Graphene-Based Steganographically Aptasensing System for Information Computing, Encryption and Hiding, Fluorescence Sensing and in Vivo Imaging of Fish Pathogens

Qiu Yan Zhu,Fu Rui Zhang,Yan Du,Xin Xing Zhang,Jiao Yang Lu,Qing Feng Yao,Wei Tao Huang,Xue Zhi Ding,Li Qiu Xia

Inspired by information processing and communication of life based on complex molecular interactions, some artificial (bio)chemical systems have been developed for applications in molecular information processing or chemo/biosensing and imaging. However, little attention has been paid to simultaneously and comprehensively utilize the information computing, encoding, and molecular recognition capabilities of molecular-level systems (such as DNA-based systems) for multifunctional applications. Herein, a graphene-based steganographically aptasensing system was constructed for multifunctional application, which relies on specific molecular recognition and information encoding abilities of DNA aptamers (Aeromonas hydrophila and Edwardsiella tarda-binding aptamers as models) and the selective adsorption and fluorescence quenching capacities of graphene oxide (GO). Although graphene–DNA systems have been widely used in biosensors and diagnostics, our proposed graphene-based aptasensing system can not only be utilized for fluorescence sensing and in vivo imaging of fish pathogens (A. hydrophila and E. tarda), but can also function as a molecular-level logic computing system where the combination of matters (specific molecules or materials) as inputs produces the resulting product (matter level) or fluorescence (energy level) changes as two outputs. More importantly and interestingly, our graphene-based steganographically aptasensing system can also serve as a generally doubly cryptographic and steganographic system for sending different secret messages by using pathogen-binding DNA aptamers as information carriers, GO as a cover, and a pair of keys, that is, target pathogen as a public key, the encryption key used to encode or decode a message in DNA as a private key. Our study not only provides a novel nanobiosensing assay for rapid and effective sensing and in vivo imaging of fish pathogens, but also demonstrates a prototype of (bio)molecular steganography as an important and interesting extension direction of molecular information technology, which is helpful in probably promoting the development of multifunctional molecular-level devices or machines.

Effect of non-covalent binding of tannins to sodium caseinate on the stability of high-internal-phase fish oil emulsions

Yanchen Wang,Yikai Huang,Ying Sun,Mantong Zhao,Zhongyuan Liu,Haohao Shi,Xueying Zhang,Yongqiang Zhao,Guanghua Xia,Xuanri Shen

In this study, we designed the noncovalent binding of sodium caseinate (SC) to tannic acid (TA) to stabilize high internal phase emulsions (HIPEs) used as fish oil delivery systems. Hydrogen bonding was the dominant binding force, followed by weak hydrophobic interaction and weak van der Waals forces, as demonstrated by FTIR, fluorescence spectroscopy, and molecular docking experiments, with a binding constant of 3.25 × 10 6 , a binding site of 1.2, and a static quenching of the binding. Increasing SC:TA from SC to 2:1 decreased the particle size from 107.37 ± 10.66 to 76.07 ± 2.77 nm and the zeta potential from −6.99 ± 2.71 to −22 ± 2.42 mV. TA increased the interfacial tension of SC, decreased the surface hydrophobicity from 1.3 × 10 4 to 1.6 × 10 3 and improved the oxidation resistance of SC. The particle size of high internal phase emulsions stabilized by complexes with different mass ratios (SC:TA from 1:0 to 2:1) increased from 4.9 ± 0.02 to 12.9 μm, the potential increased from −32.37 ± 2.7 to −35.07 ± 2.58 mV, and the instability index decreased from 0.75 to 0.02. Thicker interfacial layers could be observed by laser confocal microscopy, and an increase in the storage modulus indicated a formation of a stronger gel network. SC:TA of 1:0 showed emulsion breakage after 14 d of storage at room temperature. SC:TA of 2:1 showed the lowest degree of oil-water separation after freeze-thaw treatment. Especially, the most stable high endo -phase emulsion (at SC:TA of 2:1) prepared at each mass ratio was selected for further stability exploration. The emulsion particle size increased only from 15.63 ± 0.06 to 22.27 ± 0.35 μm at salt ion concentrations of 50–200 mM and to 249.33 ± 31.79 μm at 300 mM. The instability index and storage modulus of the high endo -phase emulsions increased gradually with increasing salt ion concentrations. At different heating temperatures (55–85 °C), the instability index of the high internal phase emulsion gradually decreased and the storage modulus gradually increased. Meanwhile, at 50 °C for 15 d of accelerated oxidation, the content of hydroperoxide decreased from 53.32 ± 0.18 to 37.48 ± 0.77 nmol/g, about 29.7 %, and the thiobarbituric acid value decreased from 1.06 × 10 3 to 0.8 × 10 3 , about 24.5 %, in the high endo -phase emulsions prepared by 2:1 SC:TA compared to the fish oils, and the SC-stabilized high endo-phase only emulsion broke at the sixth day of oxidation. From the above findings, it was concluded that the high internal phase emulsion prepared with SC:TA of 2:1 can be used as a good delivery system for fish oil.

Hyaluronic acid-poly(glyceryl)10-stearate nanoemulsion for co-delivery of fish oil and resveratrol: Enhancing bioaccessibility and antioxidant potency

Sha Ao,Xiang Luo,Mengyu Huang,Hongze Wu,Yuanyuan Chen,Haonan Chen,Jiafei Li,Yanyan Zhou,Xuguang Yin,Tao Cai,Qun Yang,Liping Deng,Kewu Zhu

Hyaluronic acid (HA), an endogenous polysaccharide comprising alternating D -glucuronic acid and N -acetylglucosamine units, is renowned for its high hydrophilicity, biocompatibility , and biodegradability. These attributes have rendered HA invaluable across medical and drug delivery fields. HA can be altered through physical, chemical, or enzymatic methods to improve the properties of the modified substances. In this work, we synthesized a derivative via the esterification of HA with poly(glyceryl)10-stearate (PG10-C18), designated as HA-PG10-C18. This novel derivative was employed to fabricate a nano co-delivery system (HA-PG10-C18@Res-NE) for fish oil and resveratrol (Res), aiming to enhance their stability and bioaccessibility. An exhaustive investigation of HA-PG10-C18@Res-NE revealed that the HA-modified system displayed superior physicochemical stability, notably in withstanding oxidation and neutralizing free radicals . Moreover, in vitro simulated digestion underscored the system's enhanced bioaccessibility of Res and more efficient release of free fatty acids . These outcomes underscore the strategic advantage of HA in modifying PG10-C18 for nanoemulsion formulation. Consequently, HA-PG10-C18 stands as a promising emulsifier for encapsulating lipophilic bioactives in functional foods, nutraceuticals , and pharmaceuticals.

Impact of Co-60 γ-ray irradiation on the cross-linking and stability of fish collagen: Structural changes and digestibility

Junwei Qin,Litong Liu,Chengdu Miao,Bifeng Lan,Tao Liao,Xiaofei Tian,Zhenqiang Wu

Irradiation can promote the cross-linking of collagen to enhance its biological properties, but the molecular changes in irradiated collagen have few been investigated. This study aimed to explore the impact of irradiation treatment on the molecular structure stabilization and cross-linking degree of skin collagen. The results showed that collagen in solution was more susceptible to Co-60 gamma irradiation than that in fish skin and in sponge forms. The solubility of collagen in solution decreased by more than 80% and the carbonyl content increased by more than 4 times compared with the control group after 12 kGy irradiation. Spectroscopic analysis and sodium dodecyl sulfate - polyacrylamide gel electrophoresis (SDS-PAGE) showed that collagen in solution was cross-linked, especially at high irradiation doses. However, the triple helical structure of skin collagen and collagen sponge remained intact. Furthermore, irradiation also reduced the sensitivity of collagen to collagenase. Gastrointestinal digestion results showed that the antioxidant activity of collagen increased at 1–3 kGy doses and decreased at 9–12 kGy doses. Collagen irradiated at high doses (9–12 kGy) was more difficult to digest and had a larger average particle size. This study provides a potential new approach for the preparation of stable fishery-derived collagen with excellent antioxidant activity and resistance to digestion by irradiation treatment.

Impacts of live and artificial feed on histology, biochemical indicators, gene expression, and bacterial resistance in mandarin fish (Siniperca chuatsi)

Shunli Che,Jiawei Chen,Hai Zhang,Weizhen Xu,Yanwei Li,Xueming Dan,Zequan Mo

The mandarin fish ( Siniperca chuatsi ) is a significant freshwater carnivorous species in Chinese aquaculture industry, and its farming scale is continuously expanding. The use of feed in aquaculture has become an increasingly common practice. However, the impacts of substituting artificial feed for live bait on fish's biochemical and immune responses are poorly understood. In this study, two hundred forty mandarin fish (weight: 5.60 ± 0.41 g) were divided into two groups and fed live bait or artificial feed (LB and AF groups) over a 63-day aquaculture experiment. We compared the differences between the two groups in terms of histology, biochemical indicators, gene expression, and bacterial resistance. The results showed that artificial feed promoted enhanced growth, evidenced by higher weight ( p  < 0.05). The AF group exhibited higher liver and intestinal somatic indices ( p  < 0.05), and histological examination revealed denser cytoplasmic content in liver cells, less fragmentation of renal tubular epithelial cells, and less detachment of intestinal epithelial cells in the AF group. Regarding biochemical indicators and gene expression, the AF group showed better performance in glucose regulation and lipid metabolism. The AF group maintained glucose balance ( p  < 0.05) and effectively regulated cholesterol transport ( p  < 0.05), promoting lipolysis ( p  < 0.05) while inhibiting lipogenesis ( p  < 0.05). In contrast, live bait consumption resulted in reduced lipolysis ( p  < 0.05), increased lipogenesis ( p  < 0.05), impaired endoplasmic reticulum function ( p  < 0.05), heightened inflammation ( p  < 0.05), and diminished antioxidant capacity ( p  < 0.05). Additionally, the LB group exhibited lower survival rates and lysozyme levels during bacterial challenges. Overall, artificial feed was more beneficial for the growth, regulate physiology and enhance disease resistance of S. chuatsi , highlighting its potential to improve fish health and increase aquaculture yield.

Multifunctional fish-skin collagen-based hydrogel sealant with dual-dynamic-bond cross-linked for rapid hemostasis and accelerated wound healing

Pei-Fang Cai,Bing-De Zheng,Ya-Ling Xu,Bao-Xia Li,Zhi-Yu Liu,Ya-Yan Huang,Jing Ye,Mei-Tian Xiao

Collagen (COL) is the most widespread functional protein. Designing and developing dual-dynamic-bond cross-linked COL adhesive hydrogel sealants with multifunctional is highly advantageous for achieving a superior wound closure effect and hemostasis. In this study, we developed hybrid hydrogels consisting of fish-skin COL, oxidized sodium alginate (OSA), borax and polyvinyl alcohol (PVA) to enhance full-thickness wound healing. The hydrogels were furnished with first-rate self-healing capabilities through the dual-dynamic-bond cross-linking of dynamic Schiff base bonds (COL-OSA) and diol boric acid bonds (OSA-borax) with reversible breakage and re-formation. Moreover, the incorporation of PVA stimulated the formation of hydrogen bonds in the system, bolstering the stability of the hydrogel framework. The prepared hydrogel manifests self-healing, injectability, multifunctional adhesiveness and biodegradability. In vivo assessment of the hemostatic capacity of COSP 20 hydrogel was superior to gauze both in the mice liver injury model and mice tail amputation model. In addition, a full-thickness skin wound model in mice revealed that the COSP 20 hydrogel facilitated faster wound closure by accelerating reepithelialization, COL deposition and angiogenesis. These findings illustrate the potential of hybrid fish-skin COL-based hydrogels to enhance wound healing and promote rapid tissue repair, and provide new possibilities for the effective utilization of marine fishery resources.

Room-temperature synthesis of yellow-emitting fluorescent silicon nanoparticles for sensitive and selective determination of crystal violet in fish tissues

Yangxia Han,Yonglei Chen,Juanjuan Liu,Xiaoying Niu,Yunxia Ma,Sudai Ma,Xingguo Chen

Silicon nanoparticles (SiNPs) have attracted increasing attentions due to their unique properties and related applications. However, previously reported SiNPs generally showed emission band only in the blue- or green-light regions, which limited their applications in biochemical samples. So far, synthesis of longer wavelength emissive SiNPs remains a challenge. In this paper, the yellow-emitting fluorescent SiNPs were prepared via a facile room temperature procedure, which avoided heating and pressurizing operation, multistep and time-consuming post-modified procedures. The obtained SiNPs displayed excellent water-solubility, pH-stability, salt-tolerance, and photo-stability. Significantly, the fluorescence of SiNPs could be remarkably quenched by crystal violet (CV) via synergistic effect of inner filter effect (IFE) and fluorescence resonance energy transfer (FRET). Based on this phenomenon, a method for the determination of CV was established. Under the optimum conditions, the method showed a wide linearity range from 0.075 to 15.0 μg/mL with a limit of detection of 25 ng/mL. Furthermore, the method was successfully applied to the analysis of CV in fish tissues samples.

An ammonia-sensitive fluorescence sensor based on polyvinyl alcohol-graphene quantum dots/halloysite nanotubes hybrid film for monitoring fish freshness

Jiaran Zhang,Jie Yu,Hao Yin,Zhixin Jia,Ce Shi,Yuntao Yue

A fluorescent hybrid film composed of nitrogen-doped graphene quantum dots (N-GQDs) loaded on halloysite nanotubes (HNTs) (N-GQDs/HNTs nanocomposite) as a sensitive element and polyvinyl alcohol (PVA) as a film-forming matrix was designed for freshness detection. The PVA-N-GQDs/HNTs hybrid film exhibited significantly enhanced fluorescence attributed to the loading of N-GQDs onto the surface of HNTs through electrostatic interactions and hydrogen bonding , effectively reducing their aggregation. The fluorescence of the hybrid film could be quenched by ammonia via photoinduced electron transfer (PET), with good linearity in the range of 20 ppm to 500 ppm ammonia and a limit of detection (LOD) of 0.63 ppm. In addition, the hybrid film was applied to monitor the freshness of seawater fish and freshwater fish stored at refrigeration and room temperature to evaluate the practicality of this approach. The developed hybrid film showed promise for nondestructive and on-site monitoring of fish spoilage.

Chunchun Wang,Jiayi Guo,Qun Liu,Xu Zeng,Yue Liu,Yanping Deng,Yanli Lin,Xialing Wu,Hongju Deng,Linjing Chen,Wuyin Weng,Yucang Zhang

Accidental bleeding is an unavoidable problem in daily life. To avoid the risk of excessive blood loss, it is urgent to design a functional material that can quickly stop bleeding. In this study, an efficient wound dressing for hemostasis was investigated. Based on the characteristics that Ca 2+ and fish skin collagen (FSC) could activate the coagulation mechanism, hemostatic cotton was prepared by solvent replacement method using CaCl 2 , FSC, soluble starch (SS), and polyvinyl alcohol (PVA) as raw materials. The cytotoxicity test showed the Ca 2+ PVA/FSC-SS hemostatic cottons had good biocompatibility. The activated partial thromboplastin time (APTT) of Ca 2+ PVA/FSC-SS(4) was 35.34 s, which was 22.07 s faster than that of PVA/FSC-SS, indicating Ca 2+ PVA/FSC-SS mediated the endogenous coagulation system. In vitro coagulation test, Ca 2+ PVA/FSC-SS(4) could stop bleeding rapidly within 39.60 ± 5.16 s, and the ability of wound healing was higher than commercial product (Celox). This study developed a rapid procoagulant and hemostatic material, which had a promising application in a variety of environments.

Locust bean gum improved the gelling properties of fish gelatin in acidic environments and its application in acid gel gummies

Wu Fen-Fen,Tu Zong-Cai,Cheng Jie,Wang Hui,Hu Zi-Zi,Sha Xiao-Mei

To address the “gel weakening” of fish gelatin (FG) under acidic conditions, this study investigates the enhancement of gel performance in FG through the incorporation of locust bean gum (LBG) and explores its feasibility for application in acidic gel systems. Results show that the gel properties of FG were significantly improved through LBG modification. At pH 3−7, gel strength and melting temperature of LBG-modified FG (FG-LBG) surpassed those of unmodified FG (524.68 ± 1.54 g, 28.86 ± 0.03 °C). Specifically, at pH 3, after LBG modification, the gel strength and melting temperature of FG increased by 62.65% and 29.44%, respectively. LBG alters the conformation of FG via electrostatic and hydrogen bonding interactions, forming a denser polymer network. The acidic gel gummies prepared from the FG modified with LBG exhibit superior gel properties and texture characteristics. This study provides a basis for the modification of FG under acidic conditions.

Wireless portable intelligent nanosensor for pretreatment-free, anti-interference, and real-time on-site detection of furazolidone in fish products

Danting Yan,Boyu Zhu,Jingjing Wang,Mengke Wu,Fang Xie,Peipei Qi,Hongping Gong,Xingyu Lin

Furazolidone (FZD) is a banned antibiotic in food-producing animals due to its carcinogenicity and toxicity. Its illegal use and residue persistence in foods remains a health risk, highlighting the need for a rapid and reliable detection. Electrochemical sensing offers rapidity and sensitivity; however, its use is limited by bulky instrumentation and low accuracy in complex food matrices, making on-site testing difficult. To overcome these limitations, this work developed a wireless portable intelligent nanosensor for direct, pretreatment-free detection of FZD in food. The nanosensor features a mesoporous silicon film microelectrode (MSFM) with nanopores, which significantly enhances selectivity through size exclusion, electrostatic repulsion, hydrophilic interaction, and nanoscale confinement effects. Integrated with a smartphone via Bluetooth, the nanosensor allows immediate visualization of detection results. Under optimized conditions, the nanosensor can directly detect trace amounts of FZD in fish samples in real time without the need for enrichment or preprocessing. It exhibited a wide linear range from 0.1 to 100.0 μmol/L and a detection limit of 0.05034 μmol/L, with excellent anti-fouling ability, reproducibility (RSD 4.50 %), and 15-day signal stability. This innovative wireless sensing technology holds great promise for extending laboratory-based detection techniques to on-site and market environments, particularly in resource-constrained areas.
