Graphene hydrogel has shown greatly potentials in bone tissue engineering recently, but it is relatively weak in the practical use. Here we report a facile method to synthesize high strength composite graphene hydrogel. Graphene oxide (GO), hydroxyapatite (HA) nanoparticles (NPs) and chitosan (CS) self-assemble into a 3-dimensional hydrogel with the assistance of crosslinking agent genipin (GNP) for CS and reducing agent sodium ascorbate (NaVC) for GO simultaneously. The dense and oriented microstructure of the resulted composite gel endows it with high mechanical strength, high fixing capacity of HA and high porosity. These properties together with the good biocompatibility make the ternary composite gel a promising material for bone tissue engineering. Such a simultaneous crosslinking and reduction strategy can also be applied to produce a variety of 3D graphene-polymer based nanocomposites for biomaterials, energy storage materials and adsorbent materials.

To meet the progressive requirements for bone regeneration purpose, injectable hydrogels have attracted increasing attention in tissue regeneration and local drug delivery applications. In this study, we report a facile method to prepare injectable and degradable polysaccharide-based hydrogels doubly integrated with hydroxyapatite (HAp) nanoparticles and calcium carbonate microspheres (CMs) under physiological condition. The mechanism of cross-linking is attributed to the Schiff-base reaction between amino and aldehyde groups of carboxymethyl chitosan (CMCS) and oxidized alginate (OAlg), respectively. Synchronously, tetracycline hydrochloride (TH) loaded CMs were fabricated by the precipitation reaction with an average diameter of 6.62 μm. To enhance bioactive and mechanical properties, nano-HAp and CMs containing TH were encapsulated into the polysaccharide-based hydrogel to form injectable gel scaffolds for imitation of bone niche. The gelation time, morphology, mechanical properties, swelling ratio and in vitro degradation of the gel scaffolds could be controlled by varying HAp and CMs contents. Moreover, the composite gel scaffolds had good sustained drug release and antibacterial properties, as confirmed by drugs release calculation and antibacterial evaluation. In addition, the gel scaffolds were found to be self-healing due to dynamic equilibrium of the Schiff-base linkages. These results suggested that the prepared composite gel scaffolds hold great potential for drug delivery and regeneration of irregular bone defects.

In this study, in order to better mimick the nature of bone extracellular matrix, hyaluronic acid (HA) hydrogels having a triple degradation behavior were synthesized from 3,3′-dithiodipropionate hydrazide-modified HA (DTPH–HA) and polyethylene glycol dilevulinate (LEV–PEG–LEV) via the reaction of the ketone carbonyl groups of LEV–PEG–LEV with the hydrazide groups of DTPH–HA. The HA hydrogels were characterized by solid state 13 C NMR, FT-IR, SEM, and rheological, swelling and degradation tests. The results showed that the HA hydrogels exhibited a highly porous morphology and had pore diameters ranging from 20 to 200 μm. The equilibrium swelling ratio of the HA hydrogels was no less than 37.5. The HA hydrogels could be degraded by hyaluronidase and reducing substances or at acidic pH values. The biocompatibility of the HA hydrogels was evaluated using osteoblast-like MC3T3-E1 cells by live/dead staining and MTT assays. The results revealed that the HA hydrogels had good biocompatibility and could support the attachment and proliferation of MC3T3-E1 cells. All the results indicated that the HA hydrogels synthesized by hydrazone bond crosslinking might have great potential to be used in bone tissue engineering.

Homogeneous gelatin/hydroxyapatite (GEL/HA) nano-composites were synthesized by a novel in situ precipitation method, and its corresponding characterizations, including composition, morphology, pore structure, thermal stability, mechanical strength, and biocompatibility, were carried out. High-magnified scanning electron microscope (SEM) images indicated that nano-HA with particle size ranging from 20 to 50 nm were uniformly distributed in GEL matrix and tightly integrated with organic phase. Wide angle X-ray diffraction (XRD) analysis and transmission electron microscope (TEM) images showed that, during the process of mineralization, there existed preferred oriented growth of HA crystals along (002) and (211) crystal planes. Thermogravimetric analysis (TGA) and differential thermal analysis (DTA) indicated that, the thermal stability of GEL molecules enhanced by hybridizing with HA nanocrystals. Interconnected porous GEL/HA nanocomposites with pore size ranging between 50 and 350 μm were prepared by a freeze-drying method. This pore size was adequate for bone tissue engineering (BTE) applications. In addition, in vitro MG63 osteoblast-like cell culture illuminated that GEL/HA nanocomposites had excellent cytocompatibility and could promote proliferation of cells. These results suggested that GEL/HA nanocomposite might be an ideal bone substitute. POLYM. COMPOS., 38:1579–1590, 2017. © 2015 Society of Plastics Engineers

Injectable and biodegradable alginate-based composite gel scaffolds doubly integrated with hydroxyapatite (HAp) and gelatin microspheres (GMs) were cross-linked via in situ release of calcium cations. As triggers of calcium cations, CaCO 3 and glucono- d -lactone (GDL) were fixed as a mass ratio of 1:1 to control pH value ranging from 6.8 to 7.2 during gelation. Synchronously, tetracycline hydrochloride (TH) was encapsulated into GMs to enhance bioactivity of composite gel scaffolds. The effects of HAp and GMs on characteristics of gel scaffolds, including pH value, gelation time, mechanical properties, swelling ratio, degradation behavior and drug release, were investigated. The results showed that HAp and GMs successfully improved mechanical properties of gel scaffolds at strain from 0.1 to 0.5, which stabilized the gel network and decreased weight loss, as well as swelling ratio and gelation time. TH could be released from this composite gel scaffold into the local microenvironment in a controlled fashion by the organic/inorganic hybrid of hydrogel network. Our results demonstrate that the HAp and GMs doubly integrated alginate-based gel scaffolds, especially the one with 6% (w/v) HAp and 5% (w/v) GMs, have suitable physical performance and bioactive properties, thus provide a potential opportunity to be used for bone tissue engineering. The potential application of this gel scaffold in bone tissue engineering was confirmed by encapsulation behavior of osteoblasts. In combination with TH, the gel scaffold exhibited beneficial effects on osteoblast activity, which suggested a promising future for local treatment of pathologies involving bone loss.

Hydrogels fabricated from natural polysaccharides may serve as ideal scaffolds for tissue engineering because of their similarity to the extracellular matrices. In this study, novel hydrogel scaffolds with bubble-like porous structure were prepared from hydroxyethyl chitosan (HECS) and cellulose (CEL) by a combination of chemical crosslinking, particle-leaching using silicon dioxide particles as porogen and freeze-drying method. The morphology, compression stress-strain curves, wettability, and swelling and rheological behaviors of the HECS/CEL scaffolds were characterized by SEM, mechanical test, contact angle measurement and rheometer. HECS/CEL scaffolds had good comprehensive performances and could reach equilibrium swelling state in water within 20s. The results from in vitro biocompatibility evaluated using SEM, live/dead cell viability and MTT assays demonstrated that the HECS/CEL scaffolds could well support the attachment, spreading and proliferation of osteoblastic MC3T3-E1 cells and showed good biocompatibility. Therefore, the novel HECS/CEL scaffolds may be promising for bone tissue engineering applications.

Man-made materials are easily available, processed and modified during bone defect repairing, without worrying about potential infections, immunological incompatibility, sterility, or donor site morbidity. Collagen(Col)/nano-hydroxyapatite (nHA) composite nanofibers based on ordered nHA ceramic fibers was prepared by sintering electrostatic spinning PVP/HA composite fibers followed by impregnation with collagen solution. Without requiring strict control of temperature or humidity, this method prevented nHA aggregation during the sintering process and reduced the use of costly Col as compared with direct electrospinning. The composite nanofiber membrane with bone-like components and structures has a great potential for application in bone tissue engineering. And the method implement easily.. The mineralization and degradation characteristics of the nanofiber biocomposite are beneficial for the bone conductivity and MSCs growth and metabolism. Protein adsorption of Col/HA nanofiber biocomposite is outstanding. There is no significant difference in cells viability between the experiment group and the control group, and Col/HA nanofibers biocomposite has weak cytotoxicity to MSCs. Overall, the approach successfully prepared Col/HA nanofibers biocomposite at common electrospinning environment with uniformly distributed high content nHA, low Col concentration and less Col consumption, all of which make it an excellent choice for the bone tissue engineering.

In this research, gelatin-tussah silk fibroin/hydroxyapatite (GEL-TSF/HAp), gelatin-Bombyx mori silk fibroin/hydroxyapatite (GEL-BMSF/HAp), and gelatin/hydroxyapatite (GEL/HAp) nano-composites were synthesized by a novel in situ precipitation method. Characterizations, including surface morphology, elemental composition and distribution, structure of the crystalline phase, mechanical strength, thermal stability, and in vitro cytocompatibility, were carried out. Investigations on the crystalline phase showed that rod-like HAp crystallites in the GEL-TSF/HAp composite had a higher aspect ratio than those in the GEL-BMSF/HAp composite and the GEL/HAp composite. In addition, the GEL-TSF/HAp composite also presented better thermal stability than the other two composites revealed by differential thermal analysis (DTA) and thermogravimetric analysis (TGA). Mechanical properties testing indicated that the GEL-TSF/HAp composite had a higher elastic modulus at low strain and higher compressive modulus at high strain simultaneously than the other two composites. An in vitro cell culture showed that MG63 osteoblast-like cells on the GEL-TSF/HAp membrane took on higher proliferative potential than those on the GEL-BMSF/HAp membrane. These results indicated that compared to the GEL-BMSF/HAp composite, the GEL-TSF/HAp composite was more suitable for bone tissue engineering (BTE) applications.

The strategy of incorporating bioactive inorganic nanomaterials without side effects as osteoinductive supplements is promising for bone regeneration. In this work, a novel biomass nanofibrous scaffold synthesized by electrospinning silica (SiO2) nanoparticles into polycaprolactone/chitosan (PCL/CS) nanofibers was reported for bone tissue engineering. The nanosilica-anchored PCL/CS nanofibrous bioscaffold (PCL/CS/SiO2) exhibited an interlinked continuous fibers framework with SiO2nanoparticles embedded in the fibers. Compact bone-derived cells (CBDCs), the stem cells derived from the bone cortex of the mouse, were seeded to the nanofibrous bioscaffolds. Scanning electron microscopy and cell counting were used to observe the cell adhesion. The Counting Kit-8 (CCK-8) assay was used. Alkaline phosphatase (ALP), Alizarin red staining, real-time Polymerase Chain Reaction and Western blot tests were performed to confirm the osteogenesis of the CBDCs on the bioscaffolds. The research results demonstrated that the mechanical property of the PCL together with the antibacterial and hydrophilic properties of the CS are conducive to promoting cell adhesion, growth, migration, proliferation and differentiation. SiO2nanoparticles, serving as bone induction factors, effectively promote the osteoblast differentiation and bone regeneration. This novel SiO2-anchored nanofibrous bioscaffold with superior bone induction activity provides a better way for bone tissue regeneration. Keywords:silica;polycaprolactone;chitosan;bioscaffold;osteogenesis;bone tissue engineering

Thiolated chitosan (CS-NAC) was synthesized and the selected CS-NAC was used together with silk fibroin (SF) to produce dual network CS-NAC/SF hydrogels. The CS-NAC/SF solutions with formulated compositions were able to form hydrogels at physiological temperature and pH. Rheological measurements showed that elastic modulus of some CS-NAC/SF gels could reach around 3 kPa or higher and was much higher than their respective viscous modulus, indicating that they behaved like strong gels. Deformation measurements verified that CS-NAC/SF gels had well-defined elasticity. The optimized CS-NAC/SF gels exhibited jointly enhanced properties in terms of strength, stiffness and elasticity when compared to the gels resulted from either CS-NAC or SF. Examinations of dry CS-NAC/SF gels revealed that they were highly porous with well-interconnected pore features. Cell culture demonstrated that CS-NAC/SF gels supported the growth of chondrocytes while effectively maintaining their phenotype. Results suggest that these dual network gels have promising potential in cartilage repair.

Sol-gel derived organic/inorganic hybrids, in which organic and inorganic components form co-networks at the molecular level, have demonstrated great potential for providing improved mechanical properties and biological functions in tissue engineering applications. Here, a novel bioactive hydroxyapatite-chitosan-silica hybrid (HA-CSH) scaffold was successfully fabricated by combining the sol-gel method and 3D plotting technique. Physiochemical characterization confirmed that chitosan was hybridized homogeneously with the inorganic phase on nanoscale. The obtained scaffolds possessed precisely controllable and interconnected porous structures. The nano-sized HA formed in situ and dispersed uniformly in the hybrid network, which reduced the water absorption and increased the mechanical strength of the hybrid scaffold under humidity condition as compared to chitosan-silica hybrid (CSH) scaffold. Compression tests showed that the 3D plotted hybrid scaffolds under wet conditions had compressive strengths of 10–13 MPa and elastic moduli of 21–27 MPa and thus met the mechanical requirements of human trabecular bone. Studies on the mineralization process under simulated body fluid (SBF) conditions confirmed that the introduction of HA obviously increased the biological activity of hybrid scaffolds. In vitro cell results indicated that the HA-CSH scaffold not only supported adhesion and proliferation of mouse bone mesenchymal stem cells (mBMSCs), but also improved the osteoinductivity. The alkaline phosphatase activity and mineral deposition on the HA-CSH scaffold were higher than those on the CSH scaffold. These results suggested that the 3D plotted HA-CSH scaffold may be a promising bioactive material for bone tissues regeneration .

A great interest has been shown in the injectable scaffolds for cartilage tissue regeneration because it can fill irregularly shaped defects easily through minimally invasive surgical treatments. Herein, we developed a new injectable three-dimensional (3D) alginate hydrogel loaded with biodegradable porous poly(ε-caprolactone)–b-poly(ethylene glycol)–b-poly(ε-caprolactone) microspheres (MPs/Alg) as the calcium gluconate container to cross-link alginate. Suspensions of chondrocytes/alginate and porous microspheres turned into a gel because of the release of calcium gluconate; thus, the injectable composite hydrogels give a 3D scaffold to fit the defects perfectly and integrate the extracellular-matrix-mimicking architecture to efficiently accommodate cartilage cells in situ. Tissue repair in a full-thickness cartilage defect model was controlled at 6, 12, and 18 weeks after the implant by micro-CT and immunohistochemistry to evaluate the healing status. The defect in the MPs/Alg+ cells group achieved an almost complete repair at 18 weeks, and the repaired chondrocytes regained a normal tissue structure. Moreover, the MPs/Alg+ cells-treated group increased the quality of tissue formed, including the accumulated glycosaminoglycan and the uniformly deposited type II collagen. The results point out the promising application of the injectable MPs/Alg-chondrocytes system for cartilage tissue engineering.
