Non-small cell lung cancer (NSCLC) is the most common type of lung cancer with poor survival, in part because that half of the cases are already in advanced stage when they are diagnosed. In response to this issue, the present work constructed a novel electrochemical biosensor for sensitive detection of circulating tumor DNA (ctDNA) related to NSCLC. Specifically, polyethyleneimine (PEI) and covalent organic framework (COF TAPB-TFPB ) functionalized nitrogen-doped graphene (NG) nanocomposite (NG-PEI-COF TAPB-TFPB ) was designed and employed as sensing platform for the first time. NG-PEI-COF TAPB-TFPB with porous structure and good conductivity showed a synergistic effect on promoting electron transfer, which could provide large specific surface area and active sites to increase the immobilization of capture probe (CP). In addition, gold nanoparticles (AuNPs) decorated Fe-based metal-organic framework (Fe-MOF) were labeled with signal probe (SP) to form tracer label, which would attach to the electrode surface by sandwich hybridization of target probe (TP) with CP and SP. Consequently, Fe III from Fe-MOF could react with K 4 [Fe(CN) 6 ] to form electroactive prussian blue, generating a significantly amplified electrochemical detection signal. Under optimum conditions, the proposed DNA biosensor displayed a broad linear range for ctDNA determination from 100 fM to 100 nM with a detection limit of 7.65 fM. Moreover, the biosensor was successfully fabricated to assay ctDNA in human serum, indicating the potential for cancer diagnosis. More importantly, the high sensitivity and satisfactory stability of this biosensor make it a promising method to detect other biomolecules by changing the corresponding sequences of CP and SP.

The detection of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is crucial for preventing and controlling infectious diseases and disease treatment. In this work, a Au@Ti3C2@PEI-Ru(dcbpy)32+ nanocomposite-based electrochemiluminescence (ECL) biosensor was rationally designed, which realized sensitive detection of the RNA-dependent RNA polymerase (RdRp) gene of SARS-CoV-2. In addition, a DNA walker was also used to excise the hairpin DNAs under the action of Nb.BbvCI endonuclease. Furthermore, model DNA–Ag nanoclusters (model DNA–AgNCs) were used to quench the initial ECL signal. As a result, the ECL biosensor was used to sensitively detect the SARS-CoV-2 RdRp gene with a detection range of 1 fM to 100 pM and a limit of detection of 0.21 fM. It was indicated that the ECL biosensor had a great application potential for clinical medical detection. Furthermore, the DNA walker amplification also played a reliable candidate strategy for other detection methods.

The photophysical properties of the hydrogen bond complexes consisting of low-barrier hydrogen bond (LBHB) depend on their environmental condition. However, the stimuli responsive materials based on LBHB are still a challenging problem. Here, we report a novel material system which has high color response to CO 2 based on the position changes of the proton between the two heavy atoms in the low barrier hydrogen bond. Perylene diimide radical anion/branched polyethyleneimine hydrogen bonding complex (TFPDI-b-PEI) linked by low-barrier hydrogen bond ([O 1 -----H 1 ---N 1 ] −1 ) is obtained and it shows the maximum absorption of 789 nm. Meanwhile, the maximum absorption of TFPDI-b-PEI appears at 561 nm with purple when CO 2 is adsorbed in the branched polyethyleneimine . In essence, the geometry of LBHB in TFPDI-b-PEI can switch between two types of hydrogen bond ([O 1 ----H 1 ----N 1 ] −1 and [O 1 -----H 1 ---N 1 ] −1 ) via adjusting the interaction between the surrounding molecule and amine groups in LBHB by interaction of CO 2 with TFPDI-b-PEI. Further the TFPDI-b-PEI shows excellent color switching and colorimetric sensing performance for CO 2 aqueous solution and its gas based on this hydrogen bond dynamic characteristic.

Abstract A new electrochemiluminescence (ECL) cytosensor is proposed for the simultaneous determination of phosphatidylserine (PS) and epidermal growth factor receptor (EGFR) based on the ECL signals of metal-organic framework-5 (MOF-5) loaded CdS quantum dots and N-(aminobutyl)-N-(ethylisoluminol)-polyethylenimine capped Au and Ag nanoparticles. Apoptosis promotes the exposure of PS and reduces the expression of EGFR in cell membranes. Two spatially resolved areas on dual-disk glassy carbon electrodes were designed to eliminate the interference from different ECL probes. Using HepG2 cells treated with resveratrol to induce apoptosis, the cytosensor exhibited high sensitivity, simplicity, and high reproducibility, demonstrating its potential in drug screening and rapid apoptotic cell detection. The strategy reported provides a promising platform for the highly sensitive cytosensing and convenient screening of clinically relevant anticancer drugs. Graphical Abstract

It was a major challenge to acquire CFRPs with both great toughness and strength . Inspired by spider silk , a biomaterial with impressive toughness, we discovered that introducing highly dense weak hydrogen bonds at the interface can be applied to CFRPs toughening. Highly dense weak hydrogen bonds between the branched polyethyleneimine (PEI) and metal-organic frameworks UIO-66 at the interface of CFRPs were formed by the “bridge” of hydroxyl-rich poly (vinyl alcohol) (PVA). The break of those hydrogen bonds could dissipate substantial energy and disperse the stress, the impact toughness of CFRPs with the “PEI-PVA-UIO-66” interlayer was up to 99.53 kJ/m 2 . Compared with composites reinforced by CFs modified with UIO-66 by chemically grafting or by physically absorbing without PVA, the impact toughness of CF-PVA-MOF/EP improved by 45% and 33%, respectively. This means that the highly dense weak hydrogen bonds have superiority for CFRPs toughening. Meanwhile, the interfacial shear strength (100.85 MPa) and interlaminar shear strength (102.44 MPa) of CFRPs were realized by the innovative designed interfacial structure.

Lithium‑oxygen battery is a very promising energy storage device for its ultrahigh specific capacity. However, due to the undecomposable insulating oxidation species accumulated on the cathode and the safety issues caused by dendrite penetration, the cycling performance of lithium‑oxygen battery is seriously restricted. Herein, through interfacial polymerization (IP) method, a polyetherimide (PEI)-based separator with dense polyamide thin layer was prepared. And the PEI-based separator is cross-linked with Polyethylene polyamines (PEPA) and undergoes 1,3,5-benzenetricarbonyl chloride (TMC) amidation . According to the SEM result, the obtained polyamide layer is well cross-linked with the PEI-based separator and at the same time it owns a dense micro-pore structure, which ensures a battery structure stability and a good electrochemical performance of the separator. Moreover, the increased IP layer amidated by TMC can not only greatly increase the ionic conductivity of the polyamide/polyetherimide (P/PEI) cross-linked separator to 1.0 mS cm −1 , but also significantly decrease the interfacial impedance between the PEI and the electrode. With all these merits, the battery assembled with the representative P/PEI2 separator shows boosted cycling performance of over 90 cycles at a capacity of 1000 mAhg −1 . Besides, the electrophilic boric acid was introduced to reduce the interaction between anions and cations , resulting in effectively promoted transport efficiency of lithium ions and the cycling life of the battery can be even prolonged to 108 cycles at a capacity of 1000 mAhg −1 .

In this study, hemin-functionalized polyethyleneimine/silica nanoparticles (hemin/PEI/SiO2 NPs) served as an electrochemical label for ultrasensitive detection of prostate-specific antigen (PSA), a clinically important prostate cancer biomarker. A poly(thionine)/carbon nanotube (PTh/CNTs) multilayer film was fabricated on a glassy carbon electrode (GCE) via integrated electropolymerization and electrostatic layer-by-layer (LbL) assembly, forming a {PTh/CNTs}2/PTh modified GCE with abundant amino groups. PSA capture antibodies (Ab1) were then covalently immobilized on the modified electrode through glutaraldehyde (GA) crosslinking, leveraging the film's surface-exposed amino groups. Furthermore, hemin/PEI/SiO2 NPs were synthesized through amine-carboxyl interactions between carboxyl groups of hemin and amine groups of PEI/SiO2 NPs. The immunosensor assembly employed a sandwich-type electrochemical platform using PSA detection antibody (Ab2)-conjugated hemin/PEI/SiO2 NPs. The specific antibody-antigen binding enables efficient electron transfer across the nanoscale spacing (several nanometers) between hemin/PEI/SiO2 NPs and the {PTh/CNTs}2/PTh multilayer film. In this architecture, CNTs serve as excellent electron mediators, significantly facilitating electron transport. Moreover, the high-density immobilization of hemin on PEI/SiO2 NPs significantly amplifies the electrochemical redox currents of the multilayer film through proximity effects. This enhanced electrical signal enables highly sensitive detection of target protein antigen–antibody binding events. Consequently, by correlating the enhanced reduction peak current of the immunosensor with PSA concentration, highly sensitive detection of PSA at various concentrations can be achieved. Under optimized conditions, it exhibited a linear response to logarithmic PSA concentrations (0.01 pg mL−1 to 100 pg mL−1, R = 0.9963), with an ultralow detection limit of 3.0 fg mL−1 (S/N = 3). Furthermore, the immunosensor demonstrated outstanding analytical performance in human serum samples (96.0–103% recovery, n = 3), confirming its high accuracy for clinical applications.

Certain chemo drugs have been reported to potentially induce tumor-specific immune recognition by triggering immunogenic cell death (ICD), which provides a promising alternative way for cancer immunotherapy. However, the immunogenic effects of such treatments are still weak and robust systemic antitumor immune responses are rarely seen when these agents were used alone. Herein, we proposed a trinity immune enhancing nanoparticles (TIENs) for boosting antitumor immune responses of chemo agents. The TIENs was constructed with Food and Drug Administration (FDA) approved polylactic acid (PLA), canonical proton-sponging cationic polymer polyethyleneimine (PEI), and Toll-like receptor 9 (TLR9) agonist cytosine phosphate guanine oligodeoxynucleotide (CpG-ODN). In in vitro studies, the TIENs was proved to (1) promote antigen capturing, (2) antigen-presenting cells (APCs) activation, and (3) antigen cross-presentation. In in vivo studies, intratumorally injected TIENs greatly enhanced antitumor effect and robust immune responses of oxaliplatin and doxorubicin in murine CT26 and 4T1 tumor models, respectively. Furthermore, after decoration with a detachable shielding, the TIENs was proved to be effective in promoting the antitumor effects of chemo agents after intravenous injection. The combination of TIENs with clinically widely used chemo agents should be meaningful in boosting effective antitumor immune responses and cancer therapy.

Mitophagy is a distinct physiological process that can have beneficial or deleterious effects in particular tissues. Prior research suggests that mitophagic activity can be triggered by CaO2-PM-CsPbBr3 QDs, yet the specific role that mitophagy plays in hepatic injury induced by CaO2-PM-CsPbBr3 QDs has yet to be established. Accordingly, in this study a series of mouse model- and cell-based experiments were performed that revealed the ability of CaO2-PM-CsPbBr3 QDs to activate mitophagic activity. Golm1 was upregulated in response to CaO2-PM-CsPbBr3 QDs treatment, and overexpressing Golm1 induced autophagic flux in the murine liver and hepatocytes, whereas knocking down Golm1 had the opposite effect. CaO2-PM-CsPbBr3 QDs were also able to Golm1 expression, in turn promoting the degradation of P53 and decreasing the half-life of this protein. Overexpressing Golm1 was sufficient to suppress the apoptotic death of hepatocytes in vitro and in vivo, whereas the knockdown of Golm1 had the opposite effect. The ability of Golm1 to promote p53-mediated autophagy was found to be associated with the disruption of Beclin-1 binding to Bcl-2, and the Golm1 N-terminal domain was determined to be required for p53 interactions, inducing autophagic activity in a manner independent of helicase activity or RNA binding. Together, these results indicate that inhibiting Golm1 can promote p53-dependent autophagy via disrupting Beclin-1 binding to Bcl-2, highlighting a novel approach to mitigating liver injury induced by CaO2-PM-CsPbBr3 QDs.

Efficient enrichment glycoproteins/glycopeptides from complex biological solutions are very important in the biomedical sciences, in particular biomarker research. In this work, the high hydrophilic polyethylenimine conjugated polymaltose polymer brushes functionalized magnetic Fe3O4 nanoparticles (NPs) denoted as Fe3O4–PEI–pMaltose were designed and synthesized via a simple two-step modification. The obtained superhydrophilic Fe3O4–PEI–pMaltose NPs displayed outstanding advantages in the enrichment of N-linked glycopeptides, including high selectivity (1:100, mass ratios of HRP and bovine serum albumin (BSA) digest), low detection limit (10 fmol), large binding capacity (200 mg/g), and high enrichment recovery (above 85%). The above-mentioned excellent performance of novel Fe3O4–PEI–pMaltose NPs was attributed to graft of maltose polymer brushes and efficient assembly strategy. Moreover, Fe3O4–PEI–pMaltose NPs were further utilized to selectively enrich glycopeptides from human renal mesangial cell (HRMC, 200 μg) tryptic digest, and 449 N-linked glycopeptides, representing 323 different glycoproteins and 476 glycosylation sites, were identified. It was expected that the as-synthesized Fe3O4–PEI–pMaltose NPs, possessing excellent performance (high binding capacity, good selectivity, low detection limit, high enrichment recovery, and easy magnetic separation) coupled to a facile preparation procedure, have a huge potential in N-glycosylation proteome analysis of complex biological samples.

A new pH-responsive charge-conversion and rapidly self-dissociative multifunctional polymer lauric acid-polyethyleneimine-cyclohexanedicarboxylic anhydride-folic acid (LA-PEI-HHPA-FA, LPHF) is developed. LPHF as an amide with neighboring carboxylic acid groups becomes negatively charged at physiological pH and was used as a coating polymer to shield positively charged DNA/polyethyleneimine 25 k (DP) binary polyplexes, resulting in the formation of DPLPHF ternary polyplexes. The characterization using various methods proved that DPLPHF ternary polyplexes are stable at physiological pH, while in acidic endosomes, the coating polymer LPHF could quickly self-dissociate from the DP binary polyplexes. As compared with DP binary polyplexes, DPLPHF ternary polyplexes containing the tumor targeting ligand folic acid (FA), the absorption enhancer lauric acid (LA) and the charge-conversion entity cis-1,2-cyclohexanedicarboxylic anhydride (HHPA) exhibited more effective cellular uptake and higher transfection efficiency with lower non-therapeutic cytotoxicity in vitro. Moreover, in vivo studies revealed the significantly stronger tumor targeting and tumor inhibiting efficacy of DPLPHF ternary polyplexes. All these findings demonstrated the potential of LPHF as a promising coating polymer for enhanced gene transfection efficiency of DP binary polyplexes in vitro and in vivo.

Excessive reactive oxygen species (ROS) generated by wounds and bacterial infection are the great obstacles for diabetic wound closure. Here, we present a novel dual ROS-scavenging and antibacterial microneedle array patch with M2 macrophage-derived extracellular vesicles (M2-EVs) encapsulation for diabetic wound healing. The microneedle patch with a core–shell structure is composed of hyaluronic acid (HA) and PFP hydrogel crosslinked by polyvinyl alcohol (PVA)/2-formylphenylboronic acid (2-FPBA)/polyetherimide (PEI). When exposed to high levels of ROS existed in the wound, the dynamic covalent bonds of the hydrogel network can dissociate and subsequently release M2-EVs from the HA core. Benefitting from ROS-scavenging properties, antibacterial activities, and macrophage polarization driven by M2-EVs, the microneedle patch could effectively promote wound closure in diabetic animal models. These results indicated that the proposed ROS-scavenging microneedle patches with EVs delivery are promising for variety of difficult-to-heal wounds in clinical application.
