Scallop hydrolysates/κ-carrageenan hydrogels improve the alleviating effect of curcumin on DSS-induced colitis

Jia-Nan Yan,Yu-Qiao Wang,Lin Li,Zhu-Jun Zhang,Ling-Yi Gao,Bin Lai,Ce Wang,Li-Chao Zhang,Hai-Tao Wu

Scallop ( Patinopecten yessoensis ) male gonad hydrolysates/ κ -carrageenan (SK) double cross-linking network hydrogels were constructed as curcumin (Cur) delivery vehicles with good gel mechanical property and homogeneous honeycomb microstructures. Compared with Cur + DSS group, weight loss and colon length in SK-Cur + DSS group increased about 1.04 and 1.06 folds, while the disease active index (DAI) scores decreased about 13 %. Moreover, SK-Cur showed superior effect on intestinal barrier improvement, oxidative stress modulation, and inflammatory expression regulation, reflected by 1.1 folds increment of epithelial tight junction proteins, 16 % and 27 % decline in MPO and iNOS levels, 20 % decline in NF-κB expression, 4 %, 7 %, 6 % decline in TNF-α, IL-1β, IL-6 expression in serum, and 9 %, 20 %, 7 % decline in TNF-α, IL-1β, IL-6 expression in colon tissue. Furthermore, SK-Cur enriched alpha diversity and restored taxonomic composition with increasing Bacteroides and Verrucomicrobia abundance accompanied by decreasing Firmicutes in DSS-induced UC mice, roughly to a normal gut flora state.

Utilization of soybean protein isolate hydrolysates as carriers: Improved encapsulation efficiency and stability of curcumin

Danhua Ma,Xiaoying Zhang,Naveed Mahmood,Qingkui Zhao,Yang Li,Shuang Zhang

This study aimed to explore the potential of soybean protein isolate hydrolysates (SPIH) prepared via Alcalase as delivery carriers and develop novel SPIH-Cur nanoparticles. Hydrolysis caused the varying degrees degradation in the 7S and 11S subunits, significantly enhancing SPI's antioxidant activity. The reduction in particle size and the exposure of hydrophobic groups in SPIH contributed to the formation of stable SPIH-Cur nanoparticles, due to their well binding capacity to curcumin (Cur). The 30 min SPIH-Cur sample exhibited the highest encapsulation efficiency (83.09 %), owing to its high binding affinity (Ka = 9.56 × 10 3  M −1 ). Encapsulation by SPIH also significantly improved Cur's thermal and light stability. Moreover, FTIR, fluorescence spectra, and molecular docking analyses revealed that the formation of SPIH-Cur were primarily driven by hydrophobic forces and hydrogen bonds. Above results provide a foundation for fabricating nanoparticles that deliver lipophilic bioactive compounds with high encapsulation efficiency and stability derived from SPIH.

Preparation and mechanism of Lactobacillus plantarum and curcumin co-encapsulated in alginate-gelatin hydrogel beads

Rongmi Zhou,Fangfang Ni,Shiyi Wu,Xinyu Wang,Jing Ming,Lantong Zhang,Lijuan Dong,Zhijun Song,Gerui Ren,Min Huang,Shuangbin Xu,Qiong Zhang,Khaled F. El-Massry,Hujun Xie

Lactobacillus plantarum (LP) and curcumin (CUR) were co-encapsulated in sodium alginate– Lactobacillus plantarum– curcumin–gelatin (ALG–LP–CUR–GE) hydrogel beads to evaluate the synergistic function of probiotics and prebiotics. The encapsulation efficiency of LP and CUR peaked at 84.6 % and 94.7 %, respectively. Results of scanning electron microscope, Fourier transform infrared spectrometry, and X-ray diffraction confirmed the encapsulation of LP and CUR in the hydrogel beads. In vitro digestion suggested that both CUR and LP can achieve successful release in the simulated intestinal fluid. The release kinetics of CUR showed that it exhibited predominantly Fickian release and non-Fickian release in gastric fluid and intestinal fluid, respectively. The application of co-encapsulated hydrogel beads in mimic milk tea illustrated that the viable count of LP remained above the minimum recommended threshold of 10 7  CFU/mL, offering a promising avenue for the delivery of synergistic probiotic and prebiotic functionalities.

Supercritical CO2 assisted micronization of curcumin-loaded oil-in-water emulsion promising in colon targeted delivery

Zi-Dan Wang,Hu-Hong Peng,Yi-Xin Guan,Shan-Jing Yao

Bioavailability of oral administration is quite limited due to hydrophobicity of most of cancer drugs. Emphasis has been placed on the development of oral drug delivery systems to enhance solubility and stability of chemotherapeutics. In this work, a novel oral drug delivery system based on emulsion was implemented to encapsulate curcumin , in which ethylcellulose was employed as oil phase polymer while gum arabic was selected as emulsifier and wall material. After processed by ultrasonic emulsification , oil-in-water emulsion loaded with curcumin was transformed from liquid state to solid particles by an improved supercritical CO 2 assisted atomization technique (SAA-HCM) to achieve its long-term stability. Effects of SAA-HCM operating conditions on sizes and distributions of microparticles were investigated in detail and particle formation mechanism was discussed. The chemical stability and amorphous form of microparticles were identified by solid characterization. In vitro drug release behavior of curcumin-loaded microparticles exhibited controlled release and enzyme triggered properties, which implies the potential in the application of colon targeted delivery. This work expands the application of SAA-HCM technique to emulsion, which will be of great importance in the design of water-insoluble drug loaded microparticles with elaborate structures.

Synthesis and Characterization of Curcumin-Functionalized HP-β-CD-Modified GoldMag Nanoparticles as Drug Delivery Agents

Lian Ting,Peng Mingli,Vermorken Alphons J. M,Jin Yanyan,Luo Zhiyi,Van de Ven Wim J. M,Wan Yinsheng,Hou Peng,Cui Yali

Curcumin, a polyphenol extracted from turmeric (Curcuma longa), has emerged as a potent multimodal cancer-preventing agent. It may attenuate the spread of cancer and render chemotherapy more effective. However, curcumin is neither well absorbed nor well retained in the blood, resulting in low efficacy. In an attempt to enhance the potency and to improve the bioavailability of curcumin, new delivery agents, hydroxypropyl-beta-cyclodextrin (HP-β-CD)-modified GoldMag nanoparticles (CD-GMNs) were designed and synthesized to incorporate curcumin. The CD-GMNs were characterized by Fourier Transform Infrared Spectroscopy (FT-IR), Thermo-gravimetric Analysis (TGA), X-ray Diffraction (XRD), Dynamic Light Scattering measurements (DLS), Transmission Electron Microscopy (TEM) and Vibrating Sample Magnetometer (VSM) analyses. For the magnetic carrier of CD-GMNs, the content of HP-β-CD was 26.9 wt%. CD-GMNs have a saturation magnetization of 22.7 emu/g with an average hydrodynamic diameter of 80 nm. The curcumin loading, encapsulation efficiency and releasing properties in vitro were also investigated. The results showed that the drug encapsulation ratio was 88% and the maximum curcumin loading capacity of CD-GMNs was 660 μg/5 mg. in vitro drug release studies showed a controlled and pH-sensitive curcumin release over a period of one week. Collectively, our data suggest that HP-β-CD-modified GoldMag nanoparticles can be considered to form a promising delivery system for curcumin to tumor sites. Targeting can be achieved by the combined effects of the application of an external magnetic field and the effect on drug release of lower pH values often found in the tumor microenvironment.

Fabrication of rhamnogalacturonan-I enriched pectin-based emulsion gels for protection and sustained release of curcumin

Laiming Zhang,Jiaqi Zheng,Yi Wang,Xingqian Ye,Shiguo Chen,Haibo Pan,Jianle Chen

Rhamnogalacturonan-I (RG-I) was considered to be incompatible with gel formation. Due to its important health benefits, our previous study reported synergistic gelation of RG-I enriched pectin from citrus membranes with sequential acidic and alkaline treatment. In the present study, we further fabricated RG-I enriched pectin-based emulsion gels with the synergistic gelation for hydrophobic bioactive encapsulation. The effect of oil fraction and interfacial compositions including pectin itself and Tween 20 on the physical properties and delivery characteristics of emulsion gels was investigated. As to interfacial compositions, pectin reduced syneresis and swelling ratio and increased water holding capacity . Oil fraction had no marked influence on syneresis and swelling ratio, but significantly increased water holding capacity. Particularly, all the emulsion gels showed an elastic behavior and a full structure recovery after shearing. Increase in pectin instead of Tween 20 onto the surface of oil droplets and oil fraction markedly improved their gel strength. Curcumin was encapsulated within the oil phase of emulsion gels. Improvement in gel strength provided enhanced protection of curcumin against heat-induced degradation. In vitro digestion revealed that interfacial compositions decelerated curcumin release by increasing the proportion of pectin onto the surface, but didn't change its bioaccessibility eventually. Increase in oil fraction accelerated curcumin release, but reduced its bioaccessibility. Overall, fabrication of RG-I enriched pectin-based emulsion gel with pectin itself as interfacial composition and low oil fraction would contribute to improving curcumin bioavailability for its sustained release, which should be further validated in vivo .

Curcumin has been reported to possess multiple bioactivities, such as antioxidant, anticancer, and anti-inflammatory properties, however the clinical application of curcumin has been significantly limited by its instability and poor metabolism. Modification of curcumin has led to discovery and development of lots of novel therapeutic candidates. In recent years acute and chronic inflammation has been the focus of numerous studies in various diseases. Here, we synthesized a series of asymmetrical curcumin analogs with highin vitrochemical stability, and their anti-inflammatory activity was evaluated in LPS-stimulated macrophages. According to the bio-screening results and QSAR analysis, these analogs exhibited potent activities against LPS-induced TNF-α and IL-6 release. Among the analogs of the potent anti-inflammatory activity, compounds3b8and3b9exhibited significant protection and possess enhanced anti-inflammatory activity thereby attenuated the LPS-induced septic death in mice. Keywords:curcumin;asymmetrical curcumin analogs;anti-inflammation;QSAR;cytokines

Using gelatin/curcumin nano-fiber membranes as scaffolds in a subcutaneous model for tissue engineered cartilages

Kang Bo Kyoung,Yu Zheyuan,Chen Weiming,Jiang Taoran,Shim Yoong-hun,Gao Jiaman,Zhou Guangdong,Cao Dejun

Engineered cartilage has several applications in treating cartilage ossification, however, its use is restricted clinically. We explored the feasibility of engineered cartilage in constructing tissues using gelatin/curcumin nano-fiber membranes as scaffolds in subcutaneous models. We constructed cartilage with gelatin nano-fiber membrane (control group) and gelatin/curcumin nano-fiber membrane (experimental group) as scaffolds. After the material was implanted into the back of BALB/c mice, gross view observation was performed. Histological examination was performed 3 and 12 weeks after implantation in vivo, and cartilage formation at different time points was compared. Gross observation showed that compared to the control group, the vascularization of nearby tissues in the experimental group was significantly inhibited. The Scanning electron microscope observation showed that the chondrocytes in both groups adhered well. The growth curve of the chondrocytes showed that curcumin had no significant effect on cell growth. Histological observation showed that the cell-material complexes in both groups had cartilage lacuna formation at 3 and 12 weeks. However, compared with that of the control group, the experimental group showed obvious absorption and thicker cartilage matrix with more homogenization. Gelatin/curcumin scaffolds were successfully used to construct engineered cartilage tissues in subcutaneous animal models. Our findings demonstrate that curcumin-loaded scaffolds have great clinical applications.

Enhanced Bioavailability and Anticancer Effect of Curcumin-Loaded Electrospun Nanofiber: In Vitro and In Vivo Study

Wang Chuan,Ma Chao,Wu Zhenkai,Liang He,Yan Peng,Song Jia,Ma Nan,Zhao Qinghua

Nanofibers have attracted increasing attention in drug delivery and other biomedical applications due to their some special properties. The present study aims to prepare a fiber-based nanosolid dispersion system to enhance the bioavailability of curcumin (CUR). CUR-loaded polyvinyl pyrrolidone (CUR@PVP) nanofibers were successfully prepared via electrospinning. Scanning electron microscopy (SEM) was employed to observe the morphology of the nanofibers, and the SEM image showed that the drug-loaded nanofibers were smooth, and no CUR clusters were found on the surface of the nanofibers. The results of X-ray diffraction (XRD) demonstrated that the CUR was evenly distributed in the nanofibers in an amorphous state. Fourier transform infrared (FTIR) spectroscopy analysis indicated that intermolecular hydrogen bonding occurred between the CUR and the polymer matrix. In vitro dissolution profiles showed that CUR@PVP nanofiber could be quickly dissolved in phosphate-buffered saline (PBS) solution, while negligible dissolution was observed in pure CUR sample. Importantly, in vitro cell viability assays and in vivo animal tests revealed that the nanosolid dispersion system dramatically enhanced the bioavailability and showed effective anticancer effect of the CUR.

Hybridization of Curcumin Analogues with Cinnamic Acid Derivatives as Multi-Target Agents Against Alzheimer’s Disease Targets

Eirini Chainoglou,Argyris Siskos,Eleni Pontiki,Dimitra Hadjipavlou-Litina

The synthesis of the new hybrids followed a hybridization with the aid of hydroxy-benzotriazole (HOBT) and 1-Ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDCI.HCL) in dry DMF or thionyl chloride between curcumin analogues and cinnamic acid derivatives. IR,1H-NMR,13C-NMR, LC/MS ESI+, and elemental analysis were used for the confirmation of the structures of the novel hybrids. The lipophilicity values of compounds were calculated theoretically and experimentally via the reversed chromatography method as RMvalues. The novel derivatives were studied through in vitro experiments for their activity as antioxidant agents and as inhibitors of lipoxygenase, cyclooxygenase-2, and acetyl-cholinesterase. All the compounds showed satisfying anti-lipid peroxidation activity of linoleic acid induced by 2,2′-azobis(2-amidinopropane) hydrochloride (AAPH). Hybrid3ewas the most significant pleiotropic derivative, followed by3a. According to the predicted results, all hybrids could be easily transported, diffused, and absorbed through the blood–brain barrier (BBB). They presented good oral bioavailability and very high absorption with the exception of3h. No inhibition for CYP1A2, CYP2C9, CYP2C19, CYP2D6, and CYP3A4 was noticed. According to the Ames test, all the hybrids induced mutagenicity with the exception of3d. Efforts were conducted a) to correlate the in vitro results with the most important physicochemical properties of the structural components of the molecules and b) to clarify the correlation of actions among them to propose a possible mechanism of action. Docking studies were performed on soybean lipoxygenase (LOX) and showed hydrophobic interactions with amino acids. Docking studies on acetylcholinesterase (AChE) exhibited: (a) hydrophobic interactions with TRP281, LEU282, TYR332, PHE333, and TYR336 and (b) π-stacking interactions with TYR336.Keywords:curcumin analogues;cinnamic acids;Hybrids;Alzheimer’s disease;therapy

Curcumin inhibits viability and promotes apoptosis by modulating miR-17/caspase-9 pathway in colorectal cancer

Jun Tang,Jingfang Yang

Purpose: To investigate the potential role of curcumin in osteosarcoma (OS) cell migration and invasion, and also to explore the underlying mechanism of action. Methods: OS cell lines, Saos-2 and U2OS, were treated with various concentrations of curcumin (0, 10, 20, 50 μM) for 48 h. Transwell assay was used to determine the migratory and invasive activities of human OS cells while miR-33b-5p expression levels were measured using quantitative real time polymerase chain reaction (qRT-PCR). Expressions of metastasis-related genes, such as matrix metalloproteinases (MMPs), were quantified by qRT-PCR or western blot. The interaction between miR33b-5p and Sirtuin 6 (SIRT6) was evaluated by luciferase assay. Results: Curcumin inhibited the migratory and invasive activities of huamn OS cells, and downregulated MMP2 and MMP9 (p < 0.05). Moreover, in curcumin-treated OS cells, miR-33b-5p expression was upregulated while SIRT6 expression was significantly downregulated (p < 0.05). Overexpression of miR-33b-5p downregulated SIRT6 mRNA and protein expression, whereas transfection with miR-33b-5p inhibitor upregulated SIRT6 expression in OS cells (p < 0.05). Furthermore, curcumin suppressed SIRT6 expression by upregulating miR-33b-5p expression in OS cells. Finally, overexpression of SIRT6 or miR-33b-5p inhibitor reversed the suppression of OS cell metastasis induced by curcumin (p < 0.05). Conclusion: Curcumin inhibits SIRT6 expression via upregulating miR-33b-5p expression, which leads to the inhibition of OS cell migration and invasion. Thus, curcumin is a potential therapeutic agent for the management of OS.

Design, synthesis, and evaluation of mono-carbonyl analogues of curcumin (MCACs) as potential antioxidants against periodontitis

Ya Zhao,Zhiwei Zheng,Menghan Zhang,Yi Wang,Rongdang Hu,Weijia Lin,Chenyang Huang,Chuchu Xu,Jianzhang Wu,Hui Deng

Background and objective The application of curcumin is limited by its instability. Mono-carbonyl analogues of curcumin (MCACs) are structurally stable, yet the intermediate bridging ketones in their skeletons account for increased toxicity. This study aimed to synthesize and screen MCACs that exhibit low cytotoxicity and high antioxidant ability, and the effects of MCACs on experimental periodontitis were also investigated. Materials and methods The cytotoxicity of MCACs on MC3 T3-E1 was determined by MTT assay. The antioxidant capacity was investigated by the cell viability against H 2 O 2 -induced damage and the level of reactive oxygen species (ROS) and malondialdehyde (MDA). The localization and protein expression of nuclear factor erythroid 2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1) was detected by immunofluorescence and western blot, respectively. In addition, MCAC was intragastrically administrated in rats with ligature-induced experimental periodontitis. The effects were assessed by bone resorption, as well as the immunohistology staining of inflammatory and oxidative stress markers. Results MCACs with cyclopentanone and containing pyrone showed lower toxicity than natural curcumin were synthesized ( 1A - 10A , 1H - 10H ), among which, 1A exhibited the most potent cytoprotective effect against H 2 O 2 -induced damage. Such effects could be explained by the reduced MDA and ROS level, possibly through the nucleus translocation of Nrf2 and the induction of HO-1. Micro-CT results further indicated that 1A significantly reduced bone loss, along with an increased level of Nrf2 and HO-1, and decreased TNF-α and IL-1β. Conclusion The present study has synthesized a novel antioxidant MCAC 1A with good biosafety and stability. MCAC 1A could serve as a host response modulator with preventive and protective effects on periodontitis.
