Enhanced permeability and antifouling performance of cellulose acetate ultrafiltration membrane assisted by l-DOPA functionalized halloysite nanotubes

Keguang Mu,Dalun Zhang,Ziqiang Shao,Dujian Qin,Yalong Wang,Shuo Wang

l -Dopa functionalized halloysite nanotubes (HNTs) were prepared by the self-polymerization of l -dopa in the weak alkaline condition. Then different contents of l -dopa coated HNTs (LPDHNTs) were blended into cellulose acetate to prepare enhanced performance ultrafiltration membranes via the phase inversion method . The HNTs and LPDHNTs were characterized by FTIR, XPS, and TEM anysis. And the membranes morphologies, separation performance, antifouling performance, mechanical properties and hydrophilicity were also investigated. It was found that the composite membranes exhibited excellent antifouling performance. The pure water flux of 3.0   wt% LPDHNTs/CA membrane increased from 11.4   L   m −2   h −1 to 92.9   L   m −2   h −1 , while the EA rejection ratio of the membrane was about 91.2%. In addition, the mechanical properties of the resultant membranes were strengthened compared with the CA ultrafiltration membrane.

Facile synthesis and characterization of poly(levodopa)-modified silica nanocomposites via self-polymerization of levodopa and their adsorption behavior toward Cu2+

Huang Qiang,Liu Meiying,Guo Ren,Mao Liucheng,Wan Qing,Zeng Guangjian,Huang Hongye,Deng Fengjie,Zhang Xiaoyong,Wei Yen

In this study, a facile surface functionalization method was applied to synthesize poly-levodopa (PDOPA)-modified silica nanocomposites (denoted as SiO 2 -PDOPA). The adsorption capacity of SiO 2 -PDOPA was found to be higher than that of unmodified SiO 2 NPs. The successful preparation of SiO 2 -PDOPA was confirmed by Fourier transform infrared spectroscopy, transmission electron microscopy, and thermo gravimetric analysis. The adsorption behavior was investigated using SiO 2 -PDOPA as adsorbents and Cu 2+ as a model heavy metal pollutant. Various adsorption parameters, including contact time, solution pH, temperature, and initial Cu 2+ concentrations were studied. The results showed that pH could markedly affect the adsorption process of SiO 2 -PDOPA to Cu 2+ . The optimum pH for Cu 2+ adsorption was found to be 7.0. The adsorption kinetic data were analyzed using pseudofirst-order, pseudosecond-order, and intraparticle diffusion models. The adsorption isotherms could be described by Langmuir and Freundlich isotherm models. The fitting results showed that the adsorption kinetics and isotherms were better described by the pseudosecond-order and Langmuir model, respectively. The values of thermodynamics constants, including entropy change (Δ S 0 ), enthalpy change (Δ H 0 ), and Gibbs free energy (Δ G 0 ) were determined at different temperatures. Results suggested that the adsorption process of SiO 2 -PDOPA to Cu 2+ is a feasible, endothermic, and spontaneous process.

Tianyu Li,Zhichao Ye,Yu Cai,Tingting Tu,Bin Zhang,Shanshan Zhang,Lu Fang,Xiyu Mao,Shiyi Xu,Xuesong Ye,Bo Liang

Conductive hydrogels have shown promise in a wide range of electrochemical sensing applications. Decorating conductive hydrogel on the electrode surface is a key technical challenge. Herein, we have constructed a method to achieve better and stable attachment between conductive hydrogel and electrode by surface rebuilding technique. Electrochemical gelation of poly(3,4-ethylenedioxythiophene): poly(4-styrenesulfonate) (PEDOT:PSS) hydrogels on gold electrodes that have undergone surface rebuilding method were then made into several biosensors for electrochemical sensing of H 2 O 2 , glucose, dopamine (DA) and levodopa (L-Dopa) respectively. The H 2 O 2 sensor shown a linear range from 0 to 1 mM with ultrahigh sensitivity of 1988.66 μA⋅cm −2 ⋅mM −1 . The sensor responded to glucose ranging from 0 to 14 mM with high sensitivity of 198.047 μA⋅cm −2 ⋅mM −1 . The sensor responded to DA ranging from 0 to 1 mM with high sensitivity of 145.047 μA⋅cm −2 ⋅mM −1 , and to L-Dopa ranging from 0 to 0.5 mM with high sensitivity of 982.21 μA⋅cm −2 ⋅mM −1 . We initially verify the gold surface rebuilding method in solving the problem of difficult combination of electrochemical mediated hydrogel and electrode, the method could be explored further for various promising applications in hydrogel electronics in the future.

Simultaneous electrochemical detection of levodapa, paracetamol and L-tyrosine based on multi-walled carbon nanotubes

Zai-Yu Li,Dan-Yang Gao,Zhi-Yong Wu,Shuang Zhao

Herein multi-walled carbon nanotubes (MWCNTs) were processed by ultrasonication and freeze-drying method. The morphology of the processed MWCNTs was examined by scanning electron microscopy. An original electrochemical sensor for the simultaneous detection of levodapa (LD), paracetamol (PA) and L-tyrosine (Tyr) was developed by dropcasting a mixture of processed MWCNTs and Nafion on a glassy carbon electrode. The as-prepared sensor was studied by cyclic voltammetry and differential pulse voltammetry. The peak currents of LD, PA and Tyr significantly increased compared to those obtained at bare glassy carbon electrodes or unprocessed MWCNTs modified electrodes. The peaks of LD, PA and Tyr were well-defined and obviously separated from each other. The linear ranges for detection of LD, PA and Tyr were 2.0–300.0, 2.0–180.0, and 2.0–120.0 μM, with a detection limit of 0.6, 0.5 and 0.8 μM (S/N = 3), respectively. Finally, the sensor was applied to detect LD, PA and Tyr in serum samples, and the results were satisfactory.

Fabrication of Electrochemical Sensor for Acetaminophen Based on Levodopa Polymer and Multi-walled Carbon Nanotubes Complex

Hui Yang,Zhiqiang Wei,Suna He,Tian Li,Yanfeng Zhu,Lengxin Duan,Yan Li,Jiangang Wang

In this work, we describe the fabrication of electrochemical sensor for acetaminophen (ACOP) based on levodopa (L-Dopa) polymer and multi-walled carbon nanotubes (MWCNTs) complex modified carbon paste electrode (Poly (L-Dopa)/MWCNTs/CPE). L-Dopa Polymer and MWCNTs complex is prepared by in situ electrochemical polymeric deposition and the electrochemical polymeric deposition behavior of L-Dopa on the surface of MWCNTs was investigated by cyclic voltammetry (CV). A novel method for the determination of ACOP is developed based on a well-defined electrochemical response signal of ACOP at Poly (L-Dopa)/MWCNTs/CPE by differential pulse voltammetry (DPV). Under the optimized conditions, the value of the DPV oxidation peak current linearly depends on the concentration of ACOP in the range from 0.06 to 30 μM. The linear equation for ACOP detection is I pa (μA) =0.0718c (μM) +0.0307 (r=0.9990) and the detection limit is 0.02 μM (S/N=3). The recoveries for ACOP are from 98.2% to 99.4% with relative standard deviation (RSD) between 3.9% and 4.6% in diluted serum samples.

Direct Electrodeposition to Fabricate Vertically-Oriented Graphene Nanosheets Modified Electrode and its Application for Determination of Levodopa in the Presence of Uric Acid and Ascorbic Acid

Yanju Wu,Huijun Song,Kui Lu,Yong Ye,Mingxiu Lv,Yufen Zhao

The electrodeposition of reduced graphene oxide (rGO) film with preferred vertical orientation was fabricated on a glassy carbon electrode by using pulse potential method in a graphene oxide solution. The rGO film was applied in developing a high-sensitive electrochemical sensor for detection of levodopa (L-dopa). The new electrochemical sensor displayed a significant response to L-dopa because of its excellent electric conductivity and large specific surface area. Moreover, the determination of L-dopa on this modified electrode was not interfered by the coexistence of ascorbic acid (AA) and uric acid (UA). Under the selective conditions, the modified electrode showed a linear voltammetric response to L-dopa within a certain concentration ranging from 5 . 0 × 1 0 − 7 − 1 . 5 × 1 0 − 4 mol L − 1 , with the detection limit of 1 . 0 × 1 0 − 7 mol L − 1 . Besides, the modified electrode also exhibited an excellent selectivity, stability, reproducibility and repeatability.

Physicochemically modulated fluorescence-scattering ratiometric sensor for selective and visual detection of levodopa

Jin Yuan Zhang,Ying Zhang,Yu Zou,Ze Li Bo Xu,Bo Zhang,Wang Ren

In this study, a facile fluorescence-scattering ratiometric sensor was designed for visual and selective detection of levodopa (LD) via a clever physicochemical modulation scheme. The alkalized products of LD can rapidly react with polyethyleneimine (PEI) to exhibit an intense blue fluorescence and decrease the second-order scattering (SOS) signal of PEI. As the concentration of LD increased, the fluorescence intensity at 420 nm increased and the SOS intensity at 675 nm decreased synchronously. Thus the fluorescence-scattering ratiometric sensor was constructed by virtue of the two simultaneously changed signals. Furthermore, red light-emitting Au nanoclusters (AuNCs) were added into the above mixture solution to enlarge the SOS signal and provide a stable red background fluorescence. The intensity ratio of fluorescence to SOS ( F /( S / S blank )) is linear dependent on C LD in the wide range of 50.0–––30000.0 nM, and LD as low as 50.0 nM can be identified with the naked eye via change of fluorescence color. The developed ratiometric sensor is smart, simple and efficient, and has been applied to the convenient assay of LD in real samples. The proposed physicochemical modulation strategy provides a new and facile path for selectively and visually identifying the target from its analogues.

Cortico-striatal gamma oscillations are modulated by dopamine D3 receptors in dyskinetic rats

Wang Pengfei,Bi Yuewei,Li Min,Chen Jiazhi,Wang Zhuyong,Wen Huantao,Zhou Ming,Luo Minjie,Zhang Wangming

Long-term levodopa administration can lead to the development of levodopa-induced dyskinesia. Gamma oscillations are a widely recognized hallmark of abnormal neural electrical activity in levodopa-induced dyskinesia. Currently, studies have reported increased oscillation power in cases of levodopa-induced dyskinesia. However, little is known about how the other electrophysiological parameters of gamma oscillations are altered in levodopa-induced dyskinesia. Furthermore, the role of the dopamine D3 receptor, which is implicated in levodopa-induced dyskinesia, in movement disorder-related changes in neural oscillations is unclear. We found that the cortico-striatal functional connectivity of beta oscillations was enhanced in a model of Parkinson’s disease. Furthermore, levodopa application enhanced cortical gamma oscillations in cortico-striatal projections and cortical gamma aperiodic components, as well as bidirectional primary motor cortex (M1) ↔ dorsolateral striatum gamma flow. Administration of PD128907 (a selective dopamine D3 receptor agonist) induced dyskinesia and excessive gamma oscillations with a bidirectional M1 ↔ dorsolateral striatum flow. However, administration of PG01037 (a selective dopamine D3 receptor antagonist) attenuated dyskinesia, suppressed gamma oscillations and cortical gamma aperiodic components, and decreased gamma causality in the M1 → dorsolateral striatum direction. These findings suggest that the dopamine D3 receptor plays a role in dyskinesia-related oscillatory activity, and that it has potential as a therapeutic target for levodopa-induced dyskinesia.

Microneedle patch based on molecular motor as a spatio-temporal controllable dosing strategy of L-DOPA for Parkinson’s disease

Xin Zhou,Bowen Li,Mingming Guo,Wenchang Peng,Deping Wang,Qinglu Guo,Shuchao Wang,Dong Ming,Bin Zheng

Controlling medication according to the condition has become a universal treatment regimen for Parkinson's disease (PD) worldwide. However, the general oral administration method is prone to a variety of digestive system discomfort, easy to be broken down by digestive juices and intestinal microorganisms, resulting in low absorption efficiency, and a serious delay in the efficacy of the drug. Here, we developed a microneedle patch (MNP) that could release drugs with controlled temporal and spatial precision triggered by near-infrared (NIR) light for the treatment of PD. The microneedles (MNs) took Gelatin-methacryloy (GelMA) as the matrix, and therapeutic drugs (L-DOPA) were stored in the mesopore of the upconversion micron-rods (UCMRs), dispersing in the GelMA. The molecular motor, as a poreblocker and gate switch, isomerizes under the excitation of ultraviolet and visible light from UCMRs for controlling drug release. This kind of MNP penetrates the epidermis in a painless, non-invasive, and non-infectious way. Under the exposure of NIR light, the L-DOPA in the MNs will be released as needed, and then it seeps into the blood and brain for relieving the symptoms of PD. In the treatment of Parkinson's mice model, this regimen significantly restored motor function, and further mechanistic studies revealed that it reduced neuroinflammation and dopaminergic neuronal death in the substantia nigra. Besides, the released L-DOPA directly entered the blood, which reduces the side effects on the gastrointestinal tract and improves the utilization rate of the drug. These attractive superiorities indicated that the MNP with controlled drug delivery system may be candidate strategies for the treatment of PD and can be used in various related biomedical fields.

Lyotropic Liquid Crystalline Based Nasal Spray for Improved Parkinson's Treatment: Enhanced Superior Nasal Tract Deposition and Antioxidation Strategy

Xiao Xia,Ziqiao Zhong,Chunfu Wei,Guanlin Wang,Ziyu Zhao,Jingyu He,Zizhao Zhai,Yue Zhou,Wenhao Wang,Haihua Guo,Yuzhen Jin,Cenfeng Liu,Xiao Yue,Chuanbin Wu,Ying Huang,Xuejuan Zhang

The blood–brain barrier is one of the serious challenges in the treatment of Parkinson's disease (PD), which severely hinders the drug's brain bioavailability. In this study, a lyotropic liquid crystalline (LLC) based in situ gel nasal spray loaded with the first-line drug Levodopa (LDA) for PD is developed to resolve this issue based on a nose-to-brain drug delivery approach. Specifically, the two key stages of LLC-based in situ gel nasal spray, droplet deposition, and drug retention are tailored. By adjusting LLC's precursor solution's viscosity, the droplet deposition on the superior nasal tract can be increased by 18 times. Through nasal mucus-triggered sol–gel transition, the LDA nasal cavity retention is prolonged by three times. The epigallocatechin gallate is incorporated into the LLC-based in situ gel nasal spray to inhibit the autoxidation of LDA and alleviate the oxidative stress response in PD. The results show that the LLC-based in situ gel nasal spray has a 26-fold higher brain bioavailability of LDA compared to oral administration. The PD rats show a significant improvement in behavioral and cognitive disorders after administration. The LLC-based in situ gel nasal spray is proved to work by reducing oxidative damage. This study is anticipated to offer a promising strategy for the clinical application of LDA.

Improving Treatment for Parkinson's Disease: Harnessing Photothermal and Phagocytosis-Driven Delivery of Levodopa Nanocarriers Across the Blood-Brain Barrier

Kaili Liang,Li Yang,Jiawei Kang,Bo Liu,Ding Zhang,Liyan Wang,Wei Wang,Qing Wang

Parkinson's disease (PD) poses a significant therapeutic challenge, mainly due to the limited ability of drugs to cross the blood-brain barrier (BBB) without undergoing metabolic transformations. Levodopa, a key component of dopamine replacement therapy, effectively enhances dopaminergic activity. However, it encounters obstacles from peripheral decarboxylase, hindering its passage through the BBB. Furthermore, levodopa metabolism generates reactive oxygen species (ROS), exacerbating neuronal damage. Systemic pulsatile dosing further disrupts natural physiological buffering mechanisms. In this investigation, we devised a ROS-responsive levodopa prodrug system capable of releasing the drug and reducing ROS levels in the central nervous system. The prodrug was incorporated within second near-infrared region (NIR-II) gold nanorods (AuNRs) and utilized angiopep-2 (ANG) for targeted delivery across the BBB. The processes of tight junction opening and endocytosis facilitated improved levodopa transport. ROS scavenging helped alleviate neuronal oxidative stress, leading to enhanced behavioral outcomes and reduced oxidative stress levels in a mouse model of PD. Following treatment, the PD mouse model exhibited enhanced flexibility, balance, and spontaneous exploratory activity. This approach successfully alleviated the motor impairments associated with the disease model. Consequently, our strategy, utilizing NIR-Ⅱ AuNRs and ANG-mediated BBB penetration, coupled with the responsive release of levodopa, offers a promising approach for dopamine supplementation and microenvironmental regulation. This system holds substantial potential as an efficient platform for delivering neuroprotective drugs and advancing PD therapy.

Accurate Neurotransmitter Release by Ultrasound-Responsive Nanomaterials for Alleviating L-DOPA-Induced Dyskinesia in Parkinson's Disease

Shengwei Jia,Xiaoxia Li,Fang Wang,Hao Zhang,Qun-Dong Shen

Functional nanomaterials show great promise in next-generation targeted non-genetic neuromodulation and on-demand neurotransmitter release with superior spatiotemporal accuracy. L-DOPA-induced dyskinesia (LID) is a common complication during the treatment process of Parkinson's disease. Dysfunction in the dynamics of dopamine release plays a pivotal role in the disease. A strategy for LID treatment by using ultrasound-responsive functional nanomaterials for controlled release of nitric oxide is proposed, which can activate the nitric oxide (NO)-soluble guanylate cyclase (sGC)-cyclic guanosine monophosphate (cGMP) signaling pathway. This alleviates the abnormal synaptic plasticity caused by excessive and pulsatile dopamine, and diminishes hypersensitivity of the D 1 dopamine receptors, thereby reducing the behavioral disorders associated with LID. These findings offer a novel perspective for therapy of neurodegenerative diseases.
