Effects of artemisinin sustained-release algaecides on in-situ cyanobacterial inhibition and microbes-floating plants dominated ecosystem functions in artificial landscape lake

Yushen Ma,Xiaoming Zhu,Lixiao Ni,Cunhao Du,Wenlu Sang,Chu Xu,Jiahui Shi,Yiping Li,Shiyin Li,Yalun Pang,Linyun Zhang

The artemisinin sustained-release algaecides (ASAs) have been proven to be a safe and effective mean of inhibiting cyanobacteria in laboratory experiments. However, their effectiveness and impacts on ecosystem functions (EF) in natural waters are still unclear. In this study, the in-situ inhibitory effect of ASAs on cyanobacteria in natural waters was investigated over a period of 110 days to assess EF impacts dominated by microbes and floating plants. The results indicated that ASAs had a long-term inhibitory effect on cyanobacteria. ASAs did not affect sediment but increased TOC and TP in the water column in 2–10 days. Microbial diversity and network analyses indicated that ASAs enhanced bacterial diversity, network complexity, and hub-bacteria in networks. Metabolic pathway predictions and CCA analysis showed that ASAs maintained the stability of EF by enhancing the metabolic capacities of bacteria, and the relationships between metabolic microorganisms and environmental factors. PLS-PM revealed that ASAs primarily drove bacterial resistance to cyanobacteria, which was the key mechanism for its long-term inhibition of cyanobacteria. However, the early outbreak of floating plants was not conducive to the long-term inhibition of cyanobacteria by ASAs. This study provides new insights into the mechanisms and ecological impacts of cyanobacterial inhibition by ASAs in complex aquatic environments.

Nanocomposite hydrogels stabilized by self-assembled multivalent bisphosphonate-magnesium nanoparticles mediate sustained release of magnesium ion and promote in-situ bone regeneration

Kunyu Zhang,Sien Lin,Qian Feng,Chaoqun Dong,Yanhua Yang,Gang Li,Liming Bian

Hydrogels are appealing biomaterials for applications in regenerative medicine due to their tunable physical and bioactive properties. Meanwhile, therapeutic metal ions, such as magnesium ion (Mg 2+ ), not only regulate the cellular behaviors but also stimulate local bone formation and healing. However, the effective delivery and tailored release of Mg 2+ remains a challenge, with few reports on hydrogels being used for Mg 2+ delivery. Bisphosphonate exhibits a variety of specific bioactivities and excellent binding affinity to multivalent cations such as Mg 2+ . Herein, we describe a nanocomposite hydrogel based on hyaluronic acid and self-assembled bisphosphonate-magnesium (BP-Mg) nanoparticles . These nanoparticles bearing acrylate groups on the surface not only function as effective multivalent crosslinkers to strengthen the hydrogel network structure, but also promote the mineralization of hydrogels and mediate sustained release of Mg 2+ . The released Mg 2+ ions facilitate stem cell adhesion and spreading on the hydrogel substrates in the absence of cell adhesion ligands, and promote osteogenesis of the seeded hMSCs in vitro . Furthermore, the acellular porous hydrogels alone can support in situ bone regeneration without using exogenous cells and inductive agents, thereby greatly simplifying the approaches of bone regeneration therapy. Statement of Significance In this study, we developed a novel bioactive nanocomposite hydrogel based on hyaluronic acid and self-assembled bisphosphonate-magnesium (BP-Mg) nanoparticles. Such hydrogels are stabilized by the multivalent crosslinking domains formed by the aggregation of Ac-BP-Mg NPs, and therefore show enhanced mechanical properties , improved capacity for mineralization, and controlled release kinetics of Mg 2+ . Moreover, the released Mg 2+ can enhance cell adhesion and spreading, and further promote the osteogenic differentiation of hMSCs. Owing to these unique properties, these acellular hydrogels alone can well facilitate the in vivo bone regeneration at the intended sites. We believe that the strategy reported in this work opens up a new route to develop biopolymer-based nanocomposite hydrogels with enhanced physical and biological functionalities for regenerative medicine.

Mechanical properties, sustained release, and oxygen scavenging properties of nanocomposite films loaded with bimetallic nanoparticles (Fe2O3/TiO2) in extra virgin olive oil

Mahsa Kheirandish,Majid Javanmard dakheli,Maryam Mizani,Alireza Salehirad

The aim of this study was the synthesis of bimetallic nanoparticles based on Fe 2 O 3 /TiO 2 and its use in the poly(lactic acid) (PLA) films as an oxygen scavenger in extra virgin olive oil (EVOO) packaging. Bimetallic nanocomposites were prepared by two different precipitation methods (precipitation with ammonia and sodium hydroxide). The characteristics of bimetallic nanoparticles precipitated with sodium hydroxide (Na-Ti 0.01 Fe 0.048 O 0.08 ) and bimetallic nanoparticles precipitated with ammonia (NH-Ti 0.01 Fe 0.022 O 0.09 ) were compared. Relative amounts of elements in bimetallic nanocomposites and their morphological characteristics were determined using field emission scanning electron microscopy coupled with energy-dispersive X-ray spectrometer. Porosity volume and surface area of bimetallic nanoparticles were calculated using adsorption–desorption isotherms and the Brunauer–Emmett–Teller method. The formation/characterization of bimetallic nanoparticles and their location in the matrix of PLA-based nanocomposite film was studied using X-ray diffraction and Fourier transform infrared. In nanocomposite films based on PLA, bimetallic nanoparticles lead to better oxidative stability (peroxide value, p -anisidine index, K 232 , and K 270 ) of the EVOO and oxygen scavenging during storage compared to free nanoparticles. Mechanical properties of nanocomposite films were improved due to bimetallic nanoparticles, which were better for Na-Ti 0.01 Fe 0.048 O 0.08 . In vitro release modeling of the bimetallic nanoparticles in EVOO proved that Fickian diffusion is the dominant mechanism, and the Peleg model was the best description of the release behavior of nanoparticles.

Silica Microcapsules Containing Waste Soybean Cooking Oil with Enhanced Thermal Stability, Environmental Resistance, and Sustained-Release for Superior Bitumen Anti-Aging Performance

Peng Yang,Xiao-Nan Su,Jun-Feng Su,Qin-Yu Xing,Xin-Yu Wang,Qian Sun,Zhi-Yong Tan

Bitumen oxidative aging leads to pavement deterioration through increased stiffness and cracking, necessitating effective rejuvenation strategies. Current approaches utilizing waste soybean cooking oil (WSCO) are limited by rapid evaporation and leaching. This study presents an innovative solution through silica microencapsulation of waste soybean cooking oil (microWSCOs) synthesized via optimized oil-in-water emulsion with tetraethyl orthosilicate. The microcapsules exhibit uniform spherical morphology (10-50 μm diameter) with high encapsulation efficiency (92±3%) and thermal stability (<5% mass loss at 180°C for 2 h). Controlled release kinetics under simulated traffic loading (0.7 MPa, 10 Hz) demonstrate 62.4±2.1% cumulative rejuvenator release after 10,000 cycles, effectively restoring aged bitumen properties: softening point reduction of 15.2±0.8°C, penetration increase of 28.3±1.5%, and complex modulus decrease of 34.7±2.3%. Environmental resistance testing reveals 89.2±3.5% WSCO retention following 500 h UV irradiation (0.89 W/m²) and 7-day water immersion. The microencapsulation system demonstrates dual sustainability benefits by valorizing waste oil while significantly extending pavement service life through targeted, long-term rejuvenation.

Enhanced oil recovery in fractured low-permeability reservoirs by a novel gel system prepared by sustained-release crosslinker and water-soluble thixotropic polymer

Xu Li,Chunsheng Pu,Huilong Wei,Feifei Huang,Yun Bai,Chao Zhang

Polymer gels have been widely used in improving oil recovery and decreasing excessive water production in heterogeneous reservoirs after the long-term water flooding process. However, the high initial viscosity of polymer and the short gelation time still restricts the performance of polymer-based in situ cross-linked gels for in-depth conformance control. Herein, a novel gel system formed by in-situ crosslinking of water-soluble thixotropic polymer (WTP) and sustained-release crosslinker (SRC) was proposed. The WTP was synthesized by acrylamide, 2-acrylamido-2-methylpropanesulfonic acid, and N-[3-(dimethylamino)propyl] methacrylamide. It exhibits low viscosity under high shear rates and becomes thick under low shear rates, which ensures good injectability during the injection process and high gelation strength after cross-linking. The SRC was prepared by coating poly(ethyleneimine) (PEI) into the W/O/W multiple emulsions . The sustained-release mechanisms were well elaborated by confocal laser scanning microscopy (CLSM) observations. Only after the demulsification of multiple emulsions, the PEI would be released from the internal aqueous phase to the external aqueous phase. Therefore, these emulsions can be employed to deliver crosslinkers to specified targets in deep regions and prolong the release of crosslinkers to prevent undesirable crosslinking reactions near the injecting wells. According to the environmental scanning electron microscope (ESEM) observations and viscosity measurements, the low viscosity and an incompact spatial network could be observed in SRC/WTP gel system in the first 17 d, after which the PEI molecules were released to the external aqueous phase and cross-linked with WTP, forming the solid gel with small pores and yielding a high viscosity of 8793 mPa⋅s. Rheology tests have demonstrated that the elastic modules and the liner viscoelastic region of the novel gel system was nearly two times and twenty times than the traditional polymer gel, indicating higher gelation strength and better shear resistance. The core flooding tests showed that the SRC/WTP gel system had good injectability and could effectively plug the millimeter and submillimeter-sized cracks in low-permeability cores. 0.6 pore volume (PV) of SRC/WTP gel system with 3 gel slugs and a low injection rate can yield the best plugging performance in fractured core samples. And the gelation time shortens with the decrease of fracture apertures . Most importantly, the field tests in Ordos Basin verified the desirable EOR performance of the SRC/WTP gel system in fractured low-permeability reservoirs.

Highly efficient alginate sodium encapsulated chlorpyrifos/copper(II) Schiff base mesoporous silica sustained release system with pH and ion response for pesticide delivery

Chen Huayao,Lin Yueshun,Zhou Hongjun,Zhou Xinhua,Gong Sheng,Xu Hua

Salicylaldehyde modified mesoporous silica (SA–MCM-41) was prepared through a co-condensation method. Through the bridge effect from copper ion, the chlorpyrifos (CH) was supported on copper(II)/Schiff base mesoporous silica (Cu–MCM-41). And then CH–Cu–MCM-41 was encapsulated by alginate sodium (ALG) to form a highly efficient sustained-release system (ALG–CH–Cu–MCM-41). The sustained-release performance of ALG–CH–Cu–MCM-41 at different pH values and different NaCl concentrations was investigated. The as-synthesized system showed significant pH responsiveness. Under the condition of pH ≤ 7, the release rate of chlorpyrifos decreased with pH increasing. Meanwhile, its release rate under weak base conditions was slightly larger than under weak acid conditions. The sustained release curves of ALG–CH–Cu–MCM-41 were consistent with the sustained release curves of CH–Cu–MCM-41 but with a smaller release rate because of the block effect from ALG. Meanwhile, the drug release rate of the sustained release system was also affected by ion concentration. Their sustained-release curves could be described by the Korsmeyer–Peppas equation.

Fabrication of Bone Morphogenic Protein-2 and Icariin-Containing Sustained-Release Microcapsule and Evaluation of its Osteogenic Differentiation Capacity in MC3T3-E1 Cells

Zhen Hua,Cheng Jian,Shi Dong-jian,Chen Ji-wei,Peng Hong-cheng,Liu Ming-ming

Abstract The delivery of cytokines and Chinese medicines using biodegradable microspheres may promote bone regeneration. We designed a sustained-release microcapsule containing bone morphogenetic protein-2 (BMP2) and icariin (ICA) and evaluated its effects on osteogenic differentiation in MC3T3-E1 cells. ICA was preloaded onto sodium alginate-grafted phenylboronic acid (Alg-PBA) particles. Alg-PBA-ICA was used to coat the surface of SiO 2 -NH 2 colloids by electrostatic interaction, and then SiO 2 colloids were etched using HF. The Alg-PBA-ICA vesicles were immersed in a BMP2 solution, and Alg-PBA-ICA/BMP2 microcapsules were obtained. The optimal concentration of ICA and BMP2 for osteogenic differentiation induction was selected. We evaluated the microcapsules' osteogenic differentiation capacity. The microcapsules displayed intact morphology without obvious broken debris. The size of Alg-PBA-ICA@SiO 2 microcapsules was 210 ± 2 nm; the surface charge was –15.70 ± 2.12 mV. BMP2 (100 ng/mL) combined with 10 nM ICA had the strongest effect on cell behavior. The osteo-inductive effect at this concentration was stronger than that of the other combinations. The mRNA and protein expression levels of osteogenic genes increased after treatment with the microcapsules following 7 days of stimulation. This new drug delivery system has powerful osteo-inductive effects on MC3T3-E1 cells.

In vitro and in vivo evaluation of a novel collagen/cellulose nanocrystals scaffold for achieving the sustained release of basic fibroblast growth factor

Weichang Li,Yong Lan,Rui Guo,Yi Zhang,Wei Xue,Yuanming Zhang

Tissue-engineered dermis is thought to be the best treatment for skin defects; however, slow vascularization of these biomaterial scaffolds limits their clinical application. Exogenous administration of angiogenic growth factors is highly desirable for tissue regeneration. In this study, biodegradable gelatin microspheres (GMs) containing basic fibroblast growth factor (bFGF) were fabricated and incorporated into a porous collagen/cellulose nanocrystals (CNCs) scaffold, as a platform for long-term release and consequent angiogenic boosting. The physicochemical properties of these scaffolds were examined and the in vitro release pattern of bFGF from scaffolds was measured by ELISA. Collagen/CNCs scaffolds with and without bFGF-GMs were incubated with human umbilical vein endothelial cells for 1 week, results showed that the scaffolds with bFGF-GMs significantly augmented cell proliferation. Then, four different groups of scaffolds were implanted subcutaneously into Sprague–Dawley rats to study angiogenesis in vivo via macroscopic observation, and hematoxylin and eosin and immunohistochemical staining. The results suggested that the collagen/CNCs/bFGF-GMs scaffolds had a significantly higher number of newly formed and mature blood vessels, and the fastest degradation rate. This study demonstrated that collagen/CNCs/bFGF-GMs scaffolds have great potential in skin tissue engineering.

Development and Validation for the Estimation of Assay and Forced Degradation Impurities of Goserelin Acetate Sustained-Release Implant Using HPLC and LC–MS

Huang Dan,Tian Yufei,Zheng Yili,Li Mengyu,Xu Xia,Chen Hongyuan,Rui Wen

Forced degradation studies aim to systemically characterize the degradation products of an active pharmaceutical ingredient (API) and its formulations under harsh conditions that accelerate degradation. These studies are especially valuable for sustained-release products in selecting appropriate packaging materials and storage conditions in the field of pharmaceutics. To demonstrate the degradation pathways and degradation products of the API and products, mass spectrometry is used for further characterization of the degradation products. Goserelin acetate sustained-release implant, which is prepared by hot melt extrusion using biodegradable material polylactic acid-glycolic acid copolymer as a carrier, is used for the treatment of early breast cancer in premenopausal and perimenopausal women. In this study, a simple and sensitive high-performance chromatography method was established following the ICH guidance. We conducted forced degradation experiments to determine the conditions of the product. The forced degradation studies of the goserelin acetate sustained-release implant showed the presence of a single impurity during thermal degradation, as observed in the HPLC analysis. To elucidate the structure of the impurity and characterize its degradation pathway, we performed further characterization using LC–MS.

Phytoferritin functions in two interface-loading of natural pigment betanin and caffeic acid with enhanced color stability and the sustained release of betanin

Yidan Zhang,Xinmei Sha,Jiangnan Hu,Jincan Wu,Yue Li,Junrui Ma,Haili Sun,Rui Yang

Betanin, a natural red pigment, is sensitive and prone to fading and discoloration, affecting its stability and bioavailability. Phytoferritin is a nano-diameter protein with unique interior-/exterior-interfaces. By the unique interfaces and pH-induced self-assembly of ferritin, a ferritin–betanin complex (FB) with an encapsulation efficiency of 17.66 ± 1.24% was prepared. The caffeic acid–FB (CFB) was further fabricated by attaching ferritin with caffeic acid, and the binding number n of caffeic acid was 88.47 ± 9.49, with a binding constant K of (1.63 ± 0.33) × 104 M−1. Fluorescence and Fourier transform infrared analysis indicated that the encapsulation of betanin and the binding of caffeic acid influenced the ferritin structure. The interaction between caffeic acid and ferritin was mainly through van der Waals forces and hydrogen bonds. TEM and DLS showed that the globular structure and diameter (12 nm) remained in CFB. Furthermore, the ferritin and caffeic acid exhibited a synergistic effect in enhancing thermal, light, and ferric ion stabilities, and controlled the betanin release in a more sustained manner in the simulated gastrointestinal tract. In addition, the antioxidant capacity of CFB was enhanced compared with free betanin. This study promotes the bioavailability of betanin by two interface-loading of ferritin, and guides the use of ferritin nanoparticles as a nanocarrier for pigment stabilization.

pH-Sensitive mesoporous silica nanoparticles anticancer prodrugs for sustained release of ursolic acid and the enhanced anti-cancer efficacy for hepatocellular carcinoma cancer

Tao Li,Xiufen Chen,Yajun Liu,Lulu Fan,Liqing Lin,Yu Xu,Sijia Chen,Jingwei Shao

Ursolic acid (UA) as a nature product exhibits good anti-cancer activity, low toxicity, and good liver protection features. However, the low-solubility and poor bioavailability restrict its further clinical application. To overcome this problem, a pH-sensitive prodrug delivery system (UA@MSN-UA) that incorporated acid-sensitive linkage between drug and silica-based mesoporous nanosphere (MSN) was successfully designed and synthesized. The physicochemical properties of the UA@MSN-UA nanoparticles were investigated for shape, particle size, zeta potential , nitrogen adsorption-desorption and infrared (IR) spectroscopy. The nanoparticles were further evaluated for in vitro cytotoxicity, including proliferation inhibition, cell cycle distribution and apoptotic effects against human hepatocellular carcinoma HepG2 cells. The TEM image showed that the size of synthesized MSN nanoparticle was a near-spherical shape with ~   100   nm diameter. In vitro cytotoxicity testing demonstrated that UA@MSN-UA nanoparticles prodrug exhibited higher proliferation inhibition, cell cycle arrest at the G2/M phase and significantly caused the early and late apoptosis in HepG2 cells, which would be contributed to high loading capacity, high cellular uptake and sustained release of UA. Overall, the UA-modified MSN prodrug delivery system can be a promising drug carrier for improving the bioavailability of UA, and further enhance its anti-cancer efficacy.

Temperature and magnetism bi-responsive molecularly imprinted polymers: Preparation, adsorption mechanism and properties as drug delivery system for sustained release of 5-fluorouracil

Longfei Li,Lin Chen,Huan Zhang,Yongzhen Yang,Xuguang Liu,Yongkang Chen

Temperature and magnetism bi-responsive molecularly imprinted polymers (TMMIPs) based on Fe 3 O 4 -encapsulating carbon nanospheres were prepared by free radical polymerization , and applied to selective adsorption and controlled release of 5-fluorouracil (5-FU) from an aqueous solution. Characterization results show that the as-synthesized TMMIPs have an average diameter of about 150   nm with a typical core–shell structure, and the thickness of the coating layer is approximately 50   nm. TMMIPs also displayed obvious magnetic properties and thermo-sensitivity. The adsorption results show that the prepared TMMIPs exhibit good adsorption capacity (up to 96.53   mg/g at 25   °C) and recognition towards 5-FU. The studies on 5-FU loading and release in vitro suggest that the release rate increases with increasing temperature. Meanwhile, adsorption mechanisms were explored by using a computational analysis to simulate the imprinted site towards 5-FU. The interaction energy between the imprinted site and 5-FU is −   112.24   kJ/mol, originating from a hydrogen bond , Van der Waals forces and a hydrophobic interaction between functional groups located on 5-FU and a NIPAM monomer. The electrostatic potential charges and population analysis results suggest that the imprinted site of 5-FU can be introduced on the surface of TMMIPs, confirming their selective adsorption behavior for 5-FU.
