In the past decade, biomimetic calcium phosphate (CaP) ceramics have been considered as practicable grafts and biomaterial substitutes in repairing jaw bone defect after tumor resection or traffic accident. Nowadays, increasing incidence of biomedical material-associated infection has raised a concern when applying these materials. In this work, a new porous CaP scaffold with antibacterial coating was proposed. This biomimetic scaffold was composited with berberine (BBR), Ag nanoparticles (nAg), and silk fibroin (SF). The microstructures and phase composition of the scaffolds were analyzed. The cytocompatibility and osteogenic potential of the prepared samples were evaluated in vitro . The scaffolds held hierarchical structure: the first-level porous CaP ceramic with micron pores ranged from 250 to 600 µm; the second-level spongy-like structure with abundant capillary pores ranged from 500 nm to 10 µm; and the third-level structure was achieved by filling BBR, nAg, and SF gel coatings into the above porous structures. The experimental results showed that the antimicrobial capability of single BBR coating is inconspicuous. However, the introduction of nAg could significantly promote the antibacterial effect of scaffolds. At the same time, such scaffolds showed improved osteoinductivity. This new biomimetic CaP scaffold with antibacterial and osteoinductive properties may be a promising candidate for bone tissue engineering.

Simple and rapid high-performance liquid chromatography methods were developed for the determination of berberine (BB) in various rat tissues so as to evaluate a P-gp inhibitor, glycyrrhetinic acid (GA), on BB’s oral bioavailability. Acetonitrile was used to extract BB from tissues and showed different extraction recoveries in diverse tissues. The intra- and interday precision and accuracy were less than 10%. Long-term stability, pre (post) -preparative stability, and freeze-thaw stability were evaluated, and the results showed it could meet the need of this study. The proposed methods were subsequently applied to investigate the possible drug-drug interaction of GA and BB in vivo from the aspect of tissue distribution. The results showed that no significant difference was found between the group of low dose and high dose at the same time point. The tissue distributions show a saturated model, i.e., the content of BB in tissue tends to be constant while its dose is more than 200 mg/kg. Besides, the contents of BB ranged from high to low according to the order of the liver, kidney, and spleen. The BB content in the liver is especially high as compared to other tissues.

Berberine (BBR), a kind of quaternary ammonium benzylisoquinoline alkaloids with multiple pharmacological activities, has been regarded as a promising lipid-lowering agent in the field of drug repurposing. Particularly, the chemical modification at the C-9 position of BBR can remarkably improve its lipid-lowering efficacy. In this study, thirteen novel BBR derivatives were rationally designed, synthesized, and evaluated by preliminary pharmacological tests. The results showed that most compounds exhibited more potent hypolipidemic activities when compared with BBR and simvastatin. Among these compounds, compound 2h-1 and 2h-2 exhibited better activity profiling in these four tests involving with inhibition of total cholesterol (TCHO), triglyceride (TG), and low-density lipoprotein cholesterol (LDLC) and the increase of high-density lipoprotein cholesterol (HDLC). Correspondingly, the BBR analogs with 9-O-cinnamic moiety probably exhibited potent lipid-lowering activity, and should be exploited as an important versatile template for the development of BBR-like lipid-lowering agents.

The purpose of this study was to investigate the therapeutic effect of berberine (BBR) on MNNG-induced chronic atrophic gastritis (CAG) and the possible mechanism of BBR through TGF-β1/PI3K signal pathway. GES-1 were pretreated with MNNG for 2 h before BBR treatment in all procedures. Cell viability was quantified by cell counting kit-8, and GES-1 morphology and proliferation were detected by high content screening (HCS) assay. The rat model of CAG was established by MNNG, and the therapeutic effect of BBR on stomach histopathology and serum supernatant were analyzed in vivo. In addition, the possible mechanism of BBR was further discussed, and the expression of related genes and proteins in TGF-β1/PI3K signal pathway was detected. The results showed that BBR could significantly improve the survival rate and morphological changes of GES-1, improve the gastric tissue injury of CAG rats, and reduce the expression of G-17 and inflammatory factors IL-8, TNF-α, IL-6 and IL-1β. In addition, BBR down-regulated the expression of TGF-β1 axis-related signals such as TGF-β1, PI3K, p-Akt/Akt, p-mTOR/mTOR and P70S6K, and promoted the expression of PTEN, LC3-II and Beclin-1. In Conclusion, BBR can improve CAG which may be closely related to TGF-β1/PI3K signal pathway.

Type 2 diabetes (T2D) often leads to metabolic disorders. Hyperchloraemia, a common electrolyte metabolic disorder and caused by excessive intake of chloride-containing acidic agents. Thus, while berberine hydrochloride (BH) elicits a good hypoglycaemic effect, its nearly 10% chloride ion component is a potential hazard for T2D patients. In this study, we prepared four berberine organic acid salts (BOAs), including berberine citrate, berberine malate, berberine succinate, and berberine fumarate. T2D rats were administered BH or BOAs (500 mg/kg d) for 6 weeks. The body weights and levels of total cholesterol, triglycerides, fasting blood glucose, fasting insulin, and insulin resistance index in administered rats were normal. Levels of Na + , K + , Cl − , HCO 3 − , pH, pCO 2 and ‘ base excess (BE) in BH group showed significant differences compared with normal rats. These results suggest that BOAs are comparable to BH in terms of hypoglycaemic effects, but superior with regard to safety from hyperchloraemia in T2D rats.

Berberine (Ber), an isoquinoline alkaloid, is a potential drug therapy for ulcerative colitis (UC) because of its anti-inflammatory activity, high biological safety, and few side effects. Nevertheless, its clinical application is hindered by its limited water solubility and low bioavailability. Currently, compared to synthetic nanocarriers, exosomes as carriers possess advantages such as low toxicity, high stability, and high specificity. Human placental mesenchymal stem cell-derived exosomes (HplMSC-Exos) have emerged as a promising drug delivery system, offering intrinsic anti-inflammatory and antioxidant activities. Therefore, we engineered MSC-Exos loaded with Ber (Exos-Ber) to enhance the solubility and bioavailability of Ber and for colon targeting, revealing a novel approach for treating UC with natural compounds. Structurally and functionally, Exos-Ber closely resembled unmodified Exos. Both in vitro and in vivo investigations confirmed the antioxidant and anti-inflammatory properties of Exos-Ber. Notably, Exos-Ber exhibited reparative effects on injured epithelial cells and reduced cellular apoptosis. Furthermore, Exos-Ber concurrently demonstrated anti-inflammatory and antioxidant activities, contributing to the mitigation of UC, possibly through its modulation of the MAPK signaling pathway. Overall, our findings demonstrate the potential of Exos-Ber as a promising therapeutic option for alleviating UC, highlighting its capacity to enhance the clinical applicability of Ber.

Berberine is separated from medical plants and food raw materials, and shows excellent anti-diabetic activity and could be used as a food additive in some countries and regions. However, its inhibitory mechanisms on α-amylase and α-glucosidase have hardly ever been revealed in vitro. In this study, enzymatic reaction kinetics, fluorescence quenching, circular dichroism spectroscopy, and molecular docking are used to study the inhibitory effects of berberine on α-amylase and α-glucosidase. Berberine has great inhibitory effects on α-amylase and α-glucosidase (IC 50 = 50.83 µg mL –1 , IC 50 = 198.4 µg mL –1 , respectively), and inhibition types of α-amylase and α-glucosidase are non-competitive inhibition and competitive inhibition, respectively. Fluorescence spectra show that there is static quenching between berberine and α-amylase or α-glucosidase. Besides, hydrophobic interaction is the main interaction type. These results are further evidenced and visualized by molecular docking. This study provides a different theoretical basis for berberine in anti-diabetic activity in vitro and expands interaction research about hydrophobic compounds and α-amylase as well as α-glucosidase.

Berberine (BBR) has seen growing application in ophthalmology, yet the ocular toxicity of BBR and its metabolites remains poorly understood. This study aimed to evaluate the ocular toxicity of BBR and its major metabolite M1 and unravel their underlying mechanisms. Ocular toxicity was evaluated in human corneal epithelial cells and wild-type AB zebrafish. Mechanistic studies utilized fluorescence imaging, biochemical quantitative assays, and qPCR analyses in AB zebrafish and transgenic mitochondrial fluorescent zebrafish (strainTg(Xla.Eef1a1:mlsEGFP)). Both BBR and M1 induced significant ocular toxicity across models, with BBR showing higher toxicity than M1. Mechanistic analyses revealed their toxicity stemmed from photoreceptor cell damage and Sirtuin 3 (SIRT3) inhibition, triggering a cascade of pathological events: mitochondrial dysfunction, oxidative stress, autophagic dysfunction, apoptosis, and inflammation. This study provides a reference for individualized risk assessment and clinical management of BBR-based therapies and paves the way for developing BBR derivatives with reduced ocular toxicity.

In order to solve the problem of bacterial resistance, increase the antibacterial performance and delay the aging of paper, the berberine modified trimethoprim carboxymethyl cellulose was prepared using trimethoprimethylate carboxymethyl cellulose as raw material and trimethoprimethylate lactate (TMP) as antibacterial modifier by Schiff Base reaction. At the same time, berberine (BBR) was used as graft monomer to prepare a multifunctional protective solution integrating strengthening, antibacterial and good biocompatibility. The optimal process conditions were determined by single factor experiment and response surface method. The structure and properties of the product were characterized by FT-IR, XRD, TG and SEM and the antibacterial properties were evaluated by AGAR disk diffusion method. The results showed that when the mass ratio of dialdehyde carboxymethyl cellulose and modified berberine was 7:15, reaction temperature was 40 °C, and pH was 6.0, the optimum synthetic conditions was obtained. And the minimum inhibitory concentration of the product against Staphylococcus aureus and Escherichia coli were 0.2 mg/mL, minimum diameter of antibacterial zone of 6.7 mm and 7.1 mm, respectively, with good antibacterial effect. This paper lays a foundation for further research on the safe and effective antiquities antibacterial agent, and provides a reference for its further development in paper protection.

Novel ethyl cellulose/chitosan microspheres (ECCMs) were prepared by the method of w/o/w emulsion and solvent evaporation. The microspheres were spherical, adhesive, and aggregated loosely with a size not bigger than 5 μm. The drug loading efficiency of berberine hydrochloride (BH) loaded in microspheres were affected by chitosan (CS) concentration, EC concentration and the volume ratio of V (CS)/ V (EC). ECCMs prepared had sustained release efficiency on BH which was changed with different preparation parameters. In addition, the pH value of release media had obvious effect on the release character of ECCMs. The release rate of BH from sample B was only a little more than 30% in diluted hydrochloric acid (dHCl) and that was almost 90% in PBS during 24 h. Furthermore, the drug release data were fitted to different kinetic models to analyze the release kinetics and the mechanism from the microspheres. The released results of BH indicated that ECCMs exhibited non-Fickian diffusion mechanism in dHCl and diffusion-controlled drug release based on Fickian diffusion in PBS. So the ECCMs might be an ideal sustained release system especially in dHCl and the drug release was governed by both diffusion of the drug and dissolution of the polymeric network.

Berberine (BBR), a quaternary isoquinoline alkaloid found in a Chinese herb medicine from Berberis species plants, was utilized to synthesize three new biocompatible berberine-based pharmaceutical salts, featuring three mid-chain fatty acid anions derived from animal and vegetable oils. Three salts [BBR][FA] were namely berberine caproate [BBR][CAP], berberine heptylate [BBR][HEP] and berberine octoate [BBR][OCT]. These compounds were characterized using 1 H and 13 C NMR, FTIR, ESI-MS, and thermogravimetric analyses. The solubility and octanol/water partition coefficient were evaluated in water and simulated body fluids (simulated gastric, simulated intestinal and simulated body fluids). The water solubility of [BBR][FA] was higher than that of salt precursor berberine chloride [BBR][Cl]. The minimum inhibitory concentration and minimum bactericidal or fungicidal concentration against three strains ( Staphylococcus aureus , Bacillus subtilis and Candida albicans ) were also examined. The results showed that they have similar antimicrobial activity to [BBR][Cl]. Furthermore, transport and volumetric properties (density and conductivity) were determined to investigate their aggregation and solvation behaviors in aqueous solution. The limiting molar conductivity, Ʌ 0 , critical micelle concentration, cmc , apparent molar volume at infinite dilution, V 2 , φ 0 , the apparent molar expansibility at infinite dilution, E φ 0 , etc for [BBR][FA] were calculated. Effect of the length in alkyl chain of fatty acid anions in [BBR][FA] and temperature on the obtained properties as well as the nature of the interactions in aqueous [BBR][FA] solutions have been discussed.

Plastic pollution and food spoilage have drawn public attention, which may cause a great threat to the global environment and human health. Therefore, natural antimicrobial packaging has a pivotal share in solving these problems. In this study, biodegradable and antibacterial packaging films were successfully prepared via incorporating self-assembled berberine-cinnamic acid nanoparticles (BC NPs) into the film matrixes. With the addition of BC NPs, the UV-shielding property, hydrophobicity, and thermal stability of packaging were apparently improved. Meanwhile, the films also held considerable mechanical property and good biocompatibility, where the migrations of films in two food simulants were all far less than the permitted limits. Furthermore, nanocomposite films (NFs) exhibited strong antibacterial ability against both E. coli and S. aureus . By measuring the fluorescence spectrum and ROS production of films, we certificated NFs were AIE-active, and the antimicrobial activity of NFs was significantly enhanced under white light. Finally, the packaging test further indicated NFs could inhibit microbial growth of chicken samples well. The sustainable NFs will be ideal candidate materials to replace plastics and extend the shelf life of food products.
