A novel low-molecular-weight pectin from sand pear (Pyrus pyrifolia Nakai) pulp improves the quality of yogurt: a comparative study with fructooligosaccharide

Xiaoping Yang,Jing Fan,Chuanhui Fan,Jingguo Zhang,Wei Du,Xin Mei,Jianbin Shi,Hongmei Shi,Hongju Hu,Wei Liu,Qiliang Chen

Pectin was extracted from sand pear pulp with a yield of 0.51 ± 0.02 g/kg. • The molecular weight of pectin was 7733 g/mol with DE 71%, noted as LMW-pectin. • LMW-pectin changed the total acids and textural structure of yogurt. • LMW-pectin affected the odor and taste of the yogurt significantly. • LMW-pectin changed the rate of Bacillus via bacterial diversity analysis. In this study, low-molecular-weight pectin (LMW-pectin) was extracted from sand pear pulp, and its effect on the quality of yogurt was studied compared to the commercial fructooligosaccharide (FOS). The structural properties of LMW-pectin were relative molecular weight: 7733 ± 4 g/mol, galacturonic acid content: 732 ± 9 g/kg, and degree of esterification: 71 ± 2%. Compared with the control, LMW-pectin at 7.0 g/kg slightly increased the water-holding capacity (from 45.71% to 46.53%) and significantly reduced the total acid content (from 6.42 g/kg to 5.24 g/kg) and hardness (from 114.27 N to 76.73 N). LMW-pectin also reduced the gel strength of yogurt slightly. It also could modulate the tastes (particularly bitterness and astringency) and aromas via increasing the concentrations of specific esters, making the yogurt demonstrate cheesy, fragrant odors. LMW-pectin increased the relative abundance of Bacillus from 0.91% to 6.78%. LMW-pectin increased the concentrations of medium-chain fatty acids, odd-chain fatty acids, and unsaturated fatty acids. LMW-pectin also increased the scores of taste and odor in yogurt significantly. In contrast, FOS only affected gel strength, electronic tongue values, and the concentrations of volatile compounds. LMW-pectin can serve as a functional additive for improving the texture, flavor, and nutritional quality of yogurt, providing a practical strategy for utilization in the dairy industry.

Interfacial behavior of quinoa protein-tannic acid particles regulated by high-methoxyl pectin for multi-responsive high internal phase emulsions as hesperetin carriers

Guangfan Qu,Jiangling Zhang,Feiyan Yang,Xu Wang,Shuguo Sun

HMP significantly reduced the oil-water interfacial tension of QT particles. • QTH particles shows a high viscoelastic interface film on the O/W interface. • Pickering HIPEs prepared from QTH particles exhibit multi-stimuli responsiveness. • Multi-responsive Pickering HIPEs improve the bioavailability of hesperetin. Protein-polyphenol composite particles are ideal carriers for bioactive substances. However, their tendency to aggregate and precipitate under weakly acidic conditions limits their broader application. This study investigated the effect of interfacial behavior of quinoa protein isolate-tannic acid (QPI-TA) particles regulated by high-methoxyl pectin (HMP) via self-assembly on the properties of Pickering high internal phase emulsions (HIPEs). Multispectral analysis demonstrated that HMP changed the hydrophobic microenvironment in QPI-TA (QT) particles, disrupted hydrogen bond structures, and triggered reassembly to form QPI-TA-HMP (QTH) composite particles. Interfacial adsorption kinetics, dilatational rheology, and quartz crystal microbalance with dissipation (QCM-D) analysis demonstrated that HMP significantly increased the interfacial pressure and adsorption rate of QT particles. The interfacial film formed by QTHs particles showed enhanced compactness, thickness, viscoelasticity, and mechanical strength relative to that of QT particles alone. Moreover, Pickering HIPEs stabilized by QTHs composite particles exhibited excellent stability and intelligent responsive properties under various oil phases, pH, centrifugation, temperature and storage conditions. Simulated in vitro digestion results demonstrated that both the release of free fatty acids and the bioaccessibility of hesperetin in hesperetin-loaded HIPEs were dramatically improved. This study demonstrates that incorporating anionic polysaccharides to optimize interfacial behavior in multi-responsive Pickering HIPEs provides a promising strategy for effective encapsulation and controlled delivery of hesperetin. Dielectric barrier discharge plasma-assisted extraction of Rhamnogalacturonan-I pectin from pomelo peel: Structural characterization and application in an emulsion gel delivery system

Jing Wen,Jingyang Wang,Huining Lai,Zhengang Zhao

Pomelo peel, a major byproduct of pomelo processing, was utilized to isolate rhamnogalacturonan-I (RG-I) pectin via a novel approach combining low-concentration alkaline extraction with dielectric barrier discharge (DBD) plasma treatment, followed by characterization. The extracted pectin (DAP) was characterized by its esterification degree, monosaccharide composition, molecular weight, X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), triple-helix structure, and scanning electron microscopy (SEM). Results revealed that DBD increased its RG-I content while preserving its overall structure. Additionally, the effects of DAP, oil phase, CaCO₃ concentration, and glucono-δ-lactone (GDL) concentration on the properties of the resulting emulsion gel were investigated. In vitro release studies indicated that the emulsion gel followed the Korsmeyer-Peppas model, demonstrating controlled-release behavior. After simulated in vitro digestion, 70% of the encapsulated naringenin available for release in intestine. This study provides a theoretical foundation for the green extraction of RG-I pectin from pomelo peel and application in delivery systems.

Effect of localized electrochemical pH and temperature synergistic modification on the structural and antibacterial properties of pectin/polyvinyl alcohol/zinc oxide nanorod films

Zhanwei Yang,Jin Wang,Hualei Chen,Hecheng Meng,Xiaobing Guo,Shujuan Yu

Two low-methoxy pectins (LMPs) were obtained by local electrochemical pH modification using an H-type double-layer water bath sealed electrochemical cell at the voltage of 180 V for 3 h. The weight-average molecular weight (Mw) of citrus peel pectin (CPP) prepared in the anodic part at room temperature (CPP-A 5/RT ) and in the cathodic part at 5 °C (CPP-C 5/RT ) were 346 kDa and 328 kDa, respectively, and the degrees of methylation (DM) were 36.8 % and 11.9 %. Moreover, the second-order kinetic model was most appropriate for the degradation processes, as free radicals were generated in the anodic part and β -elimination occurred in the cathodic part. Subsequently, CPP-A 5/RT and CPP-C 5/RT were utilized to fabricate food packaging film blending with polyvinyl alcohol (PVA), bcZnO (ZnO coupled with bentonite and colophony) nanorods , and Ca 2+ ions by casting method. Then the prepared films were studied for their ability to maintain the freshness of strawberries. The addition of Ca 2+ ions and bcZnO nanorods increased the thickness, water contact angle (WCA), and mechanical properties of the composite films, while decreased water vapor permeability (WVP). Therefore, the CPP-based films, supplemented with bcZnO nanorods and crosslinked with Ca 2+ ions by “egg-box” model, can serve as an antibacterial food packaging material for food preservation.

A novel strategy for producing low-sugar pomegranate jam with better anthocyanin stability: Combination of high-pressure processing and low methoxyl & amidated pectin

Yurou Yun,Jian Li,Fei Pan,Yijun Zhou,Xiangqiao Feng,Jun Tian,Shengbao Cai,Junjie Yi,Linyan Zhou

Low-sugar jams (LSJ) are promoted as a healthier food option due to the healthy trend, and significant color changes negatively affects the evaluation of these products. To reveal protective effect of low methoxyl & amidated pectin (LMA) on anthocyanin in jam treated by high-pressure processing (HPP), this study compared the changes in microbes and quality attributes of LSJ prepared with LMA and high-sugar pomegranate jam (HSJ) prepared with high methoxyl pectin (HMP) after HPP at 600 MPa for 5 min during accelerated storage at 35 °C for 30 days, and molecular dynamics simulation was conducted to reveal the interaction between LMA and anthocyanin. LSJ-HPP had better color quality with higher values of L* , a* , and total anthocyanin content (TAC), the corresponding percent polymeric color and browning index were 41% and 16% lower than those of HSJ-HPP, respectively. TAC in LSJ-HPP, HSJ, and HSJ-HPP decreased to 9.5, 8.6, and 6.0 mg/kg (pomegranate puree) after storage, respectively. Cyanidin-3- O -glucoside formed more H-bonds with LMA than HMP, while their total interaction energy and the binding frequency and number were also higher. This study provided a foundation for the processing and storage of high quality LSJ, and also broadened the application field of HPP technology.

The role of amide groups in the mechanism of acid-induced pectin gelation: A potential pH-sensitive hydrogel based on hydrogen bond interactions

Shuhan Feng,Jianyong Yi,Youchuan Ma,Jinfeng Bi

Gelation of amidated pectin in the acid environment with the absence of calcium has been found, but the mechanism and gel properties are still not fully understood, especially the role of amide groups. In this study, a series of pectins with different degrees of amidation (DA) were prepared by ammonia reaction, and their structural characteristics including neutral sugar contents, molecular weight contribution, and molecular morphology were fully identified. Acid-induced hydrogels with various DA were then obtained to investigate the impact of amino groups on pectin gelation. Results showed that the higher the DA, the faster the gelation process, and the more stable the gel network. Dynamic rheological and pH stability tests confirmed the essential roles of low pH value (<3.5) and high DA (>49.76%) on the amidated pectin gelation without calcium. Hydrogen bonds were identified as the main interaction for the amidated gel mechanism and enabled the thermo-reversible properties of amidated gels. High content of amide groups allowed the formation of a denser gel network structure with greater mechanical strength, stronger thermo-reversibility, and higher pH stability.

An approach of pectin from Citrus aurantium L. for superabsorbent resin with superior quality for hygiene products: Salt resistance, antibacterial, nonirritant and biodegradability

Peng Zhou,Xiangzhou Li,Zhi Jiang,Jun Zhou,Guang He,Limin Qu

In this paper, a kind of superabsorbent resin (SAR) with superior quality for hygiene products was developed using Fructus Aurantii Immaturus pectin (FAIP) from Citrus aurantium L.. FAIP-g-AM/AMPS SAR was established by free radical graft co -polymerization with FAIP as skeleton structure, N, N′-Methylene-bis (acrylamide) (MBA) as the cross-linker. Meanwhile, the functional monomers of acrylamide (AM) and 2-acrylamido-2-methylpropane sulfonic acid (AMPS) were introduced. The structure and morphology of FAIP-g-AM/AMPS were characterized by FTIR , 13 C NMR, XRD, SEM and TG-DSC analysis. The results confirmed that the AFIP-g-AM/AMPS SAR was successfully prepared, which exhibited a three-dimensional (3D) network structure and an excellent thermal stability . The absorption and retention capacity of FAIP-g-AM/AMPS was comparable to or even better than commercial diapers and sanitary napkins. Significantly, FAIP-g-AM/AMPS itself exhibited excellent antibacterial and safety. FAIP-g-AM/AMPS has an inhibition ratio of 97.1 % for Escherichia coli ( E. coli ) and 98.5 % for Staphylococcus aureus ( S. aureus ), and was non-irritating and non-allergic to the skin. In addition, FAIP-g-AM/AMPS presented amazing biodegradability and a weight loss reached 37.1 % after 30 days by soil burial test. The research provides a safe and high-performance SAR, which expected to be used in hygiene products such as baby diapers, adult incontinence pads and sanitary napkins.

Fabrication and digestive characteristics of high internal phase Pickering emulsions stabilized by ovalbumin-pectin complexes for improving the stability and bioaccessibility of curcumin

Lechuan Wang,Hong Zhang,Hanyu Li,Huajiang Zhang,Yujie Chi,Ning Xia,Zhenshun Li,Longwei Jiang,Xiaonan Zhang,Ahmed M. Rayan

In this study, ovalbumin (OVA) interacted with pectin (PE) to form soluble electrostatic complexes to improve the functional properties of high internal phase Pickering emulsions (HIPEs) under extreme conditions. The results showed that the stability of the OVA-PE soluble complexes-stabilized HIPEs was significantly better than that of the free OVA-stabilized HIPEs and was modulated by the biopolymer ratio. In particular, the complexes at an OVA:PE ratio of 1:1 (C-1:1) may form particulates with a core-shell structure by a flocculation mechanism. The C-1:1-stabilized HIPEs had the smallest oil droplet size (11.34 ± 1.14 μm) and the best resistance to extreme environmental stresses due to their strong, rigid structure and dense interfacial architecture. The in vitro digestion results showed that the bioaccessibility (from 18.3% ± 0.5% to 38.8% ± 4.8%) of curcumin improved with increasing PE content. Our work is helpful in understanding OVA-PE complexes as stabilizers for HIPEs and their potential applications in food delivery systems.

Oil-in-water emulsions prepared using high-pressure homogenisation with Dioscorea opposita mucilage and food-grade polysaccharides: guar gum, xanthan gum, and pectin

Zeyue Ren,Xiaojing Li,Fanyi Ma,Yun Zhang,Weiping Hu,Md Zaved Hossain Khan,Xiuhua Liu

In this study, oil-in-water emulsions made of medium-chain triglycerides (MCT), mucilage from Dioscorea opposita (DOM), and food-grade polysaccharides (guar gum [GG], xanthan gum [XG] and pectin [Pec]) were prepared using high-pressure homogenisation. The droplet size distributions, microstructure, turbidity, interfacial tension, creaming index, and stability of emulsions were investigated and compared with those of DOM, GG, XG and Pec. The results showed that 0.4 wt% food-grade polysaccharides (GG, XG, and Pec) with 2 wt% DOM contributed more to the stability of the emulsion during storage. In particular, low concentrations of pectin and DOM emulsions presented smaller droplet size distribution, in the range of 86.34–111.30 nm. Hence, DOM has synergistic effects with food-grade polysaccharides, which could improve the stability of emulsions, suggesting that mucilage from Dioscorea opposita has good potential for use as a natural emulsifier.

Preparation and characterization of pectin/chitosan beads containing porous starch embedded with doxorubicin hydrochloride: A novel and simple colon targeted drug delivery system

Jianzhong Zhu,Li Zhong,Wenxue Chen,Yizhe Song,Zhengming Qian,Xianying Cao,Qiang Huang,Bin Zhang,Haiming Chen,Weijun Chen

The objective of this study was to design a simple, colon targeted drug delivery system by using porous starch (PS), pectin and chitosan. Porous maize starch was prepared by hydrolysis with a combination of α-amylase and amyloglucosidase, and it was characterized by scanning electron microscopy, revealing the formation of porous structures in the starch granules. A Brunauer-Emmett-Teller (BET) specific surface area analysis indicated that the specific surface area of the PS (0.8768–0.9448 m 2 /g) was 19.88–21.42 times larger than that of native maize starch (0.0441 m 2 /g). The average pore diameter of the PS granules, as calculated by the Barrett-Joiner-Halenda (BJH) method, was 40.52–62.42 nm. A favorable adsorptive potential of the PS granules was verified by water and soybean oil tests. Doxorubicin was loaded into PS granules, which were then coated with a pectin/chitosan complex solution. The results of confocal laser scanning microscopy (CLSM) demonstrated that doxorubicin was successfully absorbed into the PS granules. In addition, an in vitro simulated digestion method demonstrated the effectiveness of this delivery design, as only a 13.80% release rate of doxorubicin was observed in the upper gastrointestinal tract, whereas release rates of 17.56% and 67.04% were observed for pectin/PS/doxorubicin and pectin/doxorubicin beads, respectively. It was concluded that the use of PS and a pectin/chitosan coating is an effective method for colon targeted drug delivery compared with the simple polysaccharide system.

Multi-stimuli-responsive pectin-coated dendritic mesoporous silica nanoparticles with Eugenol as a sustained release nanocarrier for the control of tomato bacterial wilt

Guo Xueping,Li Huiyan,Li Zhihao,Cui Ziqi,Ma Guangming,Nassor Aisha Khalfan,Guan Yi,Pan Xiaohong

Background Environmentally responsive nanoscale biocide delivery system enhances smart, regulated, and synergistic biocide application with precise biocide release. In this study, pectin-modified dendritic mesoporous silica nanoparticles (DMSNs) was used as a carrier to successfully construct a microenvironment-responsive (pH, temperature and enzyme) eugenol nano-biocide delivery system for the control of Ralstonia solanacearum infection. Results The results showed that the specific surface area, pore size and surface activity of DMSNs significantly influence the biocide loading of eugenol, and the biocide loading capability was up to 72.50%. Eu@DMSNs/Pec had significant pH and pectinase stimulating effects, with varying release amounts under different temperature conditions. Compared with eugenol alone, Eu@DMSNs/Pec significantly enhanced the efficacy of eugenol. DMSNs assisted eugenol to induce peroxidation damage, produce ROS (•O 2 − , •OH and 1 O 2 ), achieve synergistic antibacterial effects, and had better rain erosion resistance and foliar retention rate based on pectin wettability and adhesion. Eu@DMSNs/Pec-FITC showed demonstrated efficient transport characteristics in tomato roots, stems and leaves, which enhanced the control effect on tomato bacterial wilt. In addition, Eu@DMSNs/Pec exert minimal influence on tomato seed germination and root growth, and have low toxicity to non-target organisms such as earthworms. Therefore, Eu@DMSNs/Pec environment-responsive nano-controlled release nanocarrier can effectively achieve accurate biocide release and reduce biocide dosage. Conclusion This work not only provides a pectin-modified DMSNs-based eugenol nanoscale biocide delivery system in response to specific environmental conditions of R. solanacearum infection but also elucidates the eugenol biocide loading, selective release ability and antibacterial mechanism of the system.
