Identification of Polyphenol Oxidase Inhibitors from the Root of Pueraria Lobata: Inhibitor Profiles, Kinetics Analysis and Molecular Interaction

Liu Junping,Liang Chaorui,Jiang Hongwei,Yu Ziqi,Zou Liqiang,Zhou Lei,Liu Wei

Enzymatic browning induced by polyphenol oxidase (PPO) plays a significant role to quality reduction in fruit and vegetables. PPO inhibitors derived from natural products are gaining attention for their strong inhibitory abilities and low toxicity. This study aims to identify the component of phenolic-rich extract from Pueraria lobata root and evaluate their inhibitory capacity on PPO. The results showed that nine phenolics were identified by UPLC-QE-Orbitrap-MS method as PPO effective inhibitors based on their IC 50 values, in which kaempferol showed the strongest inhibitory effect. Among the inhibitors, kaempferol, daidzein, phloretin and luteolin with different inhibitory abilities were selected as representative flavonols, isoflavones, chalcones and flavonoids compounds to investigate the kinetics analysis and molecular interaction. Kaempferol and daidzein exhibited competitive inhibition on PPO, while phloretin and luteolin showed mixed-type and non-competitive inhibition, respectively. According to the fluorescence spectra results, these inhibitors were bound to PPO and quenching fluorescence, among which kaempferol showed the highest K b value and accompanied by the blueshift. Molecular docking results indicated that the inhibitors interact with PPO primarily through hydrophobic interactions, van der Waals forces and hydrogen bonds, in which kaempferol showed the strongest binding ability with the lowest binding energy. This research provides a theoretical basis for the practical application of Pueraria lobata root extracts as natural PPO inhibitors in the food industry.

Inhibition of cinnamic acid and its derivatives on polyphenol oxidase: Effect of inhibitor carboxyl group and system pH

Hongwei Jiang,Lei Zhou,Yue Wang,Guangxian Liu,Shengfeng Peng,Wenzhi Yu,Yuqing Tian,Junping Liu,Wei Liu

Phenolic acids are promising inhibitors of polyphenol oxidase (PPO), but the effects of carboxyl group and pH on their inhibition effects are still unclear. In this study, methyl cinnamate, cinnamic acid and 4-carboxycinnamic acid were investigated by their inhibitory effects with pH varied from 6.8 to 5.0. Results showed that 4-carboxycinnamic acid had the strongest inhibitory effect on PPO, followed by cinnamic acid and methyl cinnamate. Acidic pH enhanced the inhibitory effects of cinnamic acid and its derivatives on PPO, and the enhancement degree, IC 50 and K i declining degree were followed as 4-carboxycinnamic acid > cinnamic acid > methyl cinnamate. Methyl cinnamate exhibited competitive inhibition on PPO, while cinnamic acid and 4-carboxycinnamic acid exhibited mixed-type inhibition. Inhibitors induced slight changes in the secondary and tertiary structures of PPO, which were enhanced by acidic pH. Molecular docking results showed that 4-carboxycinnamic acid exhibited the strongest binding ability, and the main interaction forces were around carboxyl groups, and acidic pH enhanced the binding effect through more interactions and lower binding energy. This study could provide new insights into industrial application of cinnamic acid and its derivatives for the control of enzymatic browning of fruits and vegetables.

Metal polyphenol network-modified chitin nanocrystals reinforce versatile biomass films: Active packaging, intelligent visualization and controlled photothermal sterilization

Huanle Chen,Chuanhe Tang

The development of food packaging based on natural biomass is a growing focus for researchers and consumers. Beyond the traditional role of food preservation, there is an urgent demand for active packaging and intelligent sensory capabilities. In response, we have developed an integrated system using all-natural materials. Centered on the concept of nanofilled reinforced biomass films, the system integrates active molecules (tannic acid, TA) and an intelligent sensing system (metal polyphenol network, MPN) onto chitin nanocrystals (CNC). This innovation addresses the challenge of compositing CNC with commonly used anionic polysaccharide matrices, represented by sodium alginate (SA). The resulting composite films exhibit unprecedented breakthroughs, including a 30.6% increase in tensile strength and a 136.3% increase in elongation at break, demonstrating improved mechanical properties. In addition, the films possess active packaging features such as antimicrobial, antioxidant, water-blocking, and biosafety properties. Notably, MPN has been integrated into food packaging and is being developed as a multifunctional platform for real-time visual monitoring of food quality and controllable visible light photothermal sterilization for the first time. This platform not only meets the criteria for active packaging and intelligent packaging but also demonstrates that the scientific and rational regulation of the properties of various materials and their combinations, using food-grade raw materials and production technologies, can significantly enhance material performance. This work offers generic reference value for future advances in material performance in food packaging.

One-step assembly of conductive coatings from polyphenol and Nanocarbon-PIL: A versatile approach for fabricating multifunctional sensors

Zhongtao Xu,Yuhan Chen,Kaiyu Li,Yao Zhang,Yuan Liang,Hai Wan,Wenbin Jin,Shuohan Huang,Peng Wei,Yuwei Chen,Yanping Wang,Yong He,Yumin Xia

A highly conductive strategy for even coating carbon nanomaterials is in great demand for various applications. We introduce a conductive modification technique that is universally applicable to a range of substrates. It involves enhancing surface electronegativity by dip-coating various substrates with tannic acid (TA) and ferric chloride (FeCl 3 ) solutions. The process was further refined by the ability of imidazolyl poly (ionic liquid) (PIL-Cl) to disperse carbon black (CB) in water. The conductive modification of various substrate surfaces was facilitated based on electrostatic interactions between the TA-Fe 3+ coating and the CB-(PIL-Cl) dispersion. Meanwhile, to demonstrate the potential of this approach in fabricating materials for wearable sensors, we have fabricated conductive PET fabrics (PTFA@CB). These PTFA@CB fabrics serve as strain sensors, capable of tracking human movement. Additionally, the multi-layer fabric stack design can function as a pressure sensor, providing feedback on pressure coordinates and detecting gripping motions. In addition, the TA-Fe 3+ coating makes PTFA@CB significantly hydrophilic, which improves their responsiveness to humidity. The method described in this paper can be extended to deposit carbon nanomaterials onto various substrates with diverse shapes and properties. The process we developed offers a simple, convenient, and environmentally friendly approach for preparing conductive substrates, with the potential for scalable production.

Polyphenol improve the foaming properties of soybean isolate protein: Structural, physicochemical property changes and application in angel cake

Yi-Lun Wang,Shi-Cheng Dai,Zi-Teng Lian,Xiao-Yi Cheng,Xiao-Hong Tong,Huan Wang,Liang Li,Lian-Zhou Jiang

With the increasing demand for food foaming, how to enhance the foaming properties of protein has gradually become the research focus. This work studied the effect of synephrine (SY) on foaming properties, structure properties, and physicochemical properties of soybean protein isolate (SPI). When the mass ratio of SY to SPI was 1:2, compared with SPI alone, the foam capacity and foam stability of the SY-SPI complex were significantly enhanced. Optical microscopy and confocal laser scanning microscope showed that the improvement in foaming performance was mainly due to the reduction of bubble size and uniform protein distribution. Circular dichroism spectrum and fluorescence spectra indicated that the hydrogen bond of SPI was destroyed and blue shifted with the addition of SY. What's more, the absolute value of Zeta potential, solubility, and hydrophobicity all increased, while the particle size decreased. As a result of molecular docking, surface hydrogen bonds, Van der Waals forces and hydrophobic interactions are the main driving forces. The addition of SY and SPI improved the specific volume and texture of angel cake. This study shows that SY has the potential to be developed into a new type of blowing agent.

Preparation and characterization of lactoferrin-polyphenol conjugate with stabilizing effects on fish oil high internal phase Pickering emulsions

Ying Sun,Mantong Zhao,Zhongyuan Liu,Haohao Shi,Xueying Zhang,Yongqiang Zhao,Zhenhua Ma,Gang Yu,Guanghua Xia,Xuanri Shen

The combination of protein and polyphenol is an effective approach to improve the stability of protein emulsions. The lactoferrin (LF)-(−)-epigallocatechin-3-gallate (EGCG) covalent complex (LF-EGCG) was first prepared by alkali-induced reaction, then the structure and physicochemical properties between LF-EGCG and non-covalent complex (LF + EGCG) were compared, and finally the stability of complexes to fish oil high internal Pickering emulsions (HIPPEs) was tested. Results showed that LF-EGCG had stronger antioxidant activity, higher thermal stability, and better surface wettability than LF + EGCG. Meanwhile, the complexes showed no cytotoxicity within the tested concentration range (12.5–200 μg/mL). The HIPPEs stabilized with LF-EGCG possessed smaller droplet size, higher ζ-potential, and more uniform oil/water proton distribution. Covalent treatment also enhanced the storage, thermal, freeze-thaw and physical stability of LF HIPPEs. Furthermore, due to the higher antioxidant activity and denser microstructure, LF-EGCG HIPPE can more effectively inhibit the oxidation of fish oil.

The gut microbiota-aromatic hydrocarbon receptor (AhR) axis mediates the anticolitic effect of polyphenol-rich extracts from Sanghuangporus

Shi Zhong,Yu-Qing Sun,Jin-Xi Huo,Wen-Yi Xu,Ya-Nan Yang,Jun-Bo Yang,Wei-Jie Wu,Yong-Xin Liu,Chong-Ming Wu,You-Gui Li

Inflammatory bowel disease (IBD) is a significant global health concern. The gut microbiota plays an essential role in the onset and development of IBD. Sanghuangporus (SH), a traditional Chinese medicinal mushroom, has excellent anti-inflammatory effects and is effective at modulating the gut microbiota. Despite these attributes, the specific anticolitic effects of SH and the mechanisms through which the gut microbiota mediates its benefits remain unclear. Herein, we demonstrated that polyphenol-rich extract from SH effectively alleviated the pathological symptoms of dextran sodium sulfate (DSS)-induced colitis in mice by modulating the gut microbiota. Treatment with SH distinctly enriched Alistipes , especially Alistipes onderdonkii , and its metabolite 5-hydroxyindole-3-acetic acid (5HIAA). Oral gavage of live A. onderdonkii or 5HIAA potently mitigated DSS-induced colitis in mice. Moreover, both 5HIAA and SH significantly activated the aromatic hydrocarbon receptor (AhR), and the administration of an AhR antagonist abrogated their protective effects against colitis. These results underscore the potent efficacy of SH in diminishing DSS-induced colitis through the promotion of A. onderdonkii and 5HIAA, ultimately activating AhR signaling. This study unveils potential avenues for developing therapeutic strategies for colitis based on the interplay between SH and the gut microbiota. Graphical Sanghuangporus is a medical mushroom with potent anticolitic effects. It modulates the gut microbiota and specifically enriches Alistipes onderdonkii , which can produce 5-hydroxyindole-3-acetic acid, thus activating the aryl hydrocarbon receptor pathway and ameliorating colitis. Highlights Sanghuangporus (SH) polyphenol extract potently alleviated symptoms of colon colitis in dextran sodium sulfate-induced colitis mice. The gut microbiota especially Alistipes onderdonkii mediates the anticolitic effect of SH. The SH-enriched microbial metabolite 5-hydroxyindole-3-acetic acid exhibited an anticolitic effect via the activation of the aryl hydrocarbon receptor. The “gut microbiota-metabolites-signaling pathway” provides an effective perspective for the study of complex Chinese medicine systems.

Regulatory T Cells Engineered with Polyphenol-Functionalized Immunosuppressant Nanocomplexes for Rebuilding Periodontal Hard Tissue under Inflammation-Challenged Microenvironment

Bo Zhang,Guidong Gong,Yunxiang He,Jialing Liu,Bo Wang,Yifei Li,Jie Fang,Zhihe Zhao,Junling Guo

Global aging heightens the risk of oral disorders, among which periodontitis is the major cause of tooth loss in the aging population. The regeneration of damaged periodontal hard tissue is highly challenging due to the existence of the refractory local inflammation. Owing to the potent anti-inflammatory capabilities, regulatory T cells hold great promise in immunotherapies for tissue regeneration. However, the transferred regulatory T cells can alter their phenotypes and functions in local inflammatory milieu, significantly impairing their therapeutic efficacy. Herein, we introduce a novel regulatory T cell-based nanobiohybrid system bearing polyphenol-functionalized rapamycin nanocomplexes. The sustained in situ release of immunosuppressant rapamycin from the cell-attached nanocomplexes maintains the anti-inflammatory phenotype of regulatory T cells in the inflammatory milieu. The synergistic actions of the anti-inflammatory cytokines secreted and the immunosuppressant released guide a pro-resolving polarization of macrophages and enhance osteogenic differentiation of bone marrow-derived stromal cells. The stabilized phenotype of the regulatory T cells dramatically promoted the resolution of periodontal inflammation and the repair of the hard tissue (alveolar bone) in vivo . Overall, these studies highlight a potent regulatory T cell-based nanobiohybrid therapy to treat periodontitis by modulating periodontal immune microenvironment.

Two-electron oxidized polyphenol chemistry-inspired superhydrophilic drug-carrying coatings for the construction of multifunctional nasolacrimal duct stents

Wenzhe Xiao,Binjian Wang,Xuemei Wang,Guanghong Zhang,Yihao Zhao,Zezhen Zhang,Linhua Li,Chao Qu

Nasolacrimal duct obstruction due to infection, inflammation, or excessive fibroblast proliferation may result in persistent tearing, intraocular inflammation, or even blindness. In this study, surface engineering techniques are applied to nasolacrimal duct stents for the first time. Based on the functioning of marine mussels, “one-pot” and “stepwise” methods were employed to construct a novel multifunctional superhydrophilic PDA/RAP coating using dopamine and rapamycin. Micron-sized rapamycin crystals combined with nano-sized polydopamine particles form a micro–nano topographical structure. Therefore, acting synergistically with in situ-generated hydrophilic groups (amino, carboxyl, and phenolic hydroxyl), they impart excellent and long-lasting superhydrophilicity to the nasolacrimal duct stent. The PDA/RAP coating effectively maintained the stability of the initial microenvironment during stent implantation by inhibiting the onset of acute inflammation and infection during the early stages of implantation. Meanwhile, the rapamycin crystals, supported by the superhydrophilic platform, exhibited a sustained-release capability that helped them to better exert their anti-inflammatory, antibacterial, and anti-fibroblast proliferative properties, ensuring conducive conditions for the rapid repair of nasolacrimal duct epithelial cells, verified by a series of experiments. In conclusion, the PDA/RAP hydrophilic coating has anti-inflammatory, antifibrotic, antibacterial, and antithrombotic properties, offering a new strategy to address restenosis following clinical nasolacrimal duct stent implantation.

Ultra-Long-Term Anti-Inflammatory Polyphenol Capsule to Remodel the Microenvironment for Accelerating Osteoarthritis Healing by Single Dosage

Shaoyin Wei,Zeyu Shou,Dong Yang,Linxiao Sun,Yan Guo,Yang Wang,Xingjie Zan,Lianxin Li,Chunwu Zhang

Osteoarthritis (OA) is a common chronic inflammatory disease that leads to disability and death. Existing therapeutic agents often require frequent use, which can lead to drug resistance and long-term side effects. Polyphenols have anti-inflammatory and antioxidant potential. However, they are limited by their short half-life and low bioavailability. This work presents a novel pure polyphenol capsule for sustained release of polyphenols, which is self-assembled via hydrophobic and hydrogen bonds. The capsule enhances cellular uptake, scavenges reactive oxygen and nitrogen species, reduces inflammatory markers, and remodels the OA microenvironment by inhibiting the p38 MAPK pathway. The capsule overcomes the limitations of short half-life and low bioavailability of polyphenols and achieves single-dose cure in mouse and dog OA models, providing an optimal therapeutic window for OA repair. Taking advantage of simple manufacturing, convenient administration, and pure polyphenol composition, these capsules show great potential for clinical treatment of osteoarthritis and chronic inflammatory diseases.

A sprayable alginate hydrogel based on copper-tannic acid metal-polyphenol for promoting acute and infected wound healing

Ruigang Zhou,Mengyan Zhou,Weimei Wang,Junjie Huang,Weilong Peng,Lianggang Wang,Jun Chen,Ruonan Bo,Mingjiang Liu,Jingui Li

Bacterial-infected wounds may lead to delayed repair due to increased inflammation, especially wounds infected by drug-resistant bacteria, which remains a major clinical challenge. Sprayable hydrogels can treat large and irregular dynamic wounds and have attracted significant attention as wound dressings. Herein, sprayable hydrogels were formed by rapid crosslinking of atomized sodium alginate (SA) and high-concentration calcium ion solution, and copper-tannic acid metal-polyphenol network (CuTA) was loaded as the active ingredient. The hydrogel system achieved rapid crosslinking (10 s) through interaction with 10 % CaCl₂ solution, while deep eutectic solvent (DES) enhanced moisturizing capacity. In vitro studies demonstrated the broad-spectrum antibacterial effect of DES + Gel/CuTA sprayable hydrogel, with an inhibition rate of over 99 %. The excellent hemostatic performance of DES + Gel/CuTA sprayable hydrogel was also demonstrated, with the bleeding volume being less than 0.2 g, which was lower than 0.46 g in the control group. In three animal models of acute, infected and burn-infected skin wounds, DES + Gel/CuTA sprayable hydrogel showed significant effects in promoting wound healing. In conclusion, the developed sprayable hydrogels offered several advantages, including antibacterial and anti-inflammatory properties and promoting key stages of wound healing, such as cell migration, M2 polarization, collagen deposition, and angiogenesis. This sprayable hydrogel can resist infection and promote healing, showing its great potential in treating infected wounds.

Ultrasonic regulation of β-lactoglobulin/puerarin complex: structural modification, digestive properties, and allergenicity suppression through controlled polyphenol interactions

Peng Man,Jinghua Pei,Sijia Chen,Xiue Han,Huajiang Zhang

This study explored the effects of ultrasound on β-lactoglobulin (β-LG)/puerarin (PUE) complexes, focusing on structural, digestive, and allergenic properties. Molecular simulations revealed hydrogen bonding as the primary interaction between PUE and β-LG. Ultrasound reduced β-LG crystallinity (via XRD), further enhanced by PUE encapsulation. Moderate ultrasound (300 W) increased thermal denaturation temperature by 8.2 °C (DSC) and enhanced digestibility to 78.3 ± 1.2 % (vs. 62.5 ± 2.1 % in untreated group), while excessive power (500 W) caused structural damage. Ultrasonic treatment increased the β-sheet content of β-LG, led to a reorganization of its tertiary structure. Allergenicity assays demonstrated significant reductions in IgE and IgG binding capacity for 300 W-treated complexes. These findings highlighted moderate ultrasound (300 W) as an effective strategy to modulate functionality and reduce allergenicity in protein-polyphenol colloids, offered insights for food science applications.
