Waterborne polyurethane (WPU) dispersions were prepared by post-chain extension and crosslinking reactions using 2-[(2-aminoethyl)-amino]ethyl sulfonic acid sodium (AAS), diethylenetriamine (DETA), triethylenetetramine (TETA) and AAS/DETA as post-chain extenders, and their influence on the properties of WPUs was studied. Results showed the introduction of trifunctional DETA chain extenders significantly improved the molecular weight, T-peel strength, and water resistance of WPUs, suggesting that DETA was an effective crosslinking agent. The WPU dispersions with a 2 : 3 molar ratio of AAS/DETA chain extenders exhibited excellent properties. At a molar ratio of 1.6 for NCO groups to OH groups, a stable WPU dispersion possessed higher solid content of 42.8%. Furthermore, only 4 wt% organic solvents were used in the preparation of WPU dispersions. Results revealed that the obtained WPU dispersions are a potential candidate for binders in water-based inks, without removing organic solvents. This study may provide some insight or guidance for the design and preparation of water-based ink binders.

Antigen peptide-based immunotherapy holds significant promise for tumor treatment by eliciting targeted immune responses against tumor cells. However, current methods often face limitations in achieving effective antigen delivery and immune activation. Addressing these challenges through novel delivery systems and immunomodulatory strategies is crucial for improving the clinical efficacy of this therapy. Herein, a supramolecular system based on the self-assembly of the human papillomavirus (HPV) E7 antigen peptide is established by utilizing a Schiff base-mediated self-assembly approach. The E7 antigen peptide nanoparticles (E7 NPs) are further functionalized by loading chlorin e6 (Ce6) and decorating with bovine serum albumin (BSA), generating BSA/E7/Ce6 NPs. The BSA/E7/Ce6 NPs exhibited remarkable photodynamic effects in the inhibition of tumor cells and significantly improved tumor accumulation. Especially, the BSA/E7/Ce6 NPs improved antitumor immune responses of the E7 antigen by effectively activating cytotoxic T lymphocytes. The combination therapy integrating BSA/E7/Ce6 NPs with photodynamic therapy demonstrated a 1.8-fold increase in CD3+CD8+ T cells compared to monotherapy groups, leading to significantly enhanced antitumor effects. Our findings demonstrate that supramolecular antigen peptide nanomaterials can be established as drug delivery systems while retaining their original antigenicity, providing a versatile strategy for realizing enhanced antitumor immunotherapy.

Epithelial–mesenchymal transition (EMT) is a critical process for cancer progression, which is crucial in inhibiting the immunity in tumors and further boosting tumor metastasis. The suppression of EMT represents a promising strategy for inhibiting metastatic tumors. Herein, a series of new canadine platinum(IV) conjugates with potent antiproliferative and antimetastatic activities were developed, which activated by suppressing EMT and provoking immune response in tumors besides causing DNA injury. The complexes could covalently conjugate to DNA and induce mitochondria-mediated apoptosis via Bcl-2/Bax/caspase3 signaling. The EMT process was remarkably inhibited by suppressing the Wnt/β-catenin pathway, reversing the inflammatory tumor microenvironment, and inhibiting the HIF-1α pathway, which further resulted in the inhibited angiogenesis in tumors. Moreover, the antitumor immunity was elevated by blocking immune checkpoints PD-L1 and CD47 accompanied by the improvement of CD3+ and CD8+ T lymphocytes and the macrophage polarization from M2- toward M1-type simultaneously in tumors.

Children exposed to common aeroallergens may develop asthma that progresses into adulthood. Inflammation regulated by T helper 2 (Th2) cells, a specific subpopulation of CD4+ T lymphocytes, is involved in asthmatic injury. Herein, our microarray data indicated that microRNA-451a-5p (miRNA-451a) expression decreased by 4.6-fold and ETS proto-oncogene 1 (ETS1) increased by 2.2-fold in the peripheral blood lymphocytes isolated from asthmatic children (n = 4) as compared to control individuals (n = 4). The negative correlation between miRNA-451a and ETS1 was further validated in 40 CD4+ T cell samples (10 healthy vs. 30 asthmatic samples). In vitro, naïve CD4+ T cells isolated from control individuals were cultured under Th2 cell polarizing condition. miRNA-451a expression decreased while ETS1 increased in CD4+ T cells in the setting of Th2 cell polarization. Moreover, miRNA-451a knockdown enhanced Th2 cell polarization – cells positive for both GATA3 (GATA binding protein 3, a Th2-transcription factor) and CD4 increased, and the generation of Th2 cell cytokines, interleukin (IL)5 and IL13, increased. In contrast, miRNA-451a overexpression inhibited Th2 cell differentiation. Interestingly, dual-Luciferase assay proved ETS1 as a novel target of miRNA-451a. Moreover, enforced expression of ETS1 partially restored miRNA-451a-induced inhibition of IL5 and IL13, and increased the GATA3+CD4+ cell population. Collectively, our work demonstrates that downregulation of miRNA-451a upregulates ETS1 expression in CD4+ T cells, which may contribute to Th2 cell differentiation in pediatric asthma

Rationale: Cyclic dinucleotides (cDNs) are a promising class of immunotherapeutic agent targeting stimulator of interferon genes (STING). However, enzymatic instability and transmembrane barriers limit the extensive clinical application of cDNs. Thus, a novel delivery system, composed of a neutral cytidinyl lipid DNCA and a cationic lipid CLD (Mix) that interacts with cDNs via H-bonding, pi-stacking and electrostatic interaction, is developed and optimized to overcome the above issues. Methods: The optimal composition of Mix for cDNs encapsulation was explored with RAW-Lucia ISG cells. The physicochemical properties of resulted nanoparticles were characterized. To validate the anti-tumor immunity of cDNs/Mix both in vitro and in vivo , immunogenic cell death (ICD) related markers and tumor inhibition efficacy were evaluated in cancer cells and tumor models, respectively. The mechanism by which cdG/Mix exerted the antitumor effects was explored by flow cytometric analysis and in vivo depletion. Results: Based on our developed and optimized delivery system, neutral cytidinyl lipid DNCA/cationic lipid CLD (Mix), cdG (500 nM in vitro , 1-10 μg in vivo )/Mix not only more potently stimulated production of IFNβ and related cytokines including CXCL9 and CXCL10, promoted ICD, led to NK and CD8 + T cell activation, inhibited tumor growth in both EO771 and B16F10 models and increased their survival rate (~43%), but also obviously reversed the T cell exhaustion (Tex) in tumor, meanwhile down regulated the mRNA expression of Tox and Nr4a , which are key regulators of Tex. Conclusion: cdG/Mix triggered ICD in various cancer cells and reversed the Tex systemically in tumor-burden mice, which would be a promising alternative strategy for cancer immunotherapy.

Background Chronic nonbacterial prostatitis (CNP) has a high incidence, low cure rate, and unclear pathogenesis. Here, we aimed to systematically identify effective diagnostic and therapeutic targets for CNP.Methods Prostate tissues were obtained from established mouse models and negative controls and were used for mRNA array sequencing and immunohistochemistry (IHC) staining. Predominant pathways were identified based on pathway enrichment analysis and pharmaceutical experiments. We also investigated the functional role of CXCL12 on CP, a critical factor belonging to the predominant chemotaxis pathway, and employed IHC staining to explore the influence of the CXCL12/CXCR4 axis on the activation of the NF-κB, AKT, and STAT3 signaling pathways. Serum samples derived from both CNP cases and healthy controls were used to determine the secretion level of CXCL12.Results By employing mRNA array sequencing and immunohistochemistry, we found that CXCR4, CXCL12, CD44, and OFLM4 were highly expressed in the infiltrated inflammatory T cells of the prostate tissues generated from CNP mice, while they were rarely expressed on the epithelial cells. Based on the pathway enrichment results, we applied pathway inhibitors to suppress the activity of these classic pathways. We found that targeting the CXCL12/CXCR4 axis with its specific antagonist AMD3100 remarkably alleviated inflammatory infiltration of the prostate in CNP models. Similar results were obtained when we replaced AMD3100 with adenovirus-associated virus (AAV)-shCxcl12. To clarify the potential mechanisms of how the CXCL12/CXCR4 axis influences the pathogenesis of CNP, we tested the classical downstream pathways. The results suggested that p-Akt, p-STAT3, and p-NF-κB were more highly expressed on the inflammatory cells of the prostate derived from the CNP model and were partly suppressed after applying AMD3100 or delivering AAV-shCxcl12, indicating that the CXCL12/CXCR4 axis potentially functioned through AKT/NF-κB and STAT3 signaling to influence the pathogenesis of CNP.Conclusion Our study provides potential diagnostic biomarkers and therapeutic targets for CNP.

The broader clinical application of Bispecific T-cell engagers (BiTEs) is hindered by their short half-life, on-target off-tumor toxicity, and limited therapeutic effect for solid tumors. Herein, we constructed a bimetallic-enriched triple-kill nanobomb manganese/Co 2+ -dopamine@BiTE/HPT (MnO 2 /Co-DA@BiTE/HPT) based on metal-polyphenol to improve the immunosuppressive tumor microenvironment by activating innate and adaptive immunity, thereby enhancing the treatment efficacy of BiTEs (PD-L1/CD3). A hyaluronic acid-modified PD-L1 aptamer (HPT) was introduced to improve the active targeting of the nanobombs and bind with PD-L1 overexpressing colorectal cancer. Bimetallic (Mn 2+ /Co 2+ ) activated the STING pathway; simultaneously, photothermal therapy (PTT) induces DNA fragmentation to cooperate with bimetallic to amplify the STING signal to “heat” the “cold” tumor microenvironment. The “hot” tumor with a large amount of T-cell infiltration facilitated BiTE recruitment of T cells to kill tumor cells. Furthermore, the efficient therapeutic potency of the triple-kill nanobombs (STING, BiTE, and PTT) was determined in subcutaneous colorectal cancer, distal, lung metastasis, and postoperative recurrence models, which indicated that MnO 2 /Co-DA@BiTE/HPT could improve the immune microenvironment, produce long-term immune memory, inhibit tumor growth, and prevent tumor recurrence and metastasis.

Antigen-enriched tumor vaccines represent a promising strategy for cancer immunotherapy. Nevertheless, their clinical efficacy is often limited by insufficient antigen presentation and low immunogenicity. Here, a novel nanovaccine was developed that synergistically combines mitochondrial fission inhibition, tumor-dendritic cell fusion, and metal-phenolic nanoparticle technology. Tumor cells were pretreated with the mitochondrial fission inhibitor Mdivi‑1, yielding antigen‑enriched cell membranes (CM(M)). Subsequently, these CM(M) were fused with the dendritic cell membranes (DCM) overexpressing B7 biomolecules to create a tumor-dendritic cell fusion membrane (DCCM(M)). R848-loaded metal-phenolic nanoparticles (TF/R848) were further coated with the fusion membrane to create a dual-functional nanovaccine (TF/R848@DCCM(M)). This nanovaccine increases immunogenicity and optimizes antigen presentation. The nanovaccine promotes dual T-cell activation via direct antigen presentation and DC-mediated cross-presentation. TF/R848@DCCM(M) showed remarkable anticancer activity in melanoma-bearing mouse models, greatly reducing tumor development and extending longevity. Moreover, the nanovaccine enhanced the effects of immune checkpoint inhibitors, offering a promising strategy for combination immunotherapy. This study presents a novel, efficient platform for cancer immunotherapy, positioning a versatile and powerful approach for next-generation tumor vaccines.

Altered Treg/Th17 balance indicated disturbed T-cell homeostasis in offspring. • Maternal TBBPA exposure was associated with colonic inflammation in F1 offspring. • Maternal TBBPA exposure was associated with altered gut microbiota and SCFA profiles. • Exploratory metabolomics identified bile acid biosynthesis as a top-ranked pathway. Tetrabromobisphenol A (TBBPA), a widely used brominated flame retardant, has been associated with adverse health effects. An exploratory NHANES analysis of urinary bisphenol A (BPA) was performed to provide general population-level context for bisphenol-related metabolic and inflammatory associations because urinary TBBPA measurements were unavailable. We investigated whether maternal TBBPA exposure (170 mg·kg⁻¹·day⁻¹) was associated with developmental alterations in offspring intestinal homeostasis. Maternal TBBPA exposure was associated with colonic inflammatory features, increased pro-inflammatory cytokine expression, and reduced Treg/Th17 ratios in F1 offspring. Gut microbiota composition was altered, including phylum- and genus-level differences, accompanied by altered fecal short-chain fatty acid composition and concentrations. Exploratory untargeted metabolomics identified nominally altered MS/MS-supported putatively annotated metabolite features. Primary bile acid biosynthesis had the lowest raw pathway-level p value and was therefore considered the top-ranked exploratory pathway, although no pathway remained significant after pathway-level FDR correction. Collectively, maternal TBBPA exposure was associated with developmental alterations in gut microbiota composition, microbial metabolites, fecal metabolic profiles, and intestinal T-cell homeostasis in F1 offspring, providing insight into the potential developmental intestinal immunotoxicity of TBBPA. Download: Download high-res image (223KB) Download: Download full-size image Tetrabromobisphenol A (TBBPA) is a widely used brominated flame retardant that is now ubiquitously detected in environmental media, wildlife, and humans. Due to its persistence, widespread use, and potential for maternal transfer, TBBPA may pose sustained risks during sensitive developmental periods. Increasing evidence links TBBPA exposure to developmental, metabolic, and immune-related adverse effects, raising concern over its long-term health implications. Its environmental ubiquity and potential developmental toxicity underscore the need for continued monitoring, exposure control, and strengthened environmental risk assessment.

The escalating demand for multifunctionality in carbon fiber reinforced poly(ether ether ketone) (CF/PEEK) composites has intensified the need for next-generation structural materials. However, achieving a synergistic integration of functional performance and structural integrity—rather than a trade-off—remains a critical challenge. Herein, tetrapod-shaped ZnO whiskers decorated with 2D Ti 3 C 2 T x MXene nanosheets (T-ZnO@MXene) were fabricated via electrostatic self-assembly and incorporated into CF/PEEK composites. This 3D conductive/enhanced network enabled the simultaneous enhancement of mechanical properties, electromagnetic interference (EMI) shielding, and electrothermal deicing capabilities. The optimized CF/T-ZnO@MXene/PEEK composites exhibited a flexural strength of 1970.0 MPa, a flexural modulus of 142.3 GPa, and an interlaminar shear strength of 123.2 MPa, representing improvements of 33.4 %, 45.1 %, and 56.7 % over pristine CF/PEEK, respectively. Concurrently, the through-thickness electrical conductivity surged for 7.7 S/m, corresponding to a 76-fold enhancement, while the EMI shielding effectiveness attained 56.7 dB (a 40.7 % increase). In addition, the composites demonstrated efficient electrothermal conversion and reliable deicing performance. Overall, this work provides an effective route to harmonize structural robustness with multifunctional capabilities in CF/PEEK composites, opening new opportunities for advanced aerospace applications.

Background The immunosuppressive microenvironment and limited immune cell infiltration into the tumor bed contribute to the proliferation, metastasis, and invasion of breast cancer (BC) cells. Reprogramming the tumor immune microenvironment has emerged as a promising therapeutic target for BC, but remains challenging in clinical practice. Artemisia annua , a medicinal plant, has shown immune-enhancing and anti-tumor activities, although its potential therapeutic applications in BC remain underexplored. Methods Extracellular vesicles (EVs) were isolated from fresh Artemisia annua using gradient centrifugation and characterized by transmission electron microscopy (TEM), nanoparticle tracking analysis (NTA), Zetasizer Nano ZS90, ultra-high-performance liquid chromatography-mass spectrometry (UHPLC-MS/MS), and high-performance liquid chromatography (HPLC). Single-cell RNA sequencing (scRNA-seq) analysis was performed to investigate the mechanism of AEVs on tumor growth in vivo and mRNA sequencing (mRNA-seq) was employed to further explore the mechanism of AEVs-induced polarization shift from M2-like to M1-like macrophages. In vitro and in vivo assays were conducted to assess the polarization of macrophages and recruitment of T lymphocytes into the tumor bed. Results AEVs were successfully isolated and characterized. In vivo, AEVs inhibited tumor growth by shifting macrophage polarization from the M2-like to the M1-like phenotype, and synergistically enhancing the recruitment of CD8 + and CD4 + T cells into the tumor microenvironment. AEVs activated the NF-κB signaling pathway while inhibiting the PPARγ pathway, thereby promoting the M1-like polarization of macrophages. Polarized M1-like macrophages secreted chemokines (CCL5 and CCL3), which facilitated T lymphocyte infiltration into the tumor bed. Notably, AEVs reshaped the BC immune microenvironment without inducing systemic toxicity. Conclusions AEVs from Artemisia annua efficiently reprogrammed the immune microenvironment of breast cancer by inducing macrophage polarization and enhancing T lymphocyte infiltration. This study lays the foundation for using AEVs as a potential immunotherapy for BC and highlights the medicinal value of Artemisia annua in cancer treatment.

T-2 toxin, a trichothecene mycotoxin produced by Fusarium species, is a global cereal contaminant and an important environmental risk factor for cartilage damage and Kashin–Beck disease (KBD). Although hydrolysis is a major metabolic pathway of T-2 toxin, the human enzymes responsible and their toxicological relevance remain poorly understood. In this study, human liver microsomes, recombinant hydrolases, and selective inhibitors were used to identify the key enzymes mediating T-2 hydrolysis, and LC–MS/MS was applied to quantify T-2 and its major metabolites. Human chondrocytes were further used to evaluate the cytotoxicity of T-2 and its hydrolytic metabolites (HT-2 toxin, neosolaniol, and T-2 tetraol). T-2 toxin was predominantly hydrolyzed to HT-2 toxin, primarily by carboxylesterases (CES1, CES2), with inhibitor-based evidence supporting a potential role for arylacetamide deacetylase (AADAC). The toxicity ranking was T-2 toxin > HT-2 toxin > neosolaniol > T-2 tetraol. T-2 toxin markedly impaired autophagy and extracellular matrix metabolism in chondrocytes, whereas its metabolites showed weaker cytotoxicity. These findings indicate that human CES1 and CES2, together with other hydrolases including AADAC, contributing to the hydrolysis and detoxification of T-2 toxin and may represent potential targets for mitigating T-2–associated cartilage damage and KBD risk.
