Recently, two-dimensional layered transition metal dichalcogenide materials have been very fascinating in electrochemical fields due to their unique electronic and electrochemical properties. In the present work, a novel VS2–graphene (VS2–GR) composite is firstly prepared by a facile one-step hydrothermal method. A sensitive, universal and label-free electrochemical aptamer-based sensor (aptasensor) is then designed to detect the platelet-derived growth factor BB (PDGF-BB) based on VS2–GR coupled with Exo III-aided autocatalytic signal amplification. In the absence of PDGE-BB, the aptamer hybridized with the complementary DNA (cDNA) and the single-strand signal DNA labeled with biotin at 5′ end cannot be cleaved by the Exo III. When the above mixture is applied on the tetrahedron-structured probe (T-DNA) modified AuNP/VS2–GR electrode, signal DNA hybridizes with the protruding of T-DNA and a large number of avidin-HRP are adsorbed on the modified electrode by the specific binding of avidin–biotin, which leads to a strong current response by the catalysis of HRP to the mixture of hydrogen peroxide and hydroquinone (H2O2 + HQ). However, in the presence of PDGF-BB, the aptamer is preferentially associated with PDGF-BB, which facilitates the formation of double-strand DNA between cDNA and signal DNA. The duplex DNA is digested by Exo III from the 3′ blunt terminus of signal DNA and the cDNA is released. The liberative cDNA then reacts with the remaining signal DNA in the solution to perform a new cleavage process. Finally, the cyclic hybridization–hydrolysis process leads to a great decrease of biotin on the electrode, thus reducing the quantity of signal molecule HRP and causing an obvious decline of current response. This proposed protocol exhibits high specificity, good sensitivity, and good stability for PDGF-BB detection. Moreover, the aptasensor can be applied as a universal assay for the detection of various targets including proteins, DNAs, and metal ions.

Human Immunodeficiency Virus (HIV) compromises the human immune system and remains one of the deadliest infectious diseases globally. Nucleic acid testing is a powerful tool for the early detection of HIV infection, capable of real-time HIV DNA detection without the limitation of a window period. In this study, a DNA nanostructure-based biosensor that leveraged interactions between functionalized gold nanoparticles (AuNPs) and three-dimensional (3D) DNA walkers were developed for ultrasensitive fluorescence detection of HIV DNA. The biosensor comprised blocked-walking particles (BPs) with locked DNAzyme and substrate particles (SPs) with quenched carboxyfluorescein (FAM) fluorophores. Upon encountering HIV DNA, the blocking strands hybridized more strongly with the HIV DNA, exposing the DNAzyme, which subsequently cleaved FAM on the SPs from the AuNPs and restoring fluorescence. As all substrate strands on SPs were cleaved, the BPs interacted with new SPs, significantly amplifying the fluorescent signal. The fluorescence intensity showed a linear correlation with HIV DNA concentration ranging from 0.02 to 6.00 nM, with a detection limit of 12.8 pM. The performance of biosensor was evaluated using spiked serum samples, demonstrating good precision and recovery rates. The DNA walker biosensor leveraged particle-to-particle interactions for rapid signal amplification, offering great potential for sensitive biosensors.

DNA nanostructures have attracted great attention due to their precisely controllable geometry and great potential in various areas including bottom-up self-assembly. However, construction of higher-order DNA nanoarchitectures with individual DNA nanostructures is often hampered with the purity and quantity of these “bricks”. Here, we introduced size exclusion chromatography (SEC) to prepare highly purified tetrahedral DNA nanocages in large scale and demonstrated that precise quantification of DNA nanocages was the key to the formation of higher-order DNA nanoarchitectures. We successfully purified a series of DNA nanocages with different sizes, including seven DNA tetrahedra with different edge lengths (7, 10, 13, 17, 20, 26, 30 bp) and one trigonal bipyramid with a 20-bp edge. These highly purified and aggregation-free DNA nanocages could be self-assembled into higher-order DNA nanoarchitectures with extraordinarily high yields (98% for dimer and 95% for trimer). As a comparison, unpurified DNA nanocages resulted in low yield of 14% for dimer and 12% for trimer, respectively. AFM images cleraly presented the characteristic structure of monomer, dimer and trimer, impling the purified DNA nanocages well-formed the designed nanoarchitectures. Therefore, we have demonstrated that highly purified DNA nanocages are excellent “bricks” for DNA nanotechnology and show great potential in various applications of DNA nanomaterials.

A self-powered microRNA (miRNA) biosensing device is fabricated with high-energy enzymatic biofuel cell (EBFC) and cruciform DNA (cDNA). Sulfur-selenium co-doped graphene/gold nanoparticles (S–Se-GR/AuNPs) is synthesized and used as supporting substrate of biocathode and bioanode . The glucose oxidase (GOD) molecules are bonded to carboxyl-functionalized AuNPs through the condensation reaction between the amino groups in enzyme and carboxyl groups on the AuNPs to form the bioanode. The ultra-thin porous carbon shell/AuNPs-complementary strand of cDNA (UPCS/AuNPs-cDNA) is meticulously designed. The potassium ferricyanides are then inserted in mesoporous UPCS/AuNPs as the biocathode electron acceptor , and the cruciform DNA bioconjugate is synthesized as signal amplifier. When target miRNA is added, the hybridization reaction happens between miRNA and capture probe DNA on the biocathode to open the capture probe DNA chain. Cruciform DNA bioconjugate is immobilized onto the biocathode through base pairing with the capture DNA on the biocathode, which can release electron acceptor [Fe(CN) 6 ] 3- , resulting in the dramatically increase of the open circuit voltage of the EBFCs. The self-powered biosensor responds linearly in the miRNA level range of 0.5–10000 fM with a detection limit of 1.5 × 10 −16 mol L −1 . Detection of miRNA in spiked serum samples is also realized with the self-powered biosensor, demonstrating its great potential in the clinical applications.

A sensitive dual-target electrochemical biosensor was developed integrating circular bipedal DNA walkers with a three-dimensional hollow carbon nanospheres-tetrametallic nanoparticle (3D HCNs-TT NPs) modified platform, enabling concurrent detection of two critical epidermal growth factor receptor (EGFR) exon 19 mutations: E746_A750 deletion (Mut DNA A) and L747_S752delinsS (Mut DNA B). The circular bipedal DNA walkers address the limitations of low amplification efficiency in traditional single-legged DNA walkers and mitigate the sensitivity reduction caused by excessively long walking strands. Additionally, we used the 3D HCNs-TT NPs nanocomposite materials to modify the electrodes, which significantly increased the electron transfer rate and provided more active sites for molecular probe loading, thereby further enhancing the performance of the detection platform. This biosensor demonstrated good performance, with detection limits as low as 3.4 fM (Mut DNA A) and 2 fM (Mut DNA B) (S/N = 3). The designed sensor was applied for detection using clinical samples, including cells with gene mutations and diluted human serum, and these results confirm the sensor’s potential for clinical translation, offering a potential tool for early non-small cell lung cancer (NSCLC) diagnosis and personalized tyrosine kinase inhibitors (TKI) therapy guidance. In order to meet the demand of analyzing low concentrations of DNA with high sensitivity, herein, a magnetic separated-type fluorescent biosensor has been constructed for ultrasensitive DNA assay based on chemically driven redox-cycling system. In this study, the substrate o-phenylenediamine dihydrochlorlde (OPD) could be oxidized to produce 2, 3-diaminophenazine (DAP) with strong fluorescence emission at 555 nm. In the chemically driven redox-cycling system, Cu2+ oxidized OPD to yield DAP and Cu+/Cu0. On one hand, the produced Cu0 could further oxidize OPD to form DAP. But on the other, the generated Cu+ could react with newly imported H2O2 to undergo a Fenton-like reaction to form hydroxyl radicals (•OH) and Cu2+ ion. The reproduced Cu2+ ion and generated •OH could in turn oxidize OPD to form more DAP, which touching off a chemically driven chemical redox-cycling amplification reaction. By leveraging of the magnetic separation-mode, the chemically driven redox-cycling strategy allowed quantitative detection of H1N1, exhibiting a linear ranging from 1 pM to 1 nM with a low limit of detection of 300 fM. The magnetic separation-mode could effectively minimize the background signal to obtain greatly improved sensitivity. It’s promising that the chemically driven redox-cycling strategy would provide a novel way in the fields of biosensing and biological samples assay.

Maximizing DNA loading on magnetic nanoparticles (MNPs) is crucial for their successful utilization in gene transfer, DNA isolation, and bio-analytical applications. This enhancement is typically achieved by altering particle size and surfaces as well as charge density and ionic strength. We demonstrate a novel route for promoting DNA loading on amino-modified silica-coated magnetic nanoparticles (ASMNPs) by prior condensation of elongated DNA to a compact globule before adsorption. The enhanced DNA-loading capacity, as demonstrated by a reduction in the number of ASMNPs needed to achieve complexation, was presumably due to the elimination of DNA wrapping around nanoparticles and substantially reduced electrostatic interactions of DNA with nanoparticles because the compacted DNA globule conformation decreases its exposed surface charge. The maximum loading capacity of ASMNPs for condensed DNA was 4.4 times greater than that for elongated coiled DNA, achieving the highest ever reported value of 385 μg mg −1 . Practical applications for plasmid DNA isolation from cleared lysate confirmed the reliability of the proposed method.

Amyloid-β oligomers (AβOs) was the core-biomarker of Alzheimer's disease (AD), and the detection of AβOs is very important for the early diagnosis of AD. However, existing tests for AβOs are majorly suffering from complex process and poor sensitivity. Thus, an adjustable AβOs electrochemiluminescence (ECL) aptasensor based on the synergistic effect of self-enhanced metal-organic gel (AgCNS) and triple-helix DNA system (THS) was successfully constructed. AgCNS was prepared by an extremely simple one-pot method and was an innovative luminophore with excellent ECL performance. The AgCNS-labeled complementary sequence ( [email protected] ) was interlaced with the unlabeled aptamer (Apt) carrying two short-arms fixed on the gold electrode (GE) to form the THS. Along with the specific-binding of AβOs and Apt, the THS was disrupted and adjusted flexibly between “on” and “off”, resulting in significant changes in the ECL signals. Thus, ECL detection of AβOs was sensitively achieved with a detection limit as low as 0.23 fM and the different forms of Aβ can be specifically distinguished. The aptasensor also exhibited satisfactory selectivity, stability and reproducibility. Moreover, when proposed method and ELISA-kit were simultaneously applied to artificial cerebrospinal fluid (A-CSF) samples, the obtained results were completely consistent, reflecting the potential clinical application value of this work.

Aristolochic acids are one of the major compounds in aristolochia plants, which are nephrotoxic and carcinogenic. A method was established for the detection and identification of aristolochic acids and their DNA adducts in four different herbs using ultra-high performance liquid chromatography-ion mobility quadrupole time-of flight mass spectrometry. Solid phase extraction conditions were optimized to improve the sensitivity of the experiment by using 40 mg of C 18 as adsorbent and 100 μL ethanol as elution solvent. At a collision energy of 10-40 eV, these compounds and cleavage patterns were precisely identified and analyzed by secondary fragmentation and collision cross section values. The obtained mass spectrometry data were then analyzed by targeted metabolomics, including principal component analysis, partial least squares-discriminant analysis and hierarchical clustering analysis, and importing the samples in the established model, the confidence values can reach 0.61 and 0.76. All in all, this method can provide a useful tool for the detection of aristolochic acids and deoxyribonucleic acid adducts. In conclusion, this method was successfully used for the detection and identification of aristolochic acids and their DNA adducts.

Acute liver failure (ALF) involves extensive necrosis of liver cells and severe impairment of liver function. Hepatocyte transplantation holds promise for treating ALF by swiftly supporting liver functions and promoting liver regeneration. However, the scarcity of suitable cell sources requires strategies to obtain enough functional hepatocyte-like cells (HLCs) and optimize their in vivo transplantation efficiency. A DNA hexahedral nanostructure (DHN) is developed loaded with microRNA-122 to efficiently induce hepatic differentiation of human adipose-derived mesenchymal stem cells into HLCs. These HLCs can serve as alternative hepatocyte sources, as confirmed by expression of liver-specific genes and proteins, and the restoration of liver functions. To enhance in vivo survival efficiency of HLCs, a versatile scaffold is also created by 3D printing the calcium-cross-linked mixture bioink composed of sodium alginate, gelatin, and silk fibroin with excellent ROS scavenging capabilities. The scaffold is infused with chitosan-DHN hydrogel containing HLCs for tissue engineering orthotopic transplantation in CCl 4 -induced ALF mice. The transplanted composite scaffold-HLCs successfully repair tissue necrosis in the liver injury area of mice and regulate expressions of genes and proteins associated with inflammation, oxidative stress, and hepatocyte function. Collectively, this study offers a novel approach and strategy for identifying alternative hepatocyte sources and treating ALF.

Meat adulteration has become a significant concern due to its potential to diminish the nutritional value and trigger allergic reactions. To address the issue of fraudulent adulteration in beef ( Bos taurus ) products, a visual strategy is proposed for multiplex detection of chicken ( Gallus gallus ), duck ( Anas platyrhynchos ), and pork ( Sus scrofa ) components in beef. First, the DNA could be easily extracted from the meat samples using the microneedle within 1 min and used as recombinase polymerase amplification (RPA) template. Next, three primer pairs specific to chicken, duck, and pork DNA are designed with unique oligonucleotide tags and a spacerC9 to halt polymerase activity, enabling the RPA amplicons to possess a single-stranded DNA tail. These tail sequences precisely hybridize with nanoparticle-labeled detection probes and pre-immobilized capture probes on test lines, generating a colorimetric signal on the lateral flow strip (LFS). The enhanced LFS utilizes three distinct Au/Ag nanoparticles, including Au@Ag (yellow), gold nanoflowers (cyan), and gold nanospheres (magenta), each targeting chicken, duck, and pork DNA, respectively. The test output can be easily interpreted through a “magenta yellow cyan” color code. The sensitivity of the multicolor LFS is further improved by the incorporation of DNA-Au/Ag nanoaggregates. Four functional oligonucleotides with complementary sequences are combined to form DNA-Au/Ag nano aggregates, allowing for increased coupling of Au/Ag and enhancing detection sensitivity. Cross-reactivity tests with ten non-target species demonstrate the excellent specificity of the assay. The method can detect chicken, duck, or pork at levels as low as 0.1 g/kg in simulated adulterated beef samples. The entire process, including rapid DNA extraction (1 min), RPA reaction (15 min), and LFS detection (5 min), is completed within 21 min. This platform is user-friendly, eliminating extensive training and specialized equipment, making it ideal for rapid on-site detection of multiplex adulteration by personnel in various fields.

With the development of exonuclease, the exonuclease has been used to construct a variety of aptasensor and to realize the signal amplification. Among them, based on silver nanoparticles (Ag NPs) and exonuclease I (Exo I)-assisted cycle signal amplification strategy, we designed a novel high-sensitivity dual-target electrochemical biosensor to detect Pb 2+ or Hg 2+ in water. In the presence of Hg 2+ , the Hg 2+ was fixed to the aptamer chain by thymine-Hg 2+ -thymine (T-Hg 2+ -T), resulting in the decrease of signal. When Pb 2+ was present, DNA single strand S2 dissociated and was bound to Pb 2+ , which automatically triggered Exo I to selectively cut the single chain from the recognition site to achieve the cyclic amplification of the electrochemical signal. The interaction between aptamer and Exo I was investigated by gel electrophoresis. Under the optimum conditions in the scan range -0.20 to 0.60 V, the biosensor had high sensitivity with a linear range of 100 pg/L to 10.0 mg/L, Pb 2+ or Hg 2+ , and the detection limits were 17.0 pg/L ( R 2 = 0.993) and 12.0 pg/L ( R 2 = 0.993), respectively. The relative standard deviation (RSD) of the sensor was 0.5–2.6%, and the recovery of spiked standard solutions was between 98.3 and 110%. The cycle amplification strategy supported by this enzyme has promising applications in detection of the two metal ions in various fields. Graphical Abstract
