Rotational Paper-Based Microfluidic-Chip Device for Multiplexed and Simultaneous Fluorescence Detection of Phenolic Pollutants Based on a Molecular-Imprinting Technique

Ji Qi,Bowei Li,Xiaoyan Wang,Longwen Fu,Liqiang Luo,Lingxin Chen

In this study, we first present rotational paper-based microfluidic chips (RPADs) combined with a molecular-imprinting (MIP) technique to detect phenolic pollutants. The proposed rotational paper-based microfluidic chips could implement qualitative and quantitative analysis of two different phenolic contaminants, 4-nitrophenol (4-NP) and 2,4,6-trinitrophenol (TNP), simultaneously. Qualitative and quantitative analysis could be implemented simultaneously through fluorescence-intensity changes depending on the structures of quantum dots combined with a molecular-imprinting technique. Moreover, the rotational paper-based microfluidic chips provide a low cost, flexible, and easy way to operate the entire process conveniently. Under the optimal conditions, the proposed sensors showed high sensitivity and selectivity. Our final experimental results illustrated that the detection limits of 4-NP and TNP in the paper-based quantum-dot MIP (PQ-MIP) RPADs ranged from 0.5 to 20.0 mg/L, with detection limits of 0.097 and 0.071 mg/L, respectively. This novel rotational paper-based microfluidic device shows great potential and versatility for multiplexed, portable, and rapid testing of environmental and biological samples in the future.

Chip-based liquid phase microextraction combined with electrothermal vaporization-inductively coupled plasma mass spectrometry for trace metal determination in cell samples

Han Wang,Zhekuan Wu,Yuan Zhang,Beibei Chen,Man He,Bin Hu

Metal determination in precious biological samples is critical to metallomics research. Microfluidic chips provide an excellent platform for the miniaturization of extraction. In this work, a chip-based liquid phase microextraction (chip-based LPME) device was fabricated and combined with electrothermal vaporization-inductively coupled plasma mass spectrometry (ETV-ICP-MS) for the determination of trace Cu, Zn, Cd, Hg, Pb and Bi in cell samples and human serum samples. With sodium diethyldithiocarbamate (DDTC) as both the chemical modifier for low-temperature ETV-ICP-MS and the extracting reagent for the chip-based LPME, the chip parameters influencing the extraction efficiency including central channel width, branch channel ratio and central channel length were optimized by orthogonal design experiments. Under the optimized conditions, the limits of detection (LODs) are in the range of 6.6–89.3 pg mL−1 with the RSDs ranging from 4.23–8.15% (n = 7, c = 1 ng mL−1) for the target metal ions. The linear range covers three magnitudes with correlation coefficients higher than 0.99. The proposed method was validated by analyzing the target metal ions in certified reference materials of GBW07605 human hair and GBW09152 human serum, and metal determination in HepG2 cells, Jurkat T cells and a human serum sample were achieved with the recoveries in the range of 86.6–119%. The developed method is fast, has a low sample and reagent consumption and is highly sensitive, thus it is quite suitable for the analysis of precious biological samples such as cells.

A Surface-Enhanced Raman Scattering Optrode Prepared by in Situ Photoinduced Reactions and Its Application for Highly Sensitive On-Chip Detection

Shaoyan Wang,Chunyu Liu,Hailong Wang,Gang Chen,Ming Cong,Wei Song,Qiong Jia,Shuping Xu,Weiqing Xu

A surface-enhanced Raman scattering (SERS)-active optical fiber sensor combining the optical fiber waveguide with various SERS substrates has been a powerful analytical tool for in situ and long-distance SERS detection with high sensitivity. The design and modification of a high-quality SERS-active sensing layer are important topics in the development of novel SERS-active optical fiber sensors. Here, we prepared a highly sensitive SERS-active optrode by in situ fabrication of a three-dimensional porous structure on the optical fiber end via a photoinduced polymerization reaction, followed by the growth of photochemical silver nanoparticles above the porous polymer material. The fabrication process is rapid (finished within 1 h) and can be on line under light control. The porous structure supports vast silver nanoparticles, which allows for strong electromagnetic enhancement of SERS. Interestingly, the preparation of this SERS optrode and its utilization for SERS detection can all be conducted in a microfluidic chip. The qualitative and quantitative on-chip SERS sensing of organic pollutants and pesticides has been achieved by this SERS optrode-integrated microfluidic chip, and its high detection sensitivity makes it a promising factor in the analysis of liquid systems.

A silica-based SERS chip for rapid and ultrasensitive detection of fluoride ions triggered by a cyclic boronate ester cleavage reaction

Jian Zhang,Lifang He,Peirong Chen,Chao Tian,Jianping Wang,Bianhua Liu,Changlong Jiang,Zhongping Zhang

Chemical sensing for the convenient detection of trace aqueous fluoride ions (F−) has been widely explored with the use of various sensing materials and techniques. It still remains a challenge to achieve ultrasensitive but simple, rapid, and inexpensive detection of F− for environmental monitoring and protection. Here we reported a novel surface-enhanced Raman scattering (SERS) nanosensor, fluorescein phenylboronic acid covalently linked to 1,4-dimercapto-2,3-butanediol modified Au@Ag NPs by a cyclic boronate ester (Flu-PBA-Diol-Au@Ag NPs), for the rapid and ultrasensitive detection of F−. Once the Flu-PBA approached the surface of Au@Ag NPs, the Raman signals of Flu-PBA were remarkably enhanced due to the strong SERS effect. However, the presence of F− will induce the cleavage reaction of the cyclic boronate ester into the trifluoroborate anion (3F-Flu-PBA) and diol. The 3F-Flu-PBA molecules exfoliated from the surface of Au@Ag NPs, and the SERS signals of the nanosensor were quenched. Following the sensing mechanism, a silica-based SERS chip has been fabricated by the assembly of Flu-PBA-Diol-Au@Ag NPs on a piece of silicon wafer. The silica-based SERS chips showed high sensitivity for aqueous F−, and the limit of detection (LOD) could reach as low as 0.1 nM. Each test using the SERS chip only needs a droplet of 20 μL sample and is accomplished within ∼10 min. The silica-based SERS chip has also been applied to the quantification of F− in tap water and lake water.

The detection method for small molecules coupled with a molecularly imprinted polymer/quantum dot chip using a home-built optical system

Liu Yixi,Wang Yong,Liu Le,He Yonghong,He Qinghua,Ji Yanhong

A method to detect small molecules with a molecularly imprinted polymer/quantum dot (MIP-QD) chip using a home-built optical fluidic system was first proposed in this study. Ractopamine (RAC) was used as the model molecule to demonstrate its feasibility. The sensing of the target molecule is based on the quenching amount of the quantum dots. The method is facile, cost-saving, easy for miniaturization and avoids the cumbersome steps that are needed to get the fluorescent quenching curve using a spectrofluorometer. Most importantly, more details and accurate response time can be obtained by use of this method. The experimental results show that the prepared chips with low cost are highly selective and the home-built detection system allows the fast binding kinetics. The recorded quenching process was used to study the kinetic uptake of RAC onto the MIP-QD chip and the specificity towards RAC. The system can further be utilized to study the effect of the solvent, pH and temperature on the selectivity of the prepared MIP. The methodology could be extended to other similar studies with different molecules. Schematic illustration of the molecularly imprinted polymer/quantum dot chip capturing the target molecule

Integration of laminar flow extraction and capillary electrophoretic separation in one microfluidic chip for detection of plant alkaloids in blood samples

Yingzhi Hu,Huaidong Peng,Youshao Yan,Shuyu Guan,Shumei Wang,Paul C.H. Li,Yue Sun

The design, construction and testing for integration of liquid-liquid extraction (EX) and capillary electrophoretic (CE) separation on one glass microchip was reported. In this EX-CE chip, a 1.5 cm-long and 200 μm-wide EX channel was used for extraction based on the two-phase laminar flow , followed by a single-cross CE unit for on-line analysis without any auxiliary devices. One side of the EX channel surface for the organic solvent phase was selectively modified to be hydrophobic while the surface of the other side for the aqueous phase remained hydrophilic, and the extraction product reservoir is also used as the sample reservoir for the subsequent chip separation in the CE channel. With the surface-directed liquid flow behavior and liquid level adjustment in various reservoirs of the EX-CE chip, no disturbance occurred between the extraction (EX) and capillary electrophoretic (CE) units. A small heating block was placed under the chip to accelerate solvent evaporation after liquid-liquid extraction. Sanguinarine (SAN), a plant alkaloid , was used as a model analyte to evaluate the performance of the EX-CE chip. The influences of organic solvent type and liquid flow speed on the extraction efficiency were investigated. Rhodamine 123 (Rh123) was used as an internal standard for quantification of Sanguinarine (SAN) in a physiological buffer (e.g. PBS) or blood samples. A good linearity in the concentration range of 0.05 μg mL −1 to 0.1 mg mL −1 for SAN in PBS was obtained, with the detection limit of 0.5 ng mL −1 . Good repeatibilities for migration times (RSD of SAN is 0.63%, Rh123 is 0.91%, n = 5) and peak area ratio of SAN to Rh123 (RSD is 1.3%, n = 5) were obtained. For blood sample analysis, only 20 μL of sample was needed, and the whole analysis was finished in 17 min. In addition to the advantages in fast analysis speed, minimum sample handling, potential automation, the reported method showed an on-line sample pre-concentration capability.

Jing Zhang,Beibei Chen,Han Wang,Man He,Bin Hu

Trace heavy metals have great impact on biological system; therefore, it is essential to develop suitable analytical methods for the determination of trace heavy metals in biological samples to elucidate their biochemical and physiological functions in organisms. Herein, we presented a chip-based array monolithic microextraction system and combined it with inductively coupled plasma mass spectrometry (ICPMS) for online analysis of trace Hg, Pb, and Bi in real-world biological samples. Six ethylenediamine modified poly(glycidyl methacrylate-co-ethylene glycol dimethacrylate) (poly(GMA-co-EDMA-NH2)) capillary monolithic columns were embedded parallelly in microchannels of a microfluidic chip for array monolithic microextraction. Various parameters affecting the chip-based array monolithic microextraction of target metals were investigated. The sample throughput of the proposed method was 16 h–1, with the limits of detection for Hg, Pb, and Bi of 23, 12, and 13 ng L–1, respectively. The developed method was validated by the determination of trace Hg, Pb, and Bi in HepG2 cells and human urine samples, and the recoveries for the spiked samples were in the range of 90.4–102%. This chip-based array monolithic microextraction system is easy to prepare, and the proposed online analytical system provides a new platform for trace elements analysis in biological samples with the merits of high sample throughput, high sensitivity, and low sample/reagents consumption.

On-chip integrated multiple microelectromechanical resonators to enable the local heating, mixing and viscosity sensing for chemical reactions in a droplet

Hemi Qu,Yang Yang,Ye Chang,Zifan Tang,Wei Pang,Yanyan Wang,Hao Zhang,Xuexin Duan

On-chip integrating several functional components for developing integrated lab-on-a-chip microsystem remains as a challenge. In this work, by employing multiple microelectromechanical resonators both as actuators and sensors, on-chip heating, mixing and chemical reaction monitoring are successfully demonstrated. Mechanism studies using COMSOL simulations indicate that the local heating and mixing are induced by the acoustic wave attenuation during its transmission in liquid. On-line chemical reaction monitoring is realized by viscosity sensing using the same resonator through impedance analysis. Classic Diels-Alder reaction in a single droplet was performed to verify the feasibility of using such microsystem for mixing, heating and online reaction monitoring at microscale.

Design of membrane-based microfluidic chip device based on bionic leaf and adsorption detection of tetracycline antibiotics by specific gel membrane

Yang Yuan,Xu Zhu,Yang Liu,Zihan An,Hui Jia,Jie Wang

In this study, we developed copper alginate-carbon nanotube/copper sulfide-graphene oxide gel membranes based on the design inspired by the surface structure of leaves and plant transpiration in nature and through the modulation of high-density polymer networks. This light-driven water evaporation-specific adsorption gel membrane is capable of using solar energy to drive water evaporation from contaminated surface water to access tetracycline antibiotics for detection. Combined with the designed biomimetic adsorption detection chip, in situ monitoring of tetracycline antibiotics in water bodies was achieved. Compared with the traditional mass spectrometry detection method, it shows more outstanding performance in terms of time, environmental protection and technology. The microscale adsorption mechanism was further explored on the molecular scale by density functional theory (DFT). The results showed that tetracycline antibiotics were adsorbed on the aerogel membrane mainly through π-πEDA interactions in sandwich (S) configuration and parallel displacement (PD) configuration, which ensured high adsorption efficiency. Under the optimized conditions, the tetracycline concentration detected by the sensor showed a good linear relationship with the cyclic voltammetry curve, and the lowest detection limit was 1.59*10 −4  μM. This adsorption detection strategy can reduce the influence of organic matter, inorganic matter, and other classes of antibiotics on the detection of tetracycline antibiotics, and provide a new idea for the high-efficiency photo-thermal actuation and in-situ testing of tetracycline antibiotics.

Detection of Hg2+ in environmental water conditions by using a reusable SERS-based microfluidic chip with a high specificity and sensitivity

Yansheng Liu,Mengqi Wang,Guofu Wang,Xiaobo Jia,Jin Zhou,Hongqi Li,Haixin Chang,Zhaoxu Li

This work reported a surface-enhanced Raman spectroscopy (SERS)-based microfluidic chip that detects mercury ions (Hg 2+ ) in water with high sensitivity and good reproducibility. Silver nanoparticles (AgNPs) are easily fabricated on a Si substrate using a thin, thermally treated Ag film. Cy3 functionalized single-stranded DNA (Cy3-ssDNA) serves as the probe, which is immobilized on the AgNPs (Cy3-ssDNA/AgNPs), generating a SERS-based sensing surface. Due to the strong interaction between thymine (T) bases and Hg 2+ , in the presence of these ions, the ssDNA forms a T-Hg 2+ -T hairpin structure, which enhances the SERS signal. This method exhibits a limit of detection (LOD) of 1 × 10 −13  M. Furthermore, the proposed SERS chip demonstrates exceptional selectivity for mercury ions, as well as good reusability. The reusability of the SERS microfluidic chip was evaluated using the L-cysteine, which has a stronger affinity than T for Hg 2+ . By applying L-cysteine, the chip can be reused 10 times for the detection of Hg 2+ at concentrations as low as 1 × 10 −8  M. The method proposed in this study shows good sensitivity and holds good potential for application in the detection of Hg 2+ .

Hydrazone-linked covalent organic framework functionalized with cysteine as a fluorescence sensor and Exploration of paper chip for p-nitrophenol detection

Shan Yang,Xinyue Li,Yifang Liao,Yibing Ji,Ruijun Li

The large use and emission of p -nitrophenol (p-NP) seriously pollute the environment and endanger human health. In this work, a hydrazone-linked fluorescent covalent organic framework (BATHz-COF) was simply synthesized at room temperature and covalently linked N-acetyl-L-cysteine (NALC) via the "thiol-ene" click reaction, where carboxyl groups were introduced to improve dispersion and fluorescence intensity . As a rapid, good selectivity and reusability fluorescence sensor, the obtained COF-NALC has been used for quantitative analysis of p-NP predicated on the internal filtering effect (IFE). Under optimal conditions, COF-NALC enabled quantitative detection of p-NP with a linear range of 5–50 μM and the detection limit was 1.46 μM. The application of COF-NALC to the detection of p-NP in river water samples was successful, and the satisfactory recoveries were 98.0%–109.3%. Furthermore, the fluorescent COF paper chips constructed by in situ growth were combined with a smartphone to build a visual platform for the quick and real-time detection of p-NP, providing an excellent illustration for the development of intelligent fluorescence sensing in environmental analysis .

An enzymatic reaction-based SERS saliva analysis microporous array chip for chiral differentiation and high-throughput detection of D-amino acids

Lu Feng,Li Limao,Shen Kang,Qian Yayun,Zhang Pengfei,Yang Yan,Zhu Qunshan,Huang Yong,Yan Chunxiang,Wei Wei

A Raman-active boronate modified surface-enhanced Raman scattering (SERS) microporous array chip based on the enzymatic reaction was constructed for reliable, sensitive, and quantitative monitoring of D-Proline (D-Pro) and D-Alanine (D-Ala) in saliva. Initially, 3-mercaptophenylboronic acid (3-MPBA) was bonded to Au-coated Si nanocrown arrays (Au/SiNCA) via Au–S bonding. Following this, H 2 O 2 obtained from D-amino acid oxidase (DAAO)-specific catalyzed D-amino acids (D-AAs) further reduced 3-MPBA to 3-hydroxythiophenol (3-HTP) with a new Raman peak at 882 cm −1 . Meanwhile, the original characteristic peak at 998 cm −1 remained unchanged. Therefore, the I 882 /I 998 ratio increased with increasing content of D-AAs in the sample to be tested, allowing D-AAs to be quantitatively detected. The Au/SiNCA with large-area periodic crown structure prepared provided numerous, uniform “hot spots,” and the microporous array chip with 16 detection units was employed as the platform for SERS analysis, realizing high-throughput, high sensitivity, high specificity and high-reliability quantitative detection of D-AAs (D-Pro and D-Ala). The limits of detection (LOD) were down to 10.1 µM and 13.7 µM throughout the linear range of 20–500 µM. The good results of the saliva detection suggested that this SERS sensor could rapidly differentiate between early-stage gastric cancer patients and healthy individuals. Graphical Abstract
