Chitosan-based hydrogel dressings with antibacterial and antioxidant for wound healing

Yongyan Yang,Ying Ma,Haodong Wang,Chongruihan Li,Chaoqun Li,Ruiting Zhang,Shuangling Zhong,Wenqi He,Xuejun Cui

Bacterial infection and free radical oxidative stress at the wound site could easily cause cascade inflammation and hinder the healing process of the wound. In this study, chitosan-cysteine-gallic acid (CCG) hydrogel with antibacterial and antioxidant properties was synthesized by chitosan (CS), cysteine (Cys), and gallic acid (GA) for a preliminary evaluation of its therapeutic efficacy in a mouse model of full-layer skin defect. In vitro analysis showed that the CCG hydrogel had good antibacterial activity and blood compatibility. In vivo , the CCG hydrogel wound dressings accelerated wound healing, stimulate angiogenesis, increase collagen deposition and anti-inflammatory factor expression. The CCG hydrogel wound dressing is designed to promote the regeneration of damaged skin tissue and is expected to become a potential candidate for clinical treatment.

Chitosan-modified iron fillings materials for remediation of arsenic-contaminated soil

Yunfeng Xu,Xiaoxun Huang,Chuyin Liu,Dongning Kong,Guangren Qian

Arsenic (As) contamination of soils adversely affects human health and ecosystems and is an environmental issue of serious concern worldwide. Efficient and economical remediation methods are urgently needed. In this research, a chitosan-modified scrap iron filings (CFF) composite was prepared by using industrial scrap iron filings and chitosan to stabilize As-contaminated soil. The optimal modification conditions were explored, and CFF was applied to As-contaminated soil to investigate its remediation effect and stabilization mechanism. The results showed that the optimal preparation conditions of CFF were chitosan concentration of 3.75 % (w/w), chitosan to scrap iron filings ratio of 1:1, optimal modification temperature of 40 °C, iron filings particle size of 35–80 mesh, and 10 % glutaraldehyde crosslinking for 6 h, and the stabilization efficiency of As was 73.98 %. The adsorption behavior of CFF was consistent with the pseudo-second-order kinetic model and the Langmuir isotherm model, and the best adsorption effect was achieved at pH = 5–6. The possible mechanisms of As stabilization by CFF were surface complexation, hydrogen bonding synergy, precipitation, electrostatic attraction, and van der Waals forces.

Biodegradable aminated alginate-chitosan porous polymer for efficient indoor formaldehyde capture

Furong Li,Tao Zhu,Bo Yuan,Jun Yang,Qiuge Ju,Weifang Li,Zefu Ye,Xudong Xu,Zhujun Zhu

Formaldehyde, even at low concentrations, poses a significant indoor pollutant risk, underscoring the need for efficient and sustainable sorbents. This study introduces a novel, multifunctional, and fully biodegradable adsorbent derived from waste shrimp shells. The shells were converted into soluble chitosan, which was then physically crosslinked with branched amine-modified alginate, resulting in a porous biuret-modified alginate/chitosan (BCC) polymer. BCC, with a 6 wt% biuret content, demonstrated exceptional adsorption performance for low-concentration formaldehyde, achieving a capacity of 1.97 mg·g −1 —159% higher than alginate and 149 % greater than activated carbon. The adsorption kinetics followed the pseudo-second-order model, and the Sips isotherm model suggested a chemical adsorption-dominated mechanism, with physical adsorption acting as a supplementary process. Additionally, BCC exhibited excellent reusability, maintaining high adsorption efficiency after four regeneration cycles, and incorporated a colorimetric signaling function, with a detection threshold of 0.07 mg·m −3 and a sensitivity of 0.02 mg·m −3 . These findings provide valuable insights and a theoretical basis for developing effective, safe, and environmentally friendly formaldehyde adsorbents.

Chitosan bis(halophenylcarbamate)-(isobutyrylamide) based chiral stationary phases for enantiomeric separation

Sheng Tang,Qin Bin,Zi-Wei Feng,Wei Chen,Shao-Hua Huang,Zheng-Wu Bai

The goal of the present study is to systematically investigate the enantioseparation performance and eluent tolerance of chiral stationary phases (CSPs) derived from chitosan bis(halophenylcarbamate)-(isobutyrylamide)s. Hence, N-isobutyrylated chitosan was prepared from completely deacetylated chitosan with a high molecular weight. The N-isobutyrylated chitosan was further derivatized with various halo-substituted phenyl isocyanates, consequently affording a series of chitosan bis(halophenylcarbamate)-(isobutyrylamide)s, which were coated onto 3-aminopropyl silica particles to obtain a class of CSPs. The chiral recognition ability of these CSPs was evaluated with nineteen racemates. It was found that the chitosan bis(halophenylcarbamate)-(isobutyrylamide)s showed powerful enantioseparation performance, and those with 4-chlorophenylcarbamate, 3,4-dichlorophenylcarbamate and 4-trifluoromethoxyphenylcarbamate seemed to exhibit comparable or even more superior enantioseparation ability for the tested racemates in comparison with the well-known derivatives of cellulose and amylose. In addition, the dissolvability and swelling of the prepared chitosan bis(halophenylcarbamate)-(isobutyrylamide)s in organic solvents were discussed in particular. The eluent tolerance of the CSP prepared from chitosan bis(3,4-dichlorophenylcarbamate)-(isobutyrylamide), which exhibited relatively more favorable enantioseparation performance among the prepared CSPs, was investigated. It was observed that the tested CSP could be analyzed with 100% ethyl acetate, 100% chloroform and n-hexane/THF (70/30, volume ratio) without a significant loss of recognition and enantioseparation ability. As a consequence, we can safely draw the conclusion that chitosan bis(halophenylcarbamate)-(isobutyrylamide)s prepared from chitosan with a high molecular weight can show good enantioseparation performance as well as satisfactory eluent tolerance.

Chitosan modified halloysite nanotubes as emerging porous microspheres for drug carrier

Xiaoyu Li,QianYang,Jing Ouyang,Huaming Yang,Shi Chang

Natural halloysite (Hal) nanotubes were well dispersed and thoroughly emulsified to prepare the porous microspheres Hal-chitosan (Hal-CTS) in a water/oil microemulsion, which was further loaded with aspirin (Asp) as a model drug to investigate the drug release behavior. The charge-mediated adsorption of molecules to the surface of Hal could facilitate the dispersion of Hal in aqueous media and enabled Hal to assemble into microspheres. The microspheres had interconnected pores, large pore volume, high specific surface area and hierarchical pore distribution, which was beneficial to the entrance of Asp molecules. The porous microspheres showed excellent loading capacity for Asp as high as 42.4   wt%, around 20 times higher than the pristine Hal (2.1   wt%). The release results indicated that Hal-CTS/Asp nanocomposites had the lower release in the simulated gastric fluid, whereas more rapid release rate and higher release amount in the simulated intestinal fluid, which could help to maintain effective concentration of Asp in the body and reduce the maximum extent of the side effects to the stomach. The as-prepared Hal-CTS/Asp nanocomposites could have potential application in the drug carrier systems due to their natural and excellent biocompatibility.

Gentamicin modified chitosan film with improved antibacterial property and cell biocompatibility

Yang Liu,Peihong Ji,Huilin Lv,Yong Qin,Linhong Deng

Gentamicin modified chitosan film (CS-GT) was produced using a three-step procedure comprising: (i) the chitosan solution was air-dried to form a chitosan (CS) film, (ii) using citric acid to generate the amide and carboxyl groups on the surface of CS, (iii) the CS with surface carboxyl groups was modified by grafting of gentamicin. After modification, this CS-GT film has excellent hydrophilicity and biocompatibility. It is very evident that the gentamicin grafting treatment significantly improves the antibacterial properties of the CS film. Our preliminary results suggest that this novel gentamicin modified chitosan film, which can be prepared in large quantities and at low cost, should have potential application in biomedical applications.

Preparation and characterization of chitosan-graft-poly(L-lactic acid) microparticles

Shiping Zhan,Nan Liu,Weijing Wang,Shuhua Chen,Jingchang Wang

To improve the properties of chitosan (CS) and poly(L-lactic acid) (PLLA) and obtain fully biodegradable materials, CS- g -PLLA copolymers were prepared using 1-(3-Dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride (EDC)/N-hydroxyl succinimide (NHS) as a coupling agent. The copolymers were characterized by Fourier transform infrared analysis (FTIR), 1 H nuclear magnetic resonance ( 1 H NMR), elemental analysis, differential scanning calorimetry (DSC), X-ray diffraction (XRD) and scanning electron microscopy (SEM). The results obtained by FTIR and 1 H NMR showed that CS and PLLA were grafted successfully via an amide bond. DSC and XRD results showed that the thermal stability of CS had been significantly improved by grafting PLLA to the molecular chains of CS and the crystallinity of the CS- g -PLLA copolymers decreased significantly. Elemental analysis showed that the achieved the maximum degree of substitution of PLLA was 60.88%, while the concentration of CS was 2 mg/mL, the PLLA molecular weight was 10,000, and the EDC/NHS ratio was 2:1. Images from SEM demonstrated that the copolymers had a spherical shape and smooth surface. Moreover, the products were well dispersed without any aggregation. POLYM. ENG. Chitosan-based spray-dried mucoadhesive microspheres for sustained oromucosal drug delivery

Wang-Zhan Jiang,Yang Cai,Hao-Ying Li

The main issues attracting substantial consideration when delivering drugs to the oromucosa include the enhancement of mucoadhesion of drug formulations in order to elongate their residence time on the site of delivery and the sustained release of drug from the formulations following deposition. In this study, the mucoadhesive biopolymers of chitosan and its derivatives, including high molecular weight , low molecular weight, and carboxylic derivative, were employed as the excipients to prepare spray-dried microspheres for the oromucosal drug (e.g. benzydamine hydrochloride, BZH) delivery with the aim to enhance the delivery efficiency. All chitosan-modified spray-dried powders showed the size <   10   μm with greater surface roughness , moisture content <   10% w/w, and improved flowability . The addition of chitosan in the formulations can tremendously extend the duration of drug release. More importantly, the mucoadhesive interaction of chitosan-modified microparticles was strongly enhanced, and it can be further improved as the augmentation of chitosan contents in the formulations. Our data clearly demonstrate that the inclusion of chitosan in spray-dried microspheres elongates the duration of drug release and also enhance the mucoadhesive interaction force of microspheres to the mucosal membrane, which may be of benefit in the treatment of local conditions.

Chitosan-Assisted Crystallization and Film Forming of Perovskite Crystals through Biomineralization

Yang Yang,Chen Sun,Hin-Lap Yip,Runcang Sun,Xiaohui Wang

Graphical Crystal palace : Perovskite crystals with controlled morphology and properties are formed in a chitosan template by three steps including electrostatic adsorption/chelation, nucleation, and growth. The exposed functional groups of chitosan coordinate strongly with Pb 2+ and some of these stabilized Pb 2+ ions interact with I − ; nucleation occurs and perovskite crystals start to grow in the chitosan network. Biomimetic mineralization is a powerful approach for the synthesis of advanced composite materials with hierarchical organization and controlled structure. Herein, chitosan was introduced into a perovskite precursor solution as a biopolymer additive to control the crystallization and to improve the morphology and film-forming properties of a perovskite film by way of biomineralization. The biopolymer additive was able to control the size and morphology of the perovskite crystals and helped to form smooth films. The mechanism of chitosan-mediated nucleation and growth of the perovskite crystals was explored. As a possible application, the chitosan–perovskite composite film was introduced into a planar heterojunction solar cell and increased power conversion efficiency relative to that observed for the pristine perovskite film was achieved. The biomimetic mineralization method proposed in this study provides an alternative way of preparing perovskite crystals with well-controlled morphology and properties and extends the applications of perovskite crystals in photoelectronic fields, including planar-heterojunction solar cells.

Aqueous Hg(II) immobilization by chitosan stabilized magnetic iron sulfide nanoparticles

Mingyang Sun,Guanghuan Cheng,Xinlei Ge,Mindong Chen,Cheng Wang,Liping Lou,Xinhua Xu

Stabilized iron sulfide (FeS) nanoparticles have been proven effective in the adsorption of Hg from the water environment. However, previous work with these nanoparticles determined that the separation from the treated water was difficult and time-consuming. In this study, nanoscale FeS-Fe 3 O 4 nanocomposites were firstly synthesized with chitosan as the stabilizer (CTO-MFeS). Then, the Hg adsorption capacity and mechanism were studied. Results showed that the size of the prepared nanoparticles was about 20   nm and the specific surface area was 21.3   m 2 /g. Hg removal by the CTO-MFeS nanoparticles involved both adsorption and precipitation. Further investigation with XPS showed that Hg 2   + was adsorbed on the surface of the CTO-MFeS nanoparticles and reacted with CTO-MFeS to form HgS and [Fe (1   −   x) Hg x ]S. It was also found as pH decreased below 4, the adsorption capacity of CTO-MFeS was significantly reduced that might be due to the dissolving of Fe. Additionally, the presence of Cl − resulted in the transformation of Hg 2   + to HgClx 2-x (x   =   1, 2, 3, 4) that competed with OH in solution for Hg 2   + and therefore inhibited the adsorption of Hg. Our findings provide additional information that may be useful for a theoretical basis for Hg treatment in water environment.

Surface-modified PLGA nanoparticles with chitosan for oral delivery of tolbutamide

Yongli Shi,Jintao xue,Liyun Jia,Qian Du,Jie Niu,Dongyang Zhang

The main purpose of present study was to develop novel chitosan-modified polylactic-co-glycolicacid nanoparticles (CS@PLGA NPs) for improving the bio-availability of tolbutamide (TOL). The TOL-loaded CS@PLGA NPs (TOL-CS@PLGA NPs) were fabricated with the solvent evaporation method. The cargo-free CS@PLGA NPs showed a diameter of 228.3   ±   2.5   nm monitored with a laser light particlesizer, and the transmission electron microscope (TEM) photographs revealed their “core-shell” structures. The Zeta potential of the original PLGA NPs and the cargo-free CS@PLGA NPs was measured to be −20.2   ±   3.21   mV and 24.2   ±   1.1   mV, respectively. The changes in Zeta potential indicated the CS chains were coated on the surfaces of the original PLGA NPs. The thermal gravity analysis (TGA) curves suggested that the CS chains improved the thermostability of the original PLGA NPs. The results of cells viability indicated the cargo-free CS@PLGA NPs were nontoxicity. The in vitro release profiles suggested that TOL-CS@PLGA NPs could release TOL in pH 7.4 phosphate buffer solution (PBS) at a sustained manner. Streptozotocin (STZ) was employed to build the diabetic rat models. The physiological changes in the islet β cells confirmed the obtaining of diabetic rats. After treatment by gavage, the TOL-CS@PLGA NPs showed an excellent hypoglycemic effect. Therefore, the TOL-CS@PLGA NPs had a potential application in oral delivery of TOL.
