A label-free AuNP bioprobe-assisted CRISPR/Cas12a colorimetric platform for high-throughput detection of Staphylococcus aureus ST398

Baoqing Zhou,Qinghua Ye,Moutong Chen,Chufang Wang,Xinran Xiang,Ying Li,Jumei Zhang,Youxiong Zhang,Juan Wang,Shi Wu,Qihui Gu,Yu Ding,Qingping Wu

Rapid and accurate screening of foodborne pathogens in contaminated food is with great significance for food safety. AuNPs-based colorimetric assay is an attractive strategy in the field of food safety inspection, but is limited by tedious probe preparation, low color rendering resolution, and narrow color gamut. Herein, we proposed an e nhanced c olorimetric p latform based on the C RISPR/Cas12a system and label-free D NA-AuNP p robe (CDP-ECP) for high-throughput detection of Staphylococcus aureus ST398. In this strategy, a novel and specific gene targeting S. aureus ST398 was identified and efficient AuNP-bioprobe was successfully prepared. Through a PCR/RAA amplification, the trans -ssDNA cleavage ability induced by target DNA product triggered an aggregation performance for the prepared AuNP-bioprobe pairs, which could realize target colorimetric detection within 50 min. Under optimal conditions, the colorimetric assay could detect as low as 6.4 × 10 2  cfu/mL of S. aureus ST398 in pure culture. Specific gene mining and target-induced trans -cleavage character gave this method a superior specificity and anti-interference. More importantly, this colorimetric method can be extended to a high-throughput platform for rapid detection of spiked and natural sample by using a common microplate reader. Since it is simple to operate, low cost, high sensitivity and specificity, this new platform is fully qualified for clinical diagnosis, environmental monitoring onsite testing requirements.

A ternary AuNP/NiFe-LDH/MWCNT heterostructure enhanced electrochemical biomimetic sensor for sensitive detection of enrofloxacin in aquatic foods

Shuanghuan Gu,Sen Wang,Yongju Zhang,Zhixiang Xu,Longhua Xu

The development of sensitive and selective electrochemical sensors for antibiotic detection is critical to ensuring food safety. Herein, we report a novel biomimetic electrochemical sensor using a ternary AuNP/NiFe-LDH/MWCNT heterostructure combined with molecularly imprinted polymers (MIPs) for detecting enrofloxacin (ENR) in food. The AuNP/NiFe-LDH/MWCNT composite was prepared by in-situ reducing HAuCl 4 on a NiFe-LDH/MWCNT backbone, forming a heterojunction structure that enhances charge separation and electron transfer. Electropolymerization of pyrrole with ENR produced highly selective MIP layers. The synergistic integration of high conductivity, abundant catalytic sites, and selective molecular recognition endows the sensor with excellent electrochemical performance. The sensor exhibits a wide linear range (0.05–100.0 μM) and an ultralow limit of detection (LOD) 0.025 μM. It showed superior selectivity, reproducibility, stability, and applicability in complex aquatic food samples, with recoveries of 94.6 % to 103.9 %. This work paves the way for practical, high-precision monitoring of trace antibiotics in food safety applications.

The controllable synthesis and enhanced gas sensing performances of AuNP-modified ZnSnO3 hollow nanocubes toward highly sensitive toluene detection

Jun Li,Yongjiao Sun,Zhaomin Tong,Zhenting Zhao,Wendong Zhang,Jie Hu,Lin Chen

Morphology control and noble-metal modification have become effective ways to improve the gas sensing performances of mixed-metal-oxide sensors. In this study, ZnSnO3 nanocube structures with different morphologies were synthesized via combining NaOH-assisted dissolution and a calcination process, and they were then modified with various amounts of Au nanoparticles through a chemical reduction method. Some important parameters were systematically investigated to optimize the gas sensing performance toward toluene detection. Noticeably, the as-prepared Au2NPs@ZnSnO3 hollow nanocube sensor exhibits an ultra-high response (80.82@100 ppm), fast response and recovery times (11 s/12 s, respectively), a low limit of detection (10 ppb), excellent repeatability, long-term stability, and good anti-humidity properties. The enhanced gas sensing mechanism of the Au2NPs@ZnSnO3 sensor is also thoroughly discussed, which can be attributed to the unique hollow nanocube structure and the excellent electron sensitization and chemical sensitization properties of AuNPs. As a result, the optimum AuNP-modified ZnSnO3 sensing material is promising for high-sensitivity toluene detection for practical applications.

A label-free membrane-based biosensor array with AuNP-modified PDMS for sensitive and specific detection of alpha-fetoprotein

Zhao Zhongfu,Zhao Dong,Wang Haoyu,Yu Xingbo,Wu Sitong,Ma Jianan,Guo Xing,Ji Jianlong,Zhou Chuangang,Sang Shengbo

The surface stress–based transduction enabled by antibody functionalization has emerged as a promising paradigm for label-free biosensor development. However, the inherent limitations of conventional detection methods (e.g., high cost, complex modification, insufficient sensitivity) and the challenge of achieving high-throughput analysis hinder their practical clinical application. Herein, a high-throughput label-free biosensor array based on multi-component integration of gold nanoparticles (AuNPs), polydimethylsiloxane (PDMS) membrane, and conductive silver paste is proposed for sensitive and specific detection of alpha-fetoprotein (AFP), a pivotal biomarker for hepatocellular carcinoma (HCC). The specific immunoreaction between immobilized anti-AFP antibodies and target AFP induces compressive surface stress, which triggers PDMS membrane deformation and regulates the electrical resistance of the AuNP conductive layer, realizing the stress-electric coupling transduction. Systematic optimization of antibody surface density (50 μg/mL) and AuNP layer thickness (0.67 μm) maximizes antigen capture efficiency and mechano-electrical transduction performance. The proof-of-concept application for AFP detection demonstrates remarkable analytical performance: a linear detection range of 0–50 ng/mL, a low limit of detection (LOD) of 0.225 ng/mL, high specificity against interfering biomolecules. Furthermore, accurate detection of AFP in clinical serum samples (relative error < 5.05%, RSD < 2.97%) confirms its practical applicability. All these results indicate that the integrated design and parameter optimization of the AuNP-PDMS-based biosensor array enhance the performance of surface stress biosensors, providing a cost-effective platform for the sensitive detection of clinical cancer biomarkers. Entropy-Driven Circuit Integrated with Ligases to Regulate DNA-AuNP Network Disintegration for Colorimetric Detection of Single Nucleotide Polymorphisms

Yunshan Zhang,Tuo Huang,Fang Yang,Qianglong Tan,Jing Ye,Xianzhong Feng,Diming Zhang

In recent years, entropy-driven circuit (EDC) dynamic DNA networks have garnered significant attention in nucleic acid detection owing to their simplicity, efficiency, and flexible design. Nevertheless, conventional EDC reactions face a constraint in achieving optimal signal amplification due to a solitary and feeble driving force. To overcome this limitation, we innovatively devised a gold nanoparticle (AuNP) dispersion-enhanced EDC (Au-EDC) approach, pioneering a novel colorimetric signal amplification and output system. The system was harmoniously integrated with the ligase chain reaction (LCR) for precise single nucleotide polymorphism (SNP) genotyping. Specifically, LCR was selectively executed solely on the positive strand of the mutant target (MT), facilitating precise point-to-strand information transduction. Subsequently, the LCR product triggered the Au-EDC cycling reaction, causing the DNA-AuNPs network to progressively disintegrate and release a pronounced colorimetric signal. This strategic design ingeniously harnessed the entropy increase that occurs as AuNPs undergo a transition from aggregated to dispersed states, offering a supplemental impetus for the EDC cycle. The integrated LCR-Au-EDC system excelled in detecting MT at concentrations as low as 320 fM and differentiating pooled samples with mutation frequencies as low as 0.1%. Moreover, the system accurately performed SNP genotyping on the real genomes derived from soybean leaves. Consequently, this study not only develops a colorimetric signal amplification and output sensing system based on EDC reactions but also provides a cost-effective and efficient SNP genotyping tool.

A AuNP-capped cage fluorescent biosensor based on controlled-release and cyclic enzymatic amplification for ultrasensitive detection of ATP

Wei Wang,Xin Li,Kai Tang,Zhiling Song,Xiliang Luo

Gold nanodevices have attracted extensive interest in the detection of specific targets within cells. However, constructing gold sensing devices that can be activated by the simulation of remote applications remains a huge challenge. Here, we report a Au nanoparticle (AuNP)-capped cage fluorescent biosensor based on controlled-release and Exonuclease III (Exo III) assisted cyclic enzymatic amplification that can be activated by adenosine triphosphate (ATP). In the system, AuNPs were used as the building blocks to cap the pores of Au nanocages (AuNCs) loaded with Rhodamine B (RhB) molecules through the hybridization of DNA. The RhB fluorescent molecules were finally released with the help of Exo III in the presence of ATP for detection purposes. Ultimately, the biosensor leads to a wide linear ATP detection range from 1.0 × 10−9 to 1.0 × 10−7 M with a limit of detection (LOD) down to 0.88 nM. In addition, it also has good selectivity for ATP to distinguish between ATP and ATP analogues such as cytidine triphosphate (CTP), guanosine triphosphate (GTP), and uridine triphosphate (UTP). Therefore, as a convenient and sensitive biosensor, it is expected to be widely used in the biomedical field.

Magnetic bead-based AuNP labelling combined with inductively coupled plasma mass spectrometry for sensitively and specifically counting cancer cells

WeiJuan Yang,Zhiming Xi,Xiaoxue Zeng,Ling Fang,Wenjing Jiang,Yongning Wu,LiangJun Xu,FengFu Fu

Early diagnostic methods are the most effective way to increase the survival rate of cancer patients. In this study, we present a novel method for sensitively and specifically counting cancer cells using a magnetic bead (MB)-based AuNP–aptamer labelling technique and inductively coupled plasma mass spectrometry (ICP-MS) detection. A MB-based AuNP–aptamer labelling technique can specifically recognize cancer cells and tag AuNPs to the cancer cells, which provides a platform for indirectly counting cancer cells via the detection of Au with ICP-MS. The method reported in this study combines the merits of a MB-based AuNP–aptamer labelling technique and ICP-MS detection, resulting in better specificity and stability, high sensitivity, short analysis time, and robust resistance to a complicated matrix. Especially, the employment of ICP-MS detection and the presence of MBs that facilitate the magnetic separation of target cells, not only greatly shorten the analysis time but also greatly enhance the resistance of our method to a complicated matrix. The proposed method can be used to count as few as 100 human hepatocellular carcinoma SMMC-7721 cells in 1 mL of serum with a recovery of 93–104% and a RSD < 4% (n = 5) within 3 hours. The success of this study provides a promising rapid approach for the early detection of human hepatocellular carcinoma cells in clinical diagnosis.

Electrodeposited rGO/AuNP/MnO2 Nanocomposite-Modified Screen-Printed Carbon Electrode for Sensitive Electrochemical Sensing of Arsenic(III) in Water

Yanqing Wu,Tao Zhang,Lishen Su,Xiaoping Wu

Herein, a simple and portable electrochemical sensor based on a reduced graphene oxide/gold nanoparticle/manganese dioxide (rGO/AuNP/MnO2) nanocomposite-modified screen-printed carbon electrode (SPCE) was constructed by the facile stepwise electrodeposition method and used for electrochemical detection of As(III). The resultant electrode was characterized for its morphological, structural, and electrochemical properties using scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), energy dispersive X-ray spectroscopy (EDX), cyclic voltammetry (CV), and electrochemical impedance spectroscopy (EIS). From the morphologic structure, it can be clearly observed that the AuNPs and MnO2alone or their hybrid were densely deposited or entrapped in thin rGO sheets on the porous carbon surface, which may favor the electro-adsorption of As(III) on the modified SPCE. It is interesting that the nanohybrid modification endows the electrode with a significant decrease in charge transfer resistance and an increase in electroactive specific surface area, which dramatically increases the electro-oxidation current of As(III). This improved sensing ability was ascribed to the synergistic effect of gold nanoparticles with excellent electrocatalytic property and reduced graphene oxide with good electrical conductivity, as well as the involvement of manganese dioxide with a strong adsorption property in the electrochemical reduction of As(III). Under optimized conditions, the sensor can detect As(III) via square wave anodic stripping voltammetry (SWASV) with a low limit of detection of 2.4 μg L−1and a linear range of 25–200 μg L−1. The proposed portable sensor shows the advantages of a simple preparation procedure, low cost, good repeatability, and long-term stability. The feasibility of rGO/AuNPs/MnO2/SPCE for detecting As(III) in real water was further verified. Keywords:electrochemical sensor;gold nanoparticles;metal oxides;nanocomposites;reduced graphene oxide;screen printed carbon electrode;arsenic

Target-Responsive DNA Nanoclaw for the On-Site Identification of Chinese Medicines with Naked Eye

Yuanhuan Yang,Feng Gao,Ying Liang,Lichao Guo,Yang Pan,Peng Cao,Yue Zhang

The identification of Chinese medicinal herbs occupies a crucial part in the development of the food and drug market. Although molecular identification based on real-time PCR offers good versatility and uniform digital standards compared with traditional methods, such as morphology, the dependence on large-scale equipment hinders spot detection and marketable applications. In this study, we developed a DNA nanoclaw for colorimetric detection and visible on-site identification of Chinese medicines. When specific miRNA is present, the DNAzyme is activated and cleaves the substrate strand, triggering the catalytic hairpin assembly (CHA) reaction and forming branched DNA junctions on AuNP-I. This can then capture AuNP-II through hybridization and facilitate their aggregation, resulting in a noticeable color change that is observable to the naked eye. By harnessing the dual amplification of DNAzyme and CHA, this highly sensitive nanoprobe successfully achieved specific identification of Chinese medicines. This offers a new perspective for on-site testing in the herbal market.

Adsorption-Driven SERS Enhancement by Ti3C2Tx Quantum Dot-AuNP Assembled Interfaces for Sensitive Detection of Sudan Dyes in Chili Powder

Ruoyu Zhang,Guanghua Li,Hongwei Zhang,Junfeng Zhang,Yonghuan Yan,Weijun Kang,Peng Xue,Lianfeng Ai,Kai Kang

Background Sudan dyes, as illegally added food colorants, pose significant risks to human health due to their potential toxicity and carcinogenicity. Conventional chromatographic and mass spectrometric methods for Sudan dye detection typically require labor-intensive sample pretreatment and involve substantial time and economic costs. Surface-enhanced Raman scattering (SERS) has emerged as a rapid and sensitive analytical technique; however, its application to lipophilic analytes remains challenging because of the limited affinity between hydrophobic targets and SERS-active substrates, hindering effective signal enhancement. Therefore, developing adsorption-enhanced SERS substrates for efficient enrichment and sensitive detection of hydrophobic dyes is of great significance for food safety monitoring. Results In this study, a high-performance SERS platform based on Ti 3 C 2 T x quantum dot (MQD)-AuNP assembled interfaces was developed, with preparation conditions carefully optimized to ensure stable performance. Adsorption performance was evaluated using multiple isotherm and kinetic models, demonstrating an ultrahigh adsorption capacity (>4000 mg/g). The MQD/AuNPs substrate exhibited ∼14-fold enhancement in SERS intensity compared to AuNPs alone, with excellent signal uniformity (mapping RSD < 15%) and reproducibility (RSD < 20%). The platform also showed a broad linear response range and stable signal output across repeated measurements. Using 1595 cm -1 as the characteristic peak, the method achieved a detection limit of 10 μg/kg and a quantification limit of 100 μg/kg. In chili powder, recoveries ranged from 95.2-120.5%, confirming reliable quantitative performance. Compared with conventional chromatographic methods and reported SERS approaches, this platform maintains comparable sensitivity while significantly improving detection efficiency. Significance This work establishes an adsorption-driven SERS enhancement strategy by integrating high-capacity MQDs with plasmonic nanostructures. The synergistic combination of analyte enrichment and signal amplification enables rapid and sensitive detection without increasing analytical cost. This platform provides a practical and efficient approach for on-site screening of lipophilic contaminants in complex food systems.

Adhesive AuNP tape-mediated hierarchical assembly of multicenter DNA nanocomplexes for tumor cell nucleus-targeted staged drug delivery in vivo

Congcong Li,Yansha Gao,Qian Li,Shasha Luo,Wenqiang Liao,Zai-Sheng Wu

Developing DNA nanostructures as delivery vectors into organelles is a key technical barrier because of their innate poor pharmacokinetic properties. Herein, we propose a unique concept for high precision cancer therapy by using small gold nanoparticles (AuNPs) as adhesive nano-tapes to fold a long string of aptamer-attached tetrahedral DNA pearls (Apt-Nano-Tetra) into a spherical multicenter DNA nanostructure that can serve as a nucleus-targeted drug delivery carrier (Apt-ADMC) with a 75~85 times enhanced doxorubicin (Dox)-loading capacity. After specifically entering target tumor cells, Dox-Apt-ADMC is disassembled into individual AuNPs surrounded by short Apt-Nano-Tetra, finally reaching cell nucleus and causing cell apoptosis. Even if Dox-Apt-ADMC is treated with fetal bovine serum for 12 h, the drug delivery ability is not compromised, contributing to high functional stability in circulation system. As such, the systemically-administrated Dox-Apt-ADMC shows 7 times enhanced tumor accumulation and almost 100% inhibits tumor growth without detectable systemic toxicity.

Construction of the Au Nanoparticle/Graphene Oxide/Au Nanotube (AuNP/GO/AuNT) Sandwich Membrane for Surface-Enhanced Raman Scattering Sensing

Haibao Zhang,Yongxing Lin,Chunhong Qiao,Liang Wang,Cheng Cai,Hui He,Xingyou Tian

Au nanotube-based composite membrane served as surface-enhanced Raman scattering (SERS) substrate with an ultralarge aspect ratio possesses an excellent flexibility and widely tunable surface plasmon resonance, and by introducing graphene oxide (GO) as a spacer layer, the SERS enhancement of the composite membrane is obviously better than those from the individual blocks of the Au nanotubes (AuNTS) membrane and the Au nanoparticle/graphene oxide (AuNP/GO) membrane. Such a “sandwich” (AuNP/GO/AuNT) structured membrane has a high SERS sensitivity and a wide tunability by controlling the size of Au nanoparticles and the thickness of graphene oxide, and the detection limits of the AuNP/GO/AuNT substrate for R6G and NBA are as low as 10–12 and 10–7 M, respectively; the large enhancement is attributed to the adsorption and chemical mechanism of graphene oxide and the physical mechanism of the Au nanoparticles and nanotubes (the electromagnetic field coupling between them).
