Functionalized alginate-based bioinks for microscale electrohydrodynamic bioprinting of living tissue constructs with improved cellular spreading and alignment

Qiu Zhennan,Zhu Hui,Wang Yutao,Kasimu Ayiguli,Li Dichen,He Jiankang

Bioprinting has been widely investigated for tissue engineering and regenerative medicine applications. However, it is still difficult to reconstruct the complex native cell arrangement due to the limited printing resolution of conventional bioprinting techniques such as extrusion- and inkjet-based printing. Recently, an electrohydrodynamic (EHD) bioprinting strategy was reported for the precise deposition of well-organized cell-laden constructs with microscale filament size, whereas few studies have been devoted to developing bioinks that can be applied for EHD bioprinting and simultaneously support cell spreading. This study describes functionalized alginate-based bioinks for microscale EHD bioprinting using peptide grafting and fibrin incorporation, which leads to high cell viability (>90%) and cell spreading. The printed filaments can be further refined to as small as 30 μm by incorporating polyoxyethylene and remained stable over one week when exposed to an aqueous environment. By utilizing the presented alginate-based bioinks, layer-specific cell alignment along the printing struts could be observed inside the EHD-printed microscale filaments, which allows fabricating living constructs with cell-scale filament resolution for guided cellular orientation. Protocol for engineering bone organoids from mesenchymal stem cells

Jian Wang,Dongyang Zhou,Ruiyang Li,Shihao Sheng,Guangfeng Li,Yue Sun,Peng Wang,Yulin Mo,Han Liu,Xiao Chen,Zhen Geng,Qin Zhang,Yingying Jing,Long Bai,Ke Xu,Jiacan Su

Bone organoids are emerging as powerful tools for studying bone development and related diseases. However, the simplified design of current methods somewhat limits their application potential, as these methods produce single-tissue organoids that fail to replicate the bone microarchitecture or achieve effective mineralization. To address this issue, we propose a three-dimensional (3D) construction strategy for generating mineralized bone structures using bone marrow-derived mesenchymal stem cells (BMSCs). By mixing BMSCs with hydrogel to create a bone matrix-mimicking bioink and employing projection-based light-curing 3D printing technology, we constructed 3D-printed structures, which were then implanted subcutaneously into nude mice, away from the native bone microenvironment. Even without external stimulation, these implants spontaneously formed mineralized bone domains. With long-term culture, these structures gradually matured into fully differentiated bone tissue, completing both mineralization and vascularization. This in vivo bone organoid model offers a novel platform for studying bone development, exploring congenital diseases, testing drugs, and developing therapeutic applications.

Zili Gao,Shenglong Ding,Tingting Fan,Wenhui Huang,Xiyuan Zhao,Xin Liu,Wenli Liu,Mingzhu Zhang,Qi Gu

Summary Here, we present a protocol for 3D printing heart tissues using thiol-norbornene photoclick collagen (NorCol). We describe steps for synthesizing NorCol, preparing bioink and the support bath, and cell-laden printing. We then detail procedures for the loading of C2C12 cells into NorCol, ensuring structural integrity and cell viability after printing. This protocol is adaptable to various cell lines and allows for the printing of diverse complex structures, which can be used in drug screening and disease modeling.

Improving quantitative control and homogeneous distribution of samples on paper-based analytical devices via drop-on-demand inkjet printing

Meng-Meng Liu,Xin Lian,Zi-Zhen Guo,Hui Liu,Yun Lei,Yao Chen,Wei Chen,Xin-Hua Lin,Ai-Lin Liu,Xing-Hua Xia

A standard desktop printer with multiple ink cartridges can accurately deposit a broad variety of biomaterials on microfluidic paper-based analytical devices (μPADs) which have been extensively applied to environmental monitoring and screening of food and beverage contamination. Finding ways to realize sample quantitative control by tuning the CMYK value, however, remains challenging. Herein, we studied the influence of the CMYK value on the ink volume jetted by ink cartridges. The regularity research on a single-color and two-colors was performed in two print mode-grayscale printing and color printing. The results demonstrated that the number of ink dots increased with the increase of the gray value and opacity value, which means that the amount of the bio-ink increases with the increase of the CMYK value. The 3,3′,5,5′-tetramethylbenzidine–horseradish peroxidase–hydrogen peroxide, glucose oxidase–horseradish peroxidase and bull serum albumin–citrate buffer–tetrabromophenol blue systems were chosen as examples to prove the print regularity. Samples and assay reagents can be quantitatively deposited on a substrate by adjusting the CMYK value with as many as four ink cartridges. The present approach has been successfully applied to assay the targets in real serum samples, showing the potential application of the most common office piezoelectric printer in μPADs.

Crystallization enhanced thermal-sensitive hydrogels of PCL-PEG-PCL triblock copolymer for 3D printing

Yuecheng Cui,Ronghua Jin,Yang Zhou,Meirong Yu,Yun Ling,Li-Qun Wang

Temperature-sensitive hydrogels with mild gel-forming process, good biocompatibility and biodegradability have been widely studied as bioinks and biomaterial inks for 3D bioprinting. However, the hydrogels synthesized via copolymerization of aliphatic polyesters and polyethylene glycols have low mechanical strength and cannot meet the needs of 3D printing. In this paper, we propose a strategy of enhancing the strength of hydrogels by introducing crystallization between blocks to meet the requirements of 3D bioprinting inks. A series of polycaprolactone-polyethylene glycol-polycaprolactone (PCL-PEG-PCL) triblock polymers were prepared by ring-opening polymerization, of which the strong crystallinity of polycaprolactone blocks improved the printability and enhanced the mechanical properties of the ink. It was found that the resulted hydrogels were temperature-responsive, and the PCL blocks could form a crystalline phase in the state of the hydrogel, thereby significantly increasing the modulus of the hydrogel. Moreover, the mechanical strength of the hydrogel could be adjusted by changing the composition ratio of each block of the copolymer. The 3D printing results showed that the PCL-PEG-PCL hydrogel with crystallinity can not only be extruded and printed via temperature adjustment, but also the three-dimensional structure can be effectively maintained after 3D printing. The gels demonstrated good cell compatibility, and the cell survival rate was maintained at a high level.

3D printing of high-strength photo-crosslinking flaxseed gum bioink for cartilage regeneration

Kegang Shu,Zuquan Huang,Xiaomin Pei,Pek Yin Michelle Yew,Shanshan Wei,Yuan Yang,Ying Lan,Dan Kai,Li Zheng,Jinmin Zhao

Three-dimensional (3D) bioprinting provides a new possibility for personal customization of cartilage tissues. Although biocompatible, most natural biopolymer inks have poor mechanical strength to bear repeated extrusion , incomparable with load-bearing cartilage. By utilizing a heteropolysaccharide called flaxseed gum (FG), a strong photo-crosslinked methacrylated FG (FGMA) bioink was synthetized and integrated with stem cells for cartilage defect therapy. As a hybrid bioink, FGMA has favorable 3D printability and shown to exhibit high mechanical strength and superior fatigue resistant ability. 4% FGMA2 (MA substitution = 8.1%) has the modulus of about 41 times of GelMA and could maintain its structure integrity under 60% deformation after 2000 cycles. FGMA2 has a degradation period of about 66 days, similar to that of GelMA. In vitro study shows that FGMA2 has excellent biocompatibility with stem cells and chondrogenic potential, both beneficial to cartilage regeneration in the cartilage lesion model. More importantly, in vivo study demonstrated that the regenerated neo-cartilage tissue by FGMA2 displayed the similar morphology and matchable mechanical strength to the natural cartilage after 8 weeks. In conclusion, FGMA has demonstrated this potential as a high-performance bioink for 3D printing for tissue/organ regeneration.

Crosslinker-free silk/decellularized extracellular matrix porous bioink for 3D bioprinting-based cartilage tissue engineering

Xiao Zhang,Yang Liu,Chunyang Luo,Chenjun Zhai,Zuxi Li,Yi Zhang,Tao Yuan,Shilei Dong,Jiyong Zhang,Weimin Fan

As cartilage tissue lacks the innate ability to mount an adequate regeneration response, damage to it is detrimental to the quality of life of the subject. The emergence of three-dimensional bioprinting (3DBP) technology presents an opportunity to repair articular cartilage defects. However, widespread adoption of this technique has been impeded by difficulty in preparing a suitable bioink and the toxicity inherent in the chemical crosslinking process of most bioinks. Our objective was to develop a crosslinker-free bioink with the same biological activity as the original cartilage extracellular matrix (ECM) and good mechanical strength. We prepared bioinks containing different concentrations of silk fibroin and decellularized extracellular matrix (SF-dECM bioinks) mixed with bone marrow mesenchymal stem cells (BMSCs) for 3D bioprinting. SF and dECM interconnect with each other through physical crosslinking and entanglement. A porous structure was formed by removing the polyethylene glycol from the SF-dECM bioink. The results showed the SF-dECM construct had a suitable mechanical strength and degradation rate, and the expression of chondrogenesis-specific genes was found to be higher than that of the SF control construct group. Finally, we confirmed that a SF-dECM construct that was designed to release TGF-β3 had the ability to promote chondrogenic differentiation of BMSCs and provided a good cartilage repair environment, suggesting it is an ideal scaffold for cartilage tissue engineering.

Extracellular matrix bioink boosts stemness and facilitates transplantation of intestinal organoids as a biosafe Matrigel alternative

Zi-Yan Xu,Jin-Jian Huang,Ye Liu,Can-Wen Chen,Gui-Wen Qu,Ge-Fei Wang,Yun Zhao,Xiu-Wen Wu,Jian-An Ren

Organoids hold inestimable therapeutic potential in regenerative medicine and are increasingly serving as an in vitro research platform. Still, their expanding applications are critically restricted by the canonical culture matrix and system. Synthesis of a suitable bioink of bioactivity, biosecurity, tunable stiffness, and printability to replace conventional matrices and fabricate customized culture systems remains challenging. Here, we envisaged a novel bioink formulation based on decellularized extracellular matrix (dECM) from porcine small intestinal submucosa for organoids bioprinting, which provides intestinal stem cells (ISCs) with niche-specific ECM content and biomimetic microstructure. Intestinal organoids cultured in the fabricated bioink exhibited robust generation as well as a distinct differentiation pattern and transcriptomic signature. This bioink established a new co-culture system able to study interaction between epithelial homeostasis and submucosal cells and promote organoids maturation after transplantation into the mesentery of immune-deficient NODSCID-gamma (NSG) mice. In summary, the development of such photo-responsive bioink has the potential to replace tumor-derived Matrigel and facilitate the application of organoids in translational medicine and disease modeling.

An injectable bioink with rapid prototyping in the air and in-situ mild polymerization for 3D bioprinting

You Zhou,Shenglong Liao,Yanji Chu,Bin Yuan,Xinglei Tao,Xiaohua Hu,Yapei Wang

Bioprinting is an attractive technology for building tissues from scratch to explore entire new cell configurations, which brings numerous opportunities for biochemical research such as engineering tissues for therapeutic tissue repair or drug screening. However, bioprinting is faced with the limited number of suitable bioinks that enable bioprinting with excellent printability, high structural fidelity, physiological stability, and good biocompatibility, particularly in the case of extrusion-based bioprinting. Herein, we demonstrate a composite bioink based on gelatin, bacterial cellulose (BC), and microbial transglutaminase (mTG enzyme) with outstanding printing controllability and durable architectural integrity. BC, as a rheology modifier and mechanical enhancer component, endows the bioink with shear-thinning behavior. Moreover, the printed structure becomes robust under physiological conditions owing to the in situ chemical crosslinking catalyzed by mTG enzyme. Lattice, bowl, meniscus, and ear structures are printed to demonstrate the printing feasibility of such a composite bioink. Furthermore, the 3D-printed cell-laden constructs are proved to be a conducive biochemical environment that supports growth and proliferation of the encapsulated cells in vitro. In addition, the in vivo studies convince that the composite bioink possesses excellent biocompatibility and biodegradation. It is believed that the innovation of this new composite bioink will push forward the bioprinting technology onto a new stage.

Biocatalytic CsPbX3 Perovskite Nanocrystals: A Self-Reporting Nanoprobe for Metabolism Analysis

Menglu Li,Yujing Zeng,Xinyu Qu,Mohammed Jalalah,Saeed A. Alsareii,Chao Li,Farid A. Harraz,Genxi Li

CsPbX 3 perovskite nanocrystals (NCs), with excellent optical properties, have drawn considerable attention in recent years. However, they also suffer from inherent vulnerability and hydrolysis, causing the new understanding or new applications to be difficultly explored. Herein, for the first time, it is discovered that the phospholipid membrane (PM)-coated CsPbX 3 NCs have intrinsic biocatalytic activity. Different from other peroxidase-like nanozymes relying on extra chromogenic reagents, the PM-CsPbX 3 NCs can be used as a self-reporting nanoprobe, allowing an “add-to-answer” detection model. Notably, the fluorescence of PM-CsPbX 3 NCs can be rapidly quenched by adding H 2 O 2 and then be restored by removing excess H 2 O 2 . Initiated from this unexpected observation, the PM-CsPbX 3 NCs can be explored to prepare multi-color bioinks and metabolite-responsive paper analytical devices, demonstrating the great potential of CsPbX 3 NCs in bioanalysis. This is the first report on the discovery of nanozyme-like property of all-inorganic CsPbX 3 perovskite NCs, which adds another piece to the nanozyme puzzle and opens new avenues for in vitro disease diagnostics.

High-cytocompatible semi-IPN bio-ink with wide molecular weight distribution for extrusion 3D bioprinting

Li Meiqi,Shi Tingchun,Yao Danyu,Yue Xiuyan,Wang Haoxuan,Liu Kezhou

The development of 3D printing has recently attracted significant attention on constructing complex three-dimensional physiological microenvironments. However, it is very challenging to provide a bio-ink with cell-harmless and high mold accuracy during extrusion in 3D printing. To overcome this issue, a technique improving the shear-thinning performance of semi-IPN bio-ink, which is universally applicable to all alginate/gelatin-based materials, was developed. Semi-IPN bio-ink prepared by cyclic heating–cooling treatment in this study can reduce the cell damage without sacrificing the accuracy of the scaffolds for its excellent shear-thinning performance. A more than 15% increase in post-printing Cell viability verified the feasibility of the strategy. Moreover, the bio-ink with low molecular weight and wide molecular weight distribution also promoted a uniform cell distribution and cell proliferation in clusters. Overall, this strategy revealed the effects of molecular parameters of semi-IPN bio-inks on printing performance, and the cell activity was studied and it could be widely applicable to construct the simulated extracellular matrix with various bio-inks.

Alginate/gelatin-based hybrid hydrogels with function of injecting and encapsulating cells in situ

Pengfei Ren,Dandan Wei,Min Liang,Li Xu,Tianzhu Zhang,Qianli Zhang

Multi-network hydrogels with high strength and toughness have attracted increasing attention. Herein, a hybrid hydrogel consisting of alginate, gelatin, and polyacrylamide was constructed with the combination of advantages of natural and synthetic polymers. Alginate grafted with host-guest complex of βCD/Ad-AAm was first prepared, namely Alg-βCD/Ad-AAm, then further crosslink with gelatin methacryloyl (GelMA) to form hydrogel via one-step UV light initiation. The hydrogel produced by this method has more uniform and well-crosslinked networks. The hydrogels demonstrated uniform porosity, adjustable hydrophilicity (water contact angle within 32.7–91.5°), and desired mechanical properties (maximum tensile strain of 242.8%, tensile strength of 75.9 kPa, and Young's modulus of 28.5 kPa). The hydrogel also possessed self-healing ability and pH sensitivity, showing higher mechanical tensile strength at lower pH. The temperature-adjustable viscosity of pre-gel solution (sol-gel transition point of 20.4 °C) endowed it to be 3D printed as a bioink, and the printed scaffold exhibited good resilience and toughness. Moreover, HUVEC, L929, and 3T3 cells were cultured on hydrogel surfaces for 28 days and were enveloped within the hydrogels for 3D culture, indicating excellent cytocompatibility of the hydrogels. Therefore, this hybrid hydrogel system can be used potentially in cell culture scaffold and tissue engineering.
