A Dual-Functional BSA-MnO2 Quantum Dots-Based Ratiometric Sensor for the Detection of Acid Phosphatase

Wei Xu,Wei Yang,Yin Dai,Jianwen Fei,Chenxi Zhang,Yani Zheng,Wanying Zhu,Jun Zhang,Junli Hong,Xuemin Zhou

Ratiometric fluorescence sensors with quantum dots (QDs) as signal reporters have been widely used in biomarkers testing. However, there are few reports on the construction of a ratio fluorescence sensor based on the dual action of enzyme reaction and QDs. In this study, a “two-in-one” multifunctional catalytic QDs (BSA-MnO2 QDs) developed by hydrothermal method displayed excellent fluorescent properties and high oxidase-like activity. Its optimal quantum yield and Michaelis–Menten kinetics constants Km were 10.7% and 0.072 mM, respectively. In the presence of an enzyme substrate, the ratiometric signal can be generated indirectly by BSA-MnO2 QDs catalyzing, which was more conducive to the construction of a ratio fluorescence biosensor and made the experiment result more accurate. Based on these advantages, an acid phosphatase (ACP) ratiometric fluorescence biosensor was designed. In the absence of ACP, o-phenylenediamine (OPD) was catalyzed to produce 2,3-diaminophenothiazine (DAP) (the oxidized fluorescent product of OPD) that has an inner-filter effect with BSA-MnO2 QDs. Conversely, in the presence of ACP, l-ascorbic acid-2-phosphate was dephosphorylated to produce ascorbic acid, which inhibited the oxidase-like activity of BSA-MnO2 QDs and prevented the catalytic oxidation of OPD to DAP. The method can achieve the assay of ACP with a detection limit of 0.038 mU/mL. In addition, this effect can be achieved when other natural enzymes catalyze substrates to produce reducing substances. In this study, a universal ratio fluorescence probe (BSA-MnO2 QDs) was discovered and prepared for the first time, which has wide application prospects.

CuPc-Fe@BSA nanocomposite: Intracellular acid-sensitive aggregation for enhanced sonodynamic and chemo-therapy

Qingchen Bai,Miao Wang,Kai Wang,Jingwei Liu,Fengyu Qu,Huiming Lin

Due to their rigid π-conjugated macrocyclic structure, organic sonosensitizers face significant aggregation in physiological conditions, hindering the production of reactive oxygen species (ROS). An acid-sensitive nanoassembly was developed to address this issue and enhance sonodynamic therapy (SDT) and emission. Initially, copper phthalocyanine (CuPc) was activated using a H 2 SO 4 -assisted hydrothermal method to introduce multiple functional groups (–COOH, –OH, and −SO 3 H), disrupting strong π–π stacking and promoting ROS generation and emission. Subsequently, negatively charged CuPc-SO 4 was incorporated into bovine serum albumin (BSA) to form CuPc-Fe@BSA nanoparticles (10 nm) with Fe 3+ ions serving as linkers. In acidic conditions, protonation of CuPc-SO 4 and BSA weakened the interactions, leading to Fe 3+ release and nanostructure dissociation. Protonated CuPc-SO 4 tended to self-aggregate into nanorods. This acidity-sensitive aggregation is vital for achieving specific accumulation within the tumor microenvironment (TME), thereby enhancing retention and SDT efficacy. Prior to this, the nanocomposites demonstrated cycling stability under neutral conditions. Additionally, the released Fe ions exhibited mimicry of glutathione peroxidase and peroxidase activity for chemotherapy (CDT). The synergistic effect of SDT and CDT increased intracellular oxidative stress, causing mitochondrial injury and ferroptosis. Furthermore, the combined therapy induced immunogenic cell death (ICD), effectively activating anticancer immune responses and suppressing metastasis and recurrence.

A low-fouling electrochemical biosensor based on BSA hydrogel doped with carbon black for the detection of cortisol in human serum

Xiaohang Yu,Weichen Meng,Yang Li,Xiliang Luo

Electrochemical biosensors with high sensitivity can detect low concentrations of biomarkers, but their practical detection applications in complex biological environments such as human serum and sweat are severely limited by the biofouling. Herein, a conductive hydrogel based on bovine serum albumin (BSA) and conductive carbon black (CCB) was prepared for the construction of an antifouling biosensor. The BSA hydrogel (BSAG) was doped with CCB, and the prepared composite hydrogel exhibited good conductivity originated from the CCB and antifouling capability owing to the BSA hydrogel. An antifouling biosensor for the sensitive detection of cortisol was fabricated by drop-coating the conductive hydrogel onto a poly(3,4-ethylenedioxythiophene) (PEDOT) modified electrode and further immobilizing the cortisol aptamer. The constructed biosensor showed a linear range of 100 pg mL −1 - 10 μg mL −1 and a limit of detection of 26.0 pg mL −1 for the detection of cortisol, and it was capable of assaying cortisol accurately in complex human serum. This strategy of preparing antifouling and conductive hydrogels provides an effective way to develop robust electrochemical biosensors for biomarker detection in complex biological media.

Liposomal nano-encapsulation of bFGF combined with injectable BSA hydrogel for efficient burn wound healing

Yuhan Tu,Yangyang Ning,Hong Wang,Yang Li,Zhiang Yao,Shanhui Tao,Wenjing Yang,Bin Li,Xiaofen Li,Huacheng He,Shijun Li

Rapid and scar-free healing of burn wounds is an urgent clinical issue. Basic fibroblast growth factor (bFGF) has been proven to promote the healing of burn wounds by accelerating ECM remodeling and angiogenesis. However, exudates from burn wounds can accelerate bFGF degradation, thereby affecting its bioactivity. This study proposes an effective protection strategy for bFGF that involves encapsulating bFGF in nanoliposomes (bFGF-NLip) and then incorporating bFGF-NLip into a bovine serum albumin (BSA) hydrogel. This hybrid hydrogel system (bFGF-NLip@B) could maintain the activity of bFGF, achieve sustained release, and allow phospholipids and cholesterol to penetrate the skin, thereby enabling bFGF to function in the dermis. The experimental results showed that the hydrogel was injectable with good mechanical properties and biocompatibility. In a mouse scald wound model, owing to the sustained release of bFGF and skin permeation function of the nanoliposomes, the hydrogel promoted granulation formation, collagen deposition, vascular regeneration, and re-epithelialisation, ultimately accelerating wound healing. In addition, the hydrogel effectively inhibited scar formation. This system provides novel insights into the delivery of bFGF and scar-free healing of burn wounds.

BSA-Cu3(PO4)2 hybrid nanoflower—an efficient and low-cost nanoenzyme for decolorization of organic pollutants

Liu Cheng,Zheng Jingtian,Zhang Benxing,Zhong Xianhua,Wang Wei,Li Zhangyong

The Fenton reaction is one of the most effective methods for treating organic wastewater, which is extremely harmful to humans but difficult to treat. However, finding simple, low-cost, and efficient catalysts for the Fenton reaction remains a challenge. In this study, a BSA-Cu 3 (PO 4 ) 2 hybrid nanoflower (NF) was synthesized to investigate its peroxidase-like activity for the treatment of organic wastewater. Its morphology, composition, and crystallization had been fully studied and the results confirmed that the NFs were successfully prepared. Subsequently, the origin of the peroxidase-like activity of the NFs was further analyzed, with the results suggesting two reasons: (i) the transformation between Cu(I) and Cu(II) and (ii) nano-effects. Additionally, Congo red was selected as the organic pollutant to simulate the decolorization of wastewater. After 3 h, the decolorization efficiency reached 96%. Furthermore, the NFs exhibited good storage performance, maintaining approximately 90% relative activity after storage for 30 days. In summary, the NFs have great application prospects in the treatment of organic wastewater. Graphical abstract

BSA-templated synthesis of Ir/Gd bimetallic oxide nanotheranostics for MR/CT imaging-guided photothermal and photodynamic synergistic therapy

Chao Cai,Feng Tian,Junping Ma,Zipei Yu,Mo Yang,Changqing Yi

Precision medicine urges the development of theranostics which can efficiently integrate precise diagnosis and effective therapy. In this study, a facile synthesis of Ir/Gd bimetallic oxide nanotheranostics (termed BSA@Gd2O3/IrO2 NPs) with good biocompatibility was demonstrated using a biomineralization method where bovine serum albumin (BSA) served as a versatile template. BSA@Gd2O3/IrO2 NPs exhibited high longitudinal relaxivity (5.2 mM−1 s−1) and X-ray absorption capability (14.5 Hu mM−1), illustrating them to be a good contrast agent for magnetic resonance (MR) and computed tomography (CT) dual-modal imaging. Moreover, BSA@Gd2O3/IrO2 NPs can act as not only a photothermal conversion agent with ultrahigh efficiency (66.7%) as well as a good photosensitizer, but also an effective catalase to decompose endogenous H2O2 to produce O2, thus relieving hypoxia and enhancing the phototherapeutic effect. Both in vitro and in vivo experiments demonstrated the high effectiveness of BSA@Gd2O3/IrO2 NPs in MR/CT dual-modal imaging and photothermal and photodynamic synergistic tumor treatments. This work sheds new light on the development of versatile nanotheranostic systems using mild and robust biomineralization methods.

Smart and dual-targeted BSA nanomedicine with controllable release by high autolysosome levels

Zhanxia Zhang,Changsheng Dong,Guanzhen Yu,Wei Cheng,Yupei Liang,Yongfu Pan,Hegen Li,Hongbin Ji

Targeting modifications and smart responsiveness of nanomedicines can enable anticancer drugs to be selectively delivered to and controllably released in tumour cells or tissues, which can reduce the treatment’s toxicity and side effects. Good biocompatibility is crucial for the clinical application of any nanomedicine. In this study, a double-targeting molecule, an RGD peptide- and 4-(2-aminoethyl) morpholine-modified, doxorubicin (DOX)-loaded bovine serum albumin (BSA) nanomedicine, that can be controllably released by the high levels of autophagic lysosomes in tumour cells was developed. The size of the spherical BSA nanoparticles is approximately 60 nm. In vitro experiments indicated that the RGD peptide- and 4-(2-aminoethyl) morpholine-modified, DOX-loaded BSA nanomedicine has a better therapeutic effect than free DOX. In vivo experiments suggested that the BSA nanomedicine can successfully suppress the progression of PC9 xenograft tumours. This phenomenon may be attributable to the endocytosis of a relatively large amount of nanomedicine and the effective release of the loaded chemotherapeutic agent, as induced by high levels of autolysosomes. Collectively, the results of this study provide a smart approach for increasing therapeutic efficacy using a double-targeting molecule-modified BSA nanomedicine.

Menthol-modified BSA nanoparticles for glioma targeting therapy using an energy restriction strategy

Liang Jianming,Zhu Ying,Gao Caifang,Ling Chengli,Qin Jing,Wang Qi,Huang Yongzhuo,Lu Weigen,Wang Jianxin

Current chemotherapeutics for glioma are not sufficiently effective due to their low tolerance and poor blood–brain barrier (BBB) permeability. Energy restriction based on co-delivery of albendazole (Abz) and nano-silver has been proven to suppress tumor growth without adverse effects. Accordingly, menthol was conjugated to BSA (MeB) to overcome the BBB-permeability issue. In this study, Abz-loaded MeB-silver nanoparticles (MBS-Abz) were developed by self-assembly of MeB, albendazole and nano silver for glioma targeting therapy. The nanoparticle entered the brain across the BBB and specifically accumulated in the glioma region. MeB delivered the nanoparticles across the brain capillary endothelial cell monolayer by promoting internalization and BBB disruption. MBS-Abz hindered ATP generation via the inhibition of glycolytic and mitochondrial pathways both in vitro and in vivo. The anti-glioma mechanisms of energy restriction were related to the cytotoxicity, proliferation inhibition, cell cycle arrest, and apoptosis induced by ATP exhaustion, and the effects of MBS-Abz were significantly better than those induced by monotherapy nanoparticles or unmodified nanoparticles. These results demonstrated that by combining the energy restriction effect of albendazole and nano silver, as well as the BBB penetration ability of menthol, MBS-Abz achieves superior anti-glioma efficacy and can be an effective strategy for glioma therapy.

Regulation on the aggregation-induced emission (AIE) of DNA-templated silver nanoclusters by BSA and its hydrolysates

Wei-Xian Wang,Yuqing Wu,Hong-Wei Li

The aggregation-induced emission (AIE) phenomenon of the metal nanoclusters (NCs) has been discovered quite recently, which is considered to be creative to improve the optical properties of NCs upon ligand aggregation. In the present study, we report an AIE system for double-stranded DNA-templated silver nanoclusters (AgNCs-dsDNA) by using bovine serum albumin (BSA) in solution, which induce a moderate emission enhancement of AgNCs (5-fold) and a blue-shift through a loosen interaction. In addition, significant luminescence enhancement (30-fold) is further stimulated by the addition of digestive enzyme to the system by altering the surface structure of the aggregated particles. The processes are observed directly through transmission electron microscopy (TEM), where the dispersed AgNCs spheres are aggregated in a large incompact sheet-like film by BSA; while further trypsin addition lead them into larger particles (>50   nm) with higher density. And the intrinsic mechanism is explained well by UV–vis absorption and time-resolved luminescence spectra. The observed luminescence enhancement in step-II is attributed to the AIE through stronger interaction between the hydrolysates of BSA and the ligand dsDNA. Therefore, the optical properties of AgNCs-dsDNA are regulated well by the addition of BSA and trypsin in different amounts, which relates directly to the degree of aggregation and can be extended to other metal nanoclusters by employing the pairs of protein and related enzyme.

The protective effect of biomineralized BSA-Mn3O4 nanoparticles on HUVECs investigated by atomic force microscopy

Xinyue Guo,Weiqi Zhou,Yanxue Guan,Juan Qin,Bailin Zhang,Miaomiao Zhang,Jilin Tang

Manganese-based nanozymes have been widely used in the field of cell protection due to their various enzyme-mimicking activities, but their effect on the mechanical properties of cells is not yet known. Here, bovine serum albumin-modified Mn3O4 nanoparticles (BSA-Mn3O4 NPs) with good biocompatibility were synthesized by a one-step biomineralization method using BSA as a template. BSA-Mn3O4 NPs possess scavenging activity against superoxide free radicals (O2˙−), hydroxyl radicals (˙OH) and hydrogen peroxide (H2O2). The excellent reactive oxygen species (ROS) scavenging activity of BSA-Mn3O4 NPs enables them to effectively reduce the intracellular ROS level, thus mitigating the damage of oxidative stress on human umbilical vein endothelial cells (HUVECs). Subsequently, the intracellular antioxidant mechanism of the BSA-Mn3O4 NPs was further investigated. The results show that the BSA-Mn3O4 NPs could inhibit the depolymerization of F-actin, help cells maintain their normal morphology, and reduce the decrease in Young's modulus of cells caused by oxidative stress.

A combination of polypeptide and magnetic molecular imprinted electrochemical sensor based on BSA-AuNPs/Fe3O4@ZIF-8 for the detection of norfloxacin

Yuanqian Xiong,Xiaoyu Gao,Yu Wang,Xiangrui Deng,Chunhong Xiong,Jinsheng Zhang,Qianhui Gu,Ganhui Huang

Norfloxacin (NOR) is now extensively used for the treatment of human and animal diseases. Unfortunately, large accumulations of NOR will pose a significant threat to the human body. Thus, it is urgent to develop a rapid and sensitive detection technique for the trace determination of NOR. In this study, an electrochemical molecular imprinting sensor with great properties was fabricated for the determination of NOR in milk and environmental water. The incorporation of bovine serum proteins produced a hybrid recognition interface consisting of molecularly imprinted polymers and peptides that provided good recognition properties for norfloxacin. The Fe 3 O 4 @ZIF-8 was distributed orderly on the electrode after the embedding of the magnets inside the electrode, then the BSA was coated on the electrode with the electrodeposited AuNPs through Au-S bonding. The sensor constructed in this study achieved ultra-sensitive detection of norfloxacin with an effective area of 0.08433 cm 2 , a limit of detection (LOD) of 0.108 pmol·L −1 , and a wide linear range (1 ∼ 100, 100 ∼ 1000 and 1000 ∼ 8000 pmol·L −1 ). The prepared sensors exhibited satisfactory performance in the detection of milk and environmental water, with acceptable recoveries ranging from 96.24 % to 104.88 %, and the relative standard deviation (RSD) was no more than 4.84 %.

Near-infrared MnCuInS/ZnS@BSA and urchin-like Au nanoparticle as a novel donor-acceptor pair for enhanced FRET biosensing

Hong Xing,Tianxiang Wei,Xin Lin,Zhihui Dai

An enhanced near-infrared (NIR) FRET biosensing system based on the novel donor-acceptor pair (MnCuInS/ [email protected] and urchin-like Au NPs) was developed for the sensitive detection of HER2 protein. In this strategy, MnCuInS/ZnS quantum dots (QDs) were chosen due to their NIR emission, high quantum efficiency , and low toxicity. Amounts of MnCuInS/ZnS QDs were encapsulated in BSA to form MnCuInS/ [email protected] nanoparticles, which can easily transfer MnCuInS/ZnS QDs from organic phase into aqueous solution, and the formed nanoparticles were applied as the energy donor which significantly enhanced the fluorescence intensity . As for the energy acceptor, urchin-like Au nanoparticles (Au NPs) with novel structure, good stability and excellent quenching ability towards NIR fluorescence were selected. Thus, the NIR MnCuInS/ [email protected] and urchin-like Au NPs were designed as a novel donor-acceptor pair for FRET assay. By combining the optical advantages of NIR QDs encapsulated BSA nanoparticles with the excellent fluorescence quenching ability of urchin-like Au NPs, the proposed FRET-based biosensor realized enhanced FRET effect for highly sensitive detection of HER2 in human serum samples. A wide detection range (2–100 ng mL −1 ) and a low detection limit (1 ng mL −1 ) were obtained. This sensing system can decrease the interference of other biomolecules in NIR region, can be applied to other biomarker determination in vitro, and also shows great potential for in vivo diagnosis.
