Myo-inositol-1-phosphate synthase (Ino-1) functions as a protection mechanism in Corynebacterium glutamicum under oxidative stress

Can Chen,Keqi Chen,Tao Su,Bing Zhang,Guizhi Li,Junfeng Pan,Meiru Si

Graphical The ino-1 deletion mutant showed obviously higher ROS levels than the WT(pXMJ19) and the complement Δino-1(pXMJ19-ino-1) strains under oxidative stress conditions. And clearer protein carbonyl groups were detected in the Δino-1(pXMJ19) strains after exposure to ROS-generating agents compared to that in the WT(pXMJ19) strains. Reactive oxygen species (ROS) generated in aerobic metabolism and oxidative stress lead to macromolecules damage, such as to proteins, lipids, and DNA, which can be eliminated by the redox buffer mycothiol (AcCys-GlcN-Ins, MSH). Myo -inositol-phosphate synthase (Ino-1) catalyzes the first committed step in the synthesis of MSH, thus playing a critical role in the growth of the organism. Although Ino-1s have been systematically studied in eukaryotes, their physiological and biochemical functions remain largely unknown in bacteria. In this study, we report that Ino-1 plays an important role in oxidative stress resistance in the gram-positive Actinobacteria Corynebacterium glutamicum . Deletion of the ino-1 gene resulted in a decrease in cell viability, an increase in ROS production, and the aggravation of protein carbonylation levels under various stress conditions. The physiological roles of Ino-1 in the resistance to oxidative stresses were corroborated by the absence of MSH in the Δ ino-1 mutant. In addition, we found that the homologous expression of Ino-1 in C. glutamicum yielded a functionally active protein, while when expressed in Escherichia coli BL21(DE3), it lacked measurable activity. An examination of the molecular mass (Mr) suggested that Ino-1 expressed in E. coli BL21(DE3) was not folded in a catalytically competent conformation. Together, the results unequivocally showed that Ino-1 was important for the mediation of oxidative resistance by C. glutamicum .

Three-Step Ferroelastic Transitions from Hexagonal to Triclinic Phases in a Hybrid Perovskite: (1-Fluoromethyl-1-methylpyrrolidine)[CdCl3]

Shuai Chen,Ding-Chong Han,Le Ye,Wei-Xiong Zhang

Hybrid ferroelastic crystals have emerged as a hot research topic in recent years owing to their prospective applications in piezoelectric sensors, mechanical switches, and optoelectronic devices. Nevertheless, most of the documented materials exhibit one-step or two-step ferroelastic phase transition(s), and those with multistep ferroelastic transitions are extremely scarce. We present a new hexagonal molecular perovskite based on a fluoro-substituted flexible cyclic ammonium cation, (1-fluoromethyl-1-methylpyrrolidine)[CdCl3] (1), undergoing unusual three-step ferroelastic phase transitions from hexagonal paraelastic phase to orthorhombic, monoclinic, and triclinic ferroelastic phases at 388, 376, and 311 K, respectively, with Aizu notation of 6/mmmFmmm, mmmF2/m, and 2/mF-1, featuring spontaneous strain of 0.002, 0.023, and 0.110, respectively. Furthermore, variable-temperature single-crystal diffraction reveals that the phase-transition mechanism in 1 principally originates from intriguing dynamic change of organic cations and synchronous displacement of inorganic chains. This scarce instance of multistep hybrid ferroelastic provides important clues for finding advanced ferroelastic materials.

Shogaol triggers apoptosis and ferroptosis in Ewing sarcoma by targeting the polo-like kinase 1/nucleophosmin 1 axis

Zhengyu Wu,Huimou Chen,Yu Zhou,Sidong Zhang,Zhihua Li,Yadong Song,Jing Hu

Ewing sarcoma is an aggressive pediatric bone cancer with poor prognosis, highlighting the urgent need for novel therapeutics. This study demonstrates the potent anti-tumor activity of shogaol, a bioactive compound from ginger, against Ewing sarcoma, using both in vitro and in vivo model systems. In vitro, shogaol significantly inhibited cell proliferation and induced both apoptosis and ferroptosis in Ewing sarcoma cells, as evidenced by increased lipid reactive oxygen species, Malondialdehyde, and Fe 2+ levels, as well as altered expression of BAX, BCL2, GPX4, and SLC3A2. In vivo , shogaol significantly inhibited tumor cell growth in a subcutaneous xenograft mouse model using SKNMC cells. Using transcriptome sequencing and proteomics, we found that shogaol targeted the PLK1/NPM1 axis: it downregulated polo-like kinase 1 (PLK1), leading to reduced phosphorylation of NPM1 at Thr199. This facilitated the increased interaction between the E3 ligase TRIM28 and NPM1, leading to its ubiquitination and subsequent degradation, which in turn inhibited the MAPK signaling pathway. Key findings were validated by CETSA, isothermal dose-response fingerprinting, co-immunoprecipitation, and ubiquitination assays. These results demonstrate that shogaol exerts dual cytotoxic effects through the PLK1/NPM1 axis, presenting a potential natural therapeutic approach for Ewing sarcoma.

Synergistic adsorption effect on Co3O4(1 1 0) surface to promote the ethanol sensing properties: Experiment and theory

Songlin Li,Chen Wang,Zhaohui Lei,Shanfu Sun,Jie Gao,Pengfei Cheng,Hai Wang

A deep understanding of the relationship between the properties and structure of transition metal oxide semiconductor gas sensors is essential for optimizing sensor design. Comparing and analyzing the sensing properties of nanomaterials with different morphologies and crystal facets by combining the experiment analysis and the density functional theory (DFT) calculations could identify and understand the corresponding mechanisms at the atomic or ionic level. Here, grown Co 3 O 4 nanosheet (Co 3 O 4 -NS) with exposed (1   1   1) facets, and Co 3 O 4 nanorods (Co 3 O 4 -NR) with exposed (1   1   0) facets on alumina ceramic tubes were prepared and studied in sensing ethanol. The adsorption characteristics of ethanol on the sensor surface were analyzed by DFT calculation. The results demonstrated that the sensing performance of Co 3 O 4 -NR was superior to Co 3 O 4 -NS. The adsorption of hydroxyl groups in ethanol was better than the methyl groups on the surface. The oxygen and hydrogen atoms from the hydroxyl groups in ethanol had the lowest adsorption energies (E ads  = −3.44 eV) when they interact with the Co atom and the indirectly adjacent low-coordinate oxygen atoms on the (1   1   0) surface, respectively. The synergistic adsorption of target gases by exposed surface atoms induced superior sensing properties of the material.

(NH4)2SO4-assisted synthesis of thin-walled Ti-rich hollow titanium silicalite-1 zeolite for 1-hexene epoxidation

Mengyao Li,Yi Zhai,Xubin Zhang,Fumin Wang,Guojun Lv,Ahishakiye Rosine,Mengyue Li,Qing Zhang,Yongkui Liu

The preparation of hollow zeolite with a large cavity structure to improve the accessibility to the active sites of catalyst is of great significance. A thin-walled Ti-rich hollow Titanium silicalite-1 (TS-1) Nanobox was synthesized by dissolution-recrystallization using Silicalite-1 (S-1) as the parent zeolite and (NH 4 ) 2 SO 4 as the dual-function crystallization-mediating agent. The wall thickness of the zeolite is about 30 nm, the hollow percentage (the ratio of the cavity area in the middle of the zeolite to the overall area of the zeolite) can reach 0.57, and the Si/Ti ratio is 39.63. The formation mechanism of hollow TS-1 was studied. As the parent zeolite, S-1 is easier to dissolve than TS-1, making it easier for the hollow zeolite to get a complete cavity. When the molar ratio of (NH 4 ) 2 SO 4 /TEOS reaches 0.25, the dissolution rate matches the crystallization rate best, and a perfect hollow structure is obtained. The pH of the initial gel is the key factor for the hollow structure. The catalytic activity and stability were tested by 1-hexene epoxidation. The conversion of 1-hexene using HTS-1#S0.25 as the catalyst is much higher than that using hollow TS-1 prepared by the conventional method as the catalyst. HTS-1#S0.25 remains a high oxidation activity and high selectivity after five recycle experiments. In addition, the type of anion also has a great influence on the reactivity. This method can provide a new idea for the preparation of hollow TS-1.

Enantioselective resolution of (±)-1-phenylethyl acetate using the immobilized extracellular proteases from deep-sea Bacillus sp. DL-1

Lu Dong,Shujuan Qi,Jianwei Jia,Yun Zhang,Yunfeng Hu

Bacillus sp. DL-1 was isolated from the deep sea of the Western Pacific Ocean and behaved very good resistance to NaCl. The extracellular proteases of Bacillus sp. DL-1 were found to exhibit excellent enantioselectivity for the kinetic resolution of (±)-1-phenylethyl acetate. To improve the stability of the enzyme, the immobilized extracellular proteases were preparated by using 60 g kieselguhr and 1-L crude fermentation broth containing extracellular proteases of Bacillus sp. DL-1, shaking at 25 °C, 200 r/min for 10 h. Every gram of kieselguhr adsorbed 25.7 mg extracellular protease and the enzymatic activity recovery was 79.86%. The immobilized proteases preserved about 35.8% of its activity after 6 repeated uses, which were also used as biocatalyst to asymmetrically hydrolyse (±)-1-phenylethyl acetate for the preparation of (R)-1-phenylethanol and (S)-1-phenylethyl acetate with high optical purities. The effects of pH, temperature, enzyme concentration, substrate concentration, reaction time and additives (metal ions/surfactants) on the resolution were investigated by single factor experiments. Under the optimal reaction conditions (10 mM (±)-1-phenylethyl acetate, 40 mg/mL immobilized extracellular proteases, pH 7.5 (Tris-HCl), 5% (v/v) methanol and 45 °C for 2 h), (R)-1-phenylethanol was generated with the e.e.p being > 97%, and the yield being 53%, respectively. Analogously, under the optimal reaction conditions (10-mM (±)-1-phenylethyl acetate, 360 mg/mL immobilized extracellular proteases, pH 6.0 (PB), 5% (v/v) DMSO and 35 °C for 1.5 h), (S)-1-phenylethyl acetate was generated with the e.e.s being over 99% and the yield being 79%, respectively. Compared with the extracellular proteases from Bacillus sp. DL-2, the immobilized extracellular proteases from Bacillus sp. DL-1 exhibited higher hydrolytic activity and could asymmetrically hydrolyse (±)-1-phenylethyl acetate by using higher substrate concentrations, shorter reaction times to obtain higher yields. Notably, the extracellular proteases of Bacillus sp. DL-1 were demonstrated to behave the same enantio-preference as those of most other reported esterases/lipases. Proteases from deep-sea Bacillus sp. DL-1 are promising biocatalysts for the synthesis of valuable chiral chemicals.

Experimental and correlated liquid–liquid equilibrium data for (water + 1,6-hexanediol + 1-butanol or 2-methyl-1-propanol)

Xingchuan Yang,Xiangmeng Chen,Chunmei Cao,Li Xu,Guoji Liu

Solubility and liquid–liquid equilibria data for ternary systems for (water + 1,6-hexanediol + 1-butanol/2-methyl-1-propanol) were obtained at (293.15–323.15) K under atmospheric pressure. The LLE data were determined using a double-jacketed glass vessel, and the solubility curves were acquired by the cloud-point method. The distribution coefficient and separation factors were calculated and the reliability of experimental data was assessed by the Othmer–Tobias equation and Hand equation. The measured LLE data were fitted by NRTL and UNIQUAC models and binary parameters were obtained by using of Aspen Plus software. The calculated root-mean-square deviation values show that the calculated results from models showed good agreement with experimental data.

Elevated glutamate impedes anti-HIV-1 CD8 + T cell responses in HIV-1-infected individuals on antiretroviral therapy

Wang You-Yuan,Zhen Cheng,Hu Wei,Huang Hui-Huang,Li Yan-Jun,Zhou Ming-Ju,Li Jing,Fu Yu-Long,Zhang Peng,Li Xiao-Yu,Yang Tao,Song Jin-Wen,Fan Xing,Zou Jun,Meng Si-Run,Qin Ya-Qin,Jiao Yan-Mei,Xu Ruonan,Zhang Ji-Yuan,Zhou Chun-Bao,Yuan Jin-Hong,Huang Lei,Shi Ming,Cheng Liang,Wang Fu-Sheng,Zhang Chao

CD8 + T cells are essential for long-lasting HIV-1 control and have been harnessed to develop therapeutic and preventive approaches for people living with HIV-1 (PLWH). HIV-1 infection induces marked metabolic alterations. However, it is unclear whether these changes affect the anti-HIV function of CD8 + T cells. Here, we show that PLWH exhibit higher levels of plasma glutamate than healthy controls. In PLWH, glutamate levels positively correlate with HIV-1 reservoir and negatively correlate with the anti-HIV function of CD8 + T cells. Single-cell metabolic modeling reveals glutamate metabolism is surprisingly robust in virtual memory CD8 + T cells (TVM). We further confirmed that glutamate inhibits TVM cells function via the mTORC1 pathway in vitro. Our findings reveal an association between metabolic plasticity and CD8 + T cell-mediated HIV control, suggesting that glutamate metabolism can be exploited as a therapeutic target for the reversion of anti-HIV CD8 + T cell function in PLWH.

Peptide MSI-1 inhibited MCR-1 and regulated outer membrane vesicles to combat immune evasion of Escherichia coli

Xinyue Ye,Jian Wang,Pengfei Xu,Xiaoqian Yang,Qixue Shi,Genyan Liu,Zhaoshi Bai,Changlin Zhou,Lingman Ma

Polymyxin resistance is conferred by MCR-1 (mobile colistin resistance 1)-induced lipopolysaccharide (LPS) modification of G − bacteria. However, the peptide MSI-1 exerts potent antimicrobial activity against mcr-1 -carrying bacteria . To further investigate the potential role of MCR-1 in improving bacterial virulence and facilitating immune evasion, and the immunomodulatory effect of peptide MSI-1, we first explored outer membrane vesicle (OMV) alterations of mcr-1 -carrying bacteria in the presence and absence of sub-MIC MSI-1, and host immune activation during bacterial infection and OMV stimulation. Our results demonstrated that LPS remodelling induced by MCR-1 negatively affected OMV formation and protein cargo by E. coli . In addition, MCR-1 diminished LPS-stimulated pyroptosis but facilitated mitochondrial dysfunction, further aggravating apoptosis in macrophages induced by OMVs of E. coli . Similarly, TLR4-mediated NF-κB activation was markedly alleviated once LPS was modified by MCR-1. However, peptide MSI-1 at the sub-MIC level inhibited the expression of MCR-1, further partly rescuing OMV alteration and attenuation of immune responses in the presence of MCR-1 during both infection and OMV stimulation, which can be exploited for anti-infective therapy.

Enantioselective Resolution of (±)-1-Phenylethanol and (±)-1-Phenylethyl Acetate by a Novel Esterase from Bacillus sp. SCSIO 15121

Liang Jiayuan,Zhang Yun,Sun Aijun,Deng Dun,Hu Yunfeng

A novel microbial esterase BSE01281 identified from the Indian Ocean was cloned, expressed, and functionally characterized. Esterase BSE01281 could enanoselectively resolve (±)-1-phenylethanol and (±)-1-phenylethyl acetate through two types of enzymatic reactions. After the optimization of enzymatic reactions, BSE01281 could efficiently generate ( R )-1-phenylethyl acetate with high enantiomeric excess (>99 %) and high conversion (42 %) after 96 h trans-esterification reactions. Additionally, BSE01281 could also produce ( R )-1-phenylethanol ( e.e . > 99 %) and ( S )-1-phenylethyl acetate ( e.e . > 95 %) at a conversion of 49 % through direct hydrolysis of inexpensive racemic 1-phenylethyl acetate for 8 h. Optically pure ( R )-1-phenylethanol generated from direct enzymatic hydrolysis of racemic 1-phenylethyl acetate by BSE01281 is not easily prepared by dehydrogenases, which generally follow the “Prelog’s rule” and give ( S )-1-phenylethanol instead.

PARP-1 inhibitor alleviates cerebral ischemia/reperfusion injury by reducing PARylation of HK-1 and LDH in mice

Ya-Ling Chen,Yi Wang,Qiu-Yu Fang,Tong Wang,Cong Chen,Tong-Yao Gao,Ming Wu,Wei-Ping Zhang,Yun-Bi Lu

Poly (ADP-ribose) polymerase-1 (PARP-1) activity significantly increases during cerebral ischemia/reperfusion. PARP-1 is an NAD + -consumption enzyme. PARP-1 hyperactivity causes intracellular NAD + deficiency and bioenergetic collapse, contributing to neuronal death. Besides, the powerful trigger of PARP-1 causes the catalyzation of poly (ADP-ribosyl)ation (PARylation), a posttranslational modification of proteins. Here, we found that PARP-1 was activated in the ischemic brain tissue during middle-cerebral-artery occlusion and reperfusion (MCAO/R) for 24 h, and PAR accumulated in the neurons in mice. Using immunoprecipitation, Western blotting, liquid chromatography-mass spectrometry, and 3D-modeling analysis, we revealed that the activation of PARP-1 caused PARylation of hexokinase-1 and lactate dehydrogenase-B, which, therefore, caused the inhibition of these enzyme activities and the resulting cell energy metabolism collapse. PARP-1 inhibition significantly reversed the activity of hexokinase and lactate dehydrogenase, decreased infarct volume, and improved neuronal deficiency. PARP-1 inhibitor combined with pyruvate further alleviated MCAO/R-induced ischemic brain injury in mice. As such, we conclude that PARP-1 inhibitor alleviates neuronal death partly by inhibiting the PARylation of metabolic-related enzymes and reversing metabolism reprogramming during cerebral ischemia/reperfusion injury in mice. PARP-1 inhibitor combined with pyruvate might be a promising therapeutic approach against brain ischemia/reperfusion injury.

An Exporter of Leucine, LeuE Functions as an Exporter of 1-Aminocyclopropane-1-Carboxylic Acid in Escherichia coli

Xue-Chun Li,Hong-Jie Xie,Zheng-Xiong Zhou,Bin Sun,Jun-Jie Lin,Xiao-Dan Li,Ren Wang

1-aminocyclopropane-1-carboxylic acid (ACC), a crucial intermediate in the ethylene biosynthetic pathway, plays a decisive role in regulating plant developmental processes and stress responses. In heterologous biosynthesis systems, ACC biosynthesis is inhibited due to its antibacterial activity. Enhancing ACC export confers advantages to ACC-producing biotechnological platforms and microbial cell factories. While bacterial transporters for canonical amino acids in Corynebacterium glutamicum and Escherichia coli have been extensively studied, their roles in ACC transport remain unexplored. To identify the exporters mediating ACC transport in E. coli , relevant experiments were performed in this study. The results showed that the Leucine-exporter LeuE acts as a high-efficiency ACC exporter in E. coli . Subsequently, structural modeling coupled with site-directed mutagenesis revealed that L160 is a critical residue in determining the ACC transport specificity of LeuE. The L160Y mutation enhanced E. coli tolerance to ACC, elevating the threshold from 50   mM to 100   mM. Structural modeling predicted that the L160Y mutation-induced compaction reduced the transporter channel diameter by 28% (from 9.551   Å to 6.876   Å), a change that may enhance the binding affinity of ACC. Notably, a significant reduction in intracellular ACC levels was observed in both the LeuE-overexpressing and LeuE (L160Y)-overexpressing strains. These findings enable large-scale biosynthesis of ACC and its derivatives via engineered microbial systems, thereby facilitating cost-effective development of ACC-based pharmaceuticals and agrochemicals.
