The gelatin/gellan gum based-bilayer emulsion film was developed in this work to improve the survivability of Bifidobacterium longum during the storage process. The baobab seed oil (BO) was added to the gelatin (GE) matrix to develop emulsion film as the barrier outer layer. The blueberry anthocyanin extract (BE) was incorporated into the gellan gum (GG)-based inner layer to enhance the viability of B. longum . The SEM and FTIR results revealed that the probiotics were successfully entrapped in BO/BE-loaded bilayer film. The greatest survivability and viable cell numbers of the B. longum during the storage period were observed in the BO/BE loaded bilayer film. Furthermore, the stability of the colorful patterns by electrochemical writing was also evaluated in this work. Finally, the GE/BO-GG/BE/BM maintain satisfactory probiotic viability in steamed bread coating application. Hence, the GE/BO-GG/BE/BM bilayer film could be considered a novel material to deliver and protect the probiotics in food applications.

Background The interplay between gut microbiota and tumor microenvironment (TME) in the pathogenesis of colorectal cancer (CRC) is not well explored. Here, we elucidated the functional role of Bifidobacterium adolescentis ( B.a ) on CRC and investigated its possible mechanism on the manipulation of cancer-associated fibroblasts (CAFs) in CRC. Methods Different CRC animal models and various cell line models were established to explore the function of B.a on CRC. The single-cell RNA sequencing (scRNA-seq) or flow cytometry was used to detect the cell subsets in the TME of CRC. Western blot, quantitative real-time polymerase chain reaction (qRT-PCR), or immunofluorescence staining were performed to examine the activation of Wnt signaling and growth arrest specific 1 (GAS1) on CD143 + CAFs. Chromatin immunoprecipitation quantitative real-time PCR (CHIP-qPCR) was performed to investigate the regulation of transcription factor 4 (TCF4) on GAS1. Multi-immunofluorescence assay examined the expression level of CD143 and GAS1 on tissue microarray. Results We found that B.a abundance was significantly reduced in CRC patients from two independent cohorts and the bacteria database of GMrepo. Supplementation with B.a suppressed Apc Min/+ spontaneous or AOM/DSS-induced tumorigenesis in mice. scRNA-seq revealed that B.a facilitated a subset of CD143 + CAFs by inhibiting the infiltration of Th2 cells, while promoting the TNF-alpha + B cells in TME. CD143 + CAFs highly expressed GAS1 and exhibited tumor suppressive effect. Mechanistically, GAS1 was activated by the Wnt/β-catenin signaling in CD143 + CAFs. B.a abundance was correlated with the expression level of CD143 and GAS1. The level of CD143 + CAFs predicted the better survival outcome in CRC patients. Conclusions These results highlighted that B.a induced a new subset of CD143 + CAFs by Wnt signaling-regulated GAS1 to suppress tumorigenesis and provided a novel therapeutic target for probiotic-based modulation of TME in CRC.

Melanin produced by melanocytes protects our skin against ultraviolet (UV) radiation-induced cell damage and oxidative stress. Melanin overproduction by hyperactivated melanocytes is the direct cause of skin hyperpigmentary disorders, such as freckles and melasma. Exploring natural whitening agents without the concern of toxicity has been highly desired. In this study, we focused on aBifidobacterium longumstrain, ZJ1, isolated from a Chinese centenarian, and we evaluated the anti-melanogenic activity of the distinctive extracts of ZJ1. Our results demonstrated that whole lysate (WL) and bacterial lysate (BL) of ZJ1 ferments efficiently reduce α-melanocyte-stimulating hormone (α-MSH)-induced melanin production in B16-F10 cells as well as the melanin content in zebrafish embryos. BL and WL downregulate melanogenesis-related gene expression and indirectly inhibit intracellular tyrosinase activity. Furthermore, they both showed antioxidant activity in a menadione-induced zebrafish embryo model. Our results suggest that ZJ1 fermentation lysates have application potential as therapeutic reagents for hyperpigmentary disorders and whitening agents for cosmetics. Keywords:pigmentation;Bifidobacterium longum;anti-melanogenic;tyrosinase;zebrafish;antioxidant

Nanoplastics (NPs) exposure can cause severe hepatic injuries. Gut microbiota is considered a contributing factor to multiple hepatic injuries. However, its role in NPs‐induced hepatic injuries remains unclear, and microbial intervention strategies are required. Our results reveal that oral exposure to polystyrene NPs reduces gut probiotic Bifidobacterium pseudolongum ( B.p ) and its metabolite inosine. Gut microbiota from NPs‐administered mice partially reproduces NPs‐related impairment of gut homeostasis and hepatic injury in recipient mice. Moreover, B.p colonization improves NPs‐induced gut homeostasis impairment and hepatic injury, and its protective effects are reproduced by supplementation with inosine. Mechanically, B.p colonization increases hepatic level of inosine and subsequently normalizes the expression of its target A2AR. Meanwhile, increased inosine inhibits the miR155/SOCS1/NF‐κB pathway and represses NPs‐induced M1 macrophage polarization. CGS21680 , an agonist of A2AR, effectively represses lipopolysaccharide (LPS)‐induced M1 macrophage polarization and inhibits the miR155/SOCS1/NF‐κB pathway in vitro. Further, miR155 knockout inhibits NPs‐induced M1 macrophage polarization, but does not influence the suppression of NPs on A2AR. These findings suggest that B.p ‐derived inosine can repress NPs‐induced M1 macrophages polarization by inhibiting the miR155/SOCS1/NF‐κB pathway via targeting A2AR. Altogether, this study further clarifies the role of gut microbiota in NPs‐induced hepatic injury and provides a potential microbial therapeutic strategy. Probiotic B.p colonization elevated gut‐derived inosine level in the liver, while elevated inosine activated A2AR and subsequently blocked NPs‐induced polarization of M1 macrophages by repressing the miR155/SOCS1/NF‐κB pathway. This reduced the release of inflammatory cytokines and thereby, mitigated NPs‐induced hepatic injury.

Inflammatory bowel diseases (IBDs) are frequently accompanied by anxiety and depression, largely driven by perturbed gut-brain axis signaling. However, current oral therapies remain constrained by the spatial and functional separation between intestinal inflammation and central nervous system dysfunction. Here, we present a comprehensive gut-brain dual region integrated therapeutic strategy based on functionalized Bifidobacterium longum hydrogel (INPs@BL@Gel), in which baicalin and tyrosine are coordinated with Fe(III) to form infinite coordination polymers (ICPs), coated with inulin, assembled onto Bifidobacterium longum (BL), and subsequently encapsulated within a pH- and matrix metalloproteinase-responsive silk fibroin-gelatin hydrogel. INPs@BL@Gel exhibits high drug-loading, effective gastric protection, inflammation-triggered release, and long-term intestinal colonization. Within the inflamed intestine, BL and components synergistically suppress inflammatory responses, restore gut microbiota homeostasis, and promote intestinal barrier repair through multi-target integrated therapy. Importantly, BL combined with components markedly enhances the production of beneficial neuroactive metabolites such as homovanillic acid and short-chain fatty acids, which integratedly regulate neuroinflammation, preserve synaptic function, and facilitate blood-brain barrier repair via the gut-brain axis. In vivo studies demonstrate that INPs@BL@Gel not only exerts potent therapeutic efficacy against colitis and effectively alleviates associated depression, but also reshapes the gut microbiota and restores barrier integrity, achieving a remarkable comprehensive therapeutic effect.

Bifidobacterium longumsubsp.longumXZ01 (BLSL1) is a new strain (isolated from the intestines of healthy people and deposited with the preservation number GDMCC 61618). An exopolysaccharide, S-EPS-1, was successfully isolated from the strain and then systematically investigated for the first time. Some structural features of S-EPS-1 were analyzed by chemical component, HPLC, ultraviolet, infrared, and nuclear magnetic resonance spectrum analyses. These analyses revealed that S-EPS-1 is a neutral heteropolysaccharide with an α-configuration. It contains mainly mannose and glucose, as well as small amounts of rhamnose and galactose. The molecular weight of S-EPS-1 was calculated to be 638 kDa. Several immunoregulatory activity assays indicated that S-EPS-1 could increase proliferation, phagocytosis, and NO production in vitro. In addition, S-EPS-1 could upregulate the expression of cytokines at the mRNA level through TLR4-mediated activation of the NF-κB signaling pathway in RAW 264.7 cells. Finally, S-EPS-1 was demonstrated to exhibit antioxidant activity by ABTS+•scavenging, DPPH•scavenging, and ferric-ion reducing power assays. Furthermore, S-EPS-1 can protect cells from oxidative stress and shows no cytotoxicity. These beneficial effects can be partly attributed to its antioxidant ability. Thus, the antioxidant S-EPS-1 may be applied as a functional food in the future. Keywords:Bifidobacterium longumsubsp.longumXZ01;exopolysaccharide;isolation;structural analysis;antioxidant activity;immunoregulatory activity

Anti-aging bioproducts from natural active ingredients represent a core future direction in food research. Cardiovascular aging drives most age-related disorders. This study investigated the anti-aging effects of a postbiotic preparations derived from brown algae extract fermented by Bifidobacterium adolescentis Life Age 20 (LA20FM). Multi-model were used, including doxorubicin (DOX)-induced senescent canine (CVECs) and feline vascular endothelial cells (FVECs), senescence-accelerated mouse prone 8 (SAMP8) mice, Caenorhabditis elegans , and naturally aged dogs and cats. In vitro, LA20FM markedly attenuated DOX-induced endothelial senescence, reduced the secretion of senescence-associated secretory phenotype factors, and restored endothelial function-related markers. In aged SAMP8 mice, LA20FM alleviated vascular remodeling, reduced circulating senescence- and inflammation-related biomarkers, and improved physiological performance. In C. elegans , LA20FM extended lifespan by approximately 30%, enhanced reproductive capacity, and significantly reduced intracellular reactive oxygen species levels. Comparable improvements in physiological parameters were also observed in aged dogs and cats. Transcriptomic analyses of CVECs and SAMP8 vascular tissues revealed that LA20FM treatment was accompanied by downregulation of NF-κB-related inflammatory signaling. Phorbol 12-myristate 13-acetate rescue experiments and early time-point analyses further supported NF-κB signaling as an early and functionally important mediator of its protective effects. Metabolomic analysis further revealed metabolic remodeling characterized by reduced lipid-associated metabolic activity and enhanced amino acid biosynthesis and antioxidant-related metabolism. Collectively, these findings indicate that LA20FM alleviates canine and feline vascular endothelial senescence and improves aging-associated phenotypes across multiple experimental models, supporting its potential as a postbiotic preparation for canine and feline nutritional intervention against vascular functional decline and further aging.

Chronic inflammation is a factor in the development of cancer, and probiotics play a role in preventing or treating inflammation as an adjuvant therapy. To investigate potential probiotics for the prevention of colitis-associated colorectal cancer (CAC), Bifidobacterium bifidum H3-R2 and Lactococcus lactis KLDS4.0325 were used to examine the effects on colon cancer cells and in an inflammation-related cancer animal model. The results revealed that B. bifidum H3-R2 in combination with L. lactis KLDS4.0325 caused apoptosis in colon cancer cells by increasing caspase-3 and caspase-9 protein levels, enhancing Bax expression, and lowering Bcl-2 expression. In addition, the combination of the two strains relieved the tissue damage; reduced proinflammatory cytokines, myeloperoxidase (MPO) activity, and hypoxia-inducible factor 1-alpha (HIF-1α) level; upregulated anti-inflammatory cytokines; increased colonic tight junction protein expression; regulated intestinal homeostasis by inhibiting NLRP3 inflammasome signaling pathway; and improved the imbalance of gut microbiota in animal models. Moreover, the combination of the two strains had a greater preventive impact than each strain alone. These findings are supportive of clinical studies and product development of multi-strain probiotic preparations for diseases associated with colitis.

Incorporating Bifidobacterium into fermented milk alters the balance between Lactobacillus bulgaricus and Streptococcus thermophilus . We investigated the bacterial interaction and metabolism post‐fermentation and during 21‐day storage. Utilizing non‐targeted metabolomics and electronic nose technology, we assessed impacts on product quality and flavor. Bifidobacterium significantly increased the viability of the other two species, with AI‐2 levels rising in the mixed culture. Metabonomic analysis revealed distinct metabolic profiles, with Bifidobacterium ‐fermented milk showing enriched key metabolites. Volatile compounds like ketones, aldehydes, esters, alcohols, and acids were identified, with 2‐heptanone and 2‐pentanone as initial discriminators and 2‐pentanone and acetaldehyde as key flavor compounds after storage. This study advances understanding of symbiotic interactions and metabolite profiles in fermented dairy ecosystems. The level of autoinducer‐2 (AI‐2) is maximized in dairy products fermented with Bifidobacterium species. Bifidobacterium significantly promoted the survival rate of Lactobacillus bulgaricus and Streptococcus thermophilus . The contents of caffeic acid and D‐galactose increased significantly after co‐culture. 2‐pentanone is the key flavor substance of three groups of fermented milk in two periods.

This study aimed to investigated the structural and physicochemical properties of two pectic polysaccharide fractions (RTFP1-2 & RTFP2) isolated from Rosa roxburghii Tratt fruit by anion-exchange and size-exclusion chromatography purification, as well as their protection on Bifidobacterium spp. against antibiotic damage. RTFP1-2 had a relatively lower molecular weight of 1.12 × 10 5 Da and RTFP2 had a higher molecular weight of 5.06 × 10 5 Da. Chemical composition analysis revealed that both RTFP1-2 and RTFP2 consisted of galacturonic acid (53.13 %, 64.73 %), galactose (27.26 %, 20.04 %), arabinose (3.57 %, 2.94 %), rhamnose (2.32 %, 1.67 %) and glucose (13.73 %, 10.62 %), respectively. These two pectic polysaccharides mainly contained homogalacturonan and rhamnogalacturonan I domain. RTFP1-2 and RTFP2 were resistant to in vitro gastrointestinal digestion with no more than 18 % (w/w) hydrolysis. Minimum inhibitory and bactericidal concentration (MIC and MBC) determination showed RTFPs could notably promote bifidobacterial survivals against three commonly used antibiotics with MIC increasing to ≥ 64 μg/mL and MBC to ≥ 512 μg/mL. The protection attributed to the viscous layers formed by the RTFPs surrounding the bacterial cells to prevent antibiotics from accessing to the bacteria. The viscous RTFP1-2 and RTFP2 also promoted the cohesion among bacterial cells and enhanced bacterial biofilm formation. The results suggested that RTFPs could be potential prebiotic additives used alone or supplemented with probiotic bacteria.

Probiotics play a vital role in human health, but maintaining their high viability during processing and oral administration remains challenging. In this study, we utilized inulin and tannic acid (TA)/Ca 2+ metal-phenolic networks (MPNs) as coating materials to encapsulate anaerobic Bifidobacterium lactis (BL) using a layer-by-layer self-assembly strategy to improve their tolerance to the adverse gastrointestinal microenvironment and colonization in the intestinal tract. The results demonstrated that the three-dimensional gel coating formed by inulin could protect the MPNs coating from degradation by gastric acid and improve the ability of BL to resist gastric acid and bile salts, significantly increasing the survival rate of probiotics. Additionally, MPNs could effectively scavenge free radicals and resist cell damage caused by oxidative stress. Importantly, TA in MPNs could form strong interactions (hydrogen bonds) with mucin in the intestine, greatly prolonging the residence time of BL (more than 96 h). Moreover, the raw materials used to prepare BL@TA@Inulin are food-grade, thus ensuring high biosafety. In summary, BL@TA@Inulin represents a novel probiotic single-cell encapsulation approach to achieve highly active and adhesive delivery of probiotics, with potential applications in developing healthy foods and treating gastrointestinal tract-related diseases.

Probiotics have shown potential for alleviating circadian disruption, but efficient screening models for identifying effective strains are lacking, and the mechanisms by which probiotics modulate circadian disruption and related diseases require further investigation. In this research, we developed an in vitro screening model targeting clock gene expression and identified Bifidobacterium longum CCFM1238 as an effective strain for alleviating circadian disruption in sleep-deprived mice. B. longum CCFM1238 exhibited a rhythm-regulating effect similar to melatonin in vitro and alleviated both intestinal and hypothalamic clock gene disruption (Bmal1, Clock, Per3, Cry1 and Rev-erbα) in sleep deprivation-induced circadian disruption mice. Further studies showed that B. longum CCFM1238 reduced inflammatory cell infiltration in colonic tissue, increased colonic goblet cell numbers, and repaired sleep deprivation-induced intestinal inflammation. In cognitive ability tests, B. longum CCFM1238 ameliorated spatial recognition and memory deficits induced by circadian disruption and attenuated extensive microglial activation in the hippocampal region. Additionally, B. longum CCFM1238 increased the abundance of beneficial bacteria (Akkermansia, Alistipes, and Bifidobacterium) and modulated levels of key metabolites (sphingosine, adenosine, guanosine, N,N-dimethylglycine, and inosine) strongly associated with key microbiota. These findings suggest that B. longum CCFM1238 may target clock gene expression to alleviate circadian disruption and modulate the gut microbiota to provide neuroprotection and gut barrier protection against circadian disruption.
