Synthesis and biological evaluation of Cu(II), Zn(II), and Ni(II) 3-(4-nitrophenyl)acrylic acid complexes with diamines as potential urease inhibitors

Zhong-Ming Yang,Hui Zhu,Juan Sun,Shao-Song Qian,Meng-Na Cai,Hai-Liang Zhu

Five new Cu(II), Zn(II), and Ni(II) 3-(4-nitrophenyl)acrylic acid complexes were synthesized and evaluated for inhibitory activity on jack bean urease. All five complexes were structurally determined by single crystal X-ray analysis. Compared with the positive reference acetohydroxamic acid (IC50 = 13.25 μM), Cu(II) complexes 3 and 4 showed the strongest inhibitory activity against jack bean urease (IC50 = 1.23 and 1.17 μM). Ni(II) and Zn(II) complexes also exhibited inhibitory activities (IC50 = 10.09–13.10 μM).

Sequential recovery of Cu(II), Cr(III), and Zn(II) from electroplating sludge leaching solution by an on-line biosorption method with dosage controlling

Jing Wang,Ru-yi Zhou,Jun-xia Yu,Hui-sheng Wang,Qiu-yan Guo,Ke-qi Liu,Hao-dong Chen,Ru-An Chi

Hydrometallurgy leaching is a common method used to recycle heavy metals from electroplating sludge , but the subsequent separation of heavy metals from the complex leaching solution is difficult. In this study, Cu(II), Zn(II), and Cr(III) were sequentially recycled from electroplating sludge by an on-line biosorption method with dosage controlling. The results showed that the three metals mainly existed in their labile forms and could be leached by HNO 3 . A series of amine-modified bagasse biosorbents were prepared, and diethylenetriamine-modified sorbent was chosen and fixed in a column to remove heavy metals from the leaching solution. Sorbent dosage experiments showed Cu(II) was recycled exclusively at a dosage of 1.5 g/L, while Zn(II) and Cr(III) were recycled simultaneously at a dosage of 3.8 g/L when treating 800 mL of a leaching agent containing 13.4, 9.9, and 7.1 mg/L of Cu, Zn, and Cr. A linear relationship between the dosage and amount of adsorbed metal ions was obtained. The XPS results illustrated that the column had an adsorption affinity order of Cu(II)>Zn(II), Cr(III)≫Ca(II), Mg(II). Cu(II) was enriched in the foot layer, and Zn(II) and Cr(III) were enriched in the middle layer of the column, while co-ions could not be absorbed. According to these results, Cu(II), Zn(II), and Cr(III) could be sequentially recycled by using these columns in-series.

Degradation of sulfamethoxazole by a new modified Fenton-like process using Cu(II)-nitrilotriacetic acid complex as catalyst at neutral pH in aqueous medium

Khouloud Talbi,Lamia Mammeri,Bouchra Lekikot,Mohamed El Hadi Benssassi,Tahar Sehili

Fenton process is among the most effective advanced oxidation process in wastewater treatment , but the necessity to work at acidic pH, in order to avoid metal precipitation, limits its large-scale application. In this work, a new Fenton-like process using copper(II)-nitrilotriacetic acid (Cu II -NTA) complex as catalyst, was used for the activation of hydrogen peroxide (H 2 O 2 ) to degrade sulfamethoxazole (SMX) in aqueous medium under neutral pH conditions. The results showed that the Cu II -NTA/H 2 O 2 process was not efficient in the degradation of SMX. The reason being that the generation of HO radicals is still limited by the extremely slow Cu II -NTA/Cu I -NTA redox cycle. Therefore, the combination of hydroxylamine (HA) accelerate the Cu I recovery to induce the continuous production of HO radicals by the decomposition of H 2 O 2 . It was evident that the steady-state Cu I concentration in Cu II -NTA/HA/H 2 O 2 process was much elevated than the Cu II -NTA/H 2 O 2 process due to the quick regeneration of Cu I -NTA complex by HA. Kinetic profiles were evaluated under a wide range of experimental conditions in order to assess the effects of the initial concentrations, working pH, Cu(II):NTA molar ratio and reaction temperature. The results showed that the degradation rates were enhanced with the increase of H 2 O 2 and Cu II -NTA concentration during SMX oxidation. SMX was fully (83.64 %) degraded for an initial SMX concentration of 0.01 mM and an optimal Cu(II):NTA molar ratio of 1:1. Solution pH influenced SMX degradation and the highest degradation rate constant was achieved at pH 7.2. The radical scavenging experiments indicates that hydroxyl radical (HO ) has major role for SMX degradation. The presence of metal ions such as Mg II , Ca II and Mn II had no impacts or little inhibitory effect on the Cu II -NTA complex catalyzed Fenton-like reaction, however, a complete degradation of SMX was achieved in the presence of Fe III ions. Also, the effects of various inorganic anions such as Cl − , SO 4 2− , NO 3 − and HCO 3 − on the oxidation efficiency of SMX were studied. The efficiency of mineralization of aqueous solution of SMX was evaluated by measuring total organic carbon (TOC). These results demonstrate that Cu II -NTA catalyzed Fenton-like oxidation may be an efficient approach for remediation of wastewater containing antibiotics.

The diversity of the coordination bond generated a POSS-based fluorescent probe for the reversible detection of Cu(II), Fe(III) and amino acids

Kun Zhang,Xiaoni Wang,Minggang Tian,Zhiming Gou,Yujing Zuo

In recent years, it has been found that Cu2+, Fe3+, and amino acids play an irreplaceable and subtle role in organisms and have attracted the considerable attention of many researchers. Therefore, it is vital to design visual indicators to reveal the relationships between metal ions and amino acids. However, there have been few reports on this vigorous subject. Fortunately, based on the different coordination effects between metal ions and boron groups, we have designed an accessible fluorescent probe (PSI-A). Borane was introduced as an ion-sensitive group to form a novel POSS-based fluorescent probe, which achieves fascinating performance, in situ dynamic multiple detection, excellent photostability, and enervative biological toxicity. PSI-A exhibited predominant selectivity and sensitivity to Cu2+/amino acids and Fe3+/amino acids sequence reactions in HepG2 cells and zebrafish. The fluorescence of PSI-A was quenched by Cu2+, which can be recovered by adding Asp, Ser, Arg, Ace or Trp. Additionally, the fluorescence of PSI-A quenched by Fe3+ can be restored after adding Asp. PSI-A is available to monitor Cu2+/amino acids and Fe3+/amino acids sequence reactions and can be repeated for at least three consecutive cycles without a fatigued performance. Therefore, this multifunctional fluorescent probe may have prospective application potentials in the biological field.

A facile green and one-pot synthesis of grape seed-derived carbon quantum dots as a fluorescence probe for Cu(II) and ascorbic acid

Jiawei Li,Ouwen Xu,Xiashi Zhu

In this study, an on–off–on fluorescence probe for the detection of trace Cu(II) and ascorbic acid (AA) based on biomass-derived sulfur and nitrogen double heteroatom-doped carbon dots (N,S-CDs) was designed. For the first time, the probe (N,S-CDs) was prepared from grape seeds and thiourea as the precursor. Cu(II) was added to the carbon point solution, the fluorescence intensity (FL) of N,S-CDs was strongly quenched (switch OFF) and the fluorescence probe turned to “ON” (switch ON) with the addition of AA. Under the optimal conditions, the as-synthesized N,S-CDs had a good detection performance for Cu(II) and AA assay with the linearity ranges from 150–500 μg mL−1 and 0.1–400 μg mL−1, and the LODs were 0.048 mg L−1 and 0.036 mg L−1, respectively. The as-prepared N,S-CDs exhibited a low cytotoxicity and a good biocompatibility, which show their potential for application in the biological imaging of living cells.

Bioactivity and computational studies on the induction of urease inhibition by three Cu(II) complexes with a fluorinated Schiff base and different secondary ligands

Wei Zhang,Hu Wang,Chao Ding,Yizhu Lei,Chaochuang Yin,Renshu Wang,Qiming Yang,Tingting Wu,Mei Zhang

Three mononuclear Cu(II) complexes, [Cu(C 16 H 12 NO 3 F)(H 2 O)] ( C1 ), [Cu(C 16 H 12 NO 3 F)(C 10 H 9 N 3 )]·H 2 O·0.5CH 3 OH ( C2 ) and [Cu(C 16 H 12 NO 3 F)(C 3 H 4 N 2 )] ( C3 ) with a fluorinated Schiff base ligand (C 16 H 12 NO 3 F = 5-fluorosalicylaldehyde-L-phenylalanine) and different secondary ligands (C 10 H 9 N 3  = 2,2′-dipyridylamine; C 3 H 4 N 2  = imidazole) were synthesized and structurally characterized. Theoretical calculation of the three complexes was carried out by density functional theory (DFT) Becke's three-parameter hybrid (B3LYP) method using the Gaussian 09 package. The calculation results are in accordance with experimental results. In addition, the inhibitory activities of C1 - C3 were tested in vitro against jack bean urease. At the same time, molecular docking was investigated to determine the probable binding mode. The experimental values and docking simulation exhibited that C1 showed strong inhibitory activity (IC 50  = 0.68 ± 0.09 μM) compared with the positive reference acetohydroxamic acid (IC 50  = 27.73 ± 2.93 μM). Moreover, a 100-ns molecular dynamics (MD) simulation was performed to evaluate the stability of urease (PDB ID: 3LA4) and the binding stability of the 3LA4 /C1 and 3LA4/ C3 using the Desmond 2021 from Schrödinger. Their structure-inhibitory activity relationship was further discussed from the perspective of molecular docking, theoretical calculation and molecular dynamics simulation.

Design, synthesis, crystal structure and cytotoxicity studies of colorimetric fluorescent “OFF-ON” probes for rapid detection of hydrogen sulfide based on Cu(II) complex

Xuefang Shang,Xuejing Yue,Yanmei Chen,Congshu Li,Hongli Chen,Tianyun Wang

Four colorimetric fluorescent probes based on copper(II) complexes were synthesized and the molecular structures were characterized by X-ray diffraction, ESI-HRMS and elemental analysis . When exposed to HS − , the synthesized probes showed significant color changes from yellow to red and remarkable increase of fluorescence intensity (over 80-fold). What's more, the interaction of host-guest could be completed in 2 min. However, no clear color and fluorescence intensity changes were observed in the presence of other anions (AcO − , H 2 PO 4 − , F − , Cl − , Br − , I − , SO 4 2− , SO 3 2− , CO 3 2− , Cys and GSH). The synthesized colorimetric fluorescent probes could detect HS − rapidly and conveniently. Cytotoxicity studies indicated that the synthesized fluorescent probes were low cytotoxicity in Hela cells and may be used to detect H 2 S level in vivo.

A novel polyamine-type starch/glycidyl methacrylate copolymer for adsorption of Pb(II), Cu(II), Cd(II) and Cr(III) ions from aqueous solutions

Youning Chen,Wei Zhao,Huan Wang,Xiaohua Meng,Linjie Zhang

A novel polyamine-type starch/glycidyl methacrylate (GMA) copolymer with a high capacity for the adsorption of heavy metal ions was prepared via graft copolymerization of GMA and corn starch and a subsequent amination reaction between amino group of diethylenetriamine and epoxy group in GMA. The copolymers were characterized by Fourier transform infrared spectrometry, X-ray diffraction and scanning electron microscopy, and adsorption properties on modified starch of Cu(II), Pb(II), Cd(II) and Cr(III) were studied. By analysing the relationship between adsorption capacity and pH, adsorption isotherms and adsorption kinetics, it is proved that the adsorption of the four metal ions is mainly based on the chemical adsorption of coordination. The maximum adsorption capacities of the copolymer were up to 2.33, 1.25, 0.83 and 0.56 mmol g−1 for Cu(II), Pb(II), Cd(II) and Cr(III), respectively. The adsorption of the four concerned metal ions was hardly affected by common coexisting ions such as Na(I), K(I), Ca(II) and Mg(II), whereas it was slightly decreased when Fe(II) and Zn(II) coexisted in the solution, which illustrates the selective adsorption of Cu(II), Pb(II), Cd(II) and Cr(III) from wastewater. After 10 cycles of adsorption–desorption experiments, there was no significant change in the adsorption capacity, indicating that the polyamine-type starch/GMA copolymer has high adsorption capacity and good reusability.

Fabrication of xanthate-modified chitosan/poly(N-isopropylacrylamide) composite hydrogel for the selective adsorption of Cu(II), Pb(II) and Ni(II) metal ions

Shuping Wu,Fan Wang,Haikuan Yuan,Lijuan Zhang,Shimin Mao,Xijian Liu,Njud S. Alharbi,Sohrab Rohani,Jie Lu

A novel xanthate-modified chitosan/poly( N -isopropylacrylamide) (X-CS/NIPAAm) composite hydrogel was fabricated by a facile two-step method for the selective adsorption of Cu(II), Pb(II) and Ni(II) metal ions from aqueous solutions. The equilibrium adsorption data fitted well to the Langmuir model and the adsorption kinetic data followed the pseudo-second-order model. The maximum equilibrium adsorption capacities of composite hydrogel to Cu(II), Pb(II) and Ni(II) at 293   K were about 115.1, 172.0 and 66.9   mg   g −1 , respectively. The thermodynamics parameters showed that the adsorption processes to Cu(II), Pb(II) and Ni(II) were spontaneous and exothermic. The interactions between metal ions for multi-components systems were evaluated by the values of Q b /Q s and Q t /Q s . The adsorption mechanism was analyzed by XPS spectra and the Hard–Soft, Acids–Bases (HSAB) theory. The X-CS/NIPAAm composite hydrogel could be regenerated and repeatedly utilized at least five cycles without obvious adsorption capacity loss.

Adsorptive removal of Cu(II) from aqueous solutions using a novel macroporous bead adsorbent based on poly(vinyl alcohol)/sodium alginate/KMnO4 modified biochar

Zhixing Xiao,Lijuan Zhang,Long Wu,Dan Chen

In this study, a novel adsorbent was synthesized through the entrapment of KMnO 4 modified biochar (MB) in poly(vinyl alcohol)/sodium alginate hydrogel bead (PSMB) and then tested to determine its capacity for removal of Cu(II) from aqueous solutions. The maximum Cu(II) adsorption capacity estimated with Langmuir isotherm modeling was 87.07 mg/g of PSMB, which was higher than that of previously reported adsorbents. PSMB had a good selectivity towards Cu(II) as well as high anti-interference ability. The main species for Cu(II) adsorption likely were the inner-sphere Cu-O complexes, as indicated by the results of spectral analyses. The PSMB was easily collected and exhibited a minimal loss of adsorption capacity after 10 adsorption/regeneration cycles. The effective removal of Cu(II) from environmental water samples with relative low treatment cost indicated a possibility for practical application.

Versatile hyaluronic acid modified AQ4N-Cu(II)-gossypol infinite coordination polymer nanoparticles: Multiple tumor targeting, highly efficient synergistic chemotherapy, and real-time self-monitoring

Shihong Shen,Youshen Wu,Ke Li,Ya Wang,Jimin Wu,Yun Zeng,Daocheng Wu

A novel strategy for the preparation of infinite coordination polymer nanoparticles (ICPs) based nanomedicines was developed, with which hyaluronic acid modified AQ4N-Cu(II)-gossypol nanoparticles (HA@AQ4N-Cu(II)-gossypol NPs) were obtained. This is a highly efficient nanomedicine, in which gossypol serves as a chemotherapeutic agent and a self-carrier material; Cu(II) serves as the connecting point and anti-tumor enhancer; AQ4N not only serves as a chemotherapeutic agent and self-carrier material, but also as the self-monitor based on its inherent fluorescence. HA@AQ4N-Cu(II)-gossypol NPs possessed a spherical shape with a dynamic size of 88.7 ± 7.4 nm, and the total drug-loading content and drug encapsulation efficiency are 77.41% and 100%, respectively. This nanomedicine has a multiple tumor-targeting ability caused by HA-receptor mediation and pH-responsive drug release. A significantly low combination index (0.097) of AQ4N and gossypol is ascertained. In vivo experiments indicate that it accumulates and significantly releases drugs at the tumor region. With the use of only one-fiftieth of AQ4N and half of gossypol of the generally administered dose, they can achieve significantly high anti-tumor efficiency with negligible side effects. Importantly, the switching-type changed fluorescence of AQ4N can be used for in vivo real-time self-monitoring of the drug release and distribution, which allows us to adjust the administration dose and time for different tumor types and stages for individual therapy.

A comparison of adsorption capacity of chitosan– rhamnolipid–hydrotalcite-like compound composites synthesized by two methods towards highly efficient removal of Cu(II) ions

Yan Li,Xiaoming Mao,Yaqin Liang,Mengzhao Li

Copper pollution in global water resources is becoming increasingly severe, posing a serious threat to human health and aquatic ecosystems. In this paper, two chitosan (CS)–rhamnolipid (RL)–hydrotalcite-like compound (HTlc) composites, designated as CS@(RL-HTlc) and (RL-HTlc)/CS, were synthesized using two distinct methods: emulsion crosslinking–coprecipitation–ion exchange and coprecipitation–ion exchange–emulsion crosslinking, respectively, to investigate their adsorption properties for Cu(II) ions. Characterization revealed that the CS@(RL-HTlc) composite possessed a crosslinked CS core with an RL-HTlc shell structure, while the (RL-HTlc)/CS microsphere exhibited a porous structure with a rough surface. The adsorption properties of these composites for Cu(II) ions were thoroughly investigated and exhibited significantly enhanced adsorption performances due to the synergistic effect between CS and RL-HTlc. Specifically, the coordination of Cu(II) ions with N/O-containing functional groups and the isomorphous substitution of Mg(II) by Cu(II) ions within the HTlc layers. Additionally, the surface-induced precipitation of Cu(OH) 2 was identified as another adsorption mechanism for CS@(RL-HTlc). Interestingly, the (RL-HTlc)/CS composite, with a larger specific surface area and a lower isoelectric point, exhibited a higher adsorption capacity compared to CS@(RL-HTlc). Moreover, (RL-HTlc)/CS outperformed CS@(RL-HTlc) as a more suitable candidate for repeated use in wastewater treatment.
