Host-guest interfacial recognition of alginate-based β-cyclodextrin/dendrobine supra-amphiphiles reinforced the physicochemical stability and sustained-release properties of Pickering emulsions

Tong Ye,Qile Chen,Shujuan Yang,Haifang Liu,Yan Zhou,Yuhong Feng,Jiacheng Li,Yujuan Wang,Gaobo Yu,Jun Dai

Achieving high bioavailability of dendrobine (DDB) necessitates the development of simplified available and efficient delivery systems. Pickering emulsions (PEs) derived from biomass represent a promising option. However, the physicochemical properties of PEs interfacial films were insufficient to prevent DDB leakage, thereby reducing bioavailability. Herein, a supramolecular host-guest interfacial recognition strategy was proposed in-situ between amphipathic sodium alginate-functionalized cyclodextrin (SAE-CD) and hydrophobic DDB at oil-water interface, further forming the SAE-based supra-amphiphiles to efficient stabilize the high internal phase Pickering emulsions (HIPPEs) with gel-like characteristics. A multiscale methodology was empolyed to investigate the interfacial assembly behavior and emulsification properties of supra-amphiphilic SAE-CD/DDB interfacial system, focusing on molecular interactions, interfacial adsorption, and overall stability. Notably, the SAE-CD/DDB-based supramolecular assembly/disassembly behaviors could be self-adjusted for regulating the aggregation particle size and thickness of interfacial self-assembled films. The SAE-CD/DDB co-stabilized HIPPEs exhibited favorable drug release capabilities, enabling sustained effects of DDB. Overall, the SAE-CD/DDB co-stabilized HIPPEs demonstrated excellent properties in terms of stability, drug loading capacity, and sustained release performance, highlighting their potential for in oral delivery and sustained-release systems.

Multi-stimuli-responsive pectin-coated dendritic mesoporous silica nanoparticles with Eugenol as a sustained release nanocarrier for the control of tomato bacterial wilt

Guo Xueping,Li Huiyan,Li Zhihao,Cui Ziqi,Ma Guangming,Nassor Aisha Khalfan,Guan Yi,Pan Xiaohong

Background Environmentally responsive nanoscale biocide delivery system enhances smart, regulated, and synergistic biocide application with precise biocide release. In this study, pectin-modified dendritic mesoporous silica nanoparticles (DMSNs) was used as a carrier to successfully construct a microenvironment-responsive (pH, temperature and enzyme) eugenol nano-biocide delivery system for the control of Ralstonia solanacearum infection. Results The results showed that the specific surface area, pore size and surface activity of DMSNs significantly influence the biocide loading of eugenol, and the biocide loading capability was up to 72.50%. Eu@DMSNs/Pec had significant pH and pectinase stimulating effects, with varying release amounts under different temperature conditions. Compared with eugenol alone, Eu@DMSNs/Pec significantly enhanced the efficacy of eugenol. DMSNs assisted eugenol to induce peroxidation damage, produce ROS (•O 2 − , •OH and 1 O 2 ), achieve synergistic antibacterial effects, and had better rain erosion resistance and foliar retention rate based on pectin wettability and adhesion. Eu@DMSNs/Pec-FITC showed demonstrated efficient transport characteristics in tomato roots, stems and leaves, which enhanced the control effect on tomato bacterial wilt. In addition, Eu@DMSNs/Pec exert minimal influence on tomato seed germination and root growth, and have low toxicity to non-target organisms such as earthworms. Therefore, Eu@DMSNs/Pec environment-responsive nano-controlled release nanocarrier can effectively achieve accurate biocide release and reduce biocide dosage. Conclusion This work not only provides a pectin-modified DMSNs-based eugenol nanoscale biocide delivery system in response to specific environmental conditions of R. solanacearum infection but also elucidates the eugenol biocide loading, selective release ability and antibacterial mechanism of the system.

Dual antioxidant delivery based on liposome sustained release system: Molecular mechanisms for inhibiting lipid oxidation and benzo[a]pyrene formation in Chinese sausage

Jingnan Lu,Lizhang Wu,Ran Wang,Xingguang Chen,Kezhou Cai,Baocai Xu

Lipid oxidation significantly deteriorates the quality of Chinese sausage during storage and is a key factor in the formation of benzo[a]pyrene (B(a)P), a carcinogen. To mitigate this, we developed a sustained-release nanomaterial (GA/Res-L) by encapsulating lipophilic resveratrol (Res) and hydrophilic gallic acid (GA) in liposomes. GA/Res-L effectively suppressed lipid hydroperoxides (LOOH) and malondialdehyde (MDA) formation, delaying browning and maintaining color. It also reduced the generation of B( a )P intermediates and secondary oxidation products. SEM analysis indicated that GA/Res-L preserved sausage texture by reducing internal voids. Antioxidant assays showed strong radical scavenging abilities. Release kinetics demonstrated that GA followed first-order kinetics, while Res followed zero-order kinetics. Density functional theory (DFT) calculations show that the binding energy of GA/Res-L is −35 kcal/mol, and the stability of GA and Res in liposomes is enhanced by π-π stacking and hydrogen bonding. This study offers a new strategy for using natural antioxidants to enhance meat product safety.

Preparation of Tofacitinib-Loaded Poly(lactic-co-glycolic acid) Sustained Release Nanoparticles by High-Gravity Nanoprecipitation Technique and Its Performance in Rheumatoid Arthritis

Yali Zhang,Xiaoxue Wang,Jianjun Zhang

Rheumatoid arthritis (RA), an autoimmune disease, is related to environmental factors, sex hormones, genetics, and other factors, affecting 0.5–1% of the global population. The JAK-STAT pathway plays an important role in the signal transduction of inflammatory cytokines and chemokines in the pathogenesis of RA. Tofacitinib (TF), the first approved JAK inhibitor for treating RA, can reduce inflammatory reactions, improve RA conditions, prevent irreversible joint injury, and improve the quality of life of patients. However, the traditional oral administration of TF causes poor compliance and severe side effects owing to the long-term and high-frequency treatment. Therefore, highly efficient pharmaceutical formulations for TF are necessitated. Here, a novel TF-loaded poly(lactic-co-glycolic acid) (PLGA) sustained release nanoparticle (NanoTF-PLGA) was prepared using a high-gravity nanoprecipitation technique (HGNPT), and its therapeutic effect on RA was verified in vitro and in vivo. The influence of the different operation parameters of HGNPT on the particle size of PLGA, including the rotating speed of the rotating bed, the volume ratio of solvent to antisolvent, and the concentration of the surfactant, was investigated using an orthogonal design. The optimum reaction conditions for preparing PLGA nanoparticles were selected by optimizing the formula, and TF was then added to the reaction mixture to prepare NanoTF-PLGA. The results demonstrated that NanoTF-PLGA had a smaller size (603.8 nm), a more uniform shape, a higher drug loading efficiency, and long-term sustained release properties (over 14 days). Furthermore, in vitro experiments proved that NanoTF-PLGA could enter RAW264.7 cells by cell uptake, demonstrated good biocompatibility, inhibited the migration of RAW264.7 cells, and simultaneously inhibited the expression of IL-6, IL-1β, and TNF-α in a dose-dependent manner. In vivo experiments showed that subcutaneous administration of NanoTF-PLGA significantly alleviated articular cartilage injury and reduced the level of inflammatory cell infiltration into the joint cavity. This study presents a promising technique for preparing sustained drug formulations that can be extended to numerous therapeutic drugs.

Core–shell structural PLGA nanoparticle of astaxanthin fabricated via micelle template for sustained release in vitro and long-term hepatoprotective effect in vivo

Zhu Yuan,Lu Peiqi,Liao Youwu,Adu-Frimpong Michael,Zou Zhihui,Jin Zhou,Wang Chengwei,Peng Min,Peng Houping,Xu Jingze,Xu Ying,Yu Jiangnan,Xu Ximing

This study aimed to prepare core–shell structural PLGA nanoparticles of astaxanthin (ASTA-PLGA@M) via micelle template for sustained release in vitro and long-term hepatoprotective effects in vivo. The morphology, mean particle size, zeta potential, polydispersity index (PDI), and drug loading efficiency of optimized formulation were investigated. Meanwhile, the physicochemical characterizations including X-ray diffraction (XRD) and attenuated total reflectance-Fourier transform infrared (ATR-FTIR) of ASTA-PLGA@M were evaluated to prove the successful encapsulation of astaxanthin. The in vitro release of astaxanthin from ASTA-PLGA@M in four different media was sustained slowly for 120 h. An in vivo release study also demonstrated that ASTA-PLGA@M nanoparticles enhanced oral bioavailability significantly. In addition, the hepatoprotective effects of astaxanthin on oxidative stress (OS) accompanied by apoptosis in acute hepatic damage caused by carbon tetrachloride (CCl 4 ) in mice were investigated. ASTA-PLGA@M nanoparticles provide a clear elevating effect on the activity of SOD and inhibit the increase of MDA during acute liver damage caused by CCl4. Moreover, histopathological analysis was conducted to study the long-term hepatoprotective effects of ASTA-PLGA@M for further application of astaxanthin in functional food or clinical use.

In vitro drug release and antibacterial activity evaluation of silk fibroin coated vancomycin hydrochloride loaded poly (lactic-co-glycolic acid) (PLGA) sustained release microspheres

Shengtang Li,Xuewen Shi,Bo Xu,Jian Wang,Peng Li,Xin Wang,Jinpeng Lou,Ziyao Li,Chengwei Yang,Songkai Li,Ping Zhen

Currently, the treatment of osteomyelitis poses a great challenge to clinical orthopedics. The use of biodegradable materials combined with antibiotics provides a completely new option for the treatment of osteomyelitis. In this study, vancomycin hydrochloride (VANCO) loaded poly (lactic-co-glycolic acid) (PLGA) microspheres were prepared by a double emulsion solvent evaporation method, and the in vitro drug release behaviors of the drug loaded microspheres were explored after coating with different concentrations of silk fibroin (SF). Drug loading, encapsulation efficiency, Scanning electron microscopy, particle size analysis, Fourier transform infrared spectroscopy, hydrophilicity, in vitro drug release, and in vitro antibacterial activity were evaluated. The results showed that the drug loading of vancomycin loaded PLGA microspheres was (24.11 ±1.72)%, and the encapsulation efficiency was (48.21 ±3.44)%. The in vitro drug release indicated that the drug loaded microspheres showed an obvious initial burst release, and the drug loaded microspheres coated with SF could alleviate the initial burst release in varying degrees. It also can reduce the amount of cumulative drug release, and the effect of microspheres coated with 0.1% concentration of SF is the best. The time of in vitro drug release in different groups of drug loaded microspheres can be up to 28 days. The microspheres coated with (0.1%SF) or without (0%SF) SF showed a cumulative release of (82.50±3.51)% and (67.70±3.81)%,respectively. Therefore, the surface coating with SF of vancomycin loaded microspheres can alleviate the initial burst release, reduce the cumulative drug release, potentially prolong the drug action time, and improve the anti-infection effect.

Study on the preparation of sustained-release thiamethoxam microspheres by blending microcrystalline wax with tapioca starch ester or dehydroabietic acid ester as the matrix

Linlin Wang,Qiang Hu,Yanmin Huang,Qipeng Xiong,Yong Chen,Chunfang Gan,Yuanfei Zhang,Guoqin Cui,Jianguo Cui

The controlled release formulations (CRFs) are considered an effective way to solve damage to the environment caused by traditional pesticide formulations. To change the defects of traditional neonicotinoid formulations that dissolve quickly in soil, three types of thiamethoxam (TM) CRFs microspheres with content of 20% TM were prepared using microcrystalline wax (MK) as the matrix, laurate acid tapioca starch ester (MSK) and stearyl dehydroabietic acid ester (MDK) as the regulators of ingredient release. The release behavior of CRFs microspheres in water and soil showed that the microspheres had superior stability and different TM sustained-release periods, and TM release of the microspheres in soil was faster than that in water. The release rate is TM/MDK > TM/MSK > TM/MK. In water, the release of thiamethoxam technical was finished after 38 hours. However, for TM/MK, the release rate was 94% after 240 hours, and the release time was extended by 6 times. Meanwhile, TM/MDK has a particular pH-responsive release. Research shows that using microcrystalline wax as the matrix, by adding MSK or MDK to adjust the release of ingredients, pesticide CRFs microspheres with different release periods can be prepared to achieve the purpose of controlling the release of pesticides.

Mussel-Inspired Surface Immobilization of Heparin on Magnetic Nanoparticles for Enhanced Wound Repair via Sustained Release of a Growth Factor and M2 Macrophage Polarization

Jiang Wu,Junyi Zhu,Qiuji Wu,Ying An,Kangning Wang,Tengxiao Xuan,Junwen Zhang,Wenxiang Song,Huacheng He,Liwan Song,Jie Zheng,Jian Xiao

Efficient reconstruction of a fully functional skin after wounds requires multiple functionalities of wound dressing due to the complexity of healing. In these regards, topical administration of functionalized nanoparticles capable of sustainably releasing bioactive agents to the wound site may significantly accelerate wound repair. Among the various nanoparticles, superparamagnetic iron oxide (Fe3O4) nanoparticles gain increasing attractiveness due to their intrinsic response to an external magnetic field (eMF). Herein, based on the Fe3O4 nanoparticle, we developed a fibroblast growth factor (bFGF)-loaded Fe3O4 nanoparticle using a simple mussel-inspired surface immobilization method. This nanoparticle, named as bFGF-HDC@Fe3O4, could stabilize bFGF in various conditions and exhibited sustained release of bFGF. In addition, an in vitro study discovered that bFGF-HDC@Fe3O4 could promote macrophage polarization toward an anti-inflammatory (pro-healing) M2 phenotype especially under eMF. Further, in vivo full-thickness wound animal models demonstrated that bFGF-HDC@Fe3O4 could significantly accelerate wound healing through M2 macrophage polarization and increased cell proliferation. Therefore, this approach of realizing sustained the release of the growth factor with magnetically macrophage regulating behavior through modification of Fe3O4 nanoparticles offers promising potential to tissue-regenerative applications.

Construction of Multifunctional Zinc Ion Sustained-Release Biocoating on the Surface of Carbon Fiber Reinforced Polyetheretherketone with Enhanced Anti-inflammatory Activity, Angiogenesis, and Osteogenesis

Shanshan Zhao,Wenying Dong,Yilong Wang,Xingyu Zhou,Junhui Jiang,Ruibo Hu,Tong Lin,Dahui Sun,Mei Zhang

Current treatments of carbon fiber-reinforced polyetheretherketone (CFRPEEK) as orthopedic implants remain unsatisfactory due to the bioinert surface. The multifunctionalization of CFRPEEK, which endows it with regulating the immune inflammatory response, promoting angiogenesis, and accelerating osseointegration, is critical to the intricate bone healing process. Herein, a multifunctional zinc ion sustained-release biocoating, consisting of a carboxylated graphene oxide, zinc ion, and chitosan layer, covalently grafts on the surface of amino CFRPEEK (CP/GC@Zn/CS) to coordinate with the osseointegration process. The release behavior of zinc ions theoretically conforms to the different demands in the three stages of osseointegration, including the burst release of zinc ions in the early stage (7.27 μM, immunomodulation), continuous release in the middle stage (11.02 μM, angiogenesis), and slow release in the late stage (13.82 μM, osseointegration). In vitro assessments indicate that the multifunctional zinc ion sustained-release biocoating can remarkably regulate the immune inflammatory response, decrease the oxidative stress level, and promote angiogenesis and osteogenic differentiation. The rabbit tibial bone defect model further confirms that, compared to the unmodified group, the bone trabecular thickness of the CP/GC@Zn/CS group increases 1.32-fold, and the maximum push-out force improves 2.05-fold. In this study, a multifunctional zinc ion sustained-release biocoating constructed on the surface of CFRPEEK that conforms to the requirements of different osseointegration stages can be an attractive strategy for the clinical application of inert implants.

Sustained release of PTX-incorporated nanoparticles synergized by burst release of DOX⋅HCl from thermosensitive modified PEG/PCL hydrogel to improve anti-tumor efficiency

Shuxin Xu,Weiwei Wang,Xijing Li,Jianping Liu,Anjie Dong,Liandong Deng

As drug therapies become increasingly sophisticated, the synergistic benefits of two or more drugs are often required. In this study, we aimed at improving anti-tumor efficiency of paclitaxel (PTX)-incorporated thermo-sensitive injectable hydrogel by the synergy of burst release of doxorubicin hydrochloride (DOX⋅HCl). Thermosensitive injectable hydrogel composed of nanoparticles assembled from amphiphilic copolymer poly(ε-caprolactone-co-1,4,8-trioxa[4.6]spiro-9-undecanone)-poly(ethylene glycol)-poly(ε-caprolaone-co-1,4,8-trioxa[4.6]spiro-9-undecanone) (PECT) was fabricated. Hydrophobic PTX and hydrophilic DOX⋅HCl were loaded simultaneously in the thermo-sensitive injectable hydrogel by a two-stage entrapment. Thermosensitive gelling behaviors of drug-loading PECT nanoparticle aqueous dispersions were studied. In vitro release profiles of PTX and DOX⋅HCl and in vivo anti-tumor effect by dual drugs from PECT hydrogel were investigated. The results showed that hydrophilic and hydrophobic drugs could be successfully entrapped in PECT hydrogel simultaneously without affecting its thermo-sensitive behavior. In vitro release profiles demonstrated the burst release of DOX⋅HCl and the sustained release of PTX. Anti-tumor effect was improved by a fast and tense attack caused by the burst release of hydrophilic DOX⋅HCl from hydrogel, which was continued by the sequent sustained release of PTX-incorporated nanoparticles and remnant DOX⋅HCl. Unintentionally, entrapped in PECT hydrogel, hydrophilic DOX⋅HCl was observed to have a sustained releasing pattern in vitro and in vivo .

Fabrication of porous carbon microspheres with numerous spherical microstructures directly from waste Camellia oleifera shells and their application in sustained-release of 5-fluorouracil

Wentian Ma,Kexin Li,Huiqin Guo,Liushui Yan,Yuhua Dai,Xubiao Luo,Yong Yao

Porous carbon microspheres with numerous spherical microstructures were successfully fabricated directly from waste Camellia oleifera shells by hydrothermal carbonization combined with annealing treatment and potassium hydroxide chemical activation technique in the assistance of phloroglucinol. Numerous aromatic carbon microspheres were constructed after hydrothermal carbonization due to the structure-directing effect of phloroglucinol. The spherical microstructures remain intact after suffering from potassium hydroxide chemical activation because the aromatic carbon framework was further graphitized by annealing treatment. The mesoporous carbon microspheres were successfully obtained from microporous carbon microspheres through the secondary potassium hydroxide chemical activation. As valuable porous materials, the as-prepared microporous and mesoporous carbon microspheres were successfully applied to the sustained-release of anticancer drug 5-fluorouracil. The as-prepared microporous and mesoporous carbon microspheres showed some obvious advantages such as low-cost, high-capacity, and strong biological affinity compared with traditional drug sustained-release agent.

An injectable liposome for sustained release of tanshinone IIA to the treatment of acute blunt muscle injury by augmenting autophagy and alleviating oxidative stress

Jinwu Wang,Jie Cai,Xingyu Wang,Gaosheng Zhu,Yongzeng Feng,Hua Chen,Leyi Cai

Acute blunt skeletal muscle injury occurs frequently in sports and traffic accidents, and even leads to muscle necrosis and impaired functionality. Current treatment options for muscle injuries remain suboptimal and often result in delayed/incomplete recovery of damaged muscles. Tanshinone IIA is extracted from Salvia Miltiorrhizae, which is effective in the treatment of injury repair. But the clinical application of tanshinone IIA is limited due to its low water solubility, low permeability to biofilm and low bioavailability. In this study, tanshinone IIA liposomes were prepared to improve the bioavailability and sustained release of tanshinone IIA. The particle size, dispersion coefficient, zeta potential, encapsulation efficiency (EE) and drug loading (DL) of tanshinone IIA liposomes were 150.67 ± 27.23 nm, 0.20 ± 0.015, -8.73 ± 2.28 mV, 70.32 ± 4.04% and 15.63%, respectively. The results of quantitative real-time polymerase chain reaction (QRT-PCR) showed that tanshinone IIA liposome significantly promoted the expression of vimentin and reduce MHCIIB expression compared with other groups (P < 0.05). Western blotting showed that tanshinone IIA liposome could effectively promote the expression of autophagy-related proteins (VPS34, Beclin 1 and CTSD) and decrease p62 expression levels to treat injured muscle. Through HE, immunohistochemistry, ELISA and serological tests, we found that tanshinone IIA liposome not only effectively promoted the expression of desmin, but also reduced the expression of collagen-I and inhibited the production of pro-inflammatory factors, such as tumor necrosis factor α (TNF-α) and interleukin-6 (IL-6) (P < 0.05). In addition, tanshinone IIA liposome therapy significantly reduced the level of malondialdehyde (MDA) and increased the activity of superoxide dismutase (SOD) after muscle injury compared with other groups (P < 0.05). In conclusion, tanshinone IIA liposome possesses an effective therapeutic effect on acute blunt muscle injury in rats by augmenting autophagy and alleviating oxidative stress. The continuous release of tanshinone IIA encapsulated by liposomes for disease treatment provide a new idea for the efficient and safe use of drugs with low lipid solubility and bioavailability for the treatment of acute blunt muscle injury and repair of other injuries.
