Coating of Layer-by-Layer Assembly Based on Chitosan and CMC: Emerging Alternative for Quality Maintenance of Citrus Fruit

Chenyu Niu,Lingling Liu,Amr Farouk,Cunkun Chen,Zhaojun Ban

Citrus fruits are susceptible to microbial infection during storage and transportation, leading to weight loss, quality deterioration and even physiological decay. In this study, a layer-by-layer (LbL) self-assembly edible coating based on chitosan and Sodium carboxymethyl cellulose (CMC) was prepared using electrostatic deposition. Postharvest citrus fruits (Citrus reticulatecv. ‘Chunxiang’) were coated either with the LbL coating, which exhibited an increased transmission rate of water vapor, or with single chitosan coating. The data of some physiological indexes of citrus coated with LbL coating and single coating were compared after being stored at 13 ± 2 °C and at relative humidity (RH) at 80–85% (shelf-life condition) for 10 days. Results demonstrated that the LbL deposited coating was effective at maintaining morphological and microstructural attributes, increasing the surface brightness, firmness and the content of titratable acid and ascorbic acid, as well as reducing the weight loss after storage by about 0.8%. Results in the present study indicated that the LbL edible coating could have the potential to maintain postharvest citrus quality during storage. Keywords:chitosan;CMC;FTIR;SEM;postharvest citrus;quality

Chemiluminescence method based on the KIO4−K2CO3−Mn2+ reaction for rapid and sensitive determination of forchlorfenuron in dried fruit

Baisen Chen,Rongxia Tan,Yufei Hu,Gongke Li

Forchlorfenuron is a low-toxic phenylurea plant growth regulator. Excessive intake of forchlorfenuron can lead to metabolic disorders of the matrix and be harmful to human health. The chemiluminescence intensity of the KIO 4 –K 2 CO 3 –Mn 2+ reaction decreased in the presence of forchlorfenuron. Based on this result, a rapid and sensitive chemiluminescence method was established to determine forchlorfenuron by combining it with a batch injection static device. The injection speed, injection volume and reagent concentration of the forchlorfenuron–KIO 4 –K 2 CO 3 –Mn 2+ chemiluminescence reaction were optimized. Under these optimized conditions, the linear range of the method was 1.0–200.0 μg/L, and the limit of detection was 0.29 μg/L (S/N = 3). The chemiluminescence method for the determination of forchlorfenuron could be completed in 10 s. The method was applied to detect the residual forchlorfenuron in dried fruit samples, and the results are consistent with high-performance liquid chromatography-mass spectrometry. This method has the advantages of high sensitivity, rapid response, less reagent consumption, and convenient operation. It will provide a new perspective for chemiluminescence for the rapid and sensitive determination of forchlorfenuron in various complex samples.

Characterization of the volatile profile of feijoa (Acca sellowiana) fruit at different ripening stages by HS-SPME-GC/MS

Xinyue Song,Fen Dai,Jiarong Yao,Zhen Li,Zhongping Huang,Huijun Liu,Zuoyi Zhu

This study investigated the changes of volatile organic compounds (VOCs) in feijoa fruits during ripening. The VOCs were determined by head space solid phase microextraction and gas chromatography coupled to mass spectrometry (HS-SPME-GC/MS). Results showed that 78 VOCs were identified, mainly containing esters, terpenoids, aldehydes and alcohols. Only the minority of VOCs had relatively higher contents in unripe feijoa fruits, while most VOCs were highly accumulated in mid-ripe and ripe feijoa fruits. In the early stage, aldehydes and alcohols were the main flavor compounds which provided fruity and green aroma. The contribution of esters and terpenoids increased during the ripening process, leading to strong fruity and flower fragrance. Hexanal was the main volatile component of unripe feijoa fruits, while methyl benzoate, ethyl benzoate and 3-carene were the main volatile components of mid-ripe and ripe feijoa fruits. According to partial least square-discriminant analysis (PLS-DA), 17 VOCs with variable importance in the projection (VIP) > 1 could be used as significant markers to distinguish feijoa fruits during ripening. This study provided a theoretical support for the further study of the formation mechanism of VOCs in feijoa fruits during the ripening process.

An advanced ratiometric molecularly imprinted sensor based on metal ion reoxidation for indirect and ultrasensitive glyphosate detection in fruit

Xiaoyu Su,Kaiyi Zheng,Xiaoyu Tian,Xuan Zhou,Xiaobo Zou,Xuechao Xu,Zongbao Sun,Wen Zhang

An indirect and ultrasensitive ratiometric molecularly imprinted (MIP) sensor, based on metal ion reoxidation, is introduced for glyphosate (GLY) determination in fruit. As high-performance signal amplification substrates, carbon nanotubes (MWCNTs) and gold nanoparticles (AuNPs) are conveniently modified on GCE. The artificial antibody-MIP membrane, presents typical three-dimensional structure to GLY template. Built-in reference methylene blue (MB) is directly electropolymerized on MWCNTs-Au/GCE. Particularly, Cu 2+ and GLY interestingly form chelate complex, and the Cu 2+ ( I Cu ) in Cu(Ⅱ)-GLY-complex can be reoxidized, and indirectly quantizes GLY. The reference signal ( I MB ) presents noteworthy stability with different GLY levels, and the ratiometric readout ( I Cu / I MB ) is recognized as a more trustworthy indicator to quantize GLY. Proposed sensor presents broad range as 1.73 ∼ 400 ng/mL, and limit of detection is well found as 0.24 ng/mL (S/N = 3). Finally, as-fabricated method is verified with standard HPLC in real-fruit-sample, and the errors and recovery rates are calculated as 3.4% ∼ 6.7% and 94.4% ∼ 104.6%, respectively.

Thermoultrasound and microwave-assisted freeze-thaw pretreatments for improving infrared drying and quality characteristics of red dragon fruit slices

Edidiong Joseph Bassey,Jun-Hu Cheng,Da-Wen Sun

The effects of thermoultrasound (US-FT), microwave (MW-FT), and room temperature (RT-FT) freeze–thaw pretreatments were evaluated for improving drying kinetics and quality during infrared drying (IRD) of red dragon fruit slices (RDFS). Results indicated that microstructural alterations induced by the different pretreatments improved the moisture removal rate and effective diffusivity, and significantly reduced the drying time. US-FT pretreatments prompted more efficient drying and presented an overall enhancement in the quality of RDFS, particularly at low temperatures of 25 and 50 ℃, while enhancements in TFC, FRAP, and CUPRAC were associated with RT-FT. High-power (500 W) MW-FT pretreatments improved colour and structural properties, while low-power (100, 300 W) improved TPC, TFC, ascorbic acid, betalains, and antioxidant activity. Overall, cellular and chemical alterations prompted by pretreatments improved the drying process but presented adverse effects on betaxanthin. The study presented the fundamental background for improving the IRD of foods from the use of improved thawing approaches during freeze–thaw pretreatments.

Preparation of branched RG-I-rich pectin from red dragon fruit peel and the characterization of its probiotic properties

Meng-yuan Zhang,Jun Cai

Red dragon fruit peel is a pectin-rich fruit waste that is a potential source of prebiotics and whose different sources and structures will influence its prebiotic function. Thus, we compared the effects of three extraction methods on the structure and prebiotic function of red dragon fruit pectin, the results showed that the citric acid extracted pectin produced a high Rhamnogalacturonan-I (RG-I) region (66.59 mol%) and more side-chains of Rhamnogalacturonan-I ((Ara + Gal)/Rha = 1.25), which can promote bacterial proliferation significantly. The side-chains of Rhamnogalacturonan-I may be an important factor in that pectin can promote the proliferation of B. animalis. Our results provide a theoretical basis for the prebiotic application of red dragon fruit peel.

Effects of Microporous Packaging Combined with Chitosan Coating on the Quality and Physiological Metabolism of Passion Fruit after Harvest

Zhong Zhiwei,Zhou Lei,Yu Kaibo,Jiang Fenghua,Xu Jing,Zou Liqiang,Du Liqing,Liu Wei

The postharvest quality of passion fruit deteriorates rapidly because of intense physiological metabolism and serious water loss. It is difficult to solve the problems of water loss and microbial growth of passion fruit during room temperature storage at the same time by use of a single chitosan coating or microporous packaging technology. Therefore, a dual modified atmosphere microenvironment based on microporous packaging (MP) and a chitosan coating (CH) was established, and its effects on the quality and physiological metabolism of passion fruit were evaluated. The results indicate that MP can effectively reduce water loss and that the CH inhibits microbial growth. Combined application (MP-CH) is found to significantly inhibit fruit weight loss, which shows a value of only 11.17% on Day 12, while the weight loss of the control is as high as 51.57%. The ascorbic acid and total phenol contents of passion fruit in MP-CH are 5.63 and 37.00 mg·100 g −1 , respectively, on Day 12, which are markedly higher than those in the control. The activities of superoxide dismutase, peroxidase and catalase in MP-CH on Day 12 are 3.20, 2.22, and 1.52 times higher than those in the control, respectively, which effectively improve the antioxidant level and reduce the accumulation of malondialdehyde and hydrogen peroxide. Furthermore, MP-CH delays the degradation of protopectin, cellulose, and hemicellulose by inhibiting the activities of pectin methylesterase, polygalacturonase, cellulase, and β-glucosidase. Consequently, MP-CH is a promising method for extending the shelf life of passion fruit to 12 days at 25 °C by regulating water loss and physiological metabolism.

Sensitive and handy detection of pesticide residue on fruit surface based on single microsphere surface-enhanced Raman spectroscopy technique

Yiting Feng,Xiuli Wang,Yinghao Chang,Jia Guo,Changchun Wang

Hypothesis: Surface-enhanced Raman spectroscopy (SERS) has become an emerging and reliable tool for detecting pesticide residues due to its high sensitivity, fast testing speed and easy sample handling. SERS active substrates are the key to achieve efficient and sensitive detection. However, for the most widely used noble metal nanoparticles, there are problems of high noble metal nanoparticle usage and random aggregation. The micron-scale Raman spot is focused on multiple randomly aggregated nanoparticles during the test, resulting in poor reproducibility. Therefore, the development of micron-scale cost-effective SERS substrates with good reproducibility and simple detecting method is of great significance in practical detection. Experiments: Through deposition of silver nanoparticles (Ag-NPs) by chemical reduction on the surface of monodisperse sulfonated polystyrene (SPS) microspheres, micron-sized PS@Ag-NPs core–shell microspheres were prepared with excellent SERS activity. After that, two simple protocols (Method I and Method II) were explored for the determination of thiram on apple epidermis. Findings: Based on our developed strategy of the single microsphere SERS technique, we successfully fabricated uniform PS@Ag-NPs substrate with high SERS activity and excellent detection sensitivity. The single microsphere SERS technique possesses the capability of anti-dilutability and the utilization of ultra-low PS@Ag-NPs microsphere dosage, realizing qualitative and quantitative detection of thiram on apple with detection limits far below the standard stipulated by China and the European Union.

Novel composite nanomaterials based on magnetic molecularly imprinted polymers for selective extraction and determination of rutin in fruit juice

Fangqi Wang,Xu Ni,Jingjing Zhang,Qikun Zhang,Huning Jia,Hua He,Pierre Dramou

In this work, with the attempt to further improve the selectivity, magnetism and loading proportion of existing adsorbents, a novel composite (MGO/MHNTs@MIPs) was synthesized by electrostatically combining molecularly imprinted polymer based on the surface of magnetic halloysite nanotubes (MHNTs@MIPs) with magnetic graphene oxide (MGO). Then some characterizations were done to prove its successful synthesis. Besides, the bonding experiment showed that it possessed a loading capacity of up to 132 mg·g −1 , and the adsorption behavior of MGO/MHNTs@MIPs was elucidated by Langmuir isotherm model and Pseudo-second order model. By comparing its adsorption capacity to analogues, we concluded that the MGO/MHNTs@MIPs with the MHNTs@MIPs as basic elements exhibited higher selectivity (imprinting factor = 2.25) than that of MGO/MHNTs@NIPs based on MHNTs@NIPs for template rutin. Furthermore, a series of solid phase extraction conditions were optimized, and then the materials were used for the extraction and detection of rutin in fruit juice under the optimal conditions.

Integrative transcriptomic and metabolomic alterations unravel the effect of melatonin on mitigating postharvest chilling injury upon plum (cv.Friar) fruit

Ranran Xu,Limin Wang,Kexin Li,Jiankang Cao,Zhilei Zhao

Plum fruit often suffer severe chilling injury (CI) during storage. Plum ( Prunus salicina Lindl. cv. Friar) fruit were treated with melatonin at various concentrations and stored at 0 ℃ for 6 w and additional 3 d of shelf-life at 23 ℃. Observations revealed that the treatment with 1.0 mmol L −1 melatonin mitigated CI of the fruit by reducing flesh-reddening and ‘ethylene burst’. The melatonin treatment also suppressed fruit softening and maintained energy status. Results from UPLC-MS assay showed that several individual phenolic compounds involved in the biosynthesis of flavonoids and anthocyanins, especially, the red pigment of cyanidin-3- O -glucoside, were stimulated by cold stress and its removal, but the stimulation was effectively inhibited by melatonin treatment, and thereby flesh-reddening was alleviated. Results from RNA-sequencing unraveled that cold stress and its removal altered transcriptomic profiling of the plum fruit, but the melatonin treatment hindered the transcription of genes related to the anthocyanin biosynthesis. Correlation analysis indicated that seven MYBs were positively correlated with anthocyanin biosynthesis, which were generally down-regulated in the melatonin-treated plums. The integrative analysis of transcriptomic and metabolic alterations revealed differentially but delicately coordinated inhibition of postharvest physiological metabolisms of ‘Friar’ plums by melatonin in response to cold stress.

Preparation and characterization of ultrasound-assisted essential oil-loaded nanoemulsions stimulated pullulan-based bioactive film for strawberry fruit preservation

Arif Rashid,Abdul Qayum,Qiufang Liang,Lixin Kang,Husnain Raza,Zhuzhong Chi,Runhao Chi,Xiaofeng Ren,Haile Ma

Ultrasound is a high-energy approach that can help with homogenization and dispersion in cavitation. In this study, nanoemulsions of curcumin and orange essential oil were prepared with ultrasound treatment at different times. The ultrasound-treated nanoemulsions for 10 min exhibited the smallest droplet size, the best storage, and higher thermal stability. The pullulan-based film with ultrasound-assisted nanoemulsions exhibited improved water vapor permeability and moisture content and the highest tensile strength and elongation at break. The structural analysis showed that ultrasonic treatment enhanced the H-bond, resulting in a more orderly molecular arrangement and intermolecular compatibility. Furthermore, the bioactive film had the maximum oil retention time. It possessed excellent bacteriostatic properties against Escherichia coli and Staphylococcus aureus due to the smallest oil droplets and uniform distribution in the film matrix. Besides, the weight loss and deterioration of the strawberry fruit were effectively reduced, thus prolonging the shelf life.

NC-1 coating combined with 1-MCP treatment maintains better fruit qualities in honey peach during low-temperature storage

Wangshu Zhang,Luyang Jing,Huiyun Chen,Sheng Zhang

Summary The effect of carnauba wax-based nanoemulsion coating-1 (NC-1) combined with 1-methylcyclopropene (1-MCP) treatment on the storage of honey peach fruit ‘Yulupantao’ for 28 days under 0 ± 0.5 °C, and 90% RH conditions was studied. The results showed that NC-1 or 1-MCP treatment alone and in combination were proved to be more effective in maintaining fruit qualities compared to the control group with no treatment. However, the application with the combination of NC-1 and 1-MCP treatment had the most significant effect on reducing mass loss and decay rate, maintaining firmness, total soluble solids (TSS), ascorbic acid (AsA) and titratable acid (TA) contents, suppressing malondialdehyde (MDA) accumulation and conductivity ascend. Moreover, the activities of peroxidase (POD) were maintained, and polyphenol oxidase (PPO) and phenylalanine ammonialyase (PAL) were effectively suppressed in NC-1, 1-MCP and combined treatment than that of control. Therefore, the combination of NC-1 and 1-MCP treatment could be an efficient way to maintain the qualities of peach fruit during low-temperature storage after harvest.
