Curcumin (Cur) is a promising drug for neurological diseases. Nevertheless, the application of Cur has been limited due to its difficulty in penetrating blood-brain barrier (BBB). Intranasal drug delivery, a noninvasive alternative delivery of Cur, can effectively help Cur cross BBB and inert into central nervous system directly. Odorranalectin (OL) which is the smallest lectin can prolong the residence time of Cur in the nasal mucosa and promote cellular uptake. In this work, a nasal delivery system incorporating OL modified Cur-loaded nanoparticles (Cur–OL–NPs) was developed and expected to bypass BBB and promote the absorption of Cur. We conjugated OL to polyethylene glycol-poly (lactic-co-glycolic acid) (PEG–PLGA), and combined polyethylene glycol-poly (γ-benzyl-L-glutamate) (PEG–PBLG) and OL–PEG–PLGA to prepare nanoparticles to improve the stability, bioavailability and targeting of Cur. Compared with unmodified NPs, increased efficiency of Cur–OL–NPs cellular uptake by Calu-3 cells had been obtained with no severe toxicity. Furthermore, in vivo pharmacokinetic studies also showed that Cur–OL–NPs had higher relative bioavailability. Thus, it is concluded that the results indicated that OL–NPs as carriers of Cur had a promising future in nasal drug delivery system.

Acrolein (ACR) is a toxic unsaturated aldehyde that is formed during different steps of thermal food processing. Here, we explored the kinetics of curcumin and ACR and elucidated the pathway of curcumin trapping ACR by preparing a mono-adduct of ACR (CMA-1) conjugated with curcumin. The synergistic scavenging effect and mechanism of curcumin combined with quercetin on ACR was analyzed by liquid chromatography–tandem mass spectrometry (LC–MS/MS). Comparing the uses of curcumin and quercetin both individually and in combination, we found that quercetin in combination resulted in more curcumin being transformed into CMA-2, while curcumin in combination made the amount of di-ACR conjugated to quercetin (QDA) increase. We also added combined curcumin and quercetin into grilled chicken wings to demonstrate that curcumin and quercetin could scavenge ACR by forming their own ACR adducts and antioxidant activity during the process. Our results have noted a new strategy, in which some combinations of dietary polyphenols might contribute to the removal of toxic ACR produced during thermal food processing.

There is growing interest in developing biomaterial-coated liposome delivery systems to improve the stability and bioavailability of curcumin, which is a hydrophobic nutraceutical claimed to have several health benefits. The curcumin-loaded rhamnolipid liposomes (Cur-RL-Lips) were fabricated from rhamnolipid and phospholipids, and then chitosan (CS) covered the surface of Cur-RL-Lips by electrostatic interaction to form CS-coated Cur-RL-Lips. The influence of CS concentration on the physical stability and digestion of the liposomes was investigated. The CS-coated Cur-RL-Lips with RL:CS = 1:1 have a relatively small size (412.9 nm) and positive charge (19.7 mV). The CS-coated Cur-RL-Lips remained stable from pH 2 to 5 at room temperature and can effectively slow the degradation of curcumin at 80 °C; however, they were highly unstable to salt addition. In addition, compared with Cur-RL-Lips, the bioavailability of curcumin in CS-coated Cur-RL-Lips was relatively high due to its high transformation in gastrointestinal tract. These results may facilitate the design of a more efficacious liposomal delivery system that enhances the stability and bioavailability of curcumin in nutraceutical-loaded functional foods and beverages. Keywords:liposomes;chitosan;curcumin;rhamnolipids;stability;in vitro digestion

This study investigated the chitosan (CS)-octenyl succinic anhydride (OSA) starch complex as a stabilizer for Pickering emulsions. The effects of OSA concentration (0.5wt%-2.5wt%) and viscosity (OSA1, OSA2, OSA3) on the stability and physiochemical properties of emulsion were examined. The results showed that CS-OSA with 2.0wt% concentration OSA stabilized emulsion exhibited optimal emulsifying capacity. Contact angle analysis, Fourier transform infrared spectrometer (FTIR) and X-difference diffractometer (XRD) were utilized to explore the stabilized mechanism of CS-OSA on emulsion, results confirmed the CS and OSA interactions, and strong adsorption of CS-OSA particles at the oil-water interface. Furthermore, CS-OSA stabilized emulsion loading curcumin were assessed, results showed that curcumin loaded emulsion exhibited high stability and better antioxidant capacity than free CS during 10-day storage. The overall study offers valuable insights into the utilization of CS-OSA emulsifiers in delivery application for lipophilic bioactive molecules.

The aim of this research was to determine chemical composition and repellent and insecticidal activities of the essential oil of Kaempferia galanga L. rhizomes against the booklouse, Liposcelis bostrychophila Badonnel, and to isolate insecticidal or repellent constituents from the oil. The essential oil was obtained by hydrodistillation and analyzed by gas chromatography–mass spectrometry. Twenty-eight components of the oil were identified. The major compounds in the oil were ethyl-ρ-methoxycinnamate (38.6%), ethyl cinnamate (23.2%), 1,8-cineole (11.5%), trans-cinnamaldehyde (5.3%), and borneol (5.2%). Based on bioactivity-guided fractionation, four active constituents were isolated from the oil and identified as 1,8-cineole, ethyl cinnamate, ethyl ρ-methoxycinnamate, and trans-cinnamaldehyde. The essential oil exhibited contact toxicity against the booklouse with an LC50 value of 68.6 μg/cm2. Ethyl cinnamate (LC50 = 21.4 μg/cm2) exhibited stronger contact toxicity than ethyl ρ-methoxycinnamate and trans-cinnamaldehyde (LC50 = 44.6 and 43.4 μg/cm2, respectively) while 1,8-cineole showed weak acute toxicity. The essential oil also possessed fumigant toxicity against the booklouse with a LC50 value of 1.5 mg/liter air. 1,8-Cineole and trans-cinnamaldehyde (LC50 = 1.1 and 1.3 mg/liter, respectively) possessed stronger fumigant toxicity against the booklouse than ethyl cinnamate and ethyl ρ-methoxycinnamate (LC50 = 10.2 and 10.2 mg/liter air, respectively). trans-Cinnamaldehyde was strongly repellent to booklice, whereas ethyl cinnamate and ethyl ρ-methoxycinnamate were weakly repellent and 1,8-cineole did not repel booklice. The results indicate that the essential oil and its constituent compounds have potential for development into natural insecticides or fumigants and repellents for control of insects in stored grains.

A novel polysaccharide (ZOP) was extracted from Zingiber officinale with ultrasonic assisted extraction method. ZOP monosaccharide composition and mole ratio is GlcA: GalA: Glc: Gal: Ara = 1.97:1.15:94.33:1.48:1.07. Then, the particle size of ZOP-NPs prepared by nano-precipitation method was 230.5 nm, and the polydispersity index (PDI) was 0.260. Using ZOP and ZOP-NPs as reductants and stabilizers, ZOP-AgNPs and ZOP-NPs-AgNPs were prepared. They were characterized by ultraviolet-visible spectrophotometer (UV-Vis), fourier transform infrared spectroscopy (FT-IR), scanning electron microscope (SEM), transmission electron microscope (TEM), and X-ray diffraction (XRD). The silver chelation rate of polysaccharide silver nanoparticles (AgNPs) ranged from 68.70 to 82.12%. ZOP-AgNPs (0.5%, w/v; 1%, w/v) and ZOP-NPs-AgNPs (0.5%, w/v; 1%, w/v) exhibited a narrow particle size distribution of 31.1, 34.6, 25.1 and 27.6 nm, respectively. And the zeta potential values of them were −19.4, −21.6, −19.7 and −23.8mV, respectively. The antioxidant and antibacterial activities of ZOP-NPs-AgNPs were superior to those of ZOP, ZOP-NPs and ZOP-AgNPs.

Most proteins given orally have low bioavailability and are easily eliminated by rapid metabolism in vivo. In order to immobilize the drug at the site of administration and delay its release, a natural, gentle release system was designed. In this study, a heteropolysaccharide (ZOP) was isolated fromZingiber officinaleusing an ultrasonic assisted extraction method. ZOP Ara = 1.97: 1.15: 94.33: 1.48: 1.07. The ZOP/Chitosan (CS) composite hydrogel was synthesized using epichlorohydrin (ECH) as a cross-linking agent. The structure, morphology, and water-holding capacity of the composite hydrogel were characterized. The data showed that the addition of ZOP improved the hardness and water-holding capacity of the material. A swelling ratio test showed that the prepared hydrogel was sensitive to pH and ionic strength. In addition, the degradation rate of the hydrogel in a phosphate-buffered saline (PBS) solution with a pH value of 1.2 was higher than that in PBS with pH value of 7.4. Similarly, the release kinetics of Bovine serum albumin (BSA) showed higher release in an acidic system by the hydrogel composed of ZOP/CS. The hydrogel prepared by this study provided a good microenvironment for protein delivery. In summary, this composite polysaccharide hydrogel is a promising protein-drug-delivery material. Keywords:Zingiber offtcinale;polysaccharide;chitosan;hydrogels;drug release;degradation

Introduction Gambogic acid (GA) is proved to have anti-tumor effects on gastric cancer. Due to poor solubility, non-specific biological distribution, toxicity to normal tissues and short half-life, it is hard to be applied into the clinic. To overcome these issues, we developed a thermosensitive and injectable hydrogel composed of hydroxypropyl cellulose, silk fibroin and glycerol, with short gelling time, good compatibility and sustained release, and demonstrated that the hydrogel packaged with gambogic acid nanoparticles (GA-NPs) and tumor-penetrating peptide iRGD could improve the anti-tumor activity. Methods The Gelling time and micropore size of the hydrogels were regulated through different concentrations of glycerol. Controlled release characteristics of the hydrogels were evaluated with a real-time near-infrared fluorescence imaging system. Location of nanoparticles from different carriers was traced by confocal laser scanning microscopy. The in vivo antitumor activity of the hydrogels packaging GA-NPs and iRGD was evaluated by investigating tumor volume and tumor size. Results The thermo-sensitive properties of hydrogels were characterized by 3–4 min, 37°C, when glycerol concentration was 20%. The hydrogels physically packaged with GA-NPs and iRGD showed higher fluorescence intensity than other groups. The in vivo study indicated that the co-administration of GA-NPs and iRGD by hydrogels had higher antitumor activity than the GA-loaded hydrogels and free GA combining with iRGD. Free GA group showed few antitumor effects. Compared with the control group, the body weight in other groups had no obvious change, and the count of leukocytes and hemoglobin was slightly decreased.Discussion The hydrogel constructed iRGD and GA-NPs exerted an effective anti-tumor effect possibly due to retention effect, local administration and continuous sustained release of iRGD promoting the penetration of nanoparticles into a deep part of tumors. The delivery system showed little systemic toxicity and would provide a promising strategy to improve anti-gastric cancer efficacy.

The present study was undertaken with the aim of evaluating the ginger essential oil potential and its urea-loading efficacy in the preparation of control-releasing chitosan microspheres. The ginger oil for hydro-distillation was extracted by applying Clevenger apparatus, chemical composition was characterized by FTIR, and the GC-MS was used to identify the oil compounds. Urea-loaded chitosan microsphere formulations were formulated through modified emulsification and cross-linking procedure. The various factors and levels related to preparation of urea-loaded chitosan microspheres were evaluated using response surface test. The present study was also an effort to observe the yield and content of nitrogen. Additionally, FTIR and SEM were applied to characterize the microspheres. The results show the yield of ginger oil as 6.0%. Main compounds identified by GC-MS were gingerol, decanal, isoshogaol, octanal, and others. FTIR also showed that some typical bands appeared in ginger oil. The nitrogen content % in urea-loaded microspheres showed a significantly higher value in proportion to higher oil volume and cross-linking. However, by decreasing the amount of urea and enhancing acetic acid, nitrogen content was decreased. The lower oil concentrations improved the microsphere surface structure and morphology. Response surface analysis showed that the optimum preparation conditions are as follows: 2.094% of Span 80, 2.302% of acetic acid, and cross-linking agent ratio of formaldehyde to glutaraldehyde 1:8.15. Under these conditions, the theoretical value of nitrogen content of urea-loaded chitosan microspheres was 4.683%. According to release test, the urea diffused uniformly in the microspheres after 48 h. It is concluded that the use of ginger oil in control release of fertilizers is the right option in the formulations of control-releasing micro-fertilizers.

The non-standard use of curcumin could cause some adverse drug reactions, such as diarrhea, nausea and skin allergies. Thus, the curcumin determination was fundamental to disease treatment and prevention. Herein, a facile and efficient fluorescent probe was developed based on carbon dots, which was prepared through hydrothermal method (o-phenylenediamine and N-isopropylacrylamide as the reaction raw materials). Characteristics of the as-fabricated carbon dots (NCDs) were studied through some analysis techniques, such as UV–vis absorption spectroscopy, transmission electron microscopy, Fourier transform infrared instrument, X-ray photoelectron spectroscopy and fluorescence spectrophotometer. Fluorescence quenching phenomenon could be observed after addition of curcumin. This as-prepared fluorescent probe displayed a significant response for the determination of curcumin with a satisfactorily lower detection limit of 0.017 μM and a considerable linear range of 0.5–50 μM compared to reported literatures. Because of the preeminent repeatability and anti-jamming capability, the as-developed CDs suggested mighty potentiality for actual applications of curcumin detection in real samples and temperature sensing.

This study aimed to develop a surfactant-free microemulsion (SFME) which based on hydrophobic deep eutectic solvent (DES), intended to prospect its extraction effect of curcuminoids. A menthol and n-octanoic acid (ME-OA 1:2) hydrophobic DES is used as oil phase to SFME which have low viscosity, higher stability and better solubility for extracting curcuminoids. The hydrophobic DES was successfully formed which was demonstrated by 1HNMR and FT-IR. The ternary phase diagram and the microstructure of microemulsions was determined via visual titration, light scattering experiment and electrical conductivity. SFME consisting of ME-OA/Ethanol/H 2 O (1/7/2 mass ratio) had been constructed to investigate its ability to extract curcuminoids from turmeric rhizome utilizing HPLC. And the it was tested the extracted curcuminoids as a function of extraction time by UV–vis spectra. Using the single-factor experiments and response surface methodology (RSM), the optimal extraction conditions by ultrasound-assisted SFME were identified as follows: liquid/solid ratio of 12 g/g, extraction time of 63 min, and ultrasonic time (300 W) of 45 min. The total extraction amounts of curcuminoids could reach 61.39 mg/g powder under the best conditions, and the main factor to influence the final extraction effect was extraction time. The molecular dynamic simulation was performed to investigate the interaction between SFME and curcuminoids. Compared with the single component solvent system of ethanol, acetone, ME-OA and water, curcuminoids were stronger interaction energy with solvent molecules and higher hydrogen bond number in SFME. SFME formed the O/W structure of encapsulation solubilization for curcuminoids, in order to promote the dissolving of curcuminoids.

Xylan-type hemicellulose hydrolysis by an organic acid solution for the production of xylo-oligosaccharides (XOS) is efficient and eco-friendly, but the effects of different organic acids on XOS production from Toona sinensis branch (TB) biomass is limited. In this work, under the conditions of 170 °C for 60 min, 33.1 % and 38.7 % XOS yields were obtained from polysaccharides present in TB by 2 % lactic acid (LA) and 6 % propionic acid (PA), respectively. Then 77 % of the lignin was removed by hydrogen peroxide−acetic acid pretreatment system, and 39.5 % and 44.7 % XOS yield were obtained from polysaccharides in delignification TB by 2 % LA and 6 % PA, respectively. It was found that PA hydrolysis, especially from delignified TB, resulted in higher XOS yield and purity compared to LA hydrolysis. Moreover, the content of byproducts (xylose, hydroxymethyl-furfural and furfural) in PA hydrolysate was lower. Following the hydrolysis process, the simultaneous saccharification and fermentation of the TB solid residue achieved an ethanol yield of 71.5 %. This work proposed an integrated process to preferentially convert the TB hemicellulose into valuable XOS and then convert the cellulose into ethanol. This process had the advantages of eliminating the need for isolation and purification of xylan, and the potential to obtain multiple products from the same raw material.
