Preparation and Characterization of Folate-Targeted Fe3O4 Nanoparticle Codelivering Cisplatin and TFPI-2 Plasmid DNA for Nasopharyngeal Carcinoma Therapy

Juan Zhang,Huanhuan Weng,Xiangwan Miao,Quanming Li,Siqi Wang,Huifen Xie,Tao Liu,Minqiang Xie

A novel folate (FA) receptor-targeted superparamagnetic Fe3O4 nanoparticles (SPIONs) codelivering cisplatin (CDDP) and tissue factor pathway inhibitor-2 (TFPI-2) plasmid DNA (pDNA) was constructed. The core shell nanocomposites ([email protected]) were composed of superparamagnetic Fe3O4 core that binds CDDP and TFPI-2 shell that combines with folate-polyethylene glycol-polyethyleneimine (FA-PEG-PEI) via electrostatic interaction. The shell containing FA-PEG-PEI and TFPI-2 plasmid was synthesized through amidation reaction and electrostatic adsorption and the core containing SPION-CDDP was modified by aldehyde sodium alginate. Proton nuclear magnetic resonance and Fourier transform infrared spectra on FA-PEG-PEI polymers showed characteristic peaks of various metabolites in corresponding frequency. Transmission electron microscopy image of [email protected] nanoparticles demonstrated a near-monodisperse spherical morphology, while dynamic light scattering studies indicated an intensity-average diameter of 149.5 nm. Zeta potential was 14.89 ± 1.83 mv and the final concentration of loaded CDDP was 100 ug/ml. Gel electrophoresis data showed that the nanocomposite would protect TFPI-2 pDNA from being digested by DNases. Compared with CNE-2 cells, the good targetability and better gene transfection efficiency (57.9%) were detected by Prussian blue iron stain and fluorescence analysis in HNE-1 cells in vitro. The results suggested the potential application of [email protected] as a multifunctional anticancer nanomedicine on targeting therapy for FR positive NPC.

Functionalized boron nanosheets as an intelligent nanoplatform for synergistic low-temperature photothermal therapy and chemotherapy

Zi Fu,Gareth R. Williams,Shiwei Niu,Jianrong Wu,Feng Gao,Xuejing Zhang,Yanbo Yang,Yu Li,Li-Min Zhu

In this work, an innovative boron-based multifunctional nanoplatform was developed for synergistic chemotherapy/low temperature photothermal therapy (PTT). This platform is functionalized with a cRGD peptide to allow the targeting of αvβ3 integrin, which is over-expressed in the cells of tumors. The nanoparticles were further loaded with the chemotherapeutic drug doxorubicin (DOX) and a heat shock protein inhibitor (17AAG), and high loading capacities for both DOX (603 mg g−1 B-PEG-cRGD) and 17AAG (417 mg g−1) were obtained. The resultant DOX-17AAG@B-PEG-cRGD system shows both pH-controlled and near-infrared (NIR)-induced DOX and 17AAG release. It also provides significantly enhanced cellular uptake in cancerous cells over healthy cells. The presence of 17AAG allows low-temperature PTT to be combined with chemotherapy with DOX, resulting in highly effective anti-cancer activity. This has been confirmed by both in vitro assays and using an in vivo murine cancer model. It is expected that such a multifunctional nanoplatform can serve as a promising candidate for cancer therapy.

Superparamagnetic Fe3O4-PEG2K-FA@Ce6 Nanoprobes for in Vivo Dual-mode Imaging and Targeted Photodynamic Therapy

Yin Ting,Huang Peng,Gao Guo,Shapter Joseph G.,Shen Yulan,Sun Rongjin,Yue Caixia,Zhang Chunlei,Liu Yanlei,Zhou Sui,Cui Daxiang

The development of targeted nanoprobes is a promising approach to cancer diagnostics and therapy. In the present work, a novel multifunctional photo/magnet-diagnostic nanoprobe (MNPs-PEG 2K -FA@Ce6) has been developed. This nanoprobe is built using folic acid (FA), bifunctional polyethylene glycol (PEG 2K ) and photosensitizer chlorin e6 (Ce6). The MNPs-PEG 2K -FA@Ce6 nanoprobes are superparamagnetic, can be synthesized on a large scale by a one-pot hydrothermal process without further surface modification and are stable in an aqueous environment for eight months. Compared with free Ce6 nanoprobes in vitro studies, the MNPs-PEG 2K -FA@Ce6 nanoprobes significantly enhance cellular uptake efficiency and promote the effectiveness of photodynamic therapy (PDT) with the assistance of 633 nm laser irradiation. The unique nanoprobes show superior penetration and a retention time of more than six days with less accumulation in the liver allowing highly effective tumor recognition and monitoring. Additionally, there was little damage to healthy organs or tissues. These exciting new nanoprobes could be potential building blocks to develop new clinical therapies and translational medicine.

Ultrasound-triggered release of miR-199a-3p from liposome nanobubbles for enhanced hepatocellular carcinoma treatment

Xinmin Guo,Jianru Lin,Liwen Pan,Kun He,Zhihui Huang,Jialin Chen,Cuiyan Lin,Baohui Zeng,Sijia Luo,Mengdie Wang

This study was aimed to develop an efficient tumour-targeted liposome nanobubbles (LNBs) system using ultrasound-targeted nanobubble destruction for enhanced release and transfection of miRNA-199a-3p in hepatocellular carcinoma (HCC) therapy. The prepared LNBs comprised a polyethylene glycol-modified liposome shell and a perfluoropentane (PFP) core. MiRNA-199a-3p was attached to the nanocomposite surface via electrostatic adsorption, while RGD peptide functionalized the LNBs surface for enhanced HCC cell targeting, namely PFP@miR-RGD-LNBs. The LNBs were spherical with a narrow size distribution. The gene-loaded LNBs effectively condensed miR-199a-3p and protected it from enzymatic degradation. Low-intensity focused ultrasound (LIFU) promoted a fast release of miR-199a-3p from the prepared LNBs, thereby enhancing therapeutic effects. The combined application of PFP@miR-RGD-LNBs and LIFU exhibited a more potent inhibitory effect on HepG2 cells than the other groups, potentially due to LIFU promoting rapid and efficient gene release at the target site and increasing cell membrane permeability. Quantitative reverse transcription-polymerase chain reaction analysis revealed significantly increased mRNA expression levels of key apoptosis markers (Bad, Bax, Caspase-9 and Caspase-3) in the PFP@miR-RGD-LNBs + LIFU group compared to other groups. These findings suggest that the prepared LNBs are highly likely to be promising candidates for further exploration of HCC gene delivery and therapy.

Reactive oxygen/nitrogen species scavenging and inflammatory regulation by renal-targeted bio-inspired rhodium nanozymes for acute kidney injury theranostics

Yue Zheng,Huixi Yi,Zhixiong Zhan,Shan-Shan Xue,Guosheng Tang,Xiyong Yu,Dong-Yang Zhang

Acute kidney injury (AKI) results from the rapid deterioration of renal function, which is mainly treated by transplantation and dialysis, and has a high mortality rate. Inflammation induced by excess reactive oxygen/nitrogen species (RONS) plays a crucial role in AKI. Although small molecule antioxidants have been utilized to alleviate AKI, low bioavailability and side-effect of these drugs tremendously limit their clinical use. Hence, we successfully construct ultra-small (2–4 nm) rhodium nanoparticles modified with l -serine (denoted as Rh-Ser). Our results show that Rh-Ser with multiple enzyme-mimicking activities, allows remove various RONS to protect damaged kidney cells. Additionally, the ultrasmall size of Rh-Ser is conducive to enrichment in the renal tubules, and the modification of l -serine enables Rh-Ser to bind to kidney injury molecule-1, which is highly expressed on the surface of damaged renal cells, thereby targeting the damaged kidney and increasing the retention time. Moreover, Rh-Ser allows the production of oxygen at the inflammatory site, thus further improving hypoxia and inhibiting pro-inflammatory macrophages to relieve inflammation, and increasing the survival rate of AKI mice from 0 to 80%, which exhibits a better therapeutic effect than that of small molecule drug. Photoacoustic and fluorescence imaging can effectively monitor and evaluate the enrichment and therapeutic effect of Rh-Ser. Our study provides a promising strategy for the targeted treatment of AKI via RONS scavenging and inflammatory regulation.

Comprehensive Cellular Senescence Evaluation to Aid Targeted Therapies

Xiaolan Zhou,Xiaofeng Zhu,Weixu Wang,Jing Wang,Haimei Wen,Yuqi Zhao,Jiayu Zhang,Qiushi Xu,Zhaozhao Zhao,Ting Ni

Drug resistance to a single agent is common in cancer-targeted therapies, and rational drug combinations are a promising approach to overcome this challenge. Many Food and Drug Administration-approved drugs can induce cellular senescence, which possesses unique vulnerabilities and molecular signatures. However, there is limited analysis on the effect of the combination of cellular-senescence-inducing drugs and targeted therapy drugs. Here, we conducted a comprehensive evaluation of cellular senescence using 7 senescence-associated gene sets. We quantified the cellular senescence states of ~10,000 tumor samples from The Cancer Genome Atlas and examined their associations with targeted drug responses. Our analysis revealed that tumors with higher cellular senescence scores exhibited increased sensitivity to targeted drugs. As a proof of concept, we experimentally confirmed that etoposide-induced senescence sensitized lung cancer cells to 2 widely used targeted drugs, erlotinib and dasatinib. Furthermore, we identified multiple genes whose dependencies were associated with senescence status across ~1,000 cancer cell lines, suggesting that cellular senescence generates unique vulnerabilities for therapeutic exploitation. Our study provides a comprehensive overview of drug response related to cellular senescence and highlights the potential of combining senescence-inducing agents with targeted therapies to improve treatment outcomes in lung cancer, revealing novel applications of cellular senescence in targeted cancer therapies.

Hyperbranched polyglycerol-grafted WOx nanowires: Synthesis, characterization, functionalization and as effective drug targeted delivery vehicle

Binglong Yu,Jie Wang,Xiaoyun Mo,Xiaoxin Yang,Wenlong Wang,Xiulan Cai

Tungsten oxide (WO x ) is an important n-type semiconductor, which has attracted wide attention in the fields of gas sensing materials, photocatalysis , photoluminescence and electrochemistry due to its excellent electro-optical properties. However, the application potential of WO x -based nanomaterials in biomedicine has been neglected. In this paper, we firstly synthesized tungsten oxide nanowires (WO x NWs) by one-step method using tungsten chloride and triethylene glycol as raw materials, and reported an effective surface engineering strategy for polyglycerol-functionalized tungsten oxide nanowires (WO x -PG) to enhance their colloidal stability, biocompatibility , and selective toxicity to specific cancer cells in dispersion media . The WO x -PG was further derivatized and covalently combined with FA (WO x -PG-FA) as a drug targeting vector to deliver doxorubicin (DOX) to cancer cells. Compared with WO x NWs, WO x -PG exhibits smaller size (30−90 nm), more uniform distribution, better solubility and biocompatibility. The hemolysis rate of PG-modified WO x NWs was significantly reduced, being only 1.35 % at 200 μg/ml. The in vitro drug release percentage of WO x -PG-FA/DOX reached 80.78 % within 48 h at pH 5.2. In vitro toxicity assays and confocal laser scanning microscope (CLSM) results indicated that PG-functionalized WO x NWs were not cytotoxic, and WO x -PG-FA/DOX had highly selective cytotoxicity on HeLa cells. These results indicate that WO x -PG-FA is a potential drug carrier for effective targeting of cancer therapy.

Gold-Aptamer-Modified Metal–Organic Framework Drug Delivery Nanosystems for Combined Photothermal/Chemotherapy of Cancer

Ting Fang,Zihao Duan,Nan Wan,Xiaoxuan Zuo,Zhongwu Pan,Weiwei Zhang,Fei Ge,Lin Gui

The poor targeting and high toxicity of chemotherapeutic drugs make them less effective against breast cancer. In this study, in accordance with their photothermal effect, the aptamer (Apt) (AS1411) and doxorubicin (DOX) were linked to zeolite imidazole salt frame8 (ZIF-8) by using gold nanorods (Au NRs). Thereby, a multifunctional targeting nanoparticle platform (DOX/Au-Apt@ZIF-8) was constructed to deliver and effectively utilize DOX and realize the antitumor effects of the multimodal combination of chemotherapy–target–photothermal therapy. This platform was responsive to both pH and near-infrared (NIR) light. In the characterization experiments, DOX/Au-Apt@ZIF-8 exhibited good dispersibility, particle size stability, and consistent spectral characteristics. The photothermal conversion efficiency was up to 53.5%. In the cell uptake and cell cycle experiments, DOX/Au-Apt@ZIF-8 effectively targeted tumor cells and blocked their growth in the S phase. Moreover, in an in vivo study, DOX/Au-Apt@ZIF-8 exerted good inhibitory effects and photothermal effects on tumors. When combined with NIR irradiation, DOX/Au-Apt@ZIF-8 inhibited tumor tissue growth by enhancing and inducing apoptosis. Importantly, histopathological analysis revealed that the nanoparticles were less toxic to other organs and had high biosafety. As such, DOX/Au-Apt@ZIF-8 fabricated in this study is expected to offer a treatment approach against breast cancer.

Construction, Characterization, and Antidepressant Application of Blood Brain Barrier-Targeted Mahonia bealei Monomer Nanoparticles

Junhui He,Dongmei Li,Jiaxiu Xie,Kedao Lai,Juying Zhou

This study aimed to develop Mahonia bealei alkaloid (MA)-loaded nanoparticles (NPs) targeting the blood-brain barrier (BBB) for the treatment of depression. The MA@NPs were prepared via self-assembly using transferrin-modified chitosan-N, O-carboxymethyl chitosan (NOCMS-CS-Tf) as the carrier. Formulation optimization yielded uniform MA@NPs (~192 nm, +29 mV) with a drug loading of ~16% and encapsulation efficiency of ~80%. Transmission electron microscopy (TEM) and scanning electron microscopy (SEM) revealed spherical MA@NPs with an average size of ~55 nm under TEM, rough and porous surfaces, and good dispersibility. X-ray diffraction (XRD) analysis showed that the MA@NPs exhibited amorphous characteristics with reduced crystallinity after MA loading, while Fourier transform infrared spectroscopy (FTIR) confirmed successful MA encapsulation via hydrogen bonding and intermolecular interactions. The MA@NPs remained stable for over one month at 4 °C. In vitro release assays showed a sustained-release profile under conditions mimicking the depressive state. Consistently, in vivo fluorescence imaging indicated prolonged systemic retention, corroborating the sustained-release behavior of the MA@NPs. Cellular uptake experiments indicated enhanced BBB-targeting ability of MA@NPs especially under conditions of cellular stress or damage. Compared to free MA, the MA@NPs significantly alleviated lipopolysaccharide (LPS)-induced neuroinflammation in BV2 microglial cells and corticosterone (CORT)-induced neuronal damage in PC12 cells. In vivo, superior anti-neuroinflammatory effects compared to free MA were demonstrated in an LPS-induced mouse model of depression. Network pharmacology analysis, supported by qRT-PCR validation, revealed that both MA and MA@NPs activated the cAMP signaling pathway by upregulating the mRNA expression of protein kinase A (PKA), cAMP response element-binding protein (CREB), and brain-derived neurotrophic factor (BDNF). In conclusion, MA@NPs enhance the antidepressant efficacy of MA through sustained drug release, targeted brain accumulation, and modulation of neuroinflammation and neuroplasticity-related pathways.

Microenvironment responsive nanoplatform for targeted removal of cholesterol and reshaping inflammatory microenvironment in atherosclerotic plaques

Lin Shen,Chengli Jiang,Jingjing Song,Yanran Bi,Weiqian Chen,Chenying Lu,Minjiang Chen,Lingchun Lv,Risheng Yu,Jianhua Zou,Zhongwei Zhao,Xiaoyuan Chen,Jiansong Ji

The microenvironment of atherosclerotic plaques features abundant cholesterol crystals (CCs), reactive oxygen species (ROS), and pro-inflammatory M1 macrophages, leading to the progression and mortality of atherosclerotic cardiovascular disease (ASCVD). Targeted removal of CCs and scavenging of ROS are crucial for treatment of ASCVD. In this study, an intelligent nanoformulation consisting of epigallocatechin gallate (EGCG), cysteine (Cys), ursodeoxycholic acid (UDCA) and VHPK (Valine-Histidine-Proline-Lysine)-Lipo, was designed to precisely target and remodel the inflammatory microenvironment of atherosclerotic plaques. With the assistance of VHPK peptide-modified liposomes, this nanoplatform significantly enhances the targeting ability and therapeutic efficacy. UDCA promotes cholesterol dissolution and efflux and improves cholesterol clearance in foam cells by over 70 %. EGCG, an antioxidant, significantly enhances ROS clearance by approximately 50 %. EGCG/Cys/UDCA@VHPK-Lipo boosts plaque clearance efficiency by 75.43 % in vivo . RNA-seq analysis reveals that these nanoparticles upregulate ATP-binding cassette transporters A1 ( ABCA1 ) and G1 ( ABCG1 ) expression, promoting cholesterol metabolism and M2 macrophage polarization. Additionally, inflammation markers in plasma and plaque tissues were significantly reduced, indicating good biocompatibility and safety. EGCG/Cys/UDCA@VHPK-Lipo demonstrates superior plaque clearance and safety both in vitro and in vivo , offering promising clinical potential for atherosclerosis treatment.

Chemically Surface Tunable Solubility Parameter for Controllable Drug Delivery—An Example and Perspective from Hollow PAA-Coated Magnetite Nanoparticles with R6G Model Drug

Quanguo He,Jun Liu,Jing Liang,Xiaopeng Liu,Du Tuo,Wen Li

Solubility parameter-dependent drug releasing property is essential in practical drug delivery systems (DDS), and how to combine magnetic nanoparticles(NPs) and suitable polymer coating towards DDS is always a crucial and valuable challenge in biomedical application. Herein, a controllable drug delivery model with a surface having a chemically tunable solubility parameter is presented using hollow magnetite/polyacrylic acid (Fe3O4/PAA) nanocomposites as nanocarrier towards DDS. This composite is prepared by simply coating the modified hollow Fe3O4with PAA. The coating amount of PAA onto the surface of Fe3O4(measured by TGA) is about 40% (w/w). Then, Rhodamine 6G (R6G) is selected as model drug in drug delivery experiment. The efficiency of drug loading and drug release of these Fe3O4/PAA nanocarriers are evaluated under various temperature, solvent and pH values. As a result, the best drug releasing rate was achieved as 93.0% in pH = 7.4 PBS solution after 14 h. The releasing efficiency is 86.5% in acidic condition, while a lower releasing rate (30.0%) is obtained in aqueous solution, as different forms (polyacrylic acid and polyacrylate) of PAA present different solubility parameters, causing different salt and acid effects in various solvents, swelling property of PAA, and binding force between PAA and R6G. Therefore, by changing the solubility parameter of coating polymers, the drug delivery properties could be effectively tuned. These findings prove that the DDS based on magnetic particle cores and polymer encapsulation could efficiently regulate the drug delivery properties by tuning surface solubility parameter in potential cancer targeting and therapy. Keywords:hollow Fe3O4NPs;Fe3O4/PAA composite NPs;solubility parameter;controlled release;drug delivery systems

RVG29-modified docetaxel-loaded nanoparticles for brain-targeted glioma therapy

Hongchen Hua,Xuemei Zhang,Hongjie Mu,Qingqing Meng,Ying Jiang,Yiyun Wang,Xiaoyan Lu,Aiping Wang,Sha Liu,Yaping Zhang,Zhihui Wan,Kaoxiang Sun

Gliomas are the most common malignant brain tumor, but treatment is limited by the blood–brain barrier (BBB), especially for chemotherapeutic drugs. Although some chemotherapy drugs can pass through the BBB, many of these agents are toxic to normal brain tissue. To maximize therapeutic effects, chemotherapeutic drugs must accumulate at the glioma site. In this study, a specific ligand (the RVG29 peptide) that can combine with acetylcholine receptors was conjugated to polyethylene glycol-modified poly-( d , l -lactide-co-glycolide) (PEG-PLGA) to develop a targeted carrier; preparation of the targeted docetaxel nanoparticles (DTX-NPs) was performed by the nanoprecipitation method. The NPs were approximately 110  nm and had smooth surfaces. Enzyme-linked immunoassay results showed that the amount of receptor on the surface of glioma cells was 2.04-fold higher than that of nonmalignant cells, which may promote accumulation of RVG29-modified NPs at the targeting site. NPs showed targeting properties for glioma cells compared with the non-targeting NPs in an in vitro cellular uptake test. Targeted NPs also showed better BBB penetration in an in vitro model. In vivo tests indicated that RVG29-PEG-PLGA-NPs could selectively accumulate in intracranial glioma tissue. In conclusion, these results indicated that the RVG29-modified NPs have potential efficacy for glioma therapy.
