Co-delivery of doxorubicin and paclitaxel by PEG-polypeptide nanovehicle for the treatment of non-small cell lung cancer

Shixian Lv,Zhaohui Tang,Mingqiang Li,Jian Lin,Wantong Song,Huaiyu Liu,Yubin Huang,Yuanyuan Zhang,Xuesi Chen

Despite progress, combination therapy of different functional drugs to increase the efficiency of anticancer treatment still remains challenges. An amphiphilic methoxy poly(ethylene glycol)- b -poly( l -glutamic acid)- b -poly( l -lysine) triblock copolymer decorated with deoxycholate (mPEsG- b -PLG- b -PLL/DOCA) was synthesized and developed as a nanovehicle for the co-delivery of anticancer drugs: doxorubicin (DOX) and paclitaxel (PTX). The amphiphilic copolymer spontaneously self-assembled into micellar-type nanoparticles in aqueous solutions and the blank nanoparticles possessed excellent stability. Three different domains of the copolymer performed distinct functions: PEG outer corona provided prolonged circulation, middle biodegradable and hydrophilic PLG shell was designed for DOX loading through electrostatic interactions , and hydrophobic deoxycholate modified PLL served as the container for PTX. In vitro cytotoxicity assays against A549 human lung adenocarcinoma cell line demonstrated that the DOX + PTX co-delivered nanoparticles (Co-NPs) exhibited synergistic effect in inducing cancer cell apoptosis. Ex vivo DOX fluorescence imaging revealed that Co-NPs had highly efficient targeting and accumulation at the implanted site of A549 xenograft tumor in vivo . Co-NPs exhibited significantly higher antitumor efficiency in reducing tumor size compared to free drug combination or single drug-loaded nanoparticles, while no obvious side effects were observed during the treatment, indicating this co-delivery system with different functional antitumor drugs provides the clinical potential in cancer therapy.

DOX-loaded pH-sensitive mesoporous silica nanoparticles coated with PDA and PEG induce pro-death autophagy in breast cancer

Yanhong Duo,Yang Li,Changke Chen,Baiyun Liu,Xinyu Wang,Xiaowei Zeng,Hongbo Chen

The development of multifunctional nano drug delivery carriers has been one of the most effective and prevailing approaches to overcome drug non-selectivity, low cell uptake efficiency and various side effects of traditional chemotherapy drugs. Herein, we report a novel doxorubicin (DOX)-loaded mesoporous silica nanoparticle (MSN) coated with polydopamine (PDA) and polyethylene glycol (PEG) (MSNs-DOX@PDA-PEG) for the treatment of breast cancer. In this system, PDA functions as a pH-sensitive gatekeeper to control the release of DOX from MSNs in response to pH-stimulus and PEG was further grafted on the surface of PDA to increase the stability and biocompatibility under physiological conditions. The in vitro release results suggested that MSNs-DOX@PDA-PEG exhibits a high sensitivity to low pH. A cellular uptake assay showed a high cellular uptake efficiency of MSNs-DOX@PDA-PEG compared to free DOX. Furthermore, MSNs-DOX@PDA-PEG also demonstrated an improved anti-cancer efficacy compared to free DOX both in vivo and vitro breast cancer experiments. Mechanistic studies revealed that MSNs-DOX@PDA-PEG causes a stronger pro-death autophagy compared to free DOX via inhibition of the AKT-mTOR-p70S6K signaling pathway. Taken in concert, our results suggest that the novel material MSNs-DOX@PDA-PEG may represent a promising nanoformulation for breast cancer treatment.

Synergistically enhanced NF/CNTs@ PDA/PEG composite phase change materials for high-efficiency solar-thermal energy storage and conversion

Youchong Hu,Yaoqi Huang,Xiaopeng Liu,Meng Fu,Shuai Luo,Yuanyuan Li,Xiaomin Cheng

In order to solve the low thermal conductivity and leakage challenges of phase change materials (PCMs), a high-performance composite phase change material (CPCM) by integrating nickel foam (NF), carbon nanotubes (CNTs), and polydopamine (PDA) is developed. A three-dimensional (3D) porous NF/CNTs@PDA framework was constructed through oxidative self-polymerization of PDA on NF surfaces, followed by ultrasonic-assisted CNTs impregnation. CNTs-NF interfacial bonding was strengthened and interfacial thermal resistance was reduced by the PDA coating, as demonstrated in experimental results, leading to enhanced thermal conductivity. Light absorption and heat conduction were further optimized by CNTs. The optimized CPNF-9/PEG composite exhibited a thermal conductivity of 0.899 W m −1  K −1 , representing an 565.93 % enhancement over pure PEG (0.135 W m −1  K −1 ). The melting and crystallization enthalpies of the material were 121.13 J g −1 and 113.44 J g −1 respectively, and solar-thermal tests indicated a 95.37 % conversion efficiency. After 200 thermal cycles, the composite maintained stable phase change latent heat (loss rate <3 %) and structural integrity, indicating excellent cyclic durability. Furthermore, leakage resistance tests at 80 °C revealed minimal mass loss (2 % over 100 min), highlighting its suitability for high-temperature applications. The work provides a novel strategy for designing efficient solar-thermal energy storage systems, with potential applications in solar-thermal management and renewable energy conversion technologies.

“Corn-like” APT@UiO-66-NH₂/PEG composite membrane with excellent anti-fouling performance for oil/water emulsions separation

Ruirui Jin,Jingling Gong,Zhaoyang Li,Bin Xiang,Weimin Liu,Jian Li

The increase in oily wastewater exacerbates global water scarcity, triggering ecological damage and health risks. Although membrane separation technology was widely adopted in oily wastewater treatment due to its high efficiency and energy savings, membrane fouling issues lead to flux decline, increased energy consumption, and reduced membrane lifespan, thereby limiting its application. Therefore, this study proposed an advanced “water absorption-storage” strategy by in-situ growing UiO-66-NH₂ nanoparticles on attapulgite (APT) through carefully designed micro-nano-scale rough structures and chemical interactions, forming a unique biomimetic “corn-like” structure. The APT@UiO-66-NH₂/PEG composite membrane prepared in this study establishes a robust hydration barrier through synergistic interactions between its components, thereby exhibiting both outstanding anti-oil fouling capability and highly efficient, stable oil-water emulsion separation performance. Meanwhile, the in-situ growth of UiO-66-NH₂ on APT effectively resolved the issue of easy detachment between APT and UiO-66-NH₂. Moreover, the abundant hydrophilic functional groups (–OH) in polyethylene glycol (PEG) form hydrogen bonds with UiO-66-NH₂ and APT, further enhancing the structural stability. The obtained APT@UiO-66-NH₂/PEG composite membrane achieved separation efficiencies exceeding 99.8 % for various oil-in-water emulsions, with a separation flux reaching 3020 LMH/bar. After 10 separation cycles and 60 min of continuous operation, the separation performance remained essentially stable. The composite membrane demonstrated excellent chemical durability and mechanical stability in high-concentration acid, alkali, and salt solutions, as well as during bending and abrasion tests. Therefore, this study presents a simple and effective purification solution for practical oily wastewater, with the associated strategies and preparation techniques demonstrating broad application prospects.

Formulation-Driven Optimization of PEG-Lipid Content in Lipid Nanoparticles for Enhanced mRNA Delivery In Vitro and In Vivo

Wei Liu,Meihui Zhang,Huiyuan Lv,Chuanxu Yang

Background : Lipid nanoparticles (LNPs) represent one of the most effective non-viral vectors for nucleic acid delivery and have demonstrated clinical success in siRNA therapies and mRNA vaccines. While considerable research has focused on optimizing ionizable lipids and helper lipids, the impact of PEGylated lipid content on LNP-mediated mRNA delivery, especially in terms of in vitro transfection efficiency and in vivo performance, remains insufficiently understood. Methods : In this study, LNPs were formulated using a self-synthesized ionizable lipid and varying molar ratios of DMG-PEG2000. Nanoparticles were prepared via nanoprecipitation, and their physicochemical properties, mRNA encapsulation efficiency, cellular uptake, and transfection efficiency were evaluated in HeLa and DC2.4 cells. In vivo delivery efficiency and organ distribution were assessed in mice following intravenous administration. Results : The PEGylated lipid content exerted a significant influence on both the in vitro and in vivo performance of LNPs. A bell-shaped relationship between PEG content and transfection efficiency was observed: 1.5% DMG-PEG2000 yielded optimal mRNA transfection in vitro, while 5% DMG-PEG2000 resulted in the highest transgene expression in vivo. This discrepancy in optimal PEG content may be attributed to the trade-off between cellular uptake and systemic circulation: lower PEG levels enhance cellular internalization, whereas higher PEG levels improve stability and in vivo bioavailability at the expense of cellular entry. Furthermore, varying the PEG-lipid content enabled the partial modulation of organ distribution, offering a formulation-based strategy to influence biodistribution without altering the ionizable lipid structure. Conclusions : This study highlights the critical role of PEGylated lipid content in balancing nanoparticle stability, cellular uptake, and in vivo delivery performance. Our findings provide valuable mechanistic insights and suggest a straightforward formulation-based strategy to optimize LNP/mRNA systems for therapeutic applications.

Electrocatalytic Self-Sterilizing PEG@Mn-Ti3C2Tx MXene Biosensor with Dual Antifouling Capability for Ultrasensitive Detection of Cardiac Troponin I

Meiling Lian,Haoquan Wang,Liuxing Chen,Da Chen,Qi Yang,Xuezhi Qiao

Electrochemical sensing devices often suffer from biofouling due to nonspecific adsorption of biomolecules and microorganisms in complex biological matrices. To address this challenge, we developed an efficient antifouling and self-sterilizing sensing interface by grafting polyethylene glycol (PEG) onto two-dimensional Ti 3 C 2 T x MXene nanosheets for the accurate detection of cardiac troponin I (cTnI), a gold-standard biomarker for acute myocardial infarction (AMI). The Mn 2+ -intercalated Ti 3 C 2 T x (Mn-Ti 3 C 2 T x ) exhibited enhanced electrochemical activity in a three-electron oxygen reduction reaction (3e - ORR), enabling in situ self-sterilization by activating molecular oxygen to generate hydroxyl radicals (•OH), which demonstrated potent antibacterial effects against both Gram-negative and Gram-positive bacteria. Density functional theory calculations revealed that Mn doping in MXene significantly reduced the free energy barrier of *OOH intermediate, promoting H 2 O 2 formation and subsequent reduction to •OH. The PEGylated Mn-Ti 3 C 2 (PEG@Mn-Ti 3 C 2 T x ) composite combined the superior hydrophilicity of PEG for effective protein repulsion with the enhanced conductivity of Mn-Ti 3 C 2 T x , enabling sensitive biosensing. Critically, the fabricated biosensor demonstrated high specificity for cTnI against common interferents and achieved reliable detection in spiked serum samples. It exhibited a broad detection range from 0.1   pg   mL -1 to 500   ng   mL -1 with an ultralow limit of 0.03   pg   mL -1 . This work presents a novel strategy for combating biofouling and paves the way for designing robust biosensors capable of accurate and long-term operation in clinically relevant environments, holding great promise for point-of-care diagnostics.

Diamine-PEG crosslinking engineering of PVDF: Simultaneous γ-phase crystallization control and high-frequency dielectric stabilization for flexible ferroelectrics

Yisha Ma,Haijun Wang,Qianqian Yu,Yuxin Wang,Jian Hu,Tong Wang

The γ-phase of poly(vinylidene fluoride) (PVDF) exhibits exceptional dielectric polarization and thermal stability but suffers from kinetically hindered crystallization and high-frequency dielectric losses in conventional processing. We develop a diamine-polyethylene glycol (diamine-PEG) covalent crosslinking strategy that simultaneously overcomes both limitations. The sterically confined network enables rapid γ-phase crystallization (92 % purity within 4 h at 155°C) through directed T 3 GT 3 G′ chain alignment, while suppressing parasitic dipole relaxation. This yields breakthrough dielectric performance: frequency-stable permittivity (ε' ≈ 8) with ultralow loss (tan δ = 0.21 at 10 6 Hz, 60 % reduction vs. pristine PVDF) and ferroelectric switching exhibiting 2.5-fold enhancement in polarization (4.5 μC/cm 2 at 2000 kV/cm). Crucially, the films retain exceptional flexibility (307 % elongation at break with 5 wt% crosslinker vs. 90 % for neat PVDF) and solution processability, establishing a multifunctional platform for flexible power electronics requiring concurrent high-frequency stability, thermal resilience, and mechanical durability.

Liquid PEG and PEGDA as Protective Matrices for TTA-UC Functioning in Air and Their Application in Information Encryption

Shigang Wan,Dongxuan Wang,Yizhong Shi,Shuoran Chen,Changqing Ye

Graphical Two photochemically deoxygenating compounds, PEG-200 and PEGDA-575, have been used as solvents for triplet-triplet annihilation upconversion in air, with both serving as sensitized singlet oxygen scavengers. A high-level information encryption platform has been developed using Pt(OEP)/DPA/PEGDA-575 or Pt(OEP)/DPA/PEGDA-575/PEG-200 ink that enables data to be read under specific light or at a certain time and erased by light exposure. Photochemical deoxygenation offers a promising solution to the oxygen sensitivity issue in Triplet-triplet annihilation upconversion (TTA-UC). This study utilized polyethylene glycol (PEG)-200 and polyethylene glycol diacrylate (PEGDA)-575 as solvents and singlet oxygen scavengers for TTA-UC conducted in air. The upconversion efficiency of Pt(OEP)/DPA in aerated PEG-200 is similar to that in nitrogen-saturated PEG-200. Prolonged 532 nm light irradiation causes a PEGDA-575 solution of Pt(OEP)/DPA to undergo photoinitiated polymerization via homomolecular TTA, leading to a reduction in DPA's upconversion emission and an increase in Pt(OEP)’s phosphorescence. These results enabled the development of a high-level information encryption platform using the direct-writing method with Pt(OEP)/DPA/PEGDA-575 or Pt(OEP)/DPA/PEGDA-575/PEG-200 ink. The information is only readable under certain lighting conditions or at specific times and can be erased by exposure to light.

Shear-Thinning and Self-Recovery Hydrogel Inks Based on Alkyne-Functionalized Triblock Copolypeptide and PEG for Direct Ink Writing

Yongqiang Hong,Chongguang Li,Xinyue Zhang,Yan Xiao,Meidong Lang

Direct ink writing (DIW) hydrogel inks for biomedicine are expected to balance flowability during extrusion with stability after deposition while maintaining cytocompatibility. Herein, an alkyne-functionalized amphiphilic triblock copolypeptide composed of l-glutamic acid and l-phenylalanine residues was synthesized. In aqueous environments, the copolypeptide self-assembles spontaneously through the hydrophobically driven association of the l-phenylalanine blocks, resulting in a physically cross-linked network with tunable viscoelastic properties. This dynamic network imparts pronounced shear-thinning behavior and rapid self-recovery, facilitating continuous extrusion and DIW of hydrogel filaments with excellent shape retention. Additionally, alkyne-functionalized PEG was incorporated into the hydrogel ink as a reactive diluent to accelerate photo-cross-linking kinetics and improve printing fidelity. Following the printing process, the alkyne groups enable rapid thiol–yne photo-cross-linking under 405 nm light, thereby providing an additional covalent network that reinforces the printed structures and enhances their stability. In vitro cytocompatibility experiments further confirmed the favorable biocompatibility of the printed hydrogels. Overall, this hydrogel presents a cooperative copolypeptide/PEG design for DIW-printable and photocurable hydrogel inks, with potential relevance for future biomedical applications.

CS-PEG-modified long-circulating tanshinone I liposomes for enhanced lipid-lowering synergy with atorvastatin and reduced adverse effects

Li Zhang,Yize Lv,Xuedi Weng,Yan Gao,Jie Gao,Xia Cao,Michael Adu-Frimpong,Qilong Wang

Chitosan-polyethylene glycol (CS-PEG)-modified long-circulating liposomes were developed and loaded with tanshinone I (TaI@CS-PEG-L) to overcome its poor solubility and low bioavailability, as well as enhance the lipid-lowering efficacy of atorvastatin (At) and mitigate its tissues toxicity. After synthesis of CS-PEG copolymer, it was characterized with infrared and proton nuclear magnetic resonance. Thin-film hydration was used to prepare TaI@CS-PEG-L before optimization was performed using Box-Behnken design. Optimal liposomal formulation was obtained when mass ratio of lecithin:cholesterol:TaI mass ratio was 12:1:1 along with 0.2% CS-PEG, which displayed appropriate particle size (117.41 nm), polydispersity index (0.182), zeta potential (+17.31 mV), encapsulation efficiency (93.15%), and drug loading (5.19%). Besides, the liposomal preparation exhibited good long-term storage stability (over 30 days at 4 °C and 25 °C) and satisfactory gastrointestinal stability in simulated gastric (pH 1.2) and intestinal (pH 6.8) fluids for 4 h. TaI@CS-PEG-L displayed increased cellular uptake and in vitro release compared with free TaI and unmodified liposomes. Regarding pharmacokinetic studies, TaI@CS-PEG-L increased AUC0-ₜ, t1/2, and Cmax in rats by 7.26-fold, 2.43-fold, and 3.12-fold, respectively, which indicates significantly enhanced oral bioavailability. TaI@CS-PEG-L monotherapy markedly lowered serum lipid levels in hyperlipidemic mice, thereby improving liver function indices, and alleviating pathological liver injury. TaI@CS-PEG-L demonstrated synergistic lipid-lowering effects and efficiently reversed At-induced hepatic and pancreatic injuries after combination with At, thus outperforming free TaI and TaI@L. Therefore, CS-PEG-modified long-circulating liposomes could efficiently address delivery challenges of TaI, thereby yielding a formulation that could potently lower serum lipid levels and protect liver against injury. The findings of this study offer a promising ‘efficacy-enhancing and toxicity-reducing’ strategy for potential treatment of hyperlipidemia in the clinics.

Synergistic Interaction between Saturated MgCl2–H2O/PEG Electrolyte and Mg Alloy Anode Enables High-Performance Aqueous Magnesium-Ion Batteries

Wendi Lai,Caixing Wang,Rui Fang,Kanghui Liu,Wei Yang,Guowang Diao,Yanrong Wang

Compared to conventional organic electrolytes, aqueous magnesium-ion batteries (AMIBs) have emerged as a research hotspot due to their superior safety, environmental compatibility, and cost-effectiveness. However, the further development of AMIBs has been impeded by their short cycling lifespan, primarily caused by the aqueous corrosion of Mg anodes in water-containing electrolytes, which generates inorganic passivation layers (e.g., Mg(OH)2 and MgO) on the anode surface. To address this challenge, we propose a collaborative two-pronged approach, involving the adoption of a novel electrolyte system (saturated MgCl2 aqueous solution containing 25 vol % PEG, SMP-3) and the use of a Mg alloy anode. Electrochemical investigations suggest that the PEG molecules form hydrogen bonds with H2O molecules, effectively suppressing water activity and thereby enhancing anode stability. The assembled PTO/Mg alloy full cell exhibited a stable discharge plateau at 1.5 V with an initial discharge capacity of 176.24 mAh g–1 at 200 mA g–1. Remarkably, the cell demonstrated exceptional cycling stability, maintaining a capacity of 150 mAh g–1 after 300 cycles (85% capacity retention), with an average Coulombic efficiency approaching 100%. The results demonstrate the feasibility of stabilizing Mg alloy anodes through an interfacial engineering strategy based on a water-in-salt electrolyte design. This strategic approach provides a novel pathway for developing low-cost, high-performance AMIBs.

Redox-responsive mesoporous silica as carriers for controlled drug delivery: A comparative study based on silica and PEG gatekeepers

Ying Wang,Ning Han,Qinfu Zhao,Ling Bai,Jia Li,Tongying Jiang,Siling Wang

Hybrid mesoporous silica nanoparticles (MSNs) modified with polymer polyethylene glycol (PEG) through the biodegradable disulfide bonds were prepared to achieve ‘on demand’ drug release. In this system, PEG chains were chosen as the representative gatekeepers that can block drugs within the mesopores of MSNs. After the addition of glutathione (GSH), the gatekeepers were removed from the pore outlets of MSNs, followed by the release of encapsulated drugs. In this research, the effects of grafting density of gatekeepers on the drug release and biocompatibility of silica carriers were also investigated. First, PEG modified MSNs were prepared by the condensation reaction between the carboxyl groups of MSN and the hydroxyl of PEG. The structure of the resultant MSN-SS-PEG was characterized by transmission electron microscopy (TEM), nitrogen adsorption/desorption isotherms analysis and Fourier transform infrared spectroscopy (FTIR). Rhodamine B (RhB) as the model drug was loaded into MSNs. The in vitro assay results indicated that RhB was released rapidly after the addition of 10   mM GSH; M 1 -SS-PEG had the best capping efficiency compared with M 0.5 and M 1.5 groups. Moreover, hemolysis assay, serum protein adsorption and cell viability test indicated that with the increase of PEG grafting density, the biocompatibility of silica carriers increased.
