Purpose This study investigated the basic composition, in vitro digestibility, biological activities, and potential bioactive peptides of Pacific oyster (PO), Mytilus edulis (ME), Argopecten irradians (AIS), and Meretrix meretri x L. (MML) after simulated gastrointestinal digestion (SGD). Methods SGD products were prepared using the INFOGEST 2.0 model, and their nutritional properties and biological activities were analyzed. A comprehensive peptidomic profiling of the digested samples was conducted using nano-liquid chromatography-tandem mass spectrometry (nano-LC MS/MS) analysis. Results All four shellfish exhibited a nutritional profile characterized by high protein and low-fat content. The digestibility of shellfish increased progressively, while the average molecular weight (MW) of the hydrolysates gradually decreased during SGD, with the intestinal hydrolysates showing the highest overall digestibility. Biological activity analysis demonstrated that intestinal digestion products from all four species exhibited notable ABTS radical scavenging activity and α-glucosidase inhibition activity, with PO and ME exhibiting the strongest effects. A total of 349, 99, 314 and 105peptides were identified from the digestion products of PO, ME, AIS, and MML, respectively. Among these, PO and AIS showed the highest sequence similarity, sharing 69 identical peptides. By contrast, MML displayed distinct peptidomic characteristics, including a higher proportion of large MW peptides. Conclusion This study provides a comprehensive peptide catalog of shellfish digestion products and offers new insights into the biological activities of shellfish-derived peptides, supporting their potential as functional food resources.

Liubao tea (LBT), a traditional dark tea, is recognized for its health-promoting properties, particularly its potential to mitigate obesity. However, differences in functional efficacy among distinct aroma types remain unexplored. In this study, we systematically compared the anti-obesity activities and phytochemical characteristics of three aroma-type LBTs, namely, ginseng (GA), betelnut (BA), and stale (SA), to clarify the compositional and functional distinctions. Phytochemical analysis showed that all three teas were rich in polyphenols, flavonoids, organic acids, and other compounds, with GA-type LBT containing the highest polyphenol (477.32 mg/g) and flavonoid (240.83 mg/g) contents. In vitro, all LBT extracts displayed antioxidant and anti-inflammatory activities. Among them, GA exhibited the strongest effects, scavenging over 60% of free radicals, inhibiting pancreatic lipase by 89.27%, and reducing nitric oxide production in macrophages by 67.95%. In a high-fat diet-induced obese mouse model, GA-type LBT also showed the most pronounced anti-obesity outcomes, reducing weight gain by 52.98%, alleviating hepatic steatosis and systemic inflammation, and improving serum lipid profiles. Mechanistic studies revealed that LBT extracts modulated the gut microbiota by increasing beneficial microbes and decreasing harmful ones, while also regulating lipid metabolism pathways related to glycerophospholipid, glycerolipid, and arachidonic acid. In conclusion, although all three aroma types possess anti-obesity potential, GA-type LBT stands out due to its unique bioactive composition and consistently strong efficacy both in vitro and in vivo. This study identifies GA-type LBT as a promising candidate for anti-obesity dietary supplements and provides a scientific basis for the aroma-specific selection of LBT.

Covalent modification of polyphenols effectively enhances functional properties of proteins. This study conjugated potato protein isolate (PPI) with gallic acid (GA) via an alkaline method to investigate structural and functional alterations. Successful conjugation was confirmed by a significant decrease in free amino and sulfhydryl groups, coupled with a marked increase in total phenolic content. Multispectroscopic analyses indicated a loosening of the secondary structure and notable changes in the tertiary conformation. Molecular dynamics (MD) simulations corroborated these findings, revealing that GA conjugation induced conformational expansion, improved structural stability, and enhanced surface hydrophilicity. These structural modifications led to substantial functional improvements in the PPI-GA conjugates, including enhanced dispersion stability, improved emulsifying performance, strengthened antioxidant activity, and increased thermal stability. This research may provide effective strategy and technical support for the improvement of functional properties of PPI and expand its application in the food industry.

Ethnopharmacological relevance Lonicera japonica Thunb (honeysuckle), a traditional Chinese medicine, has been historically used to clear heat, detoxify, and dispel wind-heat. Modern research has identified various bioactive constituents from honeysuckle, supporting its potential in managing inflammatory and metabolic disorders. However, the anti-hyperuricemic activity and mechanism of its peptide components remain unexplored. Aim of the study: This study aimed to investigate the anti-hyperuricemic effect of honeysuckle-derived peptides and the underlying mechanisms, with a focus on uric acid production, excretion, and gut microbiota modulation. Materials and methods Nine novel peptides were identified from honeysuckle by HPLC-MS/MS and screened through molecular docking against xanthine oxidase (XOD). In vitro XOD inhibition and antioxidant assays were performed. A hyperuricemic mouse model was induced by potassium oxonate and hypoxanthine. Mice were treated with honeysuckle peptides at low, medium, and high doses. Serum biomarkers, renal and ileal histopathology, protein and mRNA expression of PGC-1α/PPARγ/ABCG2 pathway components, and gut microbiota composition (16S rRNA sequencing) were analyzed. Results Molecular docking confirmed strong binding between the peptides and XOD. In vitro , honeysuckle peptides significantly inhibited XOD activity and exhibited potent antioxidant capacity. In vivo , peptide treatment effectively reduced serum uric acid, XOD, creatinine, and blood urea nitrogen levels, alleviated renal and intestinal tissue damage, and suppressed inflammatory cytokines. The treatment also significantly upregulated the renal PGC-1α/PPARγ/ABCG2 pathway at both protein and mRNA levels. Furthermore, the peptides restored gut microbial diversity and corrected the Firmicutes/Bacteroidota ratio. Conclusion Honeysuckle peptides ameliorate hyperuricemia through a dual mechanism: inhibiting uric acid production by suppressing XOD activity and promoting its renal excretion by activating the PGC-1α/PPARγ/ABCG2 pathway, coupled with restoring gut microbiota homeostasis. This study provides a pharmacological basis for the traditional use of honeysuckle and highlights its peptides as promising candidates for managing hyperuricemia.

To mitigate post-harvest losses in field-grown fruits and thus contribute to global efforts against malnutrition and hunger, this study presents an edible coating inspired by the phase separation of ovalbumin (OVA) and chitosan (CS) complexes, resulting in distinct inner and outer functional layers. We demonstrate that the phase behavior of the OVA-CS complexes is modulated by resveratrol (RES). Specifically, RES modifies protein conformation and intermolecular interactions by enhancing hydrophobic forces and reorganizing hydrogen bonds, driving the self-assembly of a Janus-type coating during drying. Optimized at pH 5.6, the OVA-CS-RES (OCR) coating exhibits superior barrier properties, achieving the lowest water vapor permeability and oxygen permeability. Anisotropic wettability was confirmed by contact angle measurements and atomic force microscopy, with the maximum contact angle difference between the inner and outer surfaces reaching 57.2°. Scanning electron microscopy and confocal laser scanning microscopy further verified uniform coverage and effective sealing of epidermal pores on fruit surfaces. Application experiments showed that the OCR coating significantly delayed fruit oxidation, reduced weight loss, preserved texture and ascorbic acid content, and maintained aroma integrity throughout 40 days of storage. Overall, this study introduces a scalable, single-step coating strategy leveraging self-assembled functional Janus coatings to significantly extend fruit shelf life.

Diabetic foot ulcers (DFUs), prone to infection and deterioration without timely intervention, significantly increase risks of amputation and mortality. Elevated local temperature in DFUs further impedes the healing process. Although microneedles (MNs) represent a promising strategy for DFUs treatment, conventional passive drug release systems suffer from slow release kinetics and low utilization efficiency. To address these limitations, we developed a dual-thermo-responsive microneedle patch (DTMN) that actively controls drug delivery through temperature-induced structural transitions. The inner layer consists of sodium alginate-poly (N-isopropylacrylamide) (SA-PNIPAM) loaded with sucrose octasulfate sodium salt (SOS), utilizing the volume phase transition of PNIPAM to accelerate drug expulsion in response to temperature change. The outer layer comprises a polyethylene glycol/polylactic acid-glycolic acid copolymer (PEG-PLGA) loaded with urea, which undergoes gel-sol transition to facilitate controlled urea release and wound cooling. An upper electrospun nanofiber membrane made of poly (ε-caprolactone)/chitosan (PCL/CS) incorporated with tetracycline hydrochloride (TH) and SOS provides enhanced antibacterial efficacy and increased drug loading capacity. The resulting DTMN exhibits efficient drug release (85.23% SOS and 35.44% urea-derived ammonia at 24 h), remarkable antioxidant activities, potent antibacterial performance, excellent biocompatibility, and significantly enhanced wound healing. This multifunctional system offers a novel and effective strategy for the management of DFUs. Graphical A dual-thermo-responsive microneedle patch (DTMN) that actively controls drug delivery through temperature-induced structural transitions for DFUs treatment.

Abdominal aortic aneurysm (AAA) is a cardiovascular disease with extremely high mortality rate, yet its treatment options are still limited in the clinic. This study presents a carrier-free dual-drug nanoplatform to therapeutically reprogram macrophage fate for improving AAA treatment efficacy. The nanoplatform was synthesized through the self-assembly of naringenin (NGN) and rapamycin (RAP) and modified with DSPE-PEG 2K and DSPE-PEG 2K -cRGD (named RNR NPs) to confer AAA targeting capability. The RNR NPs could efficiently accumulate in AAA site after systemic administration for on-demand delivery of the NGN/RAP drug combination to vascular smooth muscle cells (VSMCs) and residing M1-like macrophages. Here the NGN and RAP components could cooperatively activate the transcription factor EB (TFEB) signaling in the AAA-residing pro-inflammatory M1-like macrophages to reprogram them into an anti-inflammatory M2-like phenotype to reverse local inflammation. Meanwhile, they could also scavenge reactive oxygen species in VSMCs and inhibit apoptosis signaling to inhibit VSMCs apoptosis and calcification. These effects could act in a cooperative manner for effectively ameliorating AAA symptoms in vivo . Overall, the RNR NPs present an approach for the efficient treatment of AAA in the clinic with optimal safety.

Surgical suture techniques are commonly used for hemostasis and wound closure, but they often fail to achieve optimal results, particularly in post-laceration hemostasis and healing. Furthermore, complications such as bacterial infections, oxidative stress, and inflammation often worsen the initial wound, leading to secondary damage. In this study, a novel tissue adhesive was developed by integrating a natural epigallocatechin-3-gallate (EGCG) and zirconium ion (Zr) chelate complex (EGCG-Zr) into a gelatin/starch hydrogel (GDSH). The resulting multifunctional tissue adhesive (EGCG-Zr@GDSH) exhibits excellent injectability, superior adhesion strength compared with commercial protein fiber-based tissue adhesives, and cytocompatibility. The hydrogel enables the sustained release of EGCG and Zr, providing effective antibacterial effects. In vivo histological analysis of an incision wound model revealed that EGCG-Zr@GDSH promotes angiogenesis and collagen deposition, scavenges reactive oxygen species, restores intracellular redox balance, and mitigates oxidative stress-induced cellular damage. Moreover, it regulates M1 macrophage polarization, thereby reducing the secretion of proinflammatory cytokines and alleviating inflammation. This hydrogel possesses antioxidant and immunomodulatory properties, indicating its application potential as a tissue adhesive and highlighting the importance of immune regulation in tissue repair and regeneration.

Crystalline nephropathy is a dynamic pathological process characterized by a self-amplifying feedback loop, where crystal-induced epithelial injury drives iterative crystal nucleation, growth, and aggregation, transforming the kidney into a persistent mineralization reactor. Current therapeutic approaches primarily rely on surgical intervention, which fails to disrupt this pathological cycle. The glomerular filtration barrier (10 nm threshold) limits the efficacy of conventional nanomedicines in renal delivery. This study constructs a 5.7 nm metal-polyphenol network, salvianolic acid B-Eu@BSA (SAB-Eu@BSA), via self-assembly methods for editing of the intrarenal crystal microenvironment, aiming to cut off the pathological cycle of renal mineralization. The SAB-Eu@BSA nanoplatform integrates a SAB Eu 3+ coordination core for dual-action: specific adsorption to the high-energy ( 1 ¯ 01) crystal faces of monohydrate calcium oxalate (COM) to inhibit its growth and promote phase transition to less toxic dihydrate phase, and broad-spectrum antioxidant activity to mitigate oxidative stress. The nanoplatform demonstrates enhanced crystal modulation efficacy, significantly reduces crystal-induced renal injury, and improves renal function in vivo. RNA sequencing analysis reveals modulation of key pathways, including oxidative stress, cell adhesion, and apoptosis. This multifunctional nanoplatform, combining crystal inhibition, oxidative stress regulation, and renal clearance, offers a promising therapeutic strategy for crystalline nephropathy.

Improving the stability of catechins against autoxidation and simultaneously exerting their biological activity to scavenge reactive oxygen species (ROS) are essential for achieving their efficient utilization in healthcare and drug therapy. Focusing on this important objective, a feasible strategy for modifying catechins with arylboronic acid has been proposed as they have good biocompatibility and also controllable release at specific sites. We developed three catechin-diarylboronate (DiarylBA) ester self-assembly carriers, in which DiarylBA serves as the receptor molecules of catechins, including a coupled benzyl ammonium boronic acid (BTEAB) or two coupled pyridinium boronic acids (PyBBA and PyPBA) linked by a spacer, and (−)-epicatechin (EC) is taken as the representative catechin molecule. Specifically, the inhibition of the auto-oxidation and the antioxidant activities for both EC and EC-DiarylBA esters were characterized sufficiently by combining spectroscopies (UV–vis, NMR), and ROS scavenging assessment. The relevant binding thermodynamics and interaction mechanism, as well as their aggregation behavior, were also studied by ITC, NMR, and light scattering. The thermodynamic and spectroscopic results all confirm the generation of EC-DiarylBA esters in a range from a weak acid of pH 6.5 to an alkaline one of pH 8.5. It was found that the esterification of EC with DiarylBA can effectively inhibit the auto-oxidation of EC even at pH 8.5 and still retain the excellent antioxidant activity of the original EC. Furthermore, the EC-BTEAB ester exhibits a favorable H2O2 responsiveness in the H2O2-overexpressed site, while EC-PyBBA and EC-PyPBA esters need to first release the EC component to effectively eliminate H2O2. Therefore, the comprehensive results of stability and antioxidant activity indicate that the EC-DiarylBA ester would be a promising candidate for enhancing EC’s medicinal and health care effects.

This study developed a green extraction paradigm for rosemary absolute oil (RAO) as a sustainable natural antioxidant to inhibit lipid oxidation in frying oils. RAO was prepared via three methods: heat reflux extraction (HRE) with n-hexane, solvent-free oil extraction (OE) using refined soybean oil, and ultrasound-assisted solvent-free oil extraction (UOE). Under optimized UOE conditions (solid-to-liquid ratio 1:10, 400 W, 25°C, 13 min), RAO yield reached 19.09%, with total phenolic content (TPC) of 347.16 ± 2.81 mg GAE/g, significantly exceeding values from HRE and OE. UOE-RAO contained elevated concentrations of bioactive compounds: carnosic acid (CA, 89.45 ± 1.89 mg/g), ursolic acid (UA, 7.43 ± 0.02 mg/g), and carnosol (16.81 mg/g), accompanied by a pronounced CA-to-carnosol conversion. In vitro antioxidant assays (DPPH) demonstrated UOE-RAO’s potent radical scavenging activity (IC 50 : 71.26 ± 0.81 μg/mL), on par with synthetic butylated hydroxytoluene (BHT). When fortifying soybean oil with 0.1% UOE-RAO, oxidative stability during 180°C frying was markedly enhanced: total polar materials (TPM) accumulation was retarded, extending oil usability from 31 h (control) to 63 h, while acid value (AV) remained consistently lower throughout frying. Differential scanning calorimetry (DSC) further validated improved oxidative resistance, with induction times for primary (12.75 ± 3.51 → 14.14 ± 0.20 min) and secondary oxidation (40.47 ± 6.02 → 61.41 ± 3.10 min) significantly prolonged. Collectively, these findings highlight that UOE enables the production of RAO with superior antioxidant efficacy, positioning it as a green, high-performance alternative to synthetic antioxidants for lipid-rich food processing, particularly high-temperature frying applications.

The development of eco-friendly and multifunctional edible coatings is crucial for mitigating postharvest losses of perishable fruits. Hence, a novel composite coating was engineered based on chitosan (CS) and pullulan (PUL) matrix, enhanced by a gallic acid-grafted chitosan (GS) conjugate synthesized through free radical grafting. The GS conjugate served as a multifunctional crosslinker, establishing a dense intermolecular network within the CS-PUL matrix primarily (CP) through hydrogen bonding, as confirmed by structural characterization. This molecular-level interaction significantly improved the mechanical strength (Tensile stress = 4.95 MPa, Elongation at break =121.21%) and barrier properties of the resulting coating. The chitosan/pullulan-gallic acid grafted chitosan (CP-GS) coating, applicable by spraying or dipping, exhibited remarkable antioxidant activity, achieving 97.8% DPPH radical scavenging (25 °C, 50% RH), the CP-GS coating significantly reduced weight loss, delayed the decline in titratable acidity, and effectively suppressed microbial growth, thereby extending the shelf life by 7 to 9 days compared to the uncoated samples. This study highlights the effectiveness of molecular grafting in designing advanced bio-based coatings and presents a promising strategy for fruit preservation using biological macromolecules.
