Red radish anthocyanin-based intelligent starch/pectin films for shrimp sub-freshness monitoring: effect of adjusting pH value of the film-forming solution

Ma Ye,Wan Jie,Luo Shunjing,Xiong Yong

In this study, intelligent films based on red radish anthocyanin as indicator and corn starch/citrus peel pectin as the matrix were developed by adjusting the pH value (3.0, 4.0, 5.0, 6.0 and 7.0) of the film-forming solution. The microstructure and infrared spectral characteristics of the films were elucidated, and their physical properties were assessed, including light barrier properties, hydrophobicity and mechanical strength. Furthermore, the films’ responsiveness to pH and ammonia was evaluated, and their application in monitoring the freshness of shrimp during storage was investigated. The results indicated that the cross-sectional distribution of the films prepared at pH 3.0 and pH 7.0 exhibited unevenness and poor compatibility, which adversely affected their mechanical and hydrophobic properties. All films demonstrated significant color changes in response to pH and ammonia, with the films fabricated at pH 3.0 and pH 4.0 displayed more pronounced color transitions. Notably, the film prepared at pH 4.0 exhibited the highest tensile strength (3.24 MPa) and could visually distinguish between fresh (red, 1 day), sub-fresh (pink, 2 days), and spoiled (purple, 3 days) shrimp during storage at 4 °C over a period of 4 days. Thus, this study presents a novel starch/pectin film based on red radish anthocyanin for shrimp sub-freshness monitoring by adjusting pH of the film-forming solution.

Pectin-coated hollow organosilicon construction of a dual pH and pectinase stimulation-responsive prochloraz delivery system for the control of rice sheath blight

Yibing Yan,Songye Li,Shanshan Wang,Yifan Peng,Kaikun Pi,Bin Sun,Liang Cheng,Bing Liu,Fangxiang Song

BACKGROUND Rice sheath blight, caused by Rhizoctonia solani ( R. solani) poses a significant threat to global rice production. Although prochloraz (PRO) is an effective fungicide, its poor photostability and low water solubility severely limit its field performance. We developed a sustainable, stimuli-responsive delivery system to enhance PRO efficacy and minimize environmental risks. RESULTS Multilayered mesoporous nanoparticles (MSN@PMO) were synthesized via a hard-template route and converted into hollow periodic mesoporous organosilicas (H-PMO) by sodium carbonate etching. PRO was encapsulated in H-PMO and sealed with pectin, yielding a dual pH/pectinase responsive formulation (Pec@PRO-H-PMO, PPHP). PPHP exhibited sustained release for 144 h under acidic and enzymatic conditions, and showed markedly improved photostability, foliar adhesion and rainfastness compared with PRO technical concentrate (PRO TC) and PRO water emulsion (PRO EW). Bioassays demonstrated that PPHP exhibited 1.8 times greater antifungal activity against R. solani compared to PRO TC. Ecotoxicological analysis revealed 11.9 times higher zebrafish median lethal concentration (LC₅₀) values than PRO TC over 80% viability of human bronchial epithelial (BEAS-2B) cells at 18.4 mg/L, suggesting improved biosafety. CONCLUSIONS The dual pH/pectinase triggered release of PPHP enhanced pesticide utilization and improved safety. This strategy presents a novel approach for developing stimuli-responsive pesticide delivery systems, aiming to strengthen crop disease control efficiency and mitigate environmental risks. © 2025 Society of Chemical Industry.

Food-grade Pickering emulsions and high internal phase Pickering emulsions encapsulating cinnamaldehyde based on pea protein-pectin-EGCG complexes for extrusion 3D printing

Tingting Feng,Chunli Fan,Xuejiao Wang,Xingwei Wang,Shuqin Xia,Qingrong Huang

Plant protein-based edible Pickering emulsions (PEs) and high internal phase PEs (HIPPEs) for three-dimensional printing (3DP) and flavor compounds delivery were investigated. The effects of cinnamaldehyde proportions in oil phase (0%, 25%, 50%, 75% and 100%) on the storage stability, rheological properties, 3DP characteristics and interfacial behavior of PEs (oil phase volume φ of 0.52) and HIPPEs (φ of 0.83) were discussed. PEs and HIPPEs with cinnamaldehyde proportions of 0% and 25% prior to printing exhibited an excellent storage stability against coalescence and creaming after at 4 °C, 25 °C and 45 °C respectively for 30 days. The rheological data showed that PEs containing 0–25% cinnamaldehyde and HIPPEs containing 0–75% cinnamaldehyde had a viscoelastic solid-like response with high storage moduli and suitable viscosity, providing emulsion-based inks with strong support performance and smooth extrusion characteristics. For the printed cuboids, cylinders and turtles based on PEs, the distinct structural collapse and profile loss of fidelity occurred when the cinnamaldehyde proportion was over 25%. However, for HIPPEs with the increase of cinnamaldehyde proportion up to 75%, the printed lines, cuboids, cylinders and turtles presented acceptable 3D printability, superior gel strength and texture properties, which showed smooth surfaces, regular shapes and refined 3D structures. Besides, the loss rate of cinnamaldehyde as the flavor component reduced within 10.02 ± 0.01% and 11.29 ± 0.01% respectively during the 3DP processing. Plant protein-based PEs and HIPPEs could be potential materials for food 3DP with enhanced the flavor retention, thus providing guidance for the practical applications in food 4D printing.

Anti-apoptotic effects of high hydrostatic pressure treated cyanidin-3-glucoside and blueberry pectin complexes on lipopolysaccharide-induced inflammation in Caco-2 cells

Chang Tan,Yanwen Kong,Yuqi Tong,Haotian Deng,Mingyue Wang,Yang Zhao,Meizhi Wan,Sixu Lin,Xiaoxu Liu,Xianjun Meng,Yan Ma

To investigate whether complexes of cyanidin-3-glucoside (C3G) and blueberry pectin (BP) can exert protective effects on lipopolysaccharide (LPS)-induced Caco-2 inflammatory cells, as well as the influence of high hydrostatic pressure (HHP) processing on the activities of the complexes, for determining their anti-apoptosis in vitro . LPS-induced Caco-2 cells were treated with C3G-BP complexes with or without HHP treatment. Results showed that C3G-BP complexes (100–100 µg/mL) inhibited the depolarization of mitochondria and reduced the production of ROS, as demonstrated by the reduced mRNA expression of IL-1β, TNF-α, and IL-8; increased expression of IL-10; and reduced iNOS, COX-2, Bcl-2, and cleaved caspase-3 levels. These findings reveal that C3G-BP exerts anti-apoptosis effects by inhibiting several mediators and cytokines involved in the inflammatory process. Furthermore, HHP-treated C3G-BP complexes better reduced cell apoptosis than untreated complexes, owing to the high pressure-mediated alterations in the monosaccharide composition of the complexes. This study provides insights for the functional changes of anthocynin-pectin complexes during food processing applications.

Gelatin-EGCG-high methoxyl pectin ternary complex stabilized W1/O/W2 double emulsions loaded with vitamin C: Formation, structure, stability, in vitro gastrointestinal digestion

Xiang Huang,Rui Tu,Hongbo Song,Kai Dong,Fang Geng,Lei Chen,Qun Huang,Yingmei Wu

Vitamin C is an essential nutritional supplement and antioxidant in food. However, the development of vitamin C in the food industry is limited due to its extremely poor chemical stability. In this study, W 1 /O/W 2 double emulsions loaded with vitamin C were prepared, and their structure, physicochemical stability, and in vitro gastrointestinal digestion were investigated. The results manifested that the encapsulation efficiency was the highest (90.23 ± 0.49 %) when the addition of vitamin C in the internal aqueous phase was 0.05 %. Storage stability revealed that no phase separation occurred and did not show stratification in the emulsion system during storage, and the physical stability was excellent. pH stability demonstrated that the W 1 /O/W 2 double emulsion loaded with vitamin C had a lower polydispersity index (PDI) value (0.19 ± 0.01) and greater absolute zeta potential value (40.37 ± 0.48) in alkaline environment (pH 8.0–12.0). The ionic stability suggested that the double emulsion was less stable in the presence of sodium ions. In vitro gastrointestinal digestion indicated that the bioavailability of vitamin C was 25 % after simulated digestion in vitro , elucidating that the W 1 /O/W 2 double emulsion loaded with vitamin C was released slowly in the small intestine and had a certain sustained-release function.

Influence of starch with different degrees and order of gelatinization on the microstructural and mechanical properties of pectin cryogels: A potential pore morphology regulator

Shuhan Feng,Jianyong Yi,Youchuan Ma,Jinfeng Bi

The applications of cryogels are defined by their porous morphology as well as mechanical properties. To achieve efficient regulation of porous properties for pectin cryogels, we selected starch as a potential polysaccharide regulator. Pectin/starch composite cryogels with different degrees of gelatinization were formulated, and two ways of starch gelatinization were considered: starch gelatinization occurred before or after pectin crosslinking during forming the hydrogel network. The results showed that high gelatinized starch (73.8 %–100.0 %) rendered pectin cryogels with denser pore morphology and higher mechanical strength. The pore diameter transferred from 160–200 μm to 40–60 μm with the degree of gelatinization, while the total porosity decreased by about 15 % and the specific surface area increased by about 100 m 2 /g. When starch gelatinization occurred before pectin crosslinking, the hydrogen bond interactions between gelatinized starch and pectin were formed to accelerate the gelation rate of the pectin Ca 2+ -dependent network. When gelatinization occurred after pectin crosslinking, the pre-formed pectin network delayed the breakdown of the starch crystalline structure during gelatinization. The qualitative regulation of the pore morphology in pectin cryogels by incorporating starches with varying degrees of gelatinization was confirmed.

Facile synthesis of nano-nanocarriers from chitosan and pectin with improved stability and biocompatibility for anthocyanins delivery: An in vitro and in vivo study

Xue Zhao,Xuedi Zhang,Shanshan Tie,Shuai Hou,Haitao Wang,Yukun Song,Rewa Rai,Mingqian Tan

Anthocyanins is a type of natural pigment with a variety of physiological activities, are unstable and prone to degradation. In order to improve the stability of anthocyanin, in this study, the nanocarriers were constructed through electrostatic self-assembly method using chitosan and pectin as raw materials for bilberry anthocyanins delivery. Atomic force microscope and scanning electron microscope analyses showed that the nanocarriers were well dispersed spheres with diameters around 100–300 nm when the mass ratio of chitosan/pectin/anthocyanin was 1:1:3. Fourier transform infrared spectroscopy and ultraviolet–visible absorption spectroscopy results revealed that the anthocyanins were successfully doped into the nanocarriers. The encapsulation efficiency of anthocyanins was 66.68%. Moreover, the anthocyanin nanocarriers exhibited increased storage stability under light and dark conditions. In vitro digestion experiments showed that the doped anthocyanins could escape from the interference of stomach acid environment and released slowly in small intestinal fluid, suggesting the controlled delivery of anthocyanins in the gastrointestinal tract. In vitro NRK cell model demonstrated that the nanocarriers could protect against acrylamide-induced damage by viability and cell apoptosis assay. In vivo Caenorhabditis elegans model further showed that the nanocarriers could protect from anthocyanin damage caused by various harmful conditions such as acrylamide, oxidative stress, heat shock and UV light. Furthermore, Caenorhabditis elegans treated with nanocarriers showed longer lifespan, stronger reproductive capability, more flexible locomotion behavior, and less autofluorescent lipofuscin particles. Together, these results suggest that chitosan-pectin nanoparticles can be used as delivery vehicles to improve the stability and bioavailability of the functional factor anthocyanin.

Electrochemical biosensor based on enzyme substrate as a linker: Application for aldolase activity with pectin-thionine complex as recognization element and signal amplification probe

Xiaonan Wang,Meiwen Wang,Yuanyuan Zhang,Xiaocao Miao,Yuanyuan Huang,Juan Zhang,Lizhou Sun

A new strategy to fabricate electrochemical biosensor is reported based on the linkage of enzyme substrate , thereby an electrochemical method to detect aldolase activity is established using pectin-thionine complex (PTC) as recognization element and signal probe . The linkage effect of fructose-1,6-bisphosphate (FBP), the substrate of aldolase, can be achieved via its strong binding to magnetic nanoparticles (MNPs)/aminophenylboronic acid (APBA) and the formation of phosphoramidate bond derived from its reaction with p -phenylenediamine (PDA) on the surface of electrode. Aldolase can reversibly catalyze the substrates into the products which have no binding capacity with MNPs/APBA, resulting in the exposure of the corresponding binding sites and its subsequent recognization on signal probe. Meanwhile, signal amplification can be accomplished by using the firstly prepared PTC which can bind with MNPs/APBA, and accuracy can be strengthened through magnetic separation. With good precision and accuracy, the established sensor may be extended to other proteins with reversible catalyzed ability.

Polydopamine-coated montmorillonite micro/nanoparticles enhanced pectin-based sprayable multifunctional liquid mulching films: Wind erosion resistance, water retention, and temperature increase/heat preservation properties

Fangzhou Xu,Qianyu Dong,Shikai Zhang,Qicong Wu,Chunchun An,Xiaoqian Li,Xinchuang Chen,Yang Chen,Xincheng Zhang,Jinrong Li,Zhi Dong

Natural biopolymer-based liquid mulching films (LMF) have received widespread attention, whereas the fragile structure and limited functionality have severely restricted their application. Herein, polydopamine-coated montmorillonite micro/nanoparticles enhanced pectin-based sprayable multifunctional liquid mulching films (P-MMT@PDA LMF) were prepared. Dopamine has abundant active sites, and its self-polymerization onto the surface of MMT improves the compatibility of MMT with pectin chains, facilitates the homogeneous dispersion of MMT@PDA in pectin polymers, and makes them more tightly entangled through hydrogen bonding. Therefore, P-MMT@PDA LMF exhibits better mechanical properties (improved by 64.94 N) and wind erosion resistance (wind speed 30 m/s, >60 min). Moreover, MMT@PDA micro/nanoparticles can fill the voids of pectin chains, thus increasing the densification and complexity of the network structure in LMF, enabling better water retention (improved by 14.67 %) and heat preservation (increased by 3.14 °C). Meanwhile, the photothermal effect of PDA endows P-MMT@PDA LMF with a warming effect (increased by 2.84 °C). Hence, this LMF promote wheat growth and demonstrate good biodegradability. These results suggest that the application of P-MMT@PDA LMF is an effective strategy in the environments of drought and cold, which is expected to provide a green solution for sustainable agricultural development and environmental protection in the future.

A dual-target pectin-like polysaccharide LRP01B from Lamiophlomis rotata induces pancreatic cancer cell apoptosis in vitro and in vivo by causing mitochondrial dysfunction

Jianhao Xie,Juan Su,Wanqi Zhou,Simin Cai,Wenfeng Liao,Jianing Zong,Junfeng Peng,Huanhuan Qu,Fei He,Kan Ding

Pancreatic ductal adenocarcinoma (PDAC) is a highly malignant cancer with limited treatment options. Recent pharmacological studies suggested that Lamiophlomis rotata might contain components with antitumor activities. In this study, we aimed to investigate the potential of polysaccharides as active components in Lamiophlomis rotata . Our findings suggest that a polysaccharide, LRP01B, from Lamiophlomis rotata induces PDAC cells apoptosis by causing mitochondrial dysfunction and oxidative stress in vitro and in vivo . LRP01B is a homogeneous polysaccharide with a molecular weight of 15.9 kDa, composed of galactose, glucose, xylose, arabinose, rhamnose, and galacturonic acid in a molar ratio of 0.7: 0.3: 0.1: 1.1: 1.0: 4.3. The backbone of LRP01B mainly consists of 1,4-linked α-Gal p A, 1,2-linked α-Rha p , and 1,3-linked α-Gal p A, with existence of arabinose and xylose on the side chain. In vivo experiments demonstrate that LRP01B significantly inhibits pancreatic tumor growth in a patient-derived xenograft nude mouse model. Mechanism studies uncover that LRP01B induces apoptosis of PDAC cells through mitochondrial dysfunction and oxidative stress by recruiting TNF-related apoptosis-inducing ligand (TRAIL) to TRAIL-R1, which leads to the binding of LRP01B to both TRAIL and TRAIL-R1. Our results demonstrate that LRP01B is a dual-targeting polysaccharide with anti-PDAC activity and may be a promising anti-PDAC drug candidate.

In vitro study of bioaccessibility, antioxidant, and α-glucosidase inhibitory effect of pelargonidin-3-O-glucoside after interacting with beta-lactoglobulin and chitosan/pectin

Vemana Gowd,Naymul Karim,Lianghua Xie,Mohammad Rezaul Islam Shishir,Yang Xu,Wei Chen

Polysaccharides and fruit extracts are applied in dairy products to enhance their nutritional property, but the effects of such formulations on the functions and biological activities are yet to be explored. Therefore, this study was aimed at evaluating the effect of interactions among milk protein (beta-lactoglobulin; BLG), polysaccharides (pectin, P; chitosan, CH), and anthocyanin (pelargonidin-3- O -glucoside; P3G) in improving the bioavailability and biological activity of P3G. After gastrointestinal digestion (GID), the content of free P3G in different model solutions were as follows: P3G-alone (73.59 μg/mL), P3G-P (66.59 μg/mL), P3G-CH (36.72 μg/mL), P3G-BLG (64.92 μg/mL), P3G-P-BLG (64.92 μg/mL), and P3G-CH-BLG (39.61 μg/mL). Less amount of free P3G in model solutions indicated increased complex formation of P3G with protein and/or polysaccharides during GID. These complexes resulted in protection and progressive release of P3G in the gastrointestinal tract. Chitosan exhibited more protection to P3G compared with P and BLG. In addition, α-glucosidase inhibitory activity and ROS scavenging activities of conjugated-P3G samples were potentially augmented after GID. However, the presence of polysaccharides and protein in the model solutions did not show any negative effect on the biological activity of P3G. Thus, pure P3G can be used as a nutritional ingredient in dairy industries.

Application of pectin‑zinc oxide hybrid nanocomposite in the delivery of a hydrophilic drug and a study of its isotherm, kinetics and release mechanism

Arun K. Kodoth,Vivek M. Ghate,Shaila A. Lewis,Vishalakshi Badalamoole

Pectin-based gel and its nanocomposite with zinc oxide have been compared for their capacity to release Donepezil for the possible use as an implantable drug delivery platform for the treatment of Alzheimer's disease. Adsorption capacities of the samples were determined as a function of pH, temperatures, concentrations of the drug, and the mass of the adsorbent. The nanocomposite exhibited significant adsorption compared to the parent gel. Adsorption data for the nanocomposite system fits well with Langmuir model and followed pseudo-first order kinetics, while that of the parent polymeric system followed pseudo-second-order kinetics. Donepezil adsorbed polymeric samples were prepared and evaluated for tensile strength , swelling index, folding endurance and characterized by FTIR, FESEM, EDS , XRD and TGA techniques. The desorption of zinc oxide was also monitored using the dynamic light scattering technique. The in vitro drug release study indicated desorption of Donepezil to the maximum extent of ~88% and 46% during 5 days period from the nanocomposite and the parent gel respectively. The developed systems showed negligible (<10%) percentage of hemolysis after incubation with sheep erythrocytes. In conclusion, the developed pectin-based nanocomposite can be explored as a potential platform for the development of implantable drug delivery systems for chronic diseases.
