Natural deep eutectic solvent-based headspace gas chromatography for determination of 14 residual solvents in tea polyphenol products

Hengyu Xu,Zheyuan Yao,Xianyu Bao,Yina Cai,Xiaobo Zuo,Chifang Peng

A headspace gas chromatography (HS-GC) method using natural deep eutectic solvent (NADES) as a green matrix medium was reported for the simultaneous determination of 14 residual solvents in tea polyphenol products. Through systematically screening, a NADES, choline chloride-glycerol (1:2, mol/mol), was selected as the optimal matrix medium in tea polyphenol dissolution. After the headspace parameters, including equilibrium temperature and time were optimized. A HS-GC method was developed and demonstrated good linearity (R² = 0.9874–0.9995) over 0.005–1000 μg/g. Limits of detection (LODs) and quantification (LOQs) ranged from 0.01 to 0.25 μg/g and 0.03 to 0.83 μg/g, respectively. In spiked tea polyphenol samples, the recoveries from 82.4% to119.0% were obtained with relative standard deviations (RSDs) of 0.14%–4.91%. And the above HS-GC method was utilized to measure 41 commercial tea polyphenol products. The results confirmed that the detected residual solvents in all the samples were below the Permissible Daily Exposure (PDE) limits. Risk assessment based on the hazard quotient (HQ, calculated as the ratio of Daily Intake to PDE) yielded values of 50.8% for benzene and 1.47% for dichloromethane. These results suggest that benzene requires prioritized control as a critical point. And we also should pay attention to dichloromethane. This study provides a green and efficient approach for residual solvent analysis in tea polyphenol products and demonstrates the potential of NADESs as sustainable matrix media in food safety testing.

Tea polyphenol-modified MWCNTs/PEDOT:PSS flexible films with “brick-mortar” structure for electrical heating and electromagnetic interference shielding

Di Zhang,Zhengxin Cao,Jiayi Li,Kai Yue,Zhixiong Han

The composite films with superior electrical heating and EMI shielding performance were obtained by a facile and novel method. • The composite film achieved superior electrothermal response under the voltage of as low as 6   V, and maintained stable performance after multiple cyclic tests. • The film attained an EMI shielding effectiveness of 24   dB, and reflection palyed an important role. • The performances were in good accordance with the COMSOL simulation results, further enabling the reliability. • The superior properties of material enabled it as a promising a material for wearable thermal management and electronic device electromagnetic compatibility fields. Flexible multifunctional composites show great potential in emerging wearable electronics. Nevertheless, most reported materials have several limitations, including high filler loading, intricate fabrication process and poor functional synergy, which cannot satisfy the requirements of stable electrothermal performance and electromagnetic protection for wearable devices. Therefore, this study proposed a “brick-mortar” structure to improve the carbon nanotube dispersion, and constructed continuous conductive pathways, achieving synchronous electrothermal and electromagnetic interference (EMI) shielding performance. Accordingly, tea polyphenol-modified multi-walled carbon nanotubes (T-MWCNTs)/poly(3,4-ethylenedioxythiophene)-poly(styrenesulfonate) (PEDOT:PSS)/polyvinyl alcohol (PVA) composite films (TPFs) were prepared via layer-by-layer deposition. The “brick” network was formed by T-MWCNTs and PEDOT:PSS provided continuous conductive paths, while PVA as the “mortar” enhanced the flexibility and interfacial adhesion. The composite film exhibited a rapid electrothermal response under 6   V, reaching to 123 °C within 90   s with superior cycling stability. It also showed an EMI shielding effectiveness of 24   dB, and reflection played an important role. These results were verified by COMSOL simulation as well. TPFs shows promising potential in wearable thermal management and electromagnetic compatibility. Download: Download high-res image (151KB) Download: Download full-size image

Anti-polyphenol oxidase mechanism of oligomeric procyanidins and its application on browning control of “Baiyu” loquat during storage

Gui-Li Huang,Tian-Tian Liu,Jia-Jia Ma,Ling-Xiang Sun,Si-Yao Sui,Xin-Yao Quan,Yu-Ning Wang

The harvested loquat fruits are prone to browning, leading to a decline in fruit quality. Inhibiting activity of polyphenol oxidase (PPO) is an important way to prevent browning. In this study, we evaluated the inhibitory impact of oligomeric procyanidins (OPC) on hydroxylation of monophenol and oxidation of o-diphenol catalyzed by PPO and effect of OPC on loquat fruits storage . The obtained results revealed that OPC could attenuate the activity of monophenolase and diphenolase. For monophenolase activity, the inhibition rate was observed to be dose-dependent with an IC 50 of 11.6 ± 3.4 μg/mL for the steady-state rate. For diphenolase activity, OPC inhibited PPO activity ( IC 50  = 15.75 ± 2.1 μg/mL) with a reversible and competitive mechanism and quenched fluorescence of PPO by forming OPC-PPO complex with one binding site in a static procedure. In addition, the data obtained from storage assays revealed that OPC could enhance the quality of harvested loquat fruits by attenuating the PPO activity, augmenting the content of total phenolics and flavonoids , and simultaneously delaying the browning of loquats. In this research, we identified a potent PPO inhibitor and expanded its potential as a food preservative.

Fluorometric and colorimetric analysis of alkaline phosphatase activity based on a nucleotide coordinated copper ion mimicking polyphenol oxidase

Hui Huang,Juan Bai,Jiao Li,Lulu Lei,Wenjing Zhang,Shujun Yan,Yongxin Li

In this work, a fluorometric and colorimetric analysis of alkaline phosphatase (ALP) activity was developed based on nanozymes. The nanozymes were composed of nucleotides (ATP, ADP and AMP) coordinated with copper ions. All three kinds of nanozymes (ATP-Cu, ADP-Cu and AMP-Cu) exhibited polyphenol oxidase (PPO)-mimic activity by catalyzing a chromogenic reaction of 2,4-dichlorophenol (2,4-DP) and 4-aminoantipyrine (4-AP). However, there were obvious differences in the PPO-like activity and the fluorescence of the three nanozymes produced from the same concentration of nucleotides (keeping the concentration of Cu2+ unchanged at 5 mM). The catalytic activities of produced ADP-Cu and AMP-Cu were obviously higher than that of ATP-Cu at a certain nucleotide concentration of 3 mM. In addition, when ATP was hydrolyzed into ADP and AMP by ALP, more nanozymes were produced and the catalytic activity of the system was enhanced, which resulted in an obvious increase of the colorimetric signal. The signal intensity was proportional to ALP concentration in the range of 0–30 U L−1, and the detection limit for ALP was 0.3 U L−1 from the colorimetric detection. Moreover, the fluorescence intensity of the produced nanozymes was also proportional to the ALP concentration in the range of 1–30 U L−1 and the detection limit was 0.45 U L−1 from the fluorescence detection. A fluorometric and colorimetric sensing ALP method was thus established. The method showed a high selectivity for ALP activity compared with proteins, amino acids and other interference components. Furthermore, the proposed method was also used to detect ALP activity in human serum samples, which showed great potential for diagnostic and practical purposes.

NIR-Triggered Metal-Polyphenol Nanoparticles Enhance HPV-Driven Cancer Immunotherapy via Immunogenic Cell Death and STING Sequential Activation

Maoyu Liu,Jindong Zhang,Shuning Chen,Jiao Zheng,Linlin Xiao,Xiaoli Liu,Yang Cao,Shenyin Zhu,Shufang Chang

HPV-associated malignancies consistently express E6/E7 oncoproteins, making these viral antigens prime targets for therapeutic vaccination. However, insufficient antigen exposure and presentation remain major obstacles for potent immunotherapy. Here, a novel metal-polyphenol network-coated human serum albumin nanoplatform (IMT@H) is engineered to co-deliver IR780 and manganese ions (Mn 2+ ) to achieve enhanced immunogenic cell death (ICD) and cGAS-STING-dependent antigen presentation. The metal-polyphenol nanostructure facilitates the pH-responsive release of Mn 2+ , which subsequently initiates Fenton-like reactions to generate hydroxyl radicals (·OH). Meanwhile, under near-infrared (NIR) light irradiation, IR780 induces mitochondrial-targeted phototherapy and concurrently produces reactive oxygen species (ROS). These processes act synergistically to amplify the oxidative damage and ICD in TC-1 tumors, leading to the release of damage-associated molecular patterns (DAMPs). These ICD-derived DAMPs cooperate with Mn 2+ to sustain activation of the cGAS-STING pathway in dendritic cells. This combinatorial strategy successfully transforms tumor antigens into endogenous vaccines, eventually inhibiting the growth of primary tumors and producing strong abscopal effects. Notably, mice primed with nanovaccines exhibit strong anti-HPV16 E7-specific immune responses and tumor resistance. With its dual therapeutic and preventive functionality, IMT@H represents a paradigm-shifting strategy for virus-driven malignancies and offers a blueprint for engineering self-adjuvanting nanovaccines against viral oncogenesis.

Tea polyphenol and HfCl4 Co-doped polyacrylonitrile nanofiber for highly efficient transformation of levulinic acid to γ-valerolactone

Rulu Huang,Huai Liu,Junhua Zhang,Yuan Cheng,Liang He,Lincai Peng

Design catalytic materials with high Lewis acid/base sites for catalytic transfer hydrogenation (CTH) of levulinic acid (LvA) to γ-valerolactone (GVL) is highly demanded but challenging. Herein, we report a concept of metal ion-polymer hybridization to design a tea polyphenol (TP) and HfCl 4 co-doped polyacrylonitrile (PAN) nanofiber (Hf@PAN-TP) via electrospinning technique . Comprehensive characterization revealed the well coordination between hafnium ions and phenolic hydroxyl groups of TP on PAN fiber, which promoted the formation of Lewis acid and Lewis base sites. As expected, a satisfactory GVL yield of 99.1% with 100% of LvA conversion was obtained using isopropanol as a hydrogen donor at 170 °C in 5 h, and the catalytic activity remained unchanged for five cycles. The kinetic study revealed the lower activation energy for CTH of LvA to GVL (33.5 kJ/mol), demonstrating the outstanding GVL formation rate of 3383.9 μmol/g . h. Based on the systematical combine of catalyst characterization and catalytic behavior, the structure-function relationship and reaction mechanism were proposed. More gratifyingly, Hf@PAN-TP was applicable for catalytic reduction of various carbonyl compounds, demonstrating a great potential for upgrading biomass-derived aldehydes/ketones.

Effect of Layered Debranning Processing on the Proximate Composition, Polyphenol Content, and Antioxidant Activity of Whole Grain Wheat

Wenwen Guan,Duqin Zhang,Bin Tan

Layered debranning processing (LDP) of whole grain wheat (WGW) could not only retain more bioactive compounds but also contributes to grain saving policy as compared with the refined wheat flour (WF). In this study, effect of different debranning rates from 0 to 13.37% on the proximate composition, polyphenol content, and the antioxidant activity were analyzed. As debranning rates increased from 0 to 13.37%, the insoluble dietary fiber content decreased from 9.94% to 6.47%, whereas the soluble dietary fiber contents increased from 3.06% to 3.98%. The free phenolic content decreased by 62.72%, while the free flavonoid content increased by 4.68% with debranning rates increasing. For the phenolic acids, protocatechuic acid and ferulic acid dominated the free and bound phenolic acid in WGW, which showed the highest contents at 6.95% and 4.45% debranning rates, respectively. As for flavonoids, naringenin (the free-state phenolic) and rutin (the bound state phenolic) in WGW had the greatest level at 4.45% debranning rate. As compared to WGW and WF, LDP significantly improved the DPPH, ABTS·+ radical scavenging activities and total antioxidant activities. In conclusion, 4.45% and 6.95% were the best debranning rates to retain polyphenol contents and antioxidant activities.

NIR-triggered tea polyphenol-modified gold nanoparticles-loaded hydrogel treats periodontitis by inhibiting bacteria and inducing bone regeneration

Zhiqiang Dong,Yiyi Lin,Shibo Xu,Linna Chang,Xingjun Zhao,Xifan Mei,Xiuqiu Gao

Periodontitis caused by bacteria is the most common oral disease. In the face of plaque formation, traditional scaling and antibiotic treatment can lead to gum recession, drug resistance and other side effects. The nanoplatform of multifunctional complex treatment is attractive for the treatment of periodontitis. In this study, epigallocatechin gallate (EGCG) was loaded into gold nanoparticle-modified hydrogels ( [email protected] ) to achieve the synergistic functions of near-infrared (NIR) photosensitization, bactericidal and periodontal tissue regeneration. [email protected] rapidly heats up to 50.7 °C within 5 min under NIR light irradiation, exerting a good photothermal effect, while the NIR spectrum can effectively control the release of tea polyphenols, enhance the antibacterial effect, promote angiogenesis and improve bone regeneration. In vitro studies showed that NIR-irradiated composites inhibited S. aureus , E. coli and S. aureus biofilms by 94 %, 92 % and 74 %, increased alkaline phosphatase activity 5-fold after 7 days and increased extracellular matrix mineralization rate 3-fold after 21 days. The rat periodontitis model successfully demonstrated that [email protected] with NIR light irradiation inhibited dental plaque biofilm by 87 % and promoted alveolar bone regeneration by 38 %. The experimental results suggest that this hydrogel with a NIR-sensitive composite nanoplatform offers new possibilities for the treatment of periodontal disease.

Protective role of polyphenol extract from highland barley against cisplatin-induced renal toxicity and mitochondrial damage in rats

Yunfan Liu,Jiazhou Zhai,Fei Qin,Liping Gao,Yongxin She,Mengqiang Wang

The aim of this study was to investigate the protective effect of polyphenol extract from Highland barley (HBPE) on cisplatin (CDDP)-induced oxidative damage and mitochondrial damage in rat kidney. The experimental rats were divided into control group, CDDP group, HBPE (600 mg/kg) group, CDDP+HBPE (150 mg/kg) group, CDDP+HBPE (300 mg/kg) group and CDDP+HBPE (600 mg/kg) group. Rats in each group were intragastrically fed with normal saline and corresponding dose of HBPE for 21 d. On the 16th day, CDDP group and CDDP+HBPE group were injected intraperitoneally with CDDP (7.5 mg/kg), and the other groups were injected with the same amount of normal saline. On the 22nd day, the rats were killed and blood samples were taken to detect the contents of blood urea nitrogen (BUN) and creatinine (CrE) in serum, and calculate the kidney index. The kidney samples were used for the measurement of malondialdehyde (MDA), glutathione (GSH), superoxide dismutase (SOD), lactate dehydrogenase (LDH), malate dehydrogenase (MDH), hexokinase (HK), and processed for histopathological examinations. The results showed that HBPE improved the abnormalities of serum CrE, BUN, antioxidant and carbohydrate metabolic enzymes in kidney induced by CDDP. HBPE can also reduce the degeneration of renal tubules and glomerular atrophy. Therefore, HBPE has a significant protective effect on oxidative stress injury and mitochondrial damage induced by CDDP in rats.

Small-size Ti3C2Tx MXene nanosheets coated with metal-polyphenol nanodots for enhanced cancer photothermal therapy and anti-inflammation

Tao Liao,Zhongyin Chen,Ying Kuang,Zhe Ren,Wenqian Yu,Wen Rao,Linwei Li,Yun Liu,Ziqiang Xu,Bingbing Jiang,Cao Li

As a controllable, simple method with few side effects, near-infrared (NIR) light-based photothermal therapy (PTT) has been proven an effective cancer therapeutic approach. However, PTT-induced inflammation is a potential negative factor. And the overexpressed heat shock proteins (HSPs) by cancer cells can protect them from hyperthermia during PTT. In this work, small-size Ti 3 C 2 T x MXene nanosheets with high photothermal conversion efficiency in the region of NIR, high cargo loading capability and good free radical scavenging capability were chosen for cancer PTT and anti-inflammation. And (-)-epigallocatechin gallate (EGCG) was applied to form EGCG/Fe metal-polyphenol nanodots on the nanosheets. EGCG being released in acid cancer cells could reduce the expression of HSPs and could be used for anti-inflammation. As a result, the complex nanosheets named MXene@EGCG could achieve enhanced cancer PTT and be anti-inflammatory. Both in vitro and in vivo studies proved the good photothermal ability of MXene@EGCG and demonstrated that it could inhibit the expression of HSPs in tumor cells and relieve PTT-induced inflammation. Therefore, the nanosheets show good results in tumor ablation with a low level of inflammation, which provides another possibility for cancer therapy. Statement of significance Photothermal therapy (PTT)-induced inflammation plays an essential role in some important stages of tumor development and is unfavorable for cancer treatment. And hyperthermia leads to the overexpression of heat shock proteins (HSPs) in cancer cells, which limits the therapeutic effect of PTT. Therefore, we coated small-size Ti 3 C 2 T x MXene nanosheets with (-)-epigallocatechin gallate (EGCG)/Fe metal-polyphenol nanodots and named them as MXene@EGCG. This system shows a good photothermal conversion efficiency at 808 nm. And it can release EGCG in cancer cells to inhibit the expression of HSPs, thus achieving an enhanced cancer PTT. Both MXene and EGCG can also diminish the PTT-trigged inflammation. Both in vitro and in vivo studies prove the good anti-cancer PTT effect and anti-inflammation capability of MXene@EGCG.

Toward function starch nanogels by self-assembly of polysaccharide and protein: From synthesis to potential for polyphenol delivery

Jiejie An,Mingzhu Liu,Zia-ud Din,Fang Xie,Jie Cai

Nanogels formed by self-assembly of natural proteins and polysaccharides have attracted great interest as potential carriers of bioactive molecules. Herein, we reported that carboxymethyl starch-lysozyme nanogels (CMS-Ly NGs) were prepared using carboxymethyl starch and lysozyme by green and facile electrostatic self-assembly, and the nanogels served as epigallocatechin gallate (EGCG) delivery systems. The dimensions and structure of the prepared starch-based nanogels ( i.e. , CMS-Ly NGs) were characterized by dynamic light scattering (DLS), ζ-potential, Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD) and thermal gravimetric analyzer (TGA). FT-IR and 1 H NMR spectra together confirmed the formation of CMS; FT-IR spectra confirmed the formation of CMS-Ly NGs; XRD spectra confirmed the disruption of the crystal structure of lysozyme after electrostatic self-assembly with CMS, and further confirmed the formation of nanogels. TGA demonstrated the thermal stability of nanogels. More importantly, the nanogels showed a high EGCG encapsulation rate of 80.0 ± 1.4 %. The CMS-Ly NGs encapsulated with EGCG exhibited regular spherical structure and stable particle size. Under the simulated gastrointestinal environmental conditions, CMS-Ly NGs encapsulated with EGCG showed the controlled release potential, which increased its utilization. Additionally, anthocyanins can also be encapsulated in CMS-Ly NGs and showed slow-release properties during gastrointestinal digestion in the same way. Cytotoxicity assay also demonstrated good biocompatibility between CMS-Ly NGs and CMS-Ly NGs encapsulated with EGCG. The findings of this research suggested the potential application of protein and polysaccharides-based nanogels in the delivery system of bioactive compounds.

Effects of p-sulfonatocalixarene and p-sulfonatocalixarene/sulfobetaine surfactant complex on the activities of bromelain and polyphenol oxidase

Shuhuai Xia,Yujun Jiang,Xia Guo,Yaqiong Wang,Wenlin Xu

Calixarene is a third-generation supra-molecular compound and has broad application prospects in clinical disease diagnosis and intervention treatment. Calixarene/surfactant systems have received increasing attention recently because the properties of these systems can be customized. However, very little is currently known about the binding of calixarene and calixarene/surfactant systems to proteins and the effects of calixarene and calixarene/surfactant systems on protein activity. In this study, we studied the binding of sulfonatocalix[n]arene (SC[n], n = 4, 6 and 8) with bromelain (BM) and polyphenol oxidase (PPO) both in the absence and presence of sulfobetaine surfactant (N-dodecyl-N,N-dimethyl-3-ammonio-1-propanesulfonate (SDDAB)) using various techniques, including protein activity measurements, molecular simulation, and fluorescence, UV–Vis, 1 H NMR, and circular dichroism spectroscopies. We found that SC[n] entered the docking pocket of the proteins in a stoichiometric molar ratio of 1:1, whereas the proteins did not enter the cavity of SC[n]. In addition, SC[n] formed a stronger bond with BM than with PPO. SC[n] decreased the activities of the proteins, where the activity of BM was inhibited to a greater extent than that of PPO. SC[n] and SDDAB formed complexes. The proteins did not destroy the binding between SC[n] and SDDAB but bonded with the SC[n]/SDDAB complex instead by interacting with SDDAB. Moreover, the protein activities could be controlled by the molar ratio of SC[n] to the surfactant: a lower ratio corresponded to higher activity.
