Novel chitosan-based composite film with Luteolin-SOD nano-micelles via nano-encapsulation for food preservation. • This composite film outperforms conventional packaging: superior inhibition of oxidation, delays spoilage, biodegradable. • Natural-ingredient film: promising substitute for traditional packaging, meeting eco-friendly and sustainable demands. Chitosan-based films are promising alternatives to petroleum-based food packaging materials but suffer from insufficient mechanical strength, poor water resistance, and limited antioxidant capacity. To address these issues, we synthesized chitosan-based nano-polymers loaded with luteolin (Lut) and superoxide dismutase (SOD) (CLS nano-micelles). We then incorporated these nano-micelles into a chitosan-based film to produce a CLS film. The results showed that, compared with the pure chitosan film, the mechanical properties of CLS film significantly improved, with tensile strength increasing from 6.14 MPa to 37.50 MPa and elongation at break increasing from 17.4% to 34.6%. The barrier properties of the CLS film were also enhanced, as both water vapor permeability and solubility decreased. Notably, the CLS film also exhibited excellent antioxidant properties (with DPPH inhibition rates increasing from 4.4% to 48.3% and ABTS inhibition rates rising from 7.2% to 74.6%) and demonstrated potent antibacterial activity against E. coli , S. aureus , and B. subtilis . It also exhibited excellent biocompatibility and demonstrated favorable biodegradability in soil. Grape preservation assays further confirmed its efficacy in maintaining fruit freshness. These results demonstrate that integrating CLS nano-micelles into a chitosan matrix significantly improves its mechanical, barrier, antioxidant, and antibacterial properties. This makes the CLS film a promising eco-friendly alternative for sustainable food packaging applications, offering an efficient and environmentally friendly solution. Amphiphilic micelles are widely used in biomedicine, which contain a hydrophilic shell and a hydrophobic core. In this study, phenylalanine poly (ester amide)s (Phe-PEA) were synthesized by solution polycondensation of phenylalanine, di alcohol, and dicarboxylic acid, which were biodegradable, environmentally friendly, and nontoxic. Phe-PEA was connected with dextran of different molecular weights to obtain a series of block polymers of different lengths (Dex-Phe-PEA). The results revealed that Dex-Phe-PEA could self-assemble into amphiphilic micelles in water with a stable spherical shape and uniform size distribution (Đ < 0.4). The micellar system exhibited negligible cytotoxicity (cell viability >90% at 80 μg/mL concentration) and enzymatic degradability. To verify the possibility of these micelles as drug carriers, the hydrophobic anticancer drug curcumin (Cur) was encapsulated in the hydrophobic core. The micelle exhibits a high encapsulation efficiency (66–84%) and drug loading (4.2–9.9%). Cur-loaded micelles showed significant inhibitory effects on HeLa cells (cell viability <34% at 80 μg/mL concentration). In vitro drug release experiments proved that the material is pH sensitive [the micelles exhibited faster Cur release at a cancerous pH (5.4, 52–62%) than at a physiological pH (7.4, 29–43%)], and is an excellent anticancer drug carrier.

To address the challenges of poor stability and weak targeting capability in polymeric micelles, which negatively impact controlled drug release performance, pH-responsive polymers PCL-PDEAEMA-PHEMA-PPEGMA (P1, P2, and P3) with distinct block lengths were designed and synthesized in this work. These copolymers were rigorously characterized using 1 H NMR, FT-IR, and GPC. Subsequently, polymer P3 was selected for modification based on comparative analyses of critical micelle concentrations (CMC) and drug-loading performance. Polymer P3 underwent esterification with lipoic acid (LA) and folic acid (FA) to yield LA-P3 and FA-P3, respectively. These derivatives were blended at a mass ratio of 1:1 (LA-P3:FA-P3), and dithiothreitol was employed to initiate self-crosslinking, resulting in the preparation of actively targeted crosslinked mixed micelles (C-MIX). The results revealed that C-MIX@PTX drug-loaded micelles prepared at a feeding ratio of 15 mg PTX/30 mg polymer exhibited a smaller size (134.7 nm) compared to non-crosslinked micelles, with a drug-loading content of 15.31 ± 1.79% and an encapsulation efficiency of 38.42 ± 5.22%. In vitro release studies demonstrated that under neutral conditions, C-MIX@PTX achieved a cumulative drug release of 16.8% at 120 h, significantly lower than non-crosslinked micelles. Under acidic/reductive conditions, C-MIX@PTX showed a cumulative drug release of 77.1% at 120 h, slightly lower compared to non-crosslinked micelles. Cytotoxicity assays confirmed that both MIX and C-MIX were biocompatible low-toxicity carrier materials, while LA-P3@PTX, FA-P3@PTX, MIX@PTX, and C-MIX@PTX exhibited inhibitory effects on HepG2 cells, FA-P3@PTX with folate-modified demonstrated enhanced efficacy.

Excessive ROS production triggers diabetic keratopathy (DK), causing dry eyes and impaired healing. Commercial Rebamipide (Reb) eye drops (Mucosta®) suffer from poor bioavailability, short retention, and irritation. To overcome these limitations, this study developed Reb-loaded nanomicelles in ion-sensitive in-situ gels (Reb-NM-ISGs). This dual-approach combines micelles for enhanced solubility and corneal penetration with a gel that forms upon contact with tear fluid, significantly prolonging drug retention. In diabetic models, Reb-NM-ISGs effectively treated dry eye symptoms (improving tear volume, prolonging tear film break time (TBUT)) and corneal injury, accelerating epithelial healing while significantly suppressing key inflammatory cytokines (IL-1β) and alleviating oxidative stress (reducing ROS and MDA, increasing GSH). Safety assessments confirmed good tolerance. Therefore, Reb-NM-ISGs represent a highly promising, safe, and patient-friendly strategy for managing DK, offering a significant advancement over current Reb therapy and providing a valuable reference for diabetic keratopathy research and ocular nanomedicine.

Combination therapy holds great promise for enhancing antitumor efficacy by achieving tumor-targeted and sequentially synergistic cytotoxicity. We developed a tumor-targeted hybrid micellar nanoplatform that co-delivers the chemotherapeutic agent pemetrexed (PEM) and carbon monoxide (CO) for synergistic chemo-gas therapy. PEM and Fe₃(CO)₁₂ were covalently conjugated to D-α-tocopheryl polyethylene glycol succinate (TPGS), yielding two amphiphilic prodrugs—TPGS-PEM and TPGS-CO—which were co-assembled into hybrid micelles. The presence of PEM enabled folate receptor-mediated active targeting, resulting in efficient uptake by 4T1 breast cancer cells. Intracellular esterase-catalyzed cleavage of the ester bond released PEM, which elevated reactive oxygen species (ROS) levels. The increase in ROS triggered site-specific CO release from the Fe–CO complex. This sequential mechanism enabled a cascade of tumor-selective chemotherapy and gas therapy, leading to enhanced apoptosis and reduced tumor proliferation. The hybrid micelles exhibited superior tumor suppression, reduced systemic toxicity, and prolonged survival in tumor-bearing mice.

Rheumatoid arthritis (RA), a chronic autoimmune disorder affecting 1% globally, urgently demands advanced therapies to overcome the systemic toxicity and limited efficacy of conventional glucocorticoids like dexamethasone (Dex). In this study, we constructed LMWH-Gly-Pro-ODA/Dex (LGPO/Dex) micelles, which synergistically integrate low molecular weight heparin (LMWH)-mediated active targeting with Gly-Pro-mediated fibroblast-activated protein (FAP)-α-responsive drug release to achieve spatiotemporal precision in RA treatment. The system operates through a three-stage cascade mechanism: (1) targeting the inflamed joints, (2) inflammation-responsive drug release to modulate pathological microenvironments (e.g., normalizing M1/M2 macrophage polarization), and (3) suppression of monocyte recruitment to prevent disease progression. LGPO/Dex micelles showed excellent RA therapeutic effects in the adjuvant-induced arthritis (AIA) model. Joint swelling, serum TNF-α, and nitric oxide (NO) levels in LGPO/Dex-treated rats showed no significant difference from healthy controls (ns) while exhibiting marked improvement over Dex monotherapy (**, P < 0.01). Notably, it also significantly reduced Dex-associated adverse effects. This study confirmed the feasibility of using FAP-α as a therapeutic target for RA and provided a new idea for RA treatment, offering a blueprint for disease-microenvironment-programmed therapeutics.

This study was designed to investigate the bioavailability and targeting of myricetrin-loaded ternary micelles modified with and without TPGS. The particle diameters of myricetrin-loaded micelles and myricetrin-loaded-TPGS micelle were 30.93 ± 1.34 nm and 26.42 ± 0.89 nm, respectively, while their respective encapsulation efficiencies (m/m, %) were 83.3 ± 1.08 and 93.8 ± 1.18. The release rate of myricetrin in the micellar system clearly exceeded the free myricetrin in the three media (pH 6.8 phosphate buffer, pH 1.2 HCl solution and double distilled water). In vivo studies displayed that the bioavailability of myricetrin mixed micelles was remarkably improved than the free drug after oral administration. Moreover, the results of tissue distribution showed that myricetrin-loaded-TPGS micelles accumulated well in the liver tissue. Based on these results, it was speculated that myricetrin-loaded-TPGS micelles might act as a promising carrier for liver targeting with improved hepatic concentration of myricetrin compared with the myricetrin-loaded micelles.

Fiber membranes with high specific surface area can be produced when using electrospinning technology combined with amphiphilic block polymers. Due to the solubility difference between the two segments of amphiphilic block polymers, it is easy to be self-assembly in solvents, which affects the morphology of the spinning fiber and the surface properties of the membrane. In this work, the relationship between the self-assembly of the amphiphilic block polymer and the morphology of the spinning fiber and the wetting performance of the spinning membrane was explored. Membrane is produced by electrospinning amphiphilic block copolymer PS-b-PHEMA (polystyrene-block-poly(2-hydroxyethyl methacrylate) by using co-solvent system. PS-b-PHEMA with controlled molecular weight (Mw) and polymer dispersity index (PDI) was synthesized via atom transfer radical polymerization (ATRP) and characterized by nuclear magnetic resonance (NMR) and gel permeation chromatography (GPC). The electrospun membrane have both super-hydrophobic and super-hydrophilic by adjusting the ratio of mixed solvents, demonstrating that the wetting performance of the fiber membrane mainly depends on the shell structure of the self-assembly micelles.

Objective: Inhibition of tumor angiogenesis has become a new targeted tumor therapy. In this study, we established a micellar carrier with a tumor neovascularization-targeting effect modified by the neovascularization-targeting peptide NGR. Methods: The targeted polymer poly(ethylene glycol)-b-poly(lactide-co-glycolide) (PEG-PLGA) modified with Asn–Gly–Arg (NGR) peptide was prepared and characterized by 1 H nuclear magnetic resonance and Fourier-transform infrared spectrometry. NGR-PEG-PLGA was used to construct curcumin (Cur)-loaded micelles by the solvent evaporation method. The physicochemical properties of the micelles were also investigated. Additionally, we evaluated the antitumor efficacy of the polymer micelles (PM) using in vitro cytology experiments and in vivo animal studies. Results: The particle size of Cur-NGR-PM was 139.70 ± 2.51 nm, and the drug-loading capacity was 14.37 ± 0.06%. In vitro cytological evaluation showed that NGR-modified micelles showed higher cellular uptake through receptor-mediated endocytosis pathways than did unmodified micelles, leading to the apoptosis of tumor cells. Then, in vivo antitumor experiments showed that the modified micelles significantly inhibited tumor growth and were safe. Conclusions: NGR-modified micelles significantly optimized the therapeutic efficacy of Cur. This strategy offers a viable avenue for cancer treatment. Export

Recently, we have reported the vesicle formation and vesicle-to-micelle transition (VMT) of sodium 2-ketooctanoate, a single-tailed keto-acid salt, in water with increasing concentration. Herein, we demonstrate that, upon prolonged standing at the vesicle-forming concentration, macroscopic fibrous structures spontaneously formed. Cryogenic transmission electron microscopy, atomic force microscopy, and laser scanning confocal microscopy revealed the microscopic pathway: vesicles aggregated and evolved through elongated intermediate structures (threadlike vesicles and chains of vesicles) into long strips, which ultimately assembled into floating fibers. As concentration was increased around the critical micelle concentration (CMC), these fibers transformed into bundles of nanofibers and threadlike micelles, and finally evolved to globular micelles above the CMC. The observed multi-step aggregation behavior is driven by the dynamic coexistence and rearrangement of multiple species, notably “acid-soap” dimers. This constitutes the first report detailing the microscopic transition pathway from fibrous structures to micelles for a single-tailed keto-acid salt amphiphile in water.

A new kind of negatively charged polymer-shielded supramolecular nano-micelles with dual-responsive property was designed for tumor treatment, which was prepared on the basis of adamantane terminated linear PAsp(DIP) and disulfide-β-cyclodextrin-terminated PAsp(EDA). The supramolecular nano-micelles comprised a 2,3-dimethylmaleic anhydride (DA) protective layer to stabilize the micelles , a pH-responsive core to package hydrophobic model drugs, and a disulfide-crosslinked interlayer to shackle the core against drug leakage under normal physiological conditions. After arriving at the tumor tissue via EPR, the targeting function could be turned on by dislodging DA groups on the surface of micelles , which allowed the drug-loaded nano-micelles to be easily phagocytized by the tumor cells, and then release the drug inside the cells induced by the increased glutathione level and acidic pH. The results indicated that the charge-conversional dual-responsive supramolecular nano-micelles showed excellent antitumor activity.

We have synthesized an electro-active amphiphilic copolymer with carbazole side chains via free radical polymerization using 7-(4-vinylbenzyloxy)-4-methyl coumarin and 9-(4-vinylbenzyl)-9H-carbazole as the monomers. The copolymer can self-assemble to form micelles (termed EACMs) in aqueous solution and can adsorb onto the surfaces of MWCNTs via π–π interactions and thereby cause the efficient dispersion of the MWCNTs in aqueous solution. The coumarin groups in the copolymer undergo UV-induced photo-crosslinking, which further improves the stability of the suspension. Moreover, the electro-active carbazole moieties in the EACMs can undergo electropolymerization to form a conducting network on the MWCNTs that significantly accelerates electron transfer. The EACM/MWCNTs hybrid was applied to the amperometric sensing of dopamine (DA) as a model analyte. After electropolymerization, the electrode exhibited good sensitivity and selectivity toward the determination of dopamine with a 0.2 μM detection limit and a wide linear range. The method described here provides a viable route to water-dispersible and stable carbon nanotubes while preserving their outstanding electrical properties. We presume that the composite described here represents a valuable tool for the construction of electrochemical sensors and electronics.
