Application of a Simple Microfluidic Chip Analysis Technology to Evaluate the Inhibitory Role of Protocatechuic Acid on Shear-Induced Platelet Aggregation

Cui He,Lihua Yu,Wenran Dan,Surong Deng,Haidong Ma,Beizhong Liu,Yuan Li

This study aimed to develop a simple microfluidic chip analysis technology to study the inhibitory effect of protocatechuic acid on shear-induced platelet aggregation. The microfluidic chip designed in this study simulates 80% fixed narrow microchannels. This microchannel narrow model uses the finite element analysis module of the three-dimensional modeling software solidwork to analyze fluid dynamic behavior. Blood treated with protocatechuic acid at 1, 2, 4, 8, or 16 µ g/mL was passed through the microchannel stenosis model at a shear rate of 10,000 s −1 . The platelet adhesion and aggregation behaviors were then measured using fluorescence microscopy and observed in real time. Simultaneously, the antiplatelet aggregation effect of protocatechuic acid was analyzed using thromboelastography and photoelectric turbidimetry. The designed stenosis model of the microfluidic chip can produce a gradient of fluid shear rate, and the gradient of fluid shear rate can induce platelet aggregation. Under this model, the degree of platelet adhesion and aggregation increased as the shear rate increased. In the experimental concentration range of 0–8 µ mol/mL, protocatechuic acid exerted a concentration-dependent inhibition of platelet aggregation. In contrast, thromboelastography and photoelectric turbidimetry failed to demonstrate an inhibitory effect. The microfluidic chip analysis technology developed in this study can be used to study the effect of protocatechin in inhibiting platelet aggregation induced by shear rate in vitro. This technology is simple to operate and can be used as a new type of antiplatelet aggregation analysis technology for screening studies of novel potential antiplatelet aggregation drugs.

Interfacial Regulation of ZIF-67 on Bacteria to Generate Bifunctional Sensing Material on Chip for Qualifying Cell-Released Reactive Oxygen Species

Zhuanzhuan Shi,Fang Shi,Yunpeng Li,Xiaoshuai Wu,Zhengyang Liu,Liang Liu,Qianqian Fu,Chang Ming Li,Chunxian Guo

Cell’s activities are highly dependent on signal molecules, of which reactive oxygen species of the superoxide anion (O2•–) and hydrogen peroxide (H2O2) are important ones that always work together to regulate biological processes such as apoptosis and oxidative stress. It is of significance to realize simultaneous qualification of O2•– and H2O2 but it still faces challenges particularly in live-cell assay with a complex environment. We report the design of a bifunctional sensing material by interfacially regulating ZIF-67 on bacteria Shewanella putrefaciens to generate cobalt nanoparticles/nitrogen-doped porous carbon nanorods (Co/N-doped CNRs) and its sensing chip for qualifying cell-released O2•– and H2O2. Co/N-doped CNRs exhibit unique properties including porous structure for significantly increased reaction surface area and coordinating Co nanoparticles for rich active sites. The bifunctional Co/N-doped CNRs is used to fabricate the electrochemical sensing chip, which achieves a fast response time (0.5 s for O2•–, 1.9 s for H2O2), a low detection limit (0.69 nM for O2•–, 2.25 μM for H2O2), and a remarkably high sensitivity (792.30 μA·μM–1·cm–2 for O2•–, 153.91 μA·mM–1·cm–2 for H2O2), among the best of reported bifunctional nanozymes.

Accurate molecular identification of different meat adulterations without carryover contaminations on a microarray chip PCR-directed microfluidic lateral flow strip device

Hanling Wang,Xianzhuo Meng,Li Yao,Qian Wu,Bangben Yao,Zhaoran Chen,Jianguo Xu,Wei Chen

Meat adulteration-based food fraud has recently become one of the global major economical, illegal, religious, and public health concerns. In this work, we developed a microarray chip polymerase chain reaction (PCR)-directed microfluidic lateral flow strip (LFS) device that facilitates the accurate and simultaneous identification of beef adulterated with chicken, duck, and pork, especially in processed beef products. To realize this goal, four pairs of amplification primers were designed and applied for specifically amplifying genomic DNA extracted from mixed meat powders in microarray chip. With the prominent advantage of this device lies in the flexible combination and integration of sample loading, detection, and reporting in microstructures, all the DNA amplicons can be individually visualized on the LFS unit, leading to the appearance of test lines (T C line, T D line, T P line, or T B line) as well as the control line (C line) for the species identification and quantification in beef products. Based on this new method, the adulterants were successfully distinguished and identified in mixtures down to 0.01% (wt.%) while the carryover aerogel contamination in routine molecular diagnostic laboratories was effectively avoided. The practicability, accuracy, and reliability of the device were further confirmed by using real-time PCR as a gold standard control on the successful identification of 50 processed ground meat samples sourced from local markets. The method and device proposed herein could be a useful tool for on-site identification of food authentication.

Design and fabrication of Zn@ZnO@Ag chip for Raman scattering analysis of norfloxacin in liquid milk, fish and animal feeds

Hanqiang Zhang,Xuejing Fan,Yangyue Ding,Lunzhao Yi,Kun Ge,Ying Gu

A novel and easy-to-prepare plasmonic nanoparticles doped semiconductor substrate—Zn@ZnO@Ag chip with ultra-high surface-enhanced Raman scattering (SERS) activity was fabricated for label-free, rapid and sensitive analysis of norfloxacin. The Zn@ZnO array was synthesized by surface oxidation at low temperature, followed by in-situ reduction to form leaf-like AgNPs on Zn@ZnO array without extra reducing agent, thus fabricating a Zn@ZnO@Ag chip. The ultra-high SERS activity is attributed to the synergistic effect of semiconductor characteristics of ZnO and surface plasmon resonance properties of leaf-like AgNPs. The possible enhancement mechanism was verified by density functional theory simulations. The proposed SERS method showed a wide linear range (3.0–500.0 μg/L) and low limit of detection (0.8 μg/L) for norfloxacin analysis. High sensitivity, good selectivity and acceptable recoveries (82.7–113.6%) in real sample analysis were obtained. This study offers a promising SERS chip-based platform for norfloxacin detection in the field.

Simultaneous amplification of DNA in a multiplex circular array shaped continuous flow PCR microfluidic chip for on-site detection of bacterial

Bo Yang,Ping Wang,Zhenqing Li,Qingxiang You,Shinichi Sekine,Junshan Ma,Songlin Zhuang,Dawei Zhang,Yoshinori Yamaguchi

Based on time to place conversion, continuous flow polymerase chain reaction (CF-PCR) can realize a rapid amplification of DNA by running the PCR reagent in a serpentine microchannel but a larger space is required for each sample, which greatly reduces the efficiency of the CF-PCR. Herein, we propose a multiplex circular array shaped CF-PCR microfluidic chip for on-site detection of bacteria. There were 12 serpentine microchannels which were distributed on the disc in an annular form, and each microchannel consisted of an inlet for sample injection, and an outlet for the detection of the PCR products based on fluorescence. Samples could be simultaneously driven into each inlet by a one-to-twelve diverter through a syringe. Moreover, the method of adding fluorescent dyes at the end of the microchannel can solve the inhibition effect of excessive fluorescent dyes on the PCR reaction. The process finished with simultaneous amplification of 12 different target genes from Porphyromonas gingivalis, Treponema denticola, Tannerella forsythia, and Escherichia coli, and on-site detection of their corresponding positives within 23 min. The fastest detectable PCR reaction time was 5.38 ± 0.2 min at a flow rate of 1 mL h−1. For E. coli, the minimum detectable concentration was 2.5 × 10−3 ng μL−1 in this microfluidic system. Such a system can increase the throughput of CF-PCR for point-of-care testing of pathogens.

On-Chip Precisely Controlled Preparation of Uniform Core–Shell Salmon Byproduct Protein/Polysaccharide Microcapsules for Enhancing Probiotic Survivability in Fruit Juice

Kuiyou Wang,Shasha Huang,Shanghua Xing,Shida Wu,Hongliang Li,Xu Zhong,Xin Na,Mingqian Tan,Wentao Su

The increasing demand for probiotic-fortified fruit juices stems from the dietary requirements of individuals with dairy allergies, lactose intolerance, and vegetarian diets. However, a notable obstacle arises from the degradation of probiotics in fruit juices due to their low pH levels and harsh gastrointestinal conditions. In response, this study proposes an innovative approach utilizing a microfluidic chip to create core–shell microcapsules that contain Lactobacillus plantarum Lp90. This method, based on internal–external gelation, forms highly uniform microcapsules that fully enclose the core, which consists of oil-in-water Pickering emulsions stabilized by salmon byproduct protein and sodium alginate. These emulsions remain stable for up to 72 h at a 1% sodium alginate concentration. The shell layer incorporates kelp nanocellulose and sodium alginate, thus improving the thermal properties. Furthermore, compared to free probiotics, the multilayer structure of the core–shell microcapsules provides a robust barrier, resulting in significantly enhanced probiotic stability. These findings introduce a novel strategy for augmenting probiotic delivery in functional fruit juice beverages, promising solutions to the challenges encountered during their development.

Anion Exchange membrane with High hydroxide ion conductivity and robust tensile strength fabricated from quaternary ammonia functionalized Pinus contorta, Dougl. Chip

Ming Lei,Qingtong Zhang,Mingchao Chi,Yuanyuan Yu,Hongrui Jiang,Shuangfei Wang,Douyong Min

The Pinus contorta, Dougl. chip was ammonianized by 3-chloro-2-hydroxypropyltrimethyl ammonium chloride. Subsequently, the functionalized chip was thermally compressed under 30 MPa at 100 °C for 72 h to form the quaternary ammonia functionalized membrane (QAFW). The results revealed that the hydroxide ion conductivity and ion exchange capacity (IEC) of the as-fabricated membrane were up to 15.2 mS cm −1 and 1.86 mmolg −1 at 25 °C under 100 % relative humidity respectively, which is higher than most of the quaternary ammonia-functionalized aromatic polymer membranes. The ratio of hydroxide ion conductivity to methanol permeability of the optimized QAFW membrane is up to 3.86 × 10 3 S s cm −3 . The decomposition temperature of QAFW membrane is ∼ 252 °C. The tensile strength of the wet QAFW membrane is as high as 60.02 MPa, which is significantly higher than that of the aromatic polymer membranes. Conclusively, a novel method herein characterized with environmentally friendly and economically attractive for the anion exchange membranes fabrication was presented.

Paper chip-based colorimetric assay for detection of Salmonella typhimurium by combining aptamer-modified Fe3O4@Ag nanoprobes and urease activity inhibition

Wei Shengnan,Li Juan,He Jingya,Zhao Wei,Wang Feng,Song Xiuling,Xu Kun,Wang Juan,Zhao Chao

A rapid and sensitive colorimetric assay is described for  Salmonella typhimurium ( S. typhimurium ) detection using urea/phenol red impregnated test paper. Aptamer-modified Fe 3 O 4 @Ag multifunctional hybrid nanoprobes (apt-Fe 3 O 4 @Ag NPs) were used to specifically captured S. typhimurium ; the nanoprobes were quickly etched by H 2 O 2 to form Ag + . The generated Ag + can inhibit the urease-catalyzed hydrolysis reaction of urea to produce NH 4 + . Consequently, the as-prepared test paper displayed a yellow color. In the presence of  S. typhimurium , the target bacteria can cause aggregation of apt-Fe 3 O 4 @Ag NPs, and the deposited Ag on the nanoprobe’s surface is shielded against H 2 O 2 -induced oxidative decomposition leading to reduced Ag + production. The catalytic activity of urease cannot be inhibited completely by inadequate amount of Ag + . An obvious color change from yellow to pink can be monitored directly using our test paper as a result of increased NH 4 + . The entire assay procedure could be completed within 1 h. A limit of detection of 48 cfu/mL is achieved with a linear range of 1 × 10 2 to 1 × 10 6  cfu/mL. The recoveries of  S. typhimurium spiked in pure milk samples were 92.48–94.05%. Schematic diagram of the proposed colorimetric assay for S. typhimurium detection based on etching of bifunctional apt-Fe 3 O 4 @Ag NPs and inhibiting catalytic activity of urease by Ag + . A color change from yellow to pink can be observed and correlated to the concentration of S. typhimurium.

Colloidal photonic crystal array chip based on nanoparticle self-assembly on patterned hydrophobic surface for signal-enhanced fluorescent assay of adenosine

Guo Rui,Wang Dan-Ni,Wei Yun-Yun,Zhang Ying-Zhi,Yang Chun-Guang,Xu Zhang-Run

A controllable approach for preparing a portable colloidal photonic crystal (CPC) array chip is presented. The approach was inspired by the confinement effect of nanoparticle self-assembly on patterned surface. Hydrophobic polydimethylsiloxane substrate with reproducible micro-region array was fabricated by soft-lithography. The substrate was employed as the patterned template for self-assembly of monodisperse polystyrene nanoparticles. The CPC units can be prepared in several minutes, and exhibit consistent reflection wavelength. By adjusting the size of polystyrene nanoparticles and the shape of micro-regions, CPC units with multiple structure, colors and geometries were obtained. The CPC array chip features fluorescence enhancement owing to the optical modulation capability of the periodic nanostructure of the self-assembled CPC. With the reflection wavelength (523 nm) of green CPC units overlapping the emission wavelength (520 nm, with excitation wavelength of 490 nm) of 6-carboxyfluorescein-labeled DNA probe, the fluorescence intensity increased more than 10-fold. For signal-amplified assay of adenosine, the concentration range of linear response was 5.0 × 10 −5  mol L −1 to 1.0 × 10 −3  mol L −1 , and the limit of detection was 1.3 × 10 −6  mol L −1 . Because of the enhancement effect of photonic crystal, the fluorescence images were more readable from the CPC array chip, compared with those from the planar substrate. The chip has potential applications in multiplex determination with high-throughput via encoding strategy based on the tunable structure, color or geometric shape. Schematic diagram of signal-enhanced fluorescent detection of adenosine based on the colloidal photonic crystal array chip (PDMS, polydimethylsiloxane; PS NPs, polystyrene nanoparticles; CPC, colloidal photonic crystal; GO, graphene oxide; FAM, 6-carboxyfluorescein).

A microfluidic chip based ratiometric aptasensor for antibiotic detection in foods using stir bar assisted sorptive extraction and rolling circle amplification

Liyong He,Zhipeng Shen,Yuting Cao,Tianhua Li,Dazhen Wu,Youren Dong,Ning Gan

A ratiometric and sensitive microfluidic chip based aptasensor was developed for antibiotic detection with kanamycin (Kana) as a model analyte. A novel stir bar assisted sorptive extraction and rolling circle amplification strategy was designed to largely amplify the signal and overcome complex matrix interference in food samples. The detection mechanism was as follows: firstly, many duplex DNA probes (a single-stranded DNA as a primer hybrid with an aptamer sequence) were modified on a stir bar. In the presence of Kana, the probes on the bar could specifically capture Kana and release the primer to trigger RCA in the presence of a circular DNA template (CDT). As the reaction proceeds, the amount of CDT decreased and the number of RCA products increased. It is worth mentioning that they can be efficiently separated and detected using a microfluidic chip. The signal ratio of RCA products and CDT (IR/IC) can be employed to qualify Kana in a wide linear range from 0.8 pg mL−1 to 10 ng mL−1 with a low detection limit of 0.3 pg mL−1. This method exhibited excellent sensitivity and selectivity and can obviously reduce the matrix interference through a ratiometric strategy combined with stir bar extraction. The aptasensor was successfully tested in milk and fish samples, confirming that it can be applied for on-site quantitation of antibiotic residues in foods.

An efficient method for CTCs screening with excellent operability by integrating Parsortix™-like cell separation chip and selective size amplification

Jin Xin,Chen Rui,Zhao Shikun,Li Peiyong,Xue Bai,Chen Xiang,Zhu Xinyuan

In this article, an attempt for efficient screening of circulating tumor cells (CTCs) with excellent operability on microfluidic chips was reported. A Parsortix™-like cell separation chip was manufactured in our lab. This chip allowed lateral flow of fluid which increased the flow rate of blood. And, an air valve controlled injection pump was manufactured which allowed eight chips working simultaneously. This greatly facilitated the blood treatment process and saved time. As for the mechanism of screening circulating tumor cells, selective size amplification was utilized. By size amplification of cancer cells, both the hardness and the size of CTCs increased which differentiated them from blood cells. And the modification procedure of beads used for size amplification of cancer cells was optimized. Finally, by integrating the commercialized Parsortix™-like cell separation chip and selective size amplification, a practical method for screening circulating tumor cells was established.

THz Spectroscopy for a Rapid and Label-Free Cell Viability Assay in a Microfluidic Chip Based on an Optical Clearing Agent

Ke Yang,Xiang Yang,Xiang Zhao,Marc Lamy de la Chapelle,Weiling Fu

Simple, rapid, and efficient cell viability assays play a fundamental role in much of biomedical research, including cell toxicology investigations and antitumor drug screening. Here, we demonstrate for the first time a rapid and label-free cell viability assay using THz spectroscopy in combination with a new optical clearing agent (OCA) and microfluidic technology. This strategy uses a considerably less absorptive OCA to replace the highly absorptive water molecules around the living cells and thus to decrease the background signal interference. Three low-viscosity oils were screened as potential OCA candidates, among which fluorinated oil was selected because of its lower absorption and lowest cytotoxicity. After the liquid medium was replaced with fluorinated oil in a microfluidic chip, an obvious THz spectral difference was observed between the fluorinated oils with and without living cells. This change in THz response was preliminarily attributed to the distinguishable signals between the cells and the fluorinated oil. In addition, we applied this method to cell viability assays of human breast cancer cells (MDA-MB-231) after treatment with different antitumor drugs. The results indicated that THz spectroscopy with the aid of the proposed water-replacement strategy presented excellent quantification of cell viability with the advantages of a rapid, label-free, nondestructive microassay, which offers significant potential to developing a convenient and practical cell analysis platform.
