Synthesizing highly compatible photosensitizer provides new directions for constructing high-efficiency photoinitiation systems (PSs). Here, a 2,4-Bis(4-(diethylamino)benzylidene)cyclobutanone (C4) was synthesized and used to form a three-component PSs (C4/HABI/MBO) with a biimidazole (HABI), 2-mercaptobenzoxazole (MBO). The photosensitizer has a more efficient energy transfer properties attributed to the higher fluorescence emission capacity, narrower energy band gap, lower free energy of electron transfer (Δ G et ) and improved matching with photoinitiator. Meanwhile, a double-cycle reaction mechanism is proposed, in which the improved matching seems to encourage this cycle to occur. As a result, the C4/HABI/MBO photoinitiation system well improved the diffraction efficiency of photopolymers up to 96% and demonstrated promising performance in recording and reproducing reflection holograms, which provide a new research idea for designing photoinitiation system.

One novel nanoplatform with multiple functions including Raman imaging and photodynamic therapy (PDT) capacities was constructed through modifying nanodiamonds (NDs) with photosensitizer chlorin e6 (Ce6). The NDs-Ce6 nanoparticles show enhanced singlet oxygen generation efficiency relative to free Ce6. Cytotoxicity tests indicate that NDs-Ce6 have negligible influence toward HeLa cells vitality under dark condition but enhanced photodynamic ablation upon 660 nm laser irradiation in comparison with free Ce6. In addition, the NDs-Ce6 could be used as Raman imaging probes toward HeLa cells. These results demonstrate that the NDs-Ce6 multifunctional nanoplatform have attractive features using for Raman imaging and PDT. Additionally, a new idea could be provided for designing the multifunctional platform from the work.

Photodynamic therapy (PDT) is an emerging method for the treatment of cancer. Combination of PDT and chemotherapy is a hot topic though the two therapies could not simultaneously exert their perfect effect in vivo. Here we report a doxorubicin-loaded photosensitizer-core pH-responsive copolymer nanocarrier with high tumor targeting and anticancer effects due to integration of PDT with chemotherapy. The pH-responsive photosensitizer-core four-armed star-shaped copolymer, [methoxy-poly(ethylene glycol)-poly(2-(N,N-diethylamino)ethyl methacrylate)-poly(ε-caprolactone)]4-zinc β-tetra-(4-carboxyl benzyloxyl)phthalocyanine (PDCZP), was prepared, which was a molecular spherical nanocarrier in aqueous media. The carriers changed from small at high pH to large at low pH (51, 105, and 342 nm at pH 7.4, 6.5, and 5.0, respectively) and the zeta potential gradually increased (7.15, 16.2, and 26.1 mV at the above pH, respectively). PDCZP had a longer emission wavelength (max. 677 nm) than the parent photosensitizer, favoring light penetration through biological tissues. The singlet oxygen (1O2) quantum yield of PDCZP was 0.41. Doxorubicin (DOX) showed rapid release from PDCZP in the acidic media. More importantly, the drug-loaded nanocarriers showed the better in vitro and in vivo anticancer effects under lighting on MCF-7, SW480 cells and HepG2 cells and the murine hepatocellular carcinoma H22 models than the other groups. PDCZP showed a high tumor targeting effect based on the enhanced permeation and retention effect and its small size. The photosensitizer-core nanocarrier is a promising photodynamic nanocarrier for integrating other therapies.

The skin microbiota is an inseparable component of the skin barrier structure, which participates in the stabilization or impairment of the barrier function as well as the development of many skin diseases. In order to maintain the stability of skin bacteria community structure, people pay more and more attention to protecting probiotics, such as bacilli, and killing pathogens, such as Gram-positive cocci. We herein report a negatively charged photosensitizer TPA-3NBA with aggregation-induced emission (AIE) characteristics, which was synthesized by one-step reaction, can selectively and rapidly stain (within 0.5 min) and effectively kill Gram-positive cocci (killing 6.0 log 10 of Staphylococcus aureus at a concentration of 1 μM) without affecting bacilli under white light irradiation. This selective bacterial imaging makes it possible to quickly identify locally bacterial infection. Furthermore, the normal concentration of divalent ion (Ca 2+ ) in human body can significantly promote the absorption of TPA-3NBA by Gram-positive cocci, thus improving its antibacterial activity. To the best of our knowledge, this is the first photosensitizer to be reported with good performance in selective bacterial imaging and removal of Gram-positive pathogenic cocci, indicating its potential application prospect in targeted antimicrobial therapy.

Fluorescence imaging-guided tumor photodynamic therapy (PDT) has received increasing attention due to its higher resolution of fluorescence imaging and noninvasive treatment of PDT. Two problems of fluorescence aggregation-caused quenching (ACQ) and tumor microenvironment hypoxia are urgently resolved in traditional PDT with fluorescence imaging function, but there is still a lack of effective molecular engineering to design delicate photosensitizers (PSs) to overcome fluorescence ACQ and tumor hypoxia, simultaneously. Herein, we develop a feasible molecular engineering to design aggregation-induced emission (AIE)-active PSs with unique free radical (type I) reactive oxygen species (ROS) by researching molecular structure-property relationship, which possesses bright fluorescence at aggregation and low O 2 -dependent under the process of generating free radical ROS. The intersystem crossing (ISC) channel is activated when frequently enhance intramolecular charge transfer (ICT) effect in electron-rich AIEgens with typical heavy atoms, which ensure enough triplet energy generation to induce superoxide anion free radical (O 2 − ·) generation. In vitro fluorescence imaging and photo triggering biotoxicity evaluation both revealed that new AIE-active photogenerator is a promising candidate for fluorescence imaging-guided PDT.

A novel reduction-responsive porphyrin monomer bearing a disulfide bond (TPPC6SAM) was synthesized, and then it was utilized to construct a series of POEGMA- b -PTPPC6SAM block copolymers via reversible addition–fragmentation transfer (RAFT) polymerization with water-soluble and biocompatible monomer, oligo(ethylene glycol) methyl ether methacrylate (OEGMA). POEGMA- b -PTPPC6SAM block copolymers could self-assemble into spherical micelles and vesicles by varying the mass ratio of hydrophilic block to hydrophobic block. Confocal laser scanning microscopy (CLSM) and flow cytometry showed that POEGMA- b -PTPPC6SAM micelles could effectively accumulate in A549 cells, compared to that of free porphyrin. In addition, minimal dark toxicity and efficacious photo-toxicity in vitro were also evaluated by MTT assay . According to the result of MTT assay, POEGMA- b -PTPPC6SAM block copolymers had a higher photo-toxicity with a lower IC50 (below 3 μg/mL). Thus, reduction-responsive POEGMA- b -PTPPC6SAM block copolymers could well improve the release of porphyrin photosensitizers and enhance the efficiency of PDT.

Photosensitizers (PSs) play a vital role in photodynamic therapy (PDT) for combating bacterial resistance and treating tumor. In this study, we report new donor–acceptor porphyrin PSs with a cationic conjugated oligomer (OPV) as a donor unit and porphyrin (TPP) as an acceptor unit by covalent linkage and achieved a fluorescence resonance energy transfer efficiency of 99% owing to their strong spectral overlap and short distance. The 1O2 yield of porphyrin derivatives is 121% (rose bengal as the standard reference) by virtue of OPVs’ excellent light-harvesting ability and high fluorescence resonance energy transfer efficiency, greatly exceeding those of oligomer and porphyrin derivatives reported in the literature. Additionally, the cationic donors significantly improved the water solubility, decreased the aggregation of porphyrin, and promoted the adherence of the PSs to cell membranes through electrostatic interactions. As a result, the D–A porphyrin PSs exhibit dramatic PDT treatment efficiency. The half-inhibitory concentration is as low as 33 and 88 nM for methicillin-resistant Staphylococcus aureus and Escherichia coli, respectively. Therefore, this study provides a new strategy to construct PSs with high 1O2 yield and an excellent treatment effect at a low dose of PSs, which is promising for application in PDT used to treat cancer and microbial infections.

Despite the increased interest of visible-light-absorbing compound Hypericin (Hyp) in photodiagnosis, photocatalysis, and photodynamic therapy (PDT) applications, a major obstacle still exists; i.e., the photoactivity is diminished due to the facile aggregation of Hyp in aqueous environment that induces excited-state quenching. Herein, we explore the excited-state property of Hyp bound to the DNA G-quadruplex by combining multiple steady-state and time-resolved spectroscopy. We find that the aggregation-induced quenching effect can be successfully prevented by appropriate G-quadruplex binders that disperse Hyp into monomer. The binding of Hyp/G-quadruplex is selective, however, exhibiting a preferential binding toward parallel G-quadruplexes (c-kit2, C14B1, STAT3, S50, and PS2.M), over antiparallel or hybrid G-quadruplex (Tel22, TBA). The excited-state property of Hyp is highly related to the binding behavior, showing a consistent trend that the better the Hyp/G-quadruplex binding, the longer the triplet 3Hyp* lifetime and the higher the efficiency to produce 1O2. For Hyp/c-kit2, the major binding mode is 5′-end stacking, which offers protection from collisional quenching reactions and ensures a stable photocycle of 3Hyp*-O2 energy transfer forming 1O2, leading to the highest 1O2 quantum yield (0.67) with superior photostability. These findings open possibilities of developing Hyp/G-quadruplex complex as a biocompatible photosensitizer for PDT applications, etc.

Photodynamic therapy (PDT), utilizing a photosensitizer (PS) to induce tumor cell death, is an effective modality for cancer treatment. PS–peptide conjugates have recently demonstrated remarkable antitumor potential in preclinical trials. However, the limited cell membrane binding affinity and rapid systemic clearance have hindered their transition to clinical applications. To address these challenges, we investigated whether in vivo covalent chemistry could enhance tumor accumulation and potentiate antitumor efficacy. Specifically, we synthesized a PS–peptide conjugate termed P-DBCO-Ce6, with chlorin e6 (Ce6) and dibenzocyclooctyne (DBCO) conjugated to a negatively charged short peptide. By employing metabolic glycoengineering and bioorthogonal reactions, P-DBCO-Ce6 achieves covalent bonding to the cell membrane, enabling prolonged retention of the PS on the cell surface and the in situ generation of reactive oxygen species (ROS) on cell membranes to kill tumor cells. In vivo studies demonstrated a 3.3-fold increase in tumor accumulation of the PS through bioorthogonal reactions compared to the control group, confirming that click chemistry can effectively enhance PS tumor accumulation. This approach allows for the effective elimination of tumors with a single treatment. The improved efficiency of this strategy provides new insights into the design of PDT systems for potential clinical applications.

“Molecular science” has long been regarded as the golden rule to guide the design of organic materials’ performances in the past many years, but some interesting phenomena of conventional aggregation-caused fluorescence quenching and new aggregation-induced emission reflect that materials’ properties would be changed from “molecule” to “aggregate”. Therefore, “molecular science” theory faces certain limitations to guide regulating the performance of materials at aggregation. In this work, it is discovered that the photosensitizer's performances contain fluorescence and reactive oxygen species, which could be affected by changing molecular atoms and aggregation form. The introduction of oxygen and selenium atoms could redshift fluorescence and improve reactive oxygen species (ROS) efficiency. In addition to the atomic effect, the ROS efficiency of photosensitizers could be affected after coating a polymeric shell, that is, the production of type II ROS singlet oxygen ( 1 O 2 ) is suppressed, while the type I ROS of superoxide anion (O 2 −• ) is improved. This work discovers that the fluorescence and ROS efficiency of photosensitizers are relevant to the atomic effect and polymeric aggregation effect, and discussing deeply the influence mechanism, which has important research significance for modulating precisely the performances of photosensitizers and promoting the development of type I photodynamic therapy.

Skin is the initial defense against infection of the body but becomes vulnerable when damaged. Subsequent bacterial infections can cause more severe ulcers. Antibiotics are useful for infected wound healing but can also speed up the evolution of antibiotic-resistant bacteria. Dressing and antibacterial photodynamic therapy (PDT) are effective methods for protecting and treating wounds, respectively. Herein, we developed a spray hydrogel based on sodium alginate hydrogels, loaded with methylene blue (MB) and closo-dodecaborate cluster (closo-B12H122–), for physical protection and antibacterial PDT. Among them, MB acted as the photosensitizer, which can produce reactive oxygen species (ROS) through the PDT process to implement the therapeutic effect toward bacterial-infected wounds. closo-B12H122– and MB can form a supramolecular photosensitizer (MB-B12), which can reduce the dark toxicity of the MB photosensitizer without significantly affecting its PDT effect. Moreover, the introduction of closo-B12H122– can upgrade the physical property of the spray hydrogel for better wound protection and can also increase the MB loading capacity for better wound healing. The hydrogel demonstrated an outstanding antibacterial ability of around 99% against Staphylococcus aureus and 95% for Escherichia coli. This PDT-spray hydrogel system can be a favorable option for protecting and treating bacterial-infected wounds.

Infected wounds are highly susceptible to bacterial contamination, which not only delays the healing process but also leads to a range of complications. NO is one of the most widely used gas molecules in wound treatment strategies due to its potent antibacterial and healing-promoting properties. However, excessive NO in infected wounds can damage cellular components and exacerbate inflammation. Therefore, precise control of NO dosage and targeted delivery to the wound site are crucial for effective treatment. Herein, a cationic hydrogel was constructed using modified guar gum and polyvinyl alcohol to enhance antibacterial effects, with guanidine-modified chitosan (Met-CS) and the photosensitizer sodium copper chlorophyllin (SCC) incorporated to create a light-triggered NO release system. Both in vitro and in vivo experiments confirmed that the proposed hydrogel exhibits excellent antibacterial performance, anti-inflammatory effects, and wound healing capabilities, demonstrating its potential for precise therapeutic applications.
