Enhanced photo-fermentative hydrogen production of Rhodopseudomonas sp. nov. strain A7 by the addition of TiO2, ZnO and SiC nanoparticles

Bingfeng Liu,Yaruo Jin,Zhijiang Wang,Defeng Xing,Chao Ma,Jie Ding,Nanqi Ren

In order to strengthen photo-fermentative hydrogen production by different photocatalytic nanoparticles , hydrogen production by photo-fermentative bacteria with addition of TiO 2 , ZnO and SiC nanoparticles in batch culture were investigated in this study. The results indicated that three nanoparticles could improve hydrogen production performance of Rhodopseudomonas sp. nov. strain A7 under respective optimal conditions. The hydrogen yield of 2.81 mol-H 2 /mol-acetate was obtained when TiO 2 nanoparticles with a concentration of 300 mg/L and size of 25 nm. The concentration of ZnO nanoparticles was at 100  mg/L, hydrogen yield reached 2.64 mol-H 2 /mol-acetate. Compared with TiO 2 and ZnO nanoparticles, SiC nanoparticles exhibited greatest potential for enhancing photo-hydrogen production. By addition of nano-SiC with concentration of 200 mg/L which was prepared at temperature of 1500 °C, the maximum hydrogen volume, average hydrogen content and hydrogen yield of strain A7 were achieved at 2272 mL-H 2 /L-culture, 85.2% and 2.99 mol-H 2 /mol-acetate, respectively. And hydrogen production was 18.6% higher than that of alone strain A7 without the addition of nanoparticles. Therefore, the addition of SiC nanoparticles is a promising strategy to improve photo-fermentative hydrogen production from wastewater.

An Endophytic Bacillus amyloliquefaciens ZN-S18 Suppresses Fusarium oxysporum f. sp. lycopersici Through Synergistic Activity of Surfactin and Iturin A

Dan-Na Ma,Yihan Niu,Mengli Chen,Jun-Nan Yao,Huiming Wu,Taiying Li

Fusarium wilt, caused by the soil-borne fungusFusarium oxysporumf. sp.lycopersici(Fol), is a devastating disease that severely constrains tomato production worldwide. In this study, an endophytic bacterium, designated ZN-S18, was isolated from healthy tomato stems and identified asBacillus amyloliquefaciensbased on phylogenetic analyses of the 16S rRNA andrpoBgenes. Dual culture assays demonstrated that ZN-S18 exhibited strong antagonistic activity against Fol and significantly inhibited fungal mycelial growth. The cell-free culture filtrate of ZN-S18 markedly suppressed Fol spore germination and mycelial growth, and the inhibitory effect on spore germination was irreversible. LC-MS analyses confirmed that ZN-S18 produces the cyclic lipopeptides surfactin and iturin A. In planta assays further demonstrated that pretreatment with ZN-S18, its culture filtrate, or the combination of surfactin and iturin A significantly reduced Fusarium wilt in tomato seedlings. Moreover, the culture filtrate disrupted fungal cell wall integrity, as indicated by the increased sensitivity of Fol to Congo red stress. Notably, the combined application of surfactin and iturin A exhibited significantly stronger antifungal activity than either compound alone under Congo red-containing conditions, suggesting a synergistic effect on fungal cell wall integrity. Collectively, these findings indicate that the endophytic strainB. amyloliquefaciensZN-S18 is a promising biocontrol agent against tomato Fusarium wilt, primarily through the synergistic action of surfactin and iturin A. This study provides an environmentally friendly alternative to chemical fungicides and establishes a foundation for future investigations into the molecular mechanisms underlying lipopeptide-mediated antagonism.

The synthesis of PLA-modified high bio-based content WPU and synergistic research on degradation performance with microorganisms (Bacillus sp.)

Song Kangjia,Liu Jie,Zhang Feifei,Lu Aide,Jin Liqiang,Zhao Zecui,Nian Xinyue,Song Jianan,Zhu Deyi

To mitigate plastic pollution and reduce reliance on fossil fuels, the development of high-performance polymeric coatings from renewable resources to mitigate plastic pollution and reduce reliance on fossil fuels remains a critical challenge. Among them, substituting petroleum-based isocyanates with bio-based 1,5-pentanediisocyanate (PDI) not only significantly reduces the carbon footprint but also provides a key green feedstock for developing high-performance biodegradable polyurethanes. In this study, bio-based poly (lactic acid) polyol (PLA) and PDI were utilized as the raw materials. By incorporating polytetramethylene glycol (PTMEG) into the soft segment of waterborne polyurethane (WPU), a series of PLA/PDI bio-based waterborne polyurethanes (PP-WPU-Ts) were synthesized. The results demonstrate that PP-WPU-3 (n PLA : n PTMEG = 5:5) achieved a high bio-based content of 53.1 wt% along with a balanced combination of mechanical properties, including a tensile strength of 4.02 MPa, an elongation at break of 1118.6%, and a toughness of 26.7 MJ/m 3 . Additionally, PP-WPU-3 exhibited outstanding wear and yellowing resistance, reaching 80,000 abrasion cycles and a grade 1 yellowing resistance (ΔE = 2.8). Most importantly, the material exhibited controllable degradation across various environments: rapid disintegration in alkaline media (65% weight loss in 6 h), efficient biodegradation in soil (54.3% in 120 days), and synergistic mineralization by Bacillus sp. (19.3% in 15 days), offering a promising end-of-life solution. The optimized PP-WPU-3 possesses balanced mechanical properties while also exhibiting excellent yellowing resistance and degradation capabilities. These characteristics validate its potential as an eco-friendly alternative for organic coatings.

Mechanistic insights into the enhancement of MgSO4-mediated leaching of ion-adsorption rare earth ores by Penicillium sp. fermentation broth

Jie Wang,Yimin Lan,Yaru Duan,Yun Fang,Yang Liu,Jun Wang,Zhenyue Zhang,Ruan Chi,Chunqiao Xiao

The fragility of the global rare earth supply chain and the urgent demand for sustainable exploitation have driven intensive research into green and high-efficiency extraction technologies. Bioleaching technology, distinguished by its environmental compatibility and resource adaptability, has emerged as a transformative alternative to conventional chemical leaching methods. This study focuses on the multiphase utilization of ion-adsorption rare earth ores by leveraging Penicillium sp. fermentation broth to promote MgSO 4 -mediated leaching through a biologically enhanced chemical process. The objectives were to elucidate the rare earth leaching mechanisms under bio-promoted chemical processes and to optimize strain cultivation and leaching parameters. Untargeted metabolomics was applied to delineate dynamic changes in differential metabolites before and after leaching, while Fourier transform infrared spectroscopy (FT-IR) and X-ray diffraction (XRD) were utilized to clarify metabolite-mineral interfacial interactions. The results demonstrated that the composite system achieved a rare earth recovery rate of 93.79 % under optimal conditions (liquid-to-solid ratio of 4:1, 6-hour leaching), significantly outperforming conventional chemical leaching. Metabolomic analysis revealed a multifunctional interaction network involving organic acid-mediated proton attack, lipid molecule-driven interfacial modification, and benzenoid compound-regulated redox balance. This study demonstrates that the fermentation broth of Penicillium sp. significantly enhances MgSO 4 -mediated leaching efficiency of ion-adsorption rare earth ores, thereby offering a sustainable pathway to address resource and environmental challenges in rare earth industries.

Exogenous DBT and overexpression of deoR improved the waste tire rubber desulfurization ability of a newly isolated Escherichia sp. strain

Yao Lu,Fang-Qi Gao,Yu-Yan Wang,Ying-Wen Fu,Zhuo-Miao Li,Juan Xie,Gao-Qiang Liu,Yong-Nan Liu

The environmental hazards caused by accumulation of waste tire rubber (TR) constitute a major global concern. Microbial desulfurization is an environmentally friendly method for treating waste TR for recycling; it is characterized by mild reaction conditions and low generation of harmful pollutants. Here, we report a new microorganism, Escherichia sp., which was isolated from waste tire surfaces and has the ability to desulfurize TR. Exogenous dibenzothiophene (DBT) increased the cell growth and TR desulfurization ability of Escherichia sp. in a desulfurization medium containing TR. Transcriptomic analysis revealed that TR-only treatment significantly upregulated the expression of the 4S desulfurization pathway gene and six transcription factors, and TR-DBT cotreatment further increased the upregulation of these genes compared with TR-only treatment in Escherichia sp. Overexpression of these six transcription factors in Escherichia sp. revealed that, deoR , was the most effective at increasing transcription of 4S genes and enhancing the TR desulfurization ability. In addition, exogenous DBT and deoR overexpression enhanced the ability of Escherichia sp. to decrease the intensities of C=C and C-S bonds and increased the intensities of S=O and O=S=O bonds. Our report establishes a TR desulfurization strategy involving exogenous DBT treatment or deoR overexpression in a new Escherichia sp. strain.

Synergistic bioaugmentation: Serratia sp. WKD and carboxymethyl cellulose-modified nano-zero-valent iron enhance malachite green decolorization by activated sludge

Linna Du,Xinjiao Dong,Huan Wu,Gang Li,Jiacheng Zhou,Yiqing Wu,Yanping Xiao,Jing Qin,Xiu Wang

The activated sludge process exhibits limited efficacy in decolorizing triphenylmethane (TPM) dyes like malachite green (MG) in textile wastewater. This study demonstrates that bioaugmentation with Serratia sp. strain WKD significantly enhances MG removal in activated sludge systems. Within 2 h, the combined system (activated sludge + strain WKD) achieved 98.5 ± 0.1 % decolorization efficiency—a 5.3-fold increase over sludge-only controls (18.5 ± 1.7 %). High-throughput sequencing revealed strain WKD's ecological dominance, reducing microbial alpha diversity (8 h) through competitive exclusion. When coupled with carboxymethyl cellulose-modified nano-zero-valent iron (CMC-nZVI), the hybrid system synergistically enriched stress-tolerant degraders ( Serratia sp. and Pseudomonas sp.), thereby accelerating decolorization. Prolonged incubation (24 h) enabled microbial diversity recovery (Shannon and Simpson indices) as detoxification alleviated environmental stress. Functional profiling identified pathway-specific modulation: critical metabolic functions (e.g., carbohydrate/amino acid metabolism) were transiently suppressed, while membrane transport activity remained intact, preserving system functionality. These findings establish strain WKD and CMC-nZVI as a synergistic bioaugmentation strategy, enabling rapid dye removal while maintaining microbial community resilience through dynamic stress-response mechanisms.

Carbonaceous nanomaterials stimulate extracellular enzyme release by the fungus Cladosporium sp. and enhance extracellular electron transfer to facilitate lignin biodegradation

Yan Liu,Hua Ma,Juan Huang,Zhe Li,Yu Pan,Yiwen Du

The release of carbonaceous nanomaterials (CNMs) into the environment inevitably affects the surrounding microenvironment, biochemical processes of microbes, and even entire ecosystems. However, the knowledge of the impact of CNMs on the environmental microbial activities and further natural organic matter biodegradation is still less known. Here we investigated the impact of three CNMs (single-walled carbon nanotubes , graphene, and oxidized graphene) on extracellular enzymes activities of a fungal strain ( Cladosporium sp.) and lignin biodegradation. All three CNMs increased the activities of three non-specific enzymes (laccase, manganese peroxidase, and lignin peroxidase) to different degrees, among which manganese peroxidase activity increased significantly, with an average promotion of 20% within 18 days. Meanwhile, structural characterization revealed the transformations of CNMs caused by fungal biodegradation. CNMs stimulation fungal enzyme activities further enhanced lignin degradation. Adsorption experiments and electrochemistry analysis demonstrated the mechanism that CNMs acted as adsorbent of extracellular enzymes as well as electron conductors, which enhanced extracellular direct electron transfer between enzymes and lignin and facilitated lignin conversion and degradation. Our results highlight the potential effect of CNMs on the lignin degradation by fungi, and require consideration when evaluating the environmental effects of CNMs on microbial communities or even carbon cycling across whole ecosystems.

4-Chlorophenol Oxidation Depends on the Activation of an AraC-Type Transcriptional Regulator, CphR, in Rhodococcus sp. Strain YH-5B

Zhang Hui,Yu Ting,Wang Yiran,Li Jie,Wang Guangli,Ma Yingqun,Liu Yu

4-Chlorophenol (4-CP) oxidation plays an essential role in the detoxification of 4-CP. However, oxidative regulation of 4-CP at the genetic and biochemical levels has not yet been studied. To explore the regulation mechanism of 4-CP oxidation, a novel gene cluster, cphRA2A1, involved in biodegradation of 4-CP was identified and cloned from Rhodococcus sp. strain YH-5B by genome walking. The sequence analysis showed that the cphRA2A1 gene cluster encoded an AraC-type transcriptional regulator and a two-component monooxygenase enzyme, while quantitative real-time PCR analysis further revealed that cphR was constitutively expressed and positively regulated the transcription of cphA2A1 genes in response to 4-CP or phenol, as evidenced by gene knockout and complementation experiments. Through the transcriptional fusion of the mutated cphA2A1 promoter with the lacZ gene, it was found that the CphR regulator binding sites had two 15-bp imperfect direct repeats (TGCA-N6-GGNTA) at −35 to −69 upstream of the cphA2A1 transcriptional start site. Notably, the sub-motifs at the −46 to −49 positions played a critical role in the appropriate interaction with the CphR dimer. In addition, it was confirmed that the monooxygenase subunits CphA1 and CphA2, which were purified by His-tag affinity chromatography, were able to catalyze the conversion of 4-CP to 4-chlorocatechol, suggesting that strain YH-5B could degrade 4-CP via the 4-chlorocatechol pathway. This study enhances our understanding of the genetic and biochemical diversity in the transcriptional regulation of 4-CP oxidation in Gram-positive bacteria.

Immobilization of Sphingomonas sp. GY2B in polyvinyl alcohol–alginate–kaolin beads for efficient degradation of phenol against unfavorable environmental factors

Bo Ruan,Pingxiao Wu,Meiqing Chen,Xiaolin Lai,Liya Chen,Langfeng Yu,Beini Gong,Chunxi Kang,Zhi Dang,Zhenqing Shi,Zehua Liu

In this study, batch experiments were carried out to evaluate the biodegradation of phenol by Sphingomonas sp. GY2B, which were immobilized in polyvinyl alcohol (PVA) – sodium alginate – kaolin beads under different conditions. The optimal degradation performance was achieved by GY2B immobilized in beads containing 1.0% (w/v) of kaolin, 10% (w/v) of PVA, 0.3% (w/v) of sodium alginate, 10% (v/v) of biomass dosage, and exposed to an initial phenol concentration of 100 mg/L. The experimental results indicated that PVA-sodium alginate-kaolin beads can accelerate the degradation rate of phenol by reducing the degradation time and also improve degradation rate. The biodegradation rate of phenol by immobilized cells (16.79 ± 0.81 mg/(L·h)) was significantly higher than that of free cells (11.49 ± 1.29 mg/(L·h)) under the above optimal conditions. GY2B immobilized on beads was more competent than free GY2B in degradation under conditions with high phenol concentrations (up to 300 mg/L) and in strong acidic (pH = 1) and alkaline (pH = 12) environments. Higher phenol concentrations inhibit the biomass and reduce the biodegradation rate, while the lower biodegradation rate at low initial phenol concentrations is attributed to mass transfer limitations. The Haldane inhibitory model was in agreement with the experimental data well, revealing that phenol showed a considerable inhibitory effect on the biodegradation by Sphingomonas sp. GY2B, especially at concentrations higher than 90 mg/L. Intra-particle diffusion model analysis suggests that adsorption of phenol by immobilized beads was controlled by both rapid surface adsorption as well as pore diffusion mechanism. It's worth noting that the presence of 1 mg/L Cr(VI) enhanced the biodegradation of phenol by free cells, while Cr(VI) showed no obvious impact on the removal of phenol by immobilized cells. In addition, immobilized cells were reused 16 times and removed 99.5% phenol, and when stored at 4 °C for 90 days, more than 99% phenol was removed. These results showed that immobilized cells can significantly improve the microbial degradation performance, and protect microorganisms against unfavorable environment. It is implied that PVA -sodium alginate-kaolin beads have great potential to be applied in a practical and economical phenolic wastewater treatment system.

Efficient preparation and production of ulvan oligosaccharides by using a new PL25 family ulvan lyase from Alteromonas sp. 76–1

Tang Tiancheng,Li Chen,Zhu Benwei,Yao Zhong

Ulvan lyase is a useful tool which can degrade ulvan into oligosaccharides with a range of biological activities. In this study, the ulvan lyase ULA-3 from Alteromonas sp. 76–1, which was a 1467-bp encoded 489-amino acid residue protein, has been cloned, heterologously expressed and biochemically characterized. ULA-3 showed the maximum activity at 45 ˚C and pH 9.0. The K m is 1.879 mg mL −1 and V max is 2.597 µmol min −1  mL −1 . Its enzyme activity was outstanding at 11.754 U mg −1 . Moreover, its metal ion stability is exceptionally good and most metal ions at a concentration of 1 mM have no inhibitory effect on its activity. It can endolytically degrade ulvan into oligosaccharides with a degree of polymerization (Dp) of 2–4 and with unsaturated double bond through the β -elimination reaction, which mainly cleaves the glycosidic bond between rhamnose and uronic acids. All in all, our study has increased the ulvan lyase database and offers the opportunity to fully utilize ulvan, a green biomass resource from marine algae.

Reduction of tellurite in Shinella sp. WSJ-2 and adsorption removal of multiple dyes and metals by biogenic tellurium nanorods

Shijuan Wu,Tengfei Li,Xian Xia,Zijie Zhou,Shixue Zheng,Gejiao Wang

Te(IV) reduction to Te(0) is a detoxification process in bacteria. Elemental Te nanostructures are new materials, however, the functions of them, such as adsorption ability, have not been fully explored. Shinella sp. WSJ-2 is a highly Te(IV)-resistant bacterium with a MIC of 8 mmol/L. It was able to rapidly reduce 0.3 mmol/L Te(IV) to Te nanorods (TeNRs) within 28 h, and its Te(IV) reduction ability was relatively stable after 14 cycles repeatedly. The size of TeNRs ranged from 50 to 120 nm. The surface of TeNRs was mainly enveloped with proteins and lipids. Moreover, the biogenic TeNRs were capable of removing multiple dyes and metals and were effectively recycled with 100 mmol/L NaCl. Adsorption equilibrium isotherms showed that the maximum adsorption amounts were 618.8, 1076.8, 496.3, 179.3, 58.7 and 275.3 mg/g for Methylene Blue , Azure B, Malachite Green, Cd(II), Cu(II) and Pb(II), respectively. In conclusion, this is the first evidence that the genus Shinella is involved in Te(IV) reduction. The strong sorption of cationic dyes and metals onto TeNRs is probably due to the electrostatic interaction. This study showed the potential of Shinella sp. WSJ-2 in the bioremediation of toxic Te(IV) and adsorption removal of multiple dyes and metals by biogenic TeNRs.

Re-direction of carbon flux to key precursor malonyl-CoA via artificial small RNAs in photosynthetic Synechocystis sp. PCC 6803

Sun Tao,Li Shubin,Song Xinyu,Pei Guangsheng,Diao Jinjin,Cui Jinyu,Shi Mengliang,Chen Lei,Zhang Weiwen

Background Photosynthetic cyanobacteria have attracted a significant attention as promising chassis to produce renewable fuels and chemicals due to their capability to utilizing solar energy and CO 2 . Notably, the enhancing supply of key precursors like malonyl-CoA would benefit the production of many bio-compounds. Nevertheless, the lacking of genetic tools in cyanobacteria, especially the knockdown strategies for essential pathways, has seriously restricted the attempts to re-direct carbon flux from the central carbohydrate metabolism to the synthesis of bioproducts. Results Aiming at developing new genetic tools, two small RNA regulatory tools are reported for the model cyanobacterium Synechocystis sp. PCC6803, based on paired termini RNAs as well as the exogenous Hfq chaperone and MicC scaffold (Hfq-MicC) previously developed in Escherichia coli . Both regulatory tools functioned well in regulating exogenous reporter gene lacZ and endogenous glgC gene in Synechocystis sp. PCC6803, achieving a downregulation of gene expression up to 90% compared with wildtype. In addition, the Hfq-MicC tool was developed to simultaneously regulate multiple genes related to essential fatty acids biosynthesis, which led to decreased fatty acids content by 11%. Furthermore, aiming to re-direct the carbon flux, the Hfq-MicC tool was utilized to interfere the competing pathway of malonyl-CoA, achieving an increased intracellular malonyl-CoA abundance up to 41% (~ 698.3 pg/mL/OD 730 nm ) compared to the wildtype. Finally, the Hfq-MicC system was further modified into an inducible system based on the theophylline-inducible riboswitch. Conclusions In this study, two small RNA regulatory tools for manipulating essential metabolic pathways and re-directing carbon flux are reported for Synechocystis sp. PCC6803. The work introduces efficient and valuable metabolic regulatory strategies for photosynthetic cyanobacteria.
