Oxidative damage to mitochondrial DNA is closely associated with the development of various human diseases. 8-Oxo-7,8-dihydroguanine (8-oxoG) is a vital biomarker of oxidative DNA damage with a putative role as an epigenetic-like modification. Herein, we reported a high-throughput sequencing analysis of 8-oxoG modifications using a chemical labeling-based method and identified multiple 8-oxoG sites in human mitochondrial DNA. We also demonstrated that Y-box binding protein 1 (YBX1), an 8-oxoG-binding protein, is potentially involved in the transcriptional regulation of mitochondrial genes containing 8-oxoG modification sites. These findings provide a solid foundation for further understanding the biological consequences of 8-oxoG modification in mammalian cells.

Osteoarthritis (OA) is a prevalent chronic joint disease characterized by progressive cartilage degradation, synovial inflammation, and subchondral bone remodeling. Current pharmacotherapies are limited by poor targeting, low bioavailability, and systemic side effects. Here, we developed a zinc-based metal–organic framework (ZIF-8) loaded with dimethyl malonate (DM), a succinate dehydrogenase inhibitor, for targeted OA therapy. The DM@ZIF-8 nanocomposites were synthesized via a one-step coprecipitation method and characterized using TEM, FT-IR, UV-Vis, DLS, and zeta potential measurements. The in vitro drug release profile showed pH-responsive behavior, with accelerated release under acidic conditions mimicking the inflammatory microenvironment. Cell viability and hemolysis assays demonstrated good biocompatibility. Macrophage polarization studies revealed that DM@ZIF-8 promoted M2 polarization, as indicated by increased CD206 and TGF-β expression and decreased CD86 and iNOS levels. In an OA rat model, DM@ZIF-8 significantly improved joint structure, reduced cartilage damage, and modulated inflammatory cytokines (increased IL-10/TGF-β, decreased TNF-α/iNOS). Histological and biochemical analysis confirmed no obvious systemic toxicity. Our findings suggest that DM@ZIF-8 is a promising nanoplatform for targeted OA therapy with improved efficacy and safety. In summary, this study utilizes ZIF-8 nanocarriers to deliver succinate dehydrogenase inhibitor (DM) for modulating macrophage polarization in the treatment of osteoarthritis.

With the rapid progress of urbanization and industrialization, water pollution has become an increasingly serious issue, particularly due to dye wastewater, which contains numerous harmful substances and is difficult to treat effectively. Dye wastewater contains many harmful substances and is difficult to treat effectively. Traditional treatment methods are often inefficient and economically unsustainable. In contrast, membrane separation technology offers high separation efficiency and environmental benefits, making it a promising approach for water purification. Therefore, in this paper, the hydrophilic modification of PVDF membranes was carried out by adding ZIF-8 and chitosan(CS), and at the same time, the membranes were endowed with the ability of light self-cleaning. ZIF-8, a metal–organic framework (MOF) material, was synthesized using water as the solvent through the reaction of zinc nitrate with 2-methylimidazole. The PVDF membranes were modified by physical co-mingling, and ZIF-8/CS/PVDF composite membranes with different contents of ZIF-8 and CS were prepared by the submerged precipitation phase transition method. The performance test results demonstrated that the composite membranes doped with ZIF-8 and CS exhibited significant improvements in water contact angle, pure water flux, and dye wastewater treatment performance. Notably, the modified membranes showed exceptional dye removal efficiency and enhanced resistance to membrane fouling. Furthermore, the results of photocatalytic self-cleaning experiments revealed that the modified membranes effectively degraded Congo red and methylene blue dyes under Xenon lamp irradiation, highlighting their excellent photocatalytic self-cleaning capability. These findings suggest that the ZIF-8/CS/PVDF-modified membrane holds great potential for water treatment and self-cleaning applications, achieving up to 93.06% dye rejection and a pure water flux of 95.89 L m −2 h −1 , more than double that of the pristine PVDF membrane.

The sluggish kinetics of the anodic oxygen evolution reaction (OER) severely limits the overall efficiency of water electrolysis. To overcome this critical bottleneck, we designed a self-supporting composite electrocatalyst by growing Ni-Fe layered double hydroxide (NiFe-LDH) nanosheets on zeolitic imidazolate framework-8 (ZIF-8) derived N doped porous carbon nanofibers (NCNFs). The in-situ pyrolysis process creates a dense, highly conductive mesoporous network within the carbon framework. Meanwhile, the vertical growth of NiFe-LDH helps alleviate catalyst agglomeration, thereby increasing active site exposure and accelerating mass transportation during reactions. Consequently, the optimized electrode exhibits excellent catalytic performance, achieving a low overpotential of 216 mV at 10 mA cm −2 , a Tafel slope of 62.3 mV dec −1 , and sustained long-term stability over 50 h at 50 mA cm −2 under the tested laboratory condition. Crucially, comprehensive density functional theory calculations rationalize the enhanced electrocatalytic kinetics by revealing strong interfacial electronic coupling. This unique electronic redistribution optimally upshifts the d band center of the active metal sites and significantly lowers the energy barrier of the rate-determining step from 2.14 eV to 1.56 eV. This work provides deep mechanistic insights and a feasible structural engineering strategy for developing highly efficient bimetallic electrocatalysts for future sustainable energy conversion applications.

Background The precise design or functionalization of metal organic frameworks (MOFs) based on the properties of organic dyes is a promising direction in environment remediation. Methods Herein, using methyl orange (MO) as the target dye, cetylpyridinium bromide (CPB) functionalized ZIF-8 (CPB@ZIF-8) was fabricated by the post-synthetic modification strategy. Significant findings The surface area and the pore volume of CPB@ZIF-8 are greatly reduced compared with pure ZIF-8. However, the introduction of amphiphilic CPB molecules into ZIF-8 can significantly enhance the adsorption performance of MO. This enhanced MO adsorption capacity can be attributed to the incorporation of more positively charged sites, hydrophobic sites and aromatic rings in the CPB-modified ZIF-8 frameworks, which provides the electrostatic attractions, the hydrophobic and π-π interactions that mainly govern the adsorption of MO on CPB@ZIF-8. The pseudo-second-order kinetic model and the Langmuir isotherm model can well represent the adsorption process of MO. The maximum adsorption capacity (195.54 mg/g) has been obtained at 30 °C, outperforming pristine ZIF-8 and various reported absorbents. The prepared CPB@ZIF-8 composite is a superior example of the tunable adsorption affinity of MOFs for anionic dyes.

Organic solvent nanofiltration (OSN), due to the characteristics of energy saving, mild operation conditions, and less waste generation, is an effective tool for purification, concentration and solvent exchange of high-value-added products. Introducing an interlayer containing ZIF-8, which features high surface areas and adjustable pore structures , can effectively regulate the interfacial polymerization process to optimize the performance of OSN membranes. However, due to the lack of nucleation sites with the substrate, it remains technologically challenging to design the ZIF-8 interlayer with structure and property persistence for interfacial polymerization under mild conditions. Inspired by the processes of birds’ nesting, the processes of mixing PEG and ZIF precursor, coating, and fast heating are working here to stimulate the in-situ growth of ZIF-8 in a facile, green and ordered way. PEG allows uniform dispersion of ZIF-8 in one step and without involving organic solvents. Furthermore, the increased binding affinity between ZIF-8 and the membrane surface endows the composite membrane with uniform morphology and significantly enhanced stability. Overall, this strategy may provide a fast, eco-friendly, and scalable approach to design composite membranes with MOF and other nanoparticulate materials as interlayers.

The Gum Ghatti (GG)/Konjac Glucomannan (KGM)/zeolitic imidazolate framework-8 (ZIF-8) composite aerogels (GKZ) were synthesized by the sol-gel method and the vacuum freeze-drying method for effective removal of ciprofloxacin (CIP) from contaminated water. The results showed that the addition of different concentrations (0, 0.4, 0.6, 0.8 wt%) of GG could promote homogeneity but weakened the molecular interaction of the aerogel system without water. Remarkably, the ZIF-8 maintained the crystal structure, as well as its dispersion on the surface of aerogel, was significantly improved. Additionally, it was also found that the incorporation of moderate GG was favored regarding CIP adsorption, and GKZ with 0.6 % GG (GKZ0.6) exhibited the highest adsorption capability for CIP around 1035.4 mg/g. Besides, the effects of various ions, ionic strength , and pH on CIP adsorption were also investigated, which provided the reference for guiding the utilization of GKZ0.6. Furthermore, at fewer cycles, the adsorption efficiency of GKZ0.6 on CIP showed less loss. These features revealed that the GKZ composite aerogels might be a promising adsorbent for CIP removal from water.

Fe 2 O 3 /zeolitic imidazolate framework-8 (ZIF-8) architectures were successfully prepared by a polyvinylpyrrolidone (PVP)-assisted method, which can be as an effective potential catalyst for promoting the thermal degradation of nitrocellulose (NC). Thermal behavior and degradation kinetics of catalyst-NC mixtures were studied using the TG/DTG and DSC methods, and found that the Fe 2 O 3 /ZIF-8-NC pyrolysis possessed a total degradation of 77.96%, a maximum pyrolysis rate temperature of 194.5 °C, and the lowest activation energy of 179.4 ± 16.1 kJ mol −1 , confirming a better thermocatalytic performance of Fe 2 O 3 /ZIF-8 when compared to Fe 2 O 3 and ZIF-8 for NC thermolysis. Furthermore, it can be inferred from the isoconversional analysis, which employs a modified fitting algorithm, that catalyst-NC mixtures and pure NC adhere to the reconstructed “n-order” reaction models. This work showed how a catalyst of the metal oxide/metal-organic frameworks (MOFs) type could improve the pyrolysis of NC, which could serve as a significant reference for the development of additional catalysts of the metal oxide/MOFs for use in NC-based energetic materials.

Zeolitic imidazolate framework-8 (ZIF-8) was coupled with AgBr using a facile deposition–precipitation method to synthesize AgBr@ZIF-8 composites. The photocatalytic activity of prepared photocatalysts was evaluated by the degradation of Rhodamine B (RhB). As expected, the photocatalytic activity of the AgBr@ZIF-8 photocatalysts was superior to that of ZIF-8 and AgBr. Among the AgBr@ZIF-8 photocatalysts, AgBr@ZIF-8 with a ZIF-8 loading of 5 wt% exhibited the highest photocatalytic efficiency of 99.5 % within 15 min. The remarkable photocatalytic activity of the AgBr@ZIF-8 composites was attributed to the heterojunction between ZIF-8 and AgBr, which reduced the recombination rate of electron-hole pairs, thereby enhancing the photocatalytic efficiency of the catalysts. According to the trapping experiment, superoxide radicals (•O 2 - ) and holes (h + ) were certified as the main species in the efficient removal of RhB.

BACKGROUND The separation of furfural from a low-concentration multicomponent and acid-containing aqueous solution is a highly demanding process; traditional distillation requires an excessive amount of energy. Herein, we demonstrated ZIF-8-derived microporous carbon (C) as a promising adsorbent for the separation of furfural using adsorption isotherms, dynamic column adsorption, adsorption kinetics and desorption studies. RESULTS In the competitive adsorption kinetics experiments with a furfural/acetic acid concentration of 50/20 g L −1 , the acetic acid adsorbed in the micropores of NC ZIF-8 1000C-800A gradually was replaced by furfural, whereas the amount of acetic acid in the mesopores of NC ZIF-8 800C-800A slowly increased. In addition, the micropores of NC ZIF-8 1000C-800A exhibited highly efficient and fast adsorption characteristics toward furfural over a wide furfural/acetic acid concentration range (0–50/0–20 g L −1 ) with the furfural adsorption capacity reaching an adsorption equilibrium within 10 min. The results show that the high Brunauer–Emmett–Teller surface area of NC ZIF-8 1000C-800A is responsible for the high furfural adsorption capacity (1205.3 mg g −1 ) and the confined microporous structure can strengthen the π–π interactions between porous C and furfural, thereby enhancing the selectivity of furfural/acetic acid (61.4) in the furfural/acetic acid (50/20 g L −1 ) dynamic column adsorption system. Meanwhile, the desorption ratio of furfural in the dynamic desorption system reached as high as 98.6%. CONCLUSION The unique micropore-confinement effect and excellent adsorption behavior make NC ZIF-8 1000C-800A an efficient adsorbent for furfural in the furfural–acetic acid system. © 2023 Society of Chemical Industry (SCI).

Semiconductor−metal–organic framework (MOF) hybrid photocatalysts become a significant material to treating dyeing wastewater recently. Zinc-based metal–organic framework (ZIF-8) stands out among numerous photocatalysts for its stability and simplicity to prepare. In this work, the gourd-shaped ZIF-8/TiO 2 nanocomposites are prepared in situ on anatase TiO 2 by precursor method, and then ZIF-8/TiO 2 nanocomposites is supported on PVDF electrospinning nanofibers by blending method to form ZIF-8/TiO 2 nanofibers (ZIF-8/TiO 2 NFs). We compare the degradation ability of Rhodamine B by ZIF-8/TiO 2 NFs with different mass ratios and load rates. When the mass ratio of ZIF-8 to TiO 2 is 2:1 and the loading rate is 10%, the photocatalytic degradation efficiency of ZIF-8/TiO 2 NFs reached 95.4%, and the degradation remained as high as 88.7% after cycling five times. The characterization results showed that ZIF-8 and TiO 2 are connected by N − Ti − O chemical bond, which narrows the band gap and rebars the recombination of the electron-hole pairs. The synergistic action of ZIF-8 and TiO 2 largely improves the photocatalytic capacity of ZIF-8/TiO 2 NFs.

Liver fibrosis (LF) is one of the major diseases that threaten human health. Until now, no effective drugs have been approved for clinical anti-liver fibrosis treatment. In this study, zeolitic imidazolate framework-8 (ZIF-8) lipid nanoparticles loaded with pirfenidone (PFD) and modified with vitamin A (VA) were constructed (VA-PFD@ZIF-8@DMPC NPs). PFD was embedded in ZIF-8 by the “one-pot” method, and the prepared ZIF-8 had a small particle size (84.3 nm) and high drug loading (54.46%). Moreover, the inherent pH sensitivity of ZIF-8 makes it stable in a normal physiological environment and collapsed in an acidic environment, thus controlling drug release and preventing drug leakage. Besides, the phospholipid layer makes the nano-drug delivery system dispersible and improves its biocompatibility . More importantly, VA is modified on the surface of nanoparticles (NPs), which can target the highly expressed retinol-binding protein receptor (RBPR) on the surface of hepatic stellate cells (HSCs), thereby accurately increasing the local drug concentration at the site of LF. In vivo experiments showed that VA-PFD@ZIF-8@DMPC NPs can reduce liver injury, improve the degree of LF, and exert specific therapeutic effects on LF. In conclusion, this nano-delivery system may become a novel and effective anti-liver fibrosis treatment.
