Mitochondrial dysfunction of nucleus pulposus cells (NPCs) drives IDD. • An injectable liposome-anchored ECM-mimetic hydrogel (ILH) is developed. • ILH repairs NPC mitochondria via SIRT1-AMPK-PGC-1α activation and ROS scavenging. • ILH reverses NPC senescence to restore disc structure and function. Mitochondrial dysfunction is a key driver of nucleus pulposus cell (NPC) senescence with crucial roles in orchestrating intervertebral disc (IVD) degeneration. However, the development of effective therapies is limited by the complex IVD structure and mitochondrial pathology. Here, an injectable liposome-anchored extracellular matrix (ECM)-mimetic hydrogel (ILH) was engineered for treating IVD degeneration via restoring mitochondrial biogenesis in senescent NPCs. Specifically, nicotinamide mononucleotide (NMN)-loaded liposomes were functionalized with the mitochondria-targeting peptide SS31 (MTLs) and incorporated together with Vitamin K 2 (VK 2 ) into an injectable hyaluronic acid-gelatin hydrogel crosslinked via dynamic Schiff base and boronate ester bonds. ILHs provide immediate mechanical support to degenerative IVDs while enabling on-demand release of encapsulated MTLs and VK 2 within the nucleus pulposus (NP). MTLs facilitates mitochondrial targeted-NMN delivery to replenish intracellular NAD + levels and activate SIRT1 signaling in senescent NPCs, thereby promoting AMPK-dependent mitochondrial biogenesis and suppressing the p53-associated senescence programs. On the other hand, SS31 alleviates cardiolipin peroxidation and preserves mitochondrial cristae integrity, while VK 2 activates the antioxidative Nrf2/HO-1 pathway to further improve mitochondrial quality control. Collectively, these coordinated effects enable comprehensive mitochondrial repair while further maintaining their structural and functional integrity in senescent NPCs, providing a promising therapeutic strategy for clinical treatment of IVD degeneration.

Acute kidney injury (AKI) causes renal tubular damage, driven primarily by mitochondrial dysfunction and reactive oxygen species (ROS)-mediated oxidative stress, leading to a cellular energy crisis. The physiological architecture of the kidney hampers targeted drug delivery, rendering metabolic restoration a therapeutic challenge. To address this, we developed a second near-infrared (NIR-II) light-driven, bioenergetic nanoplatform (UCTR) that leverages upconversion nanoparticle (UCNPs)-enhanced photosynthesis for energy replenishment and mitochondrial repair in AKI. The UCTR consists of thylakoid membrane (TM)-encapsulated UCNPs cloaked with activated renal tubular epithelial cell membranes (RECM), enabling targeted accumulation in injured tubules. The UCNPs convert deep-tissue-penetrating NIR-II light into visible wavelengths, activating photosynthetic adenosine triphosphate (ATP) and nicotinamide adenine dinucleotide phosphate (NADPH) synthesis within the TM component under irradiation. This exogenous bioenergetic supply mitigates hypoxia-induced mitochondrial energy deficits, while the AMPK/PGC-1α pathway is simultaneously activated to restore the mitochondrial membrane potential. Moreover, UCTR synergizes the antioxidative and anti-inflammatory effects of plantain, accelerating tubular repair.

Head and neck squamous cell carcinoma (HNSCC) remains a clinically challenging malignancy due to limited therapeutic options and considerable treatment-related toxicity. In this study, we present an innovative nanotherapeutic platform—monocarboxylate transporter 1 (MCT1) antibody-targeted nanoparticles co-loaded with a second near-infrared window (NIR-II, 1000–1700 nm) absorbing conjugated polymer (PDTP-TBZ-OD, denoted as PTO) and emodin (MCT@PTO-Em NPs) —that synergistically integrates emodin-induced pyroptosis with NIR-II mediated mild photothermal therapy (MPTT) to address critical limitations of current HNSCC treatments strategies. The synthesized NIR-II-absorbing conjugated polymer PTO exhibits strong absorption at 1064 nm and a high photothermal conversion efficiency (39 %), enabling deeptissue penetration and precise hyperthermia control. Accordingly, MCT@PTO-Em NPs implement a dual-modal therapeutic strategy that triggers pyroptosis through NLRP3 inflammasome activation while simultaneously enhancing tumor cell death via mild hyperthermia. Moreover, emodin encapsulated within the nanoparticles (NPs) inhibits heat-shock protein 70 (HSP70), thereby disrupting cellular thermoresistance mechanisms and sensitizing cancer cells to MPTT. Additionally, surface functionalization of the nanoparticles with MCT1-targeting antibodies enhances tumor-specific delivery and intracellular accumulation, thereby minimizing off-target effects. Both in vitro and in vivo studies demonstrate that MCT@PTO-Em NPs achieve superior tumor suppression through the synergistic interplay between pyroptosis and MPTT, while exhibiting excellent biocompatibility. This work presents a precision nanomedicine strategy that integrates molecular targeting, pyroptosis-mediated immune activation, and MPTT, offering a transformative therapeutic paradigm for HNSCC and other solid tumors.

Combining efficient photocatalytic activity with precise targeting is crucial for covalent organic framework (COF)-based nanomedicines to remodel the immunosuppressive microenvironment and sensitize “cold” tumors to immunotherapy. Herein, we constructed two isomorphic frameworks featuring distinct electronic topologies: a zwitterionic squaric acid (SA) COF with inherent electrostatic asymmetry, and a charge-neutral terephthaldehyde (TA) COF. The spatial divergence of ion centers in SA COF triggers intense molecular-level ionic displacement polarization, creating a robust internal electric field. Such unique field-effect regulation accelerates photogenerated charge separation for efficient superoxide anion production, enhances intersystem crossing for singlet oxygen generation, and promotes nonradiative energy relaxation for high-performance photothermal conversion under dual-wavelength irradiation. After PEGylation and iRGD modification, the formulated SA@PEG-iRGD exhibits excellent physiological stability and active deep tumor penetration. With near-infrared absorption, it enables high-contrast photoacoustic imaging for guided dual-wavelength (660/808 nm) phototherapy. This spatiotemporally controlled therapeutic cascade triggers lethal ferroptosis and precise photothermal ablation, potently activating immunogenic cell death. Combined with aPD-L1 therapy, it eliminates primary tumors, produces abscopal effects, and establishes immune memory against recurrence and metastasis. This study highlights the value of electronic structure engineering in COF-based precision nanomedicine.

Inorganic lanthanide nanomaterials as photoluminescent biolabels have attracted increasing attention due to their superior physicochemical properties. However, unstable conjugation of inorganic lanthanide nanomaterials with biological function units (such as antibodies) induces instability of conjugated complexes in aqueous solution, limiting their clinical application. In this study, we developed a rapid point-of-care testing (POCT) platform strategy based on coordination-enhanced time-resolved luminescence of specially nanostructural lanthanide particles for lateral flow immunoassay (CE-TRFIA). This strategy integrates a nanoprobe via a dendritic mesoporous silica nanosphere (DMSN) loading a large amount of ultrasmall amorphous europium oxide (Eu2O3) nanoparticles, which rapidly dissolve to release Eu3+ cations under neutral pH value and form luminescent complexes with photosensitizers (such as β-NTA and TOPO) in an LFIA system. This innovative strategy achieves high-sensitivity detection and long-term stability primarily through high-loading probes, excellent dissolution enhancement, stable covalent coupling, and time-resolved detection. With Procalcitonin (PCT) antigen selected as the detection sample, this approach achieves high-sensitivity detection of PCT with a limit of detection (LoD) as low as 1.9 pg/mL, significantly lower than that of commercial LFIA (0.1 ng/mL), and excellent clinical correlation (r = 0.989). The method offers chemiluminescence-level sensitivity without the need for large instruments while retaining the real-time detection characteristics of LFIA. Our results highlight CE-TRFIA as a highly sensitive, specific, and rapid POCT solution for detecting low-abundance biomarkers such as PCT, enhancing the diagnostic capabilities of traditional LFIA and offering significant potential for ultrasensitive and rapid clinical diagnostics.
