A fluorescence biosensor based on single-stranded DNA and carbon quantum dots for acrylamide detection

Qingyi Wei,Peiyao Zhang,Ting Liu,Hongbin Pu,Da-Wen Sun

As a potential carcinogen produced in food thermal processing, acrylamide (AM) can cause irreversible harm to human health. For the detection of AM in food products, a simple fluorescent biosensor based on single-stranded DNA (ssDNA) and carbon quantum dots (CQDs) was developed. Reduced fluorescence intensity of CQDs at 445 nm (excitation at 350 nm) was induced by the attachment of ssDNA. In the presence of AM, ssDNA was preferentially bound to AM by hydrogen bonding and the degree of fluorescence reduction was smaller than that without AM. Under optimized conditions, results showed that the sensing approach for detecting AM had a low detection limit of 2.41 × 10 −8 M in the standard solution, and a linear relationship ranging from 5 × 10 −3 to 1 × 10 −7 M with the determination coefficient (R 2 ) of 0.9895 was obtained. Furthermore, a good recovery percentage (91.36–98.11%) in bread crust showed the potential for practical applications of this proposed biosensor.

Selected ssDNA aptamers–graphene oxide-based fluorescent biosensor to detect sulfameter in milk

Yanping He,Jingrui Zhang,Ruifen Tian,Yuying Lu,Ling Pan,Yuzhong Zhang

The abuse of sulfameter (SME) in animal husbandry can cause drug resistance and toxic or allergic reactions in humans. Therefore, it is very important to establish a simple, inexpensive, and efficient method for detecting SME in food. In this work, we propose a single fluorescent aptamer/graphene oxide (GO)-based biosensor to detect SME residues in milk. Aptamers that specifically bind to SME were screened using capture-SELEX and a ssDNA library immobilized on magnetic beads. The 68 active candidate aptamers were chemically synthesized for specificity and affinity characterization. Among the aptamers, the aptamer sulf-1 revealed the highest affinity (K d = 77 ± 15 nM) to SME and was selected to construct a GO-based fluorescent biosensor for real milk sample detection. Under optimal conditions, the single fluorescent aptasensor had a wide linear range (R 2 was 0.997) from 7 to 336 ng/ml and a low detection limit of 3.35 ng/ml that was calculated with a 3SD/slope. The single fluorescent method was also validated using SME-fortified milk samples, showing average recoveries ranging from 99.01% to 104.60% with a relative standard deviation of less than 3.88%. These results demonstrate that this novel aptamer sensor provides an opportunity for sensitive, convenient, and accurate detection of SME residues in milk.

Ultrasensitive fluorometric biosensor based on Ti3C2 MXenes with Hg2+-triggered exonuclease III-assisted recycling amplification

Liling Lu,Xiao Han,Jingwen Lin,Yingxin Zhang,Minghao Qiu,Ying Chen,Meijin Li,Dianping Tang

Herein a rapid and sensitive fluorometric bioanalysis platform for mercury(II) (Hg2+) detection was innovatively developed using ultrathin two-dimensional MXenes (Ti3C2) as fluorescence quencher and Hg2+-induced exonuclease III (Exo III)-assisted target recycling strategy for efficient signal amplification. Initially, fluorophore-labeled single-stranded DNA (FAM-labeled probe) can be easily adsorbed onto the surface of ultrathin Ti3C2 nanosheets by hydrogen bonding and metal chelating interaction, and the fluorescence signal emitted by the FAM-labeled probe is quenched strongly owing to the fluorescence resonance energy transfer between the FAM and ultrathin Ti3C2 nanosheets. Upon sensing the target Hg2+, the protruding DNA fragment at the 3′ end of hairpin will hybridize with primer (hairpin-Hg2+-primer), and then further digested by Exo III to produce a probe (nicker). The released target Hg2+ and primer continue to participate in the next recycling, resulting in more hairpin probes becoming nickers. The combination of a large number of nickers and FAM-probe resulted in a significant increase in the fluorescence signal of the system, which was attributed to the fact that the double helix DNA was more rigid and separated from the surface of the ultrathin Ti3C2 nanosheets. The obvious fluorescence signal change of the Ti3C2-based Exo III-assisted target recycling can be accurately monitored by fluorescence spectrometry, which is also proportional to the concentration of Hg2+. Under optimum operating conditions, the peak intensity (520 nm wavelength) of fluorescence increased with increasing Hg2+ within a wide dynamic working range from 0.05 nM to 50 nM (R2 = 0.9913) with a limit of detection down to 42.5 pM. The proposed strategy uses ultrathin MXenes as a platform for binding nucleic acids, which contributes to its potential in nucleic acid hybridization-based biosensing and/or nucleic acid signal amplification bio-applications.

Metal nanoclusters combined with CRISPR-Cas12a for hepatitis B virus DNA detection

Yu Tao,Ke Yi,Haixia Wang,Kai Li,Mingqiang Li

Since hepatitis B virus (HBV) infection can cause chronic viral hepatitis, increase the risk of liver fibrosis and cirrhosis, and ultimately result in hepatoma, it is of great significance to explore convenient, rapid, and sensitive diagnostic strategies for HBV DNA detection. In this study, we developed a novel and sensitive fluorescent biosensor for HBV detection based on the sensing system consisting of CRISPR-Cas12a enzymes and metal nanoclusters as the luminescent nanoprobes, involving gold nanoclusters (AuNCs), silver nanoclusters (AgNCs), and copper nanoclusters (CuNCs). The DNA probes could effectively be applied as the template for the preparation of fluorescent metal nanoclusters. In the presence of the HBV target, the trans-cleavage capability of Cas12a was able to be initiated to degrade DNA probes, which brought about the inhibition of metal nanoclusters formation, thereby giving minimal fluorescence signals. In comparison with AuNCs and AgNCs, the CuNCs-based nanosensor exhibited much higher sensitivity. In addition, the fluorescence response of CuNCs to HBV detection was significantly faster, which could be completed within 25 min without the requirements of sophisticated equipment or harsh reaction conditions. Moreover, the practical application of this analysis method was also demonstrated to detect HBV DNA target in human serum samples with satisfactory recovery, which was promising for simple, rapid, sensitive, and label-free HBV DNA bioanalysis .

Execution of aggregation-induced emission as nano-sensors for hypochlorite detection and application for bioimaging in living cells and zebrafish

Hong Chen,Xiaojun He,Yuzhong Yu,Yuna Qian,Jianliang Shen,Shanchao Zhao

Hypochlorite (ClO − ) could be used as a diagnostic marker for inflammation and related diseases. Although there have been many reports on probes for ClO − imaging, there was still a lack of specificity and anti-interference ability. Herein, carbazole (NEC) and tetraphenylethylene (TPE) equipped with thiobarbituric acid (TBA), NEC-TBA and TPE-TBA , were synthesized and used as a fluorescence biosensor for monitoring ClO − with aggregation-induced emission (AIE) effect. we identified that TPE-TBA , with formed nanoparticles in the mean grain size at 76 nm (5 μM), was a superior probe to target ClO − over other analytes with fluorescence “turn off” strategy. Subsequently, to explore the bioimaging application, TPE-TBA was able to sense exogenous ClO − in living HeLa cells through fluorescence imaging. In zebrafish model, TPE-TBA effectively captured exogenous ClO − in the entire organization of zebrafish. Overall, these AIE-based probes merit further development as organism targeting ClO − sensors.

Fluorescent biosensor based on MB-APT combined with Pt NPs for the detection of infectious bacteria in mouse and human wounds

Lanmei Gao,Houbing Zheng,Yuanlong Hu,Yi Zhong,Linhai Jiang,Yuanzi Wu,Fen Yan,Da Huang,Jianhua Li,Fang Zhang,Zhenyu Lin,Meishui Wang,Zuquan Weng

Bacterial infection of wounds is one of great concern to patients, and rapid and correct detection of bacterial infection is crucial to ensure accurate diagnosis and early intervention. Based on the principle that glucose can only be consumed by live bacteria, a fluorescent biosensor was constructed to detect four kinds of common infectious bacteria (Acinetobacter baumannii, Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus) in skin wounds, taking advantage of magnetic bead-aptamer for recognizing, sorting and enrichment, platinum nanoparticles for signal amplification . The linear detection range of AB, EC, PA and SA were 27– 8 × 10 6 CFU/mL, 10– 2.5 × 10 7 CFU/mL, 34– 2.5 × 10 7 CFU/mL and 10– 1.0 × 10 7 CFU/mL, respectively, and the limits of detection were 27 CFU/mL, 10 CFU/mL, 34 CFU/mL and 2 CFU/mL. Furthermore, all target bacteria in samples containing 8 × 10 8 CFU/mL of other interfering bacteria have been successfully identified and quantified. The proposed method was also successfully applied to the detection of bacterial infection in skin wounds of mouse and human, including the detection of separate bacterial infection as well as a coinfection. The recovery of this method was in the range of 90.161– 109.961%. Thus, this proposed method can be a promising candidate for rapid and convenient evaluation of infectious bacteria in point-of-care settings.

A Sensitive Fluorescent Assay for Tetracycline Detection Based on Triple-helix Aptamer Probe and Cyclodextrin Supramolecular Inclusion

He Hui,Xie Chuchu,Yao Liu,Ning Ge,Wang Yonghong

Herein, an effective pyrene excimer signaled fluorescent biosensor for the determination of tetracycline based on triple-helix aptamer probe (TAP) and supramolecular inclusion of cyclodextrin was reported. The TAP was devised containing an aptamer loop, two DNA segment stems and a triplex-forming oligonucleotide (signal probe) labeled with pyrenes at 5′ and 3′ ends. The presence of target could result in its binding towards aptamer with a mighty affinity, leading to a conformation change of the TAP and whereupon the release of the signal probe. This liberty of signal probe enabled the formation of pyrene excimer, generating fluorescence signals. Further, signal amplification was fulfilled through the addition of γ-cyclodextrin which could interact with pyrene dimer, thus leading to an enhanced “on-state” of the sensing ensemble. In contrast, when the target was absent, the sensing ensemble remained “off-state” because of the long distance between two pyrene molecules. When the conditions were properly optimized, the increasing signal kept a linear dependence on target concentrations ranging from 5.0 nM to 100 nM, and the detection limit reached as low as 1.6 nM. In this way, a newly-constructed, simple, and economically affordable protocol enjoys desirable efficiency, sensitivity, specificity in biosensing. Also, its universality as another attractive behalf in assaying diverse targets was envisioned with only the need of matched aptamer replacement.

Magnetic aptamer copper nanoclusters fluorescent biosensor for the visual detection of zearalenone based on docking-aided rational tailoring

Yongxia Yu,Keren Chen,Zaihui Du,Bing Fang,Jing Zhan,Longjiao Zhu,Wentao Xu

To address the food safety issues caused by toxins, we established a fluorescent copper nanocluster biosensor based on magnetic aptamer for the visual and quantitative detection of ZEN. Specifically, we utilized the docking-aided rational tailoring (DART) strategy to analyze intermolecular force and interaction sites between zearalenone (ZEN) and the aptamer, and optimize the long-chain aptamer step by step to enhance the binding affinity by 3.4 times. The magnetic bead-modified aptamer underwent conformational changes when competing with complementary sequences to bind with ZEN. Then, the released complementary sequences will be amplified in template-free mode with the presence of the terminal deoxynucleotidyl transferase (TdT), and generating T-rich sequences as the core sequences for the luminescence of copper nanoclusters. The luminescence could be visualized and quantitatively detected through ultraviolet irradiation. The proposed label-free aptasensor exhibited high sensitivity and specificity, with a low limit of detection (LOD) of 0.1 ng/mL.

Dual DNA recycling amplification-assisted CRISPR/Cas12a cleavage for dual-channel ratiometric fluorescence biosensing of kanamycin antibiotic

Lai Shanshan,Guo Jiahong,Li Xin,Yu Xiaoxiao,Lai Guosong

Fluorescence biosensors hold significant importance for testing antibiotic residues which seriously endanger public health. However, how to adopt appropriate strategies to address the false result disadvantage involved in traditional single-channel biosensors is still a great challenge. Meanwhile, too much attention focused on designing signal amplification strategies of biosensors unavoidably decreases their detection efficiency. Herein, we combined the designed dual DNA recycling amplification strategy with CRISPR/Cas12a-mediated dual-channel signal output mode to successfully develop a novel ratiometric fluorescence biosensor for testing kanamycin (Kana) residues in complex sample matrices. The first recycling was formed from an exonuclease-assisted aptamer recognition reaction, which also triggered another cascade DNA recycling to amplify the release of the Cas12a activator. With the non-discrimination cleavage of Cas12a to cause reverse fluorescence changes of copper nanoclusters and an AMAC-labeled signal DNA, the ratiometric signal transduction strategy was constructed. Under optimal conditions, this biosensor could be applied for ultrasensitive testing of Kana antibiotics in a five-order of magnitude wide linear range with a low detection limit of 17.2 fg mL −1 . Benefiting from the self-correction function of the ratiometric signal transduction mode, it showed promising practicality in lake water and milk samples with the relative error less than 4.9% to the standard ELISA results. Besides CRISPR/Cas12a-based fluorescence output efficiency improvement, this biosensor also excluded the complicated manipulations and expensive instruments required in traditional methods. Therefore, it provides a good choice for expanding the application of fluorescence biosensing technology for practical analysis application. Graphical

A label-free aptasensor for rapid detection of clenbuterol based on SYBR Green I

Shuyan Xiao,Liang Sun,Jiping Lu,Zhongping Dong

A label-free fluorescent biosensor based on the clenbuterol binding aptamer and the fluorescent intercalator SYBR Green I (SGI) was established for the sensitive and selective detection of clenbuterol. In the absence of clenbuterol, a weak fluorescence signal of SGI was observed. In the presence of clenbuterol, the specific recognition and high-affinity binding of the aptamer with clenbuterol induced a conformational change of the aptamer, and a longer double-stranded DNA was formed. As a result, an enhancement in the fluorescence of SGI could be found. Under optimal conditions, the fluorescence change ratio increased linearly with clenbuterol concentration from 50 nM to 450 nM, with a detection limit of 18.8 nM. In addition, the proposed aptasensor showed good selectivity and was successfully applied for clenbuterol quantification in pork samples with recoveries of 90.37–104.98%.

A fluorescent biosensor based on boronic acid functionalized carbon dots for identification and sensitive detection of Gram-positive bacteria

Xiaoqing Zhang,Yue Ma,Lingling Zhao,Mei Liu

Rapid distinguishing and detection of Gram-positive and Gram-negative bacteria with high sensitivity is essential for human health worldwide. Herein, an excellent fluorescence biosensing platform for bacterial identification and detection is constructed on the basis of boronic acid functionalized carbon dots (B-CDs). The B-CDs fluorescence probes were tightly attached to the bacterial surface through the interaction of the boronic acid group with glycolipids on the outside of bacterial cells. Our experimental results showed that the B-CDs displayed a stronger binding ability toward Gram-positive bacteria, enabling the specific detection of Gram-positive bacteria. More importantly, Gram-positive bacteria and Gram-negative bacteria differ in terms of their saccharide units on the cell wall, resulting in distinct fluorescence signals, which could be employed for distinguishing Gram-positive and Gram-negative bacteria. Taking S. aureus as a representative of Gram-positive bacteria, the results showed that the linear range of detection was 2.6 × 101–5.2 × 106 CFU mL−1 and the detection limit was 7 CFU mL−1. Our research provides great potential for further development of a novel platform for bacterial determination and identification.

A novel quantum dots-based OFF–ON fluorescent biosensor for highly selective and sensitive detection of double-strand DNA

Zhengqing Liu,Shaopu Liu,Xiaodan Wang,Pingping Li,Youqiu He

We have developed a novel biosensor for the rapid determination of double-strand (ds) DNA based on the photoinduced electron transfer (PET) from glutathione (GSH) capped CdTe quantum dots (QDs) to praseodymium(III)–rutin (Pr 3+ –rutin) complex. The biosensor was based on a fluorescence “OFF–ON” mode. To provide the platform for dsDNA detection, the Pr 3+ –rutin complex was first employed as an effective fluorescence quencher to QDs through PET. Because of its strong binding affinity with Pr 3+ –rutin complex, dsDNA can break up the low fluoresced ionic ensemble, set free the luminescent QDs. Thus, the recognition of dsDNA by Pr 3+ –rutin complex can be realized through the restoration of QDs fluorescence. The relative recovered fluorescence intensity is directly proportional to the concentration of herring sperm (hs) DNA between 0.0874   μg   mL −1 and 20   μg   mL −1 , and the detection limit (3 δ / K ) is 0.0262   μg   mL −1 . The entire experimental process does not require any chemical modification and coupling of CdTe QDs and dsDNA, greatly simplifying the experimental process. Some possible reaction mechanisms are discussed.
