDT-DPP is 0.46%, and the highest temperature reached within 10 minutes after irradiation with a 660 nm laser is 60 °C. Another polymer, EDOT-DPP, has a QY of 0.48%, and its semiconductor polymer nanoparticle aqueous solution can reach 60 °C within 12 minutes under laser irradiation, achieving photothermal treatment of nude mice tumors. Both polymer NPs have good biocompatibility and promising applications in bioimaging and photothermal therapy.

Activatable NIR Fluorescence Probe for Epinephrine Detection and Bioimaging Based on Anionic Heptamethine Cyanine

Chen Luo,Youjia Chen,Jinxuan Gu,Huihui Cai,Huijin Lin,Zhigang Jin,Chaobiao Huang

Epinephrine (EP) is an essential catecholamine in the human body. Currently, most EP detection methods are not suitable for in vivo detection due to material limitations. An organic small molecule fluorescent probe based on a chemical cascade reaction for the detection of EP was designed. Anionic heptamethine cyanine dye was selected as a fluorescent dye because of its NIR fluorescence emission with excellent biocompatibility. The secondary amine of EP nucleophilically attacks the carbonate of the probe with its stronger nucleophilicity and further undergoes intramolecular nucleophilic cyclization to release the fluorophore. Other substances containing only primary amines or no β-OH lack reaction competitiveness due to their weaker nucleophilicity or inability to undergo further cyclization. The fluorescence recovery of the probe was linearly related to the EP concentration of 2–75 μmol/L. The detection limit was 0.4 μmol/L. The recovery rate was 94.78–111.32%. Finally, we successfully achieved bioimaging of EP in living cells and EP analogue in nematodes.

A smart capecitabine imprinted nanocontainer with the dual-responsive performance to pH/NIR light

Chensi Zeng,Yanfang Gong,Chenxi Qi,Xianghua Zeng,Ni Tan

Capecitabine (CAP) is a kind of prodrug that can be metabolized into 5-fluorouracil (5-FU) in vivo, so it is usually classified as the antimetabolite fluoropyrimidine deoxynucleoside analog. In this paper, CAP, upconversion nanoparticles (UCNPs), 3,5-dicarboxy-4-methacrylate-based azobenzene (MAPDIA)/acrylic acid-functionalized gelatin, N,N′-methylene bisacrylamide (MBA), and ammonium persulfate (APS) were used as template, light trigger, bifunctional monomers, cross-linking agent and initiator, respectively. By using the free-radical polymerization and the molecular imprinting technique, the potential drug delivery nanocontainer CAP-MIP with the dual-responsive performance to pH/near infrared light (NIR) was successfully prepared. Furthermore, the characterization of CAP-MIP was also carried out by the modern instrumental analysis such as FTIR, XRD, TGA, BET and so on. The stimulus-responsive release experiment in vitro indicated the maximum cumulative release rate of CAP changed from 1.71 % to 89.54 % when the pH value in medium varied from 1.2 to 8.3, and the equilibrium release time point also greatly prolonged to 420 min from 40 min, respectively. When irradiated by 980 nm monochromatic NIR light in the same solution of pH 6.8, the maximum release rate of the sample could reach 96.21 % at 180 min, which respectively corresponded to the data of 41.84 % at 420 min under the condition of the absence of additional NIR light. In addition, CAP-MIP also had the excellent specific adsorption for CAP with the selectivity factor of 2.904 and the maximum CAP-loaded capacity of 7 mg•g −1 . The above information denoted the target material should be of some potential in drug delivery system application.

Nanocellulose-mediated conductive hydrogels with NIR photoresponse and fatigue resistance for multifunctional wearable sensors

Chenyu Sang,Shaowei Wang,Xiaoyue Jin,Xiaoyu Cheng,Huining Xiao,Yiying Yue,Jingquan Han

The rapid development of hydrogels has garnered significant attention in health monitoring and human motion sensing. However, the synthesis of multifunctional conductive hydrogels with excellent strain/pressure sensing and photoresponsiveness remains a challenge. Herein, the conductive hydrogels (BPTP) with excellent mechanical properties, fatigue resistance and photoresponsive behavior composed of polyacrylamide (PAM) matrix, 2,2,6,6-tetramethylpiperidin-1-yloxy-oxidized cellulose nanofibers (TOCNs) reinforcement and polydopamine-modified black phosphorus (BP@PDA) photosensitizer are prepared through a facile free-radical polymerization approach. The PDA adhered to the BP surface by π-π stacking promotes the optical properties of BP while also preventing BP oxidation from water. Through hydrogen bonding interactions, TOCNs improve the homogeneous dispersion of BP@PDA nanosheets and the mechanical toughness of BPTP. Benefiting from the synergistic effect of PDA and TOCNs, the conductive BPTP integrates superior mechanical performances, excellent photoelectric response and photothermal conversion capability. The BPTP-based sensor with high cycling stability demonstrates superior strain sensitivity ( GF  = 6.0) and pressure sensing capability ( S  = 0.13 kPa −1 ) to monitor various human activities. Therefore, this work delivers an alternative construction strategy for generating high-performance conductive hydrogels as multifunctional wearable sensors.

NIR-activated endoplasmic reticulum stress amplifier to regulate phagocytosis for macrophage-mediated cancer immunotherapy

Yaru Zhang,Wenyu Xie,Jiamin Luo,Rui Gao,Zhimin Gao,Xinyi Wang,Yongxin Huang,Chen Lin,Meiling Tan,Zhiyao Hou

Effectively activating macrophages to engulf tumor cells has attracted intensive attention in cancer immunotherapy. However, tumor cells can upregulate anti-phagocytic signals (such as CD47 molecules) to bind with signal regulatory protein-α (SIRPα), ultimately providing a “don’t eat me” signal to evade phagocytosis. Blockading CD47 alone is insufficient to stimulate macrophages phagocytosis due to the absence of endogenous activating signals (“eat me” signal). Herein, a mesoporous silica (MSN) modified by Cu 9 S 5 nanoparticle with mimicking enzyme activities were synthesized and utilized to electrostatically load the anti-CD47 antibodies (aCD47), generating MCC nanozymes, to improve macrophage-mediated cancer immunotherapy. MCC nanozymes can target tumor cells by specifically binding to the cell-surface CD47, disabling the “don’t eat me” signal. Meanwhile, under near infrared light (1064 nm) stimulation, MCC nanozymes exhibit the enhanced glutathione oxidase-like and peroxidase-like enzymatic activities by the photothermal effect, which lead the reactive oxygen species (ROS) generation to induce endoplasmic reticulum (ER) stress amplification, resulting the exposure of calreticulin (CALR) for providing the endogenous “eat-me” signals to macrophages. In a 4T1 breast cancer model, even under mild photothermal condition, MCC nanozymes effectively inhibit tumor growth and metastasis. Collectively, this dual-regulatory strategy presents a promising approach for macrophage-mediated cancer immunotherapy.

Xue-Mei Dong,Chen Chen,Yin-Xiang Li,Hong-Chao Sun,Bin Liu,Zi-Fan Li,Kai-Li Wang,Zi-Xi He,Meng-Na Yu,Wei Huang,Ju-Qing Liu

Neuromorphic vision sensors capable of multispectral perception and efficient recognition are highly desirable for bioretina emulation, but their realization is challenging. Here, we present a cocrystal strategy for preparing an organic nanowire retinamorphic vision sensor with UV–vis–NIR perception and fast recognition. By leveraging molecular-scale donor–acceptor interpenetration and charge-transfer interfaces, the cocrystal nanowire device exhibits ultrawide photoperception ranging from 350 to 1050 nm, fast photoresponse of 150 ms, high specific detectivity of 8.2 × 1012 Jones, and responsivity of 15 A W–1, as well as retina-like photosynaptic plasticity behaviors. Utilizing the sensor nerve and convolutional neural network, the architecture achieves 90% accuracy in recognizing colorful images. The cocrystal design offers an effective method for constructing nanowire photosynases with high performance in artificial visual systems.
