Theanine Capture of Reactive Carbonyl Species in Humans after Consuming Theanine Capsules or Green Tea

Yuqing Zhong,Chen Yang,Yongling Lu,Lishuang Lv

Acrolein (ACR), methylglyoxal (MGO), and glyoxal (GO) are a class of reactive carbonyl species (RCS), which play a crucial role in the pathogenesis of chronic and age-related diseases. Here, we explored a new RCS inhibitor (theanine, THE) and investigated its capture capacity on RCS in vivo by human experiments. After proving that theanine could efficiently capture ACR instead of MGO/GO by forming adducts under simulated physiological conditions, we further detected the ACR/MGO/GO adducts of theanine in the human urine samples after consumption of theanine capsules (200 and 400 mg) or green tea (4 cups, containing 200 mg of theanine) by using ultraperformance liquid chromatography–time-of-flight–high-resolution mass spectrometry. Quantitative assays revealed that THE-ACR, THE-2ACR-1, THE-MGO, and THE-GO were formed in a dose-dependent manner in the theanine capsule groups; the maximum value of the adducts of theanine was also tested. Furthermore, besides the RCS adducts of theanine, the RCS adducts of catechins could also be detected in the drinking tea group. Whereas, metabolite profile analysis showed that theanine could better capture RCS produced in the renal metabolic pathway than catechins. Our findings indicated that theanine could reduce RCS in the body in two ways: as a pure component or contained in tea leaves.

Quantitative analysis of acetamiprid and thiacloprid in green tea using enhanced SERS and neural networks

Dongjian Li,Mulinde Timothy Ezra,Haoran Li,Yifei Chen,Chengyun Si,Xuefang Luo

Neonicotinoids (NEOs) are widely used in agricultural cultivation, and the presence of residual NEOs poses a serious threat to consumer health. Therefore, trace detection of NEOs is particularly important. In this study, a response surface experimental design was employed to develop AgNP with high enhancement factor and surface height anisotropy, effectively enhancing the Raman signals of acetamiprid (ACE) and thiacloprid (THI) through densely populated hotspots on the surface. Gaussian simulation calculations were used to assign peaks in the ACE and THI spectra. Subsequently, convolutional neural network, backpropagation neural network, and AlexNet neural network were employed to identify the ACE, THI, and mixed component spectra, successfully distinguishing the spectra of the three compounds and achieving qualitative analysis of unknown spectra. Furthermore, genetic algorithm-partial least squares was used to model the spectral data, achieving a correlation coefficient of 0.97. The method was applied for the detection of spiked tea samples, achieving a precision RSD of 4.84 %. Therefore, this sensor combined with intelligent algorithms can be used for spectral identification and trace detection of mixed pesticide samples with structurally similar compounds.

Linyao Song,Fengwei Ma,Haijiang Chen,Qiang Fei,Guangcan Tao,Siyao Wu,Dajuan Shi,Junyi Deng,Degang Zhao,Xuan Dong,Yichen Zhao,Su Xu

This study employed GC–MS, GC-IMS, and sensory evaluation to investigate dynamic changes in flavor during the solid-state fermentation of black tea by Eurotium cristatum . The results revealed a notable decrease in the bitter and astringent tastes of the black tea infusion following fermentation, while the mellow taste increased significantly. A total of 152 and 129 VOCs were detected by GC–MS and GC-IMS, respectively. And 4 key aroma-active compounds were identified by ROAV. These specific VOCs contributed floral, honey, and sweet scents, which were responsible for the fungal floral aroma in the processed black tea. Furthermore, OPLS-DA identified 31 key VOCs that played a crucial role in differentiating various fermentation stages, with day 4 recognized as a pivotal point for aroma development. The solid-state fermentation with Eurotium cristatum resulted in fermented black tea characterized by a mellow taste and a rich fungal floral aroma, enhancing the flavor quality of the tea.

Identification and Functional Analysis of PR Genes in Leaves from Variegated Tea Plant (Camellia sinensis)

Zuguo Xi,Huiyan Jia,Yifan Li,Jinqing Ma,Mengqian Lu,Zhihui Wang,Dexu Kong,Wei-Wei Deng

Tea is a healthy beverage made from the leaves of the tea plant [Camellia sinensis(L.) O. Kuntze]. The tea plant is a perennial evergreen plant that is widely distributed in tropical and subtropical regions. PR proteins (pathogenesis-related proteins, PRs) are defense-related proteins induced under pathogenesis-related conditions. Currently, there are a few studies on PRs in plants. Tea leaf color mutants have been of wide interest to scientists as special materials. A tea plant displaying variegated spontaneous albinism found in a local tea plantation in Huangshan City, Anhui Province, China, was used as the material sample in this study. An analysis of preexisting transcriptomic and proteomic data revealed the significant upregulation of three classes of PRs. In order to investigate the correlation between PRs and variegated tea plants, a series of studies were conducted onPRgenes. The results revealed the identification of 17CsPR1, 3CsPR4and 31CsPR5genes in tea plants using bioinformatics methods, and their structures and promoter sequences. The expression of threePR1, twoPR4and onePR5genes was determined to be induced in stress treatment experiments involving mechanical damage, insect bites, low temperature treatment, and fungal infections. Additionally, the ribonuclease activity of CsPR4 was successfully verified. This is the first study to report the ribonuclease activity of CsPR4 in tea plants. The results can serve as a reference for future studies on PRs in tea plants, offering new insights into information on albinism in tea plants. Keywords:identification;function analysis;pathogenesis-related protein;variegated tea plants

Efficient degradation of organic pollutants using carbon coil aerogels from waste tea as peroxymonosulfate activators

Feng Tao,Hao Chen,Hao Wang,Haobeng Huang,Ao Dong,Mengwei Zheng,Zhijun Wang

The efficient activation of peroxymonosulfate (PMS) using metal-free catalysts in advanced oxidation processes (AOPs) has garnered significant attention for its potential in organic pollutant remediation. However, the development of cost-effective, high-performance, and recyclable catalysts for PMS activation remains a major challenge. In this study, we introduce a novel class of biomass carbon microcoil aerogels (BCCA), characterized by low cost, ultralight, highly elastic, and recyclable properties, and assess their performance in PMS-driven degradation of organic pollutants in wastewater. Our results demonstrate that the BCCA-700/PMS system achieved over 95 % removal of methylene blue and methyl orange within 10 min, while also removing 96.5 % of tetracycline within 90 min. Additionally, the BCCA exhibited remarkable mechanical stability, maintaining over 80 % of its initial degradation capacity after 4 cycles and demonstrating consistent performance across a wide range of pH levels and temperatures, with minimal impact on plant growth. The carbonization temperature was found to be a critical factor influencing the specific surface area, porosity, and distribution of oxygen-containing functional groups in the BCCA, which in turn enhanced both pollutant adsorption and PMS activation. The synergistic effects of these properties significantly improved contaminant removal efficiency. These findings not only present a sustainable approach for utilizing biomass waste, but also offer an effective and environmentally friendly material for advanced water treatment.

Chemical profiling, discriminant analysis, antioxidants, and anti-inflammatory activities of Lonicerae Japonicae Flos tea varieties

Jiao Wu,Xiaoyan Du,Fangchao Tong,Shuangling Zhang,Jiaojie Guo,Changchun Zhu,Rui Cui,Qirong Bai,Mengyao Chen,Li Meng,Linlin Song,Haichuan Yu

Lonicerae Japonicae Flos (LJF), a widely consumed functional tea ingredient, was investigated for its phytochemical composition and bioactive properties. Ten LJF tea varieties were systematically analyzed for six bioactive compounds (chlorogenic acid, secoxyloganin, sweroside, cynaroside, isochlorogenic acid A, and isochlorogenic acid C) using high-performance liquid chromatography (HPLC) quantification. Multivariate statistical approaches including principal component analysis (PCA) and partial least squares-discriminant analysis (PLS-DA), were employed, with 80% of variance being explained by PCA (PC1+PC2), while isochlorogenic acid C (VIP=1.32) and secoxyloganin (VIP=1.18) were identified by PLS-DA as the most significant markers for varietal differentiation. Comprehensive bioactivity evaluation included: (1) in vitro antioxidant capacity assessment via DPPH and ABTS∙ + assays, revealing significant free radical scavenging capacities (0.38-0.71 mmol VCE/g and 1.22-6.35 mmol TE/g, respectively); (2) in vivo anti-inflammatory evaluation using zebrafish models, demonstrating superior efficacy against pathogen-induced versus wound-induced inflammation. Meaningful relationships between specific phytochemical components and observed bioactivities were established by Spearman correlation analysis and heatmap clustering. Notably, the cultivar Fenghua No.1 exhibited optimal bioactive performance, achieving peak antioxidant values and significant anti-inflammatory effects. The results validate LJF's functional tea status while highlighting enhanced bioactivity in newer cultivars, particularly Fenghua No.1, supporting their potential nutraceutical applications.

Tea Polyphenol-Derived Zinc-Rich Complexes for Enhanced Interfacial Dynamics in PEO-Based Gel Electrolytes

Tianyu Zou,Bingchun Jiang,Qian Qu,Bozhong Cao,Jingyu Xu,Shubing Zhen,Jincheng Zhang,Lei Ling,Yusheng Lu,Tong Zhang

The aqueous zinc-ion battery (AZIB) is regarded as one of the most promising energy storage solutions. However, its widespread adoption is hindered by challenges such as zinc dendrite formation and undesirable side reactions, primarily caused by excess free water molecules and ions. This study introduces an ethylene oxide (PEO)-based gel electrolyte designed to address these limitations. By incorporating a zinc-rich ionic conductor (TP-Zn) that establishes a cross-linked hydrogen bond network, we successfully reduce the crystallinity of the PEO matrix and enhance the ionic conductivity and mechanical strength of the electrolyte. The resulting PGPS@TP-Zn gel electrolyte exhibits a remarkable tensile deformation of up to 1785%. The incorporation of TP-Zn significantly alleviates interfacial challenges between the electrolyte and electrode, leading to a more uniform zinc ion flux distribution. This, in turn, improves ion transport kinetics, resulting in a high ionic conductivity of 1.32 × 10–3 S cm–1 and a zinc ion transference number of 0.86. Furthermore, the Zn||Zn symmetrical cell shows exceptional stability when using PGPS@TP-Zn, with a cycle life exceeding 3000 h at a current density of 1 mA cm–2 and a capacity of 1 mAh cm–2. In addition, the Zn||PGPS@TP-Zn||α-MnO2 full cell maintains a capacity retention of 76.9% even after 1000 cycles at a current of 2 A g–1. Remarkably, the assembled pouch cell functions normally even when bent to a maximum angle of 180 degrees. This study highlights the significant practical advantages of the PGPS@TP-Zn gel electrolyte design for advanced AZIB applications.

Oil-water interface behavior of tea polysaccharide-egg white protein nanoparticles and characterization of bigels

Yue Chen,Jingjing Huang,Yexu Wu,Xiaoqiang Chen

The O/W interface behavior of tea polysaccharide conjugate-egg white protein (TPC/EWP) nanoparticles was investigated and a bigel system was constructed. The results showed that TPC/EWP nanoparticles with better storage and thermal stability were successfully prepared, in which the mass ratio of tea polysaccharide conjugate to egg white protein of 3:1 helped to enhance the diffusion, penetration and rearrangement processes. To make O/W interfacial layer had higher viscoelasticity and expansion modulus. The nanoparticle-based bigel exhibited fine texture and was not easy to flow. In addition, the tight gel network endowed the bigels with strong rheological properties and stability. These results establish a theoretical foundation for further investigation into O/W interface behavior of TPC/EWP nanoparticles. The bigels' excellent gel properties and stability provide a new direction for applying the bigel as a novel food in the food field.

Investigation of Emulsifying Properties and Stability of Fish Gelatin and Tea Saponin Complex Emulsion System

Kaixuan Wang,Shengnan Zhan,Jianyuan Yang,Qiaoming Lou,Tao Huang

In this study, environmental stability, rheological properties, and structural characterization of co-stabilized emulsions using fish gelatin (FG) and tea saponin (TS) were investigated. The results demonstrated that the addition of TS significantly enhanced the emulsifying properties of FG, and the FG-TS 0.1% emulsion had the smallest particle size. TS and FG co-stabilized emulsions provided resistance to salt and high temperatures. Optical microscopy and CLSM showed that the addition of TS made FG more effectively adsorb at the oil–water interface, leading to the formation of more uniform oil droplet sizes. Additionally, the addition of TS increased the viscosity of FG emulsions, which reduced emulsion flocculation. Results of intrinsic fluorescence, FTIR, and surface hydrophobicity revealed that the addition of TS altered the secondary structure of FG, enhancing surface hydrophobicity and improving emulsification. In conclusion, the moderate addition of TS significantly enhanced the emulsification and rheological properties of FG, suggesting new potential applications for FG in various industries.

High internal phase Pickering emulsions stabilized by tea residue protein: Application in β-carotene encapsulation

Xiaoyan Guo,Junjie Zhang,Minghui Zhu,Xiaohan Xu,Lu Liu,Shengnan Chen,Yu Wang,Xueling Gao,Qi Chen

The increasing production and consumption of tea drinks has led to the generation of large amounts of discarded extracted tea residues. As a result, researchers have attempted to extract tea water-insoluble protein (TP) from discarded tea residues to produce food emulsifiers. Thus, in this study, high-internal-phase Pickering emulsions (HIPPEs) stabilized by TP were developed and characterized. First, the effects of salt ions on the emulsifying properties of TP were examined using interfacial tension and hydrophobicity. Fourier transform infrared spectroscopy was used to determine the suitable range of salt ions in the processing stage. Then, the particle size distribution, microstructure, rheological properties, and stability of the emulsions were systematically investigated by controlling the oil phase volume, particle concentration of TP, and emulsification method. The results showed that TP was effectively adsorbed on the oil–water interface and formed a stable particle layer, which means that TP-stable high-internal-phase Pickering emulsions (TPHIPPEs) has been successfully prepared. Further analysis showed that TPHIPPEs exhibited good stability and gelation properties. The pH range was 7–9, and the salt ion concentration was <0.5 M. Additionally, TPHIPPEs exhibited excellent temperature tolerance and antioxidant ability. Finally, the application development results revealed that the loading and retention rates of β-carotene in TPHIPPEs were significantly higher than those of the control group of camellia oil, and that TPHIPPEs exhibited good resistance to UV light and thermal degradation. This study provides new insights into the high-value utilization of tea residue resources.

Unraveling anti-aging mystery of green tea in C. elegans: Chemical truth and multiple mechanisms

Jia-ping Ke,Jia-Yi Li,Zi Yang,Hao-Yue Wu,Jing-Ya Yu,Yi Yang,Chen-Hui Chen,Peng Zhou,Fang Hua,Wei Wang,Fenglin Hu,Gang-Xiu Chu,Xiao-Chun Wan,Guan-Hu Bao

Tea drinking impacts aging and aging-related diseases. However, knowledge of anti-aging molecules other than the major catechins in complex tea extracts remains limited. Here we used Caenorhabditis elegans to analyze the longevity effects of tea extracts and constituents comprehensively. We found that the hot water extract of green tea prolonged lifespan and heathspan. Further, the MeOH fraction prolonged lifespan significantly longer than other fractions. Correlation analysis between mass spectroscopic data and anti-aging activity suggests that ester-type catechins (ETCs) are the major anti-aging components, including 4 common ETCs, 6 phenylpropanoid-substituted ester-type catechins (PSECs), 5 cinnamoylated catechins (CCs), 7 ester-type flavoalkaloids (ETFs), and 4 cinnamoylated flavoalkaloids (CFs). CFs (200 μM) are the strongest anti-aging ETCs (with the longest 73% lifespan extension). Green tea hot water extracts and ETCs improved healthspan by enhancing stress resistance and reducing ROS accumulation. The mechanistic study suggests that they work by multiple pathways. Moreover, ETCs modulated gut microbial homeostasis , increased the content of short-chain fatty acids, and reduced fat content. Altogether, our study provides new evidence for the anti-aging benefits of green tea and insights into a deep understanding of the chemical truth and multi-target mechanism.

Pu-erh tea increases the metabolite Cinnabarinic acid to improve circadian rhythm disorder-induced obesity

Shanshan Hu,Changhua Hu,Liyong Luo,Haotian Zhang,Sibo Zhao,Zhonghua Liu,Liang Zeng

Obesity is one of the circadian rhythm disorders (CRD)-mediated metabolic disorder syndromes. Pu-erh tea is a viable dietary intervention for CRD, however its effect on CRD-induced obesity is unclear. Here, we found that Pu-erh tea improved obesity in CRD-induced mice, which stemmed from the production of Cinnabarinic acid (CA). CA promoted adipose tissue lipolysis and thermogenic response ( HSL , ATGL , Pparα, CKB, UCP1) and increased adipocyte sensitivity to hormones and neurotransmitters by targeting the expression of adipose tissue receptor proteins (Q6KAT8, P51655, A2AKQ0, M0QWX7, Q6ZQ33, and mGluR4). This improved mitochondrial activity and facilitated adipose tissue metabolic processes, thereby accelerating glucolipid metabolism. Also, CA-induced alterations in gut microbes and short-chain fatty acids further improved CRD-mediated lipid accumulation. These results suggest that the increase of CA caused by Pu-erh tea, targeted to adipose tissue via the metabolite-blood circulation-adipose tissue axis, maybe a key mechanism for reducing the development of CRD-induced obesity.
