This study attempted to fabricate a novel colorimetric method for the quantitative detection of malachite green (MG) residues in fish and water samples. Based on the recognition of the detection target and the enhancement of the peroxidase-like activities of Fe 3 O 4 NPs by MG-aptamer, a colorimetric method was established for MG detection. The detection limit (3 α / κ , n = 9) was 16.7 μg kg –1 , and the detection linear range was 0.06–2.38 μmol L –1 . The recovery rates by standard addition in fish and water ranged from 90.1% to 119% with standard deviations (n = 3) of less than 6.6 %, and was well validated by a HPLC method. The detection procedure could be completed within 1 h. Therefore, the detection method was simple, stable, and reliable, and could be used to detect MG residues rapidly and accurately in fish and aquaculture water.

Copper sulfate is a widely used agent to control insects, bacteria and algae for fishery. However, excess amount of copper ions in water accumulate in aquatic products through the ecological cycle system, highly threatening food safety and public health. Therefore, it is urgent to develop a rapid and efficient method for the determination of copper content in aquatic products. In this study, we developed a label-free biosensor for Cu(II) based on a graphene field-effect transistor gated by structure-switching aptamer probes (SSA-GFET) against Cu(II) we obtained before. The detection mechanism of the biosensor is attributed to the surface charge shift and the potential change of the gate electrode upon the specific binding of Cu(II). The SSA-GFET biosensor has a low detection limit of 10 nM and a linear range of 10 nM to 3 μM to Cu(II). In addition to the excellent selectivity to Cu(II), the biosensor also showes the advantage of high recovery rate for detection of Cu(II) in real fish samples. Because of the detection characteristics of label-free SSA-GFET, it has great advantages in the field of food safety and environmental detection.

In this work, fish oil-gelatin core-shell electrospun nanofibrous membranes (ENMs) were explored as edible films for the encapsulation of hydrophobic fish oil and hydrophilic vitamin C (VC). The results demonstrated that fish oil-VC/gelatin core-shell ENMs could be successfully fabricated by a monoaxial emulsion electrospinning technique. The addition of VC didn't change core-shell structure but increased fragility of nanofibers. Excessive VC/gelatin mass ratios (≥4:20) increased the nanofiber adhesiveness during the storage at room temperature. No obvious chemical interactions occurred among fish oil, gelatin, and VC in the core-shell ENMs. VC addition decreased the loading capacity and had no obvious effects on encapsulation efficiency of fish oil in the ENMs. VC not only prevented lipid oxidation but also converted lipid hydroperoxides to other products during the electrospinning process. Fish oil-VC/gelatin core-shell ENMs were not disintegrated in ultrapure water but were disintegrated into particles in both gastric and intestinal fluids. The fish oil release from fish oil-VC/gelatin core-shell ENMs and disintegrated particles in the simulated intestinal phase of the gastrointestinal tract model and in the simulated intestinal model showed triphasic behaviors. This work provided a potential route to develop functional foods with encapsulation of both hydrophobic and hydrophilic nutrients.

This study was aimed to investigate the structural and immunological properties of parvalbumin from mandarin fish during the Maillard reaction. The microwave-assisted the Maillard reaction was optimized by orthogonal designed experiments. The results showed that the type of sugar and heating time had a significant effect on the Maillard reaction (p< 0.05). The SDS-PAGE analysis displayed that the molecular weight of parvalbumin in mandarin fish changed after being glycated with the Maillard reaction. The glycated parvalbumin was analyzed by Nano-LC-MS/MS and eleven glycation sites as well as five glycation groups were identified. By using the indirect competitive ELISA method, it was found that microwave heating gave a higher desensitization ability of mandarin fish parvalbumin than induction cooker did. In vitro gastric digestion experiments showed that microwave-heated parvalbumin was proved to be digested more easily than that cooked by induction cookers. The microwave-assisted Maillard reaction modified the structure of parvalbumin and reduced the immunoreactivity of parvalbumin of mandarin fish. Keywords:parvalbumin;Maillard reaction;glycation site;structural and immunological properties;in vitro

Fish farming plays a vital role in providing food, nutrition, and employment globally. However, this industry faces security challenges, necessitating the use of fungicides and preservatives, such as bronopol, to increase product yields. Bronopol (2‑bromo-2-nitropropan-1,3-diol; CAS:52–51–7) is widely used in various fields, including food production, cosmetics, and, more recently, aquaculture. Currently, there is a limited number of techniques available for detecting bronopol in aquaculture products. This is primarily due to bronopol's instability, susceptibility to degradation, and tendency to form precipitates that pose challenges in extraction from aquaculture products. For this issue, this study presents a comprehensive method for detecting bronopol content in aquaculture tissues using liquid chromatography-tandem mass spectrometry (LC‒MS/MS). The methodology was optimized, involving extraction with Cu-Zn precipitant, cleanup using a small HLB column, separation on a T3 column, and gradient elution with water and acetonitrile mobile phases. The quantitative approach was employed without the use of an internal standard, following the external standard method. The spiked recoveries at 3 fortification levels (0.1, 0.2, and 1 mg/kg) ranged from 87.1 % to 108.1 % with relative standard deviations RSD ≤ 9.0 %. By applying this method to fresh fish, shrimp, crab, and shellfish samples from a local supermarket, no residues of bronopol were detected, ensuring the reliability of the results. The simplicity, rapidity, and high sensitivity of the method make it a suitable alternative to conventional techniques for bronopol detection. Moreover, the successful validation of the method's recovery and precision supports its potential application in monitoring and preventing the misuse of bronopol in aquaculture, thereby safeguarding aquaculture product quality and protecting public health.

In this study, naphthalene-modified magnetic nanoparticles (Fe 3 O 4 @Nap) were simply prepared based on specific chelation interaction between phosphate groups and metal ions on Fe 3 O 4 surface. The resultant Fe 3 O 4 @Nap were characterized by FTIR, BET, SEM, TEM, NAM, TGA, and VSM techniques. With Fe 3 O 4 @Nap as adsorbent, the polycyclic aromatic hydrocarbons (PAHs) were efficiently extracted by magnetic solid-phase extraction (MSPE) from environmental water and fish samples through the π-π interaction between modified naphthalene groups and PAHs, followed by their determination by GC–MS/MS. The key parameters influencing the extraction efficiency were investigated. Under the optimized conditions, the Fe 3 O 4 @Nap-based MSPE/GC-MS/MS method proposed in this paper was evaluated and applied for analyzing PAHs in environmental water and fish samples. And the proposed MSPE/GC-MS/MS method exhibited good linearities for water samples (in the range of 0.1–10 ng/mL, R 2 >0.9945) and for fish samples (in the range of 1–100 ng/g, R 2 > 0.9905). The limits of detection (LODs) for water and fish samples were 0.004–0.031 ng/mL and 0.07–0.28 ng/g, respectively. Additionally, this method exhibited desirable accuracy and precision. The PAH recovery values from water and fish samples ranged from 81.5% to 109.6% with inter- and intra-day relative standard deviations (RSDs) of less than 12.8%. The MSPE/GC-MS/MS method was successfully applied to the analysis of real environmental water and fish samples. Overall, the newly synthesized Fe 3 O 4 @Nap exhibited high sensitivity, specificity, reusability, repeatability, and it could efficiently extract PAHs from environmental water and fish samples by MSPE.

In this paper, the effects of different modification orders of microbial transglutaminase (MTGase) and contents of pectin (0.1–0.5%,w/v) on the gelling and structural properties of fish gelatin (FG) and the modification mechanism were studied. The results showed that the addition of pectin could overcome the phenomenon of high-MTGase-induced lower gelling strength of gelatin gels. At a low pectin content, the modification sequences had non-significant influence on the gelling properties of modified FG, but at a higher pectin content (0.5%,w/v), P0.5%-FG-TG had higher gel strength (751.99 ± 10.9 g) and hardness (14.91 ± 0.33 N) values than those of TG-FG-P0.5%(687.67 ± 20.98 g, 12.18 ± 0.45 N). Rheology analysis showed that the addition of pectin normally improved the gelation points and melting points of FG. The structural results showed that the fluorescence intensity of FG was decreased with the increase in pectin concentration. Fourier transform infrared spectroscopy analysis indicated that the MTGase and pectin complex modifications could influence the secondary structure of FG, but the influenced mechanisms were different. FG was firstly modified by MTGase, and then pectin (P-FG-TG) had the higher gelling and stability properties.

In this study, atmospheric cold plasma (CP) was applied to treat dried fish products at 65 kV and 60 Hz, and its effects on the composition associated with the physicochemical properties were investigated. The CP treatment for 5 min induced the fracture and contraction of fish muscle fibers and increased the microporous structure, and the moisture content of the samples reduced from 39.88 % to 37.88 %. Compared to the control group, the 5-min CP treatment decreased the puncture hardness from 10.42 to 8.43 N ( p < 0.05), and increased the final rehydration ratio from 20.86 % to 28.84 %. In addition, the 5-min CP treatment increased cathepsin B + L by 58 %, and increased trichloroacetic acid (TCA) soluble peptides content by 12 % ( p < 0.05), which indicated that CP contributed to protein degradation. Besides, the CP treatment also promoted lipid oxidation (TBARs value increased from 1.12 to 1.96 mg/Kg) and the production of dicarbonyl compounds (3-deoxyglucuronide and 2,3-butanedione were increased from 0.56 and 0.51–0.95 and 1.75 μg/g, respectively) ( p < 0.05). In conclusion, CP is promising for improving storage stability, chewiness and rehydration of dried fish products, which associated with moderate protein degradation and lipid oxidation in fish muscle.

A novel fluorescent membrane based on carbon dot embedded photonic crystal molecularly imprinted polymer (PCMIP@CDs) was developed for fluorometric and colorimetric dual-mode determination of sulfadimethoxine with excellent selectivity, quick response, and reusability. In the presence of sulfadimethoxine, fluorescence quenching and red shift of diffraction peak from PCMIP@CDs were observed because of the photo-induced electron transfer and structural change of PCMIP@CDs, accompanied by a visual color change from blue to green. The fluorescence quenching efficiency and the red-shift of the diffraction peaks were positively correlated with the logarithms of the sulfadimethoxine concentration at 10 −12 –10 −5 and 10 −10 –10 −5 mol L −1 , respectively. The sensor achieved a limit of detection of 0.08 pmol L −1 (fluorescence method) and 2.75 pmol L −1 (colorimetric method, 3 σ , n = 9). The developed PCMIP@CD membrane was applied for the detection of sulfadimethoxine in fish samples with favorable recovery of 84.8–102.9%.

In this study, diacylglycerol (DAG) emulsions stabilized by different concentrations of sodium caseinate (SCE) or Tween 80 (TWE) were prepared and characterized, and the effects of prepared emulsions as fillers on the gel properties, microstructures and molecular interactions of myofibrillar proteins (MP) gel were investigated. The particle size of SCE decreased gradually with the amount of sodium caseinate, and the minimum value was obtained with 2.5% sodium caseinate. The filling of SCE enhanced the whiteness, water-holding capacity (WHC), gel strength, and texture ( P < 0.05), and facilitated the formation of a more compact gel network with sufficient emulsion droplets to be fixed in it. This positive effect was based on various interactions between SCE droplets and MP, and showed sodium caseinate dosage dependence within a certain range. In addition, TWE obtained the most effective particle size at 1.0% Tween 80 dosage, which had no adverse effect on gel properties. However, when Tween 80 concentration exceeded 1.5%, MP gel's water retention, strength, and texture properties showed a certain downward trend, accompanied by the weakening of gel network and the loss of more oil droplets. These results support that DAG emulsion stabilized by moderate sodium caseinate is a potential active filler, which can not only enhance the content of unsaturated fatty acids in gels, but also improve the gel properties.

Food poisoning caused by histamine ingestion is one of the prevalent allergies associated with fish consumption in the world. Reliable detection of histamine from fish by a portable platform was of urgent importance to food safety. A portable technology for on-site monitoring of histamine in tuna was established through combined azo-derivatized thin-layer chromatography (TLC) with surface-enhanced Raman scattering (SERS) spectroscopy. The real tuna meat sample was directly applied onto the portable sensor for the separation of histamine and azo-derivatizing of histamine was reacted on the TLC plate. The colorless histamine was visualized by azo-derivatization after spraying Pauly reagent onto the diatomite TLC plate. The molecule information and concentration of the histamine was measured and calculated by SERS spectra. Diatomite TLC plate was capable of separating histamine with 1.32 × 10 −7 M of Au colloid for the SERS enhancement. Accordingly, the limit of detection of histamine from mixture sample could achieve 2.8 × 10 −4 ppm. These results indicated that the portable azo-derivatized TLC-SERS sensor not only visualizes the histamine but also improves the intensity of the Raman spectra. The azo-derivatized TLC-SERS sensor could be applied for rapid, convenient, and ultrasensitive point-of-care sensing of histamine in fish.

Biogenic amines (BAs) in fish or fish products may render the food toxic, which allow to be used as an indication for microbial spoilage. Thus, it is crucial to construct a rapid and sensitive analytical method for detection of BAs. In this work, based on rhodamine 6G-loaded and MnO 2 nanosheets-coated mesoporous silica nanospheres (MSNs), a highly sensitive fluorescence probe was designed and developed. Wherein, a “signal on” fluorescence was achieved by using R6G (λ emi : 553 nm) as an energy donor and MnO 2 nanosheets as the energy acceptor. As BAs exist, diamine oxidase will catalyze BAs to generate hydrogen peroxide (H 2 O 2 ), which can reduce MnO 2 to Mn 2+ , resulting with the dissociation of MnO 2 nanosheets and the signal recovery of R6G. Under optimal conditions, the analytical method obtained a wide linear detection range of 10 ∼ 400 nM with a detection limit of 5.97 nM (S/N = 3, n = 3). As a proof-of-application study, the analytical method was verified by spiked fish samples with recoveries of 93.97 to 101.57 % and was accurate at 95 % confidence level, indicating its good performance in detection of BAs and promising applications in food safety.
