Pharmacokinetic Characteristics of 6-Hydroxykaempferol-3,6-Diglucoside in Normal and Diabetic Cardiomyopathy Mice

Rui Yao,Fuxin Han,Xiaoju Zhang

6-Hydroxykaempferol-3,6-diglucoside is an important flavonoid constituent of Carthamus tinctorius L., with potential antioxidant and myocardial protective effects. However, its pharmacokinetic characteristics in normal and diabetic cardiomyopathy (DCM) mice remain unknown. This study was designed to explore the pharmacokinetic differences of this component between normal mice and DCM mice induced by a high-fat diet combined with streptozotocin. All mice were intragastrically administered at a dose of 20 mg/kg. Plasma samples were collected at different time points, and the plasma concentration was determined by LC–MS/MS. Compared with normal mice, DCM mice showed significantly decreased AUC 0− t and C max , indicating reduced absorption and bioavailability, while plasma clearance was markedly increased, suggesting accelerated elimination. No significant differences in T max and t 1/2 were observed. In contrast to our previous study, another core safflower component, HSYA, exhibited enhanced absorption and delayed elimination under DCM conditions, showing an opposite trend. These findings suggest that DCM may exert distinct regulatory effects on different bioactive ingredients of safflower, providing a comprehensive experimental basis for the clinical application of safflower-containing preparations.

Metabolite Profiling and Identification of Semaglutide in Liver S9 Across Species and Rat Plasma

Yawen Cong,Chongzhuang Tang,Zhiqiang Li,Yang Gao,Lijuan Chen,Haibo Xu,Xingxing Diao

Semaglutide, a glucagon-like peptide-1 receptor agonist, stands as a paradigm of successful peptide drug development. Although absorption, distribution, metabolism and excretion characteristics of semaglutide have been thoroughly studied in both animals and humans, a reliable in vitro screening model for evaluating metabolic behavior of semaglutide and other peptides remains an unmet need in drug development. This study established a novel ultrahigh performance liquid chromatography coupled to high-resolution mass spectrometry method to elucidate the species-specific metabolic pathways of semaglutide. Semaglutide was incubated with liver S9 fractions from human, monkey, dog, rat, and mouse for 1 h, respectively. For the in vivo study, rats were subcutaneously injected with semaglutide (10 mg/kg) for subsequent plasma collection. Data-dependent acquisition was performed to obtain the fragment ions, enabling identification of drug-related materials. Notably, a diagnostic fragment ion at m / z 469, formed from the cleavage of side chain, facilitated metabolite screening. Consequently, a total of 31 metabolites were detected and characterized, constituting a comprehensive metabolic profile of semaglutide in liver S9 fractions and rat plasma. Major metabolic pathways involved hydrolysis of peptide backbone. Our findings provided a robust methodological framework for the screening and metabolism prediction of peptide candidates, supporting the comprehensive safety and efficacy assessment.

Metabolomics Analysis Reveals That Anoectochilus roxburghii Extract Enhances Immunity in Immunosuppressed Rats by Modulating Lipid Metabolism and Amino Acid Metabolism

Xu Fang,Ying Yang,Xinmei Cao,Yi Tao

Anoectochilus roxburghii (AR) has been reported to exhibit immunomodulatory activity. However, the underlying mechanism has not yet been fully elucidated. The present study employed a gas chromatography–mass spectrometry–based metabolomics approach, combined with histopathological and biochemical analyses, to investigate the metabolomic changes and therapeutic effects of AR in cyclophosphamide-induced immunosuppressed rats. The rats were divided into four groups: control group, model group, positive group, and AR-treated groups. The immunosuppressed model was established by intraperitoneal injection of cyclophosphamide (CTX, 150 mg/kg/day). The model group showed significant reductions in spleen and thymus indices, along with elevated levels of tumor necrosis factor-α (TNF-α) and interleukin-2 (IL-2) but decreased interleukin-10 (IL-10). AR treatment restored spleen and thymus indices and normalized TNF-α, IL-2, and IL-10 levels compared to the model group. Pattern recognition was employed to identify potential biomarkers. Pattern recognition analysis identified potential biomarkers: 26, 25, and 28 differential endogenous metabolites in the model, positive control, and AR-treated groups, respectively. Among these, 15 endogenous metabolites were validated using standard compounds, which demonstrated excellent linearity ( R 2 > 0.9991) and precision (intraday RSD < 4.82%, interday RSD < 4.39%). Recovery rates for the standards ranged from 95.40% to 104.91%, with RSD values below 5.97%. Pearson correlation analysis revealed that lactic acid, threonine, 5-oxoproline, palmitic acid, and arachidonic acid are positively correlated with spleen and thymus indices, whereas valine, urea, octaethylene glycol, and 2-palmitoyl glycerol are negatively correlated with these indices. Collectively, these findings demonstrate that AR extract enhances immunity in immunosuppressed rats by modulating lipid and amino acid metabolism.

Pharmacodynamic Substance Basis and Anti-Cervicitis Efficacy Study of Jing Dai Ning Jiao Nang Based on UHPLC-Q-TOF-MS/MS Technology

Ling Yuan,Xiao Liang,Huan Wu,Wenmin Wang,Liye Xu,Yulin Feng,Wugang Zhang

Jing Dai Ning Jiao Nang (JDNJN), a traditional Miao medicine from Guizhou Province, demonstrates clinical efficacy in treating gynecological disorders like dysmenorrhea but lacks mechanistic clarification. This study investigated JDNJN's anti-cervicitis effects and pharmacological basis using a 25% phenol mucilage-induced rat cervicitis model. The efficacy of JDNJN was evaluated through pathological examination, peripheral blood tests, HE staining, and ELISA assays. UHPLC-Q-TOF-MS/MS was used to analyze the chemical components of JDNJN, as well as prototype compounds and metabolites in the plasma, urine, and feces of rats after administration. The results showed that JDNJN ameliorated cervicitis-induced alterations in body weight, vulvar inflammation, organ index, white blood cell count, and cervical tissue structure in rats, and exerted therapeutic effects by regulating inflammatory cytokines. Additionally, 98 compounds were identified in JDNJN, primarily flavonoids, with 17 prototype compounds and 60 metabolites detected post-administration. Key metabolic pathways included deoxygenation, hydroxylation, methylation, hydroxylation combined with methylation, and sulfation. These findings establish the anti-inflammatory effect of JDNJN on cervicitis and clarify its pharmacodynamic material basis. This study provides foundational evidence for further exploration of JDNJN's active components and molecular mechanisms.

Optimization of Extraction of Yiwei Decoction by Network Pharmacology Combined With Response Surface Design and Bioactivity Evaluation

Wei Zhu,Zepeng Zhang,Zhiqiang Yan,Lei Zhang

Yiwei Decoction (YWD), a traditional Chinese medicine for nourishing yin and promoting fluid production, is a potential treatment for dry eye disease (DED). Network pharmacology identifies YWD’s active components and mechanisms for treating DED, while high-performance liquid chromatography (HPLC) detects active compounds in water-prepared YWD extracts. A Box–Behnken design with response surface methodology was used to optimize the extraction of these components. Using this method, acteoside (AC), quercetin (QR), and methylophiopogonanone A (MA) were detected as the main contributors to anti-DED, acting on SRC, AKT1, PIK3CA, and STAT3, which may affect the development of DED through lipid metabolism, atherosclerosis, and PI3K-Akt and MAPK pathways. Based on the analytic hierarchy process, taking the phytochemical composition and the proliferation rate of YWD on human corneal epithelial cells (HCECs) as comprehensive indicators, the best extraction parameters were obtained as liquid–solid ratio of 8 mL/g, extraction times of 1.636 times, and decocting time of 54.96 min. Verification based on actual conditions, the standardized score is 0.9417 ± 0.0386, which is very close to the predicted score of 0.9328, with an error of less than 1%. This reliable method aids in optimizing the extraction process of traditional Chinese medicine for specific diseases.

Chen Wang,Xueying Qin,Ying Wang,Weiping Wang,Shuai Sun,Hongsen Wang,Shiyu Li,Xiao Dai,Xianhong Zhu,Xun Gao,Kunming Qin,Qing Zhang

Choerospondias axillaris is a medicinal plant used for treating coronary heart disease (CHD) due to its broad spectrum of anti-inflammatory activities. Cyclooxygenase 2 (COX-2) and lipoxygenase 5 (5-LOX) were immobilized on magnetic nanoparticles for selective ligand-extraction of these two enzymes present in C. axillaris . Sixteen extracted components were identified and analyzed using ultraperformance liquid chromatography plus Q-Exactive Orbitrap tandem mass spectrometry (UHPLC-Q-ExactiveOrbitrap-MS/MS), including flavonoids Curcumin and Epicatechin. The metabolization of the aforementioned 16 components in rats with acute blood stasis indicated a solid pharmacodynamic foundation. This method offers an effective approach for conducting basic research on the enrichment, analysis, and efficacy evaluation of active components in complex natural products.

Optimization of Natural Deep Eutectic Solvents' Extraction Process From Atractylodes macrocephala–Astragali Radix Using the Dual Mathematical Model and Its Activity Evaluation

Wenlong Huang,Qingling Meng,Mingjiang Mao,Boxuan Wang,Chenhuan Shentu,Xiaohong Li,Jie Hao,Yujin Zhao,Jiaying Zhu,Shijie Dai,Xiaofeng Yuan

Natural deep eutectic solvents (NADESs) have garnered significant attention for their application in the extraction of natural products. This study proposes a strategy to enhance the extraction efficiency of target compounds from the Atractylodes macrocephala Koidz– Astragali Radix (AM-AR) combination, and the crude extracts were employed for subsequent in vitro and in vivo investigations. The extraction efficiencies of nine NADESs and conventional solvents were compared for isolating calycosin 7-O-glucoside, ononin, quercetin, and atractylenolide III using high-performance liquid chromatography (HPLC) analysis. The extraction parameters were then optimized through response surface methodology (RSM) and genetic algorithm–back propagation neural network (GA-BPNN) models. In addition, the antioxidant activities were evaluated in vitro using assays such as 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) free radical scavenging, and ferric reducing antioxidant power (FRAP). An in vivo rat model of middle cerebral artery occlusion (MCAO) was utilized to assess the biological activities. Among the tested solvents, NADES-7 exhibited the highest extraction efficiency, achieving a comprehensive evaluation value of 0.173 and was selected for subsequent experiments. These findings indicate that NADESs can serve as promising alternative solvents for extracting natural products from traditional Chinese medicine (TCM).

Exploring the Mechanisms of Polygonum orientale L. Against Myocardial Ischemia: An Integrated Analysis Using Ultra-High-Performance Liquid Chromatography–Quadrupole Exactive Orbitrap Mass Spectrometry, Network Pharmacology, and RNA Sequencing

Chunhua Liu,Changli Fu,Luping Tang,Jieqi Li,Jia Sun,Yuan Lu,Jie Pan,Ting Liu,Yongjun Li,Yonglin Wang,Yong Huang,Yueting Li,Meng Zhou

Polygonum orientale L. (PO) represents significant bioactivities in treating myocardial ischemia (MI); however, its underlying mechanisms remain unclear. This study aims to elucidate PO's potential mechanisms in MI using an integrated approach that combines UHPLC–Q-Exactive Orbitrap HRMS, network pharmacology, and RNA-sequencing (RNA-seq). Initially, the chemical constituents of PO were identified using UHPLC–Q-Exactive Orbitrap HRMS. Subsequently, network pharmacology, molecular docking, and RNA-seq were employed to screen potential active components of PO targeting MI and predict their molecular mechanisms. Then, the molecular mechanisms were verified using western blotting and ELISA in MI mice. A total of 45 components were identified from PO, with 14 potential active compounds interacting with 204 MI-related genes. The findings suggest that PO could alleviate heart damage. The RNA-seq results indicated 244 potential targets regulated by PO. Integrating RNA-seq and network pharmacology analyses revealed that the toll-like receptor signaling pathway plays an important role, alongside the PI3K-Akt. Notably, PO reduced the expression of TLR4 and TLR2 while increasing p-Akt and p-PI3K levels in MI mice, leading to decreased inflammatory cytokines and apoptosis-related proteins. This study provides initial evidence that PO inhibits the toll-like signaling pathway and activates PI3K–Akt signaling pathway to exert protective effects against MI.

Identifying of Anticoagulant Ingredients From Moutan Cortex Based on Spectrum-Effect Relationship Analysis Combined With GRA, PLS, and SVM Algorithms

Weijie Pan,Qianru Chen,Menghua Wu,Yue Sun,Ming Li,Shumei Wang,Xingyang Xue,Jiang Meng

Moutan Cortex (MC) is a renowned Chinese medicine used for promoting blood circulation and removing blood stasis. However, the active ingredients are unclear. This study aimed to identify and validate the active ingredients of MC. UPLC fingerprints of 23 batches of MC from various origins were analyzed. The activating blood efficacy of MC was assessed by evaluating the inhibitory effects on thrombin and factor Xa (FXa) using the chromogenic substrate method. Active ingredients were identified through spectrum-effect relationship analysis using gray relation analysis (GRA), partial least squares (PLS), and support vector machine (SVM) algorithms. Consequently, five components were identified as potential active ingredients: mudanpioside H, oxypaeoniflorin, 1,2,3,4,6-penta-O-galloyl-β-d-glucose (PGG), benzoyloxypaeoniflorin, and suffruticoside A/B/C/D. The pharmacological activities of these five active ingredients were further confirmed by measuring their thrombin inhibition ability and antithrombotic effects in zebrafish, and their interactions with thrombin and FXa were examined using molecular docking technology. Oxypaeoniflorin, benzoyloxypaeoniflorin, and PGG demonstrated significant efficacy in promoting blood circulation and resolving blood stasis, as well as strong binding affinities. This study provides a biochemical foundation for the anticoagulant effects of MC and offers valuable insights for quality control and the development of novel anticoagulant ingredients drugs.

A New Discovery: Corydalis yanhusuo Causes Idiosyncratic Hepatotoxicity and Its Potential Mechanisms

Longxin Guo,Li Lin,Jun Ling,Shengkai Zhu,Xinyu Li,Minjuan Long,Yingjie Xu,Zhanjiang Hu,Ming Niu,Xu Zhao,Xiaohe Xiao

Corydalis yanhusuo W.T.Wang (YHS) is a commonly used traditional Chinese medicine, often prescribed for treating a variety of pains. In recent years, there has been a gradual increase in the number of reports to liver injury caused by YHS and its preparations, but the exact type and mechanism of hepatotoxicity are still unclear. In the present study, we demonstrated that YHS could induce idiosyncratic drug-induced liver injury (IDILI) in the inflammatory activation models. A total of 459 differential genes and 25 differential metabolites were identified by transcriptomics and metabolomics, which were significantly enriched in the TNF and NF-κB signaling pathways as well as glycerophospholipid metabolism, sphingolipid metabolism, and arachidonic acid metabolism. In addition, YHS significantly increased the levels of TNF-α, IL-1β, and IL-6. Therefore, we believe that the mechanism of toxicity may be related to the TNF and NF-κB signaling pathways, with glycerophospholipid metabolism, sphingolipid metabolism, and arachidonic acid metabolism also playing important roles. It provides a reference for the safe and rational use of YHS in clinical practice and contributes to the precise prevention and control of the risk of liver toxicity associated with YHS.

A simple, robust and high-throughput LC–MS/MS method for the therapeutic drug monitoring of polymyxin B1, polymyxin B2, polymyxin B3, isoleucine-polymyxin B1, polymyxin E1 and polymyxin E2 in human plasma

Feng Chen,Huanhuan Li,Xiaoxia Yang,Ziwei Deng,Hongqiang Wang,Zhihua Shi,Chengfeng Qiu

To facilitate clinical therapeutic drug monitoring (TDM) of polymyxin B (PB) and polymyxin E (PE), we developed and validated a simple LC–MS/MS method for simultaneous determination of PB (including polymyxin B1 (PB1), polymyxin B2 (PB2), polymyxin B3 (PB3) and isoleucine-polymyxin B1 (ile-PB1)) and PE (including polymyxin E1 (PE1) and polymyxin E2 (PE2)) in human plasma. PB or PE was extracted from 20.0 μL plasma using a 5% (v/v) formic acid acetonitrile solution and separated on a BEH-C18 column (2.1 × 100 mm, 1.7 μm) with a mobile phase consisting of 0.8% formic acid aqueous solution and 0.2% formic acid acetonitrile solution. Gradient elution was performed over 5.5 min at a flow rate of 0.250 mL/min. Quantitative analysis was conducted in positive ion scanning mode by electrospray ionization and multiple reaction monitoring. The method validation was conducted based on bioanalytical method validation guidance, including specificity, calibration curve, precision, accuracy, recovery, matrix effect, stability and dilution integrity and all of the results satisfied the requirements. The method was simple, robust and high-throughput and is currently being used to provide a TDM service to enhancing therapeutic efficacy and safety use of the PB and PE.

Solid-phase extraction based on amino-functionalized graphene oxide nanocomposites for analysis of short-chain perfluorinated alkyl acids in human serum by ion chromatography mass spectrometry

Zhixiong Zhong,Gongke Li,Lingchuan Guo,Jing Yao,Zhe Liu,Jianchao Deng

A highly sensitive method was developed for the analysis of short-chain perfluorinated alkyl acids (PFAAs) in serum samples using solid-phase extraction (SPE) coupled with ion chromatography–electrospray ionization–mass spectrometry. The synthesized amino-functionalized graphene oxide nanocomposites were used as an SPE sorbent for the enrichment of trace analytes and purification of samples. They exhibited high selectivity to polar compounds. The suppressor was employed to remove counterions and reduce background signals of mobile phase. These two crucial steps could effectively eliminate matrix effects and enhance analytical sensitivity. The lowest limits of quantification were 2.0 μg L −1 for perfluorobutanoic acid and perfluorovaleric acid, 1.0 μg L −1 for perfluorocaproic acid and 0.50 μg L −1 for perfluorobutane sulfonic acid, respectively. The procedure was successfully applied for determination of trace PFAAs in 25 serum samples. Mean recoveries ranged from 86.3 to 101.4% with relative standard deviations of 1.6–6.8%. The method allowed an excellent separation and quantification of short-chain PFAAs that were difficult to analyze by conventional chromatography.
