Gelatin-nanocellulose stabilized emulsion-filled hydrogel beads loaded with curcumin: Preparation, encapsulation and release behavior

Siyi Liu,Yuxi Wang,Yue Huang,Mengtao Hu,Xiangxiang Lv,Yuhao Zhang,Hongjie Dai

In this study, the curcumin was firstly encapsulated in gelatin (GLT) and/or cellulose nanocrystals (CNC) stabilized emulsions, then further mixed with sodium alginate (SA) to form emulsion-filled hydrogel beads loaded with curcumin (Cur). The Cur-loaded emulsions showed a droplet size of 20.3–24.4 μm with a uniform distribution. Introducing CNC and/or SA increased the viscosity of emulsions accompanied by viscoelastic transition, while the modulus was reduced due to destruction of GLT gel. Cur was doubly immobilized in the hydrogel beads with >90 % of encapsulation efficiency. The results of simulated gastrointestinal tract experiments revealed that the beads possessed a good pH sensitivity and controlled release behavior to prolong the retention of Cur in the gastrointestinal tract. After 6 h of UV irradiation, the Cur-loaded emulsion-filled hydrogel beads showed a higher antioxidant activity than that of pure Cur, effectively delaying the photodegradation of Cur. In addition, the beads had better stability in aqueous and acidic environments, which was favorable for prolonging the release of Cur. These results suggest that the emulsion-filled hydrogel beads have great potential for the delivery of lipophilic bioactive molecules.

Jie Wang,Xuemei Gu,Xia Gao,Jing Chen,Zhiyang Lv,Siyu Zhang,Siyu Ni,Fei Shi,Xialin Chen,Liang Cao,Zhenzhong Wang,Wei Xiao

In order to enhance the therapeutic value of curcumin in liver cancer treatment, glycyrrhetinic acid-modified pH-sensitive curcumin liposomes (GA-pH-Lip@Cur) was developed.GA-pH-Lip@Cur was prepared using a thin film dispersion ultrasonication method, and the optimal formulation process was selected through single-factor experiments and a Box-Behnken design-response surface methodology. The liposomes were evaluated for their morphological appearance, particle size, in vitro release at different pH levels, and biocompatibility. The anti-tumor effect of GA-pH-Lip@Cur was assessed using cell viability assays (CCK-8). The in vivo hepatic targeting and anti-liver tumor efficacy of GA-pH-Lip@Cur were evaluated through pharmacokinetic and pharmacological experiments. The results indicated that optimized GA-pH-Lip@Cur exhibited uniform particle size distribution, good stability, pH-sensitive in vitro release with sustained behavior. Compared to conventional liposomes, GA-pH-Lip@Cur showed prolonged average retention time in vivo and significantly increased curcumin distribution in liver tissues, indicating excellent liver targeting. Both in vitro and in vivo evaluations demonstrated the effectiveness of GA-pH-Lip@Cur in inhibiting liver cancer cell proliferation and suppressing liver tumor growth in tumor-bearing mice. In conclusion, GA-pH-Lip@Cur, by leveraging the acidic tumor microenvironment and overexpression of glycyrrhetinic acid receptors in liver cells, encapsulates curcumin to improve its bioavailability, and target its delivery to the liver tumor sites.

Iron–Curcumin–Polyvinylpyrrolidone Nanoparticles With Dynamic Disassembly and Elimination Abilities for Photothermal/Chemo Combinatorial Therapy

Yue Cao,Yanchen Wang,Xin Zhang,Bo Ren,Bo Wang,Xiaodong Yang

Due to the heterogeneity of the tumor, it is difficult to achieve complete ablation of the tumor by single photothermal treatment. Moreover, their broader use is constrained by the intrinsic high uptake in the reticuloendothelial system (RES) organs, resulting in slow elimination from the body and potential toxicity. In this study, we report a single-step synthetic approach to fabricate versatile FeCP-NPs, leveraging the coordination bonds formed between bioactive polyphenolic compounds (curcumin), Fe III ions, and polyvinyl pyrrolidone (PVP). The FeCP-NPs possess the following favorable characteristics: (1) The components of FeCP-NPs originate from natural products, an endogenous element, and an FDA-approved polymer, guaranteeing good biosafety. (2) FeCP-NPs exhibit excellent photothermal properties. (3) After the photothermal treatment is completed, the dynamic disassembly strategy triggered by deferoxamine mesylate (DFO) can induce the dissociation of nanoparticles and achieve rapid clearance in vivo. (4) Curcumin released during the disassembly of FeCP-NPs can be combined with photothermal therapy to achieve complete tumor ablation. A comprehensive analysis of the safety, biodistribution, and clearance patterns of FeCP-NPs is conducted in vitro or in vivo. This work provides a nanoparticle with great clinical translational potential and provides a new solution for inhibiting tumor growth.

Dextran sulfate-coated curcumin nanocrystals for the treatment of DSS-induced ulcerative colitis in mice

Yunmei Li,Huilin Ma,Huan Shi,Biaobiao Wang,Desheng Li,He Tian,Xifan Mei,Chao Wu

Ulcerative colitis is an inflammatory disease that primarily involves intestinal inflammation and epithelial damage. The nano-targeted drug delivery system delivers drugs to the disease site, exerting effects such as inhibiting inflammatory response and reducing reactive oxygen species expression, thereby promoting recovery from ulcerative colitis. In this experiment, dextran sulfate-coated curcumin nanocrystals (NBD) were prepared for the oral treatment of ulcerative colitis (UC). NBD not only significantly enhances the water solubility and stability of curcumin but also possesses the ability of sustained release and targeting inflammatory macrophages. The sustained release effect of NBD was demonstrated by in vitro release experiments. In simulated gastric fluid, the cumulative release amount of NBD at 2 h was 21.99 ± 1.93 %, while in simulated colonic fluid, the cumulative release amount of NBD at 12 h was 84.98 ± 2.02 %. The ability of NBD to target inflammatory macrophages was verified through the transwell system, rat one-way intestinal perfusion experiment and in vivo imaging system. The in vitro and in vivo (mice) anti-inflammatory and antioxidant capacities of NBD were validated using immunofluorescence experiment, ELISA kits and reactive oxygen species-related detection kits. The results indicated that NBD could reduce inflammatory responses, promote macrophage polarization and inhibit oxidative stress. In addition, the therapeutic effect of NBD was further confirmed in this experiment by the clostridium perfringens-induced necrotizing enteritis model in chickens. In conclusion, NBD might be a potential pharmaceutical preparation for the treatment of UC.

Black Phosphorus Nanosheet-Loaded Traditional Chinese Medicine Curcumin for Acute Rhinosinusitis Therapy and Its Mechanism

Yongze Liu,Lei He,Dingding Liu,Xuefeng Jiang,Feng Chen,Xiaofeng Ma,Saijie Song,Jun Zhang,Jian Shen,Xia Gao

Acute rhinosinusitis is among the most prevalent disease across the world. Recently, researchers have found that traditional Chinese medicine (TCM) can effectively treat acute rhinosinusitis. Nanomaterial-based drug delivery systems (DDSs) have attracted widespread attention in disease treatment. In this work, we herein reported curcumin-loading black phosphorus nanosheet (BP-C@PEG) nanodrugs for acute rhinosinusitis therapy. The as-prepared BP-C@PEG showed excellent solubility and stability in aqueous solution and performed sustained release behavior. In addition, it also exhibited good anti-inflammatory action for the treatment of acute rhinosinusitis. The anti-inflammation mechanism of BP-C@PEG was also studied. We found that LPS-induced inflammation in nasal epithelial cells was eased through downregulating HDAC3 and NLRP3 inflammasomes and lowering IL-1β levels. Moreover, the in vivo experiments verified that BP-C@PEG could effectively alleviate the cilium injury and the recruitment of inflammatory cells in the nasal mucosa of mice, further demonstrating its promising therapeutic efficiency in acute rhinosinusitis. Overall, this study provides a potential approach for acute rhinosinusitis therapy by using BP-C@PEG nanodrugs, which may broaden the application of TCM in the biomedical field.

Multiple stimuli-responsive circularly polarized luminescent bio-composite films by coassembling cellulose nanocrystals and curcumin

Shaoyi Cao,Jingpeng Wang,Wenye Sun,Bang An,Mingcong Xu,Sha Luo,Chunhui Ma,Haipeng Yu,Zhijun Chen,Jian Li,Wei Li,Shouxin Liu

Cellulose nanocrystal (CNC) -based circularly polarized luminescent (CPL) materials, which are responsive to external stimuli, have attracted increasing attention in developing smart chiral photonic materials. In this study, chiral nematic bio-composite CNC films with right-handed (R-) CPL emission and tunable dissymmetry factors (g lum ) were prepared by encapsulating curcumin in chiral nematic CNC films via an evaporation-induced self-assembly strategy. The CPL active bio-composite CNC films exhibit multiple responsiveness to relative humidity (RH) and pH. It is noted that the pH response of the composite films is visualized, which is reflected in the variation of film colors, fluorescence, and CPL emission wavelengths. Additionally, with a hydrophobic treatment, the bio-composite CNC films exhibit enhanced water resistance and pH-responsive CPL in acid/base aqueous solutions. Based on the responsive circular polarization information, the bio-composite CNC films were developed as optical labels for multiple anti-counterfeiting applications. The reported environmentally friendly bio-composite CNC films provide a new reference for utilizing natural polysaccharides to build multi-mode responsive CPL materials.

Mitochondria-targeted nanoparticles based on glycated oat protein for enhanced curcumin bioavailability and antioxidant activity

Lei Zhong,Jiangrong Tian,Qiuhui Hu,Liyan Zhao,Qiping Zhan,Mingwen Zhao

Curcumin is a hydrophobic dietary supplement and can mitigate oxidative damage. However, the low bioavailability of curcumin impedes its application in functional foods. Protein-coated nanoparticles are susceptible to environmental stress and have limited ability to reach mitochondria, where reactive oxygen species are generated. To overcome this limitation, we prepared mitochondria-targeted nanoparticles by modifying glycosylated oat protein isolate with triphenylphosphonium bromide for the efficient delivery of curcumin. In this study, the curcumin was incorporated into the spherical nanoparticles with high entrapment efficiency and loading capacity. The nanoparticles increased the stability of curcumin during heat treatment and light irradiation. The bioavailability of curcumin was improved in the nanoparticles due to their strong resistance against in vitro gastric digestion and good permeability across the Caco-2 cell monolayer. The nanoparticles realized lysosomal escape and exhibited specific accumulation in mitochondria upon internalization by Caco-2 cells. The mitochondria-targeted nanoparticles with no cytotoxicity further enhanced the antioxidant activity of curcumin by preserving mitochondrial membrane potential, increasing antioxidant enzyme activities, decreasing the concentrations of intracellular reactive oxygen species and malondialdehyde. This study highlights the potential application of glycosylated proteins in developing nanocarriers for precise delivery of hydrophobic nutrients.

Novel amphiphilic rhamnogalacturonnan I-based nanomicelles for targeted delivery of curcumin to hepatocellular carcinoma cells

Yuejiao Wu,Jian Yang,Ke Shen,Liyan Liu,Jing Cao,Zhaomei Wang

Curcumin (Cur) is a bioactive nutraceutical with great potential in biological, nutritional, and medical applications. However, these applications are limited by various factors such as insufficient ingestion, low aqueous solubility, and relatively high toxicity to normal cells. To tackle these obstacles, we synthesized a novel Cur-modified-rhamnogalacturonan (RG-C) nano-micelle carrier to target-deliver Cur to hepatocellular carcinoma HepG2 cells via specific recognition of RG-I by the overexpressed surface galactin-3 receptor. Fourier transfer infrared, UV–vis, and 1 H NMR analyses confirmed the conjugation between RG and Cur RG-C loaded with Cur (RG-CC) was formed via self-assembly in an aqueous solution with a drug loading efficiency of 12.2%. RG-CC micelle was ellipsoidal or cubic with a size ranging between 100 and 200 nm by scanning electron microscopy observation. Cur release from RG-CC exhibited a controlled and pH-dependent manner with 50% at pH 5.0 in contrast to 5% at pH 7.4 after 24 h exposure. RG-CC possessed more potent anti-proliferative activity against HepG2 cells than normal embryonic kidney 293T cells. Compared to free Cur, both the anti-proliferative effect and uptake of RG-CC were significantly higher in HepG2 cells as revealed from laser confocal microscopy and flow cytometry analyses. RG-CC was a promising anticancer candidate and deserves further preclinical and clinical investigations.

pH-Sensitive Zinc Oxide Nanoparticle-Controlled Sodium Alginate/Silica Hydrogel Beads: for Controlled Curcumin Release

Xueze Zhang,Yu Xiao,Shuqi Guo,Huifang Wang,Xianghua Gao,Baolong Niu,Wenfeng Li

This study designed a pH-sensitive carrier regulated by zinc oxide nanoparticles for the delivery of the hydrophobic drug curcumin (Cur). Carboxymethyl cellulose (CMC) was used as a stabilizer and dispersant to synthesize CMC-ZnO nanoparticles via an in situ reaction. These nanoparticles were then mixed with sodium alginate (SA) and immersed in a tetraethyl silicate (TEOS) hydrolysis solution to form sodium alginate/silica-zinc oxide (Ss/CMC-ZnO) composite hydrogel beads. XRD, FTIR, SEM, and UV-vis analyses confirmed the successful synthesis and incorporation of ZnO nanoparticles into the hydrogel beads. Swelling experiments demonstrated that as the concentration of CMC-ZnO increased, when the TEOS molar concentration was 0.205 M, the maximum swelling ratio decreased from 39.83 to 14.03, with the time to reach the maximum swelling ratio extending from 4 to 6 hours. At a TEOS concentration of 0.375 M, the swelling ratio decreased from 26.77 to 12.10, with the swelling time extending from 6 to 7 hours. Hemolysis and cytotoxicity tests confirmed the hydrogel beads' good biocompatibility. Cur was incorporated using a blending method, and in vitro release experiments revealed that the Ss/CMC-ZnO hydrogel beads exhibited excellent pH sensitivity. The addition of CMC-ZnO NPs effectively prolonged the release time of curcumin, indicating potential for applications in controlled drug delivery.

Preparation and Characterization of Silica-Coated Sodium Alginate Hydrogel Beads and the Delivery of Curcumin

Yu Xiao,Lu Wang,Xueze Zhang,Yi Ren,Jianhong Wang,Baolong Niu,Wenfeng Li

In this study, to address the defects of sodium alginate (SA), such as its susceptibility to disintegration, silica was coated on the outer layer of sodium alginate hydrogel beads in order to improve its swelling and slow-release properties. Tetraethyl orthosilicate (TEOS) was used as the hydrolyzed precursor, and the solution of silica precursor was prepared by sol-gel reaction under acidic conditions. Then SA–silica hydrogel beads prepared by ionic crosslinking method were immersed into the SiO2 precursor solution to prepare SA–silica hydrogel beads. The chemical structure and morphology of the hydrogel beads were characterized by XRD, FTIR, and SEM, and the results showed that the surface of SA–silica beads was successfully encapsulated with the outer layer of SiO2, and the surface was smooth and dense. The swelling experiments showed that the swelling performance effectively decreased with the increase of TEOS molar concentration, and the maximum swelling ratio of the hydrogel beads decreased from 41.07 to 14.3, and the time to reach the maximum swelling ratio was prolonged from 4 h to 8 h. The sustained-release experiments showed that the SA–silica hydrogel beads possessed a good pH sensitivity, and the time of sustained-release was significantly prolonged in vitro. Hemolysis and cytotoxicity experiments showed that the SA-silica hydrogel beads were biocompatible when the TEOS molar concentration was lower than 0.375 M. The SA–silica-2 hydrogel beads had good biocompatibility, swelling properties, and slow-release properties at the same time.

Novel Halogenated Curcumin-Mediated Photodynamic Inactivation for the Preservation of Small Yellow Croaker (Larimichthys polyactis)

Long Jiao,Yuwei Li,Mingyu Tian,Shuyi Zhao,Xiaoye Zhang,Soottawat Benjakul,Bin Zhang

A novel class of halogenated curcumin, X-Cur (X = F, Cl, or Br), was synthesized, and its photosensitivity was evaluated. The results showed that Br-Cur with the highest singlet oxygen (1O2) generation capacity exhibited a better photodynamic inactivation (PDI) effect on the small yellow croaker (Larimichthys polyactis) than curcumin. This was attributed to the heavy atom effect of Br, which resulted in Br-Cur having the smallest singlet–triplet energy difference ΔEst(S1-T3) (0.140 eV) and the largest spin–orbit coupling value (0.642262 cm–1). When L. polyactis was treated with 0.025 wt % Br-Cur and exposed to blue LED irradiation (450 nm, 20 mW/cm2) for 20 min, the increase in the total volatile basic nitrogen content (28.23 ± 2.38 mg/100 g on day 6), pH, and total viable count (6.13 ± 0.06 log CFU/g on day 6) could be effectively controlled. Accordingly, Br-Cur is a promising photosensitizer for PDI preservation.
